Abstract Objectives Bovine seminal plasma proteins perform several functions related to sperm function. Changes in the expression pattern or abundance of seminal proteins are related to changes in the fertilizing capacity of bulls. Considering the role of seminal plasma proteins in sperm function and animal reproduction, we investigated changes in the protein abundance profile in response to sperm morphological changes using a proteomic approach. Datadescription In our present investigation, we employed liquid chromatography coupled with mass spectrometry to elucidate the proteomic composition of seminal plasma obtained from Nellore bulls exhibiting varying percentages of sperm abnormalities. Following semen collection, seminal plasma was promptly isolated from sperm, and proteins were subsequently precipitated, enzymatically digested using porcine trypsin, and subjected to analysis utilizing the Acquity nano UHPLC System in conjunction with a mass spectrometer. This dataset encompasses a total of 297 proteins, marking the inaugural instance in which a comparative profile of seminal plasma proteins in young Nellore bulls, categorized by their sperm abnormality percentages, has been delineated using LC-MS/MS. The comprehensive nature of this dataset contributes pivotal proteomic insights, representing a noteworthy advancement in our understanding of the reproductive biology of the Nellore breed.

ABSTRACT: Anti-Müllerian hormone (AMH) concentration and number of recovered oocytes (ROOC) are phenotypic parameters associated with in vitro embryo production (IVEP). More recently, anogenital distance (AGD) has been proposed as a proxy for fertility in dairy cattle that is easy to collect at a low cost. The aim of this study was to characterize the AGD and its phenotypic and genetic associations with AMH and IVEP in Bos indicus Gyr dairy cattle. The hypothesis was that the number of ROOC, in vitro–produced embryos, and AMH concentration would increase as the AGD decreases. From July to December 2021, a single morphometrical measurement of AGD was collected in 552 donors from 6 herds in Brazil. A subset of donors had AMH assayed on the same day. Only ovum pick-up events that occurred up to 12 mo preceding and 7 mo succeeding the AGD measurement were used to assess the association between AGD, AMH, and IVEP. Thus, 472 donors (1,551 ovum pick-up events and 140 donors with AMH) were considered in the analysis. A raw average was calculated for each individual donor's ROOC, viable oocytes, total produced embryos, viability rate, and embryo rate (defined as total produced embryos/viable oocytes). Abstract Horses are seasonal polyoestrous animals, and the photoperiod is the main factor modulating their reproductive activity. There is no consensus on the andrological and biochemical factors that influence breeding seasonality. To assess the involvement of climate in reproduction, Mangalarga Marchador stallions were monitored over 1 year regarding semen quality and seminal plasma proteome. Here, we show that kallikrein (KLKs) proteoforms in seminal plasma are involved in climate conditioning of reproduction. During the breeding season, greater abundance and different types of KLKs occurred simultaneously to lower sperm motility, greater semen volumes and higher concentrations of glucose and cholesterol. Considering that vasodilation due to activation of the kallikrein-kinin system and the consequent inhibition of the renin-angiotensin system may be associated with lower sperm motility, unravelling the involvement of KLK proteoforms in reproductive seasonality is a priority in horse breeding.

This study aimed to evaluate the proteomic profile of seminal plasma from young Nellore bulls. We used 20 bulls aged between 19.8 and 22.7 months, divided into two groups according to the results of the Breeding Soundness Evaluation (BSE): approved (FIT n = 10) and not approved (UNFIT n = 10). The scrotal perimeter was measured and a semen collection was performed through electroejaculation. The percentage of sperm motility, mass motility, and sperm vigor were calculated using conventional microscopy, and the percentage of sperm abnormalities was calculated using phase-contrast microscopy of all ejaculates. Seminal plasma was separated from spermatozoa using centrifugation and processed for proteomic analysis by LC-MS/MS. Seminal plasma proteins were identified using MASCOT Daemon software v.2.4.0 and label-free quantification analysis was carried out by SCAFFOLD Q+ software v.4.0 using the Exponentially Modified Protein Abundance Index (emPAI) method. Functional classification of proteins was performed based on their genetic ontology terms using KOG. Functional cluster analysis was performed on DAVID. There were no differences in scrotal perimeter and physical semen characteristics between FIT and UNFIT groups of bulls. The percentage of sperm abnormalities was higher (p < 0.05) in the UNFIT group of bulls. A total of 297 proteins were identified for the two groups. There were a total of 11 differentially abundant proteins (p < 0.05), two of them more abundant in FIT bulls (Spermadhesin-1 and Ig gamma-1 chain C region) and nine in UNFIT bulls (Vasoactive intestinal peptide, Metalloproteinase inhibitor 2, Ig lambda-1 chain C regions, Protein FAM3C, Hemoglobin beta, Seminal ribonuclease, Spermadhesin 2, Seminal plasma protein BSP-30kDa, and Spermadhesin Z13). Spermadhesin-1 was the protein with the highest relative abundance (36.7%) in the seminal plasma among all bulls, corresponding to 47.7% for the FIT bulls and 25,7% for the UNFIT bulls. Posttranslational modification, protein turnover, and chaperones were the functional categories with the highest number of classified proteins. Protein functional annotation clusters were related to Phospholipid efflux, ATP binding, and chaperonin-containing T-complex. The differentially abundant proteins in the group of FIT bulls were related to sperm capacitation and protection against reactive species of oxygen. In contrast, differentially expressed proteins in the group of UNFIT bulls were related to motility inhibition, intramembrane cholesterol removal and oxidative stress. In conclusion, the proteomic profile of the seminal plasma of FIT bulls presents proteins with participation in several biological processes favorable to fertilization, while the proteins of the seminal plasma of UNFIT bulls indicate a series of alterations that can compromise the fertilizing capacity of the spermatozoa. In addition, the relative abundance of spermadhesin-1 found in the seminal plasma of young Nellore bulls could be studied as a reproductive parameter for selection.

Classical in vitro fertilization (IVF) is still poorly successful in horses. This lack of success is thought to be due primarily to inadequate capacitation of stallion spermatozoa under in vitro conditions. In species in which IVF is successful, bicarbonate, calcium, and albumin are considered the key components that enable a gradual reorganization of the sperm plasma membrane that allows the spermatozoa to undergo an acrosome reaction and fertilize the oocyte. The aim of this work was to comprehensively examine contributors to stallion sperm capacitation by investigating bicarbonate-induced membrane remodelling steps, and elucidating the contribution of cAMP signalling to these events. In the presence of capacitating media containing bicarbonate, a significant increase in plasma membrane fluidity was readily detected using merocyanine 540 staining in the majority of viable spermatozoa within 15 min of bicarbonate exposure. Specific inhibition of soluble adenylyl cyclase (sAC) in the presence of bicarbonate by LRE1 significantly reduced the number of viable sperm with high membrane fluidity. This suggests a vital role for sAC-mediated cAMP production in the regulation of membrane fluidity. Cryo-electron tomography of viable cells with high membrane fluidity revealed a range of membrane remodelling intermediates, including destabilized membranes and zones with close apposition of the plasma membrane and the outer acrosomal membrane. However, lipidomic analysis of equivalent viable spermatozoa with high membrane fluidity demonstrated that this phenomenon was neither accompanied by a gross change in the phospholipid composition of stallion sperm membranes nor detectable sterol efflux (p > 0.05). After an early increase in membrane fluidity, a significant and cAMP-dependent increase in viable sperm with phosphatidylserine (PS), but not phosphatidylethanolamine (PE) exposure was noted. While the events observed partly resemble findings from the in vitro capacitation of sperm from other mammalian species, the lack of cholesterol removal appears to be an equine-specific phenomenon. This research will assist in the development of a defined medium for the capacitation of stallion sperm and will facilitate progress toward a functional IVF protocol for horse gametes.

Objetivou-se identificar a prevalência das principais patologias que acometem os órgãos genitais de tourinhos jovens. Foi utilizado um banco de dados de 6408 touros jovens da raça Nelore, nos meses de junho a agosto de 2004 e 2005. Os tourinhos foram avaliados por meio de exame andrológico e o sêmen coletado com auxílio do eletroejaculador. A maturidade sexual dos animais foi classificada em: animais aptos à reprodução, animais aptos à reprodução em regime de monta natural, animais temporariamente inaptos à reprodução e animais descartados. As afecções reprodutivas registradas nos animais descartados foram distribuídas em: alterações de pênis, testículos, membros anteriores ou posteriores, epidídimo, cordão espermático, alterações espermáticas, alterações do esfíncter anal, prepúcio, sistêmicas e de vesícula seminal. Análises descritiva e de Variância foram realizadas para análise dos dados. Dos 6408 touros avaliados, 309 animais foram descartados da reprodução (4,82%). A maioria dos touros descartados (31,7%) apresentou patologias relacionadas ao testículo (assimetria testicular, baixo perímetro escrotal para a idade, orquite e hidrocele, com valores de 64,28, 32,65, 2,04 e 1,02%, respectivamente), e vesículas seminais (31,39%), seguidas pelos defeitos espermáticos (17,15% dos descartados) e alterações de pênis (6,79%). Dos 309 animais descartados da reprodução, 21 (6,79% dos descartados e 0,32% do total de animais) apresentaram algum tipo de alteração peniana como persistência do frênulo (16/21) e aderências penianas, total ou parcial (5/21). O confinamento do rebanho por dois meses e a redução da idade dos animais para realização do primeiro exame andrológico, com objetivo de venda de animais precoces, podem ser os responsáveis pela maior incidência de patologias relacionadas às vesículas seminais (devido ao comportamento de sodomia dos machos) e pênis com alta incidência de aderências, característicos de animais que ainda não entraram na puberdade. De maneira geral, a baixa porcentagem de animais descartados da reprodução no rebanho estudado, indica adequado programa de seleção.

The aim of the study was to evaluate the effect of recombinant bovine somatotropin (rbST) and thyroxine (T4) on survival and growth of bovine preantral ovarian follicles (PAOF) cultured in vitro. Ovarian fragments were collected in local abattoirs and immediately fixed for classical histology and transmission electron microscopy (non-cultured control). The other fragments were then cultured in situ for seven days in minimum essential medium alone (MEM+ - cultured control) or in the presence of 1,000 ng/mL rbST and 20 ng/mL T4, isolated or associated. After seven days, there was a reduction (P0.01) of the medium used on the follicular diameter of the PAOF cultured for seven days. Ultrastructural analysis showed cell damage. In conclusion, the presence of rbST maintains the rate of morphologically normal follicles during the culture for seven days (observed by optical microscopy), but it does not exert beneficial effects on its ultrastructural integrity and oocyte and follicular growth.

This study aimed to evaluate the endometrial angiogenesis of pregnant commercial line and Piau gilts during early pregnancy. We used 27 gilts, divided into three groups according to the type of mating: Commercial (n = 9), commercial line females mated with commercial line males; Cross-mated (n = 9), Piau females mated with commercial line males; and Piau (n = 9), Piau females mated with Piau males. Each group was divided into three subgroups based on gestational age at the time of slaughter (7, 15, and 30 days of pregnancy). Immediately after slaughter, endometrial samples were obtained for histological evaluation and for analysis of the relative transcript abundance (RTA) of angiogenesis-related genes (HIF1α, FGF9, ANG1, TEK, VEGFA, ANGPT1, and ANGPT2). The number of endometrial glands was similar among groups but decreased with gestational age (p < 0.05). Piau females showed a higher number of blood vessels (p < 0.05) at 7 and 15 days of pregnancy, but no differences were observed among groups at 30 days, suggesting an influence of the male genotype on the pattern of uterine vascularization. There were no differences among groups for RTA of the FGF9, HIF1α, TEK, VEGFA, ANGPT1, and ANGPT2 genes. The HIF1α-gene RTA was higher at 7 and 15 days of pregnancy; for TEK and ANGPT1, the RTA was higher at 15 days of pregnancy; and the RTA of VEGFA and ANGPT2 genes were higher at 30 days of pregnancy. The ANG1 RTA was similar for pregnancies in the commercial and Piau groups but was higher (p < 0.05) at 15 days in the Cross-mated group, suggesting an interaction between genotypes. Overall, the pattern found for the RTA of angiogenesis-related genes was similar among the groups in this study, although some phenotypic differences could be noted, such as the highest number of blood vessels being found during early pregnancy of Piau gilts. The results of the gene RTA when crossed with phenotypic data led to conclusions that are conflicting with those reported in the literature. However, noteworthy is that angiogenesis is a complex process in which the balance between stimulatory and inhibitory factors may be related to time.

Evaluations of replacement heifers in intensively managed grazing systems in tropical conditions are warranted. Thus, we aimed to evaluate performance, muscle and mammary gland development, oocyte quality, and in vitro production of embryos of crossbred heifers grazing an intensively managed pasture and supplemented with high or low protein concentrates. Eighteen pubertal crossbred heifers (Holstein x Gyr) with an initial weight of 350 ± 8.0 kg were used in a 60-day trial. Two supplement types, 12% crude protein (CP) (S12CP) or 24% CP (S24CP), and a control treatment (mineral mixture, CON) were randomly distributed to the heifers. Throughout the experiment, four digestibility trials were performed over four consecutive days. Four ovarium pick-ups were performed to evaluate oocyte quality and in vitro embryo production. Lastly, ultrasounds of carcasses and mammary glands were performed. The intakes of dry matter (DM), digestible energy (DE), and CP were greater for supplemented (SUP) compared with CON heifers. The SUP heifers had a greater average daily gain (ADG) (645 versus 390 g/d) and rib eye area (58.78 versus 53.32 cm2) than the CON heifers. Oocyte recovery, quality, and follicle features were not affected by supplementation strategy. However, the cleavage rate (47.17% versus 30.31%) and blastocyst rate (27.91% versus 10.12%) were negatively affected by supplementation. The S12CP presented a blastocyst rate much lower than the S24CP (3.02% versus 17.23%). Carcass ultrasonography indicated a trend for greater rib eye area for S24CP and mammary ultrasonography indicated no effects of supplementation on mammary gland development. In summary, supplementation seems to be an appropriate strategy for satisfactory performance, with greater muscle deposition and no negative impacts on mammary gland development. However, in vitro embryo production was impaired when the animals received the supplementation with 12% CP.

Bovine herpesvirus 1 (BHV1) is an important bovine pathogen, responsible for respiratory diseases and reproductive problems. This study investigated the penetration capacity of BHV1 into oocytes after co-incubation for either 1 h or 24 h. Immunofluorescence assays in cumulus-oocyte complexes (COCs) and denuded oocytes (without the presence of cumulus cells) were performed and evaluated using confocal laser scanning microscopy. Blood samples and ovaries from BHV1 seronegative cows were used. The oocytes recovered were divided into two groups. Group I comprised COCs (n = 312) and denuded oocytes (n = 296), which were experimentally infected with BHV1 and incubated for 1 h at 38.5°C and 5% CO2. Group II comprised COCs (n = 425) and denuded oocytes (n = 405), which were co-incubated with BHV1 under the same conditions for 24 h. The negative control of these two groups was respectively subjected to the same protocol, except for exposure to BHV1. To our knowledge, this study provides the first evidence of BHV1 detection within COCs and denuded oocytes exhibiting intact zona pellucida when co-incubated with the virus for 24 h. Immunolocalization also confirmed the presence of BHV1 in the cytoplasm of the cumulus cells of all COCs exposed to the virus after both incubation periods. In conclusion, detection of BHV1 inside oocytes has a great meaning for the field of animal reproduction. The detection of BHV1 in different layers of cumulus cells also demonstrates that these cells are sources of viral infection.

Nonlinear mixed models were used to describe longitudinal scrotal circumference (SC) measurements of Nellore bulls. Models comparisons were based on Akaike's information criterion, Bayesian information criterion, error sum of squares, adjusted R2 and percentage of convergence. Sequentially, the best model was used to compare the SC growth curve in bulls divergently classified according to SC at 18-21 months of age. For this, bulls were classified into five groups: SC < 28cm; 28cm ≤ SC < 30cm, 30cm ≤ SC < 32cm, 32cm ≤ SC < 34cm and SC ≥ 34cm. Michaelis-Menten model showed the best fit according to the mentioned criteria. In this model, β1 is the asymptotic SC value and β2 represents the time to half-final growth and may be related to sexual precocity. Parameters of the individual estimated growth curves were used to create a new dataset to evaluate the effect of the classification, farms, and year of birth on β1 and β2 parameters. Bulls of the largest SC group presented a larger predicted SC along all analyzed periods; nevertheless, smaller SC group showed predicted SC similar to intermediate SC groups (28cm ≤ SC < 32cm), around 1200 days of age. In this context, bulls classified as improper for reproduction at 18-21 months old can reach a similar condition to those considered as good condition. In terms of classification at 18-21 months, asymptotic SC was similar among groups, farms and years; however, β2 differed among groups indicating that differences in growth curves are related to sexual precocity. In summary, it seems that selection based on SC at too early ages may lead to discard bulls with suitable reproductive potential.

O objetivo deste trabalho foi avaliar o efeito de dietas com diferentes concentrações de fósforo disponível sobre o perfil metabólico e o peso corporal de leitões em fase inicial de crescimento, mantidos em ambientes térmicos distintos. Foram utilizados 120 leitões machos castrados, de linhagem comercial e com peso corporal médio de 15 kg. Os tratamentos consistiram de dietas com 0,107, 0,214, 0,321, 0,428 e 0,535% de fósforo disponível, e de dois ambientes: termoneutro e quente. O peso corporal foi determinado no início e no final da fase experimental, e amostras de sangue foram coletadas para a medição de eletrólitos, albumina, proteína total, substâncias nitrogenadas, fosfatase alcalina total e óssea, hormônios tireoidianos, glicose e lactato. Tanto as dietas quanto os ambientes produziram efeito sobre o ganho de peso corporal e sobre os valores de sódio, potássio, cálcio total, magnésio, fósforo, fosfatase alcalina total e óssea, T4 livre e lactato. O aumento nos teores de fósforo na dieta tem efeito positivo sobre o peso corporal, nos ambientes estudados, mas desequilibra as relações sanguíneas cálcio:fósforo. O estresse por calor diminui as concentrações de glicose, fosfatase alcalina total e T4 livre no sangue, e aumenta as de lactato. Parâmetros bioquímicos sanguíneos retratam o estado metabólico dos animais e podem ser usados para avaliação do balanço nutricional e para detecção precoce de alterações metabólicas.

E-mail: hugoshiomi@hotmail.com The objective of this study was to verify three protocols of semen cryopreservation on spermatic viability after thawing from Piau swine breed (Sus scrofa), by thermo resistant test (TTR). Twenty two ejaculates from 5 Piau mature boars were collected by glove hand technique. To freezing, the ejaculates was split and submitted to three protocols (P): (P1) freezing method proposed by Furst et al. (2005), altered by diluent media, (P2) freezing method proposed by Furst et al. (2005), altered by cooled curve; and (P3) freezing method proposed by Ohata et al. (2001). After thawing, semen was submitted to TTR, been incubated at 37o C by 2 hours, motility sperm (MOT) and vigor (VIG) was analyzed at 30 minutes of interval. MOT and VIG after thawing was 20.9±12.4, 29.5±10.9 and 49.5±12.1%; 2.5±0.5, 2.9±0.4 and 3.4±0.4, respectively to P1, P2 and P3. The TTR results show gradually decrease of motility and vigor along 2 hours of test procedure utilized, with best average to protocol 3 at all time of analyze. The protocol 3 tested by Piau boars shows highest values in cellular semen cryopreservation.

The objective of this study was to investigate the relationship between testicular echotexture and seminal quality, in addition to evaluate the testicular parenchyma echogenicity pattern in young Montana bulls classified as sound and unsound for breeding. Fifty-two young Montana Tropical Compound bulls from 22 to 33 months of age were used. The animals were previously evaluated by breeding soundness evaluation and thereafter divided into two groups of breeding soundness classes: 1 = 16 animals sound for breeding; 2 = 36 animals unsound for breeding by means of physical and morphological analysis of semen. All animals underwent an ultrasound examination of the testes, and the images were analyzed with the software "Image J". ANOVA was used for statistical analysis, to determine the effect of groups in relation to testicular measurements, physical and morphological semen features and ultrasound pixel analysis. There was a difference between scrotal circumference between classes, with 39.7±2.1 cm and 37.3±3.1 cm for sound and unsound animals, respectively. Regarding testicular echotexture, mean values for the pixel intensity were 95.7 and 94.0 for sound and unsound animals for breeding, respectively, with no difference between the classes. None of the studied parameters were correlated with each other, indicating that the quantification of the pixel intensity in testicular echotexture was not effective in determining the degree of sexual maturity in Montana Tropical Compound bulls.

The objective of this study was to verify the effect of three protocols of cryopreservation on the sperm viability post-thawing of swine from the Piau breed (Sus scrofa), by means of evaluation assays in vitro and in vivo. Twenty-two ejaculates of 5 adult males of the Piau breed trough the gloved-hand method. In order to verify the sperm viability of the raw semen and post-thawing, the following traits were evaluated: sperm motility and vigor, hypoosmotic test, supra-vital color test and percentage of normal acrosomes. To freezing, the ejaculates were fractioned and submitted in: method 1, method of freezing recommended by Fürst et al. (2005), modified according to media extender; method 2, recommended by Fürst et al. (2005), modified according to the cooling curve; and method 3, recommended by Ohata et al. (2001). To test the in vivo fertility of the frozen/thawed semen, 23 intra-uterine (post-cervical) inseminations were performed in 14 females of the Piau breed, with semen from method 3. The mean values obtained with the use of methods 1 and 2 and post-thawing were inferior to those obtained with method 3. Four adult sows got pregnant (28.6% fertility rate). The freezing method recommended by Ohata et al. (2001), with balance period of 90 minutes at 22-26 ºC, enables higher results of in vitro sperm viability in comparison with the two other methods. The absence of balance period of method 1 and the cooling curve utilized in protocol 2 affect negatively the parameters of sperm viability evaluated in the present study. In spite of the satisfactory results in vitro for the freezing of semen with the utilization of method 3, it presents low fertility rate (28.6%).Objetivou-se verificar o efeito de três protocolos de criopreservação sobre a viabilidade espermática pósdescongelamento de suínos da raça Piau (Sus scrofa), por meio de testes de avaliação in vitro e in vivo. Foram coletados 22 ejaculados de 5 machos adultos da raça Piau por meio do método da mão enluvada. Para verificar a viabilidade espermática do sêmen in natura e pós-descongelamento, foram avaliados a motilidade e o vigor espermático, o teste hiposmótico, a coloração supra-vital e a porcentagem de acrossomas normais. Para o congelamento, os ejaculados foram fracionados e submetidos em: protocolo 1, método de congelamento preconizado por Fürst et al. (2005), modificado quanto aos meios diluentes; protocolo 2, método preconizado por Fürst et al. (2005), modificado quanto à curva de resfriamento; e protocolo 3, método preconizado por Ohata et al. (2001). Para testar a fertilidade in vivo do sêmen congelado/descongelado, foram realizadas 23 inseminações intrauterinas (pós-cervicais) em 14 fêmeas da raça Piau com o sêmen proveniente do protocolo 3. Os valores médios obtidos com o uso dos protocolos 1 e 2 pós-descongelamento foram inferiores aos obtidos com o protocolo 3. Quatro porcas adultas ficaram gestantes (28,6% de taxa de fertilidade). O protocolo de congelamento preconizado por Ohata et al. (2001), com período de equilíbrio de 90 minutos a 22-26 ºC, permite obter melhores resultados de viabilidade espermática in vitro em comparação aos outros dois protocolos. A ausência de período de equilíbrio do protocolo 1 e a curva de resfriamento utilizada no protocolo 2 afetam negativamente os parâmetros de viabilidade espermática avaliados no presente estudo. Apesar dos resultados satisfatórios in vitro para o congelamento de sêmen com a utilização do protocolo 3, o mesmo apresenta baixa taxa de fertilidade (28,6%).

Aiming to determine the relationship between the frequency of testicular shape and the andrological aspects in young Nellore bulls, 18,676 animals were assessed. All andrological examinations were performed between the years 2000 and 2008. Animals were classified as able for breeding, able for breeding in natural mating system, unable for breeding and discarded. The testicular shape was classified as long, fairly long, oval-long, spherical-oval, and spherical. The analysis of Pearson correlations was performed for testicular shape with scrotal circumference, testicular volume, progressive motility, sperm vigor, major defects, minor defects and total defects. Testicles with oval shape prevailed (99.61%). It was obseved that 76.34; 66.34; 64.34; 58.33 and 50.00% of the animals were classified as sound for breeding for shapes long, fairly long, oval-long, spherical-oval, and spherical, respectively. Correlations between testicular shape with scrotal circumference, testicular volume, progressive motility, sperm vigor, major, minor and total defects were 0.26; 0.08; 0.00; 0.11; -0.02; 0.02 and -0.01, respectively. Testicular shape had no influence upon the andrological examination results. Testicles of long shape were prevalent within the population.

Objetivou-se determinar a idade à puberdade e maturidade sexual e sua relação com peso corporal e mensurações testiculares em tourinhos da raça Simental criados sob regime extensivo em clima tropical. Utilizaram-se 44 tourinhos com 1 a 30 meses de idade. As mensurações corporais e testiculares foram feitas mensalmente e os ejaculados coletados quinzenalmente, a partir dos seis meses de idade, utilizando-se o método de eletroejaculação. Observou-se idade à puberdade aos 13,42 ± 3,02 meses e maturidade sexual aos 21,43 ± 6,6 meses de idade, com peso corporal e perímetro escrotal, respectivamente, de 324,2 ± 23,7 kg e 26,90 ± 1,41 cm para a puberdade e 360 ± 69,04 kg e 34,88 ± 3,9 cm para maturidade sexual. Este estudo revelou idade tardia para a cronologia dos eventos reprodutivos, com relação direta do tipo de manejo adotado.The objective of this study was to determine the age at puberty and sexual maturity and its relationship with body weight and testicular measurements in Simmental young bulls raised under pasture conditions in a tropical climate. Forty-four young bulls at 1 to 30 months of age were used. Body and testicular measurements were made monthly and ejaculates collected fortnightly, from six months of age by using the method of electroejaculation. Age at puberty was 13.42 ± 3.02 months and sexual maturity was observed at 21.43 ± 6.6 months of age, with body weight and scrotal circumference of 324.2 ± 23.7 kg and 26.90 ± 1.41 cm for puberty and 360 ± 69.04 kg and 34.88 ± 3.9 cm to sexual maturity, respectively. This study found late age to the timing of reproductive events, with direct relation to the type of management adopted.

O objetivo neste estudo foi avaliar diferentes osmolaridades e tempos de incubação durante o teste hiposmótico e suas correlações com a congelabilidade do sêmen de touros zebuínos. Foram utilizados 30 ejaculados de seis touros adultos da raça Nelore. No sêmen in natura foi feita avaliação física e morfológica, classificação quanto à coloração supravital e o teste hiposmótico. No teste hiposmótico foram utilizadas soluções com osmolaridades de 60, 100, 150 mOsm/kg e água destililada (19 mOsm/kg) om 15, 30 e 60 minutos de período de incubação a 37ºC. Após a criopreservação, as amostras foram descongeladas e avaliadas quanto aos testes hiposmótico, à coloração supravital, ao teste de termorresistência lento e à coloração fluorescente. Não foram detectadas interações entre os tempos e as osmolaridades. Não houve diferenças para os valores médios de espermatozoides reativos incubados nas soluções com diferentes osmolaridades para o sêmen congelado/ descongelado. Apenas a utilização de água destilada determinou diferença nos valores médios obtidos no teste hiposmótico realizado no sêmen in natura. Não houve diferença entre os valores médios de espermatozoides reativos incubados com diferentes tempos de incubação, tanto no sêmen in natura como no sêmen congelado/descongelado. O teste hiposmótico pode ser realizado com 15 minutos de incubação, e com uma solução que varie a osmolaridade de 60 a 150 mOsm/kg, tanto no sêmen in natura como no sêmen congelado/descongelado.The objective of this study was to evaluate different osmolarities and incubation times during the hiposmotic test and its correlation with cryopreservation of semen of Zebu bulls. It was used 30 ejaculates from six mature Nelore bulls. It was performed physical and morphological evaluation, supravital staining classification and hiposmotic test in the fresh semen. In the hiposmotic test, solutions with osmolarities of 60, 100, 150 mOsm/kg and distilled water (19 mOsm/kg) were used at 15, 30 and 60 minutes of incubation at 37ºC. After cryopreservation, the samples were thawed and evaluated for hiposmotic test, supravital staining, slow termoresistance test and for fluorescent stain. No interactions were detected between incubation times and solution osmolarities. There were no differences in average values of reactive sperm incubated in different solutions with different osmolarities for frozen/thawed semen. Only the use of distilled water determined differences in the average values obtained in the hiposmotic test performed in fresh semen. There was no difference between the mean values of reactive spermatozoa incubated with different incubation time in both fresh and frozen/thawed semen. Hiposmotic test can be accomplished in 15 minutes of incubation time and with a solution which varies osmolarity from 60 to 150 mOsm/kg in both fresh and frozen/thawed semen.

To evaluate the effect of using urea in the diet on ovarian follicular dynamics and plasma urea and progesterone concentration, an experiment was carried out with 29 Alpine goats randomly distributed into four diets, each one containing 0%; 0.73%; 1.46% or 2.24% of urea on total dry matter. The follicular dynamics was monitored daily by ultrasonography. The estrous cycles were characterized by occurrence of two, three, four or five follicular waves. The emergence day of the first follicular wave in cycles with two waves, the length of the third wave and the maximum diameter of the largest follicle of the third wave in cycles with three waves presented a quadratic behavior according to the percentage of urea in the diets. In cycles with four waves, the urea intake did not influence the follicular dynamics. The interestrus and interovulatory periods and the luteal and follicular phases length did not differ among treatments. The plasma urea concentration did not differ among diets neither among collection weeks. Plasma progesterone concentration on the estrus day and on the 11th day after estrus presented quadratic behavior, and on the 15th day after estrus, it linearly reduced acording to the percentage of urea in the diets. The supply of urea in diets at levels up to 2.24% in dietary dry matter did not influence the dry matter intake and the urea plasma concentration. For goats, the emergence day and the length of the waves, the follicular diameter and the plasma progesterone concentration on the estrus day and in the latter stages of diestrus were altered when there was supply of urea in the diet.

Objetivou-se avaliar os efeitos da adição de fontes de óleo e dos níveis de suplementação de vitamina E na ração sobre a criopreservação de sêmen suíno. Foram utilizados 24 suínos machos reprodutores Dalboar 85, com idades de 12 a 18 meses, distribuídos em delineamento inteiramente casualizado, em arranjo fatorial 2 × 3, com duas fontes de óleo (soja e salmão) e três níveis de vitamina E (150, 300 e 450 mg/kg). O sêmen foi coletado semanalmente (1 coleta/macho), diluído com meio centrifugador (1:1) e mantido a 25ºC por 2 horas. Após centrifugação (800 g, 10 minutos), o pélete foi suspenso com meio de congelamento para concentração final de 1 × 10(9) espermatozoides/mL. O sêmen foi envasado em palhetas de 0,5 mL e colocado dentro de tubo de ensaio (17ºC, 35 minutos). Após esse período, as palhetas foram retiradas do tubo e mantidas por 25 minutos, quando foram submetidas a vapor de nitrogênio por 20 minutos e mergulhadas em nitrogênio líquido. Após o descongelamento, o sêmen dos animais alimentados com a ração acrescida de óleo de soja reduziu a motilidade, o vigor, a porcentagem de espermatozoides vivos e o teste hiposmotico. A motilidade média do sêmen após a diluição e durante as etapas do congelamento foi de 85 e 84% para os animais que receberam óleo de salmão e soja, respectivamente. Entretanto, após o descongelamento, a motilidade dos espermatozoides dos animais alimentados com a ração contendo óleo de salmão e soja reduziu para 30 e 24%, respectivamente. O sêmen dos animais tratados com óleo de soja apresentou menor porcentagem de espermatozoides vivos e morfologia normal em comparação aos tratados com óleo de salmão. A suplementação com óleo de salmão na dieta de suínos promove redução das características negativas observadas no sêmen suíno descongelado.The objective of this work was to evaluate the effects of addition of oil and of levels of supplementation of E vitamin on the boar sperm cryopreservation. It was used 24 Dalboar 85 boars, at 12 to 18 months, distributed in a complete random design, in a 2 × 3 factorial arrangement, with two source of oil (soybean and salmon) and three levels of E vitamin (150, 300 and 450 mg/kg). Semen was collected weekly (1 collection/male), diluted in centrifuge media (1:1) and kept at 25ºC for 2 hours. After centrifugation (800 g, 10 minutes), the pellet was suspended in freezing medium for the final concentration of 1 × 10(9) sperm/mL. The semen was packaged into 0.5 mL straws and placed inside a test tube (17ºC, 35 minutes). After this period, the straws were removed from the tube and kept for 25 minutes more, submitted to nitrogen vapor for 20 minutes and then dip into liquid nitrogen. After thawing, the semen of animals fed the ration added with soybean oil reduced motility, vigor, percentage of live sperm and HOST. The average motility of the semen after dilution and during cryopreservation process was 85 and 84% for the animals fed salmon and soybean oil, respectively. However, after thawing, the motility of the sperms of animals fed ration with salmon and soybean oil was reduced to 30 and 24%, respectively. Semen of animals treated with soybean oil presented lower percentage of live sperm and normal morphology when compared to the ones treated with salmon oil. Supplementation of salmon oil in diet for swines reduces the negative traits observed in thawed semen of boars.

The effects of feeding urea on embryo production, quality and developmental stages and follicular status at the beginning of a superovulatory treatment of Toggenburg does fed urea diet were investigated in this study. Eighteen females were randomly allocated to receive diets with: no urea (control, n=8), and 2.4% urea dietary dry matter (n=10) UDM. The embryo recovery was performed at day 7 or 8 of estrus cycle by transcervical technique and classified according to quality and developmental stage. The follicular status was determined by transrectal ultrasonography at the first day of FSH injection. The number and quality of embryos were not affected by dietary urea concentration. Embryos from does treated with 2.4% UDM were recovered at advanced stage. Urea concentration (2.4%) decreases the number of follicles with less than 5 mm diameter. This follicle class is positively correlated to the number of recovered embryos and to the number and percentage of excellent and good embryos.Objetivou-se investigar os efeitos da ureia na produção, qualidade e estádio de desenvolvimento embrionário e status folicular ao início do tratamento superovulatório de animais alimentados com ureia na dieta. Dezoito cabras da raça Toggenburg foram distribuídas equitativamente para receberem dietas: sem ureia (controle, n=8) e 2,4% de ureia na matéria seca total da dieta (n=10) UMS. Os embriões foram recuperados entre o 7º o 8º dia após o estro pela técnica transcervical e classificados pela qualidade e pelo estádio de desenvolvimento. O status folicular foi determinado por ultrassonografia transretal no dia da aplicação da primeira dose de FSH. O número e a qualidade dos embriões não foram influenciados pela concentração de ureia na dieta. Embriões em estádio avançado de desenvolvimento foram recuperados do tratamento com 2,4% de ureia. A concentração de ureia testada (2,4%) diminui o número de folículos com menos de 5 mm de diâmetro, categoria que se correlaciona positivamente ao número de embriões coletados e ao número e à porcentagem de embriões excelentes e bons.

O estudo teve como objetivo identificar os efeitos do tempo de estocagem, da temperatura de armazenamento e da quantidade de anticoagulante sobre parâmetros hematológicos de cães. Foram utilizadas amostras do sangue de dez cães de raças variadas, clinicamente hígidos. As alíquotas foram colhidas com 1,8mg; 3,6mg; 7,2mg e 14,4mg de ácido etilenodiaminotetracético (EDTA) por mL de sangue, distribuídas em dois grupos: de 2°C a 8°C e temperatura ambiente. Após a coleta, foram avaliadas em quatro tempos: 0, 12, 24 e 48 horas. Usando um contador automático de células, foram avaliados leucócitos, eritrócitos, hemoglobina, hematócrito, volume corpuscular médio (VCM), índice de anisocitose eritrocitária (RDW), plaquetas e plaquetócrito (PCT). O valor do VCM diminuiu nas maiores concentrações de EDTA (7,2mg mL-1 e 14,4mg mL-1), com decréscimo de 2,36% na maior concentração. A temperatura e o tempo de armazenagem também ocasionaram alteração nesse parâmetro, ou seja, houve decréscimo no tempo 12 horas à temperatura de 2 a 8°C e aumento nos tempos 24 e 48 horas à temperatura ambiente (PThe study aimed to identify the effects of time, temperature of storage and excess of anticoagulant on hematological parameters of dogs. Blood samples of ten, clinically healthy dogs, of different breeds were utilized. Aliquots were stored with 1.8mg, 3.6mg, 7.2mg and 14.4mg of ethylenediaminetetraacetic acid (EDTA) per mL of blood, divided into two groups: 2°C to 8°C and room temperature. Right after collection, they were evaluated in four times: 0h, 12h, 24h and 48h. White blood cells, red blood cells, hemoglobin, packed cell volume (PCV), mean corpuscular volume (MCV), red cell distribution width (RDW), platelet and thrombocrit (PCT) were evaluated in the automatic cell counter. In the automatic cell counter analysis, the MCV increased significantly with higher concentrations of EDTA (7.2mg mL-1 and 14.4mg mL-1) peaking at 2.36%, in the highest concentration. Temperature and time also influenced the MCV, which showed a decrease in time 12 under temperature of 2°C to 8°C and an increase in time 24 and 48 at room temperature. Storage temperature influenced leukocytes and erythrocytes counts, which showed lower values at room temperature. Hemoglobin, PCV, platelet and PCT did not change significantly. The changes observed did not compromise the results obtained by automatic cell counter, showing that blood samples stored for two days remained in good condition for processing, especially when stored under refrigeration.

O objetivo deste trabalho foi avaliar o efeito de dietas, com diferentes teores de fósforo, no perfil eletrolítico sanguíneo e no ganho de peso corporal em suínos. Foram utilizados 60 suínos híbridos comerciais, machos castrados, com peso corporal médio inicial de 61,07±0,86 kg, com aproximadamente 110 dias de idade. Os animais foram distribuídos em blocos ao acaso, com 5 tratamentos e 12 repetições cada um. Os tratamentos foram constituídos de dietas com 0,135, 0,200, 0,265, 0,330 e 0,395% de fósforo disponível. No início e no final do período experimental, foi determinado o peso corporal e foram coletadas amostras de sangue, sem jejum dos animais, para a mensuração de sódio, potássio, cloreto, magnésio total, fósforo e cálcio ionizado. Observou-se resposta quadrática do peso corporal, de acordo com os tratamentos, e das concentrações séricas de fósforo, magnésio total e cálcio iônico em decorrência dos tratamentos. O ganho de peso corporal foi maior nos animais alimentados com suplementação de fósforo em teores acima de 0,135%. O melhor resultado foi obtido com a dieta de 0,330% de fósforo, porém, o aumento a partir de 0,200% na dieta ocasionou elevação na concentração sérica de fósforo.The objective of this work was to evaluate the effect of diets containing different levels of phosphorus on blood electrolytic profile and on body weight gain in pigs. Sixty hybrid pigs were used, all barrows, with initial body weight of 61.07 ± 0.86 kg and about 110 days old. The animals were distributed in a randomized block design, in 5 treatments, with 12 replicates each. The treatments consisted of diets containing 0.135, 0.200, 0.265, 0.330 and 0.395% available phosphorus. At the beginning and at the end of the experiment, body weight was determined and blood samples were collected, in the animals without fasting, to measure sodium, potassium, chloride, total magnesium, phosphorus and ionized calcium. A quadratic response in body weight was observed, according to treatments, and for serum phosphorus, total magnesium and ionized calcium in the treatments. Weight gain was higher in animals fed with phosphorus supplementation in levels above 0.135%. The best result occurred with 0.330% P supplementation, however, an increase from 0.200% in the diet caused an elevation in phosphorus serum concentration.

Objetivou-se avaliar o efeito da adição de fontes de óleo e dos níveis de suplementação de vitamina E na ração sobre as características do sêmen suíno resfriado a 17ºC e 5ºC. Foram utilizados 24 suínos machos reprodutores Dalboar 85 distribuídos em delineamento inteiramente casualizado, em arranjo fatorial 2 x 3, com duas fontes de óleo (soja e salmão) e três níveis de antioxidantes (150, 300 e 450 mg de vitamina E/kg). Nos animais sob suplementação com óleo de salmão, a motilidade e o teste hiposmótico dos espermatozoides após 24, 48 e 72 horas foram superiores aos observados nos animais alimentados com a ração com óleo de soja. O óleo de salmão aumentou o vigor em ambas as temperaturas avaliadas após 24 e 48 horas. A ocorrência de anormalidades morfológicas totais foi maior no sêmen dos animais sob suplementação com óleo de soja e resfriado a 17ºC, enquanto na menor temperatura de resfriamento (5º) não houve diferença entre os animais sob suplementação. A morfologia anormal no sêmen a 5ºC foi maior que no sêmen resfriado a 17ºC. O óleo de salmão melhora as características espermáticas do sêmen suíno resfriado a 17 e 5ºC durante 24 a 48 horas.The objective of this study was to evaluate the effect of the addition of oil sources and levels of vitamin E supplementation in the rations on the characteristics of boar semen cooled at 17 and 5ºC. It was used twenty-four Dalboar 85 boars distributed in a complete random design, in a 2 x 3 factorial scheme, with two sources of oil (soybean and salmon) and three levels of antioxidant (150, 300 and 450 vitamin E mg/kg). For animals under supplementation with salmon oil, the motility and hypoosmotic swelling test (HOST) of sperm after 24, 48, and 72 hours was higher than the ones observed in animals fed soybean oil ration. Salmon oil increased the vigor in both temperatures evaluated after 24 and 48 hours. The occurence of total morphological abnormalities was higher in the semen of animals fed soybean oil and cooled at 17ºC while in the lowest cooling temperature (5ºC) there was no difference among animals under supplementation. The abnormal morphology in the semen at 5ºC was higher than in the semen cooled at 17ºC. Salmon oil improves the sperm characteristics of the boar semen cooled at 17 and 5ºC from 24 to 48 hours.

The main purpose of the present study was to identify for candidate genes related to ovulation in swines. To do so, it was investigated in ovarian follicular cells through quantitative real-time PCR the differential expression of the following genes: steroidogenic acute regulator (STAR), GATA-binding protein 4 (GATA), prostaglandin F2α (PGF2α), progesterone receptor (P4R), follicle-stimulating hormone receptor (FSHR), and cytochrome P450 aromatase (CYP19). These genes encode hormone receptors (FSHR and P4R), hormone (PGF2α), steroidogenic proteins (STAR and CYP19) and transcription factor (GATA). Folicular cells were collected from sows with high and low number of piglets/litters during the follicular phase of the estrus cycle. There was difference in transcript abundance among low and high prolific sows for the STAR, GATA, PGF2α, P4R and CYP19 genes. For the FSHR gene, the fold change was not considered to be significantly different. Because in the present study only the transcript level of the above mentioned genes was analyzed, no inference can be made regarded to protein translation or activity. Therefore, gene sequence trials and other functional studies will be necessary to complement the present results, allowing a better understanding on biological complexity of these genes and their use as markers for prolificity in swines.O objetivo neste trabalho foi identificar genes candidatos relacionados à ovulação em suínos. Para tanto, investigou-se a expressão diferencial dos genes STAR (steroidogenic acute regulator), GATA (GATA-binding protein 4), PGF2α (prostaglandin F2α), P4R (progesterone receptor), FSHR (follicle-stimulating hormone receptor) e CYP19 (cytochrome P450 aromatase) em células foliculares ovarianas por meio de reação em cadeia da polimerase em tempo real (qRT-PCR) quantitativo em tempo real. Esses genes codificam para receptores hormonais (FSHR e P4R) hormônio (PGF2α), proteínas esteroidogênicas (STAR e CYP19) e fator de transcrição (GATA). As células foliculares foram coletadas durante a fase folicular do ciclo estral de porcas com alto e baixo número de leitões/leitegada. Houve diferença na abundância de transcritos entre porcas com alta e baixa prolificidade para os genes STAR, GATA, PGF2α, P4R and CYP19. Para o gene do FSHR, a alteração na abundância dos transcritos não foi significativamente diferente. Considerando que foi analisado somente o nível de transcrição desses genes mencionados, não se pode fazer inferências com relação à tradução ou atividade proteica. Portanto, ensaios de sequenciamento gênico e outras análises funcionais serão necessários para complementar esses achados e possibilitar melhor entendimento da complexidade biológica desses genes e seu uso como marcadores para prolificidade em suínos.

Objetivou-se verificar o estágio de maturidade sexual, a ocorrência do formato testicular e a relação entre características reprodutivas de tourinhos da raça Nelore. Foram utilizados 5.903 touros com média de 21 meses de idade submetidos a exame andrológico entre os anos de 1999 e 2003. Na ocasião do exame, foram registradas as seguintes características: perímetro escrotal (31,99 ± 2,23 cm), comprimentos testiculares esquerdo (11,21 ± 0,98 cm) e direito (11,26 ± 0,97 cm), larguras testiculares esquerda (5,92 ± 0,44 cm) e direita (5,97 ± 0,46 cm), formato testicular, variando de 1 a 5 (1,72 ± 0,46), volume testicular (632,21 ± 132,72 cm³), motilidade espermática progressiva retilínea (69,56 ± 12,31%), vigor espermático (2,87 ± 0,61) e morfologia espermática, com defeitos totais (22,19 ± 11,13%) e maiores (15,86 ± 10,45%) dos espermatozoides. As freqüências dos formatos testiculares foram longo (30,80%), longo-moderado (66,19%), longo-oval (2,49%), oval-esférico (0,02%) e esférico (0,04%). As correlações entre perímetro escrotal e características reprodutivas foram positivas. O perímetro escrotal é uma ótima característica para seleção de touros jovens da raça Nelore, que, na maioria (78,33%), apresentam-se sexualmente maturos aos 21 meses de idade.The objective of this study was to verify the stage of sexual maturity, occurrence of the testicular format and the relationship among reproductive characteristics. Data were used from 5903 Nellore bulls averaging 21 months submitted to the soundness evaluation from 1999 to 2003. At the time of the evaluation, the following characteristics were recorded: scrotal circumference at soundness evaluation (31.99 ± 2.23 cm), left testicular length (11.21 ± 0.98 cm) and right testicular length (11.26 ± 0.97 cm), left testicular width (5.92 ± 0.44 cm) and right testicular width (5.97 ± 0.46 cm), testicular format (1.72 ± 0.46), testicular volume (632.21 ± 132.72 cm³), individual motility (69.56 ± 12.31%), vigor (2.87 ± 0.61), as well as total defects (22.19 ± 11.13%) and major defects (15.86 ± 10.45%) of the spermatozoa. The frequencies of the testicular formats were long (30.80%), long-moderate (66.19%), long-oval (2.49%), oval-spherical (0.02%) and spherical (0.04%). Correlations between the scrotal circumference and the reproductive traits were positive. Scrotal circumference is a good trait for selection of young Nellore bulls. More than 70% of the animals studied are sexually mature at 21 months old.

Os objetivos neste estudo foram verificar se a própolis e o ácido ascórbico têm efeito sobre a integridade da membrana plasmática dos espermatozoides de caprinos e investigar o potencial desses antioxidantes no uso de meios diluidores de criopreservação de sêmen caprino. Foram utilizados cinco bodes adultos das raças Alpina (n = 2) e Saanen (n = 3). Após a coleta de sêmen, realizaram-se o exame físico do sêmen e morfológico dos espermatozoides, o teste supravital e o teste hiposmótico. Em seguida, o sêmen fresco foi diluído com o diluidor Bioxcell® (controle); Bioxcell® + 0,25% de extrato liofilizado de própolis; Bioxcell® + 0,5% de extrato liofilizado de própolis; Bioxcell® + 0,05% de ácido ascórbico; ou Bioxcell® + 0,25% de ácido ascórbico. Após as diluições finais, foram avaliados a motilidade e o vigor espermático obtidos com cada diluidor e posteriormente o sêmen foi submetido a envase, resfriamento e congelamento. No sêmen fresco, os aspectos físicos e morfológicos e os resultados dos testes supravital e hiposmótico não diferiram entre os animais nem entre raças. As médias gerais de motilidade e vigor espermático e dos testes supravital e hiposmótico obtidos logo após o descongelamento e após 3 horas de teste de termorresistência diferiram entre si, de modo que o diluidor contendo ácido ascórbico e o controle foram similares e superiores àqueles contendo própolis. O ácido ascórbico mantém a integridade estrutural da membrana dos espermatozoides durante o processo de criopreservação, bem como sua viabilidade após o teste de termorresistência, e pode ser uma alternativa na composição de diluentes para criopreservação de sêmen caprino; a própolis não é eficaz na manutenção da integridade e da viabilidade espermática pós-descongelamento e é tóxica aos espermatozoides nas concentrações de 0,25 e 0,5%.The objectives of this study were to verify whether propolis and ascorbic acid have an effect on plasmatic membrane integrity of goat spermatozoa and investigate the potential of these antioxidants in the use of goats spermatozoa cryopreservation extenders. Five adult boars were used of the Alpine (n = 2) and Saanen (n = 3) breeds. After semen collection, the evaluation consisted of the physical and morphological exam, live and dead cells (supravital test) and hyposmotic swelling test. Afterwards, the fresh semen was diluted with the Bioxcell® extender (control); Bioxcell® + 0.25% freeze dried propolis extract); Bioxcell® + 0.5% freeze dried propolis extract); Bioxcell® + 0.05% ascorbic acid) or Bioxcell® + 0.25% ascorbic acid). After final dilutions, the sperm motility and vigor were assessed obtained in each extender, and then the semen was sealed, cooled and frozen. In fresh semen, the physical and morphological aspects and the results of the supravital test and hyposmotic swelling test did not differ between animals or breeds. The general means of the sperm motility and vigor, supravital test and hyposmotic swelling test obtained immediately after thawing and after three hours of heat resistence test were different, so that the extender with ascorbic acid and the control were similar and higher than the extenders containing propolis. The ascorbic acid maintained the structural integrity of the spermatozoa membrane during the cryopreservation process and its viability after the heat resistance test, and may be an alternative in extender composition for cryopreservation of goat semen; the propolis was not effective in maintaining sperm integrity and viability after thawing and was toxic to spermatozoa at concentrations of 0.25 and 0.5%.

O presente estudo foi conduzido com seis jumentos reprodutores da raça Pêga, com peso médio de 272±34,9 (231-326kg), sendo subdivididos em adultos (J1, J2 e J3, média 15 anos) e jovens (J4, J5 e J6, média 3,5 anos). Os reprodutores apresentavam histórico normal de fertilidade à monta natural e, após realização de exame andrológico, foram classificados como aptos à reprodução. Foram realizadas 180 coletas de sêmen. Os reprodutores apresentaram parâmetros seminais considerados normais para a espécie. De agosto/2006 a fevereiro/2007, após a realização do exame andrológico, cada reprodutor foi submetido a três ou quatro mensurações do aparelho genital externo para: comprimento testicular (CT - crânio caudal), altura testicular (AT - dorso ventral), largura testicular (LT - médio lateral) e espessura do funículo (FUN - terço médio). Os dados de biometria testicular foram utilizados para o cálculo do índice testicular (IT) e volume testicular (VT). O IT médio observado foi de 7,4 (5,9 a 9,6), e o VT esquerdo foi de 155,5±14,4mL, enquanto o direito foi de 149,3±14,9. A medida de FUND foi de 25,3±3,3mm (20,3a 30,8mm), e de FUNE foi 24,6±1,5mm (22 a 26mm). Jumentos, mesmo com IT menor do que o proposto para garanhões; se apresentaram mais eficientes que os últimos para a produção espermática. O volume espermático, associado ao IT, pode ser usado como ferramenta auxiliar na estimativa da constatação de normalidade para testículos de jumentos. A mensuração in vivo do funículo espermático pode ser uma nova variante na utilização do exame andrológico de jumentos. Os dados descritos podem servir de valores de referência e valores práticos quanto à biometria do aparelho genital externo de jumentos.The present study was performed with six Pêga breed donkeys, weight from 231 to 326kg (272±34.9). The breeders were divided into the adult group (J1, J2 e J3, 15 years old average) and the young group (J4, J5 e J6, 3.5 years old average). . The donkeys presented normal rates of fertility at natural mating. After breeding soundness evaluation all animals were considered capable for reproduction. A hundred eight semen collections were realized, and the donkeys presented normal seminal parameters for the specie. From August/2006 to February/2007, after semen collection each donkey was submitted to three or four measurements of the extern genital tract (both right and left side) for: testis length (LT - cranio caudal); testis height (TH - dorso ventral) and thickness of spermatic funiculum (FUN - third part). The biometric data from biometric testis were used to calculate the testis indices (TI) and testis volume (TV). The TI observed was 7.4 (5.9 a 9.6) and TV was 155.5±14.4, 149.3±14.9 respectively left and right. The average FUN was 25.3±3.3mm (20.3 a 30.8mm) to the right and 24.6±1.5mm (22 a 26mm) to the left side. The donkeys presented lesser TI than those proposed for the stallions, but the asinine were more efficient for sperm production than the average reported to stallions . The testis volume associated with IT can be used as an additional tool to estimate the normality of the breeder's testis. The measurement in vivo of spermatic funiculum can be used as a new component for the breeding soundness evaluation of donkeys. The genital extern tract biometric values can support routine breeding evaluation and can be applied to news studies.

Objetivou-se avaliar a influência da condição corporal ao parto sobre os parâmetros produtivos de cabras Alpinas no início da lactação. Sessenta e oito cabras foram distribuídas em três grupos, de acordo com o escore de condição corporal (baixo, entre 1,00 e 2,75; intermediário, entre 2,75 e 3,50; e alto, entre 3,50 e 5,00) e avaliadas do parto até a oitava semana de lactação. Realizaram-se o controle diário do consumo alimentar e da produção leiteira e avaliações semanais de peso corporal, escore da condição corporal e composição do leite. Os animais com escore corporal alto apresentaram menor consumo de matéria seca, fibra em detergente neutro e proteína bruta em comparação àqueles com escores corporais baixo e intermediário. O período médio de recuperação do consumo de matéria seca dos animais das três condições corporais foi de 16 dias (2,3 semanas) do pós-parto, com estabilização até o final do experimento. Na sexta semana de lactação, o peso corporal dos animais com escore corporal baixo se igualou ao daqueles com escore corporal intermediário. Não houve efeito da condição corporal ao parto nas variações de ECC e da produção leiteira ao longo do período. O teor de gordura do leite dos animais com escore corporal alto foi menor nas duas primeiras semanas pós-parto em comparação ao dos animais com escores corporais baixo e intermediário. Elevada condição corporal ao parto influencia negativamente o consumo voluntário de cabras leiteiras no início da lactação, mas quando se utiliza dieta de alta qualidade e com altos níveis de energia, a influência da condição corporal ao parto inexiste para variações de peso, de escore da condição corporal e produção leiteira dos animais.The objective of this study was to evaluate the influence of body condition at birth on Alpine goat productive parameters in early lactation. Sixty-eight females were assigned to three groups according to the body condition score (BCS) - low (1.00 - 2.75); intermediate (2.75 - 3.50) and high (3.50 - 5.00) and evaluated from kidding to the eighth week of lactation. The feed intake and milk production were recorded on a daily basis and the body weight, body condition score and milk composition at weekly intervals. Animals with a high body condition score presented lower dry matter intake, neutral detergent fiber and crude protein compared to those with low and intermediate body condition scores. The average time for dry matter intake recovery of the animals with the three body condition scores was 16 days (2.3 weeks) postpartum, and stabilization by the end of the experiment. In the sixth week of lactation, animals with low body condition score had the same body weight of animals with intermediate body condition score. There was no effect of the body condition at birth on the body condition score changes and milk production during the experiment. The milk fat content of animals with high body condition score was lower in the first two weeks postpartum compared to animals with low and intermediate body condition score. The high body condition at birth influences negatively the ad libtum intake of dairy goats in the early lactation, but when it is used feed of high quality with high energy levels, the influence of the body condition at birth is nil to variations in weight, BCS and milk production.

Objetivou-se avaliar a influência da condição corporal ao parto sobre alguns metabólitos sanguíneos em cabras no início da lactação. As cabras (68 no total) foram distribuídas em três grupos de acordo com o escore da condição corporal (ECC) ao parto: entre 1,00 e 2,75 (animais magros); entre 2,75 e 3,50 (condição corporal intermediária); e entre 3,50 e 5,00 (animais gordos). O experimento foi realizado nos primeiros 60 dias de lactação e as coletas de sangue, após o parto e semanalmente até a oitava semana de lactação. Foram feitas análises para determinação das concentrações de ácidos graxos não-esterificados (AGNE), beta-hidroxibutirato (BHBA), glicose, colesterol total e lipoproteína de alta densidade (HDL). As concentrações plasmáticas dos metabólitos não diferiram entre os grupos. As concentrações de AGNE, nos animais dos três grupos, ao parto e nas oito primeiras semanas de lactação, foram de 658,46 e 232,90 mg/dL, respectivamente, e os valores de BHBA ao parto, de 10,31 ± 3,0 mg/dL para os animais magros; 6,88 ± 0,9 mg/dL para os intermediários; e 4,21 ± 0,8 mg/dL para os animais gordos. Os valores de glicose ao parto foram de 112,54 ± 15,4 mg/dL para os animais magros; 90,93 ± 13,5 mg/dL para os animais intermediários; e 132,47 ± 26,7 mg/dL para os animais gordos. Os valores médios de colesterol total e de HDL nas oito primeiras semanas pós-parto foram de 109,01 ± 2,5 e 54,9 ± 1,39 mg/dL para os animais magros; 83,0 ± 21,7 e 56,0 ± 1,30 mg/dL para os intermediários; e 84,3 23,9 e 54,5 ± 1,93 mg/dL para os gordos. As alterações no metabolismo energético de cabras leiteiras de média produção diminuem, independentemente da condição corporal ao parto, quando fornecida dieta de alta qualidade e com altos níveis de energia.An experiment was carried out to study the influence of the body condition score at birth on blood metabolites of goats in early lactation. Sixty-eight goats were placed in three groups according to the body condition score (BCS) at parturition: from 1.00 to 2.75 (thin animals); 2.75 and 3.50 (intermediate body condition) and from 3.50 to 5.00 (fat animals). The experimental was carried out in the first sixty days of lactation and blood sampling started after parturition at weekly intervals up to the eighth week of lactation. Analyses were made of the non-esterified fat acids (NEFA), beta-hydroxibutirate (BHBA), glucose, total cholesterol and high density lipoprotein (HDL). The plasma concentrations of the metabolites did not differ among the groups. The NEFA concentrations in the animals in the three groups at birth and in the first eight weeks of lactation were 658.46 and 232.90 mg/dL, respectively and the values of BHBA at birth were 10.31 ± 3.0 mg/dL for the thin animals, 6.88 ± 0.9 mg/dL for the intermediate animals and 4.21 ± 0.8 mg/dL for the fat animals. The glucose values at birth were 112.54 ± 15.4 mg/dL for the thin animals, 90.93 ± 13.5 mg/dL for the intermediate animals and 132.47 ± 26.7 mg/dL for the fat animals. The average of total cholesterol and HDL in the first eight weeks after birth were 109.01 ± 2.5; and 54.9 ± 1.39 mg/dL for the thin animals, 83.0 ± 21.7 and 56.0 ± 1.30 mg/dL for the intermediate animals and 84.3 ± 23.9 and 54.5 ± 1.93 mg/dL for the fat animals. The changes in the energy metabolism of dairy goats with medium production decreased, regardless of the body condition at birth, when supplied with high quality feed with high energy levels.

No presente estudo, foram avaliados os efeitos de soluções eletrolíticas administradas via sonda nasoesofágica de pequeno calibre sobre o hematócrito, o volume plasmático e as concentrações plasmáticas de proteínas totais, sódio, potássio, cloreto, magnésio total e cálcio iônico de eqüinos hígidos e desidratados experimentalmente. Foram utilizados quatro equinos adultos, mestiços, dois machos e duas fêmeas. No experimento 1 (E1Des), os animais foram desidratados experimentalmente, enquanto no 2 (E2Hig) foram utilizados equinos hígidos. Os animais foram submetidos a cada um dos seguintes tratamentos: SE - solução eletrolítica isotônica; SEGli - solução eletrolítica isotônica + glicose; SEMalt - solução eletrolítica isotônica + maltodextrina e SEMg - solução eletrolítica isotônica + sulfato de magnésio. A solução eletrolítica foi administrada na dose de 15mL kg-1 h-1, durante 12h via sonda nasoesofágica por fluxo contínuo. Os tratamentos com as soluções eletrolíticas ocasionaram expansão do volume plasmático, ocasionando a redução nos valores das proteínas plasmáticas totais e do hematócrito, enquanto os valores dos eletrólitos avaliados permaneceram na faixa de referência.In the present study, the effects of four different electrolyte solutions on the packed cell volume (PCV), plasma volume and plasma concentrations of total protein, sodium, potassium, chloride, total magnesium, and ionized calcium in healthy and experimentally dehydrated horses were evaluated. Four crossbred horses, two males and two females were used. In experiment 1 (E1Des) the animals were experimentally dehydrated, while in the second experiment (E2Hig) healthy equines were used. In both experiments the animals were subjected to the following treatments: SE - isotonic electrolyte solution; SEGlu - glucose-enriched SE; SEMalt - maltodextrine-enriched SE and SEMg - magnesium sulphate-enriched SE. The electrolyte solutions used in all treatments were administered at 15mL kg-1 h-1 during 12h through small-bore nasoesophagus tube by continuous flow. The treatments with the electrolyte solutions resulted in an increase of plasma volume and a decrease in total plasma protein, hematocrit, while the electrolytes remained within the reference values.

The aim of this study was to evaluate the efficiency of the postpartum bovine endometritis treatment by intrauterine and intramuscular administration of tetracycline. Forty six cross bred cows diagnosed by gynecological exam with postpartum endometritis were assigned randomly in two treatments: G1 (n=21) - animals were treated with one intramuscular (IM) dose of tetracycline (20 mg/Kg of body weight), while in G2 (n=25) animals were treated with one intrauterine (IU) dose of tetracycline (30 mg/Kg of body weight). No difference (p>0.05) was observed in the recovery rate between the two treatments (61.9 X 76.0%, G1 and G2, respectively). The interval from treatment until first estrous was 33.9±22.6 versus 14.8 ±10.9 days (P0.05) was observed in the number of services per conception between G1 (1.54) and G2 (1.3). The treatment cost was lower for the G1 (U$ 3.51 versus U$ 5.00). Although not to have had differences in the clinical recovery rate, the use of the oxitetracycline managed for way IU in the postpuerperal treatment of endometrites in cows revealed more efficient in the reduction of the interval between the treatment and first heat and first insemination, beyond presenting lower cost that the treatment with oxitetracycline managed for way IMKEY WORDS: Bovine, postpartum endometritis, intrauterine, intramuscular, oxitetracicline. Este trabalho teve por objetivo comparar a eficiência do uso da oxitetracilina, administrada pelas vias intramuscular (IM) e intrauterina (IU), no tratamento de endometrites em vacas no período pós-puerperal. Foram utilizados 46 animais mestiços que apresentaram quadro clínico de endometrite, distribuídos aleatoriamente em dois tratamentos: G1 (n=21) – tratado com uma dose de 30 mg/Kg PV de oxitetraciclina por via IM, e G2 (n=25) – uma infusão de 20 mg/Kg PV de oxitetraciclina por via IU. Não houve diferença (p>0,05) na taxa de recuperação dos animais, 61,9% e 76,0% para G1 e G2, respectivamente. O intervalo entre o tratamento e o primeiro estro (PPALAVRAS-CHAVES: Bovinos, endometrite pós-puerperal, intramuscular, intrauterina, oxitetraciclina.

Objetivou-se estudar a relação entre dois métodos de avaliação do potencial reprodutivo - Classificação Andrológica por Pontos (CAP) e comportamento sexual de touros da raça Nelore - e verificar a acurácia desses testes em predizer o potencial reprodutivo dos touros, avaliado pela taxa de gestação ao final da estação de monta, com duração média de 90 dias. Foram utilizados 14 touros, todos classificados como aptos à reprodução, após exame andrológico, CAP e observação do comportamento sexual a campo durante 12 horas. Onze touros foram avaliados em manejo individual, com proporção média touro:vaca de 1:46, e os três restantes, em manejo múltiplo, com proporção média touro:vaca de 1:23. A pontuação média obtida pela CAP foi 78,92 ± 8,92 (muito bom) e o escore, 9,85 ± 0,36 (excelente) para libido. Quanto maior o tempo de teste, maiores foram os escores da libido. Na primeira hora de observação, os touros foram classificados como de libido questionável e ao final do teste, como de libido excelente. As correlações da libido com circunferência escrotal (CE), características físicas e morfológicas do sêmen e taxa de gestação foram baixas. A pontuação pela CAP correlacionou-se positivamente com circunferência escrotal e características físicas do sêmen, mas não se correlacionou com características morfológicas dos sêmen. Não foi observada correlação entre CAP e taxa de gestação, nem entre libido e CAP. A taxa geral de gestação após o período de estação de monta foi de 74,69%. Os resultados indicam que tanto a CAP como o teste da libido não são eficientes para predizer o potencial reprodutivo de touros da raça Nelore e devem ser utilizados de forma complementar ao exame andrológico.The objective of this work was to study the relation between two reproductive potential evaluation methods - Andrologic Evaluation by Points - CAP and sexual behavior of Nelore bulls, and to verify the accuracy of these tests in predicting the reproductive potential of bulls through the gestation rate at the end of the breeding season, with average duration of ninety days. Fourteen bulls were used, all classified as able for reproduction, evaluated through andrologic examination, being submitted to CAP and observation of the sexual behavior at field (libido test) during 12 hours. Eleven bulls were evaluated in individual handling, with an average bull:cow ratio of 1:46 and the rest of the animals (three), evaluated in a multiple handling, with an average bull:cow ratio of 1:23. The average score obtained by CAP was 78.92 ± 8.92 (very good) and the score of 9.85 ± 0.36 (excellent) for the libido test. The longer the test was, the higher libido scores were obtained. At the first time of observation, the bulls were classified as with questionable libido and at the end of the test as with excellent libido. The correlations between libido, scrotal circumference (SC), physical and morphological characteristics of the semen and the gestation rate were low. The scores obtained through CAP was positively correlated with SC and physical characteristics of the semen; however, no correlation was observed for the morphologic characteristics. No correlation was observed between CAP and gestation rate or between libido and CAP. The overall gestation rate after the breeding period was 74.69%. The results suggest that both CAP and the libido test were not efficient to predict the reproductive potential of Nelore bulls and should be used as a complement to the andrologic evaluation.

In order to characterize the expression of genes associated with immune response mechanisms to mastitis, we quantified the relative expression of the IL-2, IL-4, IL-6, IL-8, IL-10, IFN-γ and TNF-α genes in milk cells of healthy cows and cows with clinical mastitis. Total RNA was extracted from milk cells of six Black and White Holstein (BW) cows and six Gyr cows, including three animals with and three without mastitis per breed. Gene expression was analyzed by real-time PCR. IL-10 gene expression was higher in the group of BW and Gyr cows with mastitis compared to animals free of infection from both breeds (p < 0.05). It was also higher in BW Holstein animals with clinical mastitis (p < 0.001), but it was not significant when Gyr cows with and without mastitis were compared (0.05 < p < 0.10). Among healthy cows, BW Holstein animals tended to present a higher expression of all genes studied, with a significant difference for the IL-2 and IFN-γ genes (p < 0.001). For animals with mastitis no significant difference in gene expression was observed between the two breeds. These findings suggest that animals with mastitis develop a preferentially cell-mediated immune response. Further studies including larger samples are necessary to better characterize the gene expression profile in cows with mastitis.

Avaliou-se a relação entre os testes complementares (teste hiposmótico, teste de termorresistência lento e teste de reação acrossômica) e os testes de avaliações convencionais (aspectos físicos e morfológicos) de sêmen bovino congelado/descongelado e os índices de prenhez. Os valores médios da motilidade espermática progressiva retilínea avaliados pelo teste de termorresistência foram de 53,48 (pós-descongelamento), 43,69 (60 minutos), 35,88 (120 minutos) e 33,04% (180 minutos) e a porcentagem de células reativas ao teste hiposmótico foi de 37,89%. Correlação positiva e de média intensidade foi encontrada para a motilidade espermática progressiva retilínea pós-descongelamento e o teste hiposmótico (0,21). Entretanto, a correlação da motilidade aos 180 minutos com o teste hiposmótico foi alta (0,64). A porcentagem de células que tiveram acrossoma reagido pós-descongelamento foi de 9,85%, apresentando correlações negativas de média e alta intensidade (-0,25 e -0,46, respectivamente) com a motilidade espermática progressiva retilínea pós-descongelamento e após 3 horas de incubação. Não houve correlação dos testes complementares e da motilidade pós-descongelamento com a taxa de gestação. Nenhum parâmetro considerado isoladamente serviu para avaliar a capacidade fertilizante do sêmen congelado/descongelado.The objective of this study was to evaluate the relationship between complementary (hiposmotic, thermo-resistance and acrosome reaction tests) and conventional evaluations (physical and morphologic aspects) of bovine frozen/thawed semen and conception rates. Average values for spermatic motility evaluated by thermo-resistance test were of 53.48% (post-thawed), 43.69% (60 minutes), 35.88% (120 minutes) and 33.04% (180 minutes). The percentage of reactive cells observed for the hiposmotic test was 37.89%. Average intensity was observed for post-thawing spermatic motility, positively correlated to hiposmotic test (0.21). However, correlation between motility in the 180 minutes and hiposmotic test was high (0.64). The percentage of cells presenting post-thawing acrosome reaction was 9.85%, which was negative correlated to post-thawing (-0.25) and after three hours of incubation (-0.46) spermatic motility. No correlation was observed for complementary tests and post-thawed motility and the conception rate. No one of the parameters evaluated in this study was individually able to indicate the fertilizing capacity of the frozen/thawed semen.

Objetivou-se determinar a eficiência do teste da libido realizado em curral em comparação ao teste realizado a campo e relacioná-los à taxa de gestação em estação de monta de 120 dias. Treze touros aptos à reprodução foram submetidos ao teste da libido em curral por 3 horas. Desses touros, nove foram submetidos à avaliação a campo na proporção touro:vaca de 1:33. A cada 21 dias, foi diagnosticada a gestação por meio de avaliações ultra-sonográficas. No teste em curral, observou-se que, quanto maior o tempo, maiores foram os escores de classificação e que nenhum touro efetuou serviço completo nos 10 minutos iniciais do teste. Com um touro classificado como muito bom obteve-se 84,80% de prenhez, porém touros questionáveis proporcionaram taxas de prenhez de 86,67 e 96,55% ao final da estação. As correlações entre características físicas e morfológicas do sêmen mantiveram-se próximas de zero ou foram nulas. Os principais sinais fisiológicos do comportamento sexual foram o ato de cheirar ou lamber o corpo da fêmea, cheirar ou lamber a vulva, seguidos ou não por reflexo de Flehmen. Em animais da raça Nelore, o tempo de 30 minutos de observação mostrou-se mais eficiente para a realização do teste da libido em curral. O teste da libido realizado a campo no período diurno não foi eficaz em predizer a libido de todos os animais estudados, em decorrência, provavelmente, do comportamento sexual noturno de alguns reprodutores.The objectives of this study were to determine the efficiency of the libido test in corral compared to the test performed at field and to relate them to the gestation rate observed in a breeding season of 120 days. Thirteen Nelore bulls able to reproduce were submitted to the libido test in corral for 3 hours and evaluated by scores every five minutes. Nine of them were submitted to evaluation at field using a bull:cow proportion equal to 1:33. The gestation was diagnosed every 21 days through ultrasonographic evaluation. During the test in corral, the classification scores increased as the testing time increased, but no bull accomplished complete service in the first 10 minutes of the test. A very good bull obtained 84.80% for pregnancy rate, but other questionable bulls obtained 86.67 and 96.55% at the end of the breeding season. Correlations estimates among physical and morphologic characteristics of the semen were close to zero or null. The main physiologic indicators of the bull sexual behavior were: smelling or licking the female body, smelling or licking the vulva, followed or not by the Flehmen reflex. The results suggested 30 minutes of observation as the most efficient time to evaluate the libido test in corral. The libido test at field during daytime was not effective probably because of the sexual behavior of some bulls at night.

Dez amostras de sêmen de dois reprodutores caprinos, da raça Parda-alpina, colhidas em vagina artificial, foram submetidas a quatro tratamentos para avaliação da eficiência do etilenoglicol e do glicerol, associados ou não ao EDTA, na criopreservação da célula espermática caprina. O diluente usado era à base de Tris-gema de ovo contendo 7% de glicerol (glicerol E glicerol+EDTA) ou 7% de etilenoglicol (etilenoglicol e etilenoglicol + EDTA), sendo que nos grupos glicerol+EDTA e etilenoglicol+EDTA foi associado ao diluente 0,1% de EDTA. As amostras foram mantidas por 60 minutos em geladeira a 40C, onde então era efetuada a congelação em nitrogênio (-1960C). A descongelação foi realizada em banho-maria a 370C por 30segundos. As médias obtidas para motilidade (%) a descongelação, para os grupos glicerol, glicerol+EDTA, etilenoglicol, etilenoglicol+EDTA, foram, respectivamente, 51%; 61%; 10% e 12%. Os grupos que utilizaram o glicerol como crioprotetor obtiveram melhores taxas de motilidade pós-descongelação, principalmente quando foi associado ao diluidor o EDTA (grupo glicerol+EDTA). Porém, esse resultado foi comprometido pelos maiores índices de alterações patológicas nos grupos que continham o glicerol. Este estudo teve por objetivo comparar a eficácia de quatro protocolos de congelação do sêmen caprino (glicerol, glicerol+EDTA; etilenoglicol; etilenoglicol+EDTA), através da utilização do teste hiposmótico (HOST). O sêmen foi colhido de dois machos da raça Alpina, sexualmente maduros, diluído nos diferentes meios, congelado e armazenado em nitrogênio líquido. Após a colheita, 20ìL do sêmen fresco foram incubados com 01mL de solução hiposmótica (combinação de citrato de sódio e frutose em água destilada com osmolaridade de 125mOsmol), em banho-maria a 370C por 30 minutos. Este procedimento foi repetido após a descongelação e, em seguida, uma amostra foi colocada sobre lâmina/lamínula e avaliada em contraste de fase com mil vezes de aumento. Um total de 100 células foi contado, e as médias percentuais de espermatozóides com edema ou dobramento de cauda, após o HOST, foram – para o sêmen fresco, glicerol, glicerol+EDTA; etilenoglicol; etilenoglicol+EDTA –, respectivamente, 53,89; 16,90; 10,25; 52,64 e 57,54. O congelamento do sêmen de bodes das raças Alpina e Saanen submetidos ao manejo de fotoperíodo artificial foi avaliado por meio dos testes de termorresistência (TTR), hiposmótico (HOST) e de integridade do acrossoma. Foram utilizados oito machos caprinos (quatro da raça Alpina e quatro da raça Saanen) de duas idades diferentes (jovens e adultos). A qualidade do sêmen durante as etapas do congelamento foi superior em bodes jovens de ambas as raças. Avaliada pelo TTR, a motilidade do sêmen fresco apresentou longevidade pós-descongelamento. Os resultados de motilidade espermática, obtidos imediatamente após o descongelamento, tiveram reflexos positivos sobre o TTR, indicando que os sêmens que apresentaram maior motilidade pós-coleta (86,2% vs 79,3%) e pós-descongelamento (37,7% vs 32,0%) tiveram maior longevidade seminal. Os resultados do HOST, tanto para o sêmen fresco quanto para o congelado, não diferiram entre raças e idades. Houve redução na porcentagem de espermatozóides íntegros após o congelamento e descongelamento do sêmen dos animais da raça Alpina e dos adultos Saanen. Houve redução superior a 14% nos valores do HOST para o sêmen congelado em relação aos valores observados para o sêmen fresco (38,0 vs 52,0%, respectivamente). A motilidade espermática progressiva mostrou maior sensibilidade à criopreservação que a integridade da membrana espermática, indicando que a motilidade espermática é mais afetada pelo processo de congelamento que a membrana plasmática. A integridade do acrossoma não foi influenciada pelo descongelamento. Os índices de danos acrossomais (edema, desprendimento parcial e até perda total de acrossoma) mostraram-se dentro do padrão aceitável (44,5%), tanto no pós-descongelamento como após o término do TTR.The freezing ability of semen from Alpine and Saanen goats submitted to an artificial photoperiod exposure were evaluated by the thermo-resistance (TTR) and hypoosmotic (HOST) tests and integrity of the acrosome using eight male goats, four of each breed and two age groups (young and adults). The quality of semen during the freezing process was superior for the young animals of both breeds. Objetivou-se com este estudo avaliar parâmetros reprodutivos (comportamento sexual, perímetro escrotal e aspectos quantitativos e qualitativos do sêmen) de bodes (jovens e adultos) das raças Alpina e Saanen submetidos ao manejo de fotoperíodo artificial (16 horas de luz e 8 horas de escuro durante dois meses). Os animais jovens demonstraram maior libido que os adultos, considerando-se o menor tempo de reação. Houve variação na freqüência de manifestação das características comportamentais, tanto entre raças quanto entre idades, e, em todas essas características, foi verificada maior intensidade para os animais adultos. Bodes Alpinos apresentaram menor perímetro escrotal que os bodes Saanen na mesma idade. O perímetro escrotal aumentou de 30,53 cm no período 1 (primeiros 15 dias após o final do manejo com fotoperíodo artificial) para 31,53 cm no período 2 (restante do experimento). Bodes Alpinos produziram sêmen de melhor qualidade que os Saanen de mesmo grupo de idade. Bodes das raças Alpina e Saanen criados nas condições deste experimento são sensíveis (sensibilidade mais pronunciada em animais adultos). Quanto aos parâmetros reprodutivos avaliados, às variações sazonais ao longo do ano.This study aimed to evaluate reproductive characteristics (sexual behavior, scrotal perimeter and quali-quantitative aspects of semen) of young and adults Alpine and Saanen male goats exposed to an artificial photoperiod (16 hours of light and 8 hours of darkness for 2 months). The young animals showed greater libido than the adults, based on the smallest time of reaction. Frequency of manifestation of the behavior traits varied between breeds and ages and higher intensity was observed on adult animals. Scrotal perimeter was smaller for Alpine goats than for Saanen goats at the same age. The scrotal perimeter increased from 30.53 cm in the first period (15 days after the end of the photoperiod exposure) to 31.53 cm in the second period (to the end of the experiment). Quality of semen of Alpine goats was better than that of Saanen goats at the same age. According to the reproductive traits evaluated, Alpine and Saanen goats exposed to similar conditions of this study are sensible to seasonal variation during the year.

Foram estudadas as relações entre a concentração de proteínas solúveis do plasma seminal, o teste hipoosmótico, a análise física e morfológica do sêmen e o desempenho reprodutivo de bodes da raça Alpina em regime de monta controlada. O experimento foi realizado durante os meses de fevereiro a abril de 2001. Foram realizadas 40 coletas de sêmen em quatro reprodutores adultos em regime de monta controlada. Em todas as coletas, além do exame físico e morfológico do sêmen, foram realizados os testes hipoosmóticos, isoosmóticos e a determinação da concentração de proteínas solúveis do plasma seminal. Foi detectada diferença entre os bodes na concentração de proteínas solúveis do plasma seminal, mas não houve diferença no teste hipoosmótico e no número de coberturas por prenhez. O teste hipoosmótico pode ser uma importante ferramenta para a avaliação de sêmen caprino, mas a concentração de proteínas solúveis do plasma seminal não pode ser utilizada como parâmetro para predizer a qualidade seminal e a fertilidade de bodes da raça Alpina utilizados em regime de monta natural.The relationships among soluble proteins of the seminal plasma, the hypoosmotic test, the physical and morphologic analyses of semen and the reproductive performance of male goats of the Alpine breed under controlled mating were studied using forty samples of semen from four adult male goats collected twice a week between February and April of 2001. Differences among male goats were observed for soluble protein concentration of the seminal plasma, but not for the hypoosmotic test and the pregnancy rate. These results suggest the hypoosmotic test may be used for the evaluation of goat semen, but the protein concentration of the seminal plasma can not be used as a parameter to predict the seminal quality and fertility of male goats of the Alpine breed.

Estudou-se a influência da aplicação de somatotropina bovina recombinante sobre a produção e composição do leite, os metabólicos sangüíneos e a concentração hormonal em cabras no terço médio da lactação. Foram utilizadas 24 cabras da raça Toggenburg, divididas em dois tratamentos: T1 (n=12): aplicação de 250 mg de r-bST a cada 14 dias, em um total de quatro aplicações; e T2 (n=12): aplicação de solução salina (controle). O tratamento com r-bST não aumentou a produção de leite e não influenciou os teores de gordura, proteína e extrato seco. A porcentagem de lactose no leite foi maior (4,47 ± 0,2 para T1 versus 4,34 ± 0,2% para T2) e a contagem de células somáticas menor nos animais tratados em relação aos controle (681,1 ± 689,9 para T1 versus 1.001,84 ± 610,9 [x10³ células/mL] para T2). A administração de r-bST aumentou as concentrações séricas de ácidos graxos não-esterificados de T2 (309,67 ± 169,62 x 247,34 ± 126,38 mEq/L, para T1 e T2, respectivamente) e reduziu as concentrações de uréia (86,84 ± 33,81 x 121,16 ± 42,57 mg/dL, para T1 e T2 respectivamente). A r-bST reduziu as concentrações de colesterol total e HDL (82,46 ± 19,25 x 89,29 ± 23,66 mg/dL e 155,95 ± 19,67 x 177,67 ± 32,79 mg/dL, para T1 e T2 respectivamente), enquanto as concentrações de albumina, glicose, proteínas totais, beta-hidroxibutirato e tiroxina não foram influenciadas pela r-bST, que também não influenciou o peso e o escore corporal dos animais. A r-bST aumentou os teores de lactose, reduziu a contagem de células somáticas e promoveu alterações nos metabólicos sangüíneos e no leite de cabras lactantes.

Objetivou-se avaliar os efeitos da criopreservação pelo método convencional em ovócitos imaturos e maturados in vitro. Utilizou-se ovócitos provenientes de ovários de vacas abatidas em matadouro, distribuídos em seis tratamentos: ovócitos não-congelados originados de células do cumulus oophorus (T1) e desnudados (T2) submetidos à MIV, FIV e CIV; ovócitos imaturos, originados de células do cumulus oophorus (T3) e desnudados (T4), congelados, reidratados, e ovócitos considerados normais, submetidos à MIV, FIV e CIV; ovócitos MIV, providos de células do cumulus oophorus (T5) e desnudados (T6), congelados, reidratados e submetidos à FIV e CIV. A congelação dos ovócitos foi realizada em soluções contendo 0,6; 1,2 e 1,8 mol L-1 de Etileno Glicol (EG) durante cinco minutos cada etapa. A descongelação foi realizada em banho-maria a 30 ºC por 20 segundos e, posteriormente, foram reidratados em três etapas (0,9 mol L-1 de EG + 0,3 mol L-1 de sacarose; 0,3 mol L-1 de sacarose, e sem EG e sem sacarose) de seis minutos. A principal alteração ultra-estrutural verificada nos ovócitos maturados in vitro e congelados foi a liberação prematura dos grânulos corticais e, tanto nos maturados quanto nos imaturos congelados, verificou-se vacuolização e redução das cristas mitocondriais. A taxa de maturação foi de 82,5; 75,4; 9,2 e 5,8%, para ovócitos do T1, T2, T3 e T4, respectivamente. As taxas de fecundação foram de 56,2; 0,0; 38,7; 8,6; 63,6 e 16,7% e de clivagem de 36,3; 7,9; 0,4; 0,0; 0,0 e 0,0%, para ovócitos do T1, T2, T3, T4, T5 e T6, respectivamente. Somente os ovócitos do T1 apresentaram desenvolvimento para mórulas e blastocistos (34,5%). Estes resultados indicam que as técnicas de congelação adotadas comprometeram a viabilidade do ovócito.The objective of this study was to evaluate the cryopreservation of in vitro mature or immature oocytes by conventional method. The experiment was conducted using oocytes of cows' ovaries from slaughterhouse, distributed into six treatments: unfrozen oocytes with the cumulus oophorus cells (T1) and naked (T2) which were submitted to the process of MIV, FIV and CIV; immature oocytes, with the cumulus oophorus cells (T3) and naked (T4), which were submitted to the cryopreservation, unfrozen and the MIV, FIV and CIV; oocytes with the cumulus oophorus cells (T5) and naked (T6), which were submitted to in vitro maturation, cryopreservation, and FIV and CIV. The oocytes were frozen by the conventional methods, dehydrated by the immersion in three solutions with 0.6; 1.2 and 1.8 mol L-1 of Ethylene Glycol (EG) during 5 minutes each phase. The thawing phase was done by the immersion in water-bath at 30 ºC during 20 seconds, and so, the oocytes were re-hydrated in three phases (0.9 mol L-1 EG + 0.3 mol L-1 of sacarose; 0.3 L-1 of sacarose without EG and sacarose) for six minutes each one. The mainly ultrastructural changes in cryopreserved matured oocytes were prematurely released of cortical granules. However, the frozen immature and mature oocytes showed vacuolization and disappearance of cristae mitochondrial. The frozen immature oocytes showed the maturation rate of 82.5, 75.4, 9.2 and 5.8% for T1, T2, T3 and T4, respectively. The fecundation rate were 56.2, 0.0, 38.7, 8.6, 63.6 and 16.7% and from the cleavage were 36.3, 7.9, 0.4, 0.0, 0.0 and 0.0% for T1, T2, T3, T4, T5 and T6, respectively. Morula and blastocyst were observed only for unfrozen and naked oocytes (T1) (34.5%). These results showed that the frozen protocols affect the viability of the oocytes.

Objetivou-se avaliar os efeitos da vitrificação em ovócitos de bovinos após o cultivo in vitro, utilizando o etilenoglicol como crioprotetor. Ovócitos obtidos de ovários de vacas abatidas em matadouro foram distribuídos aleatoriamente em três tratamentos. Tratamento 0 (testemunha): ovócitos não-desnudados e não-congelados. Tratamento 1: vitrificação de ovócitos imaturos não desnudados, desidratados previamente por cinco minutos em três soluções contendo 20, 20 e 40% de etilenoglicol, acrescidas de 0,3 mol L-1 de trehalose e 20% de PVP, em meio de Talp Hepes. Tratamento 2: vitrificação de ovócitos imaturos desnudados, conforme o Tratamento 1. Após o descongelamento (imersão em banho-maria a 30ºC por 20 segundos), os ovócitos foram reidratados gradativamente, mantendo-os por 6 minutos em cada uma das soluções a seguir, sucessivamente: meio Talp Hepes com 20% de etilenoglicol + 0,3 mol L-1 de trehalose + 10% de PVP e meio Talp Hepes sem etilenoglicol, trehalose e PVP, onde foram lavados três vezes. Posteriormente, os ovócitos foram cultivados a 38,5ºC, com 95% de umidade e atmosfera de 5% de CO2 por 24 horas. Após o cultivo, os ovócitos foram fecundados e os embriões cultivados in vitro por sete dias. Foi encontrada uma taxa de maturação nuclear de 81 (68/84), 19 (7/36) e 0% (0/31), nos Tratamentos 0, 1 e 2, respectivamente. As taxas de clivagem e de desenvolvimento embrionário foram de 56,4 (102/181) e 54,9% (56/102), 1,7 (1/60) e 0,0% (1/60), 0,0 (0/71) e 0,0% (0/71), nos Tratamentos 0, 1 e 2, respectivamente. Esses resultados indicam que o procedimento de vitrificação, segundo os protocolos utilizados, não é indicado para a criopreservação de ovócitos de bovinos.The objective was to evaluate the effects of vitrification of immature bovine oocytes after in vitro culture, by the use of cryoprotectors ethylene glycol. Oocytes from cows ovaries from slaughters houses were randomly alocated into three treatments. Treatment 0 (control): frozen-thawed undesnude oocytes; treatment 1, immature vitrificated undesnude oocytes dehydrated for 5 minutes in each of the following solutions of 20, 20 and 40% of ethylene glycol, respectively, associated to 0.3 Mol l-1 of trehalose and 20% of PVP, in media Talp Hepes, and, treatment 2, the same as treatment 1, but desnudes oocytes. After frozen-thawed of the oocytes (imersion in water bath at 30ºC for 20 seconds), the oocytes were gradually rehydrated, in the following sequence of solutions: media Talp Hepes with 20% of ethylene glycol + 0.3 Mol l-1 of trehalose + 10% of PVP and media Talp Hepes without ethylene glycol, trehalose and PVP, were washed three times. Ultimately, the oocytes were cultured at 38.5ºC, with 95% umidity and atmosphere of 5% of CO2 for 24 hours. After culture, the oocytes were fertilized and the embryos cultured in vitro for seven days. The nuclear maturation were 81 (68/84), 19 (7/36) and 0% (0/31), for treatments 0, 1 and 2, respectively. The cleavage and development rates were: 56.4(102/181) and 54,9% (56/102), 1,7. (1/60) and 0,0% (1/60), 0,0 (0/71) and 0,0% (0/71), for the treatments 1, 2 e 3, respectively. These results show that the vitrification procedures, by the used protocols, are not indicated for bovine oocytes cryopreservation.

Para avaliar a eficiência reprodutiva e sua relação com características produtivas e composição genética de um rebanho, criado em condições de clima tropical, foram utilizadas 113 vacas, distribuídas em sete grupamentos genéticos, como se segue: 1/2, 3/4, 7/8, 9/16 e 15/16 Holandês x Zebu, Holandês--PB¾PC e Gir. As varáveis estudadas foram: idade ao parto, estação de parição, intervalo do parto ao primeiro serviço, intervalo do primeiro serviço ao serviço fértil, número de serviços, período de serviço, intervalo de partos e eficiência reprodutiva. Com relação a características produtivas, avaliou-se o período de lactação e produção de leite por ordem de lactação e grupamentos genéticos do rebanho. Foi utilizado o programa estatístico SAEG, realizando-se as análises de regressões múltiplas, de variância e de comparação de médias pelo teste de Tukey a 5% de probabilidade de erro. A eficiência reprodutiva foi influenciada por idade ao parto, período de lactação, intervalo de partos e composição genética das vacas, porém não o foi pela produção de leite e estação do ano. Os índices produtivos não foram influenciados pela estação de parição, mas sim pela composição genética dos animais. Neste estudo, os índices reprodutivos mostraram-se satisfatórios, os animais zebuínos apresentaram os menores índices produtivos e os taurinos, a menor eficiência reprodutiva, provavelmente pela maior sensibilidade às condições de manejo.

It was aimed to study the in vitro seminal quality analyzed by complementary tests and to compare them with physical, morphological and biochemical aspects of male goat semen of the Alpine breed. This experiment took place at the Federal University of Viçosa, situated at 20º45’ S latitude and 42º51’ W longitude, Southwest of Brazil. It was done during the summer months of January and February, and three adult male goats of the Alpine breed were used in intensive conditions. The semen was collected by artificial vagina method. In all semen samples (45 ejaculates), after the physical and morphological analysis, the hiposmotic test was done. In 24 ejaculates, it were done thermo-resistance test, and in 21 ejaculates it were determined the concentration of total soluble proteins in seminal plasma. The male goats presented difference in the semen physical and morphological aspects, in the hiposmotic test and thermo-resistance test, but they did not presented difference in total soluble proteins concentration in seminal plasma. Results of the slow thermo-resistance test and hiposmotic test were positively correlated (r = 0.60). Este trabalho teve por objetivo comparar a eficiência do uso da oxitetracilina, administrada pelas vias intramuscular (IM) e intrauterina (IU), no tratamento de endometrites em vacas no período pós-puerperal. Foram utilizados 46 animais mestiços que apresentaram quadro clínico de endometrite, distribuídos aleatoriamente em dois tratamentos: G1 (n=21) – tratado com uma dose de 30 mg/Kg PV de oxitetraciclina por via IM, e G2 (n=25) – uma infusão de 20 mg/Kg PV de oxitetraciclina por via IU.
