Aquaculture practices expose fish to several factors that may generate stress, modifying the balance between the production of reactive oxygen species (ROS) and the activity of antioxidant defenses that induce cell damage. Alpha-tocopherol (VE) improves the antioxidant capacity against ROS production in fish. A 50-day trial with longfin yellowtail (Seriola rivoliana) juveniles was conducted to compare the dietary supplementation of 500 mg/kg of VE against a control diet without VE supplementation on growth, lymphoid tissue enzymatic activity, immune-system-related gene expression, and the histology of the liver and spleen. Growth, weight gain, specific growth rate, feed conversion rate, and survival did not show significant differences (p > 0.05) among treatments. Fish fed with an α-tocopherol-enriched diet showed a higher enzymatic activity of superoxide dismutase in the liver (p < 0.05) and a lower percentage of melanomacrophage coverage area in the lymphoid organs (p < 0.05). Overexpression was observed of MyD88 and il-10 in the spleen, and il-1b in the liver in fish fed 500 mg/kg of VE, as well as overexpression of Toll-like 3 in the head kidney, spleen, and liver in fish fed the control diet. Dietary supplementation with VE reduces the effects of oxidative stress and improves lymphoid tissue defense and immune-related gene expression in S. rivoliana.

We evaluated the effect of three variables (cryoprotectant temperature, light regime, and time of exposure to the cryoprotectant) throughout the equilibrium period during cryopreservation on the viability of the microalga Chaetoceros calcitrans (Paulsen). For this, the cryoprotectant dimethyl sulfoxide (DMSO) at 5% (v/v) was added at three different temperatures (4, 10, and 25°C) before placing the microalgae in cryobiological straws for freezing. Once inside the cryobiological straws, the microalgal-cryoprotectant suspensions were subjected to the following light regimes for 15 or 45 min: complete light, complete darkness, light/darkness, and darkness/light. Suspensions were then frozen under controlled conditions and stored in liquid nitrogen. The viability index proposed by Cañavate & Lubian (1995b) was used to measure microalgal viability after cryopreservation. Results indicated that it is necessary to use a cryoprotectant to ensure the viable cryopreservation of C. calcitrans. Statistical analyses showed that the temperature of the DMSO influenced the viability of cryopreserved microalgae and that there was no synergistic effect between the other variables studied. The viabilities obtained with DMSO at 25°C, 10°C, and 4°C were 34.9%, 27.8%, and 20.6%, respectively.Se evaluó el efecto de tres variables - la temperatura del crioprotector, el régimen de luz y el tiempo de exposición al crioprotector durante el período de equilibrio en el proceso de criopreservación -sobre la viabilidad de la microalga Chaetoceros calcitrans (Paulsen). Para ello se utilizó, el crioprotector dimetilsulfóxido (DMSO) al 5% (v/v), el cual fue adicionado a tres temperaturas diferentes (4, 10 y 25°C) antes de que las microalgas fueran introducidas dentro de pajuelas criobiológicas para su congelación. Una vez dentro de las pajuelas, las suspensiones de microalgas con crioprotector se sometieron durante 15 o 45 min a luz completa, oscuridad completa, luz/oscuridad y oscuridad/luz. Luego se congelaron de manera controlada y se almacenaron en nitrógeno líquido. La viabilidad de las microalgas posterior a la criopreservación fue medida a través del índice de viabilidad propuesto por Cañavate & Lubian (1995b). Los resultados indicaron que es necesario el uso de criprotector para la criopreservación viable de C. calcitrans. El análisis estadístico demostró que la temperatura del DMSO influye sobre la viabilidad de la microalga criopreservada y que no existe efecto sinérgico entre las demás variables de estudio. Las viabilidades alcanzadas usando DMSO a 25°C, 10°C y 4°C fueron de 34,9%, 27,8% y 20,6% respectivamente.

Abstract Background Intermittent fasting (IF) has shown benefits in various pathological conditions. Although its anti-inflammatory potential has been recognized, its effects on the mechanism underlying rheumatoid arthritis (RA) remain insufficiently characterized. This study aimed to investigate the effects of IF in a murine model of RA. Methods Collagen-induced arthritis (CIA) was developed in sixteen male DBA/1 mice, randomly assigned to two groups, with one undergoing IF every other day for four weeks. The effects of IF on joint inflammation and remodeling were evaluated clinically, histologically, and through tomography. Transcriptomic changes were characterized using expression microarrays, validated by RT-qPCR, and confirmed by immunohistochemistry. Additionally, modifications in gut microbiota were assessed through 16 S sequencing. Results Mice subjected to IF significantly reduced the incidence and severity of clinical arthritis. Histological and radiographic assessments confirmed a decrease in inflammation and joint damage. Transcriptomic analysis revealed that IF led to the upregulation of 364 genes and the downregulation of 543 genes, with notable reductions in inflammatory signaling pathways associated with RA-related genes, including Cd72, Cd79a, Ifna, Il33, and Bglap 2. Notably, IL33 emerged as a pivotal mediator in the inflammatory processes mitigated by fasting. Key regulators associated with IF effects, such as CEBPA, FOXO1, HIF1A, PPARG, and PPARA, were identified, indicating a complex interplay between metabolic and inflammatory pathways. Furthermore, differential expression of microRNAs and lncRNAs, including miR-15b, miR-103-2, miR-302a, miR-6985, and miR- 5624, was observed. Metagenomic analysis indicated that IF enhanced the abundance and diversity of the gut microbiome, explicitly promoting anti-inflammatory bacterial populations, notably within the genus Ruminococcaceae. Conclusion Our findings suggest that IF exerts significant anti-inflammatory and immunoregulatory effects in the context of CIA. Given its non-risky nature, further investigation into the potential benefits of IF in patients with RA is warranted. Clinical trial number Not applicable.

Objetivo. Estudiar el efecto de medicamentos homeopáticos sobre el crecimiento, supervivencia y microbiota del tracto gastrointestinal (TGI) de almeja Catarina Argopecten ventricosus. Materiales y Métodos. Se aplicaron cinco tratamientos homeopáticos derivados de bacterias [(T1) ViP-ViA 1D, (T2) ViP-ViA 7C], minerales [(T3) AcF-MsS 1D, (T4) PhA-SiT 7C], o venenos [(T5) ViT 31C] y tres controles: (C1) etanol diluido 1:99, (C2) etanol dinamizado 1C y (C3) agua destilada. La microbiota se determinó secuenciando la región V3-V5 del gen 16S rRNA. Resultados. El mayor crecimiento en longitud de la concha correspondió a T1 (117 µm d-1) y T2 (108 µm d-1), la mayor supervivencia a T3 y T5 y el mejor resultado global a T3. Las curvas de rarefacción de los grupos tratados y controles mostraron una clara separación. Se encontraron diferencias significativas (p≤0.05) entre filos (Proteobacteria > Actinobacteria > Firmicutes > Bacterloidetes> Chloroflexi y para los Géneros: Symbiobacterium > Microbacterium > Methylobacillus > Bacillus > Paenibacillus > Burkholderia > Nostoc > Methylobacterium > Leucobacter). El género Symbiobacterium fue dominante (p≤0.05) para T5, respecto a todos los tratamientos y grupos controles. La especie Microbacterium maritypicum (Actinobacteria) mostró la mayor abundancia relativa (p≤0.05) en T1 y T3 y Symbiobacterium toebii (Firmicutes) en T5 y T2 (p≤0.05), ambas con respecto al inicio del estudio T0. Conclusiones. Se presenta por primera vez la composición de la microbiota del TGI de A. ventricosus y la aplicabilidad potencial de la homeopatía para mejorar el rendimiento productivo y modular la microbiota gastrointestinal de la especie.

Abstract This study assessed the intestinal microbiota of juveniles of the White shrimp Litopenaus vannamei, whose feed was enriched with three probiotic formulations: Streptomyces sp. RL8 (RL8); a mix of Lactobacillus graminis and Streptomyces spp. RL8 and N7 (Lac‐Strep); and a mix of Bacillus spp. and Streptomyces spp. RL8 and N7 (Bac‐Strep). The analysis was performed by sequencing the V3 region of the 16S rRNA gene of treated animals and the control group before and after Vibrio parahaemolyticus challenge. After challenge, the highest Shannon diversity indexes corresponded to RL8 and Bac‐Strep (3.94 ± 0.11 and 3.39 ± 0.3, respectively) and the lowest to the control group (2.58 ± 0.26). The most abundant phyla before and after challenge were Proteobacteria, Actinobacteria, and Bacteroidetes. The principal component analysis and Statistical Analysis of Metagenomic Profiles (STAMP) showed that the gut microbiota of the groups RL8 and Bac‐Strep after challenge was different from the other experimental groups, which was characterized by a higher bacterial diversity, as well as a significant stimulation of the Bacteriovorax population and other antimicrobial producing genera that protected shrimp from infection.
