Microplastics are now ubiquitous in freshwater environments. As most previous research has focused on species-specific effects of microplastics under controlled laboratory conditions, little is known about the impact of microplastics at higher levels of ecological organisation, such as freshwater communities and their associated ecosystem functions. To fill this knowledge gap, an outdoor experiment using 40 freshwater mesocosms, each 1.57 m3, was used to determine the effects of (i) microplastic type: traditional oil-based high-density polyethylene versus bio-based biodegradable polylactic acid, (ii) concentration of microplastic particles and (iii) nutrient enrichment. The two concentrations of microplastics used were equivalent to measured environmentally occurring concentrations and concentrations known to cause toxicological effects under laboratory conditions. Freshwater communities are also at increasing risk from nutrient enrichment, which can alter community composition in favour of competitively dominant taxa. The independent and interactive effects of these treatments on pelagic community structure (phytoplankton standing stock, taxonomic richness, and composition) and ecosystem functioning (periphyton productivity and leaf litter decomposition) were assessed. Taxonomic richness and community composition were not affected by exposure to the experimental treatments and there were no significant treatment effects on phytoplankton standing stock, periphyton productivity, total or microbial leaf litter decomposition. Overall, multiple microplastic exposures, crossed with nutrient addition had little impact on the structure and functioning of semi-natural freshwater ecosystems. These findings indicate that the negative impacts of microplastics predicted from species-specific studies may not be readily realised at the ecosystem scale.

Although the concept of trophic interactions has been used for a long time, there are still considerable gaps in our understanding of the effect of various environmental factors on trophic interactions within river fish assemblages. Carbon (δ13C) and nitrogen (δ15N) stable isotope ratios of 20 species of fish belonging to both eurytopic and rheophilic ecological groups from a large temperate rivers were used to evaluate overall trophic niche use and trophic position of species, and to find out how environmental variability associated with nutrient loading affects individual and community-wide aspects of trophic structure. The study was carried out at 11 sites along the European rivers Vltava and Elbe, representing a continuous gradient of pollution and habitat degradation. Corrected Standard Ellipse Area (SEAc) was significantly larger for the group of eurytopic ecological species than for rheophilic species. Despite narrower isotopic niche space, rheophilic fish species occupied a higher trophic position, suggesting that these species use resources more enriched in 15N. Of the 11 environmental variables tested, nutrients had a significant effect on trophic niche area of species (SEAc), indicating that eutrophication is of critical importance for fish assemblages. Isotopic niche area of species was found to be positively influenced by total phosphorus, and negatively affected by concentrations of nitrate (N-NO3−) and ammonia (N-NH4+). A negative association between oxygen demand and a measure of trophic diversity - mean distance to centroid (CD)- and a measure of density and clustering of species - mean nearest neighbor distance (MNND)- were found, indicating that the oxygen demand is a key factor influencing community trophic structure. An observed pattern where nutrient loading influenced both individual species and trophic structure of the fish assemblage provides strong support for an anthropogenic influence on riverine food webs.

Abstract While many studies have considered the ability of eDNA to assess animal communities in lacustrine settings, fewer have considered riverine systems, particularly those spanning the environmental gradients present in large river basins. Such dynamic systems are challenging for eDNA biomonitoring due to differing eDNA transport distances in rivers and the effects of river chemistry. To address this challenge, we focused on the Thames River system, UK, which has exceptional historical fish records providing a baseline to test the accuracy of eDNA metabarcoding in recovering fish community structure across both fresh and tidal zones. Two primer sets targeting 12S and CO1 regions were used to capture fish communities across the Thames catchment, from the upper freshwaters to the mid estuary. eDNA was collected at 35 sites, 14 of which were simultaneously paired with traditional fish surveys for direct comparison. We demonstrated that eDNA metabarcoding consistently detected more freshwater species than traditional methods, despite extensive sampling effort using the latter. In contrast, metabarcoding did not perform as well as traditional approaches in estuarine waters, although results included the novel detection of the protected sea lamprey. We further demonstrated that minor variations in the recovery of all approaches would not impact on the assessment of simple ecological models of community structure and, thus, some variability between approaches should not be viewed as a serious hindrance to uptake. Rather, our findings support a growing consensus that eDNA can reliably detect fish communities across dynamic freshwater habitats.
