Abstract

Chronic progressive membranous nephropathy (MN) in humans is characterized by thickening of the glomerular basement membrane (GBM) with formation of spikes which contain laminin and other extracellular matrix (ECM) proteins. We have utilized two models of MN in the rat (active and passive Heymann nephritis, AICN, PHN) to define the sequential changes in composition of GBM as they relate to changes in glomerular gene expression for ECM components, altered permeability and morphological changes. Renal biopsies obtained during the course of AICN and PHN were immunostained for various ECM proteins and total glomerular RNA was hybridized with cDNA probes specific for laminin B2-chain, s-laminin, and types I and IV collagen. In addition, the ability of anti-glomerular epithelial cell (GEC) antibody and complement on rat GEC in culture to induce laminin release or laminin and s-laminin mRNA expression was determined. The results demonstrate that at weeks 12, 16, and 20 of AICN, immunostaining for laminin, s-laminin, fibronectin, entactin, and heparan sulfate proteoglycan increased in the GBM in a spike-like pattern. Concomitantly, glomerular mRNA levels of laminin B2-chain and of s-laminin increased. Type IV collagen protein and gene expression remained unchanged or decreased. No glomerular immunostaining for type I collagen occurred during AICN despite increased expression of mRNA for this collagen type. In contrast to AICN, in PHN no pronounced changes of the glomerular ECM occurred, except for transient expression of type I collagen mRNA in whole glomerular RNA and type I collagen protein the GEC cytoplasm. Stimulation of GEC in culture with anti-GEC antibody and complement also failed to induce transcription of laminin or s-laminin mRNA or the release of laminin protein. These findings suggest that the polyantigenic expansion of GBM which occurs in chronic experimental MN may be stimulated by factors different from the C5b-9 mediated processes that cause the initial proteinuria.

Abstract

Gas exchange between the atmosphere and ocean interior profoundly impacts global climate and biogeochemistry. However, our understanding of the relevant physical processes remains limited by a scarcity of direct observations. Dissolved noble gases in the deep ocean are powerful tracers of physical air-sea interaction due to their chemical and biological inertness, yet their isotope ratios have remained underexplored. Here, we present high-precision noble gas isotope and elemental ratios from the deep North Atlantic (~32°N, 64°W) to evaluate gas exchange parameterizations using an ocean circulation model. The unprecedented precision of these data reveal deep-ocean undersaturation of heavy noble gases and isotopes resulting from cooling-driven air-to-sea gas transport associated with deep convection in the northern high lati-tudes. Our data also imply an underappreciated and large role for bubble-mediated gas exchange in the global air-sea transfer of sparingly soluble gases, including O2, N2, and SF6. Using noble gases to validate the physical representation of air-sea gas exchange in a model also provides a unique opportunity to distinguish physical from biogeochemical signals. As a case study, we compare dissolved N2/Ar measurements in the deep North Atlantic to physics-only model predictions, revealing excess N2 from benthic denitrification in older deep waters (below 2.9 km). 7. Waterlogging in the temperate plantation species Eucalyptus globulus and E. nitens

Abstract

This project investigated the effect of waterlogging on the productivity of Eucalyptus globulus under field conditions, and the relative tolerance of the two temperate plantation eucalypt species, E. glob ulus and E. nitens, from the subgenus Symphyomyrtus, to waterlogging. In eucalypt plantations across Tasmania, waterlogging is a serious threat to productivity. It is estimated that at least 11 % of all commercial eucalypt plantations in Tasmania are affected to some degree, by waterlogging. The losses incurred by waterlogging during the first 2 years of plantation establishment were found to have significant long-term effects on tree productivity at two sites across the state. In the field, soil indicators of long-term waterlogging, specifically the evidence of greying, in combination with current seasonal waterlogging and development of hypoxic soil conditions were found to be the factors closely associated with severe reductions in tree height and growth. The use of extensive soil mapping and assessment prior to plantation establishment are therefore effective tools in identifying potential waterlogging problems. Under long-term waterlogging, there was no indication of the development of plant water stress during exposure to waterlogging. Waterlogged seedlings of both species exhibited reduced foliar nutrient status, with significantly reduced foliar nitrogen, phosphorus and potassium concentrations. Therefore, waterlogging-induced nutrient deficiency is a mechanism involved in the growth reduction of these species under waterlogged conditions. Various methods of fertiliser application were investigated to determine the most \effective mode in ameliorating waterlogging damage. Soil-based applications of slow release fertilisers applied either prior to or after a waterlogging event were the most successful. Fertiliser application was associated with improved growth foliar nutrition and increased aerenchymatous root production. Reduced availability of nutrients in waterlogged soils and/or reduced uptake of these nutrients are the two possible mechanisms leading to the development of foliar nutrient deficiency of waterlogged plants. It was found that under hypoxic hydroponic culture seedlings with low nutrient status were capable of an increased uptake of nutrients when transferred to high nutrient conditions. This result is significant as it identifies reduced availability of soil nutrients as the primary mechanism responsible for the development of foliar nutrient deficiency in E. globulus and E. nitens under waterlogged conditions. The relationship between aerenchymatous root development and waterlogging tolerance was investigated. It was determined that singular measurements of root adaptive capacity should not be used as the sole measure of tolerance to waterlogging in these species. The regulation of growth and maintenance of relatively high photosynthetic rates are also useful indicators of tolerance. Using this array of measures it was determined that E. nitens was inherently more waterlogging tolerant than E. globulus. Abstract

For more than half a century, researchers around the world have been engaged in attempts to achieve fusion ignition as a proof of principle of various fusion concepts. Following the Lawson criterion, an ignited plasma is one where the fusion heating power is high enough to overcome all the physical processes that cool the fusion plasma, creating a positive thermodynamic feedback loop with rapidly increasing temperature. In inertially confined fusion, ignition is a state where the fusion plasma can begin "burn propagation" into surrounding cold fuel, enabling the possibility of high energy gain. While "scientific breakeven" (i.e., unity target gain) has not yet been achieved (here target gain is 0.72, 1.37 MJ of fusion for 1.92 MJ of laser energy), this Letter reports the first controlled fusion experiment, using laser indirect drive, on the National Ignition Facility to produce capsule gain (here 5.8) and reach ignition by nine different formulations of the Lawson criterion.

This deliverable is related to task “T2.1. Worker needs, industrial requirements and use cases” of Ageing@Work, which deals with the formulation of the basis of the user-centred approach that will be followed throughout the project. This activity focuses on the detailed analysis of the users’ needs and specifications, alongside with the industry requirements, and the use cases, in order to establish the user requirements for the smart, personalized and adaptive ICT solutions for active, healthy and productive ageing with enhanced workability of the Ageing@Work project. Abstract

This article reports a commentary on the methodology surrounding the exploratory development and study efforts regarding a published article on the viability of a novel “going-in light” or Going Light medical component in support of US Army Pacific (USARPAC) Humanitarian Assistance/Disaster Relief (HA/DR) missions, namely, a BLU-MED® incremental modular equipment package along with a Rapid Deployment Medical Team (RDMT). There was an unreported and “untold story” in terms of the Going Light study, specifically, Capabilities Based Assessment going on in the background. This was really the consequence of Soldiers on the study group auto-performing in accordance with their training. Details on how the CBA process proceeded and deviations in the sequencing of that process are reported in detail as well as how the process was integrated in the Oracle Delphi.

Abstract

BACKGROUND AND OBJECTIVE: Serum uric acid may predict the onset and progression of kidney disease, but it is unclear whether uric acid is an independent risk factor for diabetic nephropathy. Our aim was to study the relationship between uric acid levels and the development of CKD components in patients with type 2 diabetes. DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: Longitudinal study of a cohort of patients with type 2 diabetes from the database of the Italian Association of Clinical Diabetologists network. From a total of 62,830 patients attending the diabetes centers between January 1, 2004, and June 30, 2008, we considered those with baseline eGFR values ≥60 ml/min per 1.73 m2 and normal albumin excretion (n=20,142). Urinary albumin excretion, GFR, and serum uric acid were available in 13,964 patients. We assessed the association of serum uric acid quintiles with onset of CKD components by multinomial logistic regression model adjusting for potential confounders. Abstract

In brief Tibial stress fractures often occur in Patients who participate in running and jumping sports. With rest, most stress fractures heal without incident, though midshaft fractures are more resistant to healing. Bone scans are usually required to confirm the diagnosis. Relative rest, strength training, proper footwear, and treating underlying risk factors are the mainstays of treatment. A new treatment option is a pneumatic brace that the patient can wear when he or she returns to play. Physicians should be prepared to counsel competitive athletes about the risks of returning to play too soon.
