Abstract

There are obvious benefits to integrating generative AI (artificial intelligence) into language learning and teaching. Those include using AI as a language tutor, creating learning materials, or assessing learner output. However, due to how AI systems under-stand human language, based on a mathematical model using statistical probability, they lack the lived experience to be able to use language with the same social aware-ness as humans. Additionally, there are built-in linguistic and cultural biases based on their training data which is mostly in English and predominantly from Western sources. Those facts limit AI suitability for some language learning interactions. Stud-ies have clearly shown that systems such as ChatGPT often do not produce language that is pragmatically appropriate. The lack of linguistic and cultural authenticity has important implications for how AI is integrated into second language acquisition as well as in instruction targeting development of intercultural communication compe-tence.

5. Distributed agency in second language learning and teaching through generative AI

Abstract

Generative AI offers significant opportunities for language learning. Tools like ChatGPT can provide informal second language practice through chats in written or voice forms, with the learner specifying through prompts conversational parameters such as proficiency level, language register, and discussion topics. AI can be instructed to give corrective feedback, create practice exercises, or develop an extended study plan. Instructors can use AI to build learning and assessment materials in a variety of media. AI is likely to make immersive technologies more powerful and versatile, moving away from scripted interactions. For both learners and teachers, it is important to understand the limitations of AI systems that arise from their purely statistical model of human language, which limits their ability to deal with nuanced social and cultural aspects of language use. Additionally, there are ethical concerns over how AI systems are created as well as practical constraints in their use, especially for less privileged populations. The power and versatility of AI tools are likely to turn them into valuable and constant companions in many peoples lives (akin to smartphones), creating a close connection that goes beyond simple tool use. Ecological theories such as sociomaterialism are helpful in examining the shared agency that develops through close user-AI interactions, as are the perspectives on human-object relations from Indigenous cultures., Comment: 26 pages. 7. Active Suppression of Quantum Dephasing in Resonantly Driven Ensembles

Abstract

We have used quantum control to suppress the impact of random atom positions on coherent population transfer within atom pairs, enabling the observation of dipole-dipole driven Rabi oscillations in a Rydberg gas with hundreds of atoms. The method exploits the reduced coupling-strength sensitivity of the off-resonant Rabi frequency, and coherently amplifies the achievable population transfer in analogy to quasi-phase-matching in non-linear optics. Simulations reproduce the experimental results and demonstrate the potential benefits of the technique to other many-body quantum control applications.

15. Chatbots in Language Learning: AI Systems on the Rise

Abstract

The use of chatbots in language learning has been on the rise. In recent Computer-Assisted Language Learning (CALL) research, there is a consensus that rule-based, scripted voice systems are optimal for language learning. Such systems integrate well into instructed language learning in that interactions with the user are predictable and controlled. Open, AI-based voice systems (such as in personal assistants like Siri) do not provide that degree of task-oriented learning. However, the argument is made here that they have the potential to provide open-ended conversational practice and language development which aligns with an ecological, usage-based perspective on language development. Abstract

The PI3K/AKT/mTOR (PAM) signaling pathway is a highly conserved signal transduction network in eukaryotic cells that promotes cell survival, cell growth, and cell cycle progression. Growth factor signalling to transcription factors in the PAM axis is highly regulated by multiple cross-interactions with several other signaling pathways, and dysregulation of signal transduction can predispose to cancer development. The PAM axis is the most frequently activated signaling pathway in human cancer and is often implicated in resistance to anticancer therapies. Dysfunction of components of this pathway such as hyperactivity of PI3K, loss of function of PTEN, and gain-of-function of AKT, are notorious drivers of treatment resistance and disease progression in cancer. In this review we highlight the major dysregulations in the PAM signaling pathway in cancer, and discuss the results of PI3K, AKT and mTOR inhibitors as monotherapy and in co-administation with other antineoplastic agents in clinical trials as a strategy for overcoming treatment resistance. Finally, the major mechanisms of resistance to PAM signaling targeted therapies, including PAM signaling in immunology and immunotherapies are also discussed.

Abstract

Ibrutinib is a first-in-class Bruton's tyrosine kinase inhibitor approved for the treatment of various B-cell malignancies and chronic graft-versus-host disease. We evaluated the safety and efficacy of ibrutinib, alone or combined with standard-of-care regimens, in adults with advanced urothelial carcinoma (UC). Once-daily ibrutinib was administered orally at 840 mg (single-agent or with paclitaxel) or at 560 mg (with pembrolizumab). Phase 1b determined the recommended phase 2 dose (RP2D) of ibrutinib, and phase 2 assessed progression-free survival (PFS), overall response rate (ORR), and safety. Thirty-five, eighteen, and fifty-nine patients received ibrutinib, ibrutinib plus pembrolizumab, and ibrutinib plus paclitaxel at the RP2D, respectively. Safety profiles were consistent with those of the individual agents. The best-confirmed ORRs were 7% (two partial responses) with single-agent ibrutinib and 36% (five partial responses) with ibrutinib plus pembrolizumab. Median PFS was 4.1 months (range, 1.0-37.4+) with ibrutinib plus paclitaxel. The best-confirmed ORR was 26% (two complete responses). In previously treated patients with UC, ORR was higher with ibrutinib plus pembrolizumab than with either agent alone (historical data in the intent-to-treat population). ORR with ibrutinib plus paclitaxel was greater than historical values for single-agent paclitaxel or ibrutinib. These data warrant further evaluation of ibrutinib combinations in UC.

Abstract

We numerically compute the lowest Laplacian eigenvalues of several two-dimensional shapes with dihedral symmetry at arbitrary precision arithmetic. Our approach is based on the method of particular solutions with domain decomposition. We are particularly interested in asymptotic expansions of the eigenvalues $\lambda(n)$ of shapes with $n$ edges that are of the form $\lambda(n) \sim x\sum_{k=0}^{\infty} \frac{C_k(x)}{n^k}$ where $x$ is the limiting eigenvalue for $n\rightarrow \infty$. Expansions of this form have previously only been known for regular polygons with Dirichlet boundary condition and (quite surprisingly) involve Riemann zeta values and single-valued multiple zeta values, which makes them interesting to study. We provide numerical evidence for closed-form expressions of higher order $C_k(x)$ and give more examples of shapes for which such expansions are possible (including regular polygons with Neumann boundary condition, regular star polygons and star shapes with sinusoidal boundary)., Comment: 20 pages

Abstract

Charge migration (CM) is a coherent attosecond process that involves the movement of localized holes across a molecule. To determine the relationship between a molecule's structure and the CM dynamics it exhibits, we perform systematic studies of para-functionalized bromobenzene molecules (X-C$_6$H$_4$-R) using real-time time-dependent density functional theory. We initiate valence-electron dynamics by emulating rapid strong-field ionization leading to a localized hole on the bromine atom. The resulting CM, which takes on the order of 1 fs, occurs via an X localized to C$_6$H$_4$ delocalized to R localized mechanism. Interestingly, the hole contrast on the acceptor functional group increases with increasing electron donating strength. This trend is well-described by the Hammett sigma value of the group, which is a commonly used metric for quantifying the effect of functionalization on the chemical reactivity of benzene derivatives. Abstract

We introduce high-harmonic sideband spectroscopy (HHSS) and show that it can be a robust probe of attosecond charge migration (CM) in a halogenated carbon-chain molecule. We simulate both the CM and harmonic-generation (HHG) dynamics using ab initio time-dependent density-functional theory. We find that CM dynamics initiated along the molecular backbone induces sidebands in the HHG spectrum driven by a delayed laser pulse that is polarized perpendicular to the molecular axis. Monitoring the spectrum as either the HHG laser frequency or the relative delay is scanned allows for the extraction of detailed information about the time-domain characteristics of the CM process., Comment: 8 pages, 7 figures

26. Emerging Spaces for Language Learning: AI Bots, Ambient Intelligence, and the Metaverse

Abstract

Looking at human communication from the perspective of semiotics extends our view beyond verbal language to consider other sign systems and meaning-making resources. Those include gestures, body language, images, and sounds. From this perspective, the communicative process expands from individual mental processes of verbalizing to include features of the environment, the place and space in which the communication occurs. It may be--and it is increasingly the case today--that language is mediated through digital networks. Online communication has become multimodal in virtually all platforms. At the same time, mobile devices have become indispensable digital companions, extending our perceptive and cognitive abilities. Advances in artificial intelligence are enabling tools that have considerable potential for language learning, as well as creating more complexity in the relationship between humans and the material world. In this column, we will be looking at changing perspectives on the role of place and space in language learning, as mobile, embedded, virtual, and reality-augmenting technologies play an ever-increasing role in our lives. Understanding that dynamic is aided by theories and frameworks such as 4E cognition and sociomaterialism, which posit closer connections between human cognition/language and the world around us.

Abstract

Cell-free RNA from liquid biopsies can be analyzed to determine disease tissue of origin. We extend this concept to identify cell types of origin using the Tabula Sapiens transcriptomic cell atlas as well as individual tissue transcriptomic cell atlases in combination with the Human Protein Atlas RNA consensus dataset. We define cell type signature scores, which allow the inference of cell types that contribute to cell-free RNA for a variety of diseases.

Abstract

Molecular characterization of cell types using single-cell transcriptome sequencing is revolutionizing cell biology and enabling new insights into the physiology of human organs. We created a human reference atlas comprising nearly 500,000 cells from 24 different tissues and organs, many from the same donor. This atlas enabled molecular characterization of more than 400 cell types, their distribution across tissues, and tissue-specific variation in gene expression. Using multiple tissues from a single donor enabled identification of the clonal distribution of T cells between tissues, identification of the tissue-specific mutation rate in B cells, and analysis of the cell cycle state and proliferative potential of shared cell types across tissues. Cell type-specific RNA splicing was discovered and analyzed across tissues within an individual.

Abstract

For more than 100 years, the fruit fly Drosophila melanogaster has been one of the most studied model organisms. Here, we present a single-cell atlas of the adult fly, Tabula Drosophilae, that includes 580,000 nuclei from 15 individually dissected sexed tissues as well as the entire head and body, annotated to >250 distinct cell types. We provide an in-depth analysis of cell type-related gene signatures and transcription factor markers, as well as sexual dimorphism, across the whole animal. Analysis of common cell types between tissues, such as blood and muscle cells, reveals rare cell types and tissue-specific subtypes. This atlas provides a valuable resource for the Drosophila community and serves as a reference to study genetic perturbations and disease models at single-cell resolution.

Abstract

Alzheimer's disease (AD) is a progressive neurodegenerative disease observed with aging that represents the most common form of dementia. To date, therapies targeting end-stage disease plaques, tangles, or inflammation have limited efficacy. Therefore, we set out to identify a potential earlier targetable phenotype. Utilizing a mouse model of AD and human fetal cells harboring mutant amyloid precursor protein, we show cell intrinsic neural precursor cell (NPC) dysfunction precedes widespread inflammation and amyloid plaque pathology, making it the earliest defect in the evolution of the disease. We demonstrate that reversing impaired NPC self-renewal via genetic reduction of USP16, a histone modifier and critical physiological antagonist of the Polycomb Repressor Complex 1, can prevent downstream cognitive defects and decrease astrogliosis in vivo. Reduction of USP16 led to decreased expression of senescence gene Cdkn2a and mitigated aberrant regulation of the Bone Morphogenetic Signaling (BMP) pathway, a previously unknown function of USP16. Thus, we reveal USP16 as a novel target in an AD model that can both ameliorate the NPC defect and rescue memory and learning through its regulation of both Cdkn2a and BMP signaling.

Abstract

Background: Deep vein thrombosis (DVT) is a common vascular problem seen in the emergency department (ED) and is commonly identified using ultrasound performed by a vascular lab, the radiology department, or at the point of care. Previous studies have assessed the utility of a two-point vs sequential technique to identify the presence of a thrombus. One particular study reported a concerning rate of isolated femoral vein thrombi that would be missed by a two-point technique.Objectives: In this study we sought to determine whether the two-point technique misses isolated femoral vein thrombi.Methods: We conducted a retrospective review of patients who had a new diagnosis of DVT in the ED diagnosed with vascular lab, radiology, or point-of-care ultrasound to assess for the presence and rate of thrombi that would be missed using a two-point scanning technique.Results: We included in our study 356 patients with a diagnosis of new DVT. In our population, 21 (5.9%; 0.95 confidence interval: 3.7%, 8.9%) patients were identified with thrombi isolated to the femoral vein.Conclusion: The two-point technique for lower extremity vascular ultrasound is insufficient for ruling out proximal DVTs in ED patients.

38. Re-Orienting CALL through the Lens of Complexity Theory

Abstract

Complexity Theory (CT) provides a useful framework for understanding Second Language Acquisition (SLA). Using an ecological model, CT studies the dynamic processes of change and emergent outcomes over time, tracing back how trajectories may have been affected by changes in and interactions among multiple variables and subsystems. Language learners do not follow linear learning paths, but rather their progress depends on a variety of interlocking variables. Developmental trajectories will look different for each learner. That is particularly the case for informal language learning, in which learners co-adapt from a wide variety of resources, leading to divergent outcomes (Godwin-Jones, 2018a). A CT approach emphasizes the dynamics of the interplay between learner variables and the people, artifacts, and services constituting the L2 learning system. It aligns well with usage-based linguistics. Viewing language learning from a CT perspective has a number of consequences for Computer Assisted Language Learning (CALL) research, explored here. 39. Learning in the Open: Integrating Language and Culture through Student Curation, Virtual Exchange, and OER

Abstract

To address waning enrollments in intermediate 202 language courses, faculty have developed a multiphasic project. In phase one, alongside instructional designers, they created an open, connected platform where 202 students curate -- search, select, and share -- authentic materials online. In phase two, upper-level students help triage and scaffold the best curations into online interactive modules. Phase three sets up live, virtual exchanges during which students discuss their curations with native speakers. In phase four, open e-textbooks, framed by communicative can-do statements, integrate curations, modules, virtual exchanges, and existing Open Educational Resources (OERs) into sustainable, relevant, and student driven learning materials. 40. 'Triage, Transition, and Transformation': Advocacy Discourse in Urban School Reform

Abstract

Advocacy coalitions have the potential to be a vehicle for community-based education reform in urban school systems, where state legislatures have increasingly adopted top-down policies such as state takeover and accountability systems. Yet, coalitions are influenced by and create their own informal and formal power structures that can include or exclude certain stakeholders and perspectives. In this study, the Advocacy Coalition Framework was used alongside critical discourse analysis of interviews, documents, and more than 50 news articles to explore how power and ideology shaped policy in the Coalition for the Future of Detroit Schoolchildren. We find that personal stories, political loyalties, and prior reform experiences shaped the narratives of Coalition members. While discourse from and about the Coalition was narrower in scope and representation during a tough legislative battle, the group's policy victories and organizational infrastructure created potential for substantive community-led reform in the years following. This suggests that community-based education reform may require advocates to strategically sequence the promotion of diverse stakeholder interests in order to achieve broad coalition goals.

41. Can Effect Sizes Give Any Clue to the Way Mentors Ascribe Numerical Grades When Assessing Trainee Teachers against the Teachers' Standards in England?

Abstract

Some teacher educators use numerical grades when assessing teaching competencies. In this situation, statistical analysis can be used to monitor consistency and look for correlations between assessment outcomes across teacher training partnerships and at different stages in training. Another approach is to calculate effect size metrics. These do not claim statistical significance but do seek to explain the practical impact of patterns in quantitative data. This study looks at number grade assessment data from a large secondary initial teacher education programme across schools working in partnership with a higher education provider in the Northwest of England. The proportion of variance between numerical grades for individual Teachers' Standards and overall teaching was calculated at each formal review point over three consecutive years. Despite the complex process involved in assessing teaching competencies against performance criteria and the potential for subjective variation between individual assessors, the data consistently demonstrated underlying patterns. These suggested that quality assurance and management of assessment issues could have been a major influence on the assessors.

Abstract

We report a resource-efficient scheme in which a single pump laser was used to achieve frequency conversion by Bragg-scattering four-wave mixing in a photonic crystal fiber. We demonstrate bidirectional conversion of coherent light between Sr+ P1/2 - D3/2 emission wavelength at 1092 nm and the telecommunication C band with conversion efficiencies of 4.2 % and 37 % for up- and down-conversion, respectively. We discuss how the scheme may be viably scaled to meet the temporal, spectral and polarisation stability requirements of a hybrid light-matter quantum network., Comment: 5 pages, 4 figures
