Abstract

Ibrutinib is a first-in-class Bruton's tyrosine kinase inhibitor approved for the treatment of various B-cell malignancies and chronic graft-versus-host disease. We evaluated the safety and efficacy of ibrutinib, alone or combined with standard-of-care regimens, in adults with advanced urothelial carcinoma (UC). Once-daily ibrutinib was administered orally at 840 mg (single-agent or with paclitaxel) or at 560 mg (with pembrolizumab). Phase 1b determined the recommended phase 2 dose (RP2D) of ibrutinib, and phase 2 assessed progression-free survival (PFS), overall response rate (ORR), and safety. Thirty-five, eighteen, and fifty-nine patients received ibrutinib, ibrutinib plus pembrolizumab, and ibrutinib plus paclitaxel at the RP2D, respectively. Safety profiles were consistent with those of the individual agents. The best-confirmed ORRs were 7% (two partial responses) with single-agent ibrutinib and 36% (five partial responses) with ibrutinib plus pembrolizumab. Median PFS was 4.1 months (range, 1.0-37.4+) with ibrutinib plus paclitaxel. The best-confirmed ORR was 26% (two complete responses). In previously treated patients with UC, ORR was higher with ibrutinib plus pembrolizumab than with either agent alone (historical data in the intent-to-treat population). ORR with ibrutinib plus paclitaxel was greater than historical values for single-agent paclitaxel or ibrutinib. These data warrant further evaluation of ibrutinib combinations in UC.

Abstract

We report a resource-efficient scheme in which a single pump laser was used to achieve frequency conversion by Bragg-scattering four-wave mixing in a photonic crystal fiber. We demonstrate bidirectional conversion of coherent light between Sr+ P1/2 - D3/2 emission wavelength at 1092 nm and the telecommunication C band with conversion efficiencies of 4.2 % and 37 % for up- and down-conversion, respectively. We discuss how the scheme may be viably scaled to meet the temporal, spectral and polarisation stability requirements of a hybrid light-matter quantum network., Comment: 5 pages, 4 figures

Abstract

We report a bright and tunable source of spectrally pure heralded single photons in the telecom O-Band, based on cross-polarized four wave mixing in a commercial birefringent optical fiber. The source can achieve a purity of 85%, heralding efficiency of 30% and high coincidences to accidentals ratio of 108. Furthermore, the possibility of building multiple identical sources is explored. Through the measurements of joint spectral intensities, we find that the fiber is homogeneous over atleast 45 centimeters and can potentially realize 4 identical sources. This paves the way for a cost-effective fiber-optic approach to implement multi-photon quantum optics experiments., Comment: 14 pages, 10 figures

Abstract

We use hollow-core fibre to preserve the spectrum and temporal profile of picosecond laser pulses in CBD to suppress 2.6 dB of amplitude noise at MHz noise frequencies, to within 0.01 dB of the shot-noise limit. We provide an enhanced version of the CBD scheme that concatenates circuits to suppress over multiple frequencies and over broad frequency ranges --- we perform a first demonstration that reduces total excess amplitude noise, between 2 - 6 MHz, by 85%. Abstract

Third-order nonlinear processes require phase matching between the interacting fields to achieve high efficiencies. Typically in guided-wave $\chi^{(3)}$ platforms this is achieved by engineering the dispersion of the modes through the transverse profile of the device. However, this limits the flexibility of the phase matching that can be achieved. Instead, we analyze four-wave mixing in a pair of asymmetric waveguides and show that phasematching may be achieved in any $\chi^{(3)}$ waveguide by coupling of a nondegenerate pump from an adjacent waveguide. We demonstrate the additional flexibility that this approach yields in the case of photon-pair generation by spontaneous FWM, where the supermode dispersion may be modified to produce pure heralded single photons -- a critical capability required for example by silicon platforms for chip-scale quantum photonics., Comment: 11 pages, 5 figures

Abstract

We report a single-stage bi-directional interface capable of linking Sr+ trapped ion qubits in a long-distance quantum network. Our interface converts photons between the Sr+ emission wavelength at 422 nm and the telecoms C-band to enable low-loss transmission over optical fiber. We have achieved both up- and down-conversion at the single photon level with efficiencies of 9.4% and 1.1% respectively. Abstract

We present a multiplexed single photon source that re-synchronises heralded single photons generated by four-wave mixing in photonic crystal fibre using a fibre delay loop controlled by a single switch. By combining the probabilities of generating heralded single photons on four consecutive pump pulses we obtain an enhancement of the per-bin single-photon delivery probability. Our source demonstrates a way in which single-photon source multiplexing may be carried out with minimised resource overhead while retaining the benefits of a robust and alignment-free platform.

17. Fibre-integrated noise gating of high-purity heralded single photons

Abstract

We present an all-fibre source of high-purity heralded single photons with an integrated conditional optical gate that reduces uncorrelated noise by almost an order of magnitude. Generating photon pairs by four-wave mixing in photonic crystal fibre, we observe with the noise gate active a factor of 7 reduction in the rate of single counts in the heralded channel with no measurable drop in coincidence count rate. In contrast to electronic post-selection of coincidence events, the real reduction in the flux of unwanted photons is beneficial for example to avoid bleaching light-sensitive samples or in generating entangled states.

Abstract

What is this summary about? This summary describes the results from the TALAPRO-2 research study (also known as a clinical trial). The TALAPRO-2 study tested the combination of two medicines called talazoparib plus enzalutamide. This combination of medicines was used as the first treatment for adult patients with metastatic castration-resistant prostate cancer. The combination of talazoparib plus enzalutamide was compared with a placebo plus enzalutamide. What is metastatic castration-resistant prostate cancer? Metastatic castration-resistant prostate cancer is a type of cancer that starts in the prostate and has spread to other parts of the body. Castration-resistant means that the cancer continues to grow even when testosterone levels in the blood are reduced to very low levels. Taking medicines to lower testosterone levels in the blood is a standard treatment for men with advanced prostate cancer. What are the aims of the TALAPRO-2 trial? TALAPRO-2 looked at if combining talazoparib plus enzalutamide would increase the length of time patients lived before their cancer got worse or they died compared with a placebo plus enzalutamide. Researchers looked at how treatment affected the size and number of tumors and the length of time before patients needed to change to a new cancer medicine. Researchers also looked at any side effects patients had during the study. What are the key takeaways? A total of 805 patients with metastatic castration-resistant prostate cancer took part in the study. Compared with patients who took a placebo plus enzalutamide, the group of patients who took talazoparib plus enzalutamide had a 37% reduced risk of their cancer getting worse or dying. Some patients had tumors that at the start of the study could be measured with scans. Sixty-two percent of patients who took talazoparib plus enzalutamide had their tumors decrease or shrink to the point that they could no longer be seen on scans versus 44% of patients who took a placebo plus enzalutamide. Patients who took talazoparib plus enzalutamide were more likely to have a longer time before they needed to change to a new cancer medicine. The most common side effects of talazoparib plus enzalutamide were low levels of red blood cells (66% of patients) and neutrophils (36% of patients), and excessive tiredness or exhaustion (34% of patients). Abstract

We present a method by which the degree of longitudinal variation in photonic crystal fibre (PCF) may be characterised through seeded four-wave mixing (FWM). Using an iterative numerical reconstruction, we created a model PCF that displays similar FWM phasematching properties across all measured length scales. Our results demonstrate that the structure of our PCF varies by less than 1% and that the characteristic length of the variations is approximately 15 cm., Comment: 11 pages, 7 figures

Abstract

Single photon sources based on spontaneous photon-pair generation have enabled pioneering experiments in quantum optics. However, their non-determinism presents a bottleneck to scaling up photonic and hybrid quantum-enhanced technologies. Furthermore, photon pairs are typically emitted into many correlated frequency modes, producing an undesirable mixed state on heralding. Here we present a complete fibre-integrated heralded single photon source that addresses both these difficulties simultaneously. We use active switching to provide a path to deterministic operation by multiplexing separate spontaneous sources, and dispersion engineering to minimise frequency correlation for high-purity single photon generation. All the essential elements -- nonlinear material with dispersion control, wavelength isolation, optical delay, and fast switching -- are incorporated in a low-loss alignment-free package that heralds photons in telecoms single-mode fibre. Abstract

Single photons are a vital resource for photonic quantum information processing. However, even state-of-the-art single photon sources based on photon-pair generation and heralding detection have only a low probability of delivering a single photon when one is requested. We analyse a scheme that uses a switched fibre delay loop to increase the delivery probability per time bin of single photons from heralded sources. We show that, for realistic experimental parameters, combining the output of up to 15 pulses can yield a performance improvement of a factor of 10. Abstract

In recent years, there has been rapid development in processing of quantum information using quantum states of light. The focus is now turning towards developing real-world implementations of technologies such as all-optical quantum computing and cryptography. The ability to consistently create and control the required single photon states of light is crucial for successful operation. Therefore, high performance single photon sources are very much in demand. The most common approach of generating the required nonclassical states of light is through spontaneous photon pair generation in a nonlinear medium. One photon in the pair is detected to "herald" the presence of the remaining single photon. For many applications the photons are required to be in pure indistinguishable states. However, photon pairs generated in this manner typically suffer from spectral correlations, which can lead to the production of mixed, distinguishable states. Additionally, these sources are probabilistic in nature, which fundamentally limits the number of photons that can be delivered simultaneously by independent sources and hence the scalability of these future technologies. One route to deterministic operation is by actively multiplexing several independent sources together to increase the probability of delivering a single photon from the system. This thesis presents the development and analysis of a multiplexing scheme of heralded single photons in high-purity indistinguishable states within an integrated optical fibre system. The spectral correlations present between the two photons in the pair were minimised by spectrally engineering each photonic crystal fibre source. A novel, in-fibre, broadband filtering scheme was implemented using photonic bandgap fibres. In total, two sources were multiplexed using a fast optical switch, yielding an 86% increase in the heralded count rate from the system.

Four-wave mixing (FWM) in optical fibre is a leading technique for generating high-quality photon pairs. We report the generation of photon pairs by spontaneous FWM in photonic crystal fibre pumped by a 1.5 GHz repetition-rate vertical-external-cavity surface-emitting laser (VECSEL). The photon pairs exhibit high count rates and a coincidence-to-accidental ratio of over 80. The VECSEL's high repetition-rate, high average power, tunability, and small footprint make this an attractive source for quantum key distribution and photonic quantum-state engineering., Comment: 17 Pages, 5 Figures

Abstract

We demonstrate a numerical technique that can evaluate the core-to-core variations in propagation constant in multicore fibre. Using a Markov Chain Monte Carlo process, we replicate the interference patterns of light that has coupled between the cores during propagation. We describe the algorithm and verify its operation by successfully reconstructing target propagation constants in a fictional fibre. Then we carry out a reconstruction of the propagation constants in a real fibre containing 37 single-mode cores. We find that the range of fractional propagation constant variation across the cores is approximately $\pm2 \times 10^{-5}$., Comment: 17 pages; preprint format; 5 figures. Current sources of heralded single photons based on nonlinear optics operate in a probabilistic manner. In order to build quantum-enhanced devices based around the use of single photons, compact, turn-key and deterministic sources are required. A possible solution is to multiplex a number of sources to increase the single-photon generation probability and in so doing reducing the waiting time to deliver large numbers of photons simultaneously, from independent sources. Previously it has been shown that, in the ideal case, 17 multiplexed sources allow deterministic generation of heralded single photons [Christ and Silberhorn, Phys. Rev. A 85, 023829 (2012)]. Here we extend this analysis to include undesirable effects of detector inefficiency and photon loss on a number of multiplexed sources using a variety of different detectors for heralding.
