Abstract Background Dissolved organic matter (DOM) plays a significant role in the biogeochemical cycle of crucial elements in aquatic ecosystem. However, it is still not clear on the spectral characteristics of water DOM in different types of alpine wetlands, which have less anthropogenic influences and intensive ultraviolet radiation. Here, we collected 107 water samples from marsh, lake, and river wetlands in the Zoige plateau, China, and analyzed the chemical characteristics, compositions, and potential sources of chromophoric DOM by combining UV–vis spectroscopy and excitation–emission matrix fluorescence spectroscopy coupled with parallel factor analysis (EEMs-PARAFAC). Results UVC and UVA fulvic-like substances were the prevailing fluorescence components in water DOM, which accounted for 23.74–71.59% and 16.76–30.01% of the total fluorescence intensity, respectively. Compared with the lake and river wetlands, fluoresce intensities of UVC and UVA fulvic-like substances in DOM were higher in marsh wetland. Marsh wetlands possessed the highest SUVA254, E2/E3, E2/E4, and E4/E6 of DOM, suggesting higher humification degree, higher relative molecular nominal size, and higher aromaticity. And the E2/E4 ratios in most water samples were higher than 12, indicating water DOM was mainly derived from autochthonous sources in alpine wetlands. Conclusions Wetland types strongly affected the spectral characteristics of water DOM in Zoige plateau. These findings may be beneficial for sustainable management of alpine wetlands. Abstract Notch signaling, renowned for its role in regulating cell fate, organ development, and tissue homeostasis across metazoans, is highly conserved throughout evolution. The Notch receptor and its ligands are transmembrane proteins containing epidermal growth factor-like repeat sequences, typically necessitating receptor-ligand interaction to initiate classical Notch signaling transduction. Accumulating evidence indicates that the Notch signaling pathway serves as both an oncogenic factor and a tumor suppressor in various cancer types. Dysregulation of this pathway promotes epithelial-mesenchymal transition and angiogenesis in malignancies, closely linked to cancer proliferation, invasion, and metastasis. Furthermore, the Notch signaling pathway contributes to maintaining stem-like properties in cancer cells, thereby enhancing cancer invasiveness. The regulatory role of the Notch signaling pathway in cancer metabolic reprogramming and the tumor microenvironment suggests its pivotal involvement in balancing oncogenic and tumor suppressive effects. Moreover, the Notch signaling pathway is implicated in conferring chemoresistance to tumor cells. Therefore, a comprehensive understanding of these biological processes is crucial for developing innovative therapeutic strategies targeting Notch signaling. This review focuses on the research progress of the Notch signaling pathway in cancers, providing in-depth insights into the potential mechanisms of Notch signaling regulation in the occurrence and progression of cancer. Additionally, the review summarizes pharmaceutical clinical trials targeting Notch signaling for cancer therapy, aiming to offer new insights into therapeutic strategies for human malignancies.

Highlights A molecular-sieving electrode coating towards low-concentrated aqueous sodium-ion batteries is constructed by applying a composite of NaX zeolite and NaOH-neutralized Nafion. Resulting from a molecular sieving effect of zeolite channels and size-shrunken ionic domains in Nafion, the as-prepared coating layer reject hydrated Na+ ions and allow fast dehydrated Na+ permeance. 200 cycles of Na2MnFe(CN)6//NaTi2(PO4)3 full cells can be achieved in a practically feasible 2 m aqueous electrolyte.

Abstract Hepatocellular carcinoma (HCC) is the most prevalent type of liver cancer. Since the tricarboxylic acid cycle is widely involved in tumor metabolic reprogramming and cuproptosis, investigating related genes may help to identify prognostic signature of patients with HCC. Data on patients with HCC were sourced from public datasets, and were divided into train, test, and single-cell cohorts. A variety of machine learning algorithms were used to identify different molecular subtypes and determine the prognostic risk model. Our findings revealed that the risk score (TRscore), based on the genes OGDHL, CFHR4, and SPP1, showed excellent predictive performance in different datasets. Pathways related to cell cycle and immune inflammation were enriched in the high-risk group, whereas metabolism-related pathways were significantly enriched in the low-risk group. The high-risk group was associated with a greater number of mutations of detrimental biological behavior and higher levels of immune infiltration, immune checkpoint expression, and anti-cancer immunotherapy response. Low-risk patients demonstrated greater sensitivity to erlotinib and phenformin. SPP1 was mainly involved in the interaction among tumor-associated macrophages, T cells, and malignant cells via SPP1–CD44 and SPP1–(ITGA5 + ITGB1) ligand-receptor pairs. In summary, our study established a prognostic model, which may contribute to individualized treatment and clinical management of patients with HCC.

Abstract RNA methylation modification plays a crucial role as an epigenetic regulator in the oncogenesis of hepatocellular carcinoma (HCC). Numerous studies have investigated the molecular mechanisms underlying the methylation of protein-coding RNAs in the progression of HCC. Beyond their impact on mRNA, methylation modifications also influence the biological functions of non-coding RNAs (ncRNAs). Here, we present an advanced and comprehensive overview of the interplay between methylation modifications and ncRNAs in HCC, with a specific focus on their potential implications for the tumor immune microenvironment. Moreover, we summarize promising therapeutic targets for HCC based on methylation-related proteins. In the future, a more profound investigation is warranted to elucidate the effects of ncRNA methylation modifications on HCC pathogenesis and devise valuable intervention strategies. Video Abstract

Takayasu arteritis (TA) is a rare systemic vasculitis of the aorta and its primary branches, which usually occurs in young women. Due to its insidious onset and lack of specific symptoms, this disease can be easily misdiagnosed or missed. Approximately 50 ​% of the patients having TA with pulmonary artery involvement develop pulmonary hypertension (PH). The 3-year survival rate among patients with TA-related PH is lower than that among patients with TA alone. Early balloon pulmonary angioplasty (BPA) can improve the clinical symptoms and survival of patients with stable TA. To the best of our knowledge, this is the first case reported in the English literature in which a “Guidezilla” catheter was used during BPA to treat stenosis and occlusion of the pulmonary artery caused by Takayasu arteritis (TA).

Wetlands, as a crucial component of terrestrial ecosystems, play a significant role in global ecological services. Aboveground biomass (AGB) is a key indicator of the productivity and carbon sequestration potential of wetland ecosystems. The current research methods for remote-sensing estimation of biomass either rely on traditional vegetation indices or merely perform integer-order differential transformations on the spectra, failing to fully leverage the information complexity of hyperspectral data. To identify an effective method for estimating AGB of mixed-wetland-plant communities, we conducted field surveys of AGB from three typical wetlands within the Crested Ibis National Nature Reserve in Hanzhong, Shaanxi, and concurrently acquired canopy hyperspectral data with a portable spectrometer. The spectral features were transformed by applying fractional-order differentiation (0.0 to 2.0) to extract optimal feature combinations. AGB prediction models were built using three machine learning models, XGBoost, Random Forest (RF), and CatBoost, and the accuracy of each model was evaluated. The combination of fractional-order differentiation, vegetation indices, and feature importance effectively yielded the optimal feature combinations, and integrating vegetation indices with feature bands enhanced the predictive accuracy of the models. Among the three machine-learning models, the RF model achieved superior accuracy using the 0.8-order differential transformation of vegetation indices and feature bands (R2 = 0.673, RMSE = 23.196, RPD = 1.736). The optimal RF model was visually interpreted using Shapley Additive Explanations, which revealed that the contribution of each feature varied across individual sample predictions. Our study provides methodological and technical support for remote-sensing monitoring of wetland AGB.

Soil nitrogen and phosphorus are directly related to soil quality and vegetation growth and are, therefore, a common research topic in studies on global climate change, material cycling, and information exchange in terrestrial ecosystems. However, collecting soil hyperspectral data under in situ conditions and predicting soil properties, which can effectively save time, manpower, material resources, and financial costs, have been generally undervalued. Recent optimization techniques have, however, addressed several of the limitations previously restricting this technique. In this study, hyperspectral data were taken from surface soils under different vegetation types in the wetlands of the Shaanxi Yellow River Wetland Provincial Nature Reserve. Through in situ original and first-order differential transformation spectral data, three prediction models for soil carbon, nitrogen, and phosphorus contents were established: partial least squares (PLSR), random forest (RF), and Gaussian process regression (GPR). The R2 and RMSR of the constructed models were then compared to select the optimal model for evaluating soil content. The soil organic carbon, total nitrogen, and total phosphorus content models established based on the first-order differential had a higher accuracy when modeling and during model validation than those of other models. Moreover, the PLSR model based on the original spectrum and the Gaussian process regression model had a superior inversion performance. These results provide solid theoretical and technical support for developing the optimal model for the quantitative inversion of wetland surface soil carbon, nitrogen, and phosphorus based on in situ hyperspectral technology.

Appropriate industrial agglomeration has several benefits, including reducing environmental pollution, promoting innovation and enhancing grain green total factor productivity (GTFP). It is an effective strategy for promoting high-quality and sustainable development in the grain industry. In this study, the slacks-based measure of the global Malmquist–Luenberger (SBM-GML) model is used to estimate China's grain GTFP using the panel data of 31 provinces from 2001 to 2020. Furthermore, a dynamic spatial econometric model is employed to empirically investigate the impact of grain industry agglomeration on grain GTFP and its regional heterogeneity. The results show that GTFP exhibits a fluctuating growth tendency, with advancements in green technology serving as the primary engine of that expansion. (2) The deepening of grain industrial agglomeration has a long-term promoting effect on the grain GTFP of local and neighbouring areas, with the long-term effect being more significant than the short-term effect, as revealed by the dynamic spatial Durbin model. (3) According to the heterogeneity analysis, industrial agglomeration's impact on grain GTFP is most noticeable in the production and sales balance. Therefore, encouraging regional cooperation and communication while raising the grain industrial agglomeration standard is crucial. The degree of regional economic growth, the state of the agricultural infrastructure and the conditions of the natural resources should all be considered by policymakers when developing distinct and focused policy assistance for each region.

Abstract Background This study investigated the differences in target volumes between preoperative magnetic resonance imaging (MRIpre) and postoperative MRI (MRIpost) for breast radiotherapy after breast‐conserving surgery (BCS) using deformable image registration (DIR). Methods and Materials Seventeen eligible patients who underwent whole‐breast irradiation in the prone position after BCS were enrolled. On MRIpre, the gross tumor volume (GTV) was delineated as GTVpre, which was then expanded by 10 mm to represent the preoperative lumpectomy cavity (LC), denoted as LCpre. The LC was expanded to the clinical target volume (CTV) and planning target volume (PTV) on the MRIpre and MRIpost, denoted as CTVpre, CTVpost, PTVpre, and PTVpost, respectively. The MIM software system was used to register the MRIpre and MRIpost using DIR. Differences were evaluated regarding target volume, distance between the centers of mass (dCOM), conformity index (CI), and degree of inclusion (DI). The relationship between CILC/CIPTV and the clinical factors was also assessed. Results Significant differences were observed in LC and PTV volumes between MRIpre and MRIpost (p 0.05) were found between CI and GTV, primary tumor volume‐to‐breast volume ratio, distance from the primary tumor to the nipple and chest wall, and body mass index. Conclusions Despite using DIR technology, the spatial correspondence of target volumes between MRIpre and MRIpost was suboptimal. Therefore, relying solely on preoperative diagnostic MRI with DIR for postoperative LC delineation is not recommended.

Constructing resistive layer with novel structure holds great potential in upgrading electromagnetic properties of soft magnetic materials in a wide frequency range. In this work, FeNi@SiO2 composites with controllable core–shell structures, including 2D-type laminar shell and 3D-type raspberry-like shell, were fabricated by modified Stöber method. The structure of SiO2 shell depends highly on the concentration of tetraethyl orthosilicate (TEOS). In detail, a laminar shell can be formed at low TEOS concentration, owing to slow condensation. While at high TEOS concentration, heterogeneous growth on powder surface and homogeneous growth in solution of silica networks are significantly accelerated, resulting in the formation of the raspberry-like shell. Improved electromagnetic performance of FeNi@SiO2 cores at 2 MHz, including high resistivity, low eddy current loss, and low loss factor, should be ascribed to the unique raspberry-like shell structure. Meanwhile, raspberry-like shell also contributes to enhanced attenuation and acceptable impedance matching in GHz range. Consequently, a wide effective absorption bandwidth of 8.92 GHz can be achieved at a thin thickness of 1.85 mm. This work provides deep insights into the growth mechanism of different SiO2 shells and sheds new light on the design of high-performance soft magnetic composites working in different frequency bands.

Abstract Hepatocellular carcinoma (HCC) is the most prevalent form of primary liver cancer, accounting for over 90% of cases. As pyruvate metabolic pathways are often dysregulated in cancer cells, investigating pyruvate metabolism-related genes may help identify prognostic gene signature and develop potential strategies for the management of patients with HCC. The mRNA expression profile, gene mutation data, and clinical information of HCC were obtained from open-source databases. A list of pyruvate metabolism-related genes was downloaded from the MSigDB dataset. Our findings revealed that certain pyruvate metabolism-related genes had copy number variations and single nucleotide variations in patients with liver cancer. Based on pyruvate metabolism-related genes, we stratified patients with HCC into three subtypes with different prognoses, clinical features, mutation profiles, functional annotation, and immune infiltration status. Next, we identified 13 key pyruvate metabolism-related genes significantly correlated with the prognosis of HCC using six machine learning algorithms and constructed a risk model. We also observed that the risk score was positively associated with a worse prognosis and increased immune infiltration. In summary, our study established a prognostic risk model for HCC based on pyruvate metabolism-related genes, which may contribute to the identification of potential prognostic targets and the development of new clinical management strategies for HCC.

Abstract Balloon pulmonary angioplasty (BPA) has been proven effective for addressing technically inoperable chronic thromboembolic pulmonary hypertension (CTEPH). However, the effectiveness of BPA in technically operable CTEPH patients who, for various reasons, did not undergo the procedure remains an area requiring exploration. This study sought to assess the safety and efficacy of BPA in such cases. We collected and reviewed data from CTEPH patients who underwent BPA in a consecutive manner. Following multidisciplinary team (MDT) decisions, patients were classified into two groups: technically inoperable (group A) and operable (group B). Group B comprised patients deemed technically suitable for pulmonary endarterectomy (PEA) but who did not undergo the procedure for various reasons. All patients underwent a comprehensive diagnostic work‐up, including right heart categorization at baseline and the last intervention. This study compared changes in hemodynamic parameters, functional capacity, and quality of life between the two groups. In total, 161 patients underwent 414 procedures at our center, with Group A comprising 112 patients who underwent 282 BPA sessions and group B comprising 49 patients who underwent 132 BPA sessions. Significantly, both groups exhibited improvements in hemodynamics, functional capacity, and quality of life. The occurrence rate of complications, including hemoptysis and lung injury, was similar [12 (63.2%) vs. 7 (36.8%), p = 0.68]. BPA demonstrated favorable outcomes in patients with proximal CTEPH who did not undergo pulmonary endarterectomy. However, the clinical impact of BPA in technically operable CTEPH was found to be less significant than in inoperable cases.

In the R22 (chlorodifuoromethane) steam-cracking process, which is used to produce a TFE (tetrafluoroethylene) monomer, distillation is employed to separate the high-purity TFE monomer from the cracked gas generated during this procedure. Traditionally, this distillation process is carried out using five towers. In this study, the traditional five-tower distillation method was transformed into a four-tower distillation method through the Aspen Plus simulation software, and this process was simulated and optimized. Meanwhile, a double-effect distillation process was designed for the transformed four-tower distillation process. The transformed distillation process not only meets the requirements of 99.999% purity for the TFE monomer and 99.99% purity for R22 recycling, but it also reduces the footprint by eliminating one distillation tower and saves 112.9002 kW of tower load, thus reducing the operating costs. This research provides valuable guidance for practical production.

This study aimed to investigate the relationships between LPCAT1 expression and clinicopathologic parameters of hepatocellular carcinoma (HCC), further, to explore the effect of LPCAT1 on overall survival (OS) in patients with HCC, and its possible mechanism. Bioinformatics analysis using high throughput RNA-sequencing data from TCGA was utilized to explore the differential expression of LPCAT1 between normal and tumor tissues, and the associations between LPCAT1 expression and clinicopathological parameters. Survival analyses and subgroup survival analyses were utilized to elucidate the effect of LPCAT1 on OS in patients with HCC. Univariate analysis and multivariate analysis were used to investigate the prognostic factors. Potential LPCAT1 related tumor genes were identified by the methodology of differentially expressed genes (DEGs) screening. GO term enrichment analysis, KEGG pathway analysis and the PPI network were used to explore the potential mechanism. LPCAT1 was significantly overexpressed in HCC tumor tissues compared with normal tissues. The LPCAT1 expression was related to tumor grade, ECOG score, AFP and TNM stage, with P values of 0.000, 0.000, 0.007 and 0.000, respectively. Multivariate analysis demonstrated that LPCAT1 expression was independently associated with OS, with an HR of 1.04 (CI: 1.01–1.06, P = 0.003). The KEGG pathway enrichment analyses showed that overlapped DEGs mainly participate in the cell cycle. Finally, we identified a hub gene, CDK1, which has been reported to act on the cell cycle, consistent with the result of KEGG enrichment analysis. Collectively, these data confirmed LPCAT1 was upregulated in HCC, and was an independent predictor of the prognosis.

Glioma is the most common primary malignant tumor of the adult central nervous system. It has high morbidity and poor survival. Myelin protein zero-like protein 1 (MPZL1) is a cell surface glycoprotein that activates numerous adhesion-dependent signaling pathways. MPZL1 plays important roles in human cancers that include metastatic process; however, it is not clear if MPZL1 plays a role in human glioma. Therefore, this study aimed to determine if silencing MPZL1 impacted the cell proliferative features of human glioma cells. First, MPZL1 expression was investigated in human glioma samples and tumor cell lines. Then the effects of small interfering RNA (siRNA)-targeting MPZL1 were analyzed on proliferation, colony formation, cell cycle progression, and invasion of human glioma cells. The results from this study demonstrated that MPZL1 was highly expressed in human glioma tissues and glioma cell lines. In addition, knockdown of MPZL1 significantly inhibited cell proliferation, colony formation, and invasiveness of glioma cells, and effectively induced cell cycle arrest at the G1 phase. Western blotting analysis indicated that silencing MPZL1 expression downregulated the expression of matrix metalloproteinase-2 (MMP-2), WNT1, caspase-3, cyclin A1, epidermal growth factor receptor (EGFR), and signal transducer and activator of transcription 3 (STAT3), and upregulated p53. The results from this study suggest that MPZL1 might be a marker for tumors and could be a potential therapeutic target for human glioma.

Abstract Background This study aimed to investigate the effect of abdominal compression on tumour motion and target volume and to determine suitable planning target volume (PTV) margins for patients treated with lung stereotactic body radiotherapy (SBRT) based on four-dimensional computed tomography (4DCT). Methods Twenty-three patients diagnosed to have a peripheral pulmonary tumour were selected and divided into an all lesions group (group A), an upper middle lobe lesions group (group B), and a lower lobe lesions group (group C). Two 4DCT scans were performed in each patient, one with and one without abdominal compression. Cone beam computed tomography (CBCT) was performed before starting treatment. The gross target volumes (GTVs) were delineated and internal gross target volumes (IGTVs) were defined. IGTVs were generated using two methods: (1) the maximum intensity projections (MIPs) based on the 4DCT were reconstructed to form a single volume and defined as the IGTVMIP and (2) GTVs from all 10 phases were combined to form a single volume and defined as the IGTV10. A 5-mm, 4-mm, and 3-mm margin was added in all directions on the IGTVMIP and the volume was constructed as PTVMIP5mm, PTVMIP4mm, and PTVMIP3mm. Results There was no significant difference in the amplitude of tumour motion in the left–right, anterior–posterior, or superior-inferior direction according to whether or not abdominal compression was applied (group A, p = 0.43, 0.27, and 0.29, respectively; group B, p = 0.46, 0.15, and 0.45; group C, p = 0.79, 0.86, and 0.37; Wilcoxon test). However, the median IGTVMIP without abdominal compression was 33.67% higher than that with compression (p = 0.00), and the median IGTV10 without compression was 16.08% higher than that with compression (p = 0.00). The median proportion of the degree of inclusion of the IGTVCBCT in PTVMIP5mm, PTVMIP4mm, and PTVMIP3mm ≥ 95% was 100%, 100%, and 83.33%, respectively. Conclusions Abdominal compression was useful for reducing the size of the IGTVMIP and IGTV10 and for decreasing the PTV margins based on 4DCT. In IGTVMIP with abdominal compression, adding a 4-mm margin to account for respiration is feasible in SBRT based on 4DCT.

Abstract All‐solid‐state lithium‐metal batteries (ASLMBs) are considered to be remarkably promising energy storage devices owing to their high safety and energy density. However, the limitations of current solid electrolytes in oxidation stability and ion transport properties have emerged as fundamental barriers in practical applications. Herein, a novel solid electrolyte is presented by in situ polymerization of salt‐concentrated poly(ethylene glycol) diglycidyl ether (PEGDE) implanted with a three‐dimensional porous L10GeP2S12 skeleton to mitigate these issues. The poly(PEGDE) endows more oxygen atoms to coordinate with Li+, significantly lowering its highest occupied molecular orbital level. As a consequence, the electro‐oxidation resistance of poly(PEGDE) exceeds 4.7 V versus Li+/Li. Simultaneously, the three‐dimensonal porous L10GeP2S12 skeleton provides a percolated pathway for rapid Li+ migration, ensuring a sufficient ionic conductivity of 7.7 × 10−4 S cm−1 at room temperature. As the bottlenecks are well solved, 4.5 V LiNi0.8Mn0.1Co0.1O2‐based ASLMBs present fantastic cycle performance over 200 cycles with an average Coulombic efficiency exceeding 99.6% at room temperature.

BackgroundOxygen pathway limitation exists in chronic thromboembolic pulmonary hypertension (CTEPH). Pulmonary endarterectomy (PEA) and balloon pulmonary angioplasty (BPA) are two effective interventions for CTEPH, but their effects and comparison of these two interventions on the oxygen pathway are not well demonstrated.MethodsCTEPH patients with available pulmonary function test, hemodynamics, and blood gas analysis before and after the interventions were included for comparison of oxygen pathway in terms of lung ventilation, lung gas exchange, oxygen delivery, and oxygen extraction between these two interventions.ResultsThe change in the percentage of the predicted forced expiratory volume in the 1 s (−3.4 ± 12.7 vs. 3.8 ± 8.7%, P = 0.006) and forced vital capacity (−5.5 ± 13.0 vs. 4.2 ± 9.9%, P = 0.001) among the PEA group (n = 24) and BPA group (n = 46) were significantly different. Patients in the PEA group had a significant increase in their arterial oxygen saturation (from 92.5 ± 3.6 to 94.6 ± 2.4%, P = 0.022), while those in the BPA group had no change, which could be explained by a significant improvement in ventilation/perfusion (−0.48 ± 0.53 vs. −0.17 ± 0.41, P = 0.016). Compared with patients post-BPA, patients post-PEA were characterized by higher oxygen delivery (756.3 ± 229.1 vs. 628.8 ± 188.5 ml/min, P = 0.016) and higher oxygen extraction (203.3 ± 64.8 vs. 151.2 ± 31.9 ml/min, P = 0.001).ConclusionPartial amelioration of the oxygen pathway limitations could be achieved in CTEPH patients treated with PEA and BPA. CTEPH patients post-PEA had better performance in lung gas exchange, oxygen delivery, and extraction, while those post-BPA had better lung ventilation. Cardiopulmonary rehabilitation may assist in improving the impairment of the oxygen pathway.

Abstract Background The application of delayed-enhancement magnetic resonance (DE-MR) simulation imaging in lumpectomy cavity (LC) delineation for prone radiotherapy in patients with an invisible seroma or a low seroma clarity score (SCS) after breast-conserving surgery (BCS) based on deformable image registration (DIR) was assessed. Methods Twenty-six patients who were suitable for radiotherapy in prone positions after BCS were enrolled, and both computed tomography (CT) and DE-MR simulation scans were acquired. The LC delineated based on titanium surgical clips on CT images was denoted as LCCT. The LC delineated based on the signal of cavity boundaries on fat-suppressed T2-weighted imaging (T2WI) and multiphase delayed-enhancement T1-weighted imaging (DE-T1WI), which was performed at 2 min, 5 min and 10 min postinjection, were denoted as LCT2, LC2T1, LC5T1 and LC10T1, respectively. Afterwards, DIR was performed to compare the volumes and locations of the LCs with MIM software. The generalized conformity index (CIgen) of inter (intra) observer (Inter-CIgen and Intra-CIgen) was also used to explore the inter(intra) observer variation for LC delineation on each image modality. Results LCCT–LC10T1 provided the best conformal index (CI) and degree of inclusion (DI), increasing by 2.08% and 4.48% compared to LCCT–LCT2, 11.36% and 2.94% for LCCT–LC2T1, and 8.89% and 7.69% for LC5T1–LCCT, respectively. The center of mass (COM) of LCCT–LC10T1 decreased by 17.86%, 6.12% and 13.21% compared with that of LCCT–LCT2, LCCT–LC2T1 and LCCT–LC5T1, respectively. The agreement of LC delineation was strongest for 10th min DE-TIWI (coefficient of variation, COV = 2.30%, Inter-CIgen = 87.06%, Intra-CIgen = 92.64%). Conclusion For patients with a low SCS (SCS ≤ 2) after BCS, it is feasible to contour the LC based on prone DE-MR simulation images. Furthermore, the LC derived from prone DE-T1WI at 10 min was found to be most similar to that derived from prone CT simulation scans using titanium surgical clips regardless of the volume and location of the LC. Inter (intra) variability was minimal for the delineation of the LC based on 10th min DE-TIWI.

Background and PurposeThis study aimed to investigate inter-/intra-observer delineation variability in GTVs of primary esophageal carcinomas (ECs) based on planning CT with reference to different combinations of diagnostic multimodal images from endoscopy/EUS, esophagography and FDG-PET/CT.Materials and MethodsFifty patients with pathologically proven thoracic EC who underwent diagnostic multimodal images before concurrent chemoradiotherapy were enrolled. Five radiation oncologist independently delineated the GTVs based on planning CT only (GTVC), CT combined with endoscopy/EUS (GTVCE), CT combined with endoscopy/EUS and esophagography (X-ray) (GTVCEX), and CT combined with endoscopy/EUS, esophagography, and FDG-PET/CT (GTVCEXP). The intra-/inter-observer variability in the volume, longitudinal length, generalized CI (CIgen), and position of the GTVs were assessed.ResultsThe intra-/inter-observer variability in the volume and longitudinal length of the GTVs showed no significant differences (p>0.05). The mean intra-observer CIgen values for all observers was 0.73 ± 0.15. The mean inter-observer CIgen values for the four multimodal image combinations was 0.67 ± 0.11. In the design of antineoplastic drugs, quinazolinone derivatives are often used as small molecule inhibitors for kinases or receptor kinases, such as the EGFR tyrosine kinase inhibitor gefitinib, p38MAP kinase inhibitor DQO-501, and BRD4 protein inhibitor PFI-1. A novel and convenient approach for the solid-phase synthesis of dihydroquinazoline-2(1H)-one derivatives was proposed and 19 different compounds were synthesized. Cytotoxicity tests showed that most of the target compounds had anti-proliferative activity against HepG-2, A2780 and MDA-MB-231 cell lines. Among them, compounds CA1-e and CA1-g had the most potent effect on A2780 cells, with IC50 values of 22.76 and 22.94 μM, respectively. In addition, in an antioxidant assay, the IC50 of CA1-7 was 57.99 μM. According to bioinformatics prediction, ERBB2, SRC, TNF receptor, and AKT1 were predicted to be the key targets and play an essential role in cancer treatment. ADMET prediction suggested 14 of the 19 compounds had good pharmacological properties, i.e., these compounds displayed clinical potential. The correct structure of the final compounds was confirmed based on LC/MS, 1H NMR, and 13C NMR.

Increasing attention has been given to the impact of extreme drought stress on ecosystem ecological processes. Ecosystem respiration (Re) and soil respiration (Rs) play a significant role in the regulation of the carbon (C) balance because they are two of the largest terrestrial C fluxes in the atmosphere. However, the responses of Re and Rs to extreme drought in alpine regions are still unclear, particularly with respect to the driver mechanism in plant and soil extracellular enzyme activities. In this study, we imposed three periods of extreme drought events based on field experiments on an alpine peatland: (1) early drought, in which the early stage of plant growth occurred from June 18 to July 20; (2) midterm drought, in which the peak growth period occurred from July 20 to August 23; and (3) late drought, in which the wilting period of plants occurred from August 23 to September 25. After 5 years of continuous extreme drought events, Re exhibited a consistent decreasing trend under the three periods of extreme drought, while Rs exhibited a non-significant decreasing trend in the early and midterm drought but increased significantly by 58.48% (p < 0.05) during the late drought compared with the ambient control. Plant coverage significantly increased by 79.3% (p < 0.05) in the early drought, and standing biomass significantly decreased by 18.33% (p < 0.05) in the midterm drought. Alkaline phosphatase, polyphenol oxidase, and peroxidase increased significantly by 76.46, 77.66, and 109.60% (p < 0.05), respectively, under late drought. Structural equation models demonstrated that soil water content (SWC), pH, plant coverage, plant standing biomass, soil β-D-cellobiosidase, and β-1,4-N-acetyl-glucosaminidase were crucial impact factors that eventually led to a decreasing trend in Re, and SWC, pH, β-1,4-glucosidase (BG), β-1,4-xylosidase (BX), polyphenol oxidase, soil organic carbon, microbial biomass carbon, and dissolved organic carbon were crucial impact factors that resulted in changes in Rs. Our results emphasize the key roles of plant and soil extracellular enzyme activities in regulating the different responses of Re and Rs under extreme drought events occurring at different plant growth stages.

Hyperspectral inversion techniques can facilitate soil quality monitoring and evaluation. In this study, the Yellow River Delta Wetland Nature Reserve was used as the study area. By measuring and analyzing soil samples under different vegetation types and collecting soil reflectance spectra, the relationships between vegetation types, soil depth, and the changes in soil total carbon (TC), total nitrogen (TN), and total phosphorus (TP) contents were assessed. The spectral data set was changed by spectral first derivative processing and division of the sample set according to vegetation type. The correlation between soil carbon, nitrogen, and phosphorus contents, and soil spectra was also analyzed, sensitive bands were selected, and the partial least-squares (PLS) method, support vector machine (SVM) method, and random forest (RF) model were used to establish the inversion model based on the characteristic bands. The optimal combination of spectral transformation, sample set partitioning, and inversion model was explored. The results showed significant differences (p < 0.05) in soil TC, TN, and TP contents under reed and saline alkali poncho vegetation, but not between soil element contents under different stratifications of the same plant species. The first derivative reflectance had higher correlation coefficients with soil TC, TN, and TP contents compared with the original reflectance, while the sensitive bands and quantities of the three elements differed. The division of the sample sets according to vegetation type and the first derivative treatment can improve the prediction accuracy of the model. The best combination of sample set plus FD plus RF for TC, TN, and TP in reed soil and sample set plus FD plus SVM for TC, TN, and TP in saline alkali pine soil provides technical support to further improve the prediction accuracy of TC, TN, and TP in wetland soil.

BackgroundClinically, many esophageal cancer patients who planned for radiation therapy have already undergone diagnostic Positron-emission tomography/computed tomography (PET/CT) imaging, but it remains unclear whether these imaging results can be used to delineate the gross target volume (GTV) of the primary tumor for thoracic esophageal cancer (EC).MethodsSeventy-two patients diagnosed with thoracic EC had undergone prior PET/CT for diagnosis and three-dimensional CT (3DCT) for simulation. The GTV3D was contoured on the 3DCT image without referencing the PET/CT image. The GTVPET-ref was contoured on the 3DCT image referencing the PET/CT image. The GTVPET-reg was contoured on the deformed registration image derived from 3DCT and PET/CT. Differences in the position, volume, length, conformity index (CI), and degree of inclusion (DI) among the target volumes were determined.ResultsThe centroid distance in the three directions between two different GTVs showed no significant difference (P > 0.05). No significant difference was found among the groups in the tumor volume (P > 0.05). The median DI values of the GTVPET-reg and GTVPET-ref in the GTV3D were 0.82 and 0.86, respectively (P = 0.006). The median CI values of the GTV3D in the GTVPET-reg and GTVPET-ref were 0.68 and 0.72, respectively (P = 0.006).ConclusionsPET/CT can be used to optimize the definition of the target volume in EC. However, no significant difference was found between the GTVs delineated based on visual referencing or deformable registration whether using the volume or position. So, in the absence of planning PET–CT images, it is also feasible to delineate the GTV of primary thoracic EC with reference to the diagnostic PET–CT image.

Objective: The aim of this study was to evaluate the safety and efficacy of anlotinib combined with chemoradiotherapy for treating esophageal squamous cell carcinoma (ESCC) using patient-derived xenografts (PDXs).Methods: PDX-bearing mice were randomly divided into five groups, as follows: control group receiving normal saline, the group receiving radiotherapy, the group receiving cisplatin combined with radiotherapy, the group receiving anlotinib combined with radiotherapy, and the group receiving anlotinib, and cisplatin combined with radiotherapy. Tumor volumes and body weights were measured three times weekly for 2 weeks. The PDXs were initially assessed by comparing the histology of the original patient tumor tissues with that of the corresponding serially passaged xenografts by hematoxylin and eosin (H&E) and P63 staining. Then, expression of Bax, c-PARP, PCNA, and CD31 was detected using immunohistochemistry, and apoptosis was detected by a TUNEL assay. Cytokines released into plasma were analyzed using protein chip technology. Finally, two case studies of ESCC patients were presented to further verify the results observed in the PDX models.Results: The pathological characteristics of the serially passaged patient tumor-derived xenografts established in our study were in line with those of the original ESCC patient samples. The group receiving anlotinib and cisplatin plus radiotherapy exhibited the strongest antitumor response among the groups. Moreover, the ideal anticancer effects of anlotinib combined with chemoradiotherapy observed in clinical patients were consistent with the results observed in the PDX models, and no serious side effects were observed during treatment.Conclusions: Combination therapy with anlotinib and chemoradiotherapy may be an effective regimen for the treatment of advanced ESCC.

Objective: To investigate the effect of anatomic and technical parameters on the incidental internal mammary lymph node (IMN) irradiation (IIMNI) dose among postmastectomy patients.Methods: We retrospectively delineated the IMN on planning CT images from 138 patients who had undergone postmastectomy radiotherapy (PMRT). We analyzed the IIMNI dose coverage and its relationship with anatomic and technical parameters.Results: The IIMNI mean dose was 32.85 ± 9.49 Gy, and 10 of 138 patients (7.25%) treated with PMRT received ≥45 Gy. In univariate analysis, the body weight, body mass index, body surface area, thoracic transverse diameter (DT), ratio of DT to the thoracic anteroposterior diameter (DAP)(RT/AP), planning target volume of IMN (PTVIMN) included in PTV (IMNin) and the ratio of IMNin to PTVIMN (RIMNin) and PTV posterior border were the parameters affecting IIMNI dose. In multivariate analysis, body weight, RT/AP, and RIMNin were correlative factors that affected IIMNI dose.Conclusions: For patients who underwent PMRT without IMN irradiation (IMNI), there was a wide variety in IIMNI doses. A minority of patients had adequate IIMNI dose coverage, and the higher IIMNI doses were associated with the less body weights and more RIMNin.

Background: Although the supine position remains the dominant position for external-beam partial breast irradiation (EB-PBI), the advantages of administering EB-PBI in the prone position have been recognized. The interobserver variability between target volumes delineated in the different positions for EB-PBI after breast-conserving surgery needs to be investigated.Methods: Twenty-seven patients suitable for EB-PBI were enrolled from July 2016 to April 2017. Supine and prone simulation CT images were sequentially acquired for all enrolled patients during free breathing. Five experienced radiotherapists delineated the target volumes for all patients on supine and prone simulation CT images. The selected parameters, including target volumes, the coefficient of variation (COV), the matching degree (MD), and so on, were calculated to analyze the interobserver variability.Results: Regardless of the patient position, the interobserver variability between tumor bed (TB) and clinical target volume (CTV) measurements in supine and prone positions were statistically significant (F = 31.34, 19.467; 44.000, 41.985; P = 0.000, 0.001; 0.000, 0.001). The interobserver variability of COVCTV was significantly greater in the supine position than in the prone position (T = 2.64, P = 0.014). Furthermore, the interobserver variabilities of MDTB and MDCTV were statistically lower in the supine position than in the prone position (Z = −3.460, −3.195, P = 0.000, 0.001).Conclusion: When delineating the target volume for EB-PBI, the interobserver variability in the prone position was lower than that in the supine position. Hence, the administration of EB-PBI in the prone position during free breathing is a reasonable option.

Accurate estimation of CH4 fluxes in alpine peatland of the Qinghai-Tibetan Plateau under extreme drought is vital for understanding the global carbon cycle and predicting future climate change. However, studies on the impacts of extreme drought on peatland CH4 fluxes are limited. Abstract: Dock8 deficiency leads to immunodeficiency, and the role of Dock8 in B-cell development and function has been revealed; however, the role of DocK8 on B-cell receptor (BCR) signaling and function of memory B cells remains elusive. In this study, we generated a Dock8 knockout mouse model and collected peripheral blood mononuclear cells from Dock8 patients to study the effect of Dock8 deficiency on the BCR signaling and activation of memory B cells with confocal microscopy and total internal reflection fluorescence microscopy. The activation of key, positive upstream BCR signaling molecules, pCD19 and phosphorylated Brutons tyrosine kinase (pBtk), is reduced. Interestingly, the total protein and activated levels of Wiskott–Aldrich syndrome protein (WASP) are decreased in Dock8-deficient mouse B cells. Our previous research has shown that WASP positively regulates cd19 transcription; furthermore, we found that Dock8 regulates cd19 transcription. What we found in Dock8 patients can be a phenotype copied from Dock8 mice. The early activation of memory B cells from Dock8 patients is disrupted with reduced BCR clustering, B-cell spreading, and signalosome recruitment into the degree of naïve B cells, as well as the transition from naïve B cells to unswitched memory B cells. Overall, our study provides a novel mechanism for Dock8 regulation of BCR signaling by regulating cd19 transcription, as well as the underlying mechanism of noncompetence of memory B cells in Dock8 patients.

In this paper, we investigate the fault-tolerant consensus problems for multi-agent systems with actuator faults. Based on the relative state information, fully distributed adaptive protocols are designed for these agents described by Lipschitz nonlinear dynamics under leaderless and leader-follower communication structures. As the special case, for linear multi-agent systems, fault-tolerant consensus problems are also solved with the proposed protocols. First, for the leaderless multi-agent systems, all agents reach an agreement on a common value with undirected connected communication graphs. Second, for the leader-follower multi-agent systems, in which the state of the leader is only available to a subset of the followers, the state of each follower converges to the state of the leader, asymptotically. A distinctive feature of this paper is that the adaptive protocols here are independent of the eigenvalues of the Laplacian matrix associated with the whole communication graph, which means the protocols can be implemented by each agent in a fully distributed fashion. Simulation examples are provided to illustrate the theoretical results.

A general strategy, i.e. simultaneous realization of the conductive segregation network microstructure and minimal surface porous macrostructure by means of selective laser sintering (SLS) 3D printing is proposed for devices fabrication. For the purpose of strategy demonstration, a flexible piezoresistive sensor is fabricated by SLS 3D printing with the use of self-made single-walled carbon nanotubes (SWCNTs) wrapped thermoplastic polyurethane (TPU) powders. The SLS-printed TPU/SWCNTs composite possesses an ultra-low conductive percolation threshold and excellent mechanical strength due to the construction of electrically conductive segregation network microstructure in polymer matrix, what is a prerequisite to a flexible piezoresistive sensor. The minimal surfaces including the Schwarz, Gyroid- and Diamond-structures have been used to design a 3D-ordered porous macrostructure of the sensor. The Schwarz structure has been found to be capable of producing the best piezoresistive properties of the SWCNTs/TPU composite sensor with a gauge factor (GF) much higher than that for the Gyroid- and Diamond-structures. This phenomenon is well understood by the relationship between the piezoresistive sensitivity and the localized strain distribution revealed by Finite Element Modeling. Abstract: As a key regulator of hippo signaling pathway, Mst kinases are emerging as one of the key signaling molecules that influence cell proliferation, organ size, cell migration, and cell polarity. In B lymphocytes, Mst1 deficiency causes the developmental defect of marginal zone (MZ) B cells, but how Mst1 regulates B-cell receptor (BCR) activation and differentiation remains elusive. Using genetically manipulated mouse models and total internal reflection fluorescence microscopy, we have demonstrated that Mst1 positively regulates BCR signaling via modulating CD19 transcriptional levels. Consistent with this, Mst1-deficient mice exhibited reduced BCR signaling, which is concurrent with defective BCR clustering and B-cell spreading on stimulatory lipid bilayers. The disruption of CD19-mediated Btk signaling by Mst1 deficiency leads to the severe defect in the differentiation of MZ and germinal center B cells. Mechanistic analysis showed that Mst1 upregulates the messenger RNA level of CD19 via regulating the transcriptional factor TEAD2 that directly binds to the consensus motif in the 3′ untranslated region of cd19. Overall, our results reveal a new function of Mst1 in B cells and the mechanism by which Mst1 regulates the activation and differentiation of peripheral B cells.

This paper solves the distributed fault detection (FD) problem for heterogeneous multi-agent systems (MAS). For a heterogeneous MAS, we adopt a distributed control law to realise cooperative output regulation (COR) when no fault occurs in the MAS, and propose a state-feedback-based FD scheme, where the adopted distributed control law and proposed FD scheme all utilise state information. Furthermore, we consider the condition that state information is unmeasurable, the output-feedback-based distributed FD scheme is proposed, and the adopted distributed control law also utilises measurement output. Finally, two numerical examples are utilised to verify that the proposed distributed FD schemes could locate and remove the faulty agent in time.

The Zoige alpine peatlands are one of the highest and largest alpine peatlands in the world and play an important role in the global carbon cycle. Drainage is the main disturbance at Zoige, and the drawdown of the water table level changes CH4 emissions. There is still much uncertainty relating to how CH4 emissions respond to multiple water table levels. Here, we simulated six gradients (−30 cm, −20 cm, −10 cm, 0 cm, 10 cm, and 20 cm) of the water table level through a mesocosm manipulation experiment in the Zoige peatlands. The water table level had a significant effect on CH4 emissions. CH4 emissions did not change with water table levels from −30 cm to −10 cm, but significantly increased as the water table level increased above −10 cm. A significant log-linear relationship (R2 = 0.44, p < 0.001) was found between CH4 emissions and a water table level range from −10 to 20 cm. This study characterized the responses of CH4 emissions to multiple water table levels and provide additional data for accurately evaluating CH4 emissions. The results of this study also have several conservation implications for alpine peatlands.

Coastal wetlands are considered as a significant sink of global carbon due to their tremendous organic carbon storage. Coastal CO2 and CH4 flux rates play an important role in regulating atmospheric CO2 and CH4 concentrations. However, the relative contributions of vegetation, soil properties, and spatial structure on dry-season ecosystem carbon (C) rates (net ecosystem CO2 exchange, NEE; ecosystem respiration, ER; gross ecosystem productivity, GEP; and CH4) remain unclear at a regional scale. Here, we compared dry-season ecosystem C rates, plant, and soil properties across three vegetation types from 13 locations at a regional scale in the Yellow River Delta (YRD). The results showed that the Phragmites australis stand had the greatest NEE (-1365.4 μmol m-2 s-1), ER (660.2 μmol m-2 s-1), GEP (-2025.5 μmol m-2 s-1) and acted as a CH4 source (0.27 μmol m-2 s-1), whereas the Suaeda heteroptera and Tamarix chinensis stands uptook CH4 (-0.02 to -0.12 μmol m-2 s-1). Stepwise multiple regression analysis demonstrated that plant biomass was the main factor explaining all of the investigated carbon rates (GEP, ER, NEE, and CH4); while soil organic carbon was shown to be the most important for explaining the variability in the processes of carbon release to the atmosphere, i.e., ER and CH4. Variation partitioning results showed that vegetation and soil properties played equally important roles in shaping the pattern of C rates in the YRD. These results provide a better understanding of the link between ecosystem C rates and environmental drivers, and provide a framework to predict regional-scale ecosystem C fluxes under future climate change.

The scarcity, richness, and other important physiological functions of D-psicose make it crucial to increase the yield of D-psicose. The production of D-psicose can be accomplished by D-psicose 3-epimerase (DPEase) from Clostridium bolteae (CbDPEase) catalyzing the substrate D-fructose. Although the catalytic efficiency of the CbDPEase has been raised via using the site-directed mutagenesis (Y68I/G109P) technique, structure-activity relationship in the wild-type CbDPEase and Y68I/G109P mutant is currently poorly understood. In our study, a battery of molecular modeling methods [homology modeling, adaptive steered molecular dynamics (ASMD) simulations, and Molecular Mechanics/Generalized Born Surface Area (MM-GB/SA)], combined with protein structure networks, were employed to theoretically characterize the reasons for the differences in the abilities of the D-fructose catalyzed by the wild-type CbDPEase and Y68I/G109P mutant. Protein structure networks demonstrated that site-directed mutagenesis enhanced the connectivity between D-fructose and CbDPEase, leading to the increased catalytic efficiency mediated by the functional residues with high betweenness. During the dissociation of the D-fructose from the Y68I/G109P mutant, planes of benzene rings of F248 and W114 could be continuously parallel to the stretching direction of D-fructose. It made the tunnel have an open state and resulted in the stable donor-π interactions between D-fructose and the benzene rings around 18Å. The stronger substrate-protein interactions were detected in the Y68I/G109P mutant, instead of in the wild-type CbDPEase, which were consistent with the binding free energy and Potential Mean of Force (PMF) results. The theoretical results illustrated the reasons that Y68I/G109P mutations increased the catalytic efficiency of CbDPEase and could be provided the new clue for further DPEase engineering.

As a critical linker between mTORC1 and mTORC2, Akt is important for the cell metabolism. The role of Akt in the function and development of B and T cells is well characterized, however, the role of Akt for development and function of iNKT cells is unknown. iNKT cells bridge the adaptive and innate immunity, and in this study, we found that the differentiation of NKT17 cells and IL17 production of NKT17 cells were disrupted in Akt2 KO mice. ICOS has been demonstrated to be critical for the differentiation of NKT17 cells and we found that ICOS mRNA and protein expression was reduced in Akt2 KO iNKT cells. As a consequence, phosphorylation of FoxO-1 was downregulated in Akt2 KO thymocytes but the sequestration of FoxO-1 in the nucleus of Akt2 KO iNKT cells was increased. The negative feedback loop between ICOS and FoxO-1 has been demonstrated in CD4+T follicular helper cells. Therefore our study has revealed a new intracellular mechanism in which Akt2 regulates ICOS expression via FoxO-1 and this signaling axis regulates the differentiation and function of NKT17 cells. This study provides a new linker between cell metabolism and function of iNKT cells.

As the central hub of the metabolism machinery, the mammalian target of rapamycin complex 2 (mTORC2) has been well studied in lymphocytes. As an obligatory component of mTORC2, the role of Rictor in T cells is well established. However, the role of Rictor in B cells still remains elusive. Rictor is involved in B cell development, especially the peripheral development. However, the role of Rictor on B cell receptor (BCR) signaling as well as the underlying cellular and molecular mechanism is still unknown. This study used B cell-specfic Rictor knockout (KO) mice to investigate how Rictor regulates BCR signaling. We found that the key positive and negative BCR signaling molecules, phosphorylated Brutons tyrosine kinase (pBtk) and phosphorylated SH2-containing inositol phosphatase (pSHIP), are reduced and enhanced, respectively, in Rictor KO B cells. This suggests that Rictor positively regulates the early events of BCR signaling. We found that the cellular filamentous actin (F-actin) is drastically increased in Rictor KO B cells after BCR stimulation through dysregulating the dephosphorylation of ezrin. The high actin-ezrin intensity area restricts the lateral movement of BCRs upon stimulation, consequently reducing BCR clustering and BCR signaling. The reduction in the initiation of BCR signaling caused by actin alteration is associated with a decreased humoral immune response in Rictor KO mice. The inhibition of actin polymerization with latrunculin in Rictor KO B cells rescues the defects of BCR signaling and B cell differentiation. Overall, our study provides a new pathway linking cell metablism to BCR activation, in which Rictor regulates BCR signaling via actin reorganization.

In order to obtain a novel preclinical anti-AD drug candidate, we synthesized a series of novel hydroxyl chalcone analogs which possessed anti-free radical activity, and screened their effects on scavenging 2,2-diphenyl-1-picrylhydrazyl (DPPH) and OH free radicals in vitro. Compound C7, 4,2'-dihydroxy-3,5-dimethoxychalcone was found to have potent activity in these anti-free radical activity tests. Further research revealed that C7 could elevate glutathione peroxidase (GSH-PX) and super oxide dismutase (SOD) levels and lower malonaldehyde (MDA) level in vivo in the Alzheimer’s model. The indication of C7’s effect on AD needs further study.

How to effectively combine remote sensing data with the eddy covariance (EC) technique to accurately quantify gross primary production (GPP) in coastal wetlands has been a challenge and is also important and necessary for carbon (C) budgets assessment and climate change studies at larger scales. In this study, a satellite-based Vegetation Photosynthesis Model (VPM) combined with EC measurement and Moderate Resolution Imaging Spectroradiometer (MODIS) data was used to evaluate the phenological characteristics and the biophysical performance of MODIS-based vegetation indices (VIs) and the feasibility of the model for simulating GPP of coastal wetland ecosystems. The results showed that greenness-related and water-related VIs can better identify the green-up and the senescence phases of coastal wetland vegetation, corresponds well with the C uptake period and the phenological patterns that were delineated by GPP from EC tower (GPPEC). Temperature can explain most of the seasonal variation in VIs and GPPEC fluxes. Both enhanced vegetation index (EVI) and water-sensitive land surface water index (LSWI) have a higher predictive power for simulating GPP in this coastal wetland. The comparisons between modeled GPP (GPPVPM) and GPPEC indicated that VPM model can commendably simulate the trajectories of the seasonal dynamics of GPPEC fluxes in terms of patterns and magnitudes, explaining about 85% of GPPEC changes over the study years (p < 0.0001). The results also demonstrate the potential of satellite-driven VPM model for modeling C uptake at large spatial and temporal scales in coastal wetlands, which can provide valuable production data for the assessment of global wetland C sink/source.

Caldicellulosiruptor lactoaceticus 6A, an anaerobic and extremely thermophilic bacterium, uses natural xylan as carbon source. The encoded genes of C. lactoaceticus 6A for glycoside hydrolase (GH) provide a platform for xylan degradation. The GH family 10 xylanase (Xyn10A) and GH67 α-glucuronidase (Agu67A) from C. lactoaceticus 6A were heterologously expressed, purified and characterized. Both Xyn10A and Agu67A are predicted as intracellular enzymes as no signal peptides identified. Xyn10A and Agu67A had molecular weight of 47.0 kDa and 80.0 kDa respectively as determined by SDS-PAGE, while both appeared as homodimer when analyzed by gel filtration. Xyn10A displayed the highest activity at 80 °C and pH 6.5, as 75 °C and pH 6.5 for Agu67A. Xyn10A had good stability at 75 °C, 80 °C, and pH 4.5-8.5, respectively, and was sensitive to various metal ions and reagents. Xyn10A possessed hydrolytic activity towards xylo-oligosaccharides (XOs) and beechwood xylan. At optimum conditions, the specific activity of Xyn10A was 44.6 IU/mg with beechwood xylan as substrate, and liberated branched XOs, xylobiose, and xylose. Agu67A was active on branched XOs with methyl-glucuronic acids (MeGlcA) sub-chains, and primarily generated XOs equivalents and MeGlcA. The specific activity of Agu67A was 1.3 IU/mg with aldobiouronic acid as substrate. The synergistic action of Xyn10A and Agu67A was observed with MeGlcA branched XOs and xylan as substrates, both backbone and branched chain of substrates were degraded, and liberated xylose, xylobiose, and MeGlcA. The synergism of Xyn10A and Agu67A provided not only a thermophilic method for natural xylan degradation, but also insight into the mechanisms for xylan utilization of C. lactoaceticus.

Ipomoea stolonifera (I. stolonifera) has been used for the treatment of inflammatory diseases including rheumatism and rheumatoid arthritis in Chinese traditional medicine. However, the anti-inflammatory activity of I. stolonifera has not been elucidated. For this reason, the anti-inflammatory activity of n-butanol extract of I. stolonifera (BE-IS) was evaluated in vivo by using acute models (croton oil-induced mouse ear edema, carrageenan-induced rat paw edema, and carrageenan-induced rat pleurisy) and chronic models (cotton pellet-induced rat granuloma, and complete Freund's adjuvant (CFA)-induced rat arthritis). Results indicated that oral administration of BE-IS significantly attenuated croton oil-induced ear edema, decreased carrageenan-induced paw edema, reduced carrageenan-induced exudates and cellular migration, inhibited cotton pellet-induced granuloma formation and improved CFA-induced arthritis. Preliminary mechanism studies demonstrated that BE-IS decreased the levels of myeloperoxidase (MPO) and malondialdehyde (MDA), increased the activity of anti-oxidant enzyme superoxide dismutase (SOD) in vivo, and reduced the production of nitric oxide (NO), prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α), interleukin (IL)-1β and IL-6 in lipopolysaccharide-activated RAW264.7 macrophages in vitro. Results obtained in vivo and in vitro demonstrate that BE-IS has considerable anti-inflammatory potential, which provided experimental evidences for the traditional application of Ipomoea stolonifera in inflammatory diseases.

The xylanolytic extremely thermophilic bacterium Caldicellulosiruptor owensensis provides a promising platform for xylan utilization. In the present study, two novel xylanolytic enzymes, GH10 endo-β-1,4-xylanase (Coxyn A) and GH39 β-1,4-xylosidase (Coxyl A) encoded in one gene cluster of C.owensensis were heterogeneously expressed and biochemically characterized. The optimum temperature of the two xylanlytic enzymes was 75°C, and the respective optimum pH for Coxyn A and Coxyl A was 7.0 and 5.0. The difference of Coxyn A and Coxyl A in solution was existing as monomer and homodimer respectively, it was also observed in predicted secondary structure. Under optimum condition, the catalytic efficiency (kcat/Km) of Coxyn A was 366 mg ml(-1) s(-1) on beechwood xylan, and the catalytic efficiency (kcat/Km) of Coxyl A was 2253 mM(-1) s(-1) on pNP-β-D-xylopyranoside. Coxyn A degraded xylan to oligosaccharides, which were converted to monomer by Coxyl A. The two intracellular enzymes might be responsible for xylooligosaccharides utilization in C.owensensis, also provide a potential way for xylan degradation in vitro.

With the aid of symbolic computation, we obtain the symmetry transformations of the (2 + 1)-dimensional Caudrey-Dodd-Gibbon-Kotera-Sawada (CDGKS) equation by Lou’s direct method which is based on Lax pairs. Moreover, we use the classical Lie group method to seek the symmetry groups of both the CDGKS equation and its Lax pair and then reduce them by the obtained symmetries. In particular, we consider the reductions of the Lax pair completely. As a result, three reduced (1 + 1)-dimensional equations with their new Lax pairs are presented and some group-invariant solutions of the equation are given.

The lipid phosphatase Sac1 cycles between endoplasmic reticulum and cisternal Golgi compartments. In proliferating mammalian cells, a canonical dilysine motif at the C-terminus of Sac1 is required for coatomer complex-I (COP-I)-binding and continuous retrieval to the ER. Starvation triggers accumulation of Sac1 at the Golgi. The mechanism responsible for Golgi retention of Sac1 is unknown. Here we show that the first of the two transmembrane regions in human SAC1 (TM1) functions in Golgi localization. A minimal construct containing only TM1 and the adjacent flanking sequences is concentrated at the Golgi. Transplanting TM1 into transferrin receptor 2 (TfR2) induces Golgi accumulation of this normally plasma membrane and endosomal protein, indicating that TM1 is sufficient for Golgi localization. In addition, we determined that the N-terminal cytoplasmic domain of SAC1 also promotes Golgi localization, even when TM1 is mutated or absent. We conclude that the distribution of SAC1 within the Golgi is controlled via both passive membrane thickness-dependent partitioning of TM1 and a retention mechanism that requires the N-terminal cytoplasmic region.
