Abstract

The interactions between acarbose and other carbohydrate recognition enzymes are investigated by probing the crystal structure of Escherichia coli maltodextrin phosphorylase complexed with acarbose. It is shown that acarbose exhibits a conformation that is similar to its major minimum free energy conformation in the maltodextrin-acarbose structure, suggesting that acarbose is able to adopt two different conformations. This flexibility allows acarbose to target a host of different enzymes.

Abstract

X-ray crystallographic studies reveal that the protein and water molecules in contact with nojirimycin tetrazole (NJT) and phosphate ion are the same in the T and the R states of glycogen phosphorylase. The phosphate ion is close enough for hydrogen-bonding with the cofactor pyridoxal 5'-phosphate group plus ionic contact with the N-1 atom of the tetrazole ring in both the states. Structural and kinetic studies indicate that NJT and the phosphate ion show properties similar to that of a transition state analogue and each helps the binding of the other.
