In an era of unprecedented global change, exploring patterns of gene expression among wild populations across their geographic range is crucial for characterizing adaptive potential. RNA-sequencing studies have successfully characterized gene expression differences among populations experiencing divergent environmental conditions in a wide variety of taxa. However, few of these studies have identified transcriptomic signatures to multivariate, environmental stimuli among populations in their natural environments. Herein, we aim to identify environmental and sex-driven patterns of gene expression in the Tasmanian devil (Sarcophilus harrisii), a critically endangered species that occupies a heterogeneous environment. We performed RNA-sequencing on ear tissue biopsies from adult male and female devils from three populations at the extremes of their geographic range. There were no transcriptome-wide patterns of differential gene expression that would be suggestive of significant, environmentally-driven transcriptomic responses. The general lack of transcriptome-wide variation in gene expression levels across the devil's geographic range is consistent with previous studies that documented low levels of genetic variation in the species. However, genes previously implicated in local adaptation to abiotic environment in devils were enriched for differentially expressed genes. Additionally, three modules of co-expressed genes were significantly associated with either population of origin or sex.

Emerging infectious diseases (EIDs) not only cause catastrophic declines in wildlife populations but also generate selective pressures that may result in rapid evolutionary responses. One such EID is devil facial tumour disease (DFTD) in the Tasmanian devil. DFTD is almost always fatal and has reduced the average lifespan of individuals by around 2 years, likely causing strong selection for traits that reduce susceptibility to the disease, but population decline has also left Tasmanian devils vulnerable to inbreeding depression. We analysed 22 years of data from an ongoing study of a population of Tasmanian devils on Freycinet Peninsula, Tasmania, to (1) identify whether DFTD may be causing selection on body size, by estimating phenotypic and genetic correlations between DFTD and size traits, (2) estimate the additive genetic variance of susceptibility to DFTD, and (3) investigate whether size traits or susceptibility to DFTD were under inbreeding depression. We found a positive phenotypic relationship between head width and susceptibility to DFTD, but this was not underpinned by a genetic correlation. Conversely, we found a negative phenotypic relationship between body weight and susceptibility to DFTD, and there was evidence for a negative genetic correlation between susceptibility to DFTD and body weight. There was additive genetic variance in susceptibility to DFTD, head width and body weight, but there was no evidence for inbreeding depression in any of these traits. These results suggest that Tasmanian devils have the potential to respond adaptively to DFTD, although the realised evolutionary response will critically further depend on the evolution of DFTD itself. Estimating past biodiversity using the fossil record is a central goal of palaeobiology. Because raw estimates of biodiversity are biased by variation in sampling intensity across time, space, environments and taxonomic groups, sampling standardization is routinely applied when estimating taxonomic diversity (e.g. species richness). However, sampling standardization is less commonly used when estimating alternative currencies of biological diversity, such as morphological disparity. Here, we show the effects of standardizing fossil time series of morphological disparity to equal sample completeness, or 'coverage', of the underlying taxon‐frequency distribution. We apply coverage‐based standardization to three published datasets of discrete morphological characters (echinoderms, ichthyosaurs and ornithischian dinosaurs), and quantify disparity using two metrics: weighted mean pairwise dissimilarity (WMPD) and the sum of variance (SOV). We also compare the effects of coverage‐based and sample‐size‐based standardization. Our results show that coverage standardization can yield estimates of disparity through time that dramatically deviate from raw estimates, both in magnitude and direction of changes. These findings demonstrate that future studies of morphological disparity should control for variation in sampling intensity to enable more reliable inferences. [ABSTRACT FROM AUTHOR]

Identifying the genetic architecture of complex phenotypes is a central goal of modern biology, particularly for disease-related traits. Genome-wide association methods are a classical approach for identifying the genomic basis of variation in disease phenotypes, but such analyses are particularly challenging in natural populations due to sample size difficulties. Extensive mark-recapture data, strong linkage disequilibrium and a lethal transmissible cancer make the Tasmanian devil (Sarcophilus harrisii) an ideal model for such an association study. We used a RAD-capture approach to genotype 624 devils at ~16,000 loci and then used association analyses to assess the heritability of three cancer-related phenotypes: infection case-control (where cases were infected devils and controls were devils that were never infected), age of first infection and survival following infection. The SNP array explained much of the phenotypic variance for female survival (>80%) and female case-control (>61%). We found that a few large-effect SNPs explained much of the variance for female survival (~5 SNPs explained >61% of the total variance), whereas more SNPs (~56) of smaller effect explained less of the variance for female case-control (~23% of the total variance). By contrast, these same SNPs did not account for a significant proportion of phenotypic variance in males, suggesting that the genetic bases of these traits and/or selection differ across sexes. Loci involved with cell adhesion and cell-cycle regulation underlay trait variation, suggesting that the devil immune system is rapidly evolving to recognize and potentially suppress cancer growth through these pathways. Overall, our study provided necessary data for genomics-based conservation and management in Tasmanian devils.

Tasmanian eucalypt forests are among the most carbon‐dense in the world, but projected climate change could destabilize this critical carbon sink. While the impact of abiotic factors on forest ecosystem carbon dynamics have received considerable attention, biotic factors such as the input of animal scat are less understood. Tasmanian devils (Sarcophilus harrisii)—an osteophageous scavenger that can ingest and solubilize nutrients locked in bone material—may subsidize plant and microbial productivity by concentrating bioavailable nutrients (e.g., nitrogen and phosphorus) in scat latrines. However, dramatic declines in devil population densities, driven by the spread of a transmissible cancer, may have underappreciated consequences for soil organic carbon (SOC) storage and forest productivity by altering nutrient cycling. Here, we fuse experimental data and modeling to quantify and predict future changes to forest productivity and SOC under various climate and scat‐quality futures. We find that devil scat significantly increases concentrations of nitrogen, ammonium, phosphorus, and phosphate in the soil and shifts soil microbial communities toward those dominated by r‐selected (e.g., fast‐growing) phyla. Further, under expected increases in temperature and changes in precipitation, devil scat inputs are projected to increase above‐ and below‐ground net primary productivity and microbial biomass carbon through 2100. In contrast, when devil scat is replaced by lower‐quality scat (e.g., from non‐osteophageous scavengers and herbivores), forest carbon pools are likely to increase more slowly, or in some cases, decline. Together, our results suggest often overlooked biotic factors will interact with climate change to drive current and future carbon pool dynamics in Tasmanian forests. [ABSTRACT FROM AUTHOR]

The research underpinning this article explores the impacts that parenting and primary caring responsibilities have upon academic careers. It takes an innovative approach by exploring three under-researched aspects of this issue: the longitudinal impacts that extend past the years immediately following the birth or adoption of a child; the differences and similarities that occur as a result of primary carer parenting across academic disciplines; and the impacts that primary carer parenting has upon the careers of those who are employed in senior academic levels. With these three aspects in mind, the research explores the strategies employed to cope with the impacts that birth or adoption has upon academic careers. In doing so, it broadens conceptual understandings of the career impacts of parenting, while offering recommendations for university administrators and leaders.

Unmodified forests are increasingly rare worldwide, with forestry a major contributor to habitat modification. Extending conservation practices beyond protected areas is important to conserve forest ecosystems.We investigate the response of native mammalian carnivores (both Order Carnivora and Family Dasyuridae) to production forests globally, including harvested native forest and timber plantations. We examine how carnivores recorded in production forests use these forests versus other land uses, particularly native and/or unharvested forest; how habitat use relates to threatened status, body size, diet and harvesting method; carnivore responses to habitat features within production forests; and carnivore denning, breeding and predation behaviour in production forests.We review 294 studies recording 132 carnivore species in production forests. Carnivores generally show higher use of unharvested native forests and lower use of agricultural land than production forests. Threatened species and large carnivores respond more negatively to production forests than non‐threatened species and small carnivores respectively. Hypercarnivores respond more negatively than omnivores to plantations compared to native forest, with no difference in the use of harvested and unharvested native forest between these dietary groups.Notably, a high proportion of carnivore species use clearfelled more than unharvested native forest. In forest with partial harvesting or reduced‐impact logging, most species show no difference in use between harvested and unharvested forest.Carnivores generally respond positively to habitat features such as riparian areas and coarse woody debris. Several carnivores were recorded denning and breeding in production forests. Production forests often influence the prey availability, hunting success and diet of carnivores.We show that many carnivores use production forests, and how they respond to production forestry varies with species traits and conservation status. We recommend that production forests are managed as valuable carnivore habitat, and highlight strategies to enhance the use of these forests by carnivores. [ABSTRACT FROM AUTHOR]

Coevolution is common and frequently governs host-pathogen interaction outcomes. Phenotypes underlying these interactions often manifest as the combined products of the genomes of interacting species, yet traditional quantitative trait mapping approaches ignore these intergenomic interactions. Devil facial tumor disease (DFTD), an infectious cancer afflicting Tasmanian devils (Sarcophilus harrisii), has decimated devil populations due to universal host susceptibility and a fatality rate approaching 100%. Here, we used a recently developed joint genome-wide association study (i.e., co-GWAS) approach, 15 y of mark-recapture data, and 960 genomes to identify intergenomic signatures of coevolution between devils and DFTD. Using a traditional GWA approach, we found that both devil and DFTD genomes explained a substantial proportion of variance in how quickly susceptible devils became infected, although genomic architectures differed across devils and DFTD; the devil genome had fewer loci of large effect whereas the DFTD genome had a more polygenic architecture. Using a co-GWA approach, devil-DFTD intergenomic interactions explained ~3× more variation in how quickly susceptible devils became infected than either genome alone, and the top genotype-by-genotype interactions were significantly enriched for cancer genes and signatures of selection. A devil regulatory mutation was associated with differential expression of a candidate cancer gene and showed putative allele matching effects with two DFTD coding sequence variants. Our results highlight the need to account for intergenomic interactions when investigating host-pathogen (co)evolution and emphasize the importance of such interactions when considering devil management strategies. Landscape modification is a major threat to carnivores worldwide, but modified landscapes can also provide important habitat for these species, as protected areas alone are insufficient. Understanding how carnivores use modified landscapes, such as production forests, can inform management strategies to improve the value of these landscapes to carnivores. Little is known about habitat selection by marsupial carnivores in production forests, where they occupy a similar ecological niche to their more well‐studied eutherian counterparts. We used GPS tracking, Hidden Markov Models, and Manly's selection ratios to identify the habitat selected in three behavioral states by the largest marsupial carnivore, the Tasmanian devil (Sarcophilus harrisii), in a timber plantation‐dominated landscape. Behavioral states were approximated as: state 1, resting or feeding at a carcass; state 2, foraging; and state 3, travel. Devils did not show preferences for any of native forest, native grassland, and plantation in any behavioral state. Within plantations, devils preferred a plantation age of 4–7 years (selection ratio [wi] = 1.52). Devils preferred roads (state 1: wi = 2.71, state 2: wi = 2.48, state 3: wi = 2.97) and plantation edges (state 1: wi = 2.38, state 2: wi = 2.24, state 3: wi = 2.78) in all behavioral states, and moved faster on roads and edges than away from them. Together, our results indicate devils use road and edges for foraging (scavenging and hunting) and travel. No measured habitat variables influenced devil home range size. To support devils in plantation landscapes, we recommend maintaining a heterogeneous landscape of different plantation ages and native remnants and reducing the risk of vehicle collisions by minimizing forestry traffic at night. Tasmanian devils share similar adaptable traits to generalist eutherian carnivore species in their use of modified landscapes. Plantations can provide valuable habitat for this and other threatened predator species. [ABSTRACT FROM AUTHOR]
