This study focuses on investigating a potential method for depositing zinc sulfide (ZnS), in thin film form at room temperature. A facile and effective approach was employed in this study, wherein a suspension of ZnS powder in methanol was applied onto flexible plastic substrates using a spray deposition method. Structural analysis of the resulting films revealed a broad X-ray diffraction peak along the (100) plane, indicating the presence of nanoscale crystallites. The band gap of the ZnS films was determined to be 3.78 eV. Furthermore, the films exhibited photo luminescence emission centered at a wavelength of 410 nm, which corresponds to the blue edge of the chromaticity diagram. [ABSTRACT FROM AUTHOR]

National and international policies and treaties require the protection and exploration of the Arctic. The maritime services play a primary role in pursuing responsible Arctic stewardship by protecting the environment and the personnel conducting operations and research in this harsh environment. The National Oceanic and Atmospheric Administration (NOAA) is an important partner to the U.S. Coast Guard (USCG) in hazard response and mitigation (including oil spills and search and rescue). During Arctic Shield exercises, as part of the USCG Research and Development Center's Arctic Technology Evaluation, manned and unmanned systems including the AeroVironment Puma™All Environment (AE) (Puma) Unmanned Aircraft System (UAS), were used to provide real-time information for maritime domain awareness and oil spill response in the Arctic. Real-time information distribution and maritime domain awareness are critical to prepare for and respond to potential environmental disasters in the Arctic. Additionally, the Puma was assessed for shipboard operations capabilities, Arctic air space coordination, deconfliction and safety issues, and real-time data visualization through the Arctic Environmental Response Management Application® as part of a larger datamanagement plan. The results are provided from the successful Puma testing during the Arctic Shield 2013 and 2014 exercises aboard the USCG Cutter (USCGC; Icebreaker) Healy. An overview of these operations is given with recommendations for future testing and technology assessments of small UAS platforms for Arctic shipboard operational deployments. Emerging evidence suggested a converging mechanism in neurodegenerative brain diseases (NBD) involving early neuronal network dysfunctions and alterations in the homeostasis of neuronal firing as culprits of neurodegeneration. In this study, we used paired-end short-read and direct long-read whole genome sequencing to investigate an unresolved autosomal dominant dementia family significantly linked to 7q36. We identified and validated a chromosomal inversion of ca. 4 Mb, segregating on the disease haplotype and disrupting the coding sequence of dipeptidyl-peptidase 6 gene (DPP6). DPP6 resequencing identified significantly more rare variants-nonsense, frameshift, and missense-in early-onset Alzheimer's disease (EOAD, p value = 0.03, OR = 2.21 95% CI 1.05-4.82) and frontotemporal dementia (FTD, p = 0.006, OR = 2.59, 95% CI 1.28-5.49) patient cohorts. DPP6 is a type II transmembrane protein with a highly structured extracellular domain and is mainly expressed in brain, where it binds to the potassium channel K v 4.2 enhancing its expression, regulating its gating properties and controlling the dendritic excitability of hippocampal neurons. Using in vitro modeling, we showed that the missense variants found in patients destabilize DPP6 and reduce its membrane expression (p < 0.001 and p < 0.0001) leading to a loss of protein. Reduced DPP6 and/or K v 4.2 expression was also detected in brain tissue of missense variant carriers. Loss of DPP6 is known to cause neuronal hyperexcitability and behavioral alterations in Dpp6-KO mice. Taken together, the results of our genomic, genetic, expression and modeling analyses, provided direct evidence supporting the involvement of DPP6 loss in dementia. We propose that loss of function variants have a higher penetrance and disease impact, whereas the missense variants have a variable risk contribution to disease that can vary from high to low penetrance. Our findings of DPP6, as novel gene in dementia, strengthen the involvement of neuronal hyperexcitability and alteration in the homeostasis of neuronal firing as a disease mechanism to further investigate.

The fundamental test for male infertility, semen analysis, is mostly a manually performed subjective and time-consuming process and the use of automated systems has been cost prohibitive. We have previously developed an inexpensive smartphone-based system for at-home male infertility screening through automatic and rapid measurement of sperm concentration and motility. Here, we assessed the feasibility of using a similar smartphone-based system for laboratory use in measuring: a) Hyaluronan Binding Assay (HBA) score, a quantitative score describing the sperm maturity and fertilization potential in a semen sample, b) sperm viability, which assesses sperm membrane integrity, and c) sperm DNA fragmentation that assesses the degree of DNA damage. There was good correlation between the manual analysis and smartphone-based analysis for the HBA score when the device was tested with 31 fresh, unprocessed human semen samples. The smartphone-based approach performed with an accuracy of 87% in sperm classification when the HBA score was set at manufacturer's threshold of 80. Similarly, the sperm viability and DNA fragmentation tests were also shown to be compatible with the smartphone-based system when tested with 102 and 47 human semen samples, respectively., Competing Interests: The authors have declared that no competing interests exist.

The technology for building functionally complete or 'whole-cell' biological simulations is rapidly developing. However, the predictive capabilities of these simulations are hindered by the availability of parameter values, which are often difficult or even impossible to obtain experimentally and must therefore be estimated. Using E. coli's glycolytic network as a model system, we describe and apply a new method which can estimate the values of all the system's 102 parameters - fit to observations from studies of proteomics, metabolomics, enzyme kinetics and chemical energetics - and find that the resulting metabolic models are not only well-fit, but also dynamically stable. An analysis of how well parameter values in the network were determined by the training data revealed that over 80% of the parameter values were not well-specified. Moreover, the distribution of well-determined values was biased to a specific part of the network and against certain types of experimental data. Our results also suggest that perturbing the functional, energetic space of parameters (rather than traditional metabolic parameters) is a superior strategy for exploring the space of biological dynamics. The estimated parameter values matched both training data and previously withheld validation data within an order of magnitude for over 85% of the data points; notably, the area of greatest frustration in the network was also the most fully determined. Finally, our estimation method showed that fidelity to physiological observations such as network response time is enforced at the cost of fit to molecular parameter values. In summary, our reformulation enables estimation of accurate, biologically relevant parameters, generates insight into the biology of the simulated network, and appears generalizable to any biochemical network - potentially including whole-cell models., (Copyright © 2018 The Authors. The ability to accurately predict ovulation at-home using low-cost point-of-care diagnostics can be of significant help for couples who prefer natural family planning. Detecting ovulation-specific hormones in urine samples and monitoring basal body temperature are the current commonly home-based methods used for ovulation detection; however, these methods, relatively, are expensive for prolonged use and the results are difficult to comprehend. Here, we report a smartphone-based point-of-care device for automated ovulation testing using artificial intelligence (AI) by detecting fern patterns in a small volume (<100 μL) of saliva that is air-dried on a microfluidic device. We evaluated the performance of the device using artificial saliva and human saliva samples and observed that the device showed >99% accuracy in effectively predicting ovulation.

Lack of accurate dietary assessment in free-living populations requires discovery of new biomarkers reflecting food intake qualitatively and quantitatively to objectively evaluate effects of diet on health. We provide a proof-of-principle for an analytical pipeline to identify quantitative dietary biomarkers. Tartaric acid was identified by nuclear magnetic resonance spectroscopy as a dose-responsive urinary biomarker of grape intake and subsequently quantified in volunteers following a series of 4-day dietary interventions incorporating 0 g/day, 50 g/day, 100 g/day, and 150 g/day of grapes in standardized diets from a randomized controlled clinical trial. Most accurate quantitative predictions of grape intake were obtained in 24 h urine samples which have the strongest linear relationship between grape intake and tartaric acid excretion (r(2) = 0.90). This new methodological pipeline for estimating nutritional intake based on coupling dietary intake information and quantified nutritional biomarkers was developed and validated in a controlled dietary intervention study, showing that this approach can improve the accuracy of estimating nutritional intakes.

Annually laminated stalagmites can be used to construct a precise chronology, and variations in laminae thickness provide an annual growth-rate record that can be used as a proxy for past climate and environmental change. Here, we present and analyse the first composite speleothem annual growth-rate record based on five stalagmites from the same cave system in northwest Scotland, where precipitation is sensitive to North Atlantic climate variability and the winter North Atlantic Oscillation (NAO). Our 3000-year record confirms persistently low growth-rates, reflective of positive NAO states, during the Medieval Climate Anomaly (MCA). Another persistently low growth period occurring at 290-550 CE coincides with the European Migration Period, and a subsequent period of sustained fast growth-rate (negative NAO) from 600-900 AD provides the climate context for the Viking Age in northern and western Europe.

Amyloid-β (Aβ) protein causes neurotoxicity and its abnormal aggregation into amyloid is a pathological hallmark of Alzheimer's disease (AD). Cellular proteins able to interact with Aβ or its precursor, AβPP (amyloid-β protein precursor), may regulate Aβ production and neurotoxicity. We identified a brain-enriched type I transmembrane protein, tomoregulin (TR), that directly binds Aβ and Aβ oligomers (AβO). TR co-immunoprecipitated with Aβ and AβO in cultured cells and co-localized with amyloid plaques and intraneuronal Aβ in the 5xFAD AD mouse model. TR was also enriched in astrocytic processes reactive to amyloid plaques. Surface plasmon resonance spectroscopy studies showed that the extracellular domain of TR binds to AβO with a high affinity (KD = 76.8 nM). Electron paramagnetic resonance spectroscopy also demonstrated a physical interaction between spin-labeled Aβ and the TR extracellular domain in solution. Furthermore, TR also interacted with AβPP and enhanced its cleavage by α-secretase. Both cellular expression of TR and application of recombinant TR extracellular domain protected N2a neurons from AβO-induced neuronal death. These data provide first evidence that neuronal and astrocytic expression of TR is intimately related to Aβ metabolism and toxicity, and could be neuroprotective through its direct interaction with Aβ and AβPP.
