Abstract

Rare variants in at least 10 genes, including BRCA1, BRCA2, PALB2, ATM, and CHEK2, are associated with increased risk of breast cancer; however, these variants, in combination with common variants identified through genome-wide association studies, explain only a fraction of the familial aggregation of the disease. To identify further susceptibility genes, we performed a two-stage whole-exome sequencing study. In the discovery stage, samples from 1528 breast cancer cases enriched for breast cancer susceptibility and 3733 geographically matched unaffected controls were sequenced. Using five different filtering and gene prioritization strategies, 198 genes were selected for further validation. These genes, and a panel of 32 known or suspected breast cancer susceptibility genes, were assessed in a validation set of 6211 cases and 6019 controls for their association with risk of breast cancer overall, and by estrogen receptor (ER) disease subtypes, using gene burden tests applied to loss-of-function and rare missense variants. Twenty genes showed nominal evidence of association (p-value < 0.05) with either overall or subtype-specific breast cancer. Our study had the statistical power to detect susceptibility genes with effect sizes similar to ATM, CHEK2, and PALB2, however, it was underpowered to identify genes in which susceptibility variants are rarer or confer smaller effect sizes. Larger sample sizes would be required in order to identify such genes.

9. Treatment of gummy smile combining crown lengthening, lip repositioning, and the use of polyester threads.

Abstract

Introduction: The case report presents a new possibility of treatment for a gummy smile in a patient with multiple etiologies, such as altered passive eruption and hypermobility upper lip. At first, crown lengthening was not sufficient to achieve the desired aesthetic result, being necessary its combination with lip repositioning. Case Presentation: Crown lengthening surgery (CLS) was performed in a 20‐year‐old woman, with a gingival display of 7.5 mm, having her gingival exposure reduced to 5.5 mm. Because the patient continued unsatisfied after 6 months, a new procedure was adopted. To reduce even more her gingival exposure, lip repositioning technique was performed associated with myotomy and the insertion of polyester threads as a physical barrier to prevent relapse. Conclusion: The result of the gingival display was reduced to 2.5 mm, removing the condition of a gummy smile after the combination of both techniques: crown lengthening, and lip repositioning. Key points: Why is this case new information? Association of the technique of lip repositioning and myotomy, the insertion of polyester threads that act as a physical barrier against recurrence. What are the keys to successful management of this case? Correct etiological diagnosis.Prior application of botulinum toxin.Respect the period of 1 month for the insertion of the polyester thread, helping to preserve the suture (limitation of movement). Abstract

The cryptic ushA gene (ushA(super.c)) in Salmonella enterica natural isolates (serotype Typhimurium) has been seen to be inactivated by a single missense mutation, the first reported gene inactivation in natural isolates of bacteria via a missense mutation. Results indicate the evolution of new enzymes involving a silent gene which produces an relatively inactive or inactive product. They also indicate evolution of a novel, although unknown, enzyme specificity by one amino acid change.

Abstract

An investigation of the in vitro and in vivo redox states of the Escherichia coli periplasmic oxidoreductases DsbA and DsbC was conducted using genetic selections. Results indicated DsbC acts as a disulfide isomerase. Furthermore, DsbA is a more effective oxidant than DsbC of protein dithiols. Proteins which are effected by DsbC deletions do not have cysteines paired consecutively.

A bacterial expressing system helps isolate the microtubule-binding region (MTBR) of the microtubule associated protein (MAP) 2 and site-directed mutagenesis helps study the interactions between microtubules and MAP. The MTBR enhances tubulin polymerization. The binding energies of the three second repeated peptide sequences remain unchanged during MAP binding to microtubules, confirming that the second sequence repeat is vital for microtubule binding.

28. Primary stability of dental implants installed using osseodensification or bone expansion drilling systems: a comparative clinical study.

Abstract

Aim: Primary stability of dental implants is a prerequisite for osseointegration, and the osteotomy technique is one of the factors that may alter it. The aim of this study was to compare the primary stability of dental implants installed using osseodensification and bone expansion drilling systems. Materials and Methods: This split-mouth comparative clinical study included 12 patients. Each patient received one implant for each of the tested systems, totaling 24 implants. Implants were installed in edentulous areas with space for two adjacent implants or in two similar contralateral areas for the installation of a single implant. The areas had to present medullar bone interposed between vestibular and palatal/lingual cortical bone. The primary outcome was the insertion torque. Results: The average torque obtained with bone expansion was 37±14 N.cm, whereas the final torque equaled 46±10 N.cm with osseodensification (p=0.02). The average torque was also higher with osseodensification than bone expansion in cases of type IV bone, in the maxilla and in cases of narrow implants. Osseodensification reached 35N. cm or more in 91.7% of cases compared to 50% after bone expansion (p=0.02). Conclusion: Primary stability is affected by the osteotomy technique. Dental implants installed after osseodensification presented higher levels of insertion torque than after a bone expansion system. [ABSTRACT FROM AUTHOR]

Abstract

La práctica habitual de actividad física (AcF) aporta beneficios en la salud, en especial en la prevención de enfermedades cardio y cerebrovasculares. La AcF parece tener un efecto más allá de la disminución de la incidencia y recurrencia del ictus, existiendo algunos estudios que la asocian a menor gravedad inicial del ictus y a mejor pronóstico funcional, aunque los estudios clínicos publicados son escasos y metodológicamente heterogéneos. Sólo estudios experimentales han relacionado la AcF con un menor volumen de la lesión isquémica, y ningún estudio experimental o clínico ha estudiado su relación con la recanalización arterial en el ictus agudo. Se cree que la práctica de AcF se comporta como un precondicionante de la isquemia, favoreciendo la tolerancia a la misma una vez que se produce, gracias a mecanismos de neuroprotección y neurorreparación. Aunque las vías exactas de señalización molecular y celular a través de las cuales la AcF aporta dichos beneficios son desconocidas, existen una serie de intermediaros implicados en procesos de vasculogénesis, angiogénesis, arteriogénesis y neurogénesis que aparecen de forma constante en la literatura en relación, por un lado, a la práctica de AcF y, por otro lado, en respuesta a la isquemia cerebral, como son las EPCs circulantes y algunos factores de crecimiento neuronal y vascular tales como los VEGF, G-CSF y BDNF. Sin embargo, no existen hasta el momento estudios clínicos en pacientes con ictus isquémico que analicen simultáneamente el papel de la AcF previa y de los citados intermediarios moleculares en el pronóstico clínico y radiológico del ictus. En este estudio prospectivo de pacientes con ictus agudo ( 12h) con oclusión de la ACM o TICA se ha observado que niveles elevados de AcF previo al ictus se asocian a menor gravedad neurológica al ingreso, a mayor tasa de recanalización arterial precoz tras tratamiento fibrinolítico endovenoso, a mejoría dramática precoz, a menor volumen final del infarto y a mejor pro, Regular physical activity (PhA) is beneficial for health, it is especially associated with the prevention of heart and brain vascular diseases. However PhA seems to have an effect beyond the decrease of stroke risk and recurrence, some studies suggest prestroke PhA is associated with lower stroke severity and better functional outcome, though there are only a few published clinical studies with heterogeneous methodology. Only experimental studies have related prestroke PhA with smaller infarct volume, though no studies in animal models or humans have evaluated its effect on arterial recanalization in acute stroke. Prestroke PhA seems to have a preconditioning effect on ischemia, favouring its tolerance once it is initiated, through neuroprotection and neurorepair mechanisms. Though it is unknown the exact molecular and cellular paths that take place in the prestroke PhA beneficial effects, there are a series of intermediaries involved in neovascularization and neurogenesis that constantly come out related with regular PhA practice or brain ischemia, as EPCs and some endothelial and neuronal growth factors like VEGF, G-CSF and BDNF. However, at the moment there are no clinical studies in humans with ischemic stroke that evaluate simultaneously the effect of prestroke PhA and the aforementioned intermediaries on the stroke clinical and radiological prognosis. This prospective study performed in acute stroke patients ( 12h) with MCA or TICA occlusion has observed that higher levels of prestroke PhA is associated with lower stroke severity at admission, higher rate of early arterial recanalization in patients treated with intravenous thrombolysis, early dramatic improvement, smaller infarct volume and better third month functional outcome, in addition to this it is also associated with a higher increase of VEGF on the seventh day after stroke onset. Abstract

Background: Multiple recent genome-wide association studies (GWAS) have identified a single nucleotide polymorphism (SNP), rs10771399, at 12p11 that is associated with breast cancer risk. Method: We performed a fine-scale mapping study of a 700 kb region including 441 genotyped and more than 1300 imputed genetic variants in 48,155 cases and 43,612 controls of European descent, 6269 cases and 6624 controls of East Asian descent and 1116 cases and 932 controls of African descent in the Breast Cancer Association Consortium (BCAC; http://bcac.ccge.medschl.cam.ac.uk/ ), and in 15,252 BRCA1 mutation carriers in the Consortium of Investigators of Modifiers of BRCA1/2 (CIMBA). Stepwise regression analyses were performed to identify independent association signals. Data from the Encyclopedia of DNA Elements project (ENCODE) and the Cancer Genome Atlas (TCGA) were used for functional annotation. Results: Analysis of data from European descendants found evidence for four independent association signals at 12p11, represented by rs7297051 (odds ratio (OR) = 1.09, 95 % confidence interval (CI) = 1.06-1.12; P = 3 × 10-9), rs805510 (OR = 1.08, 95 % CI = 1.04-1.12, P = 2 × 10-5), and rs1871152 (OR = 1.04, 95 % CI = 1.02-1.06; P = 2 × 10-4) identified in the general populations, and rs113824616 (P = 7 × 10-5) identified in the meta-analysis of BCAC ER-negative cases and BRCA1 mutation carriers. SNPs rs7297051, rs805510 and rs113824616 were also associated with breast cancer risk at P < 0.05 in East Asians, but none of the associations were statistically significant in African descendants. Multiple candidate functional variants are located in putative enhancer sequences. Chromatin interaction data suggested that PTHLH was the likely target gene of these enhancers. Of the six variants with the strongest evidence of potential functionality, rs11049453 was statistically significantly associated with the expression of PTHLH and its nearby gene CCDC91 at P < 0.05. Abstract

In the development of biopharmaceutical products, the expectation of regulatory agencies is that the recombinant proteins are produced from a cell line derived from a single progenitor cell. A single limiting dilution step followed by direct imaging, as supplemental information, provides direct evidence that a cell line originated from a single progenitor cell. To obtain this evidence, a high‐throughput automated imaging system was developed and characterized to consistently ensure that cell lines used for therapeutic protein production are clonally‐derived. Fluorescent cell mixing studies determined that the automated imaging workflow and analysis provide ∼95% confidence in accurately and precisely identifying one cell in a well. Manual inspection of the images increases the confidence that the cell line was derived from a single‐cell to >99.9%. © 2017 American Institute of Chemical Engineers Biotechnol.
