Abstract Resistance to immunotherapy in bladder cancer has greatly limited its clinical application. Through single-cell sequencing, we determined that ACSM6, an oncogene that is highly expressed in bladder cancer, promotes the abilities of proliferation, cloning, migration, and invasion. The key point is that ACSM6 can also lead to a non-inflammatory immune microenvironment by inhibiting the chemotaxis and tumor killing ability of CD8+ T cells. Survival analysis revealed that high ACSM6 expression was associated with shorter overall survival in the immunotherapy cohort. In summary, ACSM6 is expected to become a novel biomarker for predict bladder cancer progression.

Objective To evaluate the effectiveness of Roche Cobas s 201 in detecting HBV by analyzing its blood nucleic acid testing (NAT) results. Methods The results were grouped according to the enzyme-linked immunosorbent assay (ELISA) and NAT minipool test (MP), NAT individual test (ID) and repeated NAT ID test (rID), and categorized into 4 groups as ELISA+ /NAT(ID)+ , ELISA+ /NAT(rID)+ , ELISA-/NAT(ID)+ and ELISA-/NAT(rID)+ . The data were statistically analyzed to explore whether there was a difference in the detection of reactive results by repeated NAT, and the correlation between cycle threshold (Ct) and nucleic acid detection rate for NAT-reactive samples with different ELISA results. The true infection status of blood donors was further analyzed by supplementary tests, including NAT systems and chemiluminescence serological marker assays using other methodologies. Results A total of 1 691 groups of 766 293 blood donor samples were HBV NAT(MP)+ , of which 1 418 groups(83.86%) were detected with reactive results (1 418 HBV NAT+ , 7 090 NAT-), and there were still 273 groups (16.14%) that remained undetected after repeated testing[a total of 1 638 NAT-, Ct(MP): 39.49±3.62]. Of the HBV NAT+ , 881(62.13%) were ELISA+ /NAT(ID)+ , 19(1.34%) were ELISA+ /NAT(rID)+ , 451(31.81%) were ELISA-/NAT(ID)+ , and 67(4.72%) were ELISA-/NAT(rID)+ . Abstract Background Although immunotherapy shows tremendous potential in the treatment of bladder cancer (BLCA), the overall prognosis and response rates to immunotherapy in BLCA remain suboptimal. Methods We performed an extensive evaluation of glycosyltransferase expression patterns in BLCA patients by analyzing 210 glycosyltransferase-related genes. Subsequently, we established correlations between these glycosyltransferase patterns, prognosis, and tumor microenvironment (TME) phenotypes. To offer personalized patient assessments, we developed a glycosyltransferase risk score that accurately predicts prognosis, TME phenotypes, and molecular subtypes. Importantly, we developed a RNA-seq cohort, named Xiangya cohort, to validate our results. Results Two distinct patterns of glycosyltransferase expression were identified, corresponding to inflamed and noninflamed TME phenotypes, and demonstrated the potential to predict prognosis. We developed and validated a comprehensive risk score that accurately predicted individual patient prognosis in the TCGA-BLCA cohort. Additionally, we constructed a nomogram that integrated the risk score with several key clinical factors. Importantly, this risk score was successfully validated in external cohorts, including the Xiangya cohort and GSE48075. Furthermore, we discovered a positive correlation between this risk score and tumor-infiltrating lymphocytes in both the TCGA-BLCA and Xiangya cohorts, suggesting that patients with a higher risk score exhibited an inflamed TME phenotype and were more responsive to immunotherapy. Finally, we observed that the high and low risk score groups were consistent with the luminal and basal subtypes of BLCA, respectively, providing further validation of the risk score’s role in the TME in terms of molecular subtypes. Conclusions Glycosyltransferase patterns exhibit distinct TME phenotypes in BLCA. Our comprehensive risk score provides a promising approach for prognostic prediction and assessment of immunotherapy efficacy, offering valuable guidance for precision medicine.

Objective In situ leaching is a uranium extraction process in which a solution reacts with uranium-bearing minerals in a saturated aquifer and then uranium in the flowing solution is extracted through exchange. To understand the internal structure and seepage path of the 512 deposits, Methods representative wallrock and ore cores of the deposit were selected for CT scanning, and the pore throat parameters were obtained via image noise reduction filtering, image segmentation to extract pores, and the construction of pore network models. The changes in the absolute permeability, tortuosity and seepage velocity are simulated with Avizo software. Results The results showed that the porosity of the wallrock was 15.42%, the connected porosity was 9.61%, the ore porosity was 15.18%, the connected porosity was 13.82%, and the water permeability was better than that of the wallrock. The high-density materials in the wallrock are metal minerals, accounting for approximately 0.54%, and most of the high-density minerals in the ore are secondary uranium minerals, accounting for 1.06%. It can be fully in contact with the solution during the leaching process. Conclusion There is strong heterogeneity inside the pores, which causes the flow rate to gradually decrease in the flow path. The number of large pores connecting the wallrock and ore is greater than that of small pores, indicating that large pores are the main factors determining the degree of pore development. According to the velocity streamlines, although there is a blockage area in the wallrock and ore, the circulation area is dominant.

Abstract Background Screw loosening is a commonly reported issue following spinal screw fixation and can lead to various complications. The initial cause of screw loosening is biomechanical deterioration. Previous studies have demonstrated that modifications in screw design can impact the local biomechanical environment, specifically the stress distribution on bone-screw interfaces. There are several different designs of screw tips available for clinically used pedicle screws; however, it remains unclear whether these variations affect the local stress distribution and subsequent screw anchorage ability. Methods This study conducted comprehensive biomechanical research using polyurethane foam mechanical tests and corresponding numerical simulations to investigate this topic. Models of pedicle screw-fixed osteoporotic polyurethane foam were created with two different clinically used screw tip designs (flat and steep) featuring varying tip lengths, taper angles, and diameters, as well as identical flank overlap areas and thread designs. The anchorage ability of the different models was assessed through toggle and pull-out test. Additionally, numerical mechanical models were utilized to compute the stress distributions at the screw and bone-screw interfaces in the different models. Results Mechanical tests revealed superior anchorage ability in models utilizing flat-tipped screws. Furthermore, numerical modeling indicated improved anchorage ability and reduced stress concentration tendency in these models. Conclusion Changes in screw tip design can significantly impact the biomechanical anchoring capability of screws. Specifically, flatter tip pedicle screws may mitigate the risk of screw loosening by alleviating stress concentration on bone-screw interfaces.

Abstract Background This study aims to investigate the role of shear wave elastography (SWE) and connective tissue growth factor (CTGF) in the assessment of papillary thyroid carcinoma (PTC) prognosis. Methods CTGF expression was detected with immunohistochemistry. Clinical and pathological data were collected. Parameters of conventional ultrasound combined with SWE were also collected. The relationship among CTGF expression, ultrasound indicators, the elastic modulus and the clinicopathological parameters were analyzed. Results Univariate analysis showed that patients with high risk of PTC were characterized with male, Uygur ethnicity, increased expression of CTGF, convex lesions, calcified, incomplete capsule, intranodular blood flow, rear echo attenuation, cervical lymph node metastasis, lesions larger than 1 cm, psammoma bodies, advanced clinical stage, increased TSH and high value in the shear modulus (P

In the preparation of magnesium by Pidgeon process, the phenomenon slag pellets sticking on the wall of reduction pot always appear, and the glaze sticking on the inner wall of the reduction pot is difficult to remove. The mechanism of this phenomenon is studied in this work by X-ray fluorescence spectrometer (XRF) measurement, electron probe microanalyzer scanning (EPMA) analysis, differential scanning calorimetry (DSC) analysis, and thermodynamic calculations. The main components of the glaze are MgO, Ca12Al14F2O32, CaF2, CaO, and a small amount of Ca4Si2O7F2. The solid-liquid transition temperature of Ca12Al14F2O32 and CaF2 is close to the production temperature of Pidgeon process, which leads to the bonding between the slag pellets and the pot wall. The loss of CaF2 in glaze layer will reduce the total amount of liquid phase and increase the temperature at which Ca12Al14F2O32 is completely transformed into liquid phase, which causes glaze layer sticking on the inner wall of the reduction pot.

BackgroundColorectal cancer (CRC) is a malignant tumor that poses a significant threat to human health due to rising incidence and mortality rates. In recent years, immune checkpoint blockade (ICB) therapy, represented by Programmed death receptor 1 (PD-1), T-lymphocyte-associated protein 4 (CTLA-4), and others, has been widely applied in CRC and has achieved encouraging results in some patients and has become a hot topic in both clinical and basic research.ObjectiveThis study undertakes a comprehensive bibliometric analysis of ICB research in CRC, aiming to evaluate the current status, identify future trends, and provide scientific insights for researchers and decision-makers.MethodsUtilizing the Web of Science Core Collection (WoSCC), articles focusing on ICB in CRC from 2000 to 2022 were retrieved. Knowledge mapping and bibliometric analysis were conducted using tools such as CiteSpace, VOSviewer, SCImago Graphicay, and the R package bibliometrix.Results6,718 publications were analyzed from 24,846 institutions across 639 regions. Temporally, ICB research in CRC is rapidly advancing, led by the USA and China with extensive global collaborations. Sun Yat-sen University from China stands out as the institution with the highest number of publications. Professor Thierry Andre from Sorbonne University in France is identified as a prolific author in this field, engaging in extensive collaboration for clinical trials on a global scale. Publications related to this research topic were published in 1,142 academic journals, demonstrating a positive co-citation relationship. Key clustering and burst terms analysis indicate that current research on ICB in CRC has shifted from basic experiments to clinical trials and from universal healthcare to precision medicine.ConclusionICB therapies have shown substantial progress in CRC, highlighting their therapeutic potential. Research trends emphasize deeper drug mechanisms, treatment efficacy prediction, managing immune-related adverse events, and exploring novel drug delivery methods. Abstract This study aims to measure anatomical data of the capitate bone, develop an external fixator for treating late-stage osteonecrosis of lunate through Ilizarov technique, and evaluate its biomechanical performance. We selected eight wrist joint specimens to measure various parameters of the capitate bone, including its length, the distance from the junction of capitate head and body to the proximal end, as well as the width of its proximal head and distal body. Additionally, we measured these same indicators in 107 patients who had undergone wrist X-ray examination. Based on our measurements, we categorized the capitate bone into two groups and designed two types of capitate bone Ilizarov external fixator (CIEF) for it. Then, we compared it with the orthofix external fixator (OEF) through dynamic fatigue biomechanical experiments and pull-out resistance experiments. The results of the measurement revealed two categories of general patterns in the capitate bone. The first type maintains a consistent longitudinal axis between the proximal and distal ends. The second type is characterized by its proximal end being close to the radial side and its distal end being close to the ulnar side. Abstract Purpose To compare the outcomes of big-bubble deep anterior lamellar keratoplasty (BB-DALK) and penetrating keratoplasty (PKP) in the management of medically unresponsive Acanthamoeba keratitis (AK). Methods This retrospective study included 27 eyes of BB-DALK and 24 eyes of PKP from a tertiary ophthalmology care centre. Glucocorticoid eye drops were subsequently added to the treatment plan 2 months postoperatively based on the evaluation using confocal laser scanning microscopy. The clinical presentations, best-corrected visual acuity (BCVA), postoperative refractive outcomes, graft survival, and Acanthamoeba recurrence were analyzed. Results The AK patients included in the study were in stage 2 or stage 3, and the percentage of patients in stage 3 was higher in the PKP group (P = 0.003). Clinical presentations were mainly corneal ulcers and ring infiltrates, and endothelial plaques, hypopyon, uveitis and glaucoma were more common in the PKP group (P = 0.007). The BCVA and the graft survival rate showed no statistically significant differences between the two groups at 1 year after surgery. However, 3 years postoperatively, the BCVA of 0.71 ± 0.64 logMAR, the graft survival rate of 89.5%, and the endothelial cell density of 1899 ± 125 cells per square millimeter in the BB-DALK group were significantly better than those of the PKP group (P = 0.010, 0.046, and 0.032, respectively). 3 eyes (11.1%) in the BB-DALK group and 2 eyes (8.3%) in the PKP group experienced Acanthamoeba recurrence, but the rates showed no statistically significant difference between the two groups (P = 1.000). In the PKP group, immune rejection and elevated intraocular pressure were observed in 5 and 6 eyes, respectively. Conclusion Corneal transplantation is recommended for AK patients unresponsive to antiamoebic agents. The visual acuity and graft survival can be maintained after BB-DALK surgery. Acanthamoeba recurrence is not related to the surgical approach performed, whereas complete dissection of the infected corneal stroma and delayed prescribing of glucocorticoid eye drops were important to prevent recurrence.

Ochratoxin A (OTA) is a common mycotoxin that causes intestinal injury in humans and various animal species. OTA may lead to intestinal injury in offspring due to the maternal effect. The aim of this study was to investigate the mechanism of embryo injected with OTA induced jejunum injury in ducklings. The results showed that OTA disrupted the jejunum tight junctions in hatching ducklings, and promoted the secretion of inflammatory cytokines. And this inflammatory response was caused by the activation of the TLR4 signaling pathway. Moreover, embryo injected with OTA could cause damage to the intestinal barrier in 21-day-old ducks, characterized by shortened villi, crypt hyperplasia, disrupted intestinal tight junctions, increased level of LPS in the jejunum, activation of the TLR4 signaling pathway, and increased levels of pro-inflammatory cytokines. Meanwhile, OTA induced oxidative stress in the jejunum. And dysbiosis of gut microbiota was mainly characterized by an increased the relative abundance of Bacteroides, Megamonas, Fournierella, and decreased the relative abundance of Alistipes and Weissella. Interestingly, embryo injected with OTA did not induce these changes in the jejunum of antibiotics-treated 21-day-old ducks. In conclusion, embryo injected with OTA induced jejunum injury in ducklings by activating the TLR4 signaling pathway, which involvement of intestinal microbiota.

This paper considered the groundwater head fluctuation induced by tide and pumping in the coastal multi-layered aquifer system. The multi-layered aquifer system comprises an unconfined aquifer, an upper confined aquifer, and a lower confined aquifer. An aquiclude exists between each two aquifers. All the layers terminate at the coastline. The new analytical solutions describing groundwater head variation in the coastal multi-confined aquifer system are derived. Superposition principle and image methods are used for the derivation of the analytical solutions. Analytical solutions of different situations of without considering pumping, of without considering tidal effect, and of N-layered confined aquifers are also derived. The impacts of the parameters of the initial phase shift of tide, pumping rate, position of the pumping well, storage coefficient, and transmissivity on the groundwater head fluctuation are discussed. The analytical solutions are applied with application examples in fitting field observations and parameter estimations. The estimated values of the hydraulic conductivities in the upper and lower confined aquifers are within the range of the values obtained from the field experiments. The fitted results of the analytical solutions capture the main characteristics of groundwater head fluctuation affected by the tide and groundwater pumping. The study of groundwater head fluctuation in the coastal zone is helpful to understand the mechanism of seawater intrusion under the influence of tide and groundwater pumping.

Background: Drug resistance is the primary obstacle to advanced tumor therapy and the key risk factor for tumor recurrence and death. 5-Fluorouracil (5-FU) chemotherapy is the most common chemotherapy for individuals with colorectal cancer, despite numerous options. Methods: The Gene Expression Omnibus database was utilized to extract expression profile data of HCT-8 human colorectal cancer wild-type cells and their 5-FU-induced drug resistance cell line. These data were used to identify 5-FU resistance-related differentially expressed genes (5FRRDEGs), which intersected with the colorectal adenocarcinoma (COAD) transcriptome data provided by the Cancer Genome Atlas Program database. A prognostic signature containing five 5FRRDEGs (GOLGA8A, KLC3, TIGD1, NBPF1, and SERPINE1) was established after conducting a Cox regression analysis. We conducted nomogram development, drug sensitivity analysis, tumor immune microenvironment analysis, and mutation analysis to assess the therapeutic value of the prognostic qualities. Results: We identified 166 5FRRDEGs in patients with COAD. Subsequently, we created a prognostic model consisting of five 5FRRDEGs using Cox regression analysis. The patients with COAD were divided into different risk groups by risk score; the high-risk group demonstrated a worse prognosis than the low-risk group. Conclusion: In summary, the 5FRRDEG-based prognostic model is an effective tool for targeted therapy and chemotherapy in patients with COAD. It can accurately predict the survival prognosis of these patients as well as to provide the direction for exploring the resistance mechanism underlying COAD.

Abstract $$H_{\infty }$$ H ∞ state estimation is addressed for continuous-time neural networks in the paper. The norm-bounded uncertainties are considered in communication neural networks. For the considered neural networks with uncertainties, a reduced-order $$H_{\infty }$$ H ∞ state estimator is designed, which makes that the error dynamics is exponentially stable and has weighted $$H_{\infty }$$ H ∞ performance index by Lyapunov function method. Moreover, it is also given the devised method of the reduced-order $$H_{\infty }$$ H ∞ state estimator. Then, considering that sampling the output y(t) of the neural network at every moment will result in waste of excess resources, the event-triggered sampling strategy is used to solve the oversampling problem. In addition, a devised method is also given for the event-triggered reduced-order $$H_{\infty }$$ H ∞ state estimator. Finally, by the well-known Tunnel Diode Circuit example, it shows that a lower order state estimator can be designed under the premise of maintaining the same weighted $$H_{\infty }$$ H ∞ performance index, and using the event-triggered sampling method can reduce the computational and time costs and save communication resources.

This paper comprehensively introduces a new magnesium production technology the compound-vertical-retort technology, involving in the related fundamental researches, core equipment development, working flow, and technical characteristics. Scale-up test and an annual 1200-ton-magnesium demonstration-level test was conducted to confirm the rationality, reliability, and advancement of the equipment, system and process design. It is indicated that the new technology solved a series of problems of traditional silicothermic process including adhesion and glaze, short life of retort, low efficiency, high impurity of crystallized magnesium, large heat losses, and poor working environment, making a great technological breakthrough in this field. Representatively, the new well-designed ceramic-lined steel retort serves 2∼3 times in life than the normal retorts. The magnesium yield per retort is improved 4∼5 times, with purity of >99.8%. The energy consumption in reduction stage is reduced by more than 20%. The mechanical production is fully realized and operating environment is significantly improved.

Silica, as a high-quality reinforcing filler, can satisfy the requirements of high-performance green tread rubber with high wet-skid resistance, low rolling resistance, and low heat generation. However, the silica surface contains abundant silicon hydroxyl groups, resulting in a severe aggregation of silica particles in non-polar rubber matrix. Herein, we explored a carbon black (CB)/silica hybrid reinforcing strategy to prepare epoxidized natural rubber (ENR)-based vulcanizates. Benefiting from the reaction and interaction between the epoxy groups on ENR chains and the silicon hydroxyl groups on silica surfaces, the dispersion uniformity of silica in the ENR matrix was significantly enhanced. Meanwhile, the silica can facilitate the dispersity and reinforcing effect of CB particles in the ENR matrix. By optimizing the CB/silica blending ratios, we realized high-performance ENR vulcanizates with simultaneously improved mechanical strength, wear resistance, resilience, anti-aging, and damping properties, as well as reduced heat generation and rolling resistance. For example, compared with ENR vulcanizates with only CB fillers, those with CB/silica hybrid fillers showed ~10% increase in tensile strength, ~20% increase in elongation at break, and ~20% increase in tensile retention rate. These results indicated that the ENR compounds reinforced with CB/silica hybrid fillers are a promising candidate for high-performance green tread rubber materials.

The separate vertical wire (SVW) technique and the improved candy box (CB) technique have been proposed for treating inferior pole patellar fractures. However, there is still a lack of clear explanation regarding the location of the wire passing through the patella. Five models of SVW techniques were established in different positions. Finite element analysis was then conducted to determine the optimal bone tunnel position for the SVW technique. Based on these findings, six groups of finite element models were created for CB techniques. The maximum displacement and stress on both the patella and steel wire were compared among these groups under 100-N, 200-N, 300-N, 400-N, and 500-N force loads. The results indicated that, in the SVW technique, the steel wire group near the fracture end of the longitudinal bone tunnel showed minimal displacement and stress on the patella when subjected to different forces. On the other hand, in the CB technique, both the patella and wire experienced minimal stress when a transverse bone tunnel wire was placed near the upper posterior aspect of patella. In conclusion, the SVW technique may require the bone tunnel wire to be positioned near the fractured end of the lower pole of the patella. On the other hand, in CB technique, the transverse bone tunnel wire passing through the patella may be close to its upper posterior aspect. However, further validation is necessary through comprehensive finite element analysis and additional biomechanical experiments.

Background: The long-term prognosis for patients with osteosarcoma (OS) metastasis remains unfavourable, highlighting the urgent need for research that explores potential biomarkers using innovative methodologies. Methods: This study explored potential biomarkers for OS metastasis by analysing data from the Cancer Genome Atlas Program (TCGA) and Gene Expression Omnibus (GEO) databases. The synthetic minority oversampling technique (SMOTE) was employed to tackle class imbalances, while genes were selected using four feature selection algorithms (Monte Carlo feature selection [MCFS], Borota, minimum-redundancy maximum-relevance [mRMR], and light gradient-boosting machine [LightGBM]) based on the gene expression matrix. Four machine learning (ML) algorithms (support vector machine [SVM], extreme gradient boosting [XGBoost], random forest [RF], and k-nearest neighbours [kNN]) were utilized to determine the optimal number of genes for building the model. Interpretable machine learning (IML) was applied to construct prediction networks, revealing potential relationships among the selected genes. Additionally, enrichment analysis, survival analysis, and immune infiltration were performed on the featured genes. Results: In DS1, DS2, and DS3, the IML algorithm identified 53, 45, and 46 features, respectively. Using the merged gene set, we obtained a total of 79 interpretable prediction rules for OS metastasis. We subsequently conducted an in-depth investigation on 39 crucial molecules associated with predicting OS metastasis, elucidating their roles within the tumour microenvironment. Importantly, we found that certain genes act as both predictors and differentially expressed genes. Finally, our study unveiled statistically significant differences in survival between the high and low expression groups of TRIP4, S100A9, SELL and SLC11A1, and there was a certain correlation between these genes and 22 various immune cells. Conclusions: The biomarkers discovered in this study hold significant implications for personalized therapies, potentially enhancing the clinical prognosis of patients with OS.

BackgroundWhen employing the transcription-mediated amplification method for screening blood donors, there are some non-discriminatory reactive results which are screening assay reactive but HBV-DNA discriminatory assay negative. This raises concerns regarding the possibility of false positives among donors, which may lead to permanent deferral of blood donors and affect blood supply. This study aimed to elucidate the infection status of these non-discriminatory reactive blood donors and develop and validate a model to predict individualized hepatitis B status to establish an optimal screening strategy.MethodsSupplementary tests were conducted on initial non-discriminating reactive donations to determine their HBV infection status, including repeat testing, viral load, serological marker detection, and follow-up. Primary clinical variables of the donors were recorded. Based on the Akaike information criterion, a stepwise forward algorithm was used to identify the predictive factors for information and construct a predictive model. The optimal screening strategy was determined through cost-effectiveness analysis.ResultsAt the Blood Center of Zhejiang Province, 435 cases of initial non-discriminatory reactive donations were collected over two successive periods and sub-categorized through repeated testing into the following three groups: non-repeated positive group, non-discriminated positive group, and non-repeated HBV-DNA positive group. The HBV discriminatory rate increased after repeated testing (110/435, 25.29%). IntroductionWe aim to predict the pathological complete response (pCR) of neoadjuvant chemotherapy (NAC) in breast cancer patients by constructing a Nomogram based on radiomics models, clinicopathological features, and ultrasound features.MethodsUltrasound images of 464 breast cancer patients undergoing NAC were retrospectively analyzed. The patients were further divided into the training cohort and the validation cohort. The radiomics signatures (RS) before NAC treatment (RS1), after 2 cycles of NAC (RS2), and the different signatures between RS2 and RS1 (Delta-RS/RS1) were obtained. LASSO regression and random forest analysis were used for feature screening and model development, respectively. The independent predictors of pCR were screened from clinicopathological features, ultrasound features, and radiomics models by using univariate and multivariate analysis. The Nomogram model was constructed based on the optimal radiomics model and clinicopathological and ultrasound features. The predictive performance was evaluated with the receiver operating characteristic (ROC) curve.ResultsWe found that RS2 had better predictive performance for pCR. In the validation cohort, the area under the ROC curve was 0.817 (95%CI: 0.734-0.900), which was higher than RS1 and Delta-RS/RS1. The Nomogram based on clinicopathological features, ultrasound features, and RS2 could accurately predict the pCR value, and had the area under the ROC curve of 0.897 (95%CI: 0.866-0.929) in the validation cohort. The decision curve analysis showed that the Nomogram model had certain clinical practical value.DiscussionThe Nomogram based on radiomics signatures after two cycles of NAC, and clinicopathological and ultrasound features have good performance in predicting the NAC efficacy of breast cancer.

Abstract Background Maintaining effective reduction and firm fixation in inferior pole patellar fractures is a highly challenging task. There are various treatment methods available; although tension-band wiring combined with cerclage wiring (TBWC) is the mainstream approach, its effectiveness is limited. Herein, we propose and evaluate a new technique called candy box (CB), based on separate vertical wiring (SVW), for the treatment of inferior pole patellar fractures. Specifically, we provide biomechanical evidence for its clinical application. Methods Five fixation models were built: SVW combined with cerclage wiring (SVWC); TBWC; modified SVW with the middle (MSVW-A) or upper (MSVW-B) 1/3 of the steel wire reserved, and CB. A finite element analysis was performed to compare the displacement and stress under 100-N, 200-N, 300-N, 400-N and 500-N force loads. Three-dimensional printing technology was utilized to create fracture models, and the average displacement of each model group was compared under a 500-N force. Results The results of the finite element analysis indicate that CB technology exhibits significantly lower maximum displacement, bone stress, and wire stress compared to that with other technologies under different loads. The stable and safe operation of highway/railway lines is largely dependent on the dynamic behavior of subgrade fillings. Clay soils are widely used in subgrade construction and are compacted at different remolding water contents and compaction degrees, depending on the field conditions. As a result, their dynamic behaviors may vary, which have not been fully investigated until now. To clarify this aspect, a series of cyclic triaxial tests were carried out in this study with three typical remolding water contents (w = 19%, 24%, and 29%), corresponding to the optimum water content as well as its dry and wet sides, and two compaction degrees (Dc = 0.8 and 0.9), which were selected according to the field-testing data. Scanning electron microscopy (SEM) and mercury intrusion porosimetry (MIP) tests were also conducted on typical samples to investigate the corresponding soil fabric variations. The findings indicate the following: (a) The soil fabric at the optimum remolding water content and its dry side was characterized by a clay aggregate assembly with a bimodal pore size distribution. In contrast, the soil fabric on the wet side of the optimum water content consisted of dispersed clay particles with a unimodal pore size distribution. (b) As the compaction degree increased, to ensure the optimum water content and its dry side, large pores were compressed to make them smaller, while small pores remained unchanged. Comparatively, all the pores on the wet side were compressed to make them smaller. (c) For each compaction degree, as the remolding water content increased, a non-monotonic changing pattern was identified for both the permanent strain and resilient modulus; the permanent strain first decreased and then increased, while, for the resilient modulus, an initial increasing trend and then a decreasing trend were identified. In addition, a larger changing rate of the permanent strain (resilient modulus) was observed on the dry side, indicating a larger effect of the remolding water content. (d) For each remolding water content, as the compaction degree increased, the permanent strain exhibited a decreasing trend, but an increasing trend was identified for the resilient modulus. Moreover, the rate of change in the permanent strain (resilient modulus) on the dry side of the optimum water content was larger than that on the wet side. In contrast, the minimum rate of change was identified at the optimum water content. The obtained results allowed for the effects of the remolding water content and compaction degree on the dynamic behavior to be analyzed, and they helped guide the construction and maintenance of the subgrade.

Abstract This paper addresses the feature selection problem for gene expression profiles. Feature selection based on fuzzy neighborhood rough sets is very important in gene expression profiles. The process of extracting key features in gene expression profiles may have two disadvantages: (1) it may generate many redundant features, thus reducing the classification accuracy, and (2) it may ignore some information. To address the above problems, this paper proposes a fuzzy neighborhood joint entropy model based on feature selection; this model adopts the nonnegative principle of fuzzy neighborhood joint entropy to evaluate the importance of a candidate feature gene. Based on the fuzzy neighborhood joint entropy model, this paper proposes a new feature gene selection algorithm: the fuzzy neighborhood joint entropy (FNJE) algorithm. In the model, first, fuzzy neighborhood particles and fuzzy decision-making are combined with the uncertainty measure of joint entropy to construct a fuzzy neighborhood joint entropy model. Second, the importance degree of a feature is introduced as the measurement standard of candidate feature genes to evaluate the importance of each feature. Third, the algorithm uses the importance of candidate features in selecting features to reduce the redundancy of the selected features and improve the classification accuracy. In this paper, based on the UCI and gene datasets, we conduct a series of experiments. The experimental results show that the proposed algorithm can select fewer feature genes and achieve higher classification accuracy. Compared with the other four algorithms, our proposed algorithm can improve the accuracy by 0.4 $$\%$$ % $$-$$ - 10.42 $$\%$$ % and 1.16 $$\%$$ % $$-$$ - 15.18 $$\%$$ % and can reach maximum accuracy values of 90.52 $$\%$$ % and 87.02 $$\%$$ % with the linear-SVM and KNN classifiers, respectively.

Tetraploid is a very important experimental material, whether in scientific research or breeding practice. However, tetraploid strains are relatively rare. It was reported that chromosomes allocation disorder during cell division may lead to high embryonic mortality and low fertility in tetraploid fish. In this paper, tetraploid zebrafish is prepared by ploidy operation. And successfully establishes a tetraploid zebrafish cell line in vitro. To tracing status of microtubules, the tetraploid zebrafish cells are transfected with two plasmids of pBLK-α-tubulin-mCherry and pBLK-γ-tubulin-EGFP by electroporation in vitro. Observation shows that these transfected cells can be traced the changes of spindle and centrosomes during cell division in tetraploid cells. The results provide important material platform in vitro for studying the chromosome behavior in tetraploid cell proliferation, especially the formation mechanism of aneuploidy.

Gene editing technique has been widely applied for gene function characterization. However, such an approach is often time-consuming to obtain homozygous mutant. In this article, through timing the earlier injection of embryos, the gene editing efficiency is significantly improved, and together with the technique of the UV inactivated sperm and heat shock treatment for blocking the first cleavage, the gene-edited homozygous zebrafish are successfully obtained in F0 generation, rather than in the third or more generations as reported in the existing results. It is concluded that genetic purification of artificial gynogenesis can play critical role in more efficiently preparing the gene-edited homozygous individuals in fish.

The π-calculus is a basic theory of mobile communication based on the notion of interaction, which, is aimed at analyzing and modeling the behaviors of communication processes in communicating and mobile systems, and is widely applied to the security analysis of cryptographic protocol’s design and implementation. But the π-calculus does not provide seamless logical security analysis, so the logical flaws in the design and the implementation of a cryptographic protocol cannot be discovered in time. This paper introduces logical rules and logical proofs, binary tree, and the KMP algorithm and proposes a new extension of the π-calculus theory, a logical security analysis method, and an algorithm. The aim is to analyze whether there are logical flaws in the design and the implementation of a cryptographic protocol, to ensure the security of the cryptographic protocol when it is encoded into software and implemented. This paper presents the logical security proof and analysis of the TLS1.3 protocol’s interactional implementation process. Empirical results show that the additional extension theory, the logical security analysis method, and the algorithm can effectively analyze whether there are logical flaws in the design and the implementation of a cryptographic protocol.

Background. Blood safety levels have been significantly improved since the implementation of nucleic acid amplification technology (NAT) testing for blood donors. However, there remains a residual risk of transfusion transmission infections. This study aimed to evaluate the prevalence of HIV and its residual risk transmission among volunteer blood donors of Zhejiang Province, China, for five years after NAT implementation. Materials and Methods. All specimens and information were collected from voluntary unpaid donors at all blood services in Zhejiang Province, China, from January 2018 to December 2022. The HIV antibody or antigen and HIV RNA were detected using enzyme-linked immunosorbent assay and NAT, respectively. The HIV residual risk transmission was calculated using the incidence or window period model. Results. A total of 3,375,678 voluntary blood donors were detected, revealing an HIV prevalence of 9.92/100000. The HIV prevalence of blood donors in 12 blood services in Zhejiang Province was 6.11, 6.98, 7.45, 8.21, 8.36, 8.94, 9.04, 9.66, 9.73, 10.22, 11.80, and 12.47 per 100000 donors, without statistically significant difference observed among the services (p>0.05). Background: The integrity of the radial head is critical to maintaining elbow joint stability. For radial head fractures requiring surgical treatment, headless compression cannulated screw fixation is a less invasive scheme that has fewer complications. The aim of this study was to compare the mechanical stability of different fixation devices, including headless compression cannulated screws and mini-T-plates, for the fixation of transversely unstable radial head fractures.Methods: Forty identical synthetic radius bones were used to construct transverse unstable radial head fracture models. Parallel, cross, and tripod headless compression cannulated screw fixation and mini-T plate fixation were applied. The structural stiffness of each group was compared by static shear loading. Afterward, cyclic loading was performed in each of the three directions of the radial head, and the shear stability of each group was compared by calculating the maximum radial head displacement at the end of the cycle.Findings: The mini-T plate group had the lowest structural stiffness (51.8 ± 7.7 N/mm) and the highest relative displacement of the radial head after cyclic loading (p < 0.05). The tripod headless compression cannulated screw group had the highest structural stiffness among all screw groups (p < 0.05). However, there was no significant difference in the relative displacement of the radial head between the screw groups after cyclic loading in different directions (p > 0.05).Interpretation: In conclusion, the biomechanical stability of the mini-T plate for fixation of transverse unstable radial head fractures is lower than that of headless compression cannulated screws. Tripod fixation provides more stable fixation than parallel and cross fixation with headless compression cannulated screws for the treatment of transversely unstable radial head fractures.

Superhydrophobic fabrics suggest tremendous potential in some emerging fields such as electronic skin, bioelectronic sensors and special clothing due to their excellent liquid repellency and self-cleaning properties. However, currently reported superhydrophobic materials are usually prepared using biologically toxic fluorinated compounds, which severely restricts their practical applications. Herein, we propose a two-step thiol-ene click reaction strategy to fabricate fluorine-free superhydrophobic cotton fibers (Fabric-SH-PB-SiO2). The 1,2-polybutadiene (1,2-PB) with flexible rubber molecular chains and low surface energy was coated by cotton fiber through thiol-ene click reaction. Subsequently, based on this reaction, the coated cotton fiber was used to establish chemical bonds with hydrophobic silica. The Fabric-SH-PB-SiO2 possesses a rich multi-level micro-nano structure that can form stable air layer on the surface of the fabric, which showed a high hydrophobic angle of 156° and exhibited some distinct advantages such as anti-fouling and self-cleaning characteristics, high buoyancy, and reduced drag. More remarkably, the Fabric-SH-PB-SiO2 could float on the water surface after carrying 34 times its weight. Compared with unmodified fabrics, the Fabric-SH-PB-SiO2 demonstrated a high drag reduction rate of up to 102 %, among the highest values for the reported hydrophobic materials. The strategy developed here provides insights towards fabricating high-buoyancy fluorine-free superhydrophobic cotton fibers.

Triploid crucian carp is a new breed of aquaculture with rapid growth characteristics. However, its growth regulation mechanism has not yet been elucidated. In this study, our results show that the expression levels of mTOR signaling pathway-related genes (AKT1, AKT2, AKT3, mTOR, 4E-BP1 and S6K1) are higher in the muscle and intestine tissues of triploid crucian carp than those of diploid red crucian carp. Embryos of triploid crucian carp exhibit a faster growth rate comparing to red crucian carp. And in muscle effect stage, the mRNA levels of mTOR signaling pathway related genes in triploid crucian carp are higher than red crucian carp, except for AKT2. Inhibition of mTOR activity with rapamycin leads to slower embryonic development and the eliminated expression of mTOR in the embryos of neurula, muscle effect, and eye pigmentation stages. Conversely, overexpression of the upstream gene AKT2 can promote the development of zebrafish embryos and affect the expression of mTOR signaling pathway. These data highlight the significant role of mTOR signaling pathway in regulating fish embryonic development and rapid growth, and might offer new insights and avenues for exploring fish embryonic development, improving fish growth.

The weathering index of welded details, including butt-welded, non-load-carrying T-shaped and cruciform welded joints, fabricated with Q370qNH weathering steel (WS) was evaluated, and fatigue tests were conducted on the three types of welded details with a stress ratio of − 1 to determine the fatigue modes and fatigue properties. The fatigue S-N curves of three typical welded joints are fitted using the fatigue data and compared with the existing fatigue specifications, such as Eurocode 3, IIW 2007 and China Standard for Design of Steel Structures GB50017–2017. A total of 215 fatigue data are collected form the existing literatures, the difference of fatigue performance between the WS welded joints and that of traditional structural steels was analyzed. Results show that the slope parameter m of S-N curve of Q370qNH butt-welded joint is 5.50, which is quite different from the recommended value of 3. The fitting S-N curves of T-shaped welded joints and cruciform welded joints are in relatively well correlation to the S-N design curves. The fatigue performance of non-load-carrying T-shaped welded joints is significantly better than that of butt-welded joints and load-bearing cruciform welded joints. FRI analysis based on the collected fatigue data of 13 types of metallic materials in this study indicates that the actual fatigue strength of Q370qNH welding details demonstrates a certain fatigue safety margin, especially for the non-load-carrying T-shaped welded joints.

Urbanization causes immense eutrophication pressure on small urban wetlands, which play a vital role in urban ecosystems. However, our comprehension of the prolonged impacts of eutrophication on these types of wetlands continues to be significantly limited. To address this knowledge gap, we analyzed monthly physical–chemical variables and seasonal phytoplankton and zooplankton communities at the Taipa-Coloane wetland in Macau, China, from 2013 to 2019. Through the generalized additive model (GAM) and principal component analysis (PCA), we found that the aquatic system mainly exhibited interannual variations instead of seasonal variations, with nutrient loads and river discharge (turbidity and salinity) being the most prominent features affected by anthropogenic activities and climate change. Over the years, the increasing turbidity and phytoplankton showed that bottom-up effects predominated the aquatic systems. Under the bottom-up effects of eutrophication and turbidity, the phytoplankton communities showed high adaptation that MP and S1 occupied more dominance after 2016, with higher abundance but lower diversity. Moreover, the bottom-up effects further affected the structure of zooplankton communities. The dominance of rotifers filter feeders (RF) decreased over the years while middle copepods and cladocerans filter feeders (MCF) increased, implying a trend of zooplankton toward large size over time. Overall, our result reveals the bottom-up control pattern in the aquatic systems of small urban wetlands. This research provides valuable insights into the dynamics and responses of plankton communities to environmental changes in small urban wetlands, providing valuable information for future conservation efforts.

Abstract Background Necroptosis is a novel type of programmed cell death distinct from apoptosis. However, the role of necroptosis in ovarian cancer (OC) remains unclear. The present study investigated the prognostic value of necroptosis-related genes (NRGs) and the immune landscape in OC. Methods The gene expression profiling and clinical information were downloaded from the TCGA and GTEx databases. Differentially expressed NRGs (DE-NRGs) between OC and normal tissueswere identified. The regression analyses were conducted to screen the prognostic NRGs and construct the predictive risk model. Patients were then divided into high- and low-risk groups, and the GO and KEGG analyses were performed to explore bioinformatics functions between the two groups. Subsequently, the risk level and immune status correlations were assessed through the ESTIMATE and CIBERSORT algorithms. The tumor mutation burden (TMB) and the drug sensitivity were also analyzed based on the two-NRG signature in OC. Results Totally 42 DE-NRGs were identified in OC. The regression analyses screened out two NRGs (MAPK10 and STAT4) with prognostic values for overall survival. The ROC curve showed a better predictive ability in five-year OS using the risk score. Immune-related functions were significantly enriched in the high- and low-risk group. Macrophages M1, T cells CD4 memory activated, T cells CD8, and T cells regulatory infiltration immune cells were associated with the low-risk score. The lower tumor microenvironment score was demonstrated in the high-risk group. Patients with lower TMB in the low-risk group showed a better prognosis, and a lower TIDE score suggested a better immune checkpoint inhibitor response in the high-risk group. Besides, cisplatin and paclitaxel were found to be more sensitive in the low-risk group. Conclusions MAPK10 and STAT4 can be important prognosis factors in OC, and the two-gene signature performs well in predicting survival outcomes. Our study provided novel ways of OC prognosis estimation and potential treatment strategy.

We conducted a systematic investigation into the spectral and pulse characteristics of C and L-band Nonlinear Polarization Rotation (NPR) mode-locked fiber lasers effectively employing nonlinear polarization rotation technology. In our experimental setup, we achieved a stable mode-locked state at 1560.076 nm, exhibiting a 3 dB spectral bandwidth of 9.1 nm. As the pump power increased, we observed spectral shifts accompanied by shifts in the first Kelly sideband and the generation of new Kelly sidebands. In this paper, the phenomenon of spectral deviation is elucidated through the interplay of self-phase modulation, group velocity drift, and polarization-dependent isolator (PD-ISO) filter effect, with an analysis of the formation and deviation of Kelly sidebands. Notably, spectral shift persisted even when the pump power exceeded 200 mW. However, continuous pump power escalation led to soliton splitting, resulting in the formation of new soliton beams. Based on the simultaneous generation of spectral shift and pulse splitting, our study contributes to an enhanced understanding of soliton dynamics in ultrafast fiber lasers and lays a foundation for the application of high-repetition-frequency harmonic mode-locked lasers with tunable wavelengths.

The study of functional traits and their relationship to trade-offs has provided valuable insights into how plants adapt to environmental changes. Nonetheless, further research is necessary to fully comprehend the subtropical montane trade-off patterns in moss functional traits and the impact of environmental gradients on the correlation of these traits. To address this gap, we conducted a study of 11 moss species (7 families, 9 genera) in 54 patches from two subtropical mountain ranges, examining 40 functional traits related to photosynthesis, nutrients, water retention, and architecture. Through principal component analysis (PCA) and bi-variate correlation analysis, our findings reveal a strong correlation between light capture and nutrient assimilation strategies, as evidenced by the coordination between the traits of light capture and nutrient per area along a main principal component. Interestingly, we observed a trend towards smaller leaves and leaf cells in species with higher capacity for photosynthesis and metabolism, consistent with global trait spectra in vascular plants. However, we found that the trade-off between light capture and nutrient assimilation strategies was independent of water-holding capacity at shoot levels. Instead, we found that water-holding capacity was closely associated with nutrient utilization, energy metabolism, chlorophyll synthesis, and the primary process of photosynthesis. Our results highlight the multiple dimensions of functional traits in subtropical montane mosses and demonstrate that variation in these traits is driven by water availability, slope, and canopy density. Overall, our study provides valuable insights into the co-variation of moss traits and how environmental changes may impact mosses and ecosystem function.

Background: Bladder cancer is a common urological cancer associated high significant morbidity and mortality rates. Immunotherapy has emerged as a promising treatment option, although response rates vary among patients. Glycosylation has been implicated in tumorigenesis and immune regulation. However, our current comprehensive understanding of the role of glycosylation in bladder cancer and its clinical implications is limited.Methods: We constructed a training cohort based on the downloaded TCGA-BLCA dataset, while additional datasets (Xiangya cohort, GSE32894, GSE48075, GSE31684, GSE69795 and E-MTAB-1803) from Xiangya hospital, GEO and ArrayExpress database were obtained and used as validation cohorts. To identify glycosylation-related genes associated with prognosis, univariate Cox regression and LASSO regression were performed. A Cox proportional hazards regression model was then constructed to develop a risk score model. The performance of the risk score was assessed in the training cohort using Kaplan-Meier survival curves and ROC curves, and further validated in multiple validation cohorts.Results: We classified patients in the training cohort into two groups based on glycosylation-related gene expression patterns: Cluster 1 and Cluster 2. Prognostic analysis revealed that Cluster 2 had poorer survival outcomes. Cluster 2 also showed higher levels of immune cell presence in the tumor microenvironment and increased activation in key steps of the cancer immune response cycle. We developed an independent prognostic risk score (p < 0.001) and used it to construct an accurate prognostic prediction nomogram. The high glycosylation risk score group exhibited higher tumor immune cell infiltration, enrichment scores in immune therapy-related pathways, and a tendency towards a basal subtype. Conversely, the low-risk score group had minimal immune cell infiltration and tended to have a luminal subtype. These findings were consistent in our real-world Xiangya cohort.Conclusion: This multi-omics glycosylation score based on these genes reliably confirmed the heterogeneity of bladder cancer tumors, predicted the efficacy of immunotherapy and molecular subtypes, optimizing individual treatment decisions.

Background: Colorectal cancer (CRC) is a prevalent malignancy affecting the digestive tract, and its incidence has been steadily rising over the years. Surgery remains the primary treatment modality for advanced colorectal cancer, complemented by chemotherapy. The development of drug resistance to chemotherapy is a significant contributor to treatment failure in colorectal cancer. Nanodrug delivery systems (NDDS) can significantly improve the delivery and efficacy of antitumor drugs in multiple ways. However, there is a lack of visualization of NDDS research structures and research hotspots in the field of colorectal cancer, and the elaboration of potential research areas remains to be discovered.Objective: To comprehensively explore the current research status and development trend of NDDS in CRC research.Methods: Bibliometric analysis of articles and reviews on NDDS for CRC published between 2002 and 2022 using tools including CiteSpace, VOSviewer, R-bibliometrix, and Microsoft Excel was performed.Results: A total of 1866 publications authored by 9,870 individuals affiliated with 6,126 institutions across 293 countries/regions were included in the analysis. These publications appeared in 456 journals. Abnous Khalil has the highest number of publications in this field. The most published journals are the International Journal of Nanomedicine, International Journal of Pharmaceutics, and Biomaterials. Notably, the Journal of Controlled Release has the highest citation count and the third-highest H-index. Thematic analysis identified “inflammatory bowel disease”,“ “oral drug delivery," and “ulcerative colitis” as areas requiring further development. Keyword analysis revealed that “ulcerative colitis,” “exosomes,” and “as1411”have emerged as keywords within the last 2 years. These emerging keywords may become the focal points of future research.Conclusion: Our findings reveal the current research landscape and intellectual structure of NDDS in CRC research which helps researchers understand the research trends and hot spots in this field.

Abstract Although immune checkpoint blockade (ICB) therapies have been approved for bladder cancer (BLCA), only a minority of patients respond to these therapies, and there is an urgent need to explore combined therapies. Systematic multi‐omics analysis identified S100A5 as a novel immunosuppressive target for BLCA. The expression of S100A5 in malignant cells inhibited CD8+ T cell recruitment by decreasing pro‐inflammatory chemokine secretion. Furthermore, S100A5 attenuated effector T cell killing of cancer cells by inhibiting CD8+ T cell proliferation and cytotoxicity. In addition, S100A5 acted as an oncogene, thereby promoting tumor proliferation and invasion. Targeting S100A5 synergized with the efficacy of anti‐PD‐1 treatment by enhancing infiltration and cytotoxicity of CD8+ T cells in vivo. Clinically, there was a spatially exclusive relationship between S100A5+ tumor cells and CD8+ T cells in tissue microarrays. Moreover, S100A5 negatively correlated with immunotherapy efficacy in our real‐world and several public immunotherapy cohorts. In summary, S100A5 shapes a non‐inflamed tumor microenvironment in BLCA by inhibiting the secretion of pro‐inflammatory chemokines and the recruitment and cytotoxicity of CD8+ T cells. Targeting S100A5 converts cold tumors into hot tumors, thus enhancing the efficacy of ICB therapy in BLCA.

It is still a challenge to prepare flexible electromagnetic interference (EMI) shielding materials based on Ti3C2Tx MXene through a facial and scalable method. In this study, epoxied octamethyl-cyclotetrasiloxane (EPOSS) was self-assembled with Ti3C2Tx to construct EMI shielding composites via facial spraying coating. Because the EPOSS decreased the surface free energy of Ti3C2Tx in the acid-base component, the Ti3C2Tx/EPOSS suspension was able to be sprayed on carboxylated polybutadiene (CPB) uniformly. Molecular dynamics (MD) simulations were explored to demonstrate the interaction energy between EPOSS and CPB with different –COOH contents. The prepared EMI shielding composite has high abrasion resistance and good bending durability because EPOSS reinforced the interface interaction between Ti3C2Tx and CPB through covalent bonds. Based on a pre-stretching spray method, the prepared composite has high surface roughness, leading to a high specific EMI shielding effectiveness of 1800 dB cm2 g−1 at a low Ti3C2Tx loading of 8.5 wt%. Additionally, the physically protective effect of Si–O–Si cages in EPOSS provided good flame retardancy. Therefore, the proposed EPOSS-assisted Ti3C2Tx spraying strategy could prepare EMI shielding materials with high effectiveness, excellent abrasion resistance, and good flame retardancy, which has a broad prospect for the application of high-power, portable, and wearable flexible electronic devices.

Feature extraction and matching are irreplaceable parts of a typical visual simultaneous localization and mapping (VSLAM) algorithm. A variety of different approaches (e.g., ORB, Superpoint, GCNv2, etc.) have been proposed for effective feature extraction and matching. However, as far as we know, such methods still face great challenges in extreme angle and texture sparse scenarios. In this paper, an Improved Visual SLAM Algorithm Based on Graph Neural Network (GNNI-VSLAM), inspired by the strong robustness of the graph neural network in the field of image matching, is proposed to solve the problem of feature point extraction difficulties in sparsely textures scenes and feature point matching difficulties at extreme angles. First, the a priori location estimation feature extraction network is proposed to obtain fast and uniform detection and description of image feature points by a priori location estimation and to build accurate and real feature point information. Second, the feature matching network of the graph attention mechanism is proposed to aggregate feature point information through the neural network of the message passing graph, and then use the self and joint attention mechanism for adjacent image frame weighted feature matching. Then, the feature extraction and neural network are merged with the back-end nonlinear optimization, closed-loop correction and local mapping modules of the ORB-SLAM2 system to propose a complete monocular vision GNNI-VSLAM system. Finally, the proposed algorithm is verified by the public TUM dataset and the experimental results show that the absolute trajectory error of the proposed algorithm is reduced by 29.7 % compared to the GCNv2-SLAM algorithm, which shows a good mapping capabilities.

Abstract Background and aims This study investigated the natural history of esophageal squamous cell carcinoma (ESCC) in rural Chinese. We sought to help provide more data to support ESCC screenings. Methods This study was based on an existing Screening Program in Feicheng, China. Esophageal precancerous lesions were identified in 1753 cases, diagnosed from esophageal cancer screenings from 2006 to 2016. We followed up with them through endoscopic screening until October 1, 2017. Pathology results from various grades of precancerous lesions were recorded and the annual transition probabilities and incidence density of ESCC were calculated. Results As of October 1, 2017, a total of 4055.8 person‐years has been observed. The ESCC incidence density of mild, moderate, and severe dysplasia (SD) was 0.17, 0.79, and 1.77 per 100 person‐years, respectively. The median follow‐up time of mild, moderate, and SD was 3.5, 2.3, and 2.2 years, respectively. The annual transition probability of mild, moderate, and SD to the next pathological level was 0.025, 0.038, and 0.016, respectively. The ESCC incidence density of males was 2.6 times higher than females (0.58 vs. 0.22), and the older age group (56–69 age group) had a ESCC incidence density 1.2 times higher than the younger group (40–55 age group) (0.45 vs. 0.39). Conclusions The higher the grade of precancerous lesions, the higher the incidence density of ESCC. Screening of esophageal cancer in males and the elderly should be strengthened. It is recommended to reinforce follow‐up management for untreated patients with SD/carcinoma in situ. For patients with mild and moderate dysplasia in high‐risk rural Chinese populations, endoscopic follow‐up intervals can be appropriately adjusted to once every 2 years.

Oct4 was one of the important markers of cellular pluripotency. In this study, by Tol2 transgenic technology, the recombinant plasmids composed of Oct4 promoter DNA and enhanced green fluorescent protein (EGFP) gene are microinjected into zebrafish 1-cell stage embryos, and obtain the EGFP positive (EGFP+) transgenic fish. To generation homozygous transgenic fish, the eggs of EGFP+ F0 were activated by ultraviolet inactivated sperm of crucian carp, and then inhibited their first cleavage. The homozygous Oct4-EGFP+ zebrafish are successfully obtained in F1 generation. After chemical reprogramming, the clones derived from caudal fin fibroblasts of homozygous Oct4-EGFP+ zebrafish are observed green fluorescence. These results suggest that gynogenetic technology provides new strategies for the rapid preparation of homozygous transgenic fish.

Rheumatoid arthritis (RA) is an autoimmune disease that currently has an unknown cause and pathogenesis, and is associated with many complications and a high disability rate. The neutrophil extracellular trap network (NETs) is a newly discovered mechanism that allows neutrophils to capture and kill pathogens. Multiple studies in recent years have highlighted its relevance to the progression of rheumatoid arthritis. Despite the growing number of studies indicating the crucial role of NETs in RA, there has been no bibliometric review of research hotspots and trends in this area. In this study, we retrieved articles related to NETs in RA from the Web of Science Core Collection (WoSCC) database from 1985 to 2023 and used visualization tools such as Citespace, VOSviewer, Tableau Public, and Microsoft Office Excel 2021 to analyze the data. After screening, we included a total of 416 publications involving 2,334 researchers from 1,357 institutions in 167 countries/regions, with relevant articles published in 219 journals. The U.S., China, and Germany are the top 3 countries/regions with 124, 57, and 37 publications respectively. Mariana J. Kaplan is the most published author, and journals such as Frontiers in Immunology and International Journal of Molecular Sciences have had a significant impact on research in this field. The clinical application of PAD enzymes and their inhibitors, and the drug development of NETs as therapeutic targets for RA is a trend for future research. Our study provides a comprehensive bibliometric analysis and summary of NETs in RA publications, which will aid researchers in conducting further scientific research.

Myopia has significantly risen in East and Southeast Asia, and the pathological outcomes of this condition, such as myopic maculopathy and optic neuropathy linked to high myopia, have emerged as leading causes of irreversible vision loss. Addressing this issue requires strategies to reduce myopia prevalence and prevent progression to high myopia. Encouraging outdoor activities for schoolchildren and reducing near-work and screen time can effectively prevent myopia development, offering a safe intervention that promotes healthier habits. Several clinical approaches can be employed to decelerate myopia progression, such as administering low-dose atropine eye drops (0.05%), utilizing orthokeratology lenses, implementing soft contact lenses equipped with myopia control features, and incorporating spectacle lenses with aspherical lenslets. When choosing an appropriate strategy, factors such as age, ethnicity, and the rate of myopia progression should be considered. However, some treatments may encounter obstacles such as adverse side effects, high costs, complex procedures, or limited effectiveness. Presently, low-dose atropine (0.05%), soft contact lenses with myopia control features, and orthokeratology lenses appear as promising options for managing myopia. The measures mentioned above are not necessarily mutually exclusive, and researchers are increasingly exploring their combined effects. By advocating for a personalized approach based on individual risk factors and the unique needs of each child, this review aims to contribute to the development of targeted and effective myopia prevention strategies, thereby minimizing the impact of myopia and its related complications among school-aged children in affected regions.

Background: Ovarian cancer (OC) is a common tumor of the female reproductive system, while Alzheimer's disease (AD) is a prevalent neurodegenerative disease that primarily affects cognitive function in the elderly. Monocytes are immune cells in the blood that can enter tissues and transform into macrophages, thus participating in immune and inflammatory responses. Overall, monocytes may play an important role in Alzheimer's disease and ovarian cancer. Methods: The CIBERSORT algorithm results indicate a potential crucial role of monocytes/macrophages in OC and AD. To identify monocyte marker genes, single-cell RNA-seq data of peripheral blood mononuclear cells (PBMCs) from OC and AD patients were analyzed. Enrichment analysis of various cell subpopulations was performed using the “irGSEA” R package. The estimation of cell cycle was conducted with the “tricycle” R package, and intercellular communication networks were analyzed using “CellChat”. For 134 monocyte-associated genes (MRGs), bulk RNA-seq data from two diseased tissues were obtained. Cox regression analysis was employed to develop risk models, categorizing patients into high-risk (HR) and low-risk (LR) groups. The model's accuracy was validated using an external GEO cohort. The different risk groups were evaluated in terms of immune cell infiltration, mutational status, signaling pathways, immune checkpoint expression, and immunotherapy. To identify characteristic MRGs in AD, two machine learning algorithms, namely random forest and support vector machine (SVM), were utilized. Results: Based on Cox regression analysis, a risk model consisting of seven genes was developed in OC, indicating a better prognosis for patients in the LR group. The LR group had a higher tumor mutation burden, immune cell infiltration abundance, and immune checkpoint expression. The results of the TIDE algorithm and the IMvigor210 cohort showed that the LR group was more likely to benefit from immunotherapy. Finally, ZFP36L1 and AP1S2 were identified as characteristic MRGs affecting OC and AD progression. Conclusion: The risk profile containing seven genes identified in this study may help further guide clinical management and targeted therapy for OC. ZFP36L1 and AP1S2 may serve as biomarkers and new therapeutic targets for patients with OC and AD.

Abstract To study and prevent the water hazards of deep coal mines roof in the Inner Mongolia–Shaanxi (IM–S) mining area, it is essential to correctly evaluate the water-richness distributions of water-bearing aquifers in roof. This paper puts forward a sediment control method for water-richness law and zoning in the roof aquifers of deep Jurassic coals. To determine the vertical distance of direct water-bearing aquifers, the height of fractured water-conducting zone was detected by an underground network parallel electrical method. The plane and lateral spatial distribution patterns of the water-bearing aquifers and the control of the water-richness distribution was analyzed with the sediment control method. An evaluation system that consisted of four indicators, i.e., sedimentary environmental impact index, interlayer ratio of sandstone and mudstone, sandstone thickness, geophysical water-richness anomaly index was constructed. Furthermore, an Analytic Hierarchy Process (AHP) was introduced to establish the comprehensive zoning map. Finally, through the example analysis of Muduchaideng coal mine, the zoning evaluation results of water-richness were verified by the mine inflow. The findings of this study provide scientific guidance for prevention and control of mine water hazards in the IM–S mining area.

Abstract Background Long non-coding RNAs (lncRNA) have been implicated in a hand of studies that supported an involvement and co-operation in Uterine Corpus Endometrial Carcinoma (UCEC). Enhancer RNAs (eRNA), a functional subtype of lncRNA, have a key role throughout the genome to guide protein production, thus potentially associated with diseases. Methods In this study, we mainly applied the Cancer Genome Atlas (TCGA) dataset to systematically discover crucial eRNAs involving UCEC. For the key eRNAs in UCEC, we employed RT-qPCR to compare eRNA expression levels in tumor tissues and paired normal adjacent tissues from UCEC patients for validation. Furthermore, the relationships between the key eRNAs and immune activities were measured from several aspects, including the analysis for tumor microenvironment, immune infiltration cells, immune check point genes, tumor mutation burden, and microsatellite instability, as well as m6A related genes. Finally, the key eRNAs were verified by a comprehensive pan-cancer analysis. Results IGFBP7 Antisense RNA 1 (IGFBP7-AS1) was identified as the key eRNA for its expression patterns of low levels in tumor tissues and favorable prognostic value in UCEC correlated with its target gene IGFBP7. In RT-qPCR analysis, IGFBP7-AS1 and IGFBP7 had down-regulated expression in tumor tissues, which was consistent with previous analysis. Moreover, IGFBP7-AS1 was found closely related with immune response in relevant immune analyses. Besides, IGFBP7-AS1 and its target gene IGFBP7 correlated with a multi-omics pan-cancer analysis. Conclusions Finally, we suggested that IGFBP7-AS1 played a key role in impacting on clinical outcomes of UCEC patients for its possible influence on immune activity.
