Dependency ofSolanaceaebut notA. thalianaEDS1 on catalytic triad residues raises the possibility that a TNL-derived small molecule binds to theSolanaceaeEDS1 lipase-like domain, and that EDS1 lipase-like domain pocket contributions to TNL immune responses vary between lineages. Whether and how nuclear EDS1 activity connects to membrane pore-forming RNLs remains unknown.

The evaluation of environmental aspects in the early planning phase of buildings can support the reduction of the resource use and environmental impacts associated with the building sector over the whole building life-cycle. The integration of life cycle assessment (LCA) benchmarks in the planning phase is one potential measure. To derive these benchmarks a large database of existing building assessment is essential. Potential data input is available from the German Sustainable Building Council (DGNB), as it certifies more than 200 buildings annually and the certification includes a mandatory LCA. In this study, the current submission files and database of the DGNB are assessed and critically reviewed with regard to their usability for automated LCA benchmarks. First, a harmonized database is created from the large number of assessed buildings. Second, the data is examined for its suitability for benchmarking with regard to data format, structure and level of detail. The data that were declared fit for purpose were used to create an exemplary, harmonized data set with 22 office buildings. The evaluation of these data for various environmental indicators of the individual life-cycle phases shows their respective relevance and can thus serve as a benchmark. Another focus is to encourage improvement of the additional documentation like the energy source required for better benchmarking, interpretation of results and auditing of the LCA rules for building certification. The results of this study highlight the opportunities and challenges in the development of a database for benchmarking. Before long-term LCA benchmarks can be developed and deployed, a standardized and uniform submission format of results, that is indifferent regarding the used LCA software, needs to be developed. In the future the submission process should be extended by an automated quality assurance to prevent restraints from low data quality and data gaps that otherwise have to be detected manually.

Existing buildings can reach a performance enhancement and extend their nominal service life through renovation measures such as seismic rehabilitation. In particular, when buildings have almost exhausted their service life, seeking an optimal solution should consider whether costs and environmental effects are worthwhile, or new construction is preferred. In this paper, a methodology to consider seismic hazard into probabilistic approaches for life-cycle analyses is presented considering the possibility of structural enhancement over an extended building lifespan. A life-cycle-based decision support tool for building renovation measures is developed and applied to a selected case study. Unlike standard “static” analyses, which in this work show shortcomings by underestimating impacts of vulnerable buildings, such an approach brings out environmental and economic advantages of retrofit measures designed to improve the structural performance.

Building Information Modelling (BIM) supports construction processes by dealing with the variety and complexity of design in a single virtual model. The model may also be complemented by the static and energy performance of buildings. Facing the growing demand of sustainability strategies in the construction sector, the consideration of environmental information within the planning process influences the decision making of planners and stakeholders. Nevertheless, the life cycle assessment of buildings has been so far excluded in BIM, due to the high variety of accurate information and time required. In this paper, a systematic framework is presented and applied to a case study. BIM-LCA assists actors along the planning and designing phase, from the building conception as a whole, up to the elements’ details and materials’ definition. BIM and LCA intertwine in an application scheme of seven phases for integral planning and four levels of structural composition of a building. With respect to these, involved actors examine potential solutions through a tool which exploits alternative specifications in order to assess the environmental impacts. The goal of this paper is to demonstrate the application of a BIM-LCA model regarding decision making for reliable values of environmental impact in a given structural level of the building. The main findings of this framework are due to the multitude of actors and information orchestrated, namely to uncertainties which characterize the whole planning process and data handling. Through BIMLCA, actors are assisted by ensuring flexibility of models and consistency of results throughout planning and designing.

Summary Genome editing facilitated by Cas9-based RNA-guided nucleases (RGNs) is becoming an increasingly important and popular technique for reverse genetics in both model and non-model species. So far, RGNs were mainly applied for the induction of point mutations, and one major challenge consists in the detection of genome-edited individuals from a mutagenized population. Also, point mutations are not appropriate for functional dissection of non-coding DNA. Here, the multiplexing capacity of a newly developed genome editing toolkit was exploited for the induction of inheritable chromosomal deletions at six different loci in Nicotiana benthamiana and Arabidopsis. Globalization leads to an increase in goods transported by ships. Ships are scrapped once they have reached the end of their use. Over its lifetime a ship generates added value, which benefits the owner, and at the same time harms the environment through emissions. A certain imbalance in the distribution of the added value and the harm to the environment can be observed over the lifetime of a ship. The goal of this study is to depict and quantify this imbalance and give an evaluation indicator in order to transparently describe this issue. Methods used in this study are Life Cycle Assessment (LCA), for the quantification of the environmental impacts, local added value, for the quantification of economic impact and the method of eco-efficiency. A literature review was conducted in order to estimate the added value per life cycle phase. Finally these two factors were put in relation and the adjusted eco-efficiency indicator was developed. As result it can be seen, that especially the European owners of the ship during its use phase benefit the most, whereas the Asian producers and dismantlers of the ship have to suffer comparatively more environmental impacts per unit of added value.

Technological advancement culminating in a globalized economy has brought tremendous improvements for mankind in manifold respects but comes at the cost of alienation from nature. Human activities nowadays are unsustainable and cause severe damage especially in terms of global depletion and destabilization of natural systems but also harm its own social resources. In this paper, a sustainability assessment method is developed based on a bio-inspired sustainability framework that has been developed in the project TRR 141-C01 “The biomimetic promise.” It is aims at regaining the advantages of societal embeddedness in its environment through biological inspiration. The method is developed using a structured approach including requirement specification, description of the inventory models on bio-inspiration and sustainability assessment, creation of a bio-inspired sustainability assessment model and its validation. It is defined as an accompanying assessment for decision support, using a six-fold two-dimensional structure of social, economic and environmental functions and burdens. The method is applied and validated in 6 projects of TRR 141 and its applicability is exemplarily shown by the assessment of “Bio-flexi”, a biobased and biodegradable natural fiber reinforced plastic composite for indoor cladding applications. Based on the findings of the application the assessment method itself is proposed to be advanced towards an adaptive structure and a consequent outlook is provided.

Standardized DNA assembly strategies facilitate the generation of multigene constructs from collections of building blocks in plant synthetic biology. A common syntax for hierarchical DNA assembly following the Golden Gate principle employing Type IIs restriction endonucleases was recently developed, and underlies the Modular Cloning and GoldenBraid systems. In these systems, transcriptional units and/or multigene constructs are assembled from libraries of standardized building blocks, also referred to as phytobricks, in several hierarchical levels and by iterative Golden Gate reactions. This combinatorial assembly strategy meets the increasingly complex demands in biotechnology and bioengineering, and also represents a cost-efficient and versatile alternative to previous molecular cloning techniques. For Modular Cloning, a collection of commonly used Plant Parts was previously released together with the Modular Cloning toolkit itself, which largely facilitated the adoption of this cloning system in the research community. Here, a collection of approximately 80 additional phytobricks is provided. These phytobricks comprise e.g. modules for inducible expression systems, different promoters or epitope tags, which will increase the versatility of Modular Cloning-based DNA assemblies. Furthermore, first instances of a “peripheral infrastructure” around Modular Cloning are presented: While available toolkits are designed for the assembly of plant transformation constructs, vectors were created to also use coding sequence-containing phytobricks directly in yeast two hybrid interaction or bacterial infection assays. Additionally, DNA modules and assembly strategies for connecting Modular Cloning with Gateway Cloning are presented, which may serve as an interface between available resources and newly adopted hierarchical assembly strategies. The presented material will be provided as a toolkit to the plant research community and will further enhance the usefulness and versatility of Modular Cloning.

Over the last few decades, the systematic approach of knowledge transfer from biological concept generators to technical applications has received increasing attention, particularly because marketable bio-derived developments are often described as sustainable. The objective of this paper is to rationalize and refine the discussion about bio-derived developments also with respect to sustainability by taking descriptive, normative and emotional aspects into consideration. In the framework of supervised learning, a dataset of 70 biology-derived and technology-derived developments characterised by 9 different attributes together with their respective values and assigned to one of 17 classes was created. On the basis of the dataset a decision tree was generated which can be used as a straightforward classification tool to identify biology-derived and technology-derived developments. The validation of the applied learning procedure achieved an average accuracy of 90.0%. Additional extraordinary qualities of technical applications are generally discussed by means of selected biology-derived and technology-derived examples with reference to normative (contribution to sustainability) and emotional aspects (aesthetics and symbolic character). In the context of a case study from the building sector, all aspects are critically discussed.

The EU building stock is relatively old with 40% of it built before 1960. In Albania the building sector accounts for 26, 9% of final energy consumption, offering high energy saving potentials due to the great number of old residential buildings. Intervention is not always possible and, in order to achieve significant environmental savings, the national action plans cannot rely only on the physical improvement of existing buildings. This paper proposes a probabilistic LCA and LCC evaluation model using MC simulation, for the prediction of intervention options in existing buildings. The potential environmental and economic impacts of three intervention options: standard, ambitious retrofitting and new construction during the whole life cycle of a building are analysed. A framework is defined, with the purpose of estimating the value of a building in a specific time during its life cycle. Comparing the generated values of potential environmental impacts and associating them with the changes on the buildings value enables the process of deciding upon the most desirable and/or agreed combination. The results of the SLED Study 2015 on Albanian building typology are used, while the new construction model is defined according to German EnEV2014 requirements. The GWP values from the LCA/LCC assessment of the intervention scenarios, done through the SBS tool of Fraunhofer IBP, are used to create a prediction model for future alternative solutions especially in early planning phase. Decision-making through this model can encourage a sustainability strategy for energy efficiency improvement in the building sector of Albania.

Because of the tremendous challenges of the impacts caused by the globally growing economy, the targeted development of sustainable innovation is an inevitable social responsibility. Despite some advances, however, sustainability has not yet been integrated into product development on a broad scale. Although bio-inspired innovations seem to offer solutions, the transfer of sustainability through the bio-inspiration process is only conducted implicitly and the possible fulfilment of the ‘promise of bio-inspiration’ is only assessed retrospectively.

This paper represents the first stage of extending the scope of LCA to space and is intended as a discussion starter. Based on the assumption, that the future and outlast of humanity lies within the exploration and colonisation of space, the LCA methodology as of today, is discussed with regards to its capabilities to cover the impact of human activities in space. Based on this assessment, ideas whether and how LCA can be extended are outlined. Initially, an understanding of additional environmental impacts which occur in space compared to Earth is built up by the means of literature research. The state of the art of space regulations and availability of LCAs in space and for astronautics is clarified as well. Further literature research was conducted on the LCA subtopic of regionalization. Based on this and assumptions regarding future space travel, the suitability of LCA as an assessment method is validated. Afterwards, different potential development phases of LCA towards its applicability in space are defined. For activities in space, the regarded environmental impacts have to be expanded (e.g. space debris, extra-terrestrial life toxicity, etc.). Space regulations, if in place, cover only impacts of space activities on Earth so far. LCAs for space activities are not widespread yet. One reason for this is that the state of the art LCA methodology has not been expanded and existing regionalisation approaches are not easily transferable to space. Critical issues are faced in all phases of an LCA and include widening of boundaries, definition of space regions, finding suitable reference units and ethical problems. As a result, four LCA development phases are suggested: Earth-bound, solar system-bound, transition phase and intergalactic. Each phase involves different activities and goals, which result in different system boundaries and impact categories and widen the scope of LCA subsequently. It is a long way for humanity to populate space and so, it is for enabling LCA to assess these activities. The methodology of LCA is flexible and capable to make this adaptation. This paper can be seen as a starting point of a discussion opening up many questions. Some of these questions can only be answered in the future with more certainty about the development of space colonialization.

Purpose The objective of the paper is to discuss the role of a new guidance document for life cycle assessment (LCA) in the construction sector available as an online InfoHub.\ud \ud Methods This InfoHub derives from the EeBGuide European project that aimed at developing a guidance document for energy-efficient building LCA studies. The InfoHub is built on reference documents such as the ISO 14040-44 standards, the EN 15804 and EN 15978 standards as well as the ILCD Handbook. The guidance document was filled with expertise and knowledge of several experts. The focus was put on providing scientifically sound, yet practical guidance.\ud \ud Results The EeBGuide InfoHub is an online guidance document, setting rules for conducting LCA studies and giving instructions on how to do this. The document has a section on buildings—new and existing—and a section on construction products. It is structured according to the life cycle stages of the European standards EN 15804 and EN 15978, covering all aspects of LCA studies by applying provisions from these standards and the ILCD handbook, wherever applicable. The guidance is presented for different scopes of studies by means of three study types. For the same system boundaries, default values are proposed in early or quick assessment (screening and simplified LCA) while detailed calculation rules correspond to a complete LCA. Such approach is intended to better match the user needs in the building sector.\ud \ud Conclusions and recommendations This paper can be viewed as a contribution to the ongoing efforts to improve the consistency and harmonisation in LCA studies for building products and buildings. Further contributions are now needed to improve building LCA guidance and to strengthen links between research, standardisation and implementation of LCA in the construction practice.
