Abstract Bacterial stem and root rot (BSRR) in sweetpotato caused by Dickeya dadantii is one of the ten major diseases of sweetpotatoes in China. However, the molecular mechanism underlying the resistance of sweetpotato to D. dadantii remains unclear. This study adopted a resistance identification assay that conformed Guangshu87 (GS87) as BSRR-resistant and Xinxiang (XX) as susceptible. Compared to XX, GS87 effectively prevented the invasion and dissemination of D. dadantii in planta. An RNA sequencing (RNA-seq) analysis identified 54,844 expressed unigenes between GS87 and XX at four different stages. Further, it revealed that GS87 was more able to regulate the expressions of more unigenes after the inoculation with D. dadantii, including resistance (R) and transcription factors (TF) genes. Moreover, content measurements of disease resistance-related phytohormones showed that both jasmonic acids (JAs) and salicylic acids (SAs) accumulated in D. dadantii-inoculated sweetpotatoes, and JAs may negatively regulate sweetpotato resistance against D. dadantii and accumulated faster than SAs. Meanwhile, determinations of ROS production rate and relevant enzymatic/non-enzymatic activity highlighted the vital roles of reactive oxygen species (ROS) and superoxide dismutase (SOD) in confering GS87 resistance against D. dadantii. Additionally, several hub genes with high connectivity were highlighted through Protein–Protein interaction (PPI) network analysis. In summary, the findings in this study contribute to the understanding of the different responses of resistant and susceptible sweetpotato cultivars to D. dadantii infection, and it also provide the first insight into the relevant candidate genes and phytohormones involved in the resistance of sweetpotato to D. dadantii.

Abstract Single-cell transcriptome sequencing (scRNA-seq) is a powerful tool for describing the transcriptome dynamics of plant development but has not yet been utilized to analyze the tissue ontology of sweetpotato. This study established a stable method for isolating single protoplast cells for scRNA-seq to reveal the cell heterogeneity of sweetpotato root tip meristems at the single-cell level. The study analyzed 12,172 single cells and 27,355 genes in the root tips of the sweetpotato variety Guangshu 87, which were distributed into 15 cell clusters. Pseudo-time analysis showed that there were transitional cells in the apical development trajectory of mature cell types from stem cell niches. Furthermore, we identified novel development regulators of sweetpotato tubers via trajectory analysis. The transcription factor IbGATA4 was highly expressed in the adventitious roots during the development of sweetpotato root tips, where it may regulate the development of sweetpotato root tips. In addition, significant differences were observed in the transcriptional profiles of cell types between sweetpotato, Arabidopsis thaliana, and maize. This study mapped the single-cell transcriptome of sweetpotato root tips, laying a foundation for studying the types, functions, differentiation, and development of sweetpotato root tip cells.

Abstract Objective To investigate the clinical efficacy of transumbilical single-incision laparoscopic surgery in the treatment of complicated appendicitis in overweight/obese adolescents. Methods A retrospective analysis was conducted on the clinical data of 226 adolescent patients with complicated appendicitis who were admitted to our hospital from January 2014 to June 2022. Among them, 102 cases underwent transumbilical single-incision laparoscopic appendectomy as the observation group, and another 124 cases underwent conventional three-port laparoscopic appendectomy as the control group. The surgical time, intraoperative blood loss, duration of incisional pain, postoperative flatus time, length of hospital stay, surgical site infection (SSI), satisfaction with cosmetic result, and occurrence of postoperative complications were compared between the two groups. Results Both groups completed the surgery smoothly, and there were no statistically significant differences in gender, age, BMI, duration of illness, white blood cell count, and preoperative CRP value between the two groups (P > 0.05). There were no statistically significant differences in surgical time and intraoperative blood loss between the two groups (P > 0.05). Abstract Background Failure to restore missing teeth in time can easily lead to the mesial tilting of the distal abutment teeth. However, a fixed partial denture (FPD) can improve stress conduction and distribution and prevent periodontal injuries. In these more complex cases, it is necessary to consider various factors comprehensively to improve conventional treatment planning and achieve better results. Methods We selected a patient with a missing first molar and a mesial inclination of the second molar, leaving inadequate space or bone mass for implant denture restoration, necessitating an FPD for restoration. Three-dimensional finite element analysis (3D-FEA) combined with photoelastic analysis were used to explore how the inclination angle (0 ‒ 30°) and different dental restoration materials (zirconia, lithium disilicate, polymer-infiltrated ceramic network, and resin composite) affect the biomechanical behaviour of FPD‒abutments‒periodontal tissue complex. Results The stress was easily concentrated in the FPD connectors, enamel shoulder collar, periapical area, and root bifurcation. The stress on FPD and the periodontal ligament (PDL) of the second premolar increased with an increase in the elastic modulus of FPD, with an opposite trend in the abutments, the alveolar bone, and the PDL of the second molar. The stress on the FPD and alveolar bone increased with increased inclination angle of the distal abutment. The stress on two abutments and their PDL were positively correlated with the inclination angle in two stages; however, when the inclination angle > 12°, the second premolar and its PDL showed a negative correlation. Conclusions FPDs can be used for restoration within 24° of distal abutment inclination, but protecting the abutments ( 12° especially) must be taken seriously. For this purpose, an FPD material with higher strength is recommended.

Abstract Dysfunction of pancreatic δ cells contributes to the etiology of diabetes. Despite their important role, human δ cells are scarce, limiting physiological studies and drug discovery targeting δ cells. To date, no directed δ-cell differentiation method has been established. Here, we demonstrate that fibroblast growth factor (FGF) 7 promotes pancreatic endoderm/progenitor differentiation, whereas FGF2 biases cells towards the pancreatic δ-cell lineage via FGF receptor 1. We develop a differentiation method to generate δ cells from human stem cells by combining FGF2 with FGF7, which synergistically directs pancreatic lineage differentiation and modulates the expression of transcription factors and SST activators during endoderm/endocrine precursor induction. These δ cells display mature RNA profiles and fine secretory granules, secrete somatostatin in response to various stimuli, and suppress insulin secretion from in vitro co-cultured β cells and mouse β cells upon transplantation. The generation of human pancreatic δ cells from stem cells in vitro would provide an unprecedented cell source for drug discovery and cell transplantation studies in diabetes.

Digital twins and the physical assets of electric power systems face the potential risk of data loss and monitoring failures owing to catastrophic events, causing surveillance and energy loss. This study aims to refine maintenance strategies for the monitoring of an electric power digital twin system post disasters. Initially, the research delineates the physical electric power system along with its digital counterpart and post-disaster restoration processes. Subsequently, it delves into communication and data processing mechanisms, specifically focusing on central data processing (CDP), communication routers (CRs), and phasor measurement units (PMUs), to re-establish an equipment recovery model based on these data transmission methodologies. Furthermore, it introduces a mathematical optimization model designed to enhance the digital twin system’s post-disaster monitoring efficacy by employing the branch-and-bound method for its resolution. The efficacy of the proposed model was corroborated by analyzing an IEEE-14 system. The findings suggest that the proposed branch- and-bound algorithm significantly augments the observational capabilities of a power system with limited resources, thereby bolstering its stability and emergency response mechanisms.

In this protocol, we detail a seven-stage differentiation methodology for generating pancreatic delta cells (SC-delta cells) from human pluripotent stem cells (hPSCs). In the first step, definitive endoderm is generated by activin A and CHIR99021, followed by induction of primitive gut tube and posterior foregut by treatment with FGF7, SANT1, LDN193189, PdBU, and retinoic acid (RA). The subsequent endocrine generation and directed SC-delta cell induction is achieved by a combined treatment of the FGF7 with FGF2 during stage 4 and 5, together with RA, XXI, ALK5 inhibitor II, SANT1, Betacellulin and LDN193189. The planar cultivation is converted to a suspended system after stage 5, allowing cells to aggregate into delta cell-containing spheroids. The differentiation takes approximately 4-5 weeks for delta cell generation and an additional 1-2 weeks for cell expansion and evaluation. We believe that this amenable and simplified protocol can provide a stable source of SC-delta cells from efficient differentiation, facilitating further investigation of the physiological role of delta cells as well as refinement of islet cell therapeutic strategies.

Abstract The influence of entrainment, a key process characterized by the entrainment rate in cumulus parameterization, on aerosol‐cloud interactions has been widely recognized. However, despite qualitative links established between entrainment and aerosol loading, a quantitative relationship based on observational evidence remains elusive. This study utilizes aircraft observations of cumulus clouds during two field campaigns to determine the quantitative relationship between entrainment rate and aerosol loading. In both campaigns, the entrainment rate is negatively correlated with aerosol loading. It is speculated that increased aerosol loading enhances cloud edge droplet evaporation, which leads to increased buoyancy and vertical velocity within the cloud, thereby reducing the entrainment rate. Further analysis shows that the response of entrainment rate to aerosol perturbations is more significant in smaller cumulus clouds with weak buoyancy and less pronounced under opposite conditions. These findings shed new light on improving the description of aerosol‐cloud interactions in cumulus parameterizations.

Abstract Background Inflammation and obesity are the risk factors for hyperlipidaemia. Nonetheless, research regarding the association between dietary live microbes intake and hyperlipidaemia is lacking. Therefore, this study focused on revealing the relationship between them and mediating roles of inflammation and obesity. Methods Totally 16,677 subjects were enrolled from the National Health and Nutrition Examination Survey (NHANES) (1999–2010 and 2015–2020). To explore the correlation between live microbes and hyperlipidaemia as well as blood lipid levels, respectively, multiple logistic regression and linear regression were employed. Furthermore, the mediating roles of body mass index (BMI), C-reactive protein (Crp) and their chain effect were explored through mediating analysis. Results High dietary live microbes intake was the protective factor for hyperlipidaemia. Abstract Background Postoperative performance, including best corrected distance visual acuity (BCDVA) and optical metrics (from the OQAS and iTrace devices), was compared among 4 different intraocular lenses (IOLs). Methods This prospective observational study included 104 eyes from 104 subjects who underwent cataract surgery combined with implantation of 4 different IOLs: monofocal (Mon) IOLs, segmental refractive (SegRef) IOLs, diffractive (Dif) IOLs and extended depth of focus (EDoF) IOLs. Postoperative BCDVA and optical metrics were collected at the 6th month. The OQAS optical metrics included the objective scattering index (OSI), Strehl ratio (SR), modulation transfer function (MTF) cut-off frequency, and predicted visual acuity (PVA); the iTrace optical metrics included blur/double vision, glare/halo, starburst, mixed focus, night myopia, and night hyperopia. Abstract The angiotensin-converting enzyme 2 (ACE2) is a primary cell surface viral binding receptor for SARS-CoV-2, so finding new regulatory molecules to modulate ACE2 expression levels is a promising strategy against COVID-19. In the current study, we utilized islet organoids derived from human embryonic stem cells (hESCs), animal models and COVID-19 patients to discover that fibroblast growth factor 7 (FGF7) enhances ACE2 expression within the islets, facilitating SARS-CoV-2 infection and resulting in impaired insulin secretion. Using hESC-derived islet organoids, we demonstrated that FGF7 interacts with FGF receptor 2 (FGFR2) and FGFR1 to upregulate ACE2 expression predominantly in β cells. This upregulation increases both insulin secretion and susceptibility of β cells to SARS-CoV-2 infection. Inhibiting FGFR counteracts the FGF7-induced ACE2 upregulation, subsequently reducing viral infection and replication in the islets. Furthermore, retrospective clinical data revealed that diabetic patients with severe COVID-19 symptoms exhibited elevated serum FGF7 levels compared to those with mild symptoms. Finally, animal experiments indicated that SARS-CoV-2 infection increased pancreatic FGF7 levels, resulting in a reduction of insulin concentrations in situ. Taken together, our research offers a potential regulatory strategy for ACE2 by controlling FGF7, thereby protecting islets from SARS-CoV-2 infection and preventing the progression of diabetes in the context of COVID-19.

Abstract Background The effectiveness of anti-programmed cell death protein 1(PD-1)/programmed cell death 1 ligand 1(PD-L1) therapy in treating certain types of cancer is associated with the level of PD-L1. However, this relationship has not been observed in colorectal cancer (CRC), and the underlying regulatory mechanism of PD-L1 in CRC remains unclear. Methods Binding of TMEM160 to PD-L1 was determined by co-immunoprecipitation (Co-IP) and GST pull-down assay.The ubiquitination levels of PD-L1 were verified using the ubiquitination assay. Phenotypic experiments were conducted to assess the role of TMEM160 in CRC cells. Animal models were employed to investigate how TMEM160 contributes to tumor growth.The expression and clinical significance of TMEM160 and PD-L1 in CRC tissues were evaluated by immunohistochemistry(IHC). Results In our study, we made a discovery that TMEM160 interacts with PD-L1 and plays a role in stabilizing its expression within a CRC model. Furthermore, we demonstrated that TMEM160 hinders the ubiquitination-dependent degradation of PD-L1 by competing with SPOP for binding to PD-L1 in CRC cells. Regarding functionality, the absence of TMEM160 significantly inhibited the proliferation, invasion, metastasis, clonogenicity, and radioresistance of CRC cells, while simultaneously enhancing the cytotoxic effect of CD8 + T cells on tumor cells. Conversely, the upregulation of TMEM160 substantially increased these capabilities. In severely immunodeficient mice, tumor growth derived from lentiviral vector shTMEM160 cells was lower compared with that derived from shNC control cells. Furthermore, the downregulation of TMEM160 significantly restricted tumor growth in immune-competent BALB/c mice. In clinical samples from patients with CRC, we observed a strong positive correlation between TMEM160 expression and PD-L1 expression, as well as a negative correlation with CD8A expression. Importantly, patients with high TMEM160 expression exhibited a worse prognosis compared with those with low or no TMEM160 expression. Conclusions Our study reveals that TMEM160 inhibits the ubiquitination-dependent degradation of PD-L1 that is mediated by SPOP, thereby stabilizing PD-L1 expression to foster the malignant progress, radioresistance, and immune evasion of CRC cells. These findings suggest that TMEM160 holds potential as a target for the treatment of patients with CRC.

In this study, we introduce an integrated schedule of order picking and delivery for instant delivery. Order picking, including order batching and picking sequencing, is scheduled online under real-time order arrival, which integrates order delivery by depicting order location dispersion in an online order picking strategy. Order delivery, including delivery person assignment and route planning, is modeled to minimize the total duration of order fulfillment by considering the influence of the order picking completion time. A rule-based online order picking strategy is established, and a customized ant colony optimization (ACO) algorithm is proposed to optimize order delivery. Experiments on 16 simulated instances of different scales demonstrate that our online order picking schedule considering order delivery outperforms existing approaches and that the customized ACO algorithm for order delivery is effective.

BackgroundStroke is a devastating disease and ranks as the second leading cause of death and disability globally. Several studies have shown that preceding infection (PI) of upper respiratory tract are strongly associated with acute ischemic stroke (AIS). However, the clinical implications and underlying pathological mechanisms remain unclear.MethodsIn this study, 16S rRNA gene sequencing was employed to compare the structural characteristics of oropharyngeal and gut microbiota in AIS patients with or without PI and normal controls (NCs; 30 cases each), and systemic inflammatory markers were detected to explore the relationship between upper respiratory tract infections (URTIs) and subsequent stroke severity and functional outcome and the potential mechanism.ResultsWe found that patients with AIS-PI exhibited elevated serum WBC, NE, CRP, and Hcy levels, as well as a higher 90-day mRS score. Oropharyngeal and gut microbiota analysis showed that AIS and AIS-PI patients exhibited increased microbial richness in sequence. Principal coordinate analysis of the microbiota demonstrated significant differences in microbiota composition among the three groups. In AIS-PI patients, Megamonas, Megasphaera, Ruminococcaceae UCG 004, Rothia, and Streptococcus were significantly enriched in the gut. Opportunistic pathogens, including Thermus, uncultured Veillonella sp., and Oribacterium sinu, were found to be significantly enriched in the oropharynx. The dysregulated microbiota were positively correlated with systemic inflammatory markers, stroke severity, and poor prognosis. In contrast, short-chain fatty acid-producing bacteria Eisenbergiella, bacterium NLAE, Fusicatenibacter, Ruminococcaceae, and Faecalibacterium were enriched in NCs. Their abundances were negatively correlated with systemic inflammatory markers, stroke severity and poor prognosis.ConclusionOur findings suggest that PIs of the upper respiratory tract may contribute to poor short-term functional outcome in AIS patients by causing disturbance of the oropharyngeal and gut microbiota and promoting elevated systemic inflammation levels.

Forkhead box protein J1 (FOXJ1), a member of the forkhead family, is an important transcription factor regulating multiciliated cell differentiation and motile ciliogenic program. Here, we established a FOXJ1- EGFP knock-in human embryonic stem cell (hESC) line by inserting a P2A-EGFP gene cassette of FOXJ1 using CRISPR/Cas9 system. The reporter cell line retained a normal karyotype, expressed comparable pluripotent marker genes, and maintained differentiation potential. This reporter cell line enables live identification of multiciliated cells during the general lung differentiation and will be a valuable tool for studying the multiciliated cell differentiation, ciliogenesis and mechanism of related pulmonary diseases.

The modified coconut shell biochars (MCSBCs) were fabricated and their adsorptions for Pb(II) were evaluated, in which waste coconut shell was used as the raw material, both ZnCl2 and KMnO4 were applied as the inorganic modifiers. FT-IR spectra, TGA, SEM and BET techniques were utilized to characterize their properties. It was spotted that the thermal stability of UCSBC could arrive at 500 °C. The BET specific surface areas of both Zn- and Mn-modified MCSBCs (485.137, 476.734 m2/g) were highly decreased as compared with that of UCSBC (3528.78 m2/g). In contrast, the average pore diameters of both Zn- and Mn-modified MCSBCs (3.295, 3.803 nm) were smaller than that of UCSBC (3.814 nm). These findings reveal that the modification of CSBC didn't change its pore size. Their adsorptions for Pb(II) were performed and some controlling factors involving pH, contact time, starting concentration and temperature were explored. Moreover, the experiment data were fitted via linear and non-linear techniques. It was found that the Langmuir maximal adsorption amounts of un-modified coconut shell biochar (UCSBC), Zn-modified and Mn-modified MCSBCs for Pb(II) could reach 31.653, 86.547 and 93.666 mg/g, respectively. Two-parameter kinetic models exposed that Pb(II) adsorption on UCSBC, Zn-modified and Mn-modified MCSBCs obeyed both the Lagergren first-order (non-linear R2 = 0.990, 0.954, 0.953, respectively) and Avrami fractional-order (non-linear R2 = 0.989, 0.946, 0.945, respectively) kinetic models. Two-parameter and three-parameter isotherm models verified that Pb(II) adsorption on UCSBC, Zn-modified and Mn-modified MCSBCs followed the Langmuir (non-linear R2 = 0.992, 0.997, 0.993, respectively) as well as Sips (non-linear R2 = 0.992, 0.997, 0.992, respectively) isotherm models. The computation of thermodynamic parameters evidenced that the modification of UCSBC via KMnO4 and ZnCl2 can effectively rise its adsorption for Pb(II), exhibiting promising applications in the handling of metal-bearing water.

Recently, extended-spectrum β-lactamases (ESBL) producing E.coli has posed a threat to public health. Generally, captive healthy giant pandas are not exposed to antibiotics; however, they still acquire antimicrobial resistant bacteria. In order to understand whether there is an exchange of resistance genes within the ecosystems of captive giant pandas, this study explored resistance characteristics of 330 commensal E. coli isolates from feces of giant pandas, the surroundings, and breeders. Ferroptosis, a newly discovered form of regulated cell death, has garnered significant attention in the field of tumor therapy. However, the presence of overexpressed glutathione (GSH) and insufficient levels of H2O2 in the tumor microenvironment (TME) hinders the occurrence of ferroptosis. In response to these challenges, here we have constructed the self-assembled nanocomplexes (FeE NPs) utilizing epigallocatechin-3-gallate (EGCG) from green tea polyphenols and metal ions (Fe3+) as components. After grafting PEG, the nanocomplexes (FeE@PEG NPs) exhibit good biocompatibility and synergistically enhanced tumor-inhibitory properties. FeE@PEG NPs can be disassembled by H2O2 in the TME, leading to the rapid release of Fe3+ and EGCG. The released Fe3+ produces large amounts of toxic •OH by the Fenton reactions while having minimal impact on normal cells. The generated •OH effectively induces lipid peroxidation, which leads to ferroptosis in tumor cells. Meanwhile, the released EGCG can autoxidize to produce H2O2, which further promotes the production of •OH radicals and increases lipid peroxide levels. Moreover, EGCG also depletes the high levels of intracellular GSH, leading to an intracellular redox imbalance and triggering ferroptosis. This study provides new insights into advancing anticancer ferroptosis through rational material design, offering promising avenues for future research.

Sweet potato leaf tips have high nutritional value, and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities. In this study, a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties. Three varieties with distinct overall metabolic profiles (A01, A02, and A03), two varieties with distinct profiles of phenolic acids (A20 and A18), and three varieties with distinct profiles of flavonoids (A05, A12, and A16) were identified. In addition, a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips, respectively, were identified through morphological characterization. Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids. Also, the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips, with lobed leaf types having higher levels of polyphenols than the entire leaf types. The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value.

High-throughput screening is an indispensable technology in drug discovery, cancer therapy, and disease diagnosis, and it could greatly reduce time cost, reagent consumption, and labor expense. Here, four high-throughput screening methods with high sensitivity and accessibility are discussed in detail. Fluorescence, DNA, heavy metal, and nonmetal isotope barcodes, which generally label antibodies, proteins, and saccharides to identify cells, are detected by flow cytometry, second-generation DNA sequencing, mass cytometry, and second-ion mass spectrometry, respectively. Encoding binary information in barcodes, labeling individual cells by barcodes, performing the characterization of cells together, and identifying the result belonging to individual cells via barcodes are the main steps for high-throughput screening. Applications of the four digital barcodes in high-throughput screening for both in vitro and in vivo tests are described in detail, and their advantages and disadvantages are also summarized. High-throughput screening has provided a powerful platform widely accessible for multidisciplinary studies and has greatly sped up the progress of drug discovery, disease diagnosis, and cancer therapy.

Abstract Practical lithium-ion battery systems require parallelisation of tens to hundreds of cells, however understanding of how pack-level thermal gradients influence lifetime performance remains a research gap. Here we present an experimental study of surface cooled parallel-string battery packs (temperature range 20–45 °C), and identify two main operational modes; convergent degradation with homogeneous temperatures, and (the more detrimental) divergent degradation driven by thermal gradients. We attribute the divergent case to the, often overlooked, cathode impedance growth. This was negatively correlated with temperature and can cause positive feedback where the impedance of cells in parallel diverge over time; increasing heterogeneous current and state-of-charge distributions. These conclusions are supported by current distribution measurements, decoupled impedance measurements and degradation mode analysis. From this, mechanistic explanations are proposed, alongside a publicly available aging dataset, which highlights the critical role of capturing cathode degradation in parallel-connected batteries; a key insight for battery pack developers.

Interferometric synthetic aperture radar (InSAR) techniques can be used to derive spatially dense “relative” measurements of vertical land motion (VLM), whereas global navigation satellite system (GNSS) provides point-based “absolute” measurements of VLM. The combination of GNSS and InSAR observations can yield spatially dense VLM measurements in an absolute reference frame. In addition, GNSS observations can be used to correct atmospheric noise in InSAR deformation measurements and serve as a complementary measure to isolate deep and shallow subsidence components. Given the increasing spatial and temporal coverage available from InSAR satellites, there is a need to establish calibration/validation networks that enable the use of InSAR for measuring VLM in coherence-challenged areas such as many low-lying coastal lands. In this study, we provide a method for the selection of sites for new GNSS installations such that the resulting GNSS network can better serve as tie points and validation for InSAR in areas where low coherence prevents high-fidelity phase unwrapping. Our method is applied in a case study for expanding the existing GNSS network in southern Louisiana, using abandoned oil well sites as potential sites. Considering practical limitations, distribution among various land classes, and following National Geodetic Survey guidelines, our proposed GNSS network consists of 61 (45 existing + 16 new) stations spread over a 50 000 km2 area of southern Louisiana.

Background: Tripartite motif (TRIM) family proteins have more than 80 members and are widely found in various eukaryotic cells. Most TRIM family proteins participate in the ubiquitin-proteasome degradation system as E3-ubiquitin ligases; therefore, they play pivotal regulatory roles in the occurrence and development of tumors, including tumor immune escape. Due to the diversity of functional domains of TRIM family proteins, they can extensively participate in multiple signaling pathways of tumor immune escape through different substrates. In current research and clinical contexts, immune escape has become an urgent problem. The extensive participation of TRIM family proteins in curing tumors or preventing postoperative recurrence and metastasis makes them promising targets. Aim of Review: The aim of the review is to make up for the gap in the current research on TRIM family proteins and tumor immune escape and propose future development directions according to the current progress and problems. Key Scientific Concepts of Review: This up-to-date review summarizes the characteristics and biological functions of TRIM family proteins, discusses the mechanisms of TRIM family proteins involved in tumor immune escape, and highlights the specific mechanism from the level of structure-function-molecule-pathway-phenotype, including mechanisms at the level of protein domains and functions, at the level of molecules and signaling pathways, and at the level of cells and microenvironments. We also discuss the application potential of TRIM family proteins in tumor immunotherapy, such as possible treatment strategies for combination targeting TRIM family protein drugs and checkpoint inhibitors for improving cancer treatment.

Hazmat transportation in cities faces significant risks that may cause catastrophic losses to humans. From the perspective of the regulator, the main responsibility is to mitigate hazmat transport risk by determining the availability of road networks to hazmat carriers. Based on the time-variant population distribution, the hazmat transport risk was assessed via the total population exposure associated with the resident and variable populations at different times. We propose a risk-minimizing urban hazmat road network design model for multiple types of hazmats, considering time-varying traffic. The model was applied to a realistic case study of hazmat transportation in a densely populated urban area with complex traffic in Shanghai, China.

Insulin secretion by pancreatic islets plays a vital role in regulating blood glucose levels. Nevertheless, the mechanism responsible for this dynamic insulin secretion has not been completely understood, particularly at the single islet level. In this study, we have successfully developed an easy microfluidic platform that allows for the exploration of dynamic glucose-stimulated insulin secretion (GSIS) at the single islet level. With the utilization of this platform, we evaluated dynamic GSIS from single islets isolated from both normal and diabetic rats. Our results demonstrate that islets can be categorized into three types based on their dynamic GSIS: Type I exhibits a biphasic GSIS profile with a fast first phase and flat second phase; Type II also has a biphasic GSIS profile with a fast first phase but a slow increased second phase; Type III displays only a slowly increased second phase and lacks a fast first phase. RNA sequencing analysis demonstrated that the cell type and exocytosis-specific genes are consistent with the proportion of cells and insulin release kinetics among the three types of islets, respectively. Moreover, our findings suggest that high expression of Atp5pb is anti-correlated with the first phase of insulin secretion. Furthermore, we revealed that diabetic islets exhibit only the type I GSIS response, indicating a deliberate impairment of the second phase of insulin secretion. Together, this device serves as a crucial tool in the research field of islets and diabetes, allowing researchers to investigate islet functional heterogeneity and identity at the single islet level.

BackgroundVentilator-Associated Pneumonia (VAP) severely impacts stroke patients’ prognosis after endovascular treatment. Hence, this study created a nomogram to predict the occurrence of VAP after endovascular treatment.MethodsThe individuals with acute ischemic stroke and large vessel occlusion (AIS-LVO) who received mechanical ventilation and endovascular therapy between July 2020 and August 2023 were included in this retrospective study. The predictive model and nomogram were generated by performing feature selection optimization using the LASSO regression model and multifactor logistic regression analysis and assessed the evaluation, verification and clinical application.ResultsA total of 184 individuals (average age 61.85 ± 13.25 years, 73.37% male) were enrolled, and the rate of VAP occurrence was found to be 57.07%. Factors such as the Glasgow Coma Scale (GCS) score, duration of stay in the Intensive Care Unit (ICU), dysphagia, Fazekas scale 2 and admission diastolic blood pressure were found to be associated with the occurrence of VAP in the nomogram that demonstrating a strong discriminatory power with AUC of 0.862 (95% CI, 0.810–0.914), and a favorable clinical net benefit.ConclusionThis nomogram, comprising GCS score, ICU duration, dysphagia, Fazekas scale 2 and admission diastolic blood pressure, can aid clinicians in predicting the identification of high-risk patients for VAP following endovascular treatment in large vessel occlusion stroke.

Introduction Influenza is a major public health threat, and vaccination is the most effective prevention method. However, vaccination coverage remains suboptimal. Low health literacy regarding influenza vaccination may contribute to vaccine hesitancy. This study aims to evaluate the effect of health education interventions on influenza vaccination rates and health literacy.Methods and analysis This cluster randomised controlled trial will enrol 3036 students in grades 4–5 from 20 primary schools in Dongguan City, China. Schools will be randomised to an intervention group receiving influenza vaccination health education or a control group receiving routine health education. The primary outcome is the influenza vaccination rate. Secondary outcomes include health literacy levels, influenza diagnosis rate, influenza-like illness incidence and vaccine protection rate. Data will be collected through questionnaires, influenza surveillance and self-reports at baseline and study conclusion.Ethics and dissemination Ethical approval has been sought from the Ethics Committee of the School of Public Health, Sun Yat-sen University. Findings from the study will be made accessible to both peer-reviewed journals and key stakeholders.Trial registration number NCT06048406.

ObjectiveTo compare the efficacy of robot-assisted single-port-plus-one pyeloplasty (RSPY) and laparoscopic single-port pyeloplasty (LSPY) in the treatment of children with ureteropelvic junction obstruction (UPJO).MethodsThe clinical data of 47 children who underwent surgery for UPJO at the Department of the Pediatric Surgery of the Fujian Provincial Hospital from October 2020 to September 2022 were analyzed retrospectively. Of these 47 children, 27 received RSPY while 20 underwent LSPY. The baseline data, operation time, intraoperative anastomosis time, intraoperative blood loss, postoperative hospital stay, complications, total cost, preoperative and postoperative renal parenchymal thickness (PT), anteroposterior renal pelvis diameter (APD), and differential renal function (DRF) of the two groups were compared to evaluate the clinical efficacy of the two surgical methods.ResultsThe results showed that both surgical techniques were successful and no patient transitioned to open surgery. There was no significant difference between the two groups in baseline data, intraoperative blood loss, complications, APD, and PT 6 months after surgery. There was also no significant difference in APD, PT, and DRF 12 months after surgery (all P > 0.05). IntroductionThe incidence of biliary system diseases has been continuously increasing in the past decade. Biliary system diseases bring a heavy burden to humanity and society. However, the specific etiology and pathogenesis are still unknown. The biliary system, as a bridge between the liver and intestine, plays an indispensable role in maintaining the physiological metabolism of the body. Therefore, prevention and treatment of biliary diseases are crucial. It is worth noting that the microorganisms participate in the lipid metabolism of the bile duct, especially the largest proportion of intestinal bacteria.MethodsWe systematically reviewed the intestinal microbiota in patients with gallstones (GS), non-calculous biliary inflammatory, and biliary tract cancer (BTC). And searched Pubmed, Embase and Web of science for research studies published up to November 2023.ResultsWe found that the abundance of Faecalibacterium genus is decreased in GS, primary sclerosing cholangitis (PSC), primary biliary cholangitis (PBC) and BTC. Veillonella, Lactobacillus, Streptococcus and Enterococcus genus were significantly increased in PSC, PBC and BTC. Interestingly, we found that the relative abundance of Clostridium was generally reduced in GS, PBC and BTC. However, Clostridium was generally increased in PSC.DiscussionThe existing research mostly focuses on exploring the mechanisms of bacteria targeting a single disease. Lacking comparison of multiple diseases and changes in bacteria during the disease process. We hope to provide biomarkers forearly diagnosis of biliary system diseases and provide new directions for the mechanism of intestinal microbiota in biliary diseases.

Precursor-derived ceramic SiOC (PDC-SiOC) microlattices exhibit excellent oxidation resistance, high-temperature stability, and superior mechanical properties. However, the printing accuracy of the PDC-SiOC microlattices by 3D printing is still limited, and mechanical properties of the PDC-SiOC microlattices have not been studied systematically. Here, PDC-SiOC octet microlattices were fabricated by projection micro stereolithography (PμSL) 3D printing, and photoabsorber (Sudan III)’s effect on the accuracy was systematically analyzed. The results showed that the addition of Sudan III improved the printing accuracy significantly. Then, the ceramization process of the green body was analyzed in detail. The order of the green body decreased, and most of their chemical bonds were broken during pyrolysis. After that, the PDC-SiOC microlattices with different truss diameters in the range of 52–220 μm were fabricated, and their mechanical properties were investigated. The PDC-SiOC microlattices with a truss diameter of 52 μm exhibited higher compression strength (31 MPa) than those with bigger truss diameters. The size effect among the PDC-SiOC microlattices was analyzed. Our work provides a deeper insight into the manufacturing of PDC-SiOC micro-scaled architectures by 3D printing and paves a path to the research of the size effect in ceramic structures.

Ceramic microcomponents have a wide range of applications in microfluidic, sensors, etc. Stereolithography 3D printing technology provided a shortcut for the preparation of this small-size and complex component. With the development of device miniaturization, the existing stereolithography forms cannot meet the demand. To prepare smaller components, it is necessary to decrease the slice thickness (Cs). But during the stereolithography 3D printing, ceramic is usually easy to over-cure, leading to the key issue in the stereolithography of ceramic microcomponents: the mismatching between the curing thickness (Cp) and the slice thickness (Cs). To solve the problem above, this paper started from the stereolithography mechanism of the ceramic/resin slurry and adjusted the matching relationship between Cp and Cs theoretically by adding a photoabsorber. Then, from the experiments, the influence of different content of photoabsorber (carbon black) on curing thickness, printing accuracy, and sintering accuracy, were studied. Al2O3 ceramic microcomponents that met the design requirements were thus prepared. These results would help in the development of stereolithography 3D printing of ceramic microcomponents.

The impact of Ba-modified peanut shell biochar (Ba-PSB) on Pb(II) removal was studied and BaCl2 was used as a modifier. It was shown that the PSB obtained at 750 °C had the best adsorption effect, and the Ba-PSB had a larger specific surface area and a good adsorption effect on Pb(II). At pH = 5, concentration was 400 mg/L, time was 14 h, and temperature was 55 °C, the loading amount of black peanut shell biochar (BPSB), red peanut shell biochar (RPSB), Ba-BPSB, and Ba-RPSB reached 128.050, 98.217, 379.330, and 364.910 mg/g, respectively. In addition, based on the non-linear fitting, it was found that the quasi-second-order kinetic model, and isothermal model could be applied to describe Pb(II) adsorption on PSB and Ba-PSB. The adsorption behavior of PSB unmodified and modified was a spontaneous process. Moreover, chemical modification of BPSB, RPSB, Ba-BPSB, and Ba-RPSB for hindering of –COOH and –OH groups revealed 81.81, 77.08, 86.90, and 83.65% removal of Pb(II), respectively, which was due to the participation of –COOH, while 17.61, 21.70, 12.77, and 15.06% was from –OH group, respectively. The increase of cation strength (Na+, K+, Ca2+, and Mg2+) will reduce the adsorption capacity of PSB for Pb(II). HIGHLIGHTS Ba-modified PSB was prepared and its adsorption property was examined.; The modification of PSB could greatly improve its loading amount for Pb(II).; The effect of –COOH and –OH groups from PSB on Pb(II) sorption was scrutinized.; Ba-modified PSB was an excellent green adsorbent for Pb(II) removal from water.;

Abstract Background and purpose To explore the association of systemic inflammatory index (SIRI), systemic immune-inflammatory index (SII) and inflammatory prognosis index (IPI) with 90d outcomes in patients with acute ischemic stroke (AIS) after intravenous thrombolysis. Methods The patients who underwent intravenous thrombolysis were enrolled in the present study from September 2019 to December 2022. According to the relevant blood indexes obtained in 24 h after admission, the corresponding values of SIRI, SII and IPI were calculated. The correlation among SIRI, SII, IPI, and admission NIHSS scores was examined by Spearman correlation analysis. ROC curve analysis was conducted to determine the optimal cut-off value of SIRI, SII, IPI, and their corresponding sensitivity and specificity to evaluate their predictive value on admission for poor prognosis. Abstract Autophagy is a lysosome-dependent bulk degradation process essential for cell viability but excessive autophagy leads to a unique form of cell death termed autosis. Triple-negative breast cancer (TNBC) is a highly aggressive subtype of breast cancer with notable defect in its autophagy process. In previous studies, we developed stapled peptides that specifically targeted the essential autophagy protein Beclin 1 to induce autophagy and promote endolysosomal trafficking. Here we show that one lead peptide Tat-SP4 induced mild increase of autophagy in TNBC cells but showed potent anti-proliferative effect that could not be rescued by inhibitors of programmed cell death pathways. The cell death induced by Tat-SP4 showed typical features of autosis including sustained adherence to the substrate surface, rupture of plasma membrane and effective rescue by digoxin, a cardioglycoside that blocks the Na+/K+ ATPase. Tat-SP4 also induced prominent mitochondria dysfunction including loss of mitochondria membrane potential, elevated mitochondria reactive oxygen species and reduced oxidative phosphorylation. The anti-proliferative effect of Tat-SP4 was confirmed in a TNBC xenograft model. Our study uncovers three notable aspects of autosis. Firstly, autosis can be triggered by moderate increase in autophagy if such increase exceeds the endogenous capacity of the host cells. Secondly, mitochondria may play an essential role in autosis with dysregulated autophagy leading to mitochondria dysfunction to trigger autosis. Lastly, intrinsic autophagy deficiency and quiescent mitochondria bioenergetic profile likely render TNBC cells particularly susceptible to autosis. Our designed peptides like Tat-SP4 may serve as potential therapeutic candidates against TNBC by targeting this vulnerability.

Abstract Background The drastic change in an ecosystem as a threshold phenomenon caused by abrupt changes in environmental conditions is a focus of current ecological research. However, the study of ecological thresholds has generally been limited to estimating the threshold values of single factors. Using eDNA metabarcoding technology, we collected zooplankton data from Baiyangdian Lake, the largest freshwater lake in the North China Plain, to explore the zooplankton community distribution characteristics and the relevant environmental factors. We used Threshold Indicator Taxa Analysis (TITAN) to determine the thresholds of key environmental factors and to identify the factors influencing biological diversity. Results By comparing previous studies, we found that the zooplankton community composition based on eDNA metabarcoding was similar to that based on morphological methods, and that the data could be used to estimate ecological thresholds and assess risk conditions. Temperature (T), electrical conductivity (EC), and turbidity were the major environmental factors affecting the zooplankton community structure. The composition and structure of zooplankton communities in rivers and lakes were significantly different due to the influence of specific environmental factors. The results of TITAN analysis showed that there were different indicator species for T and EC in rivers and lakes. The protection thresholds of zooplankton in rivers were T = 19.0 °C and EC = 795 μS/cm, whereas the protection thresholds of zooplankton in lakes were T = 14.3 °C and EC = 1920 μS/cm. The overall values for the Baiyangdian watershed were T = 15.5 °C and EC = 1073 μS/cm. Compared with the field monitoring results, approximately 50% of the water quality index values at the sampling points in the Baiyangdian watershed exceeded the negative response threshold, indicating that Baiyangdian Lake was disturbed. Conclusions The validity of eDNA technology in biodiversity analysis was confirmed by the zooplankton community data from Baiyangdian Lake. The ecological thresholds derived by combining eDNA technology with Threshold Indicator Taxa Analysis (TITAN) are beneficial to the biological conservation of the region.

Abstract Background China is the largest producer of sweet potato in the world, accounting for 57.0% of the global output. Germplasm resources are the basis for promoting innovations in the seed industry and ensuring food security. Individual and accurate identification of sweet potato germplasm is an important part of conservation and efficient utilization. Results In this study, nine pairs of simple sequence repeat molecular markers and 16 morphological markers were used to construct genetic fingerprints for sweet potato individual identification. Combined with basic information, typical phenotypic photographs, genotype peak graphs, and a two-dimensional code for detection and identification were generated. Finally, a genetic fingerprint database containing 1021 sweet potato germplasm resources in the “National Germplasm Guangzhou Sweet Potato Nursery Genebank in China” was constructed. Genetic diversity analysis of the 1021 sweet potato genotypes using the nine pairs of simple sequence repeat markers revealed a narrow genetic variation range of Chinese native sweet potato germplasm resources, and Chinese germplasm was close to that from Japan and the United States, far from that from the Philippines and Thailand, and the furthest from that from Peru. Sweet potato germplasm resources from Peru had the richest genetic diversity, supporting the view that Peru is the center of origin and domestication of sweet potato varieties. Conclusions Overall, this study provides scientific guidance for the conservation, identification, and utilization of sweet potato germplasm resources and offers a reference to facilitate the discovery of important genes to boost sweet potato breeding.

Abstract Background Increasing evidence suggests that lncRNA (Long non‐coding RNA, lncRNA)‐mediated ceRNA (competing endogenous RNA, ceRNA) networks are involved in the occurrence and progression of colorectal cancer (CRC). However, the roles of the lncRNA–miRNA–mRNA ceRNA network in distant metastasis of CRC are still unclear. Methods In this study, we constructed a specific ceRNA network to identify potential biomarkers and therapeutic targets for distant metastasis of CRC. Specifically, RNA‐Seq data from The Cancer Genome Atlas (TCGA) were used to screen for differentially expressed lncRNAs (DElncRNAs) and mRNAs (DEmRNAs) related to metastasis. After validation and selection by qRT–PCR and univariate and multivariate analysis of the metastasis‐ and prognosis‐related lncRNAs, the regulated microRNAs (miRNAs) and coexpressed mRNAs were used to construct a ceRNA network for distant metastasis of CRC. Results Two key distant metastasis‐related DElncRNAs, AP002498.1 and LINC01871, were identified by univariate and multivariate analysis in combination with analyses of clinical data and expression levels. Furthermore, lncRNA‐associated ceRNA subnetworks were constructed from the predicted miRNAs and 13 coexpressed DEmRNAs (SERPINA1, ITLN1, REG4, L1TD1, IGFALS, MUC5B, CIITA, CXCL9, CXCL10, GBP4, GNLY, IDO1, and NOS2). The AP002498.1‐ and LINC01871‐associated ceRNA subnetworks regulated the expression of the target genes SERPINA1 and MUC5B and GNLY, respectively, through the associated miRNAs. Conclusion The DElncRNA AP002498.1 and the LINC01871/miR‐4644 and miR‐185‐5p/GNLY axes were identified as being closely associated with distant metastasis and could represent independent prognostic biomarkers or therapeutic targets in colorectal adenocarcinoma.

Abstract Cloud lifecycles are influenced by interactions among clouds and mixing with cloud‐free environments. Cloud shapes dictate the length of cloud boundaries, which affects these interactions. This study uses a large eddy simulation to examine the relationships between the shallow cumulus cloud shapes, processes at their boundaries, and lifecycles. To characterize the shape of clouds, two metrics are newly defined: one reflects the overall horizontal cloud shape, and the other defines the cloud edge irregularity. Simulated clouds experience increased irregularity, but little changes in aspect ratio. Irregularity‐driven cloud perimeter growth strongly indicates cloud splitting, with a more pronounced effect than aspect ratio‐driven perimeter growth. Smaller cloud‐shell gradients of properties across cloud boundaries are associated with more irregular clouds, suggesting enhanced mixing of clouds with the cloud‐free environment. These findings indicate that the shallow cumulus evolution is partly driven by the irregularity of the cloud edge through lateral mixing and cloud splitting.

Electroreduction of oxygen driven by renewable electricity holds significant promise for the sustainable production of value‐added hydrogen peroxide (H2O2). While water is a desirable source of protons and electrons for this reaction, its low gas solubility often limits the transportation of gas molecules and consequently leads to large concentration overpotential, resulting in unsatisfactory energy efficiencies. Herein, a facile and effective strategy to promote the 2e− oxygen reduction reaction (ORR) through microenvironment modulation is presented. In this work, it is specifically aimed to facilitate oxygen transportation at the reaction interface, particularly at high rates. To achieve this, hydrophobic polytetrafluoroethylene (PTFE) particles are introduced into a catalyst layer containing amino‐group‐functionalized carbon nanotube (CNT‐NH2) as the ORR catalyst for H2O2 production. As a result, the PTFE‐modified CNT‐NH2‐based gas‐diffusion electrode (GDE) substantially improves ORR activity. At 100 mA cm−2, the PTFE‐modified CNT‐NH2 achieves a high cathodic energy efficiency of 92%, 1.5 times higher than the pristine CNT‐NH2‐based GDE (63%). Detailed kinetic analysis reveals that this enhanced ORR performance is indeed due to the enhanced oxygen transportation induced by the persistent hydrophobic microenvironment created by the PTFE‐modified catalyst layer, reducing the concentration overpotential during ORR.

The synergistic anti-tumor therapy based on chemotherapeutic drugs and gene drugs has become a new direction of tumor treatment. However, the significantly distinct physicochemical properties and the unbalanced coordinate proportion of chemotherapeutic and genetic drugs limit the actual therapeutic efficiency of the combined drug delivery strategy. Herein, we developed a simple method to self-assemble the fluorine atom-modified antisense oligonucleotide (2′F-G1319) with chemotherapeutic pirarubicin (THP) to form the carrier-free nanomedicines (2′F-G1319/THP NMs), which could efficaciously achieve the synergistic gene therapy and chemotherapy. In this study, 2′F-G1319 and THP could be self-assembled to form uniform nanoparticles through enhanced intermolecular interactions after G1319 was modified with the fluorine atom. Both in-vitro and in-vivo experiments verified that 2′F-G1319/THP NMs exhibited significant synergistic anti-tumor effects with high biostability and biocompatibility. Owing to the simple and universal synthesis process, our constructed self-assembled nanomedicine provides a promising opportunity for the wide applications of nanomedicines and is expected to realize more effective synergistic anti-tumor therapy without the addition of non-therapeutic excipients

ABSTRACTThe implementation of process analytical technologies is positioned to play a critical role in advancing biopharmaceutical manufacturing by simultaneously resolving clinical, regulatory, and cost challenges. Raman spectroscopy is emerging as a key technology enabling in-line product quality monitoring, but laborious calibration and computational modeling efforts limit the widespread application of this promising technology. In this study, we demonstrate new capabilities for measuring product aggregation and fragmentation in real-time during a bioprocess intended for clinical manufacturing by applying hardware automation and machine learning data analysis methods. We reduced the effort needed to calibrate and validate multiple critical quality attribute models by integrating existing workflows into one robotic system. The increased data throughput resulting from this system allowed us to train calibration models that demonstrate accurate product quality measurements every 38 s. In-process analytics enable advanced process understanding in the short-term and will lead ultimately to controlled bioprocesses that can both safeguard and take necessary actions that guarantee consistent product quality.

Abstract Background The morbidity and mortality of sepsis are extremely high, which is a major problem plaguing human health. However, current drugs and measures for the prevention and treatment of sepsis have little effect. Sepsis-associated acute liver injury (SALI) is an independent risk factor for sepsis, which seriously affects the prognosis of sepsis. Studies have found that gut microbiota is closely related to SALI, and indole-3-propionic Acid (IPA) can activate Pregnane X receptor (PXR). However, the role of IPA and PXR in SALI has not been reported. Methods This study aimed to explore the association between IPA and SALI. The clinical data of SALI patients were collected and IPA level in feces was detected. The sepsis model was established in wild-type mice and PXR knockout mice to investigate the role of IPA and PXR signaling in SALI. Results We showed that the level of IPA in patients’ feces is closely related to SALI, and the level of IPA in feces has a good ability to identify and diagnose SALI. IPA pretreatment significantly attenuated septic injury and SALI in wild-type mice, but not found in knockout PXR gene mice. Conclusions IPA alleviates SALI by activating PXR, which reveals a new mechanism of SALI, and provides potentially effective drugs and targets for the prevention of SALI.

In the manufacturing process of lithium-ion power battery, winding is a very critical process. In order to ensure the safety and stability of the battery, no buckling, wrinkling, and even fracture is allowed in the process of winding. Different battery models lead to different winding methods for the jellyroll. The most suitable winding method is determined by the external radius of the jellyroll. First, the winding layer is treated as a thin-film material, the constitutive equation of jellyroll is established by considering the constraint of winding tension to obtain hoop stress and radial stress distribution function at the place of r under plane strain state. The hoop stress and the radial stress at any place of r inside the jellyroll are obtained at the end of n coils. Moreover, the hoop stresses of the jellyrolls with different external radius are analyzed by calculating examples according to the type of winding tension, and the suitable winding methods for different external radius are suggested. With the increase of the external radius of jellyroll, the constant winding tension and taper winding tension are gradually no longer suitable, and the parabolic winding tension and compound winding tension are proposed. After the winding of the power battery is completed, the stress generated by the winding tension of jellyroll is equivalent to the prestress applied to the battery, which will affect the use of the power battery.

Abstract Background Fibroblast growth factors (FGFs) and their receptors (FGFRs) play a crucial role in cell fate and angiogenesis, with dysregulation of the signaling axis driving tumorigenesis. Therefore, many studies have targeted FGF/FGFR signaling for cancer therapy and several FGFR inhibitors have promising results in different tumors but treatment efficiency may still be improved. The clinical use of immune checkpoint blockade (ICB) has resulted in sustained remission for patients. Main Although there is limited data linking FGFR inhibitors and immunotherapy, preclinical research suggest that FGF/FGFR signaling is involved in regulating the tumor microenvironment (TME) including immune cells, vasculogenesis, and epithelial-mesenchymal transition (EMT). This raises the possibility that ICB in combination with FGFR-tyrosine kinase inhibitors (FGFR-TKIs) may be feasible for treatment option for patients with dysregulated FGF/FGFR signaling. Conclusion Here, we review the role of FGF/FGFR signaling in TME regulation and the potential mechanisms of FGFR-TKI in combination with ICB. In addition, we review clinical data surrounding ICB alone or in combination with FGFR-TKI for the treatment of FGFR-dysregulated tumors, highlighting that FGFR inhibitors may sensitize the response to ICB by impacting various stages of the “cancer-immune cycle”.

Vat photopolymerization (VP), as an additive manufacturing process, has experienced significant growth due to its high manufacturing precision and excellent surface quality. This method enables the fabrication of intricate shapes and structures while mitigating the machining challenges associated with non-oxide ceramics, which are known for their high hardness and brittleness. Consequently, the VP process of non-oxide ceramics has emerged as a focal point in additive manufacturing research areas. However, the absorption, refraction, and reflection of ultraviolet light by non-oxide ceramic particles can impede light penetration, leading to reduced curing thickness and posing challenges to the VP process. To enhance the efficiency and success rate of this process, researchers have explored various aspects, including the parameters of VP equipment, the composition of non-oxide VP slurries, and the surface modification of non-oxide particles. Silicon carbide and silicon nitride are examples of non-oxide ceramic particles that have been successfully employed in VP process. Nonetheless, there remains a lack of systematic induction regarding the curing mechanisms and key influencing factors of the VP process in non-oxide ceramics. This review firstly describes the curing mechanism of the non-oxide ceramic VP process, which contains the chain initiation, chain polymerization, and chain termination processes of the photosensitive resin. After that, the impact of key factors on the curing process, such as the wavelength and power of incident light, particle size, volume fraction of ceramic particles, refractive indices of photosensitive resin and ceramic particles, incident light intensity, critical light intensity, and the reactivity of photosensitive resins, are systematically discussed. Finally, this review discusses future prospects and challenges in the non-oxide ceramic VP process. Its objective is to offer valuable insights and references for further research into non-oxide ceramic VP processes.

Camellia oleifera oil (CO oil) extracted from C. oleifera seeds has a 2300-year consumption history in China. However, there is relatively little research regarding its non-edible uses. This study determined the physicochemical properties of CO oil extracted via direct pressing, identified its main components using GC-MS, and evaluated its antioxidant, moisturizing, and anti-inflammatory activities. The results revealed that CO oil’s acid, peroxide, iodine, and saponification values were 1.06 ± 0.031 mg/g, 0.24 ± 0.01 g/100 g, 65.14 ± 8.22 g/100 g, and 180.41 ± 5.60 mg/g, respectively. CO oil’s tocopherol, polyphenol, and squalene contents were 82.21 ± 9.07 mg/kg, 181.37 ± 3.76 mg/kg, and 53.39 ± 6.58 mg/kg, respectively; its unsaturated fatty acid (UFA) content was 87.44%, and its saturated fatty acid (SFA) content was 12.56%. CO oil also demonstrated excellent moisture retention properties, anti-inflammatory effects, and certain free radical scavenging. A highly stable CO oil emulsion with competent microbiological detection was developed using formulation optimization. Using CO oil in the emulsion significantly improved the formulation’s antioxidant and moisturizing properties compared with those of the emulsion formulation that did not include CO oil. The prepared emulsion was not cytotoxic to cells and could reduce cells’ NO content; therefore, it may have potential nutritional value in medicine and cosmetics.

The 2019 novel coronavirus disease (COVID-19) has lasted for over 3 years and has seriously affected the regular life trajectory of human beings. The severe acute respiratory syndrome coronavirus 2 (SARS‑CoV‑2) has caused non-negligible harm to people’s respiratory systems and multiple organs. Although the pathogenesis of COVID-19 has been fully elucidated, there is still a lack of effective and specific treatment for COVID-19. Mesenchymal stem cells (MSCs) or MSC-derived extracellular vesicles (MSC-EVs) by far have become the most promising candidates in preclinical studies and clinical trials, and MSC-related therapies have shown their potential to treat severe COVID-19. The multidirectional differentiation potential and immunomodulatory function of MSCs have allowed the cells to exert multiple cellular and molecular actions on different immune cells and organs. It is critical to understand the therapeutic roles of MSCs before entering the clinics for COVID-19 and other diseases. This review summarizes the recent progress of particular mechanisms underlying the immunomodulatory and tissue regenerative effects of MSCs toward COVID-19. We focused on discussing the functional roles of MSC-mediated effects on immune cell responses, cell survival, and organ regeneration. Besides, the novel discoveries and recent findings of MSC clinical application in patients with COVID-19 were highlighted. This will provide an overview of the current research on the rapid development of MSC-based therapies not only for COVID-19 but also for other immune-mediated/immune-dysregulating diseases.

The variability of surface air temperature is of great importance for both society and the environment, is impacted by global warming and local-scale changes. In the arid eastern part of Washington state of USA, substantial urbanization has transformed the Tri-cities into the state’s third-largest urban cluster. This study utilizes a combination of in situ observations and reanalysis datasets to investigate the influence of land use changes on the region’s 2-meter temperature, revealing local effects that compensate for the background global warming. Within the urban fabric, distinct microclimates have emerged due to varying land use, establishing unique relationships between greenness and temperature alterations caused by land use transitions. Notably, our findings demonstrate that the observed compensating signal in the temperature of farmland locations is primarily attributed to local vegetation increases. Through these observations, this research highlights the urban impacts on local climate, offering valuable insights into the complexities surrounding land use and its consequences on the environment.

Abstract Background Severe fever with thrombocytopenia syndrome (SFTS), one kind of tick-borne acute infectious disease, is caused by a novel bunyavirus. The relationship between meteorological factors and infectious diseases is a hot topic of current research. Liaoning Province has reported a high incidence of SFTS in recent years. However, the epidemiological characteristics of SFTS and its relationship with meteorological factors in the province remain largely unexplored. Methods Data on reported SFTS cases were collected from 2011 to 2019. Epidemiological characteristics of SFTS were analyzed. Spearman’s correlation test and generalized linear models (GLM) were used to identify the relationship between meteorological factors and the number of SFTS cases. Results From 2011 to 2019, the incidence showed an overall upward trend in Liaoning Province, with the highest incidence in 2019 (0.35/100,000). The incidence was slightly higher in males (55.9%, 438/783), and there were more SFTS patients in the 60–69 age group (31.29%, 245/783). Dalian City and Dandong City had the largest number of cases of SFTS (87.99%, 689/783). The median duration from the date of illness onset to the date of diagnosis was 8 days [interquartile range (IQR): 4–13 days].
