International audience; When telomeres shorten, they are thought to lose their protective caps at a critical short length, activating the DNA damage response and recruiting DNA damage repair activities that would degrade, fuse or recombine dysfunctional telomeres. However, the structure(s) of short and dysfunctional telomeres, which respectively trigger permanent replicative senescence and/or potentially promote genome instability, remain unclear.To define the structure of telomeres at the point of dysfunction and the fate of cells carrying them, we have developed a system called FinalCut in Saccharomyces cerevisiae to induce a single telomere of defined length in cells in which we can conditionally inactivate telomerase. This allows 1/ the structural analysis of the shortest telomere in cells, 2/ the monitoring of the pace and the number of consecutive cell cycles from telomerase inactivation to cell death with our microfluidic system, and 3/ the combination of our single cell - single telomere resolved data with a mathematical model of senescence in budding yeast.Our results show that a very small fraction of cells can enter senescence in a probabilistic manner, as cells proliferate in the absence of telomerase. However, for the large majority of cells, when the shortest telomere reaches a critical threshold length, cells immediately stop proliferation. This is consistent with the model that the first telomere reaching a short threshold length is sufficient to abruptly stop cell divisions in a large majority of cells. The structure of this critically short telomere will be discussed.

Abstract

A key challenge in the actual context of fossil sources rarefaction, global warming, and of increase of the world global population, is to promote the use of molecules derived from renewable sources such as plants. Among these molecules, lipids and proteins are targets of interest. Plant lipids from oilseeds are attractive substitutes to the use of fossil oil. Till the beginning of the 20th century, numerous products used in the daily life were derived from natural renewable products. For instance, plant oil was commonly used as fuel for vehicles and was entering in the composition of paintings, lubricants etc. Unfortunately, natural oils have been progressively replaced by cheaper fossil oil in the fabrication of these products. Nowadays, fossil oils are becoming increasingly expensive being a finite comodity. It is thus important to reduce our dependence from fossil oil and develop substitution industries. Oilseeds contain important amounts of proteins which are mainly used in feed. As several kilograms of plant protein are needed to obtain one kilogram of animal protein, the interest toward using plant protein in food is reinforced. The developments of the use of plant lipids, as well as proteins are a major stakes for the competitiveness of European agriculture and industry, as well as for sustainable development. Extraction of oil and proteins from rapeseed has a significant cost, in term of energy and solvent uses, and finally affects the ultimate quality of the products (protein digestibility). In order to quantitatively extract seed reserves under mild conditions, it will be necessary to limit the amount of energy needed, and avoid any use of solvents. Ideally, seeds should be processed in a bio refinery. In this paper, we will describe how oilseeds store their reserves, and roadblocks for improving actual oilseed extraction processes. A continuum of research projects aimed at answering targeted questions will be presented, with selected results obtained.

International audience; Telomeres ensure genome integrity and are maintained by telomerase. During replicative senescence, telomerase is inactivated, telomere sequences progressively shorten, and set the limit for cell proliferation. As telomeres shorten, they are thought to lose their protective caps at a critical short length, activating the DNA damage response and recruiting DNA damage repair activities that would degrade, fuse, or recombine dysfunctional telomeres. However, the structure(s) of short and dysfunctional telomeres, which respectively trigger permanent replicative senescence or potentially promote genome instability, remain unclear. To define the structure of telomeres at the point of dysfunction and the fate of cells carrying them, we developed a system called FinalCut to induce a single telomere of defined length in cells in which we can conditionally inactivate telomerase. This allows structural analysis of this telomere, and combined with the use of our microfluidic system to track consecutive cell cycles from telomerase inactivation to cell death, we can achieve single telomere and single cell resolution. Our results show that the loss of telomere stability is progressive as the telomere shortens, and that telomeres of a given length can be protected or unprotected in separate cells. We conclude that telomere length is not the sole determinant of the switch between functional and dysfunctional telomeres. We therefore propose that the minimum length of telomeres to maintain cell viability follows a probabilistic, rather than deterministic, model.

15. Purification and protein composition of oil bodies from Brassica napus seeds

Abstract

Seed oil bodies are intracellular particles to store lipids as food reserves in oleaginous plants. Description of oil body-associated proteins of Arabidopsis thaliana has been recently reported whereas only few data are available in the case of rapeseed. Oil bodies have been prepared from two double-low varieties of Brassica napus seeds, a standard variety (Explus) and an oleic variety (Cabriolet). Oil bodies have been purified using floatation technique in the successive presence of high salt concentration, detergent or urea in order to remove non-specifically trapped proteins. The integrity of the oil bodies has been verified and their size estimated. Their protein and fatty acid contents have been determined. The proteins composing these organelles were extracted, separated by denaturing gel electrophoresis, digested by trypsin and their peptides were subsequently analyzed by liquid chromatography-tandem mass spectrometry. Protein identification was performed using Arabidopsis thaliana protein sequence database and a collection of Expressed Sequence Tag (EST) of Brassica napus generated from the framework of the French plant genomics programme “Genoplante”. This led to the identification of a limited number of proteins: eight oleosins showing a high similarity each other and representing up to 75% of oil body proteins, a 11 β hydroxysteroid dehydrogenase-like protein highly homologous to the same protein from A. thaliana, and only few contaminating proteins associated with myrosinase activity.

Abstract

This study characterizes the sequence, end structure, and length distribution of Chlamydomonas reinhardtii telomeres and shows that telomerase mutants are defective in telomere maintenance., Telomeres are repeated sequences found at the end of the linear chromosomes of most eukaryotes and are required for chromosome integrity. Expression of the reverse-transcriptase telomerase allows for extension of telomeric repeats to counteract natural telomere shortening. Although Chlamydomonas reinhardtii, a photosynthetic unicellular green alga, is widely used as a model organism in photosynthesis and flagella research, and for biotechnological applications, the biology of its telomeres has not been investigated in depth. Here, we show that the C. reinhardtii (TTTTAGGG)n telomeric repeats are mostly nondegenerate and that the telomeres form a protective structure, with a subset ending with a 3′ overhang and another subset presenting a blunt end. Although telomere size and length distributions are stable under various standard growth conditions, they vary substantially between 12 genetically close reference strains. Finally, we identify CrTERT, the gene encoding the catalytic subunit of telomerase and show that telomeres shorten progressively in mutants of this gene. Telomerase mutants eventually enter replicative senescence, demonstrating that telomerase is required for long-term maintenance of telomeres in C. reinhardtii.

Abstract

National audience; In yeast, animals and other organisms, storage lipids are maintained in the cytoplasm in specialized organelles called lipid droplets (LDs) or oil bodies. These structures consist mainly of a core of neutral lipids (triacylglycerols and/or steryl esters) enclosed in a monolayer of phospholipids, and contain a number of proteins which vary considerably with the species. In particular, some structural hydrophobic proteins stabilize the interface between lipid core and cellular aqueous environment (perilipins, apolipoproteins, oleosins). Oleosins, with their mass between 15 and 25 kDa, are the major proteins of plant LDs, and completely cover the LD surface. In the last decade, proteomic and genetic studies of this compartment have shown that LDs appear as a complex dynamic organelle with a role in metabolism control and cell signaling. LDs have received a lot of attention as their abnormal dynamics is associated with several metabolic diseases (obesity, diabetes, atherosclerosis and myopathies). In the context of green chemistry, LDs are also promising sources of neutral lipids for the development of derived products, such as biofuels and alternative molecules for food, medicine and cosmetics. For these reasons, understanding dynamics of LD and structure of LD integral proteins is of major importance. We developed a genetic approach using Saccharomyces cerevisiae to obtain heterologous expression of Arabidopsis thaliana LD proteins [1]: oleosin AtOle1 or caleosin AtClo1. These transformed yeasts overaccumulated LDs leading to a specific increase of storage lipids and these LDs harbor a high level of oleosins. These cells or their purified LDs were exposed to the Soleil synchrotron light to understand the dynamics of lipid storage and carbon fluxes by measuring the biochemical changes on single cells using the Synchrotron FTIR microspectroscopy on SMIS beamline [2]. to obtain structural data on whole cellular organelle and on oleosins inserted in LD using Sychrotron Radiation Circular Dichroism (SRCD) [3], Deep UV imaging on DISCO beamline and Synchrotron water radiolysis footprinting of accessible oleosin amino-acids on METROLOGY beamline.

Abstract

International audience; Organelle proteomics describes the study of proteins present in organelle at a particular instance during the whole period of their life cycle in a cell. Organelles are specialized membrane bound structures within a cell that function by interacting with cytosolic and luminal soluble proteins making the protein composition of each organelle dynamic. Depending on organism, the total number of organelles within a cell varies, indicating their evolution with respect to protein number and function. For example, one of the striking differences between plant and animal cells is the plastids in plants. Organelles have their own proteins, and few organelles like mitochondria and chloroplast have their own genome to synthesize proteins for specific function and also require nuclear-encoded proteins. Enormous work has been performed on animal organelle proteomics. However, plant organelle proteomics has seen limited work mainly due to: (i) inter-plant and inter-tissue complexity, (ii) difficulties in isolation of subcellular compartments, and (iii) their enrichment and purity. Despite these concerns, the field of organelle proteomics is growing in plants, such as Arabidopsis, rice and maize. The available data are beginning to help better understand organelles and their distinct and/or overlapping functions in different plant tissues, organs or cell types, and more importantly, how protein components of organelles behave during development and with surrounding environments. Studies on organelles have provided a few good reviews, but none of them are comprehensive. Here, we present a comprehensive review on plant organelle proteomics starting from the significance of organelle in cells, to organelle isolation, to protein identification and to biology and beyond. To put together such a systematic, in-depth review and to translate acquired knowledge in a proper and adequate form, we join minds to provide discussion and viewpoints on the collaborative nature of organelles in cell, their proper function and evolution. © 2010 Wiley Periodicals, Inc.
