Abstract

This annual review is the sixth of its kind since 2016 (see references). Our objective is to explore and share articles which we deem influential and significant in the field of biotransformation and bioactivation. These fields are constantly evolving with new molecular structures and discoveries of corresponding pathways for metabolism that impact relevant drug development with respect to efficacy and safety. Based on the selected articles, we created three sections: (1) drug design, (2) metabolites and drug metabolizing enzymes, and (3) bioactivation and safety (Table 1). Abstract

The conservation of rarely encountered species is notoriously difficult mainly because of the lack of information about their natural history. This is definitely the case for the spot-tailed earless lizards (Holbrookia lacerata and H. subcaudalis). Sparse distributional records and infrequent observations at known sites were used as evidence to petition these species for protection under the Endangered Species Act. For this study, we did surveys in historic and new areas to determine the current distribution of both species. We also radio-tracked individuals from two populations to estimate movement potential and home range sizes, as well as understand habitat use. Distribution surveys indicate that H. lacerata has undergone range contraction (39%); however, H. subcaudalis was historically found in 21 counties but we only observed them in five. We found home ranges to be relatively large compared to similarly sized insectivores and at least four times larger than the similar H. maculata. Both species are inhabitants of historic grasslands, but within this habitat type they used sites that were more open compared to what was available, including areas such as row-crop agriculture and frequently mowed fields. Frequent habitat disturbance seems to be important for both species. Urbanisation, invasive woody vegetation and invasive grasses are all considered threats that may have already caused declines to both species. [ABSTRACT FROM AUTHOR]

Abstract

The goal of competitive bodybuilding is to attain perfection in muscle development. The first phase of training is to build muscle, while the final phase before a competition seeks to reduce body fat, especially the skin surface, to further enhance cosmetic appearances. The training regimen of a 27-year-old male bodybuilder 30 days prior to a competition is described. Dietary changes, exercise, and steroid and diuretic drugs were used in this final phase to achieve weight and fat loss goals. The subject followed a drug regimen he received from another bodybuilder which included taking six different steroids (300-400 milligrams per week), a diuretic to achieve water loss, and a female hormone he said he used to stimulate testosterone (male hormone) production. He purchased the steroids at his gym and could not get them the last week of his training. The bodybuilder achieved a 15.6 pound weight loss and placed in the top three in his division at the competition. While the subject was on steroids his blood tests showed several abnormal values, including an increase in red blood cells and blood concentration. This may have been the result of taking both steroids, which stimulate red blood cell production, and diuretics, which decrease blood concentration through fluid loss. Blood values began to return to normal the last week of training when the steroids were unavailable. This subject also had changes in his cholesterol levels, with an increase in total cholesterol and low density lipoproteins and a decrease in high density lipoproteins, changes which are considered unfavorable. These values also reverted to normal with cessation of steroid use. Approximately one third of the weight lost was lean body tissue and not fat, which suggests that the weight loss was achieved too quickly. The authors suggest that the second phase should have been started earlier to prevent loss of any muscle bulk that was achieved during the first phase. They also suggest that the changes in lipid and blood concentrations that resulted during this training period put this man at risk for blood clot formation, with possible serious health consequences. 26. The geotectonic evolution of the Western Ethiopian shield

Abstract

The western Ethiopian Shield comprises three lithotectonic units. The Birbir domain, an assemblage of mafic to felsic intrusive and extrusive rocks and mainly volcanogenic sedimentary rocks, is enclosed between the dominantly orthogneissic Baro and Geba domains. The earliest recorded deformation event (D1) resulted in the formation of a subhorizontal gneissosity within the gneissic terranes which was synchronous with an early upper amphibolite-facies metamorphic peak (M1) at 800–770 Ma which locally caused partial melting. All terrains were subsequently deformed in the D2 event which was the result of severe E-W crustal shortening. An anticlockwise P-T-t path is implied. Subsequent D3 deformation was concentrated within mylonitised domain boundaries which record major transcurrent movement. These structures were reactivated and suffered fluid incursion resulting in isotopic reequilibration at 635–580 Ma. A second metamorphic event, M2, related to crustal thickening and consequent granite genesis, occurred after the cessation of D3 shearing. Bulk chemical analyses show that the metamorphosed plutonic and volcanic rocks of the Birbir domain are predominantly calc-alkaline and similar to those generated by subduction in modern magmatic arcs. They belong in part to the low-K series, suggesting an oceanic environment. The evolution of the region can be explained in terms of the melting of a subducting slab, intrusion, metamorphism and the formation of an oceanic island arc complex. Continued plate convergence caused severe east-west shortening and basin closure. Further attenuation gave rise to transcurrent shearing, fluid influx, a second thermal event and accretion of microcontinents.
