Goal: Parkinson's disease (PD) can lead to gait impairment and Freezing of Gait (FoG). Recent advances in cueing technologies have enhanced mobility in PD patients. While sensor technology and machine learning offer real-time detection for on-demand cueing, existing systems are limited by the usage of smartphones between the sensor(s) and cueing device(s) for data processing. By avoiding this we aim at improving usability, robustness, and detection delay. Methods: We present a new technical solution, that runs detection and cueing algorithms directly on the sensing and cueing devices, bypassing the smartphone. This solution relies on edge computing on the devices' hardware. The wearable system consists of a single inertial sensor to control a stimulator and enables machine-learning-based FoG detection by classifying foot motion phases as either normal or FoG-affected. We demonstrate the system's functionality and safety during on-demand gait-synchronous electrical cueing in two patients, performing freezing of gait assessments. As references, motion phases and FoG episodes have been video-annotated. Results: The analysis confirms adequate gait phase and FoG detection performance. The mobility assistant detected foot motions with a rate above 94 % and classified them with an accuracy of 84 % into normal or FoG-affected. The FoG detection delay is mainly defined by the foot-motion duration, which is below the delay in existing sliding-window approaches. Conclusions: Direct computing on the sensor and cueing devices ensures robust detection of FoG-affected motions for on demand cueing synchronized with the gait. The proposed solution can be easily adopted to other sensor and cueing modalities.

Abstract Key message Paternity analyses show that effective pollination of ash (Fraxinus excelsior L.) in a seed orchard and a floodplain forest affected by ash dieback is more likely to be facilitated by healthier males. Thereby, natural selection can have a positive effect on the health of future generations. Context Ongoing ash dieback and increasing fragmentation of ash populations may result in reduced pollen flow, which can reduce pollination success of future generations of ash trees. Therefore, it is essential to further improve our understanding of gene flow patterns, especially with respect to ash dieback. Aims In this study, paternity analyses were conducted in a seed orchard and a floodplain forest in Germany in 2018 to explain the relationship between pollination success and the health status of ash trees and distances of effective pollen transport. Methods Cambium samples (i.e., from twigs and stumps) were collected from 251 ash trees (putative father and mother trees) for genotyping, and the health status of each tree was documented using a scoring system to evaluate vitality. Additionally, seeds were harvested from 12 mother trees per site. Genetic analyses using nuclear microsatellites were performed to determine paternal trees. Paternities were assigned based on the likelihood model implemented in the Cervus 3.0.7 software. Results Our results showed that the average pollination distance was 76 m in the seed orchard and 166 m in the floodplain forest. In general, pollination success decreased substantially with increasing distance to the mother tree. Despite the dense tree cover in the floodplain forest, pollen were transported over long distances (greater than 550 m), suggesting that non-local sources also play a role in pollination. This is supported by the foreign pollen input identified in the seed orchard (66.5%). Self-pollination was detected only to a very small extent, and thus had no major influence on reproduction. In addition, both healthy and slightly diseased father trees showed similar mating success. However, this was not the case for the severely diseased ash trees (more than 50% of crown damage) because only a few offspring could be assigned to them. Nevertheless, in contrast to the floodplain forest, there was no significant correlation between damage classes and pollination success in the seed orchard. Conclusion Long-distance pollen transport contributes to the connectivity of ash trees in the landscape. Additionally, both healthy and slightly diseased fathers have a greater contribution to pollination, thus potentially improving the health of the next generation of ash trees. Moreover, gene flow between stepping stone populations is necessary to ensure the positive impact on the genetic diversity of ash populations in the future.

Abstract Mutations in glucocerebrosidase cause the lysosomal storage disorder Gaucher’s disease and are the most common risk factor for Parkinson’s disease. Therapies to restore the enzyme’s function in the brain hold great promise for treating the neurological implications. Thus, we developed blood-brain barrier penetrant therapeutic molecules by fusing transferrin receptor-binding moieties to β-glucocerebrosidase (referred to as GCase-BS). We demonstrate that these fusion proteins show significantly increased uptake and lysosomal efficiency compared to the enzyme alone. In a cellular disease model, GCase-BS rapidly rescues the lysosomal proteome and lipid accumulations beyond known substrates. In a mouse disease model, intravenous injection of GCase-BS leads to a sustained reduction of glucosylsphingosine and can lower neurofilament-light chain plasma levels. Collectively, these findings demonstrate the potential of GCase-BS for treating GBA1-associated lysosomal dysfunction, provide insight into candidate biomarkers, and may ultimately open a promising treatment paradigm for lysosomal storage diseases extending beyond the central nervous system.

The understanding of locomotion in neurological disorders requires technologies for quantitative gait analysis. Numerous modalities are available today to objectively capture spatiotemporal gait and postural control features. Nevertheless, many obstacles prevent the application of these technologies to their full potential in neurological research and especially clinical practice. These include the required expert knowledge, time for data collection, and missing standards for data analysis and reporting. Here, we provide a technological review of wearable and vision-based portable motion analysis tools that emerged in the last decade with recent applications in neurological disorders such as Parkinson's disease and Multiple Sclerosis. The goal is to enable the reader to understand the available technologies with their individual strengths and limitations in order to make an informed decision for own investigations and clinical applications. We foresee that ongoing developments toward user-friendly automated devices will allow for closed-loop applications, long-term monitoring, and telemedical consulting in real-life environments.

Parkinson's disease is the second most common neurodegenerative disease worldwide reducing cognitive and motoric abilities of affected persons. Freezing of Gait (FoG) is one of the severe symptoms that is observed in the late stages of the disease and considerably impairs the mobility of the person and raises the risk of falls. Due to the pathology and heterogeneity of the Parkinsonian gait cycle, especially in the case of freezing episodes, the detection of the gait phases with wearables is challenging in Parkinson's disease. This is addressed by introducing a state-automaton-based algorithm for the detection of the foot's motion phases using a shoe-placed inertial sensor. Machine-learning-based methods are investigated to classify the actual motion phase as normal or FoG-affected and to predict the outcome for the next motion phase. For this purpose, spatio-temporal gait and signal parameters are determined from the segmented movement phases. In this context, inertial sensor fusion is applied to the foot's 3D acceleration and rate of turn. Support Vector Machine (SVM) and AdaBoost classifiers have been trained on the data of 16 Parkinson's patients who had shown FoG episodes during a clinical freezing-provoking assessment course. Two clinical experts rated the video-recorded trials and marked episodes with festination, shank trembling, shuffling, or akinesia. Motion phases inside such episodes were labeled as FoG-affected. The classifiers were evaluated using leave-one-patient-out cross-validation. No statistically significant differences could be observed between the different classifiers for FoG detection (p>0.05). An SVM model with 10 features of the actual and two preceding motion phases achieved the highest average performance with 88.5 ± 5.8% sensitivity, 83.3 ± 17.1% specificity, and 92.8 ± 5.9% Area Under the Curve (AUC). The performance of predicting the behavior of the next motion phase was significantly lower compared to the detection classifiers. No statistically significant differences were found between all prediction models. An SVM-predictor with features from the two preceding motion phases had with 81.6 ± 7.7% sensitivity, 70.3 ± 18.4% specificity, and 82.8 ± 7.1% AUC the best average performance. The developed methods enable motion-phase-based FoG detection and prediction and can be utilized for closed-loop systems that provide on-demand gait-phase-synchronous cueing to mitigate FoG symptoms and to prevent complete motoric blockades.

There is only little and partially contradicting knowledge on the variation of pollen abundance at different altitudes in mountainous regions. The aim of this work is to gain new insights on the influence of wind and surrounding trees on the deposition and transport of birch (Betula spec.) pollen in the Bavarian Alps, Germany. Data on birch pollen deposition were collected at five sites using gravimetric traps along an altitudinal gradient from Garmisch-Partenkirchen (720 m a.s.l.) to the Environmental Research Station Schneefernerhaus (2650 m a.s.l) in the pollen season 2020. We compared these data with birch pollen concentration derived from a volumetric trap at Schneefernerhaus and with phenological data, i.e., flowering onset times observed at 21 birch trees at different altitudes. Wind data were gathered directly at or near each pollen trap and surrounding birch trees were mapped in the field. Whereas the pollen load was lowest at the highest location, substantially higher values were measured at medium altitudes (1300–1600 m a.s.l.). This can be explained by the pronounced mountain-valley wind system, which ensured the transport of pollen to the corresponding altitudes. We conclude that pollen levels are influenced by topography, local wind systems and the availability of pollen. Pollen levels in complex mountainous environments are therefore not substantially affected by the occurrence of birch trees in the immediate vicinity.

Fragmented ash populations due to ash dieback may lead to a limited gene flow and pollination success. Therefore, the viability of ash pollen plays a major role for the survival of the species. The extent to which the long-distance transport of pollen affects pollen viability was investigated with experiments in a climate chamber using ash pollen samples from a seed orchard in Emmendingen, Germany. Furthermore, experiments with a volumetric pollen trap were conducted. A suitable storage temperature for ash pollen was determined by using four viability tests; TTC test, pollen germination, Alexander’s stain and Acetocarmine. An optimization of the germination medium was performed. We found a strong influence of prevailing temperatures on pollen viability, which decreased faster under warmer conditions. At moderate temperatures, viable pollen could still be observed after 28 days. Thus, a possible successful pollination can also be associated to long-range transported pollen. Storage experiments showed that pollen viability could be maintained longer at temperatures of −20 °C and −80 °C than at 4 °C. In particular, the TTC test has proven to be suitable for determining viability. Therefore, properly stored pollen can be used for breeding programs to support the survival of Fraxinus excelsior.

The ongoing fragmentation of ash populations due to ash dieback requires an effective gene flow between individuals; thus, investigations on ash pollen transport are essential. In this study, comprehensive aerobiological field experiments at two seed plantations in Baden-Württemberg were conducted in 2019 and 2020 in order to study the influence of phenology and meteorology (especially wind) on pollen transport using self-constructed gravimetric pollen traps located 1.5 and 5 m a.g.l. Our main objectives were to investigate the local scale dispersion of ash pollen and to evaluate the recommended distance (i.e., 400 m) from seed plantations to other ash trees according to the German Forest Reproduction Act. Our results showed a link between pollen transport and meteorology, the onset of phenological development, and the topography of the plantation. The plantation at Schorndorf was characterized by a slope and associated cold air flows, suggesting that this could be a factor contributing to higher pollen levels at the downslope traps. In addition, in many cases, the cardinal direction associated with the highest pollen impaction was also identical with the predominant wind direction. Analyzing pollen data for single traps in detail, we found that the highest total pollen catch (31%) was measured outside the plantations in 2019, a year with very low flower development. In contrast, most pollen (33%) was caught within the plantation in 2020, which presented a much stronger pollen year than 2019 (with a factor of 11 regarding total sums). This indicates, in the lower pollen year, a potential higher contribution of trees from outside the plantation, and thus it can be recommended that seed harvesting of ash trees in the plantations should preferentially take place in full mast years. Interestingly, the total pollen deposition in Emmendingen at 5 m height showed little difference compared to the traps at 1.5 m height, but there was a large temporal difference pointing to vertical variations in pollen availability. In general, we found that ash pollen was transported for a larger distance than 400 m, but the amount of pollen decreased substantially with increasing distance. At a distance of 200 m, there was already approx. 50% less pollen captured from the air. However, even at a distance of 500 m, more than 10% of the pollen was still captured. In order to ensure cross-pollination of healthy ash trees, the distance of ash individuals or stands should not be too large, and there should be no spatial separation (e.g., by conifer stands).

Grass pollen allergy is widespread all around the globe. With an increasing number of people living in cities, the examination of grass pollen levels within cities and their surroundings has increased in importance. The aim of this study was to examine different temporal and spatial scales of grass pollen concentration and deposition across urban and semi-rural environments in the years 2019 and 2020. We installed different types of pollen traps in the city of Ingolstadt (Bavaria, Germany) and its surroundings: volumetric pollen traps at roof level to assess background pollen concentration and gravimetric pollen traps and portable volumetric traps at street level. We considered grass pollen concentration and deposition in the context of land use and management. Our data showed that the grass pollen season in 2020 was longer and more intense than in 2019. Background grass pollen concentration was generally higher at the semi-rural site in both years: peak values were eight times (2019) and more than four times (2020) higher, and Seasonal Pollen Index was more than four times and almost three times higher in 2019 and 2020, respectively. Analyses of spatial variations measured at street level revealed higher numbers for pollen deposition and concentrations at semi-rural than at urban sites. Recorded values were linked to local vegetation and the management of grass areas surrounding the traps. Analyses of diurnal variations at street level in June 2019 showed that pollen concentration for all sites, independent of their degree of urbanization, were highest at noon (22.2 pollen grains/m³ vs. 8.5 pollen grains/m³ in the morning and 10.4 pollen grains/m³ in the evening). Diurnal variations at roof level showed similarities for the same days but differed when considering the whole season. Our data suggest the importance of the management of grass areas as areas cut earlier have a decreased amount of emitted pollen.

Background and Objectives: The existence of common ash (Fraxinus excelsior) in Europe is severely endangered by ash dieback. To support its future sustainability, it is essential to improve the natural ash regeneration. The main aim of this study was to investigate the influence of light conditions, conceivably influenced by stand structure/ash dieback, on ash regeneration and the competition between ash seedlings and species growing in the understory. Materials and Methods: We selected 40 plots in a riparian forest located in Bavaria, Germany. Light-related variables (Leaf Area Index, gap fraction) were gathered with fish-eye photography, whereas other environmental factors were derived from vegetation surveys (Ellenberg indicator values). We assessed vegetation parameters such as species’ richness and coverage of the herb layer to account for competition with ash seedlings. Results: Our results indicate that ash regeneration is favoured under shady conditions. The majority of other abiotic factors were not statistically associated with the analysed ash metrics. In contrast, the coverage of grass was negatively related to LAI and positively to gap fraction. Higher herb and grass coverages were linked to a suppression of ash regeneration. A higher litter coverage was associated with a higher frequency of ash seedlings. Nonparametric partial correlation analyses demonstrated the influence of light and stressed that litter coverage is of particular importance. Conclusions: We conclude that gaps, inter alia induced by ash dieback, favour grass invasion. In turn, this invasion might suppress regeneration of ash. In this regard, rapid silvicultural management such as reforestation of gaps after dieback of mature trees is recommended. The influence of litter on interspecific competition during growth should be also considered. The pace of dieback might additionally influence the timing and quantity of litter accumulation; thus, further research should also focus on these interrelations.

Coarse particulate organic matter (CPOM) fulfills important functions in the physical and ecological system of a stream. CPOM delivery to and export from the stream has implications for the stream's morphology and sediment transport capacity as well as the energy budget and food availability. Export rates of CPOM from mountain catchments have been observed to strongly increase with rising discharge, but the mechanism leading to this strong relationship is unclear. Here, we show that log jams in the Erlenbach, a steep headwater stream in the Swiss Prealps, are an effective barrier for the transport of CPOM pieces, and thus become sites of storage of large quantities of material over time. Exceptional discharge events with return periods exceeding 20 years play a dual role in CPOM transport in the Erlenbach. First, they appear to destroy existing log jams, releasing the stored material (wood and sediment). Second, they intensify channel–hillslope coupling, thereby recruiting new logs to the channel, around which new jams can form. This allows for the formulation of a new, fully episodic end-member in a four-end-member model of CPOM dynamics of steep mountain streams based on wood delivery and export.
