The increases in both the elderly population and urbanization present new problems for planning new cities and managing old cities. To solve this challenge, innovative ideas/proposals that encourage walkability and reduce fossil energy motor transport are needed, in line with the actions advocated by Sustainable Development Goal #11 (SDG11), Sustainable Cities and Communities. Opening passages that cross blocks, creating connections between the streets, is the proposal discussed in this paper, as seen in cities like Paris, Prague, Milan, and Juíz de Fora, which have passages in their urban fabric. These can be implemented in a building’s rehabilitation or when a new building replaces it. New architectural project designs should have several characteristics that promote walkability in the city, including passages and galleries. Although previous research has obtained promising results, extending the research to different urban fabrics and block sizes is needed. Using space syntax and data analysis, we simulated and quantitatively assessed the walkability of three neighborhoods in Lisbon, Portugal. The comparison of all simulations and the discussion of the results show that the passages improve walkability. It is also noticeable that there is higher walkability in orthogonal urban fabrics than in non-orthogonal fabrics. The results for block size are inconclusive.

Abstract The pressure for food production has expanded agriculture frontiers worldwide, posing a threat to water resources. For instance, placing crop systems over hydromorphic soils (HS), have a direct impact on groundwater and influence the recharge of riverine ecosystems. Environmental regulations improved over the past decades, but it is difficult to detect and protect these soils. To overcome this issue, we applied a temporal remote sensing strategy to generate a synthetic soil image (SYSI) associated with random forest (RF) to map HS in an 735,953.8 km2 area in Brazil. HS presented different spectral patterns from other soils, allowing the detection by satellite sensors. Slope and SYSI contributed the most for the prediction model using RF with cross validation (accuracy of 0.92). The assessments showed that 14.5% of the study area represented HS, mostly located inside agricultural areas. Soybean and pasture areas had up to 14.9% while sugar cane had just 3%. Here we present an advanced remote sensing technique that may improve the identification of HS under agriculture and assist public policies for their conservation.

Powdery mildews are notorious fungal plant pathogens but only limited information exists on their genomes. Here we present the mitochondrial genome of the grape powdery mildew fungus Erysiphe necator and a high-quality mitochondrial gene annotation generated through cloning and Sanger sequencing of full-length cDNA clones. The E. necator mitochondrial genome consists of a circular DNA sequence of 188,577 bp that harbors a core set of 14 protein-coding genes that are typically present in fungal mitochondrial genomes, along with genes encoding the small and large ribosomal subunits, a ribosomal protein S3, and 25 mitochondrial-encoded transfer RNAs (mt-tRNAs). Interestingly, it also exhibits a distinct gene organization with atypical bicistronic-like expression of the nad4L/nad5 and atp6/nad3 gene pairs, and contains a large number of 70 introns, making it one of the richest in introns mitochondrial genomes among fungi. Sixty-four intronic ORFs were also found, most of which encoded homing endonucleases of the LAGLIDADG or GIY-YIG families. Further comparative analysis of five E. necator isolates revealed 203 polymorphic sites, but only five were located within exons of the core mitochondrial genes. These results provide insights into the organization of mitochondrial genomes of powdery mildews and represent valuable resources for population genetic and evolutionary studies.

Abstract Large gaps in reef distribution may hinder the dispersal of marine organisms, interrupting processes vital to the maintenance of biodiversity. Here we show the presence and location of extensive reef habitats on the continental shelf between the Amazon Reef System (ARS) and the Eastern Brazilian Reef System (ERS), two reef complexes off eastern South America. Formations located 20–50 m deep include both biogenic and geogenic structures. The presence of diverse reef assemblages suggests the widespread occurrence of rocky substrates below 50 m. These habitats represent an expansion of both the ARS and ERS and the closure of the only remaining large-scale gap (~ 1000 km) among West Atlantic reef environments. This indicates that the SW Atlantic harbors a single, yet heterogeneous, reef system that stretches for about 4000 km, and thus, represents one of the largest semi-continuous tropical marine ecosystems in the world.

The deep-sea covers over 70% of the Earth’s surface and harbors predominantly uncharacterized bacterial communities. Actinobacteria are the major prokaryotic source of bioactive natural products that find their way into drug discovery programs, and the deep-sea is a promising source of biotechnologically relevant actinobacteria. Previous studies on actinobacteria in deep-sea sediments were either regionally restricted or did not combine a community characterization with the analysis of their bioactive potential. Here we characterized the actinobacterial communities of upper layers of deep-sea sediments from the Arctic and the Atlantic (Azores and Madeira) ocean basins, employing 16S rRNA metabarcoding, and studied the biosynthetic potential of cultivable actinobacteria retrieved from those samples. Metabarcoding analysis showed that the actinobacterial composition varied between the sampled regions, with higher abundance in the Arctic samples but higher diversity in the Atlantic ones. Twenty actinobacterial genera were detected using metabarcoding, as a culture-independent method, while culture-dependent methods only allowed the identification of nine genera. Isolation of actinobacteria resulted on the retrieval of 44 isolates, mainly associated with Brachybacterium, Microbacterium, and Brevibacterium genera. Some of these isolates were only identified on a specific sampled region. Chemical extracts of the actinobacterial isolates were subsequently screened for their antimicrobial, anticancer and anti-inflammatory activities. Extracts from two Streptomyces strains demonstrated activity against Candida albicans. Additionally, eight extracts (obtained from Brachybacterium, Brevibacterium, Microbacterium, Rhodococcus, and Streptomyces isolates) showed significant activity against at least one of the tested cancer cell lines (HepG2 and T-47D). Furthermore, 15 actinobacterial extracts showed anti-inflammatory potential in the RAW 264.4 cell model assay, with no concomitant cytotoxic response. Dereplication and molecular networking analysis of the bioactive actinobacterial extracts showed the presence of some metabolites associated with known natural products, but one of the analyzed clusters did not show any match with the natural products described as responsible for these bioactivities. Overall, we were able to recover taxonomically diverse actinobacteria with different bioactivities from the studied deep-sea samples. The conjugation of culture-dependent and -independent methods allows a better understanding of the actinobacterial diversity of deep-sea environments, which is important for the optimization of approaches to obtain novel chemically-rich isolates.

Introduction: Third-degree burns are known to affect the epidermis, dermis, hypodermis and adjacent structures, causing damage to the dermal matrix. To date, the gold standard treatment is autologous skin grafting. However, its effectiveness is limited owing to its availability and cost. Depending on the severity of the wound, the maturation of the graft can take several weeks and the tcosmetic results are often unsatisfactory. Given the need to develop new therapies to improve the management and quality of life of patients, cell therapy has become a promising alternative. Objective: To describe the evolution of non-grafted third-degree burns after subcutaneous application of autologous bone marrow (BM) total nucleated cells (TNCs). Case report: We present the case of a 52-year-old male patient with a third-degree scald on the external surface of the left foot. Nine days after the burn, 2 ml of autologous BM TNCs were applied subcutaneously around the wound. An autologous skin graft was not performed; therefore, healing was by second intention. The patient had good clinical evolution without any keloid scars or complications. Conclusion: Although current treatments for third-degree burns still have limitations, recent studies have shown the effectiveness of stem cells in these types of burns by allowing accelerated healing. However, further studies are required to demonstrate their efficacy and safety.

Humanity’s way of life has been irreversibly transformed by new technological advancements during the past decades. Although such technological innovations have been gradually transposed into architecture, their full integration is not yet achieved. This article addresses the issue of incorporating cutting-edge technologies (such as smart thermostats, lighting sensors, security cameras, remote commands, graphic user interfaces, smartphones, mobile apps, gestures, voice commands, etc.) into urban small-scale residential architecture, in the future evolution context. For this purpose, a methodology was conceived that the main concepts regarding automation and information networks were researched, as well as their practice in some reference architecture cases. The guidelines for the prototype architectonic design were defined based on the previous knowledge acquired. Then, a prototype design of an intelligent home was iteratively developed as a machine for living in constant change. It was expected to contribute to increasing and disseminating knowledge in these fields, explaining their benefits and limitations. The prototype design presented in this article contributes to sensitizing architecture professionals to the importance of integrated and systematized thinking in all procedures of a smart home design.

ABSTRACT Soil apparent electrical conductivity (ECa) sensors have been used to detect spatial variability because they correlate with soil attributes. Studies with soil attributes have shown that the number of subsamples and sampling points influences mapping. However, there are no studies that investigated the influence of sampling or subsampling density on ECa maps. Therefore, this study verified the influence of ECa readings per sample point on the semivariance and kriging analysis. The data were collected from an area (2.5 ha) of coffee plants. One hundred sampling points were measured considering 20 readings each. 1, 5, 10, 15, and 20 sample point readings were tested. The influence of the number of readings per sampling point on the ECa mapping was determined using linear regression analysis at a significance level of 5%. The results obtained showed that ECa readings per sampling point significantly influence ECa maps. In addition, they demonstrated that reducing the number of readings per sampling point increases prediction errors by kriging. Thus, ECa maps determined with the highest readings per sampling point were mostly accurate.

ABSTRACT There exist two options for digital cameras that can capture the near-infrared (NIR) band. Conventional red–green–blue (RGB, visible bands) cameras with a single sensor provide NIR band visibility based on the removal of the internal NIR-blocking filter. Alternatively, multisensor cameras exist that have a specific sensor for each band. The modified RGB cameras are of a lower price. In this context, the objective of this study was to compare the performance of a modified RGB camera with that of a multisensor camera for obtaining the normalized difference vegetation index (NDVI) in an area with coffee cultivations. A multispectral camera with five sensors and another camera with only one sensor were used. The NDVI of the coffee field was also measured using the GreenSeeker handheld NDVI sensor manufactured by Trimble. The images were calibrated radiometrically based on the targets in shades of gray made of napa, and the NDVI was calculated after image calibration. The calibration curves showed a high coefficient of determination. The NDVI value obtained with the calibrated images from the cameras showed a significant correlation with the values obtained by the GreenSeeker NDVI sensor, making it possible to obtain the variability pattern of the vegetation index. However, the NDVI obtained using the multisensor camera was closer to the NDVI obtained by the GreenSeeker NDVI sensor.

Previous research has demonstrated a benefit of music training on a number of cognitive functions including verbal memory performance. The impact of school-based music programs on memory processes is however relatively unknown. The current study explored the effect of increasing frequency and intensity of classroom-based instrumental training program on verbal and visual memory across a two year period. Data from 142 participants were analysed: 82 (41 female (F), 41 male (M); mean age 8.62 years) allocated to the intensive strings music training program and 68 (37F, 31M; mean age 8.79 years) allocated to the control group (usual music classes). The Children's Memory Scale was used to test digit span, verbal learning and immediate and delayed verbal memory. Immediate visual recall was tested using the Benton Visual Designs test. Mixed model ANOVAs revealed that students receiving the intensive music training program exhibited significantly better learning and immediate recall for verbal information, after approximately one year, but not two years, after implementation of the program than did controls. No such benefit was observed following one year of a similarly novel juggling program in a sub-set of the sample. As anticipated, the intensive music training had no effect on visual memory, although an improvement in visual perceptual ability and digit span recall was observed in the first year also. The current findings have implications for educational programs (such as literacy training) which may benefit from transient improvements in verbal memory. (Contains 1 table and 1 figure.)

Background: A commonly described analgesic protocol for ovariohysterectomy (OHE) combines systemic opioids, sedatives, and non-steroidal anti-inflammatory drugs. However, systemic analgesia does not fully prevent perioperative visceral and somatic pain triggered by the surgical stimulus. Objectives: To compare the analgesic effects and quality of recovery of systemic analgesia with those of a sacrococcygeal epidural injection of lidocaine and morphine in cats undergoing elective OHE. Methods: Twenty domestic female cats were premedicated with dexmedetomidine (0.01 mg kg−1 IM) and alfaxalone (1.5 mg kg−1 IM) and randomly assigned to one of two analgesic protocols: methadone (0.2 mg kg−1 IM) in the control group CTR (n = 10) and methadone (0.1 mg kg−1 IM) + epidural lidocaine 2% (0.3 mL kg−1) + morphine 1% (0.1 mg kg−1) diluted with NaCl 0.9% to a total volume of 1.5 mL in the SCC-E group (n = 10). General anaesthesia was induced with alfaxalone (1 mg kg−1 IV) and maintained with sevoflurane in 100% oxygen. Non-invasive blood arterial pressure and cardiorespiratory variables were recorded. The quality of recovery was assessed using a simple descriptive scale. Before surgery and 1, 2, 3, 4, 6, and 8 h post-op pain was assessed using the UNESP-Botucatu multidimensional composite pain scale (MCPS) and mechanical nociception thresholds (MNT). The repeated measures analysis of variance (ANOVA) was used to compare groups over time. Comparison between groups was performed using independent samples t-test if the assumption of normality was verified, or the Mann–Whitney test. The chi-square test of independence and exact Fisher’s test were used to compare groups according to recovery quality. Results: Heart rate and systolic arterial pressure increased significantly from baseline values in the CTR group and did not change in the SCC-E group. In the CTR group, MNT and UNESP-Botucatu-MCPS scores increased significantly from baseline for all assessment points and the first 3 h, respectively, whereas this did not occur in the SCC-E group. Conclusions and clinical relevance: Based on our results, the SCC-E administration of lidocaine 2% with morphine 1% is a reasonable option to provide perioperative analgesia in cats submitted to OHE, compared to a systemic protocol alone.

Objetivo: Evaluar los resultados estéticos y funcionales, así como el grado de satisfacción, posteriores a la reconstrucción de defectos de la extremidad inferior en niños con colgajo de dorsal ancho preservador del músculo como una alternativa para estos casos. Método: Se incluyeron pacientes menores de 18 años con defectos en la extremidad inferior tratados en un periodo de 3 años. Se realizó colgajo dorsal ancho preservador de músculo en todos los pacientes. Se valoraron la apariencia estética, la fuerza muscular del área donadora y la función en el área receptora a los 3 meses de realizado el colgajo. Resultados: Se realizaron 13 colgajos de dorsal ancho preservadores de músculo. La apariencia estética de las áreas donadora y receptora fue satisfactoria en la mayoría de los casos. La mayoría de los pacientes presentaron una fuerza muscular normal (M5) en el sitio donador y pudieron deambular. Conclusiones: El colgajo de dorsal ancho preservador de músculo tiene un grado de satisfacción muy elevado, excelente simetría y preservación de la función muscular del sitio donador. Es una técnica ideal en defectos tisulares importantes de la extremidad inferior en la población pediátrica.

Beneficial microorganisms play essential roles in soil fertility, plant nutrition, and health. In this study, we examined the potential of a collection of 138 bacterial strains to promote plant growth. The strains were isolated from the rhizosphere of two monocotyledonous and two dicotyledonous plant species and from bare fallow soil, all from the same site. Our interest in this study was to investigate the diversity and the potential for growth promotion in this collection of culturable bacteria. The most common trait was phosphorus (P) solubilization from aluminium phosphate (in 66.7% of the strains), whereas solubilization of P from phytic acid (6.5%) and from iron phosphate (5.8%) was the least common and they were only detected in bacterial strains from faba bean and oilseed rape. All bacterial strains inhibited the growth of Fusarium graminearum (from 5.4% to 87.2%). In total, 10 genera were identified among the strains by 16S rRNA sequencing and Pseudomonas was the most common in monocotyledonous plants and in bulk soil, while Stenotrophomonas was dominant in the rhizosphere of the dicotyledonous plants. Combinations of bacterial strains improved the spectrum of in vitro activity in most cases, however, wheat growth was generally lower. These strains have potential to be used as biofertilizers and/or biocontrol agents and further studies should be pursued to develop them into practical solutions for a more sustainable agricultural production. Abstract Osteoporosis is a metabolic disorder characterized by a loss of bone mass and structure and increasing the risk of fragility fractures, mostly among postmenopausal women. Sheep is a recognized large animal model for osteoporosis research. An experimental group of ewes (3-4 years old) was subjected to ovariectomy (OVX) and weekly glucocorticoid (GC) application for 24 weeks and compared with a sham control group. Blood and bone marrow parameters were analyzed before and 24 weeks after OVX and GC administration. Osteopenia was confirmed through micro-computed tomography and histomorphometric analysis of L4 vertebra in the study end. Corticotroph cells give rise to aggressive and rare pituitary neoplasms comprising ACTH-producing adenomas resulting in Cushing disease (CD), clinically silent ACTH adenomas (SCA), Crooke cell adenomas (CCA) and ACTH-producing carcinomas (CA). The molecular pathogenesis of these tumors is still poorly understood. To better understand the genomic landscape of all the lesions of the corticotroph lineage, we sequenced the whole exome of three SCA, one CCA, four ACTH-secreting PA causing CD, one corticotrophinoma occurring in a CD patient who developed Nelson syndrome after adrenalectomy and one patient with an ACTH-producing CA. The ACTH-producing CA was the lesion with the highest number of single nucleotide variants (SNV) in genes such as USP8, TP53, AURKA, EGFR, HSD3B1 and CDKN1A. The USP8 variant was found only in the ACTH-CA and in the corticotrophinoma occurring in a patient with Nelson syndrome. In CCA, SNV in TP53, EGFR, HSD3B1 and CDKN1A SNV were present. HSD3B1 and CDKN1A SNVs were present in all three SCA, whereas in two of these tumors SNV in TP53, AURKA and EGFR were found. None of the analyzed tumors showed SNV in USP48, BRAF, BRG1 or CABLES1. The amplification of 17q12 was found in all tumors, except for the ACTH-producing carcinoma. The four clinically functioning ACTH adenomas and the ACTH-CA shared the amplification of 10q11.22 and showed more copy-number variation (CNV) gains and single-nucleotide variations than the nonfunctioning tumors.

Marine biofilms are known to influence the corrosion of metal surfaces in the marine environment. Despite some recent research, the succession of bacterial communities colonizing artificial surfaces remains uncharacterized in some temporal settings. More specifically, it is not fully known if bacterial colonizers of artificial surfaces are similar or distinct in the different seasons of the year. In particular the study of early biofilms, in which the bacterial cells communities first adhere to artificial surfaces, are crucial for the development of the subsequent biofilm communities. In this work, we used amplicon-based NGS (next-generation sequencing) and universal 16S rRNA bacterial primers to characterize the early biofilm bacterial communities growing on 316 L stainless steel surfaces in a Northern Portugal port. Sampling spanned 30-day periods in two distinct seasons (spring and winter). Biofilm communities growing in steel surfaces covered with an anti-corrosion paint and planktonic communities from the same location were also characterized. Our results demonstrated that distinct temporal patterns were observed in the sampled seasons. Specifically, a significantly higher abundance of Gammaproteobacteria and Mollicutes was found on the first days of biofilm growth in spring (day 1 to day 4) and a higher abundance of Alphaproteobacteria during the same days of biofilm growth in winter. In the last sampled day (day 30), the spring biofilms significantly shifted toward a dominance of photoautotrophic groups (mostly diatoms) and were also colonized by some macrofouling communities, something not observed during the winter sampling. Our results revealed that bacterial composition in the biofilms was particularly affected by the sampled day of the specific season, more so than the overall effect of the season or overall sampling day of both seasons. Additionally, the application of a non-fouling-release anti-corrosion paint in the steel plates resulted in a significantly lower diversity compared with plates without paint, but this was only observed during spring. We suggest that temporal succession of marine biofilm communities should be taken in consideration for future antifouling/anti-biofilm applications.

Abstract The uniformity of water application is an important factor in the evaluation of sprinkler irrigation systems. This uniformity depends on the type of sprinkler and its operating conditions, such as the arrangement and spacing between the sprinklers in the area; velocity and wind direction during the period of water application and the pressure variation of the irrigation system. The objective of this study was to model, analyze and compare the structure of spatial dependence, as well as the spatial variability of the water depths applied by a sprinkler irrigation system with compensating and non-compensating sprinklers, using geostatistical methods and measurements of accuracy or similarity between the applied water depth maps. The experiment was carried out in an agricultural area, in the city of Cascavel-Paraná-Brazil. A total area of 10 × 10 m was used, with 04 compensating and 04 non-compensating sprinklers installed at a height of 1.5 m. For each type of sprinkler, water levels were measured in 100 collectors spaced 1 × 1 m in the study area in 32 trials. On each test sprinkling was carried out for one hour. The conditions of wind, temperature and air humidity were evaluated at the beginning of each test and at 10-minute intervals with a climatological station. As the geostatistical analysis showed the existence of directional trends, the coordinates were incorporated as covariates to the linear spatial model in the study of the spatial dependence of the average depth of the irrigation water for the two types of sprinklers. The spatial dependence structure that best fits the data when using the compensating sprinklers was the Gaussian model and when the non-compensating sprinklers were used, it was the exponential model. The spatial variability maps of average irrigation water depth (mm) of the trials, obtained by universal kriging, revealed that for both sprinklers there was an increase in the mean level average values in the northwest-southeast direction (135° in the azimuth system) in the area under study, influenced by wind direction and velocity during the execution of the experiment.

Abstract Background Currently, the best resources for assessment of bone tissue using imaging techniques are expensive and available in few medical facilities, thus serum or urinary bone turnover biomarkers could be useful as early indicators of prognosis. However, there is a wide range of variability in bone turnover markers due to several factors, such as different ages and metabolic stages, thus it is important to have as much data published on the subject as possible. The aim of this study was therefore to generate a reference range for alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRAP) and validate the already published data. Results Serum alkaline phosphatase decreased with age, with statistical difference between the 1 month old and the other groups and between the over 8 years and the 6 months old groups. There was also a statistical difference in the ALP levels between the 3 to 5 years old gestation and lactation groups. For serum tartrate-resistant acid phosphatase, there was a statistical difference mainly between the 1 month old and the 6 months old, 6–8 years old, and above 8 years old groups. Conclusions The results obtained could represent a useful tool for future studies using sheep as an animal model for orthopedic research. The different groups presented a wide variation of serum ALP and TRAP levels, however, these variations are entirely explained by known physiology. Therefore, this detailed study confirms the prediction that unexplained changes in these bone turnover markers do not occur during the lifespan of sheep.

Abstract The introduction of nanomaterials as detection reagents has enabled improved sensitivity and facilitated detection in a variety of bioanalytical assays. However, high nanoprobe densities are typically needed for colorimetric detection and to circumvent this limitation several enhancement protocols have been reported. Nevertheless, there is currently a lack of universal, enzyme-free and versatile methods that can be readily applied to existing as well as new biosensing strategies.
