In plants, Glycoside Hydrolase (GH) Family 1 β-glycosidases are believed to play important roles in many diverse processes including chemical defense against herbivory, lignification, hydrolysis of cell wall-derived oligosaccharides during germination, and control of active phytohormone levels. Completion of the Arabidopsis thalianagenome sequencing project has enabled us, for the first time, to determine the total number of Family 1 members in a higher plant. Reiterative database searches revealed a multigene family of 48 members that includes eight probable pseudogenes. Manual reannotation and analysis of the entire family were undertaken to rectify existing misannotations and identify phylogenetic relationships among family members. Forty-seven members (designated BGLU1 through BGLU47) share a common evolutionary origin and were subdivided into approximately 10 subfamilies based on phylogenetic analysis and consideration of intron–exon organizations. The forty-eighth member of this family (At3g06510; sfr2) is a β-glucosidase-like gene that belongs to a distinct lineage. Information pertaining to expression patterns and potential functions of Arabidopsis GH Family 1 members is presented. To determine the biological function of all family members, we intend to investigate the substrate specificity of each mature hydrolase after its heterologous expression in the Pichia pastoris expression system. To test the validity of this approach, the BGLU44-encoded hydrolase was expressed in P. pastoris and purified to homogeneity. When tested against a wide range of natural and synthetic substrates, this enzyme showed a preference for β-mannosides including 1,4-β-D-mannooligosaccharides, suggesting that it may be involved in A. thaliana in degradation of mannans, galactomannans, or glucogalactomannans. Supporting this notion, BGLU44 shared high sequence identity and similar gene organization with tomato endosperm β-mannosidase and barley seed β-glucosidase/β-mannosidase BGQ60.

An extracellular exo-beta-(1,3)-glucanase (designated EXG1) was purified to apparent homogeneity from Pichia pastoris X-33 cultures by ammonium sulfate fractionation, ion-exchange chromatography, and gel filtration. The native enzyme is unglycosylated and monomeric with a molecular mass of approximately 47kDa. At its optimal pH of 6.0, the enzyme shows highest activity among physiological substrates toward laminarin (apparent Km, 3.5 mg/ml; Vmax, 192 micromole glucose produced/min/mg protein) but also hydrolyzes amygdalin and esculin, and the chromogenic substrates p-nitrophenyl-beta-D-glucopyranoside and p-nitrophenyl-beta-D-xylopyranoside. The P. pastoris EXG1 gene was cloned by a PCR-based strategy using genomic DNA as template. This intronless gene predicts an ORF that encodes a primary translation product of 414 amino acids. We believe that this preproprotein is processed sequentially by signal peptidase and a Kex2-like endoprotease to yield a mature protein of 392 amino acids (45,376 Da; pI, 4.46) that shares 36-64% amino acid identity with other yeast exo-beta-(1,3)-glucanases belonging to Glycoside Hydrolase Family 5. It also possesses the eight invariant residues and signature pattern [LIV]-[LIVMFYWGA](2)-[DNEQG]-[LIVMGST]-X-N-E-[PV]-[RHDNSTLIVFY] shown by all Family 5 members. Overexpression of the cloned EXG1 gene in Pichia cells, followed by Ni-CAM HC resin chromatography, yielded milligram quantities of homogeneous recombinant EXG1 in active form for further characterization studies.

The flavoprotein (R)-(+)-mandelonitrile lyase (MDL; EC 4.1.2.10), which plays a key role in cyanogenesis in rosaceous stone fruits, occurs in black cherry (Prunus serotina Ehrh.) homogenates as several closely related isoforms. Biochemical and molecular biological methods were used to investigate MDL microheterogeneity and function in this species. Three novel MDL cDNAs of high sequence identity (designated MDL2, MDL4, and MDL5) were isolated. Like MDL1 and MDL3 cDNAs (Z. Hu, J.E. Poulton [1997] Plant Physiol 115: 1359–1369), they had open reading frames that predicted a flavin adenine dinucleotide-binding site, multipleN-glycosylation sites, and an N-terminal signal sequence. The N terminus of an MDL isoform purified from seedlings matched the derived amino acid sequence of the MDL4 cDNA. Genomic sequences corresponding to the MDL1, MDL2, and MDL4 cDNAs were obtained by polymerase chain reaction amplification of genomic DNA. Like the previously reported mdl3 gene, these genes are interrupted at identical positions by three short, conserved introns. Given their overall similarity, we conclude that the genesmdl1, mdl2, mdl3,mdl4, and mdl5 are derived from a common ancestral gene and constitute members of a gene family. Genomic Southern-blot analysis showed that this family has approximately eight members. Northern-blot analysis using gene-specific probes revealed differential expression of the genes mdl1,mdl2, mdl3, mdl4, andmdl5.

The flavoprotein (R)-(+)-mandelonitrile lyase (MDL; EC 4.1.2.10) plays a key role in cyanogenesis in rosaceous stone fruits. An MDL gene (mdl3) and its corresponding cDNA (MDL3) were isolated from black cherry (Prunus serotina) and characterized. The mdl3 gene contains 2292 bp of the 5[prime] flanking region, the entire coding region, and 300 bp of the 3[prime] flanking region. The coding region is interrupted by three short introns, of which one possesses the unusual GC-AG splice junction dinucleotides. This gene encodes a polypeptide of 573 amino acids that includes a putative signal sequence, 13 potential N-glycosylation sites, and a presumptive flavin adenine dinucleotide-binding site. To determine whether the 5[prime] flanking region of the mdl3 gene is capable of driving MDL expression, it was fused to the β-glucuronidase reporter gene for Agrobacterium-mediated transformation into tobacco. Matching endogenous MDL expression patterns, β-glucuronidase staining was observed in maturing embryos and seeds; it also occurred in postembryonic tissues, especially in association with vascular tissues. After developing a homologous transient transformation system to facilitate identification of putative regulatory sequences, we demonstrated that 125 bp (-107 to +18) of the 5[prime] flanking sequence of the mdl3 gene is sufficient for MDL expression in protoplasts derived from immature black cherry embryos.

In black cherry (Prunus serotina Ehrh.) macerates, the cyanogenic diglucoside (R)-amygdalin undergoes stepwise degradation to HCN catalyzed by amygdalin hydrolase (AH), prunasin hydrolase, and (R)-(+)-mandelonitrile lyase (MDL). A near full-length AH cDNA clone (pAH1), whose insert encodes the isozyme AH I, has been isolated and sequenced. AH I exhibits several features characteristic of [beta]-glucosidases of the BGA family, including their likely nucleophile center (isoleucine-threonine-glutamic acid-asparagine-glycine) and acid catalyst (asparagine-glutamic acid-proline/isoleucine) motifs. The temporal expression of AH and MDL in ripening fruit was analyzed by northern blotting. Neither mRNA was detectable until approximately 40 days after flowering (DAF), when embryos first became visible to the naked eye. Both mRNAs peaked at approximately 49 DAF before declining to negligible levels when the fruit matured (82 DAF). Taken together with enzyme activity data, these time courses suggest that AH and MDL expression may be under transcriptional control during fruit maturation. In situ hybridization analysis indicated that AH transcripts are restricted to the procambium, whereas MDL transcripts are localized within cotyledonary parenchyma cells. These tissue-specific distributions are consistent with the major locations of AH and MDL protein in mature seeds previously determined by immunocytochemistry (E. Swain, C.P. Li, and J.E. Poulton [1992] Plant Physiol 100:291–300).

In macerates of black cherry (Prunus serotina Ehrh.) leaves and stems, (R)-prunasin is catabolized to HCN, benzaldehyde, and D-glucose by the sequential action of prunasin hydrolase (EC 3.2.1.21) and (R)-(+)-mandelonitrile lyase (EC 4.1.2.10). Immuno-cytochemical techniques have shown that within these organs prunasin hydrolase occurs within the vacuoles of phloem parenchyma cells. In arborescent leaves, mandelonitrile lyase was also located in phloem parenchyma vacuoles, but comparison of serial sections revealed that these two degradative enzymes are usually localized within different cells.

UDPG:thiohydroximate glucosyltransferase (EC 2.4.1.-), which catalyses the penultimate reaction in glucosinolate biosynthesis, was purified 1100-fold in 37% yield from Arabidopsis thaliana inflorescences. The enzyme possessed a native M r of 57 800 and a pI of 4.5. At its optimum pH of 6.0, it showed a K m for UDPG of 0.27 mM. Enzyme activity was stimulated 20–45% by thiol reducing agents (e.g. 2-mercaptoethanol, l -cysteine), CaCl 2 , MgCl 2 and MnCl 2 , but was greatly inhibited by ZnCl 2 , and CuCl 2 . With the exception of 1,10-phenanthroline (10 mM) which caused ca 50% inhibition, metal chelators had little effect upon glucosyltransferase activity. The sensitivity of the enzyme to several thiol blocking reagents suggests that sulphydryl groups are essential for GT activity. The purified Arabidopsis enzyme preparation lacked desulphoglucosinolate sulphotransferase activity.

Plum (Prunus domestica) seeds, which contain the cyanogenic diglucoside (R)-amygdalin and lesser amounts of the corresponding monoglucoside (R)-prunasin, release the respiratory toxin HCN upon tissue disruption. Amygdalin hydrolase (AH) and prunasin hydrolase (PH), two specific [beta]-glucosidases responsible for hydrolysis of these glucosides, were purified to near homogeneity by concanavalin A-Sepharose 4B and carboxymethyl-cellulose chromatography. Both proteins appear as polypeptides with molecular masses of 60 kD upon sodium dodecyl sulfate-polyacrylamide gel electrophoresis, but they exhibit different isoelectric points (PH, 5.6-6.0; AH, 7.8-8.2). AH and PH were localized within mature plum seeds by tissue printing, histochemistry, and silver-enhanced immunogold labeling. As was previously shown in black cherry (Prunus serotina) seeds (E.Swain, C.P. Li, J.E. Poulton [1992] Plant Physiol 100: 291-300), AH and PH are restricted to protein bodies of specific procambial cells and are absent from the cotyledonary parenchyma, bundle sheath, and endosperm cells. In contrast, the cyanogenic glycosides in both plum and black cherry seeds, which were detected by tissue printing, occur solely in the cotyledonary parenchyma and are absent from the procambium and endosperm. It is concluded that tissue level compartmentation prevents large-scale cyanoglycoside hydrolysis in intact Prunus seeds.

In black cherry (Prunus serotina Ehrh.) homogenates, (R)-amygdalin is catabolized to HCN, benzaldehyde, and d-glucose by the sequential action of amygdalin hydrolase, prunasin hydrolase, and mandelonitrile lyase. The tissue and subcellular localizations of these enzymes were determined within intact black cherry seeds by direct enzyme analysis, immunoblotting, and colloidal gold immunocytochemical techniques. Taken together, these procedures showed that the two beta-glucosidases are restricted to protein bodies of the procambium, which ramifies throughout the cotyledons. Although amygdalin hydrolase occurred within the majority of procambial cells, prunasin hydrolase was confined to the peripheral layers of this meristematic tissue. Highest levels of mandelonitrile lyase were observed in the protein bodies of the cotyledonary parenchyma cells, with lesser amounts in the procambial cell protein bodies. The residual endosperm tissue had insignificant levels of amygdalin hydrolase, prunasin hydrolase, and mandelonitrile lyase.

Biochemical changes related to cyanogenesis (hydrogen cyanide production) were monitored during maturation of black cherry (Prunus serotina Ehrh.) fruits. At weekly intervals from flowering until maturity, fruits (or selected parts thereof) were analyzed for (a) fresh and dry weights, (b) prunasin and amygdalin levels, and (c) levels of the catabolic enzymes amygdalin hydrolase, prunasin hydrolase, and mandelonitrile lyase. During phase I (0-28 days after flowering [DAF]), immature fruits accumulated prunasin (mean: 3 micromoles/fruit) but were acyanogenic because they lacked the above enzymes. Concomitant with cotyledon development during mid-phase II, the seeds began accumulating both amygdalin (mean: 3 micromoles/seed) and the catabolic enzymes and were highly cyanogenic upon tissue disruption. Meanwhile, prunasin levels rapidly declined and were negligible by maturity. During phases II (29-65 DAF) and III (66-81 DAF), the pericarp also accumulated amygdalin, whereas its prunasin content declined toward maturity. Lacking the catabolic enzymes, the pericarp remained acyanogenic throughout all developmental stages.

Mandelonitrile lyase (MDL, EC 4.1.2.10), which catalyzes the reversible dissociation of (R)-(+)-mandelonitrile to benzaldehyde and hydrogen cyanide, was purified to apparent homogeneity from mature black cherry (Prunus serotina Ehrh.) seeds by conventional protein purification techniques. This flavoprotein is monomeric with a subunit molecular mass of 57 kilodaltons. Glycoprotein character was shown by its binding to the affinity matrix concanavalin A-Sepharose 4B with subsequent elution by alpha-methyl-d-glucoside. Upon chemical deglycosylation by trifluoromethanesulfonic acid, the molecular mass was reduced to 50.9 kilodaltons. Two-dimensional gel analysis of deglycosylated MDL revealed the presence of several subunit isoforms of similar molecular mass but differing slightly in isoelectric point. Polyclonal antibodies were raised in New Zealand white rabbits against deglycosylated and untreated MDL. Antibody titers were determined by enzyme linked immunosorbent and dot immunobinding assays, while their specificities were assessed by Western immunoblot analysis. Antibodies raised against untreated lyase recognized several proteins in addition to MDL. In contrast, antisera raised against deglycosylated MDL were monospecific and were utilized for developmental and immunocytochemical localization studies. SDS-PAGE and immunoblotting analysis of seed proteins during fruit maturation showed that MDL first appeared in seeds shortly after cotyledons began development. In cotyledon cells of mature seeds, MDL was localized primarily in the cell wall with lesser amounts in the protein bodies, whereas in endosperm cells, this labeling pattern was reversed. N-terminal sequence data was gathered for future molecular approaches to the question of MDL microheterogeneity.

A 3' -phosphoadenosine 5' -phosphosulfate (PAPS):desulfoglucosinolate sulfotransferase (EC 2.8.2-) was extensively purified from light-grown cress (Lepidium sativum L.) seedlings by gel filtration and concanavalin A-Sepharose 4B, Matrex Gel Green A, and Mono Q fast protein liquid chromatography. The purified enzyme, which required bovine serum albumin for stabilization, had a native molecular weight of 31,000 +/- 5,000 and an apparent isoelectric point of 5.2. Using PAPS (K(m) 60 micromolar) as sulfur donor, it catalyzed the sulfation of desulfobenzylglucosinolate (K(m) 82 micromolar), desulfo-p-hydroxybenzylglucosinolate (K(m) 670 micromolar), and desulfoallylglucosinolate (K(m) 6.5 millimolar) at an optimal pH of 9.0. All other potential substrates tested, including flavonoids, flavonoid glycosides, cinnamic acids, and phenylacetaldoxime, were not sulfated. Sulfotransferase activity was stimulated by MgCl(2), MnCl(2) and reducing agents and inhibited by ZnCl(2), PbNO(3) NiCl(2) and the reaction product PAP. The thiol reagents N-ethylmaleimide, p-chloromercuriphenylsulfonic acid, and 5,5' -dithio-bis-(2-nitrobenzoic acid) were also potent inhibitors, but the enzyme was protected from covalent modification by beta-mercaptoethanol. The kinetics of desulfobenzylglucosinolate sulfation were consistent with a rapid equilibrium ordered mechanism with desulfobenzylglucosinolate binding first and PAPS second.

Several thousand plant species, including many economically important food plants, synthesize cyanogenic glycosides and cyanolipids. Upon tissue disruption, these natural products are hydrolyzed liberating the respiratory poison hydrogen cyanide. This phenomenon of cyanogenesis accounts for numerous cases of acute and chronic cyanide poisoning of animals including man. This article reviews information gathered during the past decade about the enzymology and molecular biology of cyanogenesis in higher plants. How compartmentation normally prevents the large-scale, suicidal release of HCN within the intact plant is discussed. A renewed interest in the physiology of these cyanogenic compounds has revealed that, in addition to providing protection for some species against herbivory, they may also serve as storage forms for reduced nitrogen.

To establish the substrate specificity of the thioglucoside glucohydrolase myrosinase (EC 3.2.3.1), this enzyme was purified to homogeneity from light-grown cress (Lepidium sativum L.) seedlings by Sephadex gel filtration. Red Dye and anion exchange (FPLC Mono Q) chro­matography, and preparative isoelectric focusing. Hydrolytic activity was shown toward only 4 of the 29 synthetic and natural O-and S-glycosides tested. Highest activity was displayed with the endogenous glucosinolates benzylglucosinolate (Km, 295 μM) and sinigrin (Km, 300 μM) at an optimum pH of 5.5 in sodium citrate buffer. The synthetic glycosides PNPG (Km, 2.0 mM) and ONPG were poorer substrates at an optimum pH of 6.5 in potassium phos­phate buffer. The enzyme was inactive with all other nitrophenyl glycosides tested including PNP-a-D-glucoside and PNP-thio-β-D-glucoside, suggesting a requirement for O-β-D-glucose as the glycone moiety within these substrates. PNPG hydrolysis was stimulated 2.6-fold by ascorbate (1 mM). The enzyme exhibited no metal ion requirement and was strongly inhibited by lead nitrate, mercury chloride, and ferric chloride at 1 mM concentration. The metal chela­tors DIECA , EDTA , o-phenanthroline, and 2,2′-dipyridyl were not inhibitory, but the thiol reagents PCMS, PCMB, and N-ethylmaleimide (at 1mM) caused 50 -80% inhibition of enzyme activity.

The catabolism of cyanogenic glycosides is initiated by cleavage of the carbohydrate moiety by one or more beta-glycosidases, which yields the corresponding alpha-hydroxynitrile. Until recently, the mode by which cyanogenic disaccharides are hydrolysed was largely unclear. Investigation of highly purified beta-glycosidases from plants containing cyanogenic disaccharides has now indicated that these compounds may be degraded via two distinct pathways, depending on the plant species. beta-Glycosidases from Davallia trichomanoides and Vicia angustifolia hydrolysed (R)-vicianin and (R)-amygdalin at the aglycone-disaccharide bond producing mandelonitrile and the corresponding disaccharide. Alternatively, hydrolysis of cyanogenic disaccharides in Prunus serotina, almonds, and Linum usitatissimum involves stepwise removal of the sugar residues. The nature of these enzymes and of other beta-glycosidases responsible for hydrolysis of simple cyanogenic monosaccharides is discussed. Hydroxynitriles may decompose either spontaneously or enzymically in the presence of a hydroxynitrile lyase to produce hydrogen cyanide and an aldehyde or ketone. The major kinetic and molecular properties of hydroxynitrile lyases purified from species accumulating aromatic and aliphatic cyanogens are reviewed. Cyanogenesis occurs rapidly only after cyanogenic plant tissues are macerated, allowing glycosides access to their catabolic enzymes. The possible nature of the compartmentation which prevents cyanogenesis under normal physiological conditions is discussed in relation to our knowledge of the tissue and subcellular localizations of cyanogens and catabolic enzymes.

In black cherry (Prunus serotina Ehrh.) seed homogenates, (R)-amygdalin is degraded to HCN, benzaldehyde, and glucose by the sequential action of amygdalin hydrolase (AH), prunasin hydrolase (PH), and mandelonitrile lyase. Leaves are also highly cyanogenic because they possess (R)-prunasin, PH, and mandelonitrile lyase. Taking both enzymological and molecular approaches, we demonstrate here that black cherry PH is encoded by a putative multigene family of at least five members. Their respective cDNAs (designated Ph1,Ph2, Ph3, Ph4, andPh5) predict isoforms that share 49% to 92% amino acid identity with members of glycoside hydrolase family 1, including their catalytic asparagine-glutamate-proline and isoleucine-threonine-glutamate-asparagine-glycine motifs. Furthermore, consistent with the vacuolar/protein body location and glycoprotein character of these hydrolases, their open reading frames predict N-terminal signal sequences and multiple potential N-glycosylation sites. Genomic sequences corresponding to the open reading frames of these PHs and of the previously isolated AH1 isoform are interrupted at identical positions by 12 introns. Earlier studies established that native AH and PH display strict specificities toward their respective glucosidic substrates. Such behavior was also shown by recombinant AH1, PH2, and PH4 proteins after expression in Pichia pastoris. Three amino acid moieties that may play a role in conferring such aglycone specificities were predicted by structural modeling and comparative sequence analysis and tested by introducing single and multiple mutations into isoform AH1 by site-directed mutagenesis. The double mutant AH ID (Y200I and G394D) hydrolyzed prunasin at approximately 150% of the rate of amygdalin hydrolysis, whereas the other mutations failed to engender PH activity.

Cotyledons of mature black cherry (Prunus serotina Ehrh.) seeds contain the cyanogenic diglucoside (R)-amygdalin. The levels of amygdalin, its corresponding monoglucoside (R)-prunasin, and the enzymes that metabolize these cyanoglycosides were measured during the course of seedling development. During the first 3 weeks following imbibition, cotyledonary amygdalin levels declined by more than 80%, but free hydrogen cyanide was not released to the atmosphere. Concomitantly, prunasin, which was not present in mature, ungerminated seeds, accumulated in the seedling epicotyls, hypocotyls, and cotyledons to levels approaching 4 [mu]mol per seedling. Whether this prunasin resulted from amygdalin hydrolysis remains unclear, however, because these organs also possess UDPG:mandelonitrile glucosyltransferase, which catalyzes de novo prunasin biosynthesis. The reduction in amygdalin levels was paralleled by declines in the levels of amygdalin hydrolase (AH), prunasin hydrolase (PH), mandelonitrile lyase (MDL), and [beta]-cyanoalanine synthase. At all stages of seedling development, AH and PH were localized by immunocytochemistry within the vascular tissues. In contrast, MDL occurred mostly in the cotyledonary parenchyma cells but was also present in the vascular tissues. Soon after imbibition, AH, PH, and MDL were found within protein bodies but were later detected in vacuoles derived from these organelles.

In black cherry (Prunus serotina Ehrh.) seed homogenates, amygdalin hydrolase (AH) participates with prunasin hydrolase (PH) and mandelonitrile lyase in the sequential degradation of (R)-amygdalin to HCN, benzaldehyde, and glucose. Four isozymes of AH (designated AH I, I', II, II') were purified from mature cherry seeds by concanavalin A-Sepharose 4B chromatography, ion-exchange chromatography, and chromatofocusing. All isozymes were monomeric glycoproteins with native molecular masses of 52 kD. They showed similar kinetic properties (pH optima, K(m), V(max)) but differed in their isoelectric points and N-terminal amino acid sequences. Analytical isoelectric focusing revealed the presence of subisozymes of each isozyme. The relative abundance of these isozymes and/or subisozymes varied from seed to seed. Three isozymes of PH (designated PH I, IIa, and IIb) were purified to apparent homogeneity by affinity, ion-exchange, and hydroxyapatite chromatography and by nondenaturing polyacrylamide gel electrophoresis. PH I and PH IIb are 68-kD monomeric glycoproteins, whereas PH IIa is dimeric (140 kD). The N-terminal sequences of all PH and AH isozymes showed considerable similarity. Polyclonal antisera raised in rabbits against deglycosylated AH I or a mixture of the three deglycosylated PH isozymes were not monospecific as judged by immunoblotting analysis, but also cross-reacted with the opposing glucosidase. Monospecific antisera deemed suitable for immunocytochemistry and screening of expression libraries were obtained by affinity chromatography. Each antiserum recognized all known isozymes of the specific glucosidase used as antigen.

The cyanogenic glycoside of young fronds and fiddleheads of the fern Davallia trichomanoides Blume was identified as (R)-vicianin (the β-vicianoside of (R)-mandelonitrile) by acid and enzymic hydrolysis, 1H-NMR and 13C-NMR spectroscopy, and by comparison with an authentic sample isolated from Vicia angustifolia seeds.

Adenosine nucleosidase (EC 3.2.2.7), which catalyses the irreversible hydrolysis of adenosine to adenine and ribose, has been isolated and purified about 40-fold from leaves of spinach beet (Beta vulgaris L.). The enzyme appeared to be specific for adenosine only among the naturally-occurring nucleosides, but comparable activity was also found with adenosine N-oxide. Adenosine hydrolysis, which had an optimum at pH 4.5, did not require phosphate ions nor was it stimulated by their presence. The Michaelis constant for this substrate was 11 μM. Whereas the rate of adenosine hydrolysis was unaffected by DL-homocysteine, L-methionine and ribose, it was sensitive to the presence of adenine, S-adenosyl-L-methionine, S-adenosyl-L-homocysteine, AMP and deoxyadenosine. The role of this enzyme in plant metabolism is discussed.

Immature fruits and leaves of black cherry (Prunus serotina Ehrh.) accumulate the cyanogenic glucoside prunasin (the β-glucoside of (R)-mandelonitrile). Cell-free extracts from these tissues catalysed the stereospecific glucosylation of (R,S)-mandelonitrile to (β)-prunasin at rates of 0.2-2.0 μmol/h/mg protein. Uridine diphosphate glucose (Km = 0.32 mᴍ) acted as glucose donor. The optimum concentration of (R,S)-mandelonitrile was 20 mᴍ. Highest activity was exhibited at pH 7.0-8.0 in Tris-phosphate buffer, and no additional cofactors were required. β- Mercaptoethanol (14.5 mᴍ), provided in the homogenization medium to prevent browning of homogenates, did not stimulate the rate of prunasin production. In addition to (R)-mandelonitrile (100%), significant activity was shown towards mandelamide (21%), benzyl alcohol (15%), mandelic acid (8%) and benzoic acid (153%), but not towards prunasin. Mandelonitrile glucosyltransferase activity was most stable at - 20 °C in the presence of 10% glycerol.

A 3- O -methyltransferase which catalyzes the methylation of caffeic acid to ferulic acid using S -adenosyl- l -methionine as methyl donor has been isolated and purified about 60-fold from cell suspension cultures of soybean ( Glycine max L., var. Mandarin). The enzyme utilized, in addition to caffeic acid ( K m = 133 μM), 5-hydroxyferulic acid ( K m = 55 μM), 3,4,5-trihydroxy-cinnamic acid ( K m = 100 μM), and protocatechualdehyde ( K m = 50 μM) as substrates. Methylation proceeded only in the meta position. The enzyme was unable to catalyze the methylation of ferulic acid, of ortho- , meta- , and para -coumaric acids, and of the flavonoid compounds quercetin and luteolin. The methylation of caffeic acid and 5-hydroxyferulic acid showed a pH optimum at 6.5–7.0. No stimulation of the reaction velocity was observed when Mg 2+ ions were added. EDTA did not inhibit the reaction. The K m for S -adencsyl- l -methionine was 15 μ m . S -Adenosyl- l -homocysteine was a potent competitive inhibitor of S -adenosyl- l -methionine ( K i = 6.9 μM).

An enzyme, catalysing the glucosylation of cyanidin at the 3-position using uridine diphosphate- D -glucose (UDPG) as glucosyl-donor, has been isolated and purified about 50-fold from young red cabbage ( Brassica oleracea ) seedlings. The pH optimum for this reaction was ca 8 and no additional cofactors were required. The reaction was inhibited by cyanidin (above 0.25 mM) and by very low concentrations of the reaction product cyanidin-3-glucoside (5 μM). The K m values for UDPG and cyanidin were 0.51 and 0.4 mM respectively. In addition to cyanidin the enzyme could also glucosylate the following compounds at the 3-position: pelargonidin, peonidin, malvidin, kaempferol, quercetin, isorhamnetin, myricetin and fisetin. In contrast, cyanidin-3-glucoside, cyanidin-3-sophoroside, cyanidin-3,5-diglucoside, apigenin, luteolin, naringenin and dihydroquercetin were not glucosylated.

Five multiple forms (forms 1–5) of mandelonitrile lyase (EC 4.1.2.10) which catalyze the decomposition of mandelonitrile to benzaldehyde and hydrogen cyanide have been extensively purified from seeds of black cherry ( Prunus serotina Ehrh.) by concanavalin A-Sepharose 4B chromatography and chromatofocusing. These forms are monomers which differ only slightly in molecular weight (57,000–59,000) and isoelectric point (4.58–4.63), but heterogeneity in their carbohydrate side-chains was suggested by concanavalin A-Sepharose 4B chromatography. The absorption spectra of the predominating forms 4 and 5 showed maxima of 278, 380, and 460 nm, indicative of flavoprotein character. Detailed comparative kinetic studies of forms 4 and 5 revealed few significant differences in behavior. Both proteins showed pH optima between 6.0 and 7.0, had identical K m values (0.17 m m ) for ( R,S )-mandelonitrile, and retained similar activities upon storage at 4 and −20 °C. Neither form exhibited a metal ion requirement and both were affected similarly by metal salts, β-mercaptoethanol, and sulfhydryl reagents. Benzole acid, p -hydroxybenzyl alcohol, and benzyl alcohol inhibited both forms.

A methyltransferase, which catalyzes the methylation of luteolin (Km, 16 μM) using S-adenosyl-l-methionine as the methyl donor, has been purified about 38-fold from cell suspension cultures of soybean (Glycine max L., var. Mandarin). The following 3,4-dihydroxy phenolic compounds were also methylated: luteolin 7-O-glucoside (Km, 28 μm), quercetin (Km, 35 μm), eriodictyol (Km, 75 μm), 5-hydroxyferulic acid (Km, 227 μm), dihydroquercetin (Km, 435 μm), and caffeic acid (Km, 770 μm). Rutin and quercetin 3-O-glucoside were poor substrates. Methylation proceeded only in the meta position. The enzyme was unable to catalyze the methylation of p-coumaric acid, m-coumaric acid, ferulic acid, isoferulic acid, sinapic acid, apigenin, or naringenin. While the isoflavones biochanin A and daidzein did not serve as substrates, texasin (6,7-dihydroxy-3′-methoxyisoflavone) was methylated (Km, 35 μm). The methylation of caffeic acid and quercetin showed a pH optimum of 8.6–8.9. The enzyme required Mg2+ ions for maximum activity (approximately 1 mm) and could be totally inhibited by EDTA (10 mm). The Km for S-adenosyl-l-methionine was 11 μm. S-Adenosyl-l-homocysteine inhibited the methylation of luteolin by S-adenosyl-l-methionine.

The tissue distributions of dhurrin [p-hydroxy-(S)-mandelonitrile-β-d-glucoside] and of enzymes involved in its metabolism have been investigated in leaf blades of light-grown Sorghum bicolor seedlings. Enzymic digestion of these leaves using cellulase has enabled preparations of epidermal and mesophyll protoplasts and bundle sheath strands to be isolated with only minor cross-contamination. Dhurrin was located entirely in the epidermal layers of the leaf blade, whereas the two enzymes responsible for its catabolism, namely dhurrin β-glucosidase and hydroxynitrile lyase, resided almost exclusively in the mesophyll tissue. The final enzyme of dhurrin biosynthesis, uridine diphosphate glucose:p-hydroxymandelonitrile glucosyltransferase, was found in both mesophyll (32% of the total activity of the leaf blade) and epidermal (68%) tissues. The bundle sheath strands did not contain significant amounts of dhurrin or of these enzymes. It was concluded that the separation of dhurrin and its catabolic enzymes in different tissues prevents its large scale hydrolysis under normal physiological conditions. The well documented production of HCN (cyanogenesis), which occurs rapidly on crushing Sorghum leaves, would be expected to proceed when the contents of the ruptured epidermal and mesophyll cells are allowed to mix.

Three forms of prunasin hydrolase (PH I, PH IIa, and PH IIb), which catalyze the hydrolysis of (R)-prunasin to mandelonitrile and d -glucose, have been purified from homogenates of mature black cherry (Prunus serotina Ehrh.) seeds. Hydroxyapatite chromatography completely resolved PH I from PH IIa and PH IIb. PH IIa and IIb, which coeluted on hydroxyapatite, were resolved by gel filtration. PH IIa was a dimer with a native molecular weight of 140,000. Both PH I and PH IIb were monomeric with molecular weights of 68,000. The isozymes appeared to be glycoproteins based on their binding to concanavalin A-Sepharose 4B with subsequent elution by α-methyl- d -glucoside. When presented several potential glycosidic substrates, these enzymes exhibited a narrow specificity towards (R)-prunasin. Km values for (R)-prunasin for PH I, PH IIa, and PH IIb were 1.73, 2.3, and 1.35 m m , respectively. PH I and PH IIb possessed fivefold greater V max K m values than PH IIa. Ortho- and para-nitrophenyl-β-D-glucosides were hydrolyzed at the same active site. All forms had a pH optimum of 5.0 in citratephosphate buffer. PH I and PH IIb were competitively inhibited by castanospermine with Ki values of 0.19 and 0.09 m m , respectively. PH activity was not stimulated by any metal ion tested and was unaffected by diethyldithiocarbamate, o-phenanthroline, 2,2′-dipyridyl, and EDTA.

A rapid, sensitive, and inexpensive assay for prunasin hydrolase activity is described. Prunasin hydrolase is incubated with saturating concentrations of prunasin in the presence of excess purified mandelonitrile lyase. Under these conditions, the mandelonitrile formed by prunasin hydrolysis is immediately converted to benzaldehyde, which may be determined spectrophotometrically at 249.6 nm. The rate of prunasin hydrolysis is determined from the rate of change of absorbance at this wavelength. A single ion-exchange chromatographic procedure is also described which allows the facile purification of mandelonitrile lyase to near homogeneity with greater than 70% recovery.

Crude extracts from young Robinia pseudoacacia seedlings, shoots, and callus tissue catalyze the para-O-methylation of apigenin to acacetin using S-adenosyl-ʟ-methionine as methyl donor. Optimum activity was exhibited at pH 9.0, and Mg2+ was not required for maximum activity. EDTA (10 mᴍ) did not affect the reaction rate, but 47% inhibition was observed with SAH (100 μᴍ). β-Mercaptoethanol (5 mᴍ) was required in the homogenization medium for optimum O-methyltransferase activity. Apigenin (Km, 50 μᴍ) was the best substrate, but significant activity was shown towards caffeic acid, 5-hydroxyferulic acid, naringenin, and quercetin. Paracoumaric, ferulic, and sinapic acids were not methylated. The Km for S-adenosyl-ʟ-methionine was 31 μᴍ. Our demonstration of a para-O-methyltransferase activity methylating apigenin, but not para-coumaric acid, strongly supports the conclusion that the B-ring methylation pattern of acacetin is determined at the C15-level in Robinia pseudoacacia.

Mature black cherry (Prunus serotina Ehrh.) seeds accum ulate high levels of the cyanogenic disaccharide (R)-amygdalin. Extracts from these seeds contain two β-glycosidases which have been identified and completely resolved by DEAE-cellulose ion-exchange chromatography. Amygdalin hydrolase hydrolyzed (R)-am ygdalin at an optimum pH of 5.5, releasing (R)-prunasin and D-glucose. This enzyme showed highest activity towards (R)-am ygdalin and failed to hydrolyze (R)-prunasin. linamarin, β-gentiobiose and cellobiose. A distinct β-glycosidase, prunasin hydrolase, displayed a pronounced preference for (R)-prunasin, hydrolyzing this cyanogenic monosaccharide at an optimum pH of 6.5 to mandelonitrile and D-glucose. Prunasin hydrolase was inactive towards (R)-am ygdalin, linamarin, and β-gentiobiose. Both enzymes showed significant activity towards the artificial substrates β-ONPGlu and β-PNPGlu but did not hydrolyze α-PNPGlu. In view of the pronounced specificity of these enzymes towards endogenous cyanogens, it is concluded that upon disruption of black cherry seeds (R)- amygdalin is catabolized to mandelonitrile in a stepwise manner (the sequential mechanism) by amygdalin hydrolase and prunasin hydrolase with (R)-prunasin serving as intermediate. Young fronds of Davallia trichomanoides are rich sources of (R)-vicianin (the β-vicianoside of (R)-mandelonitrile). A β-glycosidase, vicianin hydrolase, has been partially purified from frond extracts by ion-exchange chromatography. At the optimum pH of 6.0, this enzyme showed highest hydrolytic activity with (R)-vicianin, although both (R)-am ygdalin and (R)-prunasin could be hydrolyzed at approximately 15% of the rate observed with (R)-vicianin. It failed to hydrolyze β-gentiobiose, cellobiose, linamarin and α-PNPGlu. Closer exam ination revealed that (R)-vicianin and (R)-amygdalin were hydrolyzed at the aglycone-disaccharide bond (the simultaneous mechanism) yielding mandelonitrile and the respective disaccharides vicianose and β-gentiobiose

An enzyme catalysing the methylation of caffeic acid to ferulic acid, using S-adenosyl-l-methionine as methyl donor, has been extracted from leaves of spinach beet and purified 75-fold to obtain a stable prepation. 2. 2. The enzyme showed optimum activity at pH 6.5, and did not require the addition of Mg2+ for maximum activity. 3. 3. It was most active with caffeic acid, but showed some activity with catechol, protocatechuic acid and 3,4-dihydroxybenzaldehyde. The Km for caffeic acid was 68 μM. 4. 4. The Km for S-Adenosyl-l-rmmethionine was 12.5 μM. S-Adenosyl-l-homocysteine (Ki = 4.4 μM) was a competitive inhibitor of S-adenosyl-l-methionine. 5. 5. The synthesis of S-adenosyl-l-homocysteine from adenosine and l-homocysteine and its consequent effect on caffeic acid methylation were demonstrated with a partially-purified preparation from spinach-beet leaves, which possessed both S-adenosyl-l-homocysteine hydrolase (EC 3.3.1.1) and adenosine nucleosidase (EC 3.2.2.7) activities. This preparation was also able to catalyse the rapid breakdown of S-adenosyl-l-homocysteine to adenosine and adenine; the possible significance of this reaction in relieving the inhibition of caffeic acid methylation by S-adenosyl-l-homocysteine is discussed.

An earlier report that various sugars, especially d -mannitol and d -sorbitol, severely inhibited phenylalanine ammonia-lyase (PAL) 1 activity from tobacco in vitro has been reappraised. This apparent inhibition resulted from complexing of the sugars with borate, used as buffer system in that study, leading to an undetected pH decrease. We find that d -mannitol, d -sorbitol, d -glucose and sucrose are not inhibitory to PAL preparations obtained from sweet-clover leaves or from protoplasts of tobacco and barley, when alternative buffers are used for the assay. Moreover, the inhibition of barley PAL due to borate—sugar complex formation was completely eliminated by readjusting the pH of the reaction mixture to 8.8 before assaying.

After removal of myrosinase activity by concanavalin A-Sepharose 4B chromatography, cell-free extracts of light-grown cress (Lepidium sativum L.) seedlings, catalyzed the sulfation of desulfobenzylglucosinolate (K(m), 0.23 millimolar) to benzylglucosinolate using PAPS (K(m), 1 millimolar) as sulfur donor. Sulfotransferase activity, which was optimal at pH 9.0, was stimulated by MgCl(2), MnCl(2), beta-mercaptoethanol, and dithiothreitol and was inhibited by ZnSO(4) and SH-reagents. The enzyme also sulfated desulfoallyglucosinolate to allylglucosinolate (sinigrin) but was inactive towards all phenylpropanoids and flavonoids tested.

Two S-adenosyl- l -methionine:3,4-dihydric phenol 3-O-methyltransferases were present in crude extracts from soybean (Glycine max L., var. Mandarin) cell suspension cultures. The enzymes could be clearly distinguished by differences in (a) stability upon storage, (b) elution pattern upon gel chromatography, (c) substrate specificity, and (d) changes in specific activity during the growth cycle of a cell culture. The results from studies of the latter two criteria suggest that one of the methyltransferases, which is specific for substituted cinnamic acids, is related to the formation of lignin, whereas the other is involved in the flavonoid pathway.

The localization of phenylalanine ammonia-lyase [EC 4.3.1.5] within sweet clover (Melilotus alba) leaves was investigated. Apical buds and axillary leaves contained 15 to 30 times more enzyme activity than did mature leaves. Mesophyll protoplasts were prepared by digesting young leaves with Cellulysin and Macerase and were gently ruptured yielding intact chloroplasts. These chloroplast preparations exhibited neither phenylalanine ammonia-lyase nor o-coumaric acid O-glucosyltransferase activities. The general enzymic properties of sweet clover leaf phenylalanine ammonia-lyase were similar to those described for this enzyme isolated from other plant species. The conversion of l-phenylalanine to trans-cinnamic acid, which occurred at an optimum pH of about 8.7, was strongly inhibited by the metabolites trans-cinnamic and o-coumaric acids. In contrast, o-coumaric acid glucoside, coumarin, p-coumaric acid, and melilotic acid had no significant effect on the reaction rate.

Vicianin hydrolase, which catalyzes the hydrolysis of vicianin (K(m), 4.9 millimolar) to (R)-mandelonitrile and vicianose at an optimum pH of 5.5, was extensively purified from the young fronds and fiddleheads of the squirrel's foot fern (Davallia trichomanoides Blume) using DEAE-cellulose and Ultrogel HA chromatography. The native molecular weight of the enzyme was 340,000, and the isoelectric point was 4.6 to 4.7. SDS-PAGE analysis yielded three polypeptides with molecular weights of 56,000, 49,000, and 32,500. The enzyme hydrolyzed only a narrow range of glycosides and, among cyanogenic glycosides, exhibited a strict requirement for (R)-epimers and a preference for disaccharides over monosaccharides. (R)-Amygdalin, (R)-prunasin and p-nitrophenyl-beta-d-glucoside were hydrolyzed at 27, 14, and 3%, respectively, of the rate of vicianin hydrolysis. Mixed substrate studies showed that (R)-vicianin, (R)-prunasin, and p-nitrophenyl-beta-d-glucoside competed for the same active site. The enzyme was significantly inhibited by castanospermine, delta-gluconolactone, and p-chloromercuriphenylsulfonate. Failure to recognize concanavalin A-Sepharose 4B and to stain with periodic acid-Schiff reagent indicated that the enzyme was not a glycoprotein.

Myrosinase (beta-thioglucoside glucohydrolase, EC 3.2.3.1) was purified to apparent homogeneity from light-grown cress (Lepidium sativum L.) seedlings. This enzyme, which catalyzes hydrolysis of the glucosinolate sinigrin (K(m), 115 micromolar) at an optimum pH of 5.5 in sodium citrate buffer, had a native molecular weight of 130 +/- 5 kilodaltons and an isoelectric point of 4.7 to 4.9. SDS-PAGE revealed two polypeptides with molecular weights of 62 and 65 kilodaltons. Both subunits contained carbohydrate as shown by periodic acid-Schiff staining. The purified enzyme hydrolyzed p-nitrophenyl-beta-d-glucoside (K(m), 2.0 millimolar) at an optimum pH of 6.5 in phosphate buffer. The indolizidine alkaloid castanospermine, a known inhibitor of O-glycosidases, competitively inhibited the hydrolyses of sinigrin (thioglucosidase activity) and p-nitrophenyl-beta-d-glucoside (O-glucosidase activity) with K(i) values of 5 and 6 micromolar, respectively. In contrast, the related polyhydroxyalkaloids swainsonine and deoxynojirimycin were without effect upon these hydrolyses.

Cell-free extracts from very young primary leaves of parsley plants ( Petroselinum hortense Hoffm.) catalyse the oxidation of naringenin (5,7,4′-trihydroxyflavanone) to apigenin (5,7,4′-trihydroxyflavone). Enzyme activity is found in the 150,000 g supernatant. The reaction requires, in addition to oxygen and Fe 2+ ions, (an)other heat stable and dialysable nonproteinaceous cofactor(s). The reaction can be inhibited by 10 −4 m o -phenanthroline. It has a pH optimum of about 7.5. The enzyme preparation also catalyses the oxidation of 7,4-dihydroxyflavanone to the corresponding flavone. The isomeric chalcone (4,2′,4′-trihydroxychalcone) cannot serve as substrate. Only after cyclization to the flavanone with chalcone-flavanone isomerase is oxidation to flavone observed. The enzyme catalyzing this oxidation cannot be a peroxidase, because the reaction is not H 2 O 2 -dependent and is not inhibited by catalase, cyanide, or 1.4·10 −2 m mercaptoethanol.

Two forms of the beta-glucosidase amygdalin hydrolase (AH I and II), which catalyze the hydrolysis of (R)-amygdalin to (R)-prunasin and D-glucose, have been purified over 200-fold from mature black cherry (Prunus serotina Ehrh.) seeds. These proteins showed very similar molecular and kinetic properties but could be resolved by chromatofocusing and isoelectric focusing. AH I and II were monomeric (Mr 60,000) and had isoelectric points of 6.6 and 6.5, respectively. Their glycoprotein character was indicated by positive periodic acid-Schiff staining and by their binding to concanavalin A-Sepharose 4B with subsequent elution by alpha-Me-D-glucoside. Of the natural glycosidic substrates tested, both enzymes showed a pronounced preference for the endogenous cyanogenic disaccharide (R)-amygdalin. They also hydrolyzed at the same active site the synthetic substrates p-nitrophenyl-beta-D-glucoside and 4-methylumbelliferyl-beta-D-glucoside but were inactive towards (R)-prunasin, p-nitrophenyl-alpha-D-glucoside, and 4-methylumbelliferyl-alpha-D-glucoside. Maximum hydrolytic activity was shown in citrate-phosphate buffer in the pH range 4.5-5.0. AH I and II were inhibited competitively by the reaction product (R)-prunasin and noncompetitively (mixed type) by delta-gluconolactone and castanospermine.

S-Adenosyl- l -homocysteine hydrolase (EC 3.3.1.1) has been isolated from spinach-beet leaves and purified 100-fold. The enzyme catalyzes both the hydrolysis of S-adenosyl- l -homocysteine to adenosine and l -homocysteine and its synthesis from these compounds. The equilibrium constant for the reaction is 1.8 × 10 −6 in relation to hydrolysis. The enzyme shows optimum activity at pH 8.5. Enzyme preparations were stabilized by the addition of bovine serum albumin. The K m for S-adenosylhomocysteine was 41 μ m in the hydrolysis reaction and for adenosine, dl -homocysteine, and l -homocysteine it was 13 μ m , 2.2 m m , and 1.2 m m , respectively. The enzyme was inhibited by S-adenosylmethionine, homocysteine, and adenine. These inhibitions and the K m values determined are discussed in relation to the regulation of the enzyme in vivo and especially its effect on methylation reactions using S-adenosylmethionine as methyl donor.

The leaves of Nandina domestica have yielded a new cyanogenic compound, characterized here as the 3-(3,4-dihydroxyphenyl) −2-propenoic acid 4′-ester of 4-(β- d -glucopyranosyloxy)-α-hydroxybenzeneacetonitrile.
