Abstract Thousands of cancer patients are currently in clinical trials evaluating antiangiogenic therapy in the neoadjuvant setting, which is the treatment of localized primary tumors prior to surgical intervention. The rationale is that shrinking a tumor will improve surgical outcomes and minimize growth of occult micrometastatic disease—thus delaying post‐surgical recurrence and improving survival. But approved VEGF pathway inhibitors have not been tested in clinically relevant neoadjuvant models that compare pre‐ and post‐surgical treatment effects. Using mouse models of breast, kidney, and melanoma metastasis, we demonstrate that primary tumor responses to neoadjuvant VEGFR TKI treatment do not consistently correlate with improved post‐surgical survival, with survival worsened in certain settings. Similar negative effects did not extend to protein‐based VEGF pathway inhibitors and could be reversed with altered dose, surgical timing, and treatment duration, or when VEGFR TKIs are combined with metronomic ‘anti‐metastatic’ chemotherapy regimens. These studies represent the first attempt to recapitulate the complex clinical parameters of neoadjuvant therapy in mice and identify a novel tool to compare systemic antiangiogenic treatment effects on localized and disseminated disease.

Metastasis represents one of the main clinical challenge in cancer treatment since it is associated with the majority of deaths. Recent technological advances allow quantification of the dynamics of the process by means of noninvasive techniques such as longitudinal tracking of bioluminescent cells. The metastatic process was simplified here into two essential components – dissemination and colonization – which were mathematically formalized in terms of simple quantitative laws. The resulting mathematical model was confronted to in vivo experimental data of spontaneous metastasis after primary tumor resection. We discuss how much information can be inferred from confrontation of theories to the data with emphasis on identifiability issues. It is shown that two mutually exclusive assumptions for the secondary growth law (namely same or different from the primary tumor growth law) could fit equally well the data. Similarly, the fractal dimension coeffcient in the dissemination law could not be uniquely determined from data on total metastatic burden only. Together, these results delimitate the range of information that can be recovered from fitting data of metastatic growth to already simplified mathematical models.

The present work describes the synthesis of new terpyridine (tpy) molecules possessing functionalized long alkyl chains at the 4'-position associated with a planar structure with considerable delocalization. Out of the three synthesized tpy derivatives (L1, L2, and L3), L2 containing an ester group at the end of the alkyl chain emerged as an excellent probe for the selective detection of Zn 2+ . The detection of Zn 2+ ions under biological conditions was achieved by the introduction of a distinct aliphatic undecanoic ester chain at the 4'-position of the core terpyridine ring, thereby making it more lipophilic. The increased lipophilicity aids in the probe performance by enabling precise Zn 2+ detection and bioimaging. The bioimaging capability of the probe L2 for Zn 2+ ions was demonstrated in ICG cell lines. The RGB method was integrated into the detection process, making it portable and low-cost. The LODs were determined to be 106 and 115 nM, respectively, for the solution-phase and test paper strips. The probe L2 was successfully utilized to determine Zn 2+ levels in pharmaceutical and water samples.

N-acyltaurines (NATs) are biologically active amphiphilic lipids. They come under the group of compounds known as N-acyl amino acids. NATs were first detected in the brain and other tissues in mice lacking the enzyme fatty acid amide hydrolase FAAH (-/-). N-arachidonoyltaurine (20:4 NAT) acts as an excellent ligand for the subset of transient receptor potential (TRP) channels, especially vanilloid type channels TRPV1 and TRPV4. Also, hydrophobic and hydrophilic regions of NATs enable them to interact with membrane lipids. Here, we have investigated the interaction of NATs, N-myristoyltaurine (NMT), and N-palmitoyltaurine (NPT) with their corresponding diacyl phosphatidylcholines (PCs), dimyristoylphosphatidylcholine (DMPC), and dipalmitoylphosphatidylchoine (DPPC). The miscibility and phase behavior of the hydrated binary mixtures have been investigated by differential scanning calorimetry (DSC). Studies on the interaction of NMT/NPT with DMPC/DPPC revealed that the two amphiphiles mix well up to 50 mol% of NAT and phase separation is observed at higher contents of the NAT. The phase transition of the equimolar mixtures of NAT:PC (50:50) studied by fluorescence, also supported the DSC results. PXRD and FTIR analysis show that the NAT:PC equimolar mixture (50:50) forms different supramolecular structures when compared to that of individual NATs and PCs. The phosphorylated histone variant, γ-H2AX, is known to play a key role in DNA damage repair. However, the clinical significance of H2AX mRNA expression in breast cancer remains unclear. Utilizing a bioinformatical approach, a total of 3594 breast cancer patients with clinical and transcriptomic data were investigated. Bioinformatical analysis showed that high expression of H2AX is associated with worse disease-free, disease-specific, and overall survival consistently in two independent cohorts. High H2AX expressing tumors were associated with upregulated DNA repair gene sets. Although H2AX was not predictive of chemotherapy response, it was significantly downregulated after effective chemotherapy or radio-chemotherapy. Notably, tumors with high H2AX expression were enriched for DNA replication and MYC targets gene sets, and associated with increased MKI67 expression, suggesting alterations in cell proliferation machinery. H2AX knockdown cells showed decreased cell proliferation as compared to the control cells. Finally, H2AX mRNA expression was higher in the metastatic clones as compared to the parental cells and in the metastatic tumors as compared to the primary tumors in patients, with higher H2AX mRNA expression found in advanced stage cancer patients. Fifty-four adult German patients suffering from idiopathic thrombotic thrombocytopenic purpura (TTP) have been examined for HLA class II. All patients presented autoantibodies against ADAMTS13 and ADAMTS13 activity levels <5%. Blood samples have been analyzed for HLA-DRB1 and DQB1 alleles using sequence-specific primer PCR and sequence-specific oligonucleotide PCR. Reference data of German bone marrow and blood donors were obtained from www.allelefrequencies.net. The results were evaluated employing two-sided binomial tests, and p values were corrected using the Benjamini-Hochberg procedure. A significant accumulation for DQB1*02:02 (p < 0.001) and DRB1*11 (p = 0.003) was found within the patient group. Twenty percent (DQB1*02:02) or 48.1% (DRB1*11) of TTP patients were tested positive for the particular HLA antigen compared to 1.2% (DQB1*02:02) or 23.5% (DRB1*11) in the control group. A tendency for a reduced occurrence of HLA-DRB1*04 was revealed (7.4% in patients compared to 24.6% in controls). An association between the HLA antigens DQB1*02:02 and DRB1*11 and disease susceptibility for idiopathic TTP has been found. A higher risk for disease outbreak within persons carrying the mentioned alleles can be assumed. The reduced occurrence of HLA-DRB1*04 in TTP patients indicates a possible protective effect of this HLA allele in disease development.

Unlabelled: 76 German patients suffering from thrombotic thrombocytopenic purpura (TTP) were interrogated about the prevalence of co-occurring autoimmune disorders. In order to analyze a possible association of TTP with the questioned diseases, a comparison of prevalence rates between the patient group and the general population has been made for each disease., Results: Compared to the estimated prevalence rates, the statistical analysis revealed an unexpected high occurrence of the following disorders within the patient group: Hashimoto's thyroiditis (23.5% within the patients compared to 0.7% within the general population, p<0.001), systemic lupus erythematosus (SLE) (6.5% in patients to 0.025% in the general population, p<0.001), immune thrombocytopenic purpura (ITP) (6.3% in patients to 0.02% in the general population, p<0.001), psoriasis (9.4% in patients to 2.5% in the general population, p=0.005) and celiac disease (3.1% in patients to 0.2% in the general population, p=0.007)., Conclusion: These findings confirm the mentioned tendency of autoimmune diseases to co-occur in one individual and argue once more for a genetic susceptibility in idiopathic TTP as well as in autoimmune disorders.

The distinct effects of cytokines on cellular growth and differentiation suggest that specific signaling pathways mediate these diverse biological activities. Fibroblast growth factors (FGFs) are well-established inhibitors of skeletal muscle differentiation and may operate via activation of specific signaling pathways distinct from recently identified mitogen signaling pathways. We examined whether platelet-derived growth factor (PDGF)-activated signaling pathways are sufficient to mediate FGF-dependent repression of myogenesis by introducing the PDGF beta receptor into a mouse skeletal muscle cell line. Addition of PDGF-BB to cells expressing the PDGF beta receptor activated the PDGF beta receptor tyrosine kinase, stimulated mitogen-activated protein (MAP) kinase, and increased the steady-state levels of junB and c-fos mRNAs. Despite the activation of these intracellular signaling molecules, PDGF beta receptor activation elicited no detectable effect on cell proliferation or differentiation. In contrast to PDGF-BB, addition of FGF-2 to myoblasts activated signaling pathways that resulted in DNA synthesis and repression of differentiation. Because of the low number of endogenous FGF receptors expressed, FGF-stimulated signaling events, including tyrosine phosphorylation and activation of MAP kinase, could be detected only in cells expressing higher levels of a transfected FGF receptor cDNA. As the PDGF beta receptor- and FGF receptor-stimulated signaling pathways yield different biological responses in these skeletal muscle cells, we hypothesize that FGF-mediated repression of skeletal muscle differentiation activates signaling pathways distinct from those activated by the PDGF beta receptor. Activation of PDGF beta receptor tyrosine kinase activity, stimulation of MAP kinase, and upregulation of immediate-early gene expression are not sufficient to repress skeletal muscle differentiation.
