Aim: Adipokines seem to play a role in bone morphogenesis, although this also depends on the mechanical forces applied to the skeleton. The aim was to assess the relationships of resting leptin and adiponectin with bone parameters and whether high muscular fitness levels affect these relationships in children with overweight or obesity. Methods: This cross-sectional study took part from 2014 to 2016 in Granada, Spain. Participants were recruited from University Hospitals, and we also used advertisements in local media and school contacts in the city. Adipokines were analysed in plasma. Muscular fitness was assessed by one repetition maximum in bench and leg press tests. Dual-energy X-ray absorptiometry was used to measure bone parameters. Results: We included 84 children (10.0 ± 1.2y; 63% boys) in this analysis. Leptin was negatively associated with lumbar spine bone mineral content (β = −0.162, p = 0.053).No significant interaction was found for muscular fitness. Simple slope estimates suggested that children performing more than 133.3 kg in leg press test ameliorated the negative association between leptin and lumbar spine bone mineral content. Conclusion: Leptin levels were negatively associated with lumbar spine bone mineral content in children with overweight or obesity. A high muscular fitness at the lower body could counteract this association. By using the recently introduced parametrization of an n-dimensional density matrix in terms of the indices of population asymmetry and the intrinsic coherences, we define descriptors in both integer and continuous forms of the effective dimension that take place for a complete description of a density matrix, thus providing accurate information beyond the rank of the density matrix. The concepts of dimensional folding, hidden dimensional purity, and dimensional entropy are introduced and discussed in view of the new approach presented. The results are applicable to any physical system represented by a density matrix, such as n-level quantum systems, qutrits, sets of interacting pencils of radiation, classical polarization states, and to transformations of density matrices, as occurs with quantum channels.

The relationship between inflammatory markers and bone turnover in adults is well known, whilst a negative association between cardiorespiratory fitness (CRF) and inflammatory markers has also been described. Hence, we tested whether the association between CRF and bone turnover markers is mediated by inflammatory markers in adults with metabolic syndrome. A total of 81 adults (58.5±5.0 yrs, 62.7% women) were included in the analysis. CRF was measured by the six-minute walking test. Serum interleukine (IL)-1β, IL-6, IL-10, tumor necrosis factor alpha, high-sensitivity c-reactive protein (hsCRP) and vascular endothelial growth factor, collagen type I cross-linked C-telopeptide, procollagen type I N-terminal propeptide (P1NP) and total osteocalcin were assessed using a sensitive ELISA kit. Body composition was assessed by dual-energy x-ray absorptiometry. Partial correlation was used to test the relationship between CRF, inflammatory markers and bone turnover markers, controlling for sex, lean mass and fat mass. Boot-strapped mediation procedures were performed and indirect effects with confidence intervals not including zero were interpreted as statistically significant. CRF was positively correlated with P1NP levels (r=0.228, p=0.044) and osteocalcin levels (r=0.296, p=0.009). Furthermore, CRF was positively correlated with IL-1β levels (r=0.340, p=0.002) and negatively correlated with hsCRP levels (r=-0.335, p=0.003), whereas IL-1β levels were positively correlated with P1NP levels (r=0.245, p=0.030) and hsCRP levels were negatively correlated with P1NP levels (r=-0.319, p=0.004). Finally, the association between CRF and P1NP levels was totally mediated by hsCRP (PM=39.9). The lack of bacteriophages capable of infecting the Listeria species, Listeria grayi, is academically intriguing and presents an obstacle to the development of bacteriophage-based technologies for Listeria. We describe the isolation and engineering of a novel L. grayi bacteriophage, LPJP1, isolated from farm silage. With a genome over 200,000 base pairs, LPJP1 is the first and only reported jumbo bacteriophage infecting the Listeria genus. Similar to other Gram-positive jumbo phages, LPJP1 appeared to contain modified base pairs, which complicated initial attempts to obtain genomic sequence using standard methods. Following successful sequencing with a modified approach, a recombinant of LPJP1 encoding the NanoLuc luciferase was engineered using homologous recombination. This luciferase reporter bacteriophage successfully detected 100 stationary phase colony forming units of both subspecies of L. grayi in four hours. A single log phase colony forming unit was also sufficient for positive detection in the same time period. The recombinant demonstrated complete specificity for this particular Listeria species and did not infect 150 non-L. grayi Listeria strains nor any other bacterial genus. LPJP1 is believed to be the first reported lytic bacteriophage of L. grayi as well as the only jumbo bacteriophage to be successfully engineered into a luciferase reporter.

Background The PhageDx™Cronobacter Assay is based on the infection of Cronobacter spp. by specific bacteriophages and expression of a luciferase reporter gene. Results are generated in as little as 18.5 h for powdered infant formula (PIF). Objective An AOAC Performance Tested MethodsSM (PTM) study was conducted to validate the PhageDx Cronobacter Assay for the detection of Cronobacter in 10, 100, and 300 g milk- and soy-based PIF test portions. Method The performance of the PhageDx method was compared to the ISO 22964:2006/2017 Microbiology of the Food Chain—Horizontal Method for the Detection of Cronobacter spp. and the U.S. Food and Drug Administration (FDA) Bacteriological Analytical Manual (BAM) Chapter 29 Cronobacter: 2012. Inclusivity/exclusivity, product consistency and stability, and robustness testing also were conducted. Results There was no significant difference between the 10, 100, or 300 g test portions for the milk and soy PIF matrixes between the PhageDx Cronobacter Assay, the ISO 22964:2006/2017, and the FDA BAM Chapter 29 Cronobacter: 2012 methods. The reporter bacteriophages were specific for Cronobacter and infected 75 strains in inclusivity testing. They did not infect 35 non-Cronobacter bacteria in exclusivity testing. Robustness testing showed that the method performed well with specific deviations from the standard protocol. Consistency and stability testing demonstrated that the recombinant phage gave consistent results across three production lots and was stable when stored under appropriate conditions for at least 3 months. Conclusions Work in the submitting and independent laboratories demonstrated that the PhageDx Cronobacter Assay meets the qualifications for PTM status. Highlights The PhageDx Cronobacter Assay is a rapid, simple, and specific test that has shown equivalence to both the FDA BAM and ISO reference methods for detecting Cronobacter spp. in PIF.

Background The PhageDx™ Listeria Assay is a simple, specific, and sensitive assay based on the infection of Listeria spp. by selected bacteriophages and the resultant expression of a luciferase reporter gene. Results are generated in as little as 24.5 h for stainless steel and ceramic environmental surfaces. Objective An AOAC Performance Tested MethodsSM (PTM) study was conducted to validate the PhageDx Listeria Assay for the detection of Listeria on stainless steel and ceramic surfaces. Method The performance of the PhageDx method was compared to that of the U.S. Food and Drug Administration (FDA) Bacterial Analytical Manual (BAM) Ch. 10. Inclusivity/exclusivity, product consistency and stability, and robustness testing also were conducted. Results Inclusivity testing demonstrated that the reporter bacteriophages were specific for Listeria ssp. and detected 58/61 Listeria strains tested, including all 34 L. monocytogenes strains. The reporter bacteriophage also was shown to not detect 46/47 non-Listeria bacteria in exclusivity testing. Robustness testing showed that the method performed well with specific deviations from the standard protocol. Consistency and stability testing demonstrated that the recombinant phage gave consistent results across three production lots and was stable when stored under appropriate conditions for at least 6 months. Matrix studies on stainless steel and ceramic surfaces showed that there was no significant difference between the PhageDx Listeria Assay and the FDA/BAM Chapter 10 reference method. Conclusions and Highlights The validation study demonstrates that the PhageDx Listeria Assay is an effective method for the detection of Listeria spp. on stainless steel and ceramic environmental surfaces and meets the qualifications for AOAC PTM status.

ImportanceChildhood obesity is a risk factor for type 2 diabetes (T2D), cardiovascular disease, and mental disorders later in life. To investigate the parallel effects on cardiometabolic and mental health in children with overweight or obesity will provide new insights on the benefits of exercise on overall health.ObjectiveThis study aimed to investigate the effects of a 20-week exercise program on cardiometabolic and mental health in children with overweight or obesity.DesignParallel-group randomized clinical trial (RCT) conducted in Granada (Spain) from November 2014 to June 2016.SettingClinical setting.ParticipantsEligibility criteria included children with overweight or obesity aged 8 to 11.9 years from Granada (Spain) and surrounding areas.InterventionThe exercise program included 3-5 sessions/week (90 min/session) of aerobic plus resistance training for 20-weeks. The wait-list control group continued with their usual routines.Main Outcomes and MeasuresCardiometabolic outcomes included body composition (fat mass, fat-free mass, and visceral adipose tissue), physical fitness (cardiorespiratory, speed-agility, and muscular), and traditional risk factors (waist circumference, blood lipids biomarkers, glucose, insulin, and blood pressure). Cardiometabolic risk score (z-score) was calculated based on age and sex reference values for triglycerides, inverted high-density lipoprotein cholesterol, glucose, the average of systolic and diastolic blood pressure, and waist circumference. An additional cardiometabolic risk score also included cardiorespiratory fitness. Mental health outcomes included an array of psychological well-being and ill-being indicators.ResultsThe ActiveBrains exercise program reduced the cardiometabolic risk score by ∼0.4 (95% confidence interval [CI95%]: −0.75, −0.03) standard deviations (SD). The exercise program had a positive effect on low-density lipoprotein cholesterol (−7.40 [CI95%: −14.82, 0.016] mg/dL), body mass index (−0.60 [CI95%: −1.07, −0.13] kg/m2), fat mass index (−0.70 [CI95%: −1.03 to −0.36] kg/m2), visceral adipose tissue (−34.05 [CI95%: −61.38, −6.73] g), and cardiorespiratory fitness (+3.07 [CI95%: 0.68, 5.45] laps) in the exercise group compared to controls. No effects were observed on mental health outcomes.Conclusions and RelevanceThe ActiveBrains exercise program improved cardiometabolic health in children with overweight or obesity, yet it had no effect on mental health. Salmonella is a major causative agent of foodborne illness and rapid identification of this pathogen is essential to prevent disease. Currently most assays require high bacterial burdens or prolonged enrichment to achieve acceptable performance. A reduction in testing time without loss of sensitivity is critical to allow food processors to safely decrease product holding time. To meet this need, a method was developed to detect Salmonella using luciferase reporter bacteriophages. Bacteriophages were engineered to express NanoLuc, a novel optimized luciferase originating from the deep-sea shrimp Oplophorus gracilirostris. NanoLuc-expressing bacteriophages had a limit of detection of 10–100 CFU per mL in culture without enrichment. Luciferase reporters demonstrated a broad host range covering all Salmonella species with one reporter detecting 99.3% of 269 inclusivity strains. Cross-reactivity was limited and only observed with other members of the Enterobacteriaceae family. In food matrix studies, a cocktail of engineered bacteriophages accurately detected 1 CFU in either 25 g of ground turkey with a 7 h enrichment or 100 g of powdered infant formula with a 16 h enrichment. Use of the NanoLuc reporter assay described herein resulted in a considerable reduction in enrichment time without a loss of sensitivity.

Linear polarimetric transformations of light polarization states by the action of material media are fully characterized by the corresponding Mueller matrices, which contain in an implicit and intricate manner all measurable information on such transformations. The general characterization of Mueller matrices relies on the nonnegativity of the associated coherency matrix, which can be mathematically formulated through the nonnegativity of its eigenvalues. The enormously involved explicit algebraic form of such formulation prevents its interpretation in terms of simple physical conditions. In this work, a general and simple characterization of Mueller matrices is presented based on their statistical structure. The link between depolarization measures and physical nature and structure of material media inducing depolarization is nowadays an open question. This article shows how the joint use of two complementary sets of depolarizing metrics, namely the Indices of polarimetric purity and the Components of purity, are sufficient to completely describe the integral depolarizing properties of a sample. Based on a collection of illustrative and representative polarimetric configurations, a clear and meaningful physical interpretation of such metrics is provided, thus extending the current tools and comprehension for the study and analysis of the depolarizing properties of material media. This study could be of interest to those users dealing with depolarization or depolarizing samples.

We analyze the spin structure of weakly and tightly focused random light beams in view of the characteristic decomposition of the $3\ifmmode\times\else\texttimes\fi{}3$ polarization matrix. We take the incident beam to be a mixture of mutually uncorrelated, orthogonally linearly polarized parts which entails zero spin for the beam but leads to a nonzero purely transverse spin in focusing via the generation of a longitudinal electric-field component. We show that the spin ensuing from the so-called discriminating component of the polarization matrix displays a vortex structure for any degree of polarization and level of focusing, while the spin distribution of the pure component may vary from linear to circular as the incident beam changes from fully polarized to completely unpolarized. The spin of the discriminating component is related to the recently introduced concept of polarimetric nonregularity and emerges only when the degree of nonregularity is different from zero. Our results demonstrate the relationship between the rich polarimetric structure of random three-component light and the composition of the optical electric-field spin.

Polarimetry is today a widely used and powerful tool for nondestructive analysis of the structural and morphological properties of a great variety of material samples, including aerosols and hydrosols, among many others. For each given scattering measurement configuration, absolute Mueller polarimeters provide the most complete polarimetric information, intricately encoded in the 16 parameters of the corresponding Mueller matrix. Thus, the determination of the mathematical structure of the polarimetric information contained in a Mueller matrix constitutes a topic of great interest. In this work, besides a structural decomposition that makes explicit the role played by the diattenuation-polarizance of a general depolarizing medium, a universal synthesizer of Muller matrices is developed. This is based on the concept of an enpolarizing ellipsoid, whose symmetry features are directly linked to the way in which the polarimetric information is organized.

Background ThePhageDx™ E. coli O157:H7 Assay originally required a 5 h enrichment period and a 10 mL test sample for 25 g ground beef. The proposed method modifications seek to include beef trim (375 g) matrix and a new method procedure for 25 g ground beef comprised of 6–7 h enrichment and 1 mL test sample. Objective To validate the method modifications for PhageDx E. coli O157:H7 Assay to include beef trim (375 g) matrix and modify enrichment time and test sample size for 25 g ground beef. Method For both matrixes, pre-warmed tryptic soya broth (TSB; 42 ± 1°C) was added in a 3:1 (media:sample size) ratio, blended, and enriched for 9–10 h (beef trim, 375 g) or 6 h (ground beef, 25 g) at 42 ± 1°C. One milliliter samples were transferred to a microfuge tube, centrifuged, and the supernatant removed. The pellet was resuspended in 0.2 mL of TSB and phage reagent was added. Samples were incubated for 2 h at 37 ± 1°C. After infection, the sample was centrifuged, and 150 µl of the supernatant was transferred to a 96-well plate. Then, lysis buffer and luciferase substrate were added and the sample read on a luminometer to determine the presence or absence of E. coli O157:H7. Results No significant differences were found between the PhageDx E. coli O157:H7 Assay method modifications and the U.S. Department of Agriculture Food Safety and Inspection Service (FSIS) Microbiology Laboratory Guidebook (MLG) reference method. Conclusions The independent study demonstrated that the PhageDx E. coli O157:H7 Assay method modifications meet the qualifications for Performance Tested Method SM status. Highlights The PhageDx E. coli O157:H7 Assay is a rapid detection method capable of detecting E. coli O157:H7 in 25 g ground beef and 375 g beef trim samples in as little as 8.5 h and 11.5 h total turn around time, respectively.

Light interaction with material systems may introduce depolarization to the incident light. This phenomenon comes from multiple scattering processes that take place inside the media and strongly depends on the particle characteristics. In the case of botany, plant leaves can be understood as depolarizing systems. A non-contact method to analyze these samples consist of illuminating them with well-known polarized light and study the scattered light to retrieve the physical characteristics of the sample. This physical study can be done by measuring the Mueller matrix of samples, in which the physical information of samples is encoded in their 16 elements and further mathematical treatment is required to extract the information. In the case of scattering systems, the depolarization content carries very valuable information but it is usually not inspected in the botanic field. A way to study depolarized content is by determining the so-called depolarization index PΔ, which gives an overall measure of the degree of depolarization of a system but it does not measure possible anisotropic dependence of the depolarization. For instance, a depolarizer equally depolarizing any fully polarized input polarization or a depolarizer that depolarizes them in a strongly heterogenous way, may lead to the same PΔ value. In contrast, the Indices of Polarimetric Purity (IPP) are a group of metrics that further synthesize the depolarizing content, taking into account the anisotropic depolarization. In this work, we describe the main physical characteristics of samples achieved by using these IPP through plant samples. Moreover, we show how IPP highlights some structures hidden in regular intensity measurements, highlighting the potential of these metrics for botanical applications.

Complete characterization of biological samples is of potential interest in different industrial and research areas, as for instance, in biomedical applications, for the recognition of organic structures or for the early detection of some diseases. During the last decades, polarimetric methods are experiencing an increase of attention in the study of biomedical tissues, and they are nowadays used in such framework to provide qualitative (polarimetric imaging) and quantitative (data processing) information for the studied samples. Polarimetric methods are based on the analysis of polarization modifications produced by light-matter interactions which can be triggered by a number of complex internal processes but can be roughly understood as the result of the combination of three pure polarimetric features of the sample: its diattenuation, retardance and depolarization. For the analysis of the depolarization content, we propose the use of the Indices of Polarimetric Purity (IPP) to describe the sample behavior. Related with the randomness of the scattering processes, IPPs provide more information of depolarizing systems than the widely used depolarization index (p^), which further synthetize the depolarization content of samples. Moreover, certain combinations of IPP parameters leads to p^. As a result, IPPs allow the revelation of some structures from tissue samples hidden in regular intensity images of even in the p^ channel, leading to better tissue classification results. In this work, we present different applications of IPPs in biomedical tissue that show its potential, which are not restricted to the biomedical framework as relevant results in plants characterization are also presented.

Optical microscopy techniques for plant inspection benefit from the fact that at least one of the multiple properties of light (intensity, phase, wavelength, polarization) may be modified by vegetal tissues. Paradoxically, polarimetric microscopy although being a mature technique in biophotonics, is not so commonly used in botany. Importantly, only specific polarimetric observables, as birefringence or dichroism, have some presence in botany studies, and other relevant metrics, as those based on depolarization, are underused. We present a versatile method, based on a representative selection of polarimetric observables, to obtain and to analyse images of plants which bring significant information about their structure and/or the spatial organization of their constituents (cells, organelles, among other structures). We provide a thorough analysis of polarimetric microscopy images of sections of plant leaves which are compared with those obtained by other commonly used microscopy techniques in plant biology.
