Abstract Understanding the genetic factors related to meat drip loss is of great importance for animal breeding and production. In this study, we employed a combination of genome‐wide association study (GWAS) mapping and RNA sequencing (RNA‐seq) data to effectively identify potentially functional single nucleotide polymorphisms (SNPs) as well as candidate genes associated with drip loss (DL) in Beijing Black pigs. Initially, we conducted a single‐ and multi‐trait GWAS on drip loss traits in 441 Beijing Black pigs at 24 (DL24) and 48 (DL48) hours postmortem using the Illumina pig 50K SNP chip. Five SNPs with annotations for four genes (FGGY, LHFPL6, OSBPL1A, and NMNAT3) were consistently identified in single or multiple trait GWAS results, indicating their potential pleiotropic effects on drip loss. Next, a comprehensive comparative transcriptomic analysis was performed on samples of Beijing Black pigs exhibiting extremely high and low drip loss, resulting in the identification of 21 differentially expressed genes (DGEs) as potential candidates. Additionally, protein–protein interaction (PPI) network analysis revealed reciprocal regulatory relationships between FOXO1, OSBPL1A, DOCK1 (identified from GWAS) and the candidate DGEs obtained from RNA‐seq data. Therefore, we propose that these genes may impact drip loss traits through gene interactions. In conclusion, our integrative analysis screened candidate genes that may affect the drip loss traits in Beijing Black pigs, which provides crucial insights into the molecular mechanisms of drip loss and serves as a theoretical reference for improving meat quality in Beijing Black pigs.

Abstract To reduce the unpleasant flavor in tea infusion made from summer fresh leaves and lessen tea resource wastage, a short‐term cycled heaping method, addressing the temperature/moisture variations during prolonged heaping, was introduced to produce high‐quality yellow tea with significantly enhanced taste and aroma. Furthermore, a hyperspectral approach was developed to anticipate such improvements. Specifically, short‐term cycled heaping reduced summer tea's astringency and bitterness while increasing sweetness. Using near‐infrared spectroscopy, the multiplicative scatter correction (MSC)–support vector machine (SVM) model predicted tea umami with 66.25% precision and richness with 77.64% accuracy via standard normal variate (SNV)–SVM, based on epigallocatechin (EGC), epicatechin (EC), and gallocatechin gallate (GCG) variations. Aroma enhancement was forecasted by a hyperspectral–electronic nose regression model, achieving over 81.50% prediction accuracy for summer tea aromas in the visible spectrum and surpassing 73.45% in the near‐infrared domain, primarily attributed to pentanal, propanal, toluene, ethyl propionate, and 2,3‐pentanedione variations. Overall, hyperspectral‐guided summer tea processing, particularly for summer‐harvested leaves, shows potential in offering a valuable screening tool, enhancing quality, and efficiency in the tea industry and promoting sustainable development.

Abstract pH value is a crucial index used to evaluate pork quality due to its direct impact on specific meat characteristics. This study investigated the genetic mechanisms influencing pH values through measurements taken from the longissimus dorsi muscle of Beijing Black pigs at 2 h (pH2h) and 24 h (pH24h) postmortem. A total of 614 Beijing Black pigs were subsequently genotyped using the Illumina Porcine 50K SNP Chip. Heritability estimates for pH2h and pH24h were found to be 0.19 and 0.25, respectively, with a genetic correlation of 0.53. Furthermore, we conducted both a genome‐wide association study (GWAS) and an RNA sequencing (RNA‐seq) analysis, the latter of which identified differentially expressed genes (DEGs) between high and low pH groups. We identified 31, 6, and 32 single‐nucleotide polymorphisms in the pH2h, pH24h, and pH2–24h traits, respectively. The GWAS results revealed the presence of the SYT5 gene in both the pH2h and pH2–24h traits, while the SNX13 gene was simultaneously identified in the pH24h and pH2–24h traits. The RNA‐seq results also found SYT5 to be highly expressed, while SNX13 did not exhibit differential expression. Moreover, Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses based on the DEGs revealed potential links between pH levels and the glycogen metabolic process as well as associations with the regulation of cell proliferation and calcium ion transmembrane transport. Ultimately, SYT5 and SNX13 emerged as key candidate genes affecting pH values at 2 and 24 h, respectively. These findings contribute to a better understanding of the genetic mechanisms affecting pork quality and safety and offer insights for enhancing meat quality through genetic improvement.

Abstract Background The objective of this research is to clarify the impact of periodontitis on overall and cardiovascular-related death rates among hypertensive individuals. Method A total of 5665 individuals with hypertension were included from the National Health and Nutrition Examination Survey (NHANES) data spanning 2001–2004 and 2009–2014. These individuals were divided into two groups based on the presence or absence of periodontitis and further stratified by the severity of periodontitis. We employed weighted multivariate Cox proportional hazards regression and Kaplan-Meier curves (log-rank test) to evaluate the impact of periodontitis on all-cause and cardiovascular mortality. Additional analyses, including adjustments for various covariates, subgroups, and sensitivity analyses, were conducted to ensure the robustness and reliability of our results. Result Over an average follow-up duration of 10.22 years, there were 1,122 all-cause and 297 cardiovascular deaths. Abstract The biodegradable polymer poly-β-hydroxybutyrate (PHB) is a promising carbon source for biological mitigation of nitrogen pollution, a significant problem in aquaculture that physical and chemical methods have not provided a comprehensive solution. Here we investigated the impact of PHB on the zero-water-change largemouth bass culture by 30- and 40-day experiments. PHB loaded into the filter circulation pump at 4 g L− 1, optimum value determined by the first experiment, significantly reduced the levels of nitrate by 99.65%, nitrite by 95.96%, and total nitrogen by 85.22% compared to the control without PHB. PHB also significantly increased denitrifying bacteria (e.g., Proteobacteria and Fusobacteria) and expression of denitrification genes (e.g., nirK and nirS) in the microbial community, improving growth and health parameters of largemouth bass. While the impact may vary in other culture systems, PHB thus demonstrated its remarkable utility in aquaculture, highlighting ecological assessment and application to larger aquaculture operations as future considerations.

The pregnane X receptor (PXR)/nuclear factor-kB (NF-kB) signaling pathway plays a critical role in regulating toxin-induced inflammation and apoptosis in mammals. Whether dietary curcumin (CUR) can prevent copper (Cu)-induced liver injury via this signaling pathway remains to be established in aquatic animals. Juvenile largemouth bass (Micropterus salmoides) were exposed to dietary Cu and CUR treatments for 8 weeks. The results showed that chronic Cu exposure induced oxidative stress, causing liver function damage and liver injury. Cu exposure stimulated inflammation by regulating nf-kb and pro-inflammatory genes such as tnfα and il-1β and promoted apoptotic signals in the liver by modulating bcl2 and casp3 mRNA levels. In addition, Pearson correlation analysis verified that inflammatory and apoptotic responses were important indicators of Cu-induced liver injury. CUR attenuated stress responses by enhancing the antioxidant system. Importantly, CUR significantly stimulated PXR mRNA and protein levels in the Cu + CUR group and suppressed NF-κB activation to inhibit the inflammatory and apoptotic signaling cascade. These results suggest that CUR may be an effective activator of PXR in teleost fishes, exerting cytoprotective effects on Cu-induced liver injury via a PXR-mediated NF-κB repression mechanism. In conclusion, this study is the first to demonstrate that CUR may act as a potent PXR ligand that exerts hepatoprotective effects against Cu-induced liver injury. The findings shed light on the specific regulatory role of the PXR/NF-κB signaling pathway in liver pathogenesis and its potential as a therapeutic target in teleost fishes.

Enteritis often limits intensive aquaculture, where bile acids (BAs), a group of steroid molecules, are used to prevent intestinal diseases. However, the functions of certain BAs are poorly understood. In this study, grass carp with enteritis (EGC) and healthy grass carp (HGC) were sampled, and the BAs' composition was analyzed. Results showed distinct differences in the BA profiles: EGC exhibited significantly elevated levels of taurodeoxycholic acid (TDCA), lithocholic acid (LCA), glycoursodeoxycholic acid (GUDCA), and glycohyocholic acid (GHCA) (P < 0.05), and decreased levels of TCDCA, taurocholic acid (TCA), and cholic acid (CA) in the intestine compared with their levels in HGC. Subsequently, five diets containing four typical BAs (CA, chenodeoxycholic acid [CDCA], deoxycholic acid [DCA], and LCA) at the same concentration (0.1 %) were formulated to feed the juvenile grass carps (6.29 ± 0.05 g) for 8 weeks. DCA alone increased body weight, reduced the feed conversion ratio (FCR), and ameliorated histological intestinal inflammatory hallmarks. Conversely, individual supplementation with CDCA, LCA, or CA decreased body weight and increased FCR and exacerbated the histological hallmarks of enteritis. Furthermore, DCA markedly reduced the mRNA expression levels of endoplasmic reticulum stress (ERS)-related genes in the intestine, including glucose-regulated protein 78 (GPR78), activating transcription factor 6 (ATF6), inositol-requiring enzyme 1, and protein kinase RNA-like ER kinase, as well as genes encoding proinflammatory cytokines, such as interleukin (IL)-1 beta, IL-8, and tumor necrosis factor α (TNF-α). Supplementation with CDCA, LCA, or CA promoted the expression of ERS- and proinflammatory-cytokine-related genes. Additionally, Actinobacteriota abundance negatively correlated with intestinal mRNA expression levels of TNF-α, ATF6, IL-1β, GPR78, and IL-8, and Candidatus-Arthromitus abundance positively correlated with these inflammatory markers (P < 0.05). In conclusion, our results demonstrated that BAs play differential roles in modulating the enteritis process and that DCA may have a specific anti-inflammatory role in grass carp.

The treatment of breast cancer (BC) remains a significant challenge, exemplified by the limitations of chemotherapy due to its high side effects. Light-responsive drug delivery systems (DDS) offer a promising approach to spatially and temporally release drugs, minimizing chemotherapy side effects and enhancing efficacy. However, achieving precise delivery and site-specific drug release poses complex challenges. Here, We present a novel nano-system combining light-responsive photothermal therapy (PTT) with photothermal-responsive chemotherapy via π − π Stacking. Conjugated polymer nanoparticles (cRGD-PTer N25/CPT NPs) is designed for spatiotemporal targeted PTT/chemotherapy. Research shows that the strong near-infrared absorption of PTer N25 endows cRGD-PTer N25/CPT NPs with excellent photothermal capabilities(57.4 %) and near-infrared laser-triggered drug release performance. The cRGD-PTer N25/CPT NPs enhanced BC-specific cellular accumulation, prolonged blood circulation, exhibited good biocompatibility, and improved PTT and chemotherapy efficacy in BC cell lines and mouse models while reducing systemic toxicity. The photothermal effect triggered the on-demand release of camptothecin (CPT), producing therapeutic effects and downregulating heat shock protein 70 (HSP 70), reducing cancer cell thermoresistance, and enhancing combined treatment efficacy. Under spatial and temporal control, the tumor growth inhibition rate of the cRGD-PTer N25/CPT NPs group reached 93.6 %, nearly eradicating tumor presence, whereas the control group exhibited only partial inhibition. Minimal cardiotoxicity and metastatic side effects are not observed. This work presents a viable strategy for designing novel controlled-release drug systems to improve breast cancer treatment efficiency.

Abstract Broad bean (Vicia faba L.) has received attention from the food and animal feed industries as a sustainable, inexpensive, and versatile plant‐based protein source. However, long‐term consumption of broad bean, especially unprocessed ones, can lead to hepatic lipid accumulation. To investigate how broad bean affected hepatic lipid metabolism and its underlying mechanisms, we conducted a multi‐omics analysis of crisp grass carp (Ctenopharyngodon idellus C. et V) model with hepatic steatosis after feeding with single broad bean for 120 days. The glycolysis inhibition, increased de novo lipogenesis, abnormalities in lipid metabolism, and activation of oxidative stress were the key processes involved in broad bean‐induced hepatic lipid accumulation. Disturbances in intestinal flora caused by damage to the intestinal barrier and increased lipopolysaccharide caused by broad bean promoted hepatic lipid accumulation. Reduction of antinutritional factors and balanced diet offer potential strategies to mitigate the liver damage caused by broad bean consumption. Overall, our findings provided a theoretical basis for the wide application of broad bean as animal feeds.

Aiming to address power consumption issues of various equipment in metro stations and the inefficiency of peak shaving and valley filling in the power supply system, this study presents an economic optimization scheduling method for the multi-modal “source-network-load-storage” system in metro stations. The proposed method, called the Improved Gray Wolf Optimization Algorithm (IGWO), utilizes objective evaluation criteria to achieve economic optimization. First, construct a mathematical model of the “sourcenetwork- load-storage” joint system with the metro station at its core. This model should consider the electricity consumption within the station. Secondly, a two-layer optimal scheduling model is established, with the upper model aiming to optimize peak elimination and valley filling, and the lower model aiming to minimize electricity consumption costs within a scheduling cycle. Finally, this paper introduces the IGWO optimization approach, which utilizes meta-models and the Improved Gray Wolf Optimization Algorithm to address the nonlinearity and computational complexity of the two-layer model. The analysis shows that the proposed model and algorithm can improve the solution speed and minimize the cost of electricity used by about 5.5% to 8.7% on the one hand, and on the other hand, it improves the solution accuracy, and at the same time effectively realizes the peak shaving and valley filling, which provides a proof of the effectiveness and feasibility of the new method.

Abstract This study assesses a 40‐day 3.25‐km global simulation of the Simple Cloud‐Resolving E3SM Model (SCREAMv0) using high‐resolution ground‐based observations from the Atmospheric Radiation Measurement (ARM) Green Ocean Amazon (GoAmazon) field campaign. SCREAMv0 reasonably captures the diurnal timing of boundary layer clouds yet underestimates the boundary layer cloud fraction and mid‐level congestus. SCREAMv0 well replicates the precipitation diurnal cycle, however it exhibits biases in the precipitation cluster size distribution compared to scanning radar observations. Specifically, SCREAMv0 overproduces clusters smaller than 128 km, and does not form enough large clusters. Such biases suggest an inhibition of convective upscale growth, preventing isolated deep convective clusters from evolving into larger mesoscale systems. This model bias is partially attributed to the misrepresentation of land‐atmosphere coupling. This study highlights the potential use of high‐resolution ground‐based observations to diagnose convective processes in global storm resolving model simulations, identify key model deficiencies, and guide future process‐oriented model sensitivity tests and detailed analyses.

Habitual dietary changes have the potential to induce alterations in the host’s gut microbiota. Mandarin fish (Siniperca chuatsi), an aquatic vertebrate species with distinct feeding habits, were fed with natural feeds (NF) and artificial feeds (AF) to simulate the effects of natural and processed food consumption on host gut microbiota assemblages. The results showed that the alpha diversity index was reduced in the AF diet treatment, as lower abundance and diversity of the gut microbiota were observed, which could be attributed to the colonized microorganisms of the diet itself and the incorporation of plant-derived proteins or carbohydrates. The β-diversity analysis indicated that the two dietary treatments were associated with distinct bacterial communities. The AF diet had a significantly higher abundance of Bacteroidota and a lower abundance of Actinomycetota, Acidobacteriota, and Chloroflexota compared to the NF group. In addition, Bacteroidota was the biomarker in the gut of mandarin fish from the AF treatment, while Acidobacteriota was distinguished in the NF treatments. Additionally, the increased abundance of Bacteroidota in the AF diet group contributed to the improved fermentation and nutrient assimilation, as supported by the metabolic functional prediction and transcriptome verification. Overall, the present work used the mandarin fish as a vertebrate model to uncover the effects of habitual dietary changes on the evolution of the host microbiota, which may provide potential insights for the substitution of natural foods by processed foods in mammals.

Diets with high carbohydrate (HC) was reported to have influence on appetite and intermediary metabolism in fish. To illustrate whether betaine could improve appetite and glucose-lipid metabolism in aquatic animals, mandarin fish (Siniperca chuatsi) were fed with the HC diets with or without betaine for 8 weeks. The results suggested that betaine enhanced feed intake by regulating the hypothalamic appetite genes. The HC diet-induced downregulation of AMPK and appetite genes was also positively correlated with the decreased autophagy genes, suggesting a possible mechanism that AMPK/mTOR signaling might regulate appetite through autophagy. The HC diet remarkably elevated transcriptional levels of genes related to lipogenesis, while betaine alleviated the HC-induced hepatic lipid deposition. Additionally, betaine supplementation tended to store the energy storage as hepatic glycogen. Our findings proposed the possible mechanism for appetite regulation through autophagy via AMPK/mTOR, and demonstrated the feasibility of betaine as an aquafeed additive to regulate appetite and intermediary metabolism in fish.

Abstract Background The Fe (II)- and α-ketoglutarate-dependent AlkB family dioxygenases are implicated in nucleotide demethylation. AlkB homolog1 (ALKBH1) is shown to demethylate DNA adenine methylation (6mA) preferentially from single-stranded or unpaired DNA, while its demethylase activity and function in the chromatin context are unclear. Results Here, we find that loss-of-function of the rice ALKBH1 gene leads to increased 6mA in the R-loop regions of the genome but has a limited effect on the overall 6mA level. However, in the context of mixed tissues, rather than on individual loci, the ALKBH1 mutation or overexpression mainly affects the expression of genes with a specific combination of chromatin modifications in the body region marked with H3K4me3 and H3K27me3 but depleted of DNA CG methylation. In the similar context of mixed tissues, further analysis reveals that the ALKBH1 protein preferentially binds to genes marked by the chromatin signature and has a function to maintain a high H3K4me3/H3K27me3 ratio by impairing the binding of Polycomb repressive complex 2 (PRC2) to the targets, which is required for both the basal and stress-induced expression of the genes. Conclusion Our findings unravel a function of ALKBH1 to control the balance between the antagonistic histone methylations for gene activity and provide insight into the regulatory mechanism of PRC2-mediated H3K27me3 deposition within the gene body region.

In China, aquaculture is the primary source of aquatic products due to the decrease in wild fishery resources. In 2018, the total output of aquatic products in China expanded to 47.6 million tons, accounting for 58% of global aquaculture production. Intensive culture methods generally use significant quantities of feed however, approximately 75% of nitrogen in the feed is retained in aquaculture water, mainly as soluble nitrogen, such as ammonia nitrogen (NH4+-N) and nitrate (NO3–-N), owing to low feed-utilization rates during cultivation. At the same time, fishes generate a substantial amount of excreta, which will cause the increase of nitrogen compounds in water and negatively affects the quality of aquatic products. Serious problems could occur if nitrogen compounds are discharged into the environment, including the eutrophication of rivers, the deterioration of drinking water sources, and hazards to human health. Furthermore, nitrates can form potentially carcinogenic compounds, such as nitrosamines and nitrosamides, and nitrate consumption can cause methemoglobinemia in infants. The Second National Census of Pollution Sources survey showed that the total nitrogen emission from aquaculture was 99 100 tons in 2017. To protect the environment and human health, it is important to remove nitrogen from aquaculture tailwater before discharging it to the surrounding waters. Biological denitrification is considered the most promising approach since nitrate can be reduced to harmless nitrogen gas by bacteria. A sufficient carbon source is necessary during the heterotrophic denitrification process. To solve the problems mentioned above, external liquid carbon sources such as methanol, acetic acid, and glucose are added to the tailwater, but they are costly, require high-energy, and have high operating requirement. In contrast, agricultural wastes as a carbon source have exhibited significant economic advantages and high efficiency. Many aquaculture tailwater treatment systems often face variations in hydraulic retention times (HRT) and influent nitrate concentration (INC), which are caused by acute changes in tailwater characteristics and production, and HRT and INC often exert a profound effect on the treatment performance of biological treatment systems. Extensive research has confirmed that adding agricultural waste (such as corncob, woodchip and rice straw) to municipal sewage and industrial wastewater can effectively improve denitrification efficiency. However, the effect of using agricultural waste as denitrifying carbon source to treat aquaculture tail water remains unclear. Banana stalk (BS), a typical agricultural waste product, is used as a denitrifying carbon source for the first time in this study. The study investigated the effects of HRT and INC on the denitrification performance of BS, and provided a theoretical basis for the application of agricultural waste in aquaculture tailwater treatment. In this study, using BS as a carbon source and a towel as biological carrier, the performance of solid-phase denitrification under dynamic flow conditions was studied by using a 1-D column experiment. In the HRT optimization experiment, INC was maintained at 50 mg/L and operated under four HRTs (16 h, 20 h, 24 h and 28 h) for 14 days. The effluent NO3–-N, nitrite (NO2–-N), NH4+-N, Total nitrogen (TN), Total phosphorus (TP), and chemical oxygen demand (COD) removal efficiency were measured every 2 days. The optimal HRT of BS-DR (banana stalk-denitrification reactor) was optimized by one-way ANOVA analysis. Then, based on the optimization of HRT, the reactor was operated for 14 days under different INC (75 mg/L, 100 mg/L, and 125 mg/L). The sampling time interval and measurement indexes were the same as those of the HRT optimization experiment. The Illumina MiSeq high-throughput sequencing method was used to sequence and analyze the two hyper-variable regions (V3-V4) of the 16S rRNA gene of bacteria in the initial and final stages of the BS-DR. The results indicated that HRT and INC are the key factors affecting the denitrification performance of BS-DR. There was no significant difference in nitrate removal efficiency when the HRT was 20 h (96.71±1.36)%, 24 h (94.57±4.73)%, and 28 h (99.41±0.64)%, but they were significantly higher than that when the HRT was 16 h (87.53±7.95)%. Therefore, the optimal HRT for BS-DR was 20 h, and no nitrite accumulation. The second set of experiments was conducted using the optimal HRT obtained from the first set of experiments. The effluent nitrate concentration (ENC) and nitrate removal rate (NRR) of BS-DR increased significantly with increase in INC (P < 0.05), and the effluent COD decreased with increase in INC, and the proper INC for BS-DR was ≤50 mg/L. It is worth noting that BS-DR could completely remove NH4+-N in both experiments. In addition, HRT significantly affects the removal efficiency of TP, but INC has little effect. According to pyrosequencing analysis, the microbial community structure of BS-DR changed after long-term operation, with the relative abundances of Proteobacteria, Bacteroidetes, Campilobacterota, and Firmicutes increasing to 31.20%, 6.67%, 3.08%, and 4.26%, respectively, ensuring the efficient operation of the reactor. On the contrary, the relative abundances of Halobacterota, Desulfobacterota, Sva0485, Chloroflexi, and Verrucomicrobiota decreased to 10.39%, 5.13%, 2.82%, 2.00%, and 1.17 %, respectively, in the reactor. In addition, at the genus level, most of the dominant bacteria at the end of reactor operation play a role in denitrification and degradation of agricultural waste, which is significantly different from that at the beginning of the reactor operation.

Long-term nonunion of bone defects has always been a major problem in orthopedic treatment. Artificial bone graft materials such as Poly (lactic-co-glycolic acid)/β-tricalcium phosphate (PLGA/β-TCP) scaffolds are expected to solve this problem due to their suitable degradation rate and good osteoconductivity. However, insufficient mechanical properties, lack of osteoinductivity and infections after implanted limit its large-scale clinical application. Hence, we proposed a novel bone repair bioscaffold by adding zinc submicron particles to PLGA/β-TCP using low temperature rapid prototyping 3D printing technology. We first screened the scaffolds with 1 wt% Zn that had good biocompatibility and could stably release a safe dose of zinc ions within 16 weeks to ensure long-term non-toxicity. As designed, the scaffold had a multi-level porous structure of biomimetic cancellous bone, and the Young's modulus (63.41 ± 1.89 MPa) and compressive strength (2.887 ± 0.025 MPa) of the scaffold were close to those of cancellous bone. In addition, after a series of in vitro and in vivo experiments, the scaffolds proved to have no adverse effects on the viability of BMSCs and promoted their adhesion and osteogenic differentiation, as well as exhibiting higher osteogenic and anti-inflammatory properties than PLGA/β-TCP scaffold without zinc particles. We also found that this osteogenic and anti-inflammatory effect might be related to Wnt/β-catenin, P38 MAPK and NFkB pathways. This study lay a foundation for the follow-up study of bone regeneration mechanism of Zn-containing biomaterials. We envision that this scaffold may become a new strategy for clinical treatment of bone defects.

A three-level multi-input DC/DC converter is proposed to solve the problems of complex interface circuit structure and high economic cost for multi-source access to the joint power supply distribution system. In this structure, multiple dc sources are integrated into a three-level DC/DC converter. In comparison with the two-stage counterpart, two active switches and boost diodes are eliminated, while two blocking diodes are added to block the reverse current from the dc-link capacitors. In addition, when the input inductors work in the discontinuous conduction mode, power sharing among different input sources can be achieved by properly selecting the inductance value. The working principle of the converter is analyzed by introducing nine working modes in detail and deriving the steady-state relationship expressions. The parameter range of the element is determined and the design process of a group of dynamic parameter values is shown. Finally, the power electronics real-time simulation platform is built based on StarSim HIL and the corresponding experimental waveforms are given to verify the topology and analysis.

Introduction: Cadmium (Cd), one of the major toxic heavy metals, causes severe deleterious effects on all living organisms from prokaryotes to eukaryotes. Cadmium deposition affects bacterial diversity and bacterial population in soil. Cadmium accumulation in plants is mainly controlled by transporters and the resulting Cd enrichment gives rise to phytotoxicity. Objective: This study aimed to mine transporters that control Cd import or accumulation in rice and uncover the underlying mechanisms that how accumulated Cd poses risks to host plant and leaf bacteria. Methods: RNA-seq analysis, histochemical assays, and elemental quantification were carried out to reveal the biological roles of OsABCG43 for Cd import. Pathogen inoculation, IC50 value, and bacterial virulence assays were conducted to disclose the effects of Cd on leaf bacteria. Results: OsABCG43 is characterized as a Cd importer controlling Cd accumulation in rice. OsABCG43 was induced under Cd stress and specifically expressed in the vasculature of leaves and roots. Overexpression of OsABCG43 caused Cd accumulation which inhibits photosynthesis and development and alters the antioxidant system, resulting in phytotoxicity. Moreover, overexpression of OsABCG43 resulted in retarded plant growth and enhanced rice sensitivity to Cd stress. Numerous differentially expressed genes were identified via RNA-seq analysis between the OsABCG43-overexpressing plants and wild type, which functioned in Cd or reactive oxygen species (ROS) homeostasis. In addition, OsABCG43 transcripts were induced by leaf bacteria Xanthomonas oryzae pv. oryzicola (Xoc) and X. oryzae pv. oryzae (Xoo). The enriched Cd directly impaired the formation of virulence factors for the leaf bacteria, preventing colonization or proliferation of Xoc or Xoo in rice leaves. Conclusion: This work reveals that OsABCG43 is expressed specifically in the vascular and plasma membrane-localized OsABCG43 functions as a Cd importer. OsABCG43-mediated import of Cd is harmful for both rice and the corresponding leaf bacteria.

Microcystin-LR (MC-LR) is a cyanobacterial metabolite produced during cyanobacterial blooms and is toxic to aquatic animals, and the liver is the main targeted organ of MC-LR. To comprehensively understand the toxicity mechanism of chronic exposure to environmental levels of MC-LR on the liver of fish, juvenile Nile tilapia were exposed to 0 μg/L (control), 1 μg/L (M1), 3 μg/L (M3), 10 μg/L (M10), and 30 μg/L (M30) MC-LR for 60 days. Then, the liver hepatotoxicity induced by MC-LR exposure was systematically evaluated via histological and biochemical determinations, and the underlying mechanisms were explored through combining analysis of biochemical parameters, multi-omics (transcriptome and metabolome), and gene expression. The results exhibited that chronic MC-LR exposure caused slight liver minor structural damage and lipid accumulation in the M10 group, while resulting in serious histological damage and lipid accumulation in the M30 group, indicating obvious hepatotoxicity, which was confirmed by increased toxicity indexes (i.e., AST, ALT, and AKP). Transcriptomic and metabolomic analysis revealed that chronic MC-LR exposure induced extensive changes in gene expression and metabolites in six typical pathways, including oxidative stress, apoptosis, autophagy, amino acid metabolism, primary bile acid biosynthesis, and lipid metabolism. Taken together, chronic MC-LR exposure induced oxidative stress, apoptosis, and autophagy, inhibited primary bile acid biosynthesis, and caused fatty deposition in the liver of Nile tilapia.

The design and fabrication of highly efficient, cheap, and stable heterogeneous catalyst for the advancement of sustainable organic transformation is a crucial goal in both academic research and industry. Herein, an efficient catalyst based on Cu-decorating on a N, P-codoped porous carbon(Cu/NPC) derived from wheat straw was successfully synthesized by a cost-effective method. The as-prepared Cu/NPC showed superior catalytic activity for the N-alkylation of amines with alcohols via the borrowing hydrogen strategy. A variety of aromatic amines were converted into the corresponding secondary amines in moderate to excellent yields. Noted that, such catalyst can be easily prepared, recovered and recycled four times without obvious loss in both activity and selectivity.

ATP-binding cassette (ABC) transporters are believed to protect aquatic organisms by pumping xenobiotics out, and recent investigation has suggested their involvement in the detoxification and efflux of nanoparticles (NPs), but their roles in fish embryos are poorly understood. In this regard, this paper summarizes the recent advances in research pertaining to the development of ABC transporter-mediated multi-xenobiotic resistance (MXR) mechanism in fish embryos and the potential interaction between ABC transporters and NPs. The paper focuses on: (1) Expression, function, and modulation mechanism of ABC proteins in fish embryos; (2) Potential interaction between ABC transporters and NPs in cell models and fish embryos. ABC transporters could be maternally transferred to fish embryos and thus play an important role in the detoxification of various chemical pollutants and NPs. There is a need to understand the specific mechanism to benefit the protection of aquatic resources.

Solid-phase denitrification shows promise for removing nitrate (NO3−-N) from water. Biological denitrification uses external carbon sources to remove nitrogen from wastewater, among which agriculture waste is considered the most promising source due to its economic and efficiency advantages. Hydraulic retention time (HRT) and influent nitrate concentration (INC) are the main factors influencing biological denitrification. This study explored the effects of HRT and INC on solid-phase denitrification using wheat husk (WH) as a carbon source. A solid-phase denitrification system with WH carbon source was constructed to explore denitrification performance with differing HRT and INC. The optimal HRT and INC of the wheat husk-denitrification reactor (WH-DR) were 32 h and 50 mg/L, respectively. Under these conditions, NO3−-N and total nitrogen removal rates were 97.37 ± 2.68% and 94.08 ± 4.01%, respectively. High-throughput sequencing revealed that the dominant phyla in the WH-DR operation were Proteobacteria, Bacteroidetes, and Campilobacterota. Among the dominant genera, Diaphorobacter (0.85%), Ideonella (0.38%), Thiobacillus (4.22%), and Sulfurifustis (0.60%) have denitrification functions; Spirochaeta (0.47%) is mainly involved in the degradation of WH; and Acidovorax (0.37%) and Azospira (0.86%) can both denitrify and degrade WH. This study determined the optimal HRT and INC for WH-DR and provides a reference for the development and application of WH as a novel, slow-release carbon source in treating aquaculture wastewater.

Faba bean water extract (FBW) and vitamin K3 (VK3) have been demonstrated to improve the muscle textural quality of fish. To better apply these two feed additives in commercial aquaculture setting, four experimental diets (control, commercial feed group; 15% FBW, 15% faba bean water extract group; 2.5% VK3, 2.5% vitamin K3 group; combined group, 15% faba bean water extract + 2.5% vitamin K3 group) were formulated to explore their combined effects of FBW and VK3 on the growth, health status, and muscle textural quality of grass carp. The growth performance, textural quality, intestinal characteristics, and oxidative and immune responses were analyzed on days 40, 80 and 120. The results showed that supplementation with higher doses of FBW and VK3 have no influence on growth-related parameters and immune parameters of grass carp. Notably, compared with the control, fish in the combined group had the highest textural qualities (hardness, chewiness and adhesiveness), followed by those in 15% FBW and 2.5% VK3 groups (P < 0.05). Also, FBW and VK3, to some extent, may lower antioxidative ability of grass carp, as illustrated by lower levels of GSH and CAT in 15% FBW, 2.5% VK3, and combined groups on day 120 (P < 0.05). In addition, enhanced lipase activity was observed in the 15% FBW group. Taken together, the combined supplementation of FBW and VK3 was demonstrated to be a more advanced option than their individual supplementation in a commercial setting owing to the resulting combined effects on both the textural quality and health status of grass carp.

With the ability to produce components with complex and precise structures, additive manufacturing or 3D printing techniques are now widely applied in both industry and consumer markets. The emergence of tissue engineering has facilitated the application of 3D printing in the field of biomedical implants. 3D printed implants with proper structural design can not only eliminate the stress shielding effect but also improve in vivo biocompatibility and functionality. By combining medical images derived from technologies such as X-ray scanning, CT, MRI, or ultrasonic scanning, 3D printing can be used to create patient-specific implants with almost the same anatomical structures as the injured tissues. Numerous clinical trials have already been conducted with customized implants. However, the limited availability of raw materials for printing and a lack of guidance from related regulations or laws may impede the development of 3D printing in medical implants. This review provides information on the current state of 3D printing techniques in orthopedic implant applications. The current challenges and future perspectives are also included.

Abstract Enantioselective recognition and separation are the most important issues in the fields of chemistry, pharmacy, agrochemical, and food science. Here, we developed two optically active diamines showing aggregation‐induced emission (AIE) that can discriminate 5 kinds of chiral acids with high enantioselectivity. Especially, a very high fluorescence intensity ratio (Il/Id) of 281 for (±)‐Dibenzoyl‐d/l‐tartaric acid was obtained through the collection of fluorescence change after interaction with chiral AIE‐active diamine. By virtue of AIE property and intermolecular acid‐base interaction, enantioselective separation was facilely realized by simple filtration of the precipitates formed by chiral AIE luminogen (AIEgen) and one enantiomer in the racemic solution. The chiral HPLC data indicated that the precipitates of AIEgen/chiral acid possessed 82% l‐analyte (the enantiomeric excess value was assessed to be 64% ee). Therefore, this method can serve as a simple, convenient, and low‐cost tool for chiral detection and separation.

The complete replacement of fresh baits (FB) by formulated artificial feeds (AF) after domestication in mandarin fish (Siniperca chuatsi) has matured in recent years, while the underlying molecular events of long-term AF diet feeding after diet shifts remained unclear. Therefore, mandarin fish, which had been successfully domesticated, were fed with the FB or AF for 180 days to explore the molecular mechanisms underlying the differences due to shifted food habit. Compared to the FB group, slight intestinal histological damage was found in the AF group of mandarin fish. An integrated analysis of miRNA-mRNA sequencing and their interaction networks was conducted between the FB and AF groups. The result showed that a total of 209 differentially expressed mRNAs (DEmRNAs) and 2 differentially expressed miRNAs (DEmiRNAs) with significance were identified. The DEmRNAs were enriched to 39 pathways including tight junction, steroid biosynthesis and protein processing in ER. Oni-Let-7a and aca-Let-7i were the two significantly identified DEmiRNAs, whose target prediction was enriched with GO terms and KEGG pathways including TGFβ signaling pathway, steroid biosynthesis, and retinol metabolism. In general, our research was the first exploration to characterize the interacted miRNA-mRNA sequencing of mandarin fish fed the FB and AF diets, which could reveal the mechanisms of long-term adaptation to new foods in organisms who subjected to a food conversion in their early life. Microbial communities in aquaculture ponds play a critical role in pond productivity and the success of fish production. Nevertheless, our current understanding of the functional diversity of bacterial microbiomes in grass carp pond water and sediments is fragmentary. This study utilized high-throughput sequencing technology and community level physiological profiling (CLPP) methods to investigate microbial communities and metabolic activity of bacterial found in water samples and sediments from eight typical grass carp (Ctenopharyngodon idella) culturing pond in southern China. The correlations between putative function using PICRUSt2 and actual metabolic profiles via Biolog EcoPlates was assessed. Carbohydrates (32.51 %), amino acids (23.13 %), and polymers (16.61 %) were the most dominant carbon sources utilized by the microbial community in the sediment; while carbohydrates (34.72 %), amino acids (25.49 %), and polymers (16.15 %) were also utilized most often in the water samples. In the sediment samples, Proteobacteria (24.67 %), Chloroflexi (15.30 %), and Firmicutes (13.74 %) were the most dominant bacteria phyla. Actinobacteriota (32.62 %), Cyanobacteria (25.06 %), and Proteobacteria (17.51 %) were the most dominant bacteria phyla in the water samples. Moreover, our results suggested that the nitrogen transformations bacterial genera (Bacillus, Bradyrhizobium, Brocadia, Halomonas, and Vogesella) were positively correlated with carbon source utilization of the microbial community in the Biolog EcoPlates in the sediment. Dechloromonas and Pseudomonas were significantly negatively correlated in the water samples. In addition, there was a significant correlation between predicted function and the actual metabolic characteristics of microbial communities in sediment and water samples. The results facilitate detailed perspective of the relationship between the microbial community structure and the function in grass carp ponds and also provide a reference for the selection of suitable carbon sources in aquaculture wastewater treatment technology.

CSFL caused by spinal dural defect is a common complication of spinal surgery, which need repair such as suture or sealants. However, low intracranial pressure symptoms, wound infection and prolonged hospital associated with pin-hole leakage or loose seal effect were often occurred after surgical suture or sealants repair. Stable, pressure resistance and high viscosity spinal dural repair patch in wet environment without suture or sealants was highly needed. Herein, a bioactive patch composed of alginate and polyacrylamide hydrogel matrix cross-linked by calcium ions, and chitosan adhesive was proposed. This fabricated patch exhibits the capabilities of promoting defect closure and good tight seal ability with the bursting pressure is more than 790 mm H2O in wet environment. In addition, the chitosan adhesive layer of the patch could inhibit the growth of bacterial in vitro, which is meaningful for the postoperative infection. Furthermore, the patch also significantly reduced the expression of GFAP, IBA-1, MBP, TNF-α, and COX-2 in early postoperative period in vivo study, exerting the effects of anti-inflammatory, analgesic and adhesion prevention. Thus, the bioactive patch expected to be applied in spinal dural repair with the good properties of withstanding high pressure, promoting defect closure and inhibiting postoperative infection.

​The zero-water exchange aquaculture has been identified as a promising method of farming to decrease the environment pressure of aquaculture and to increase profits. However, the ecological functions (e.g., nitrogen removal) and microbial biodiversity of the zero-water exchange pond aquaculture system are relatively understudied. In the present study, the zero-water exchange pond aquaculture system were constructed to investigated the microbial communities, sediment potential nitrification and denitrification production rates. And five functional genes (AOA amoA, AOB amoA, nirS, nosZ, and hzsB) were used to quantify the abundance of nitrifying and denitrifying microorganisms. The results showed that the sediment of the system had excellent potential nitrification-denitrification performance, with potential nitrification and denitrification rate were 149.77-1024.44 ng N g−1 h−1 and 48.32-145.01 ng N g−1 h−1, respectively. The absolute copy numbers of nitrogen functional genes and total bacterial 16S rRNA were 1.59×105-1.39×109 and 1.55×1010-2.55×1010copies g−1, respectively, with the dominant phyla, i.e., Proteobacteria, Actinobacteriota, Chloroflexi, Cyanobacteria, and Firmicutes. The relative abundances of the genera related to nitrification and denitrification, varied from 0.01% to 0.79% and from 0.01% to 15.54%, respectively. The potential nitrification rate was positively related to the sediment TOC concentration; and the potential denitrification rate had a positive correlation with sediment nitrate concentration. The genera Bacillus positively correlated with sediment NO3‐-N concentration, whereas Flavobacterium and Shewanella positively correlated with sediment NH4+-N concentration, which could be the functional bacteria for nitrogen removal. These findings may shed light on quantitative molecular mechanisms for nitrogen removal in zero-water exchange ponds, providing a sustainable solution to nitrogen pollution problem in the freshwater aquaculture ecosystems.

To investigate the impact of betaine on high-carbohydrate-diet-induced oxidative stress and endoplasmic reticulum (ER) stress, mandarin fish (Siniperca chuatsi) (23.73 ± 0.05 g) were fed with control (NC), betaine (BET), high carbohydrate (HC), and high carbohydrate + betaine (HC + BET) diets for 8 weeks. The results showed that betaine significantly promoted the growth of mandarin fish irrespective of the dietary carbohydrate levels. The HC diet induced oxidative stress, as evidenced by significantly elevated MDA levels. The HC diet significantly stimulated the mRNA levels of genes involved in ER stress (ire1, perk, atf6, xbp1, eif2α, atf4, chop), autophagy (ulk1, becn1, lc3b), and apoptosis (bax). However, betaine mitigated HC-diet-induced oxidative stress by modulating antioxidant enzymes and alleviated ER stress by regulating the mRNA of genes in the PERK-eIF2a-ATF4 pathway. Additionally, betaine significantly reduced the mRNA levels of becn1 and bax, along with the apoptosis rate, indicating a mitigating effect on autophagy and apoptosis. Overall, dietary betaine improved growth, attenuated HC-diet-induced oxidative stress and ER stress, and ultimately alleviated apoptosis in mandarin fish. These findings provide evidence for the use of betaine in aquafeeds to counter disruptive effects due to diets containing high carbohydrate levels.

With the advent of the information epoch and the development of Big Data, users are constantly overwhelmed in massive information online. As an effective method to deal with this dilemma of information overload, recommendation system has become a popular area of research for the past few years. However, most of the current recommendation models fail to make the most of the ample resources hidden behind auxiliary data and user reviews. For this reason, we put forward the FHRec model. We try to combine heterogeneous information network with deep learning technology to improve recommendation performance. The working mechanism is to represent the rich auxiliary data by using heterogeneous information network, learning the features of entities through network embedding; in the meantime, by adopting deep learning technology to mine the features of entities from review texts, using the attention mechanism to fuse feature vectors of entities, inputting them into the neural network for recommendation; the last step is that we use Yelp dataset and Douban movie dataset to verify the FHRec model. The experimental results show that the FHRec behaves better than the traditional comparison algorithm on the Yelp dataset and Douban movie dataset.

BackgroundDiabetes is one of the most common metabolic diseases and continues to be a leading cause of death worldwide. The NLRP3 inflammasome has been shown to exert detrimental effects on diabetic models. However, evidence linking NLRP3 inflammasome and pre-diabetes has been scarcely explored. Herein, we aimed to determine whether the NLRP3 inflammasome correlates with insulin resistance and liver pathology in a cohort of pre-diabetic subjects.Methods50 pre-diabetic subjects were randomly assigned to a Pre-diabetes Control (DC, n=25) and a Pre-diabetes exercise (DEx, n=25) group. 25 Normal subjects (NC) were selected as controls. The DEx group performed a 6-month combined Yijingjing and resistance training intervention, while DC and NC group remained daily routines. Clinical metabolic parameters were determined with an automatic biochemistry analyzer; inflammatory cytokines were quantified by the ELISA assay; the protein expressions of NLRP3 inflammasome components in PBMCs were evaluated by Western Blot.ResultsThe insulin resistance, liver injury and NLRP3 inflammasome activity were higher in pre-diabetic individuals than in normal control group. However, 6-month exercise intervention counteracted this trend, significantly improved insulin sensitivity, reduced liver injury and inhibited the overactivation of NLRP3 inflammasome in pre-diabetic subjects. Moreover, positive correlations between insulin resistance, liver pathology and NLRP3 inflammasome were also found.ConclusionsOur study suggests that exercise training is an effective strategy to alleviate insulin resistance and liver injury in elderly pre-diabetic subjects which is probably associated with the inhibition of NLRP3 inflammasome activity.

Autonomous vehicles (AVs) have the potential to improve safety, traffic capacity, and energy efficiency, but these advantages can only be realized when the AV market penetration is sufficiently high. To promote the adoption of AVs, it would be crucial for the government to take policy measures. This paper develops a two-stage model to explore the effects of subsidy and AV lanes’ policies on AV adoption. In the first stage, given the subsidy policy, the vehicle manufacturer sets AV price to pursue maximum profit while anticipating the choice of consumers for AVs or conventional vehicles (CVs), from which the AV market penetration can be accessed. Subsequently, based on the AV market penetration acquired from the first stage model, an optimization model integrating the mixed traffic assignment is developed in the second stage to determine the time-dependent progressive AV lanes’ deployment plan. The first and second stage models are solved using the simulation-optimization and genetic-algorithm-based approaches, respectively. Due to the mutual influence of the two models, an iterative optimization approach is applied to solve the whole model. Two numerical experiments are conducted, and the results demonstrate the positive effects of subsidy and AV lane policies on increasing AV market penetration. The analysis provides significant managerial insights for policymakers to promote the development of AVs.

Plants have the potency to regenerate adventitious roots from aerial organs after detachment. In Arabidopsis thaliana, de novo root regeneration (DNRR) from leaf explants is triggered by wounding signaling that rapidly induces the expression of the ETHYLENE RESPONSE FACTOR (ERF) transcription factors ERF109 and ABR1 (ERF111). In turn, the ERFs promote the expression of ASA1, an essential enzyme of auxin biosynthesis, which contributes to rooting by providing high levels of auxin near the wounding side of the leaf. Here, we show that the loss of the epigenetic regulator ULTRAPETALA1 (ULT1), which interacts with Polycomb and Trithorax Group proteins, accelerates and reinforces adventitious root formation. Expression of ERF109 and ASA1 was increased in ult1 mutants, whereas ABR1 was not significantly changed. Cultivation of explants on media with exogenous auxin equates adventitious root formation in wild-type with ult1 mutants, suggesting that ULT1 negatively regulates DNRR by suppressing auxin biosynthesis. Based on these findings, we propose that ULT1 is involved in a novel mechanism that prevents overproliferation of adventitious roots during DNRR.

Aiming at the problem that the effect of the existing fault line selection methods is mainly determined by the fault features constructed by manual extraction, and the fault feature extraction process is complex and time-consuming, a new method based on convolutional neural network (CNN) is proposed. Firstly, the fault voltage and current signal data are collected, and the time-frequency energy matrix of the fault signal is constructed by the HHT band-pass filtering method, which is regarded as the two-dimensional matrix form of input data of CNN. Then the time-frequency energy matrix is input into the CNN, and the fault features are extracted autonomously through the convolution layer and pooling layer of the network, which is used to train the network to realize fault line selection and fault phase judgment. Finally, the results show that the method not only has a high accuracy of fault line selection but also can complete the fault line selection and fault phase judgment at the same time without adjusting any parameters, which realize the shared weights of the two non-independent problems. Meanwhile, under the influence of noise interference, compensation degree, network structure change, and other factors, the proposed method has good robustness. However, compared with the FCM, SVM, DNN, and DBN, CNN can still identify the faulty line and keep the optimal accuracy in the case of two-point grounding fault, and other methods have errors inline selection or low accuracy. Therefore, the experimental results also provide a new idea for fault line selection of distribution networks.

Growth and fat deposition are complex traits, which can affect economical income in the pig industry. Due to the intensive artificial selection, a significant genetic improvement has been observed for growth and fat deposition in pigs. Here, we first investigated genomic-wide association studies (GWAS) and population genomics (e.g., selection signature) to explore the genetic basis of such complex traits in two Large White pig lines (n = 3,727) with the GeneSeek GGP Porcine HD array (n = 50,915 SNPs). Ten genetic variants were identified to be associated with growth and fatness traits in two Large White pig lines from different genetic backgrounds by performing both within-population GWAS and cross-population GWAS analyses. These ten significant loci represented eight candidate genes, i.e., NRG4, BATF3, IRS2, ANO1, ANO9, RNF152, KCNQ5, and EYA2. One of them, ANO1 gene was simultaneously identified for both two lines in BF100 trait. Compared to single-population GWAS, cross-population GWAS was less effective for identifying SNPs with population-specific effect, but more powerful for detecting SNPs with population-shared effects. We further detected genomic regions specifically selected in each of two populations, but did not observe a significant enrichment for the heritability of growth and backfat traits in such regions. In summary, the candidate genes will provide an insight into the understanding of the genetic architecture of growth-related traits and backfat thickness, and may have a potential use in the genomic breeding programs in pigs.

Crisp grass carp (CGC), the most representative improved varieties of grass carp (GC), features higher muscle hardness after feeding faba bean (Vicia faba L.) for 90–120 days. DNA methylation, a most widely studied epigenetic modification, plays an essential role in muscle development. Previous studies have identified numerous differentially expressed genes (DEGs) between CGC and GC. However, it remains unknown if the expression levels of these DEGs are influenced by DNA methylation. In the present study, we performed a comprehensive analysis of DNA methylation profiles between CGC and GC, and identified important candidate genes related to muscle development coupled with the transcriptome sequencing data. A total of 9,318 differentially methylated genes (DMGs) corresponding to 155,760 differentially methylated regions (DMRs) were identified between the two groups under the CG context in promoter regions. Combined with the transcriptome sequencing data, 14 key genes related to muscle development were identified, eight of which (gsk3b, wnt8a, wnt11, axin2, stat1, stat2, jak2, hsp90) were involved in muscle fiber hyperplasia, six of which (tgf-β1, col1a1, col1a2, col1a3, col4a1, col18a1) were associated with collagen synthesis in crisp grass carp. The difference of methylation levels in the key genes might lead to the expression difference, further resulting in the increase of muscle hardness in crisp grass carp. Overall, this study can help further understand how faba bean modulates muscle development by the epigenetic modifications, providing novel insights into the texture quality improvement in other aquaculture fish species by nutritional programming.

Selenium (Se)-enriched tea is a well-regarded natural beverage that is often consumed for its Se supplementation benefits. However, the production of this tea, particularly in Se-abundant tea plantations, is challenging due to soil acidification. Therefore, this study aimed to investigate the effects of changes in Se under acidified soil conditions. Eight tea plantation soil monitoring sites in Southern Jiangsu were first selected. Simulated acid rain experiments and experiments with different acidification methods were designed and soil pH, as well as various Al-ion and Se-ion concentrations were systematically determined. The data were analyzed using R statistical software, and a correlation analysis was carried out. The results indicated that as the pH value dropped, exchangeable selenium (Exc-Se) and residual selenium (Res-Se) were transformed into acid-soluble selenium (Fmo-Se) and manganese oxide selenium (Om-Se). As the pH increased, exchange state aluminum (Alex) and water-soluble aluminum (Alw) decreased, Fmo-Se and Om-Se declined, and Exc-Se and Res-Se increased, a phenomenon attributed to the weakened substitution of Se ions by Al ions. In the simulated acid rain experiment, P1 compared to the control (CK), the pH value of the YJW tea plantation decreased by 0.13, Exc-Se decreased by 4 ug mg−1, Res-Se decreased by 54.65 ug kg−1, Fmo-Se increased by 2.78 ug mg−1, and Om-Se increased by 5.94 ug mg−1 while Alex increased by 28.53 mg kg−1. The decrease in pH led to an increase in the content of Alex and Alw, which further resulted in the conversion of Exc-Se to Fmo-Se and Om-Se. In various acidification experiments, compared with CK, the pH value of T6 decreased by 0.23, Exc-Se content decreased by 8.35 ug kg−1, Res-Se content decreased by 40.62 ug kg−1, and Fmo-Se content increased by 15.52 ug kg−1 while Alex increased by 33.67 mg kg−1, Alw increased by 1.7 mg kg−1, and Alh decreased by 573.89 mg kg−1. Acidification can trigger the conversion of Exc-Se to Fmo-Se and Om-Se, while the content of available Se may decrease due to the complexation interplay between Alex and Exc-Se. This study provides a theoretical basis for solving the problem of Se-enriched in tea caused by soil acidification.

Abstract Background The therapeutic potential of exosomes derived from stem cells has attracted increasing interest recently, because they can exert similar paracrine functions of stem cells and overcome the limitations of stem cells transplantation. Exosomes derived from bone mesenchymal stem cells (BMSC-Exos) have been confirmed to promote osteogenesis and angiogenesis. The magnetic nanoparticles (eg. Fe3O4, γ-Fe2O3) combined with a static magnetic field (SMF) has been commonly used to increase wound healing and bone regeneration. Hence, this study aims to evaluate whether exosomes derived from BMSCs preconditioned with a low dose of Fe3O4 nanoparticles with or without the SMF, exert superior pro-osteogenic and pro-angiogenic activities in bone regeneration and the underlying mechanisms involved. Methods Two novel types of exosomes derived from preconditioned BMSCs that fabricated by regulating the contents with the stimulation of magnetic nanoparticles and/or a SMF. Then, the new exosomes were isolated by ultracentrifugation and characterized. Afterwards, we conducted in vitro experiments in which we measured osteogenic differentiation, cell proliferation, cell migration, and tube formation, then established an in vivo critical-sized calvarial defect rat model. The miRNA expression profiles were compared among the exosomes to detect the potential mechanism of improving osteogenesis and angiogenesis. At last, the function of exosomal miRNA during bone regeneration was confirmed by utilizing a series of gain- and loss-of-function experiments in vitro. Results 50 µg/mL Fe3O4 nanoparticles and a 100 mT SMF were chosen as the optimum magnetic conditions to fabricate two new exosomes, named BMSC-Fe3O4-Exos and BMSC-Fe3O4-SMF-Exos. They were both confirmed to enhance osteogenesis and angiogenesis in vitro and in vivo compared with BMSC-Exos, and BMSC-Fe3O4-SMF-Exos had the most marked effect. The promotion effect was found to be related to the highly riched miR-1260a in BMSC-Fe3O4-SMF-Exos. Furthermore, miR-1260a was verified to enhance osteogenesis and angiogenesis through inhibition of HDAC7 and COL4A2, respectively. Conclusion These results suggest that low doses of Fe3O4 nanoparticles combined with a SMF trigger exosomes to exert enhanced osteogenesis and angiogenesis and that targeting of HDAC7 and COL4A2 by exosomal miR-1260a plays a crucial role in this process. This work could provide a new protocol to promote bone regeneration for tissue engineering in the future. Graphical abstract

As a typical microscopic imaging technology, the emergence of the microscope has accelerated the pace of human exploration of the micro world. With the development of science and technology, microscopes have developed from the optical microscopes at the time of their invention to electron microscopes and even atomic force microscopes. The resolution has steadily improved, allowing humans to expand the field of research from the initial animal and plant tissues to microorganisms such as bacteria, and even down to the nanolevel. The microscope is now widely used in life science, material science, geological research, and other fields. It can be said that the development of microscopes also promotes the development of micro- and nanotechnology. It is foreseeable that microscopes will play a significant part in the exploration of the microworld for a long time to come. The development of microscope technology is the focus of this study, which summarized the properties of numerous microscopes and discussed their applications in micro and nanotechnology. At the same time, the application of microscopic imaging technology in micro- and nanofields was investigated based on the properties of various microscopes.

The pregnane X receptor (PXR) is a master xenobiotic-sensing receptor in response to toxic byproducts, as well as a key regulator in intermediary lipid metabolism. Therefore, the present study was conducted to investigate the potential role of PXR in mediating the lipid dysregulation and xenobiotic responses under Cu-induced stress in largemouth bass (Micropterus salmoides). Four groups of largemouth bass (52.66 ± 0.03 g) were treated with control, Cu waterborne (9.44 μmol/L), Cu+RIF (Rifampicin, 100 mg/kg, PXR activator), and Cu+KET (Ketoconazole, 20 mg/kg, PXR inhibitor) for 48 h. Results showed that Cu exposure significantly elevated the plasma stress indicators and triggered antioxidant systems to counteract Cu-induced oxidative stress. Acute Cu exposure caused liver steatosis, as indicated by the significantly higher levels of plasma triglycerides (TG), lipid droplets, and mRNA levels of lipogenesis genes in the liver. Liver injuries were detected, as shown by hepatocyte vacuolization and severe apoptotic signals after Cu exposure. Importantly, Cu exposure significantly stimulated mRNA levels of PXR, suggesting the response of this regulator in the xenobiotic response. The pharmacological intervention of PXR by the agonist and antagonist significantly altered hepatic mRNA levels of PXR, implying that RIF and KET were effective agents of PXR in largemouth bass. Administration of RIF significantly exacerbated liver steatosis, and such alterations were dependent on the regulations on pparγ and cd36 rather than srebp1 signaling, which suggested that PXR-PPARγ might be another pathway for Cu-induced lipid deposition in fish. Whereas, KET administration showed reverse effects on lipid metabolism as indicated by the lower hepatic TG levels, suppressed mRNA levels of pparγ and cd36. Activation of PXR stimulated autophagy and inhibited apoptosis, leading to lower hepatic vacuolization; while inhibition of PXR showed higher apoptotic signals, inhibition of autophagic genes and stimulation of apoptotic genes. Taken together, PXR played a cytoprotective role in Cu-induced hepatotoxicity through regulations on autophagy and apoptosis. Overall, our data has demonstrated for the first time on the dual roles of PXR as a co-regulator in mediating xenobiotic responses and lipid metabolism in fish, which implying the potential of PXR as a therapy target for xenobiotics-induced lipid dysregulation and hepatotoxicity.

Graft healing after anterior cruciate ligament reconstruction (ACLR) involves slow biological processes, and various types of biological modulations have been explored to promote tendon-to-bone integration. Exosomes have been extensively studied as a promising new cell-free strategy for tissue regeneration, but few studies have reported their potential in tendon-to-bone healing. In this study, a novel type of exosome derived from magnetically actuated (iron oxide nanoparticles (IONPs) combined with a magnetic field) bone mesenchymal stem cells (BMSCs) (IONP-Exos) was developed, and the primary purpose of this study was to determine whether IONP-Exos exert more significant effects on tendon-to-bone healing than normal BMSC-derived exosomes (BMSC-Exos). Here, we isolated and characterized the two types of exosomes, conducted in vitro experiments to measure their effects on fibroblasts (NIH3T3), and performed in vivo experiments to compare the effects on tendon-to-bone integration. Moreover, functional exploration of exosomal miRNAs was further performed by utilizing a series of gain- and loss-of-function experiments. Experimental results showed that both BMSC-Exos and IONP-Exos could be shuttled intercellularly into NIH3T3 fibroblasts and enhanced fibroblast activity, including proliferation, migration, and fibrogenesis. In vivo, we found that IONP-Exos significantly prevented peri-tunnel bone loss, promoted more osseous ingrowth into the tendon graft, increased fibrocartilage formation at the tendon-bone tunnel interface, and induced a higher maximum load to failure than BMSC-Exos. Furthermore, overexpression of miR-21-5p remarkably enhanced fibrogenesis in vitro, and SMAD7 was shown to be involved in the promotive effect of IONP-Exos on tendon-to-bone healing. Our findings may provide new insights into the regulatory roles of IONPs in IONP-Exos communication via stimulating exosomal miR-21-5p secretion and the SMAD7 signaling pathway in the fibrogenic process of tendon-to-bone integration. This work could provide a new strategy to promote tendon-to-bone healing for tissue engineering in the future.

Micro-Nano optics is one of the most active frontiers in the current development of optics. It combines the cutting-edge achievements of photonics and nanotechnology, which can realize many brand-new functions on the basis of local electromagnetic interactions and become an indispensable key science and technology of the 21st century. Micro-Nano optics is also an important development direction of the new optoelectronics industry at present. It plays an irreplaceable role in optical communication, optical interconnection, optical storage, sensing imaging, sensing measurement, display, solid-state lighting, biomedicine, security, green energy, and other fields. In this paper, we will summarize the research status of micro-nano optics, and analyze it from four aspects: micro-nano luminescent materials and devices, micro-nano optical waveguide materials and devices, micro-nano photoelectric detection materials and devices, and micro-nano optical structures and devices. Finally, the future development of micro-nano optics will be prospected.

Abstract Background and objectives Long non-coding RNA (lncRNA) prostate androgen-regulated transcript 1 (PART1) was previously shown to exert an oncogenic role in several human cancers. However, whether PART1 is associated with the malignant progression of pancreatic cancer remains unclear. In the current study, we aimed to identify the role and potential mechanism of PART1 in pancreatic cancer. Methods qRT-PCR was applied to detect PART1 expression in 45 cases of pancreatic cancer patients. The chi-square test was performed to assess the association between PART1 expression and clinicopathologic features, and Kaplan-Meier method was applied to evaluate overall survival. In vitro CCK-8, transwell invasion, and flow cytometry assays were applied to detect the effects of PART1 on cell proliferation, invasion, and apoptosis, respectively. Luciferase reporter and RNA immunoprecipitation assays were used to identify the regulatory mechanism between PART1 and miR-122. Results PART1 expression was upregulated in pancreatic cancer tissues and cell lines. High PART1 expression was closely correlated with tumor size, T classification, clinical stage, and vascular invasion, and predicted a poor overall survival. PART1 knockdown significantly suppressed cell proliferation and invasion abilities of pancreatic cancer but promoted cell apoptosis. PART1 was found to serve as a molecular sponge of miR-122, and miR-122 inhibition partially reversed the inhibitory phenotypes of PART1 knockdown on pancreatic cancer cells. Conclusions PART1 promotes the malignant progression of pancreatic cancer by sponging miR-122. The PART1/miR-122 axis might be a promising target for anticancer therapy in patients with pancreatic cancer.

Ricin is a highly toxic ribosome-inactivating lectin derived from castor beans. To date, no antidote is available to treat ricin-poisoned patients, and the development of a safe and effective antidote is urgently needed. First, ricin was prepared and used to construct a mouse model and a rhesus monkey model of ricin intoxication. Second, pepsin-digested F(ab′) 2 fragments of serum IgG from horses injected with Freund’s-adjuvanted purified ricin were prepared. Third, the protective efficacy was evaluated in mouse and rhesus monkey models of lethal ricin intoxication. The purity quotient of the prepared ricin and F(ab′) 2 fragments exceeded 90% and 85% in the mouse and monkey models, respectively. The LD 50 of ricin in mice and rhesus monkeys was 2.7 and 9 μg/kg, respectively. A quantity of 6.25 and 1.85 mg/kg F(ab′) 2 was sufficient to treat lethal ricin intoxication in the mice and rhesus monkeys, respectively. Finally, the effect of this therapeutic antibody on peripheral blood immune cells was examined by analysis of peripheral blood immune cells through single cell sequencing. The underlying mechanism was found to involve restraining neutrophil activation, proliferation, and differentiation. Purified F(ab′) 2 fragments administered with needle-free devices fully protect mice and rhesus monkeys against lethal doses of ricin intoxication.

In-stent restenosis caused by tumor ingrowth increases the risk of secondary surgery for patients with abdominal aortic aneurysms (AAA) because conventional vascular stent grafts suffer from mechanical fatigue, thrombosis, and endothelial hyperplasia. For that, we report a woven vascular stent-graft with robust mechanical properties, biocompatibility, and drug delivery functions to inhibit thrombosis and the growth of AAA. Paclitaxel (PTX)/metformin (MET)-loaded silk fibroin (SF) microspheres were self-assembly synthesized by emulsification-precipitation technology and layer-by-layer coated on the surface of a woven stent via electrostatic bonding. The woven vascular stent-graft before and after coating drug-loaded membranes were characterized and analyzed systematically. The results show that small-sized drug-loaded microspheres increased the specific surface area and promoted the dissolution/release of drugs. The stent-grafts with drug-loaded membranes exhibited a slow drug-release profile more for than 70 h and low water permeability at 158.33 ± 17.56 mL/cm2·min. The combination of PTX and MET inhibited the growth of human umbilical vein endothelial cells. Therefore, it was possible to generate dual-drug-loaded woven vascular stent-grafts to achieve the more effective treatment of AAA.
