Predation can effectively limit insect herbivores with cascading effects on plant community composition and diversity of tropical rainforests. Assessing variation in predation is therefore important to understand the mechanisms structuring complex rainforest ecosystems. Variation in predation with time of day may provide herbivores with temporal enemy-free space. Trichomes (plant hairs) may provide spatial enemy-free space by increasing climbing resistance for walking arthropod predators and by scattering bat echolocation calls. Artificial model prey is commonly used to measure predation pressure on insect herbivores. Whether model prey shape is sufficient to deceive predators and whether attacks represent actual predation however remain unresolved. We used artificial, plasticine prey to assess temporal and spatial variation in predation in two Panamanian rainforests and tested whether model prey shape is as important for prey recognition by predators as often assumed. We assessed the effect of prey shape and size, time of day, and trichomes on predation by comparing attacks on caterpillar- and humanoid-shaped figurines. We find higher nocturnal than diurnal predation in one but not the other forest, suggesting that herbivores may benefit from enemy-free space during the day in some forests. We find no evidence for an effect of trichomes on predation in the two plant species tested. Equal attack numbers on caterpillar- and humanoid-shaped objects challenge the idea that the visual resemblance of model prey alone is sufficient to deceive predators. We conclude that attacks on model prey represent a variety of responses to novel objects (e.g. exploration, aggression, possibly predation) and urge caution when interpreting their results. Purpose: Recent evidence suggests that at least some sarcomas arise through aberrant differentiation of mesenchymal stromal cells (MSCs), but MSCs have never been isolated directly from human sarcoma specimens.Experimental Design: We examined human sarcoma cell lines and primary adherent cultures derived from human sarcoma surgical samples for features of MSCs. We further characterized primary cultures as either benign or malignant by the presence of tumor-defining genetic lesions and tumor formation in immunocompromised mice.Results: We show that a dedifferentiated liposarcoma cell line DDLS8817 posesses fat, bone, and cartilage trilineage differentiation potential characteristic of MSCs. Primary sarcoma cultures have the morphology, surface immunophenotype, and differentiation potential characteristic of MSCs. Surprisingly, many of these cultures are benign, as they do not form tumors in mice and lack sarcoma-defining genetic lesions. Consistent with the recently proposed pericyte origin of MSCs in normal human tissues, sarcoma-derived benign MSCs (SDBMSCs) express markers of pericytes and cooperate with endothelial cells in tube formation assays. In human sarcoma specimens, a subset of CD146-positive microvascular pericytes expresses CD105, an MSC marker, whereas malignant cells largely do not. In an in vitro coculture model, SDBMSCs as well as normal human pericytes markedly stimulate the growth of sarcoma cell lines.Conclusions: SDBMSCs/pericytes represent a previously undescribed stromal cell type in sarcoma that may contribute to tumor formation. Most skeletal fragility disorders are characterized by bone loss with a concurrent gain in marrow adipocytes 1-8. This suggests that a cell that forms adipocytes at the expense of osteoblasts is central to the pathogenesis of skeletal disorders. However, this cellular point of bifurcation between adipocyte and osteoblast differentiation pathways remains unknown. Here, we identify a new cell type defined by co-expression of skeletal stem cell and adipocyte precursor markers, 9-13 (CD24+CD29+ skeletal stem cells (SSCs)), that serves as a key cellular point of bifurcation between the osteoblast and adipocyte differentiation pathways, giving rise to closely related osteoblast and adipocyte lineage-restricted precursors. CD24+CD29+SSCs comprise a small fraction of SSCs, and only this fraction displays full stemness features, including the ability to undergo serial transplantation. In line with serving as the osteoblast/adipocyte bipotent cell, the “bone to fat” tissue remodeling occurring in models of postmenopausal osteoporosis or after high fat diet exposure occur in part by reprogramming these CD24+CD29+SSCs to change their output of lineage-restricted precursors. Lastly, as subcutaneous white adipose tissue displays a similar set of CD24+CD29+ stem cells and related lineage-restricted progenitors, these findings provide a new schema explaining the stem cell basis of bone versus adipose tissue production that unifies multiple mesenchymal tissues.

Epithelioid hemangioma is a rare, histologically benign but locally aggressive primary vascular neoplasm that can rarely arise in bone. Mainstay treatment is surgical resection or curettage with bone grafting. We report a novel multidisciplinary, joint-sparing treatment approach for an epithelioid hemangioma of bone arising in the acetabulum causing severe thinning of the subchondral bone plate. After 4 sessions of transarterial bland particle and ethanol embolization, the resultant increased ossification of the tumor allowed preservation of the articular surface during surgical resection. Imaging follow-up 14 months after surgical resection showed no evidence of recurrence and continued ossification of the portions of the lesion treated only with embolization.

The growth in institutional holdings of public firms has led to increased interest in the concept of common ownership, in which the same investor owns stakes in multiple firms within the same industry. Economic theory suggests that common ownership could affect firm performance, but little empirical research has examined the nature of this effect or how a firm’s extant marketing potentially relates to this effect. This paper addresses this gap by proposing a relationship between common ownership and firm performance that is moderated by the firm’s extant marketing capabilities and its relative marketing strategic emphasis. Our empirical approach employs data from over 43 million institutional holdings to develop a measure of common ownership and accounts for empirical issues like endogeneity and unobserved heterogeneity. The results document a positive relationship between common ownership and firm performance and provide some evidence that this effect is stronger for firms with lower marketing capabilities and a relative strategic emphasis towards R&D spending. These results suggest that public policymakers should consider the firms’ extant strategic marketing when assessing regulations on common ownership.

TGCT comprises a nodular- and a diffuse-type, with the former exhibiting mostly indolent course and the latter a locally aggressive behavior. Although usually not life-threatening, TGCT may cause chronic pain and adversely impact function and quality of life (QoL). CSFR1 inhibitors are effective with benefit on symptoms and QoL but are not available in most countries. The degree of uncertainty in selecting the most appropriate therapy and the lack of guidelines on the clinical management of TGCT make the adoption of new treatments inconsistent across the world, with suboptimal outcomes for patients. A global consensus meeting was organized in June 2022, involving experts from several disciplines and patient representatives from SPAGN to define the best evidence-based practice for the optimal approach to TGCT and generate the recommendations presented herein.

There is growing recognition of the potential of large-scale restoration in the Amazon as a “nature-based solution” to climate change. However, our knowledge of forest loss and recovery beyond Brazil is limited, and carbon emissions and accumulation have not been estimated for the whole biome. Combining a 33-year land cover dataset with estimates of above-ground biomass and carbon sequestration rates, we evaluate forest loss and recovery across nine Amazonian countries and at a local scale. We also estimate the role of secondary forests in offsetting old-growth deforestation emissions and explore the temporal trends in forest loss and recovery. We find secondary forests across the biome to have offset just 9.7% of carbon emissions from old-growth deforestation, despite occupying 27.6% of deforested land. However, these numbers varied between countries ranging from 9.0% in Brazil to 23.8% in Guyana for carbon offsetting, and 24.8% in Brazil to 56.9% in Ecuador for forest area recovery. We reveal a strong, negative spatial relationship between old-growth forest loss and recovery by secondary forests, showing that regions with the greatest potential for large-scale restoration are also those that currently have the lowest recovery (e.g. Brazil dominates deforestation and emissions but has the lowest recovery). Our findings identify three important challenges for policy makers: (1) incentivising large-scale restoration in highly deforested regions, (2) protecting secondary forests without disadvantaging landowners who depend on farm-fallow systems, and (3) preventing further deforestation. Combatting all of these successfully is essential to ensuring that the Amazon biome achieves its potential in mitigating anthropogenic climate change.

Pancreatic ductal adenocarcinomas respond poorly to chemotherapy, in part due to the dense tumor stroma that hinders drug delivery. Ultrasound (US) in combination with microbubbles has previously shown promise as a means to improve drug delivery, and the therapeutic efficacy of ultrasound-mediated drug delivery is currently being evaluated in multiple clinical trials. However, most of these utilize echogenic contrast agents engineered for imaging, which might not be optimal compared to specialized formulations tailored for drug delivery. In this study, we evaluated the in vivo efficacy of phase-shifting microbubble-microdroplet clusters that, upon insonation, form bubbles in the size range of 20–30 μm. We developed a patient-derived xenograft model of pancreatic cancer implanted in mice that largely retained the stromal content of the originating tumor and compared tumor growth in mice given chemotherapeutics (nab-paclitaxel plus gemcitabine or liposomal irinotecan) with mice given the same chemotherapeutics in addition to ultrasound and acoustic cluster therapy. We found that acoustic cluster therapy significantly improved the effect of both chemotherapeutic regimens and resulted in 7.2 times higher odds of complete remission of the tumor compared to the chemotherapeutics alone.

This exegesis and the accompanying folio of original compositions explores what it means to create contemporary art music in the modern context through an inverstigation of two major themes: (1) Metamodernism and (2) Wabi Sabi aesthetics. The folio responds to these two culturally and aesthetically disparate notions of art-making as a metaxic pair, exploring them both individually as well as considering their dialectical tension.The exegesis additionally considers a broad range of compositional issues including aesthetics, musical meaning-making, personal influences, psychology and praxis, and their relation to the folio works.

A hallmark of sarcomas is genetic, biologic, and clinical heterogeneity, which is a challenge for preclinical development of therapeutic targets, clinical trial design, and patient care. Because sarcomas comprise over 100 subtypes, a broad comparative genetic analysis of subtypes has been lacking and sarcoma genetic studies have by necessity focused on one or a few subtypes. We will report the results of a comprehensive analysis of somatic genetic alterations detected by a targeted sequencing platform in a single institution cohort of 2,138 patient-specific samples of soft tissue and bone sarcoma representing 45 distinct pathological entities. We determined subtype-specific mutation and copy number (arm and gene level) events with a focus on pathways in which there are therapeutic targets under clinical or preclinical investigation. The most commonly altered pathways are cell cycle control, TP53, PI3K, receptor tyrosine kinase/RAS, and epigenetic regulators (e.g. >40% of uterine leiomyosarcoma [n=165], undifferentiated pleomorphic sarcoma [UPS, n=145] and pleomorphic liposarcoma [n=22]). Subtype-specific associations in rare subtypes include TERT amplification in 41% of intimal sarcoma (n=18) and alterations in chromatin remodeling and histone modifying complexes in uterine adenosarcoma (43% and 36%, respectively; n=14). Epigenetic pathway alterations in common sarcomas were also noted, including an increased frequency of oncogenic alterations in epigenetic regulators in dedifferentiated liposarcoma (DDLS; n=167) (25%) vs. well differentiated liposarcoma (8%; n=48). We evaluated rates of whole genome doubling (WGD), which in subtypes such as UPS and osteosarcoma (n=129) were >45%, which is more frequent than many carcinomas. WGD associated with worse overall survival in metastatic (p=0.042) but not primary samples (p=0.391). Among specific subtypes, WGD was associated with worse overall survival in metastatic UPS (p=0.022). Unsupervised clustering of samples based on genetic features revealed that in some cases sarcomas are more similar to individual tumors of distinct histology than to other sarcomas of the same subtype. For example, UPS and soft tissue leiomyosarcoma (n=125) segregated into multiple genetic clusters (high entropy) compared to others with low entropy (DDLS), which could have implications for future clinical trial interpretation and/or design. We also determined the subtype-specific rate of tumor mutation burden, which was overall low compared to many carcinomas, but notably varied between and within subtypes, with some having a long tail in the upper range of the distribution. Lastly, we assessed targetable alterations on a subtype-specific basis. Together with other ‘multiomic’ approaches, we anticipate this work will motivate preclinical studies of subtype-specific sarcoma biology and potential therapeutic targets and will inform efforts to interpret outcomes in sarcoma clinical trials with respect to underlying genetic subtypes. PREMISE Plants rely on pools of internal nonstructural carbohydrates (NSCs: soluble sugars plus starch) to support metabolism, growth, and regrowth of tissues damaged from disturbance such as foliage herbivory. However, impacts of foliage herbivory on the quantity and composition of NSC pools in long-lived woody plants are currently unclear. We implemented a controlled defoliation experiment on mature Tamarix spp.-a dominant riparian woody shrub/tree that has evolved with intense herbivory pressure-to test two interrelated hypotheses: (1) Repeated defoliation disproportionately impacts aboveground versus belowground NSC storage. (2) Defoliation disproportionately impacts starch versus soluble sugar storage. METHODS Hypotheses were tested by transplanting six Tamarix seedlings into each of eight cylinder mesocosms (2 m diameter, 1 m in depth). After 2.5 years, plants in four of the eight mesocosms were mechanically defoliated repeatedly over a single growing season, and all plants were harvested in the following spring. RESULTS Defoliation had no impact on either above- or belowground soluble sugar pools. However, starch in defoliated plants dropped to 55% and 26% in stems and roots, respectively, relative to control plants, resulting in an over 2-fold higher soluble sugar to starch ratio in defoliated plants. CONCLUSIONS The results suggest that defoliation occurring over a single growing season does not impact immediate plant functions such as osmoregulation, but depleted starch could limit future fitness, particularly where defoliation occurs over multiple years. These results improve our understanding of how woody plants cope with episodic defoliation caused by foliage herbivory and other disturbances.

This chapter discusses aspects of the historical, social, and religious environment in which Islam emerged, especially the Christian and the Arabian ‘pagan’ environment (deities worshipped, including consideration of cults in which images of gods were avoided, rituals observed in relation to temples and pilgrimage, the use of divination to seek the will of the gods or to foretell the future, and some major theological themes, such as monotheism). The Conclusions attempt to draw this evidence together and suggest that Islam incorporated elements from its earliest religious environment in northern Arabia and Bilād ash-Shām in the expression of its own unique message. It argues that Islam inevitably drew on the surrounding social and religious traditions while giving them a new meaning in a new context.

Buying behavior in the video game market can be conceptualized as a two-stage process where users first purchase a console and then purchase content for that console. As a result, the sales of consoles and content in this market are interdependent and together form a platform-mediated market. Established research on platform-mediated markets has highlighted the effects of installed base size (or the number of consumer who have adopted the platform) on content sales. In other words, how does the number of console users impact sales of content designed for that platform? Here, we extend the research on installed base effects on content sales by examining two characteristics of installed bases: installed base innovativeness and recency. Installed base innovativeness is defined by the proportion of the installed base that adopted the platform early in the platform product’s lifecycle. Installed base recency is defined as the proportion of the installed base that adopted the platform in recent weeks. We find that installed bases with higher innovativeness or higher recency purchase more content than less innovative or lower recency installed bases. We also find that installed base recency has a larger positive effect on content sales when an installed base is less innovative. These results suggest that content sales depend on more than just installed base size. In fact, there may be opportunities to increase content sales by releasing new content not just when the installed base is at its largest, but also when the installed base is considered more innovative and recent.

This thesis identifies prototypical features in reality television subgenres that lead to particular ethical problems. Through the ethical analysis of cases studies from three subgenres of reality television, I also demonstrate the relative value of various normative ethical frameworks in analysing ethical problems in reality programming. I also argue that popular, non-academic criticisms of reality television act as implicit ethical judgements that align with formal criticisms from normative ethics.My first chapter addresses the ethical problems of informed consent, agency and representation in Extreme Guide to Parenting (Bravo 2014) and Toddlers & Tiaras (TLC 2008–13), two docu-soap programs that feature children. I find that a Kantian deontological perspective largely disapproves of children’s involvement in reality television, and these objections are echoed by similar arguments from documentary ethics. A perspective from the utilitarian arguments of Jeremy Bentham and John Stuart Mill, conversely, may in some cases approve or disapprove of children’s participation in docu-soaps, depending on whether the production of these programs is seen to generally contribute to wider happiness. A utilitarian evaluation of the ethical goods and harms of docu-soaps production and reception has parallels with debates in documentary ethics over the function of documentary as a general public good.In my second chapter, I reformulate popular journalistic criticisms of makeover programs Snog Marry Avoid? (BBC Three 2008–13) and The Biggest Loser Australia (Network Ten 2006–) along explicitly ethical lines via Kantian deontology and Julia Driver’s hybrid of virtue and consequentialist ethics. I draw on theoretical perspectives regarding self-transformation and therapy culture in order to evaluate the moral and practical status of makeover programs’ humiliation of subjects and their stated therapeutic projects. I argue that the individual subjects of these programs are deployed to reinforce dominant discourses regarding class, gender, national identity and neoliberal self-governance in the United Kingdom and Australia.My third chapter studies three reflexive parodies of reality television, UnREAL (Lifetime 2015–), Sex House (Onion Digital Studios 2012), and Nathan for You (Comedy Central 2013–), to consider how and why these programs comment on perceived ethical problems in reality television and the format’s status in wider culture. I refer to theories of parody, satire, and mockumentary programming, and argue that my case studies use shocking depictions of reality television production in order to encourage audience scrutiny of their referent reality programs. Similarly to mockumentary programs, these case studies challenge reality programming’s ability to present factuality, and emphasise differences of power between reality producer, subject, and audience to critique the form.In this thesis, I argue that particular ethical issues in reality television reflect ideas and strategies embedded in reality television programming at a subgeneric level. I demonstrate the validity of Kantian deontology, utilitarian consequentialism and a hybrid consequentialist-virtue ethics framework as a means of identifying and assessing the validity of various ethical criticisms of reality television. These ethical criticisms, I argue, also circulate in popular culture, albeit at an implicit level. This study demonstrates the connection between the ethical study of reality television and the cultural and historical influences that characterise contemporary reality television.

Diorhabda carinulata (northern tamarisk beetle) has been released in the western United States as a biological control agent for the invasive plant Tamarix spp. (tamarisk). A few studies have been conducted analysing the effects of beetle defoliation on tamarisk water consumption, but predefoliation and post-defoliation comparison based on field data is scarce. The question of whether beetles substantially alter tamarisk water consumption is still open for discussion. In this study, an eddy covariance station and groundwater monitoring well were installed in a tamarisk stand along the Virgin River near Mesquite, NV, in 2010 to observe the impacts of tamarisk defoliation on evapotranspiration (ET) and groundwater levels. ET was calculated using the eddy covariance method and the White method and supported by Landsat remote sensing estimates. Data collected in 2010 established a baseline since the beetles arrived at the site in late 2010 and started feeding on foliage causing tamarisk trees to turn to brown during 2011. Repeated defoliations in 2011 and 2012 show that post-defoliation ET values and magnitude of diurnal groundwater-level fluctuations decreased compared with the predefoliation values. Estimated annual ET and annual average daily groundwater fluctuations were reduced by 18% and 35%, respectively. However, the magnitude of effects of defoliation was dependent on the growth stage of tamarisk at the time of defoliation. Also, ET recovered within a month as tamarisk established new leaves. Results from this study suggest that long-term changes in ET are highly dependent on repeated defoliation occurrences over several years. Copyright © 2015 John Wiley & Sons, Ltd.
