- Abstract
The West Antarctic Ice Sheet (WAIS) is one of the largest potential sources of future sea-level rise, with glaciers draining the WAIS thinning at an accelerating rate over the past 40 years. Due to complexities in calibrating palaeoceanographic proxies for the Southern Ocean, it remains difficult to assess whether similar changes have occurred earlier during the Holocene or whether there is underlying centennial- to millennial-scale forcing in oceanic variability. Archaeal lipid-based proxies, specifically glycerol dialkyl glycerol tetraether (GDGT; e.g. TEX86 and TEXL86), are powerful tools for reconstructing ocean temperature, but these proxies have been shown previously to be difficult to apply to the Southern Ocean. A greater understanding of the parameters that control Southern Ocean GDGT distributions would improve the application of these biomarker proxies and thus help provide a longer-term perspective on ocean forcing of Antarctic ice sheet changes. In this study, we characterised intact polar lipid (IPL)-GDGTs, representing (recently) living archaeal populations in suspended particulate matter (SPM) from the Amundsen Sea and the Scotia Sea. SPM samples from the Amundsen Sea were collected from up to four water column depths representing the surface waters through to Circumpolar Deep Water (CDW), whereas the Scotia Sea samples were collected along a transect encompassing the sub-Antarctic front through to the southern boundary of the Antarctic Circumpolar Current. IPL-GDGTs with low cyclic diversity were detected throughout the water column with high relative abundances of hydroxylated IPL-GDGTs identified in both the Amundsen and Scotia seas. Results from the Scotia Sea show shifts in IPL-GDGT signatures across well-defined fronts of the Southern Ocean. Two experiments were conducted to verify the feeding value of NutriDense (ND) and NutriDense Low-Phytate (NDLP) corn (Exseed Genetics LLC, BASF Plant Science, Research Triangle Park, NC) relative to that of yellow dent (YD) corn in swine diets. NutriDense corn is a high-protein, high-oil variety, and NDLP is a high-protein, high-oil, low-phytate variety. In Exp. 1,315 nursery pigs that initially weighed 15.2 kg were used in a 21-d growth assay. Dietary treatments were arranged in a 3 x 3 factorial; main effects were corn source (YD, ND, and NDLP) and added fat (0, 3, or 6%, as-fed basis). Diets were formulated to contain 3.83 g of lysine/Mcal using calculated nutrient values. There were no corn source x fat interactions observed. Pigs fed YD, ND, and NDLP had ADG of 750, 734, and 738 g/d and G:F of 0.64, 0.66, and 0.65, respectively. No differences (P > 0.10) in ADG were observed among the three corn sources; however, pigs fed diets containing either ND or NDLP corn had decreased ADFI (P < 0.02) and improved G:F (P < 0.05) compared with pigs fed diets containing YD corn. Increasing dietary fat increased ADG (727, 746, and 748 g/d; linear, P < 0.04) and G:F (0.62, 0.66, and 0.68; linear, P < 0.01) and decreased ADFI (linear, P < 0.01). Using the NRC (1998) value for ME in YD corn, we calculated the energy value for ND and NDLP based on G:F differences compared with pigs fed YD corn. These data indicated the ME values for ND and NDLP corn are 4.5 and 2.5% greater (3,575 and 3,505 Kcal/ kg), respectively, than for YD corn (3,420 Kcal/kg). In Exp. 2, 1,144 gilts (initial BW = 50.1 kg) were used in a commercial research facility to evaluate the effects of corn source (ND and YD) and added fat (0, 3, or 6%, as-fed basis) in a 2 x 3 factorial on pig performance and carcass traits. There was a corn source x fat interaction for ADFI and G:F. Increasing added fat resulted in greater changes in ADFI and G:F in pigs fed YD corn diets compared with those fed ND corn. Feeding ND corn increased ADG (main effect, P < 0.04), and greater percentages of added fat increased ADG (main effect; linear, P < 0.01). Results of Exp. 2 suggest that ND corn has 5.3% more ME than YD corn. The additional energy provided by ND corn improves G:F in both nursery and grow-finish pigs, and ND corn offers a means of formulating diets more concentrated in energy than YD corn. - Abstract
Clustered, regularly interspaced, short palindromic repeats-CRISPR associated (CRISPR-Cas) systems defend bacteria against foreign nucleic acids, such as during bacteriophage infection and transformation, processes which cause envelope stress. It is unclear if these machineries enhance membrane integrity to combat this stress. Here, we show that the Cas9-dependent CRISPR-Cas system of the intracellular bacterial pathogen Francisella novicida is involved in enhancing envelope integrity through the regulation of a bacterial lipoprotein. This action ultimately provides increased resistance to numerous membrane stressors, including antibiotics. We further find that this previously unappreciated function of Cas9 is critical during infection, as it promotes evasion of the host innate immune absent in melanoma 2/apoptosis associated speck-like protein containing a CARD (AIM2/ASC) inflammasome. Interestingly, the attenuation of the cas9 mutant is complemented only in mice lacking both the AIM2/ASC inflammasome and the bacterial lipoprotein sensor Toll-like receptor 2, but not in single knockout mice, demonstrating that Cas9 is essential for evasion of both pathways. These data represent a paradigm shift in our understanding of the function of CRISPR-Cas systems as regulators of bacterial physiology and provide a framework with which to investigate the roles of these systems in myriad bacteria, including pathogens and commensals.

The design and manufacture of individually customised products is generally restricted to bespoke clothing or footwear for very wealthy customers. The aim of the Custom-Fit project was to develop a fast, flexible and economically viable route for the manufacture of individually customised parts. These products not only provide improved comfort levels but also provide better functional performance, including enhanced safety for the user. This 4.5 year, European Commission subsidised €16 million project, supported by the EU, involving 30 partners across the breadth of the Europe finished in early 2009. This paper will showcase the technology developed: CAD packages which automate the design process and three new rapid manufacturing methods. It will also include case studies on a range of customised products, including customised Motorcycles helmets. The case studies not only demonstrate the performance benefits of individual customisation but also show the potential for new approaches to product design. More information at www.Custom-Fit.org.

We present a measurement of pi^+pi^-pi^+pi^- photonuclear production in ultra-peripheral Au-Au collisions at sqrt(s_{NN}) = 200 GeV from the STAR experiment. The pi^+pi^-pi^+pi^- final states are observed at low transverse momentum and are accompanied by mutual nuclear excitation of the beam particles. The strong enhancement of the production cross section at low transverse momentum is consistent with coherent photoproduction. The pi^+pi^-pi^+pi^- invariant mass spectrum of the coherent events exhibits a broad peak around 1540 pm 40 MeV/c^2 with a width of 570 pm 60 MeV/c^2, in agreement with the photoproduction data for the rho^0(1700). We do not observe a corresponding peak in the pi^+pi^- final state and measure an upper limit for the ratio of the branching fractions of the rho^0(1700) to pi^+pi^- and pi^+pi^-pi^+pi^- of 2.5 % at 90 % confidence level. The ratio of rho^0(1700) and rho^0(770) coherent production cross sections is measured to be 13.4 pm 0.8 (stat.) pm 4.4 (syst.) %., Comment: 9 pages, 7 figures

A radical change (Feenstra, Holmer, Tromans, Moos and Mieritz 2003) in manufacturing is starting to occur. The Rapid Manufacturing, which can be defined as the use of an additive manufacturing process to construct parts that are used directly as finished products or components, is set to supersede many current uses of moulds and dies. Rapid Manufacturing (Wohlers 2003) is based on new additive manufacturing techniques that produce fully functional parts directly from a 3D CAD model without the use of tooling. The ambitious scope of the European Initiative CUSTOM-FIT, a Framework 6 Integrated Project, is to create a fully integrated system for the design, production and supply of individualized custom-products. Within the cases under study there are strong connections to sporting articles. This project is being funded by the European Commission over the next four years and a half and will become central to European research concerning Rapid Manufacturing (RM). © 2006 Springer-Verlag New York.

Treatment of children with steroid-dependent nephrotic syndrome (SDNS) continues to be a challenge when relapses recur after treatment with cyclophosphamide and side effects or non-compliance make steroids and cyclosporin unsatisfactory. We treated 12 patients with intravenous vincristine for SDNS in a regime of 1-1.5 mg/m2 weekly for 4 weeks then monthly for 4 months. Four of the 5 patients in relapse when commencing vincristine remitted within 2 doses. Comparing relapse frequency in the 12 months before and after vincristine, there was a reduction from 4 to 1.5 (p=0.004) relapses per year. Median sustained remission was 5 months, but 1 frequently relapsing patient remains in remission 4 years after vincristine. Vincristine was also successfully used in 1 or 2 doses at weekly intervals for subsequent relapses in 5 patients. Side effects were minimal in most cases. Abdominal pain occurred in 2 patients who commenced vincristine at 1.5 mg/m2, but resolved when continued at 1 mg/m2. We felt vincristine had a role in a subset of children with challenging SDNS administered as 1 mg/m2 weekly for 4 weeks then 1.5 mg/m2 monthly for 4 months. Vincristine allowed steroid- and cyclosporin-sparing, contributed to long-term remission in some patients, and was especially valuable in children with poor compliance with oral medication. Many patients expressed a preference for a few doses of vincristine rather than a standard course of oral prednisolone or cyclosporin. © IPNA 2005.

Background. There is limited population-based epidemiological data on renal disease. An insight into the spectrum of clinically significant glomerulonephritis can be obtained from renal biopsy diagnoses. This is a descriptive report of biopsy-proven glomerulonephritis within a defined population. Methods. A retrospective review of the pathology reports of all native renal biopsies performed in the Australian state of Victoria in 1995 and 1997 was undertaken. Trends in the average annual age- and sex-specific incidence rates for biopsy-proven glomerulonephritis were calculated. Comparisons were made with the incidence rates of end-stage renal disease due to glomerulonephritis confirmed on renal biopsy. Results. The most common glomerulonephritides in adults are IgA disease, focal glomerulosclerosis, lupus nephritis and vasculitis, and in children are lupus nephritis focal glomerulosclerosis, IgA disease and minimal change disease. A male predominance is seen for all glomerulonephritides, except lupus nephritis, in both adults and children. An increase in incidence of disease with age, particularly in males, is seen for vasculitis and focal glomerulosclerosis. The most common glomerulonephritides on renal biopys are reflected in the most common causes of end-stage renal disease due to glomerulonephritis. Conclusions. This review has provided population-based descriptive epidemiological data on clinically significant glomerulonephritis. This data provides important clues for further studies relating to the identification of risk factors for the various types of glomerulonephritis.

A total of 180 weanling pigs (average initial BW of 5.7 kg) were used to determine the effects of stickwater on pelleting characteristics of diets and growth performance of pigs. Treatments were a non-fat control, choice white grease (CWG), and stickwater (SW) substituted for 1/3, 2/3, and 3/3 (DM basis) of the CWG. Diets were formulated to 1.7, 1.5, and 1.3% lysine with 3, 5, and 7% fat for d 0 to 7, 7 to 21, and 21 to 35, respectively. The diets were steam conditioned at 83 C in a CPM Master Model HD pellet mill equipped with a die having 4-mm openings. The diets with fat required less energy to pellet than the nonfat control (P < .001). Energy consumption decreased with as much as 2/3 replacement of the CWG, but complete replacement of CWG with SW increased energy consumption (cubic effect, P < .001). The nonfat control had greater pellet durability index (ASAE, S269.3) than diets with added fat (P < .001), but pellet durability index increased as SW was added to replace the CWG (linear effect, P < .001). When the diets were fed to pigs, there were no differences in ADG, ADFI, or gain/feed for d 0 to 7 or d 7 to 21 (P > .12), but the diets with added fat supported greater gain/feed than the nonfat control for d 21 to 35 (P < .001) and overall (P < .01). There was a trend (quadratic, P < .07) for overall gain/feed to increase when 1/3 of the CWG was replaced with SW and to decrease with total replacement of the CWG. In conclusion, replacement of CWG with SW increased pellet durability and up to 2/3 of the CWG could be replaced with SW without negative effects on growth performance. Our findings suggest that SW is an economically attractive addition to diets for nursery pigs.
