Abstract Updates of SARS-CoV-2 vaccines are required to generate immunity in the population against constantly evolving SARS-CoV-2 variants of concerns (VOCs). Here we describe three novel in-silico designed spike-based antigens capable of inducing neutralising antibodies across a spectrum of SARS-CoV-2 VOCs. Three sets of antigens utilising pre-Delta (T2_32), and post-Gamma sequence data (T2_35 and T2_36) were designed. T2_32 elicited superior neutralising responses against VOCs compared to the Wuhan-1 spike antigen in DNA prime-boost immunisation regime in guinea pigs. Heterologous boosting with the attenuated poxvirus - Modified vaccinia Ankara expressing T2_32 induced broader neutralising immune responses in all primed animals. T2_32, T2_35 and T2_36 elicited broader neutralising capacity compared to the Omicron BA.1 spike antigen administered by mRNA immunisation in mice. These findings demonstrate the utility of structure-informed computationally derived modifications of spike-based antigens for inducing broad immune responses covering more than 2 years of evolved SARS-CoV-2 variants.

Abstract Elucidating the adaptive immune characteristics of natural protection to Lassa fever (LF) is vital in designing and selecting optimal vaccine candidates. With rejuvenated interest in LF and a call for accelerated research on the Lassa virus (LASV) vaccine, there is a need to define the correlates of natural protective immune responses to LF. Here, we describe cellular and antibody immune responses present in survivors of LF (N = 370) and their exposed contacts (N = 170) in a LASV endemic region in Nigeria. Interestingly, our data showed comparable T cell and binding antibody responses from both survivors and their contacts, while neutralizing antibody responses were primarily seen in the LF survivors and not their contacts. Neutralizing antibody responses were found to be cross-reactive against all five lineages of LASV with a strong bias to Lineage II, the prevalent strain in southern Nigeria. We demonstrated that both T cell and antibody responses were not detectable in peripheral blood after a decade in LF survivors. Notably LF survivors maintained high levels of detectable binding antibody response for six months while their contacts did not. Lastly, as potential vaccine targets, we identified the regions of the LASV Glycoprotein (GP) and Nucleoprotein (NP) that induced the broadest peptide-specific T cell responses. Taken together this data informs immunological readouts and potential benchmarks for clinical trials evaluating LASV vaccine candidates.

BackgroundThe virus neutralization assay is a principal method to assess the efficacy of antibodies in blocking viral entry. Due to biosafety handling requirements of viruses classified as hazard group 3 or 4, pseudotyped viruses can be used as a safer alternative. However, it is often queried how well the results derived from pseudotyped viruses correlate with authentic virus. This systematic review and meta-analysis was designed to comprehensively evaluate the correlation between the two assays.MethodsUsing PubMed and Google Scholar, reports that incorporated neutralisation assays with both pseudotyped virus, authentic virus, and the application of a mathematical formula to assess the relationship between the results, were selected for review. Our searches identified 67 reports, of which 22 underwent a three-level meta-analysis.ResultsThe three-level meta-analysis revealed a high level of correlation between pseudotyped viruses and authentic viruses when used in an neutralisation assay. Reports that were not included in the meta-analysis also showed a high degree of correlation, with the exception of lentiviral-based pseudotyped Ebola viruses.ConclusionPseudotyped viruses identified in this report can be used as a surrogate for authentic virus, though care must be taken in considering which pseudotype core to use when generating new uncharacterised pseudotyped viruses.

Abstract This learning module is on oral findings in routine medical imaging. It is designed to improve the ability of the medical student, resident, and practicing radiologist/physician to identify pathology of the oral cavity seen on routine medical imaging, with a special emphasis on the dentition and the supporting tissues. Additionally, changes secondary to dental treatment are illustrated. Recommendations for dental follow-up, if indicated, are noted as a means of optimizing health through collaborative care. This resource is primarily a PowerPoint presentation with background information on oral health and illustrations of common imaging used in dentistry. It also includes a review of cases with imminent dental pathology. At the end of the module, an eight-question self-assessment tool is provided. This module can be utilized for individual or group seminars for medical students and residents, especially radiology residents. It is critical that medical students, residents, and ultimately physicians recognize the signs and appearance of dental disease on radiographs.

ABSTRACT This article reports on a case study of English language learning in higher education, considering the impact of language policies embedded within a mobility scholarship programme on the practices of language teachers. Based on a larger ethnographic study following the experiences of several undergraduate members of one cohort of Brazil’s Science without Borders programme for students in science and technological fields, this article describes how several language teachers engaged with the power relations of the frequently shifting policy terrain of the programme. It concludes with a discussion of possibilities for teachers in similar situations who endeavour to take a critical approach to language teaching and explores some of the tactics they might employ against those policies which they see as incompatible with their practices.

Has Visual International Relations (IR) become too distant from the content of visual artefacts? This is a paradoxical question. Visual IR is a vibrant and pluralist field exploring visuals in innumerable ways. Nonetheless, the field tends to focus on 'deep' readings of the socio-political implications of visual artefacts at the expense of a close and attentive observation and description of the events, situations, or phenomena they may depict. Simply put, visual IR usually analyses visuals-as-visuals rather than seeing them as entry points for studying the social world. But might a video of torture teach us something about the practicality of torture? Might a video of peace negotiations teach us something about their successes or failures? Can we gain a fleeting glimpse of 'reality' within visuals? We address these questions by first situating our focus on close 'visual (data) observation' in conceptual conversation with the literature's existing focus on deep interpretation. Second, we outline three approaches to visual observation as they are deployed outside IR. Third, we unpack how those approaches might be of value for IR, especially vis-à-vis the study of practice, materiality, and discourse. This paper explores lithium's role, its chemical properties, and the environmental and economic considerations associated with its extraction and recovery. We examine various lithium recovery methods, including conventional techniques such as hydrometallurgy, pyrometallurgy, and direct physical recycling, as well as emerging technologies like mechanochemistry, ion pumping, and bioleaching while emphasizing the need for sustainable practices to address environmental challenges. The review also discusses the challenges and future directions of lithium recovery. Advances in technology have streamlined lithium recovery processes and spurred the development of innovative extraction techniques. This paper concludes with an emphasis on further research essential to deepen our understanding of lithium recovery methods, their strengths and limitations, and the need to explore new strategies to meet the growing global demand for this indispensable resource. How are socially and politically controversial security practices materially-technologically scripted into our lives in ever-deeper ways? This essay proposes that acts of aesthetic design are at the heart of that process and are being deployed by technology corporations to "smooth" the diffusion of security practices, discourses, and politics across global space. To substantiate that claim, we make three moves. First, we propose an understanding of the "script" that returns to the roots of the concept in theater and the arts. That understanding stresses that our material-technological enmeshing is governed strongly by aesthetic and affective factors that operate through forms of resonance (rather that reason). In consequence—we argue—much of the power of technology conglomerates is linked to their capacity to harness these aesthetico-affective resonances. Second, we demonstrate this through a case study focused on the material-aesthetic design activities of Google, teasing out how it deploys aesthetic practices to extend its sociopolitical power. Third, we speculatively conclude by introducing the architect Keller Easterling's concept of "active form" to show how conceptualizing scripts in aesthetic terms also provides insights into how security practices are diffused across global contexts, irrelevant contextual sociopolitical differences, and seemingly without any limit. What's at stake in debates over critique and postcritique in International Relations (IR)? To subvert that question, this article is written in the format of a theatre play that stages a repeated encounter between scholars who are invested, or not, in that debate. At the centre of the conversations that take place are questions linked to academic responsibility, the nature of reflexivity, and the possibility of different political futures. The play and its characters are fictions, though they are hoped to reflect the 'narcissism of small differences' that often pervades academia. In doing so, we seek to (1) foreground how small changes in context (here, from an academic to a practitioner conference) force radical changes on how intellectual controversies are debated, (2) emphasize that the true stakes of debates such as those between critique and postcritique are often obscured by the combative nature of academic discourse, and (3) advocate for a refusal of, or active withdrawal from, the arbitrary classification of scholars into particular intellectual camps or positions. This process of fixing identities, the false idea that we are simply our academic personas, very rarely reflects either our personal-political realities nor the complex, polyphonic, and sometimes happily contradictory qualities of our evolving intellectual lives.
