Abstract

The pseudogap Anderson impurity model is a paradigm for locally critical quantum phase transitions. Within the framework of the local moment approach we study its finite-T dynamics, as embodied in the single-particle spectrum, in the vicinity of the symmetric quantum critical point (QCP) separating generalized Fermi-liquid (Kondo screened) and local moment phases. The scaling spectra in both phases, and at the QCP itself, are obtained analytically. A key result is that pure omega/T-scaling obtains at the QCP, where the Kondo resonance has just collapsed. The connection between the scaling spectra in either phase and that at the QCP is explored in detail., Comment: 12 pages, 7 figures

Abstract

Urokinase-type plasminogen activator (uPA) and its receptor (uPAR) play an important role in cell guidance and chemotaxis during normal and pathological events, uPAR is GPI-anchored and the mechanism by which it transmits intracellular polarity cues across the plasma membrane during directional sensing has not been elucidated. The constitutively recycling endocytic receptor Endo180 forms a trimolecular complex with uPAR in the presence of uPA, hence its alternate name uPAR-associated protein. Here, we demonstrate that Endo180 is a general promoter of random cell migration and has a more specific function in cell chemotaxis up a uPA gradient. Endo180 expression was demonstrated to enhance uPA-mediated filopodia production and promote rapid activation of Cdc42 and Rac. Expression of a noninternalizing Endo180 mutant revealed that promotion of random cell migration requires receptor endocytosis, whereas the chemotactic response to uPA does not. From these studies, we conclude that Endo180 is a crucial link between uPA-uPAR and setting of the internal cellular compass.

Abstract

Dielectric constant measurements were made, in the frequency range 100 kHz to 1 MHz, on the compounds glycine, diglycine, D-sorbitol, dextran 15-20,000) and. Ficoll (m.w t. 400,000) in aqueous solutions of low specific conductivity (< 200 x 10-4 ohm-1 m-1) normally buffered to pH 7.45. The values obtained were used to predict the dielectric constant of Hanks-199 tissue culture medium, to which various concentrations of these compounds had been added. Single-cell dispersions of two primary chick embryonic tissues, 7-day neural retina and 5-day limb bud, were prepared in tissue culture media of varying dielectric constant and also in suitable controls. Ionic strength, pH and. osmolarity were kept equal in experimental and control media. Selected cell dispersions were examined by means of particle electrophoresis in order to determine their electrokinetic potentials. It was found that 7-day neural retina tissue had a mean value for the zeta potential of -13.88 mV +/- 0.04 S.E. in unmodified medium, this value falling slightly in the media of differing composition. The 5-day limb bud tissue gave a value for the zeta potential of -14.37 mV +/- 0.03 S.E. in normal medium, again showing a slight falling off in the modified media. The results were interpreted as showing that no significant adsorption of added compounds was occurring onto the cell membranes, thus changing their surface properties. Cell suspensions in media of a range of dielectric constant were subjected to a laminar flow shear gradient in a Couette viscometer. The effect of this shear gradient was to bring about collisions between the cells and thus to effect aggregation of these suspensions. By measuring the total number of particles in the suspension at timed intervals a parameter was calculated for each aggregation, the collision efficiency. The collision efficiency is a measure of the rate of aggregation and also can be used to calculate the total energy of adhesive interaction of the cells. The collision efficiency was used to calculate this parameter from aggregation studies in media of varying dielectric constant. It was shown firstly that 5-day limb bud tissue gave much lower values for this parameter (termed the force constant) than did 7-day neural retina. In the control media, the former gave a value of 3.66 x 10-26 J. and the latter 5.65 x 10-23 J. Secondly it was discovered that in both tissues there was a steady increase in the adhesive interaction of the cells (as measured by the force constant) with increasing dielectric constant of the aggregation medium, the 5-day limb bud being far more sensitive than 7-day neural retina in this respect. These observations were discussed in relation to the lyophobic colloid stability theory of cell adhesion. The results were found to be consistent with this theory when account was taken of recent theoretical studies into the effect of the dielectric properties of a medium on the attractive energy of particles suspended in that medium.

Abstract

Podosomes are a singular category of integrin-mediated adhesions important in the processes of cell migration, matrix degradation and cancer cell invasion. Despite a wealth of biochemical studies, the effects of mechanical forces on podosome integrity and dynamics are poorly understood. Here, we show that podosomes are highly sensitive to two groups of physical factors. First, we describe the process of podosome disassembly induced by activation of myosin-IIA filament assembly. Next, we find that podosome integrity and dynamics depends upon membrane tension and can be experimentally perturbed by osmotic swelling and deoxycholate treatment. We have also found that podosomes can be disrupted in a reversible manner by single or cyclic radial stretching of the substratum. We show that disruption of podosomes induced by osmotic swelling is independent of myosin-II filaments. The inhibition of the membrane sculpting protein, dynamin-II, but not clathrin, resulted in activation of myosin-IIA filament formation and disruption of podosomes. The effect of dynamin-II inhibition on podosomes was however independent of myosin-II filaments. Moreover, formation of organized arrays of podosomes in response to microtopographic cues (the ridges with triangular profile) was not accompanied by reorganization of myosin-II filaments. Thus, mechanical elements such as myosin-II filaments and factors affecting membrane tension/sculpting independently modulate podosome formation and dynamics, underlying a versatile response of these adhesion structures to intracellular and extracellular cues.This article is part of the discussion meeting issue ‘Forces in cancer: interdisciplinary approaches in tumour mechanobiology’.

Abstract

This work is licensed under a Creative Commons Attribution 4.0 International License., Invadopodia are actin-rich cell membrane projections used by invasive cells to penetrate the basement membrane. Control of invadopodia stability is critical for efficient degradation of the extracellular matrix (ECM); however, the underlying molecular mechanisms remain poorly understood. Here, we uncover a new role for podoplanin, a transmembrane glycoprotein closely associated with malignant progression of squamous cell carcinomas (SCCs), in the regulation of invadopodia-mediated matrix degradation. Podoplanin downregulation in SCC cells impairs invadopodia stability, thereby reducing the efficiency of ECM degradation. We report podoplanin as a novel component of invadopodia-associated adhesion rings, where it clusters prior to matrix degradation. Early podoplanin recruitment to invadopodia is dependent on lipid rafts, whereas ezrin/moesin proteins mediate podoplanin ring assembly. Finally, we demonstrate that podoplanin regulates invadopodia maturation by acting upstream of the ROCK-LIMK-Cofilin pathway through the control of RhoC GTPase activity.
