Objective To analyze the structure of CD36, search the possible B cell epitopes and prepare multi-antigen peptides(MAP) with B cell epitopes, so as to provide a preliminary experimental basis for the preparation of CD36 antibodies using MAP with B cell epitopes. Methods The potential B cell epitopes of CD36 were analyzed by bioinformatics methods, including physical and chemical properties, secondary structure, potential phosphorylation and glycosylation sites. Eight-branch MAP with CD36 B cell epitopes were synthesized by FMOC using polylysine as the core matrix. The purity of MAPs was analyzed by reverse high-performance liquid chromatography chromatography(RP-HPLC), and the molecular weight of MAPs was determined by mass spectrometry. Results CD36 is a stable and hydrophilic alkaline protein, with multiple phosphorylation and glycosylation sites and strong antigenicity, and its secondary structure is mainly characterized by irregular curls. Four potential B cell epitopes were obtained and 4 MAPs containing potential B cell epitopes were prepared. RP-HPLC analysis showed that the purity of the MAPs were above 85%, and the molecular weight of 3 MAPs was consistent with the expected theoretical molecular weight. Conclusion CD36 on platelet has strong antigenicity. MAPs containing CD36 B cell epitopes can provide the experimental basis for the preparation and related research of CD36 antibodies.

Abstract Background Even with advances in primary health care, depressive disorders remain a major global public health problem. We conducted an in-depth analysis of global, regional and national trends in depressive disorders incidence over the past 30 years. Methods Data on the incidence of depressive disorders were obtained by sex (female, male, and both), location (204 countries), age (5–84 years), year (1990–2019) from the Global Burden of Disease Study (GBD) 2019. Further, age-period-cohort modeling was used to estimate the net drift, local drift, age, period and cohort effects between 1990 and 2019. Results In 2019, although the incidence of depressive disorders has increased by 59.3% to 290 million (95% UI: 256, 328), the age-standardized incidence rate has decreased by 2.35% to 3588.25 per 100,000 people (3152.71, 4060.42) compared to 1990. There was an emerging transition of incidences from the young and middle-aged population to the old population. From 1990 to 2019, the net drift of incidence rate ranged from −0.54% (−0.61%, −0.47%) in low-middle Socio-demographic Index (SDI) regions to 0.52% (0.25%, 0.79%) in high SDI regions. Globally, the incidence rate of depressive disorders increases with age, period effects showing a decreasing risk and cohort effects beginning to decline after the 1960s. Conclusions Our current findings reflect substantial health disparities and potential priority-setting of depressive disorders incidence in the three dimensions of age, period and cohort across SDI regions, countries. The scope of healthcare to improve the progression of depressive disorders events can be expanded to include males, females of all ages.

To investigate the changes in the quality of yak meat at different dry aging times (7, 14, 21, 28 and 35 d), in this study, sensory evaluation, texture analysis, and GC-IMS were employed to investigate the changes in texture and flavor of yak meat during different dry aging periods. Additionally, PLS-DA was utilized to identify the characteristic flavor compounds. To investigate the effects of Angel yeast, Meyerozyma guilliermondii, Saccharomyces cerevisiae, Zygosaccharomyces rouxii and Candida parapsilosis fermentation on volatile flavour compounds in bean paste, the volatile flavour substances of fermented bean soup paste were analyzed by gas chromatography-ion mobility spectrometry (GC-IMS) technique using unfermented bean soup paste as a control group. The relative odour activity value (ROAV) method was used to determine the key flavour components. Principal component analysis (PCA) and partial least squares-discrimination analysis (PLS-DA) were used to comprehensively evaluate the aroma of fermented bean soup paste. A total of 50 volatile compounds were identified, which were primarily esters and alcohols in six fermented bean sauces. These compounds contribute to the distinct mellow, fruity, and malty flavour profiles characteristic of these sauces. A kind of Agriophyllum squarrosum bread (SMMB) was developed with Agriophyllum squarrosum flour and wheat flour as main raw materials. In this study, the formula of SMMB was optimized by single factor tests and response surface test. Then the texture, color and flavor of SMMB under optimized condition were compared with traditional bread (BMB) by using texture analyzer, colorimeter and gas chromatography-ionmobility spectrometry (GC-IMS). The results showed that when the proportion of Agriophyllum squarrosum flour in the mixed powder was 19.1%, and based on the total amount of mixed powder, adding of 2.5% yeast, 14.5% sugar, 10% butter, 1% edible salt, 0.5% ameliorant as well as 50% water, the SMMB was golden in color, full in shape, exquisite and uniform in internal organization, and had a unique flavor of Agriophyllum squarrosum. The SMMB also had the highest sensory score of 86.96 under the optimized condition. Compared with BMB, the color of SMMB was obviously different with decreased lightness, but increased redness and yellowness. The hardness, cohesion and elasticity were also increased in SMMB. The main flavor substances of the two types of bread were similar, but the contents of alcohols, aldehydes and ketones in SMMB were relatively higher, while the contents of esters and heterocyclic substances were lower than that of BMB. The different relative contents of flavor substances endowed SMMB with higher mellow, citrus, grass and malty flavor, while the flavor of ester, nutty and toasty was lower. This project would provide a theoretical basis for the application of Agriophyllum squarrosum in bread.

Cooking methods could have a great influence on the flavour of food, to investigate the effect of cooking methods on the aroma of pork, this study used electronic nose combined with gas chromatography-ion mobility spectrometry (GC-IMS) for multivariate statistical analyses of the volatile compounds in pork under different cooking methods. GC-IMS was used to characterise 100 volatile compounds (including monomers and dimers) of pork under different cooking methods. Finally, a stable model was established using orthogonal partial least squares discriminant analysis. Results showed that, the cooking method is crucial for the formation of pork flavour compounds. Pork cooked by oven roasting and air fryer methods contained characteristic volatile compounds such as ethyl (E)-2-hexenoate, 1-octen-3-ol, (Z)-4-decenal, 3-nonen-2-one, (E)-2-hexen-1-ol, propyl butyrate, furazanone, and dipentenyl-M, as compared to the two cooking methods of steaming and boiling. The air fryer method pork contained higher levels of (E)-ethyl 2-hexenoate, 1-octen-3-ol, (Z)-4-decenal, and 3-nonen-2-one than the oven-roasted pork. Valeraldehyde and ethyl acetate were labelled as characteristic volatile substance compounds for both steamed and boiled cooking styles, and the results from the electronic nose sensor showed that the RPCA model was effective in identifying volatile substances in pork for each cooking style. The results of the study revealed the effects of different cooking methods on the volatile components of pork and found that the aromas of air fryer preparation and oven roasting were more similar, providing some reference for the cooking and processing of pork.

Additive manufacturing (AM) presents promising prospects for the production of laser ceramics, particularly those with composite structures. However, achieving uniform density within these ceramics is a formidable challenge, primarily due to the intrinsic limitations of uneven forces within the mold. This paper introduces the Density Spatiotemporal Evolution (DSE) model, a novel predictive tool designed to map the density distribution in laser ceramics throughout the AM process. The DSE model elucidates that ceramic density increases in proximity to the indenter and mold wall and reveals an oscillatory pattern in particle density during pressing. These insights have been corroborated through experimental measurements and simulation analyses. Leveraging the DSE model and a Simulated Annealing (SA) algorithm, an innovative AM process is proposed. The pressure exerted to each layer of ceramics is gradient distributed, i.e. 18, 18.36, 19.31, 22.08, 33.02 MPa. Preliminary experimental results indicate a significant enhancement in density uniformity, with an improvement of up to 45% in the optimized ceramics. The advancement promises to yield more homogeneous ceramic materials, potentially accelerating the progress of high-energy laser technology.

Abstract Background IgG4-related diseases are very uncommon, and its diagnosis and treatment are complicated as it encompasses multiple disciplines. Case presentation : A 77-year-old woman was admitted with a jaw mass and nausea and vomiting. Laboratory tests showed elevated serum IgG4, pituitary MRI suggested thickening of the pituitary stalk, and head and neck CT suggested orbital and mandibular masses. Patients with mandibular mass were diagnosed with Mikulicz’s disease with IgG4-related hypophysitis. We found no other evidence of causing thickening of the pituitary stalk. She was given oral prednisolone 30 mg daily, and her nausea and vomiting improved significantly, and the mandibular and ocular masses decreased in size. Conclusion Mikulicz’s disease combined with IgG4-related hypophysitis is a rare case of IgG4-RD in elderly women. IgG4-RD is one of the causes of head and neck exocrine gland mass and pituitary stalk thickening in the elderly.

Abstract Objective To establish the population pharmacokinetics (PPK) of magnesium sulfate (MgSO4)in women with preeclampsia (PE), and to determine the key covariates having an effect in magnesium pharmacokinetics in Chinese PE. Methods Pregnant women with PE prescribed MgSO4 were enrolled in this prospective study from April 2021 to April 2023. On the initial day of administration, the patients were administered a loading dose of 5 g in conjunction with 10 g of magnesium sulfate as a maintenance dose. On the second day, only the maintenance dose was administration, and maternal blood samples were taken at 0, 4, 5, and 12 h after the second day’s 10 g maintenance dose. The software Phoenix was used to estimate PPK parameters of MgSO4, such as clearance (CL) and volume of distribution (V), and to model PPK models with patient demographic, clinical, and laboratory covariates. Results A total of 199 blood samples were collected from 51 women with PE and PPK profiles were analyzed. The PPK of MgSO4 is consistent with to a one-compartment model. The base model adequately described the maternal serum magnesium concentrations after magnesium administration. The population parameter estimates were as follows: CL was 2.98 L/h, V was 25.07 L. The model predictions changed significantly with covariates (BMI, creatinine clearance, and furosemide). Furosemide statistically influences V. The creatinine clearance, BMI and furosemide jointly affects CL. Monte Carlo simulation results showed that a loading dose combined with a maintenance dose would need to be administered daily to achieve the therapeutic blood magnesium concentrations. For the non-furosemide group, the optimal dosing regimen was a 5 g loading dose combined with a 10 g maintenance dose of MgSO4. For the furosemide group, the optimal dosing regimen was a 2.5 g loading dose combined with a 10 g maintenance dose of MgSO4. Conclusions The magnesium PPK model was successfully developed and evaluated in Chinese preeclampsia population, and the dose optimization of MgSO4 was completed through Monte Carlo simulation.

Abstract To explore the molecular pathogenesis of pulmonary arterial hypertension (PAH) and identify potential therapeutic targets, we performed transcriptome sequencing of lung tissue from mice with hypoxia-induced pulmonary hypertension. Our Gene Ontology analysis revealed that “extracellular matrix organization” ranked high in the biological process category, and matrix metallopeptidases (MMPs) and other proteases also played important roles in it. Moreover, compared with those in the normoxia group, we confirmed that MMP s expression was upregulated in the hypoxia group, while the hub gene Timp1 was downregulated. Crocin, a natural MMP inhibitor, was found to reduce inflammation, decrease MMPs levels, increase Timp1 expression levels, and attenuate hypoxia-induced pulmonary hypertension in mice. In addition, analysis of the cell distribution of MMPs and Timp1 in the human lung cell atlas using single-cell RNAseq datasets revealed that MMPs and Timp1 are mainly expressed in a population of fibroblasts. Moreover, in vitro experiments revealed that crocin significantly inhibited myofibroblast proliferation, migration, and extracellular matrix deposition. Furthermore, we demonstrated that crocin inhibited TGF-β1-induced fibroblast activation and regulated the pulmonary arterial fibroblast MMP2/TIMP1 balance by inhibiting the TGF-β1/Smad3 signaling pathway. In summary, our results indicate that crocin attenuates hypoxia-induced pulmonary hypertension in mice by inhibiting TGF-β1-induced myofibroblast activation.

Abstract It is recognized as a promising therapeutic strategy for cocaine use disorder to develop an efficient enzyme which can rapidly convert cocaine to physiologically inactive metabolites. We have designed and discovered a series of highly efficient cocaine hydrolases, including CocH5-Fc(M6) which is the currently known as the most efficient cocaine hydrolase with both the highest catalytic activity against (−)-cocaine and the longest biological half-life in rats. In the present study, we characterized the time courses of protein appearance, pH, structural integrity, and catalytic activity against cocaine in vitro and in vivo of a CocH5-Fc(M6) bulk drug substance produced in a bioreactor for its in vitro and in vivo stability after long-time storage under various temperatures (− 80, − 20, 4, 25, or 37 °C). Specifically, all the tested properties of the CocH5-Fc(M6) protein did not significantly change after the protein was stored at any of four temperatures including − 80, − 20, 4, and 25 °C for ~ 18 months. In comparison, at 37 °C, the protein was less stable, with a half-life of ~ 82 days for cocaine hydrolysis activity. Additionally, the in vivo studies further confirmed the linear elimination PK profile of CocH5-Fc(M6) with an elimination half-life of ~ 9 days. All the in vitro and in vivo data on the efficacy and stability of CocH5-Fc(M6) have consistently demonstrated that CocH5-Fc(M6) has the desired in vitro and in vivo stability as a promising therapeutic candidate for treatment of cocaine use disorder.

Mixed matrix membranes (MMMs) could combine the advantages of both polymeric membranes and porous fillers, making them an effective alternative to conventional polymer membranes. However, interfacial incompatibility issues, such as the presence of interfacial voids, hardening of polymer chains, and blockage of micropores by polymers between common MMMs fillers and the polymer matrix, currently limit the gas separation performance of MMMs. Ternary phase MMMs (consisting of a filler, an additive, and a matrix) made by adding a third compound, usually functionalized additives, can overcome the structural problems of binary phase MMMs and positively impact membrane separation performance. This review introduces the structure and fabrication processes for ternary MMMs, categorizes various nanofillers and the third component, and summarizes and analyzes in detail the CO2 separation performance of newly developed ternary MMMs based on both rubbery and glassy polymers. Based on this separation data, the challenges of ternary MMMs are also discussed. Finally, future directions for ternary MMMs are proposed.

Integrated photonic devices are essential for on-chip optical communication, optical-electronic systems, and quantum information sciences. To develop a high-fidelity interface between photonics in various frequency domains without disturbing their quantum properties, nonlinear frequency conversion, typically steered with the quadratic (χ2) process, should be considered. Furthermore, another degree of freedom in steering the spatial modes during the χ2 process, with unprecedent mode intensity is proposed here by modulating the lithium niobate (LN) waveguide-based inter-mode quasi-phase-matching conditions with both temperature and wavelength parameters. Under high incident light intensities (25 and 27.8 dBm for the pump and the signal lights, respectively), mode conversion at the sum-frequency wavelength with sufficient high output power (−7 – 8 dBm) among the TM01, TM10, and TM00 modes is realized automatically with characterized broad temperature (ΔT ≥ 8 °C) and wavelength windows (Δλ ≥ 1 nm), avoiding the previous efforts in carefully preparing the signal or pump modes. The results prove that high-intensity spatial modes can be prepared at arbitrary transparent wavelength of the χ2 media toward on-chip integration, which facilitates the development of chip-based communication and quantum information systems because spatial correlations can be applied to generate hyperentangled states and provide additional robustness in quantum error correction with the extended Hilbert space.

Abstract Background Avian influenza viruses (AIVs) constitute significant zoonotic pathogens encompassing a broad spectrum of subtypes. Notably, the H4 subtype of AIVs has a pronounced ability to shift hosts. The escalating prevalence of the H4 subtype heightens the concern for its zoonotic potential, signaling an urgent need for vigilance. Methods During the period from December 2021 to November 2023, we collected AIV-related environmental samples and assessed them using a comprehensive protocol that included nucleic acid testing, gene sequencing, isolation culture, and resequencing. Results In this study, a total of 934 environmental samples were assessed, revealing a remarkably high detection rate (43.66%, 289/662) of AIV in the live poultry market. Notably, the H4N1 subtype AIV (cs2301) was isolated from the live poultry market and its complete genome sequence was successfully determined. Subsequent analysis revealed that cs2301, resulting from a reassortment event between wild and domesticated waterfowl, exhibits multiple mutations and demonstrates potential for host transfer. Conclusions Our research once again demonstrates the significant role of wild and domesticated waterfowl in the reassortment process of avian influenza virus, enriching the research on the H4 subtype of AIV, and emphasizing the importance of proactive monitoring the environment related to avian influenza virus.

Background: Nano titanium dioxides (TiO2) are widely used in drug development, food additives and packaging materials. Although several studies have demonstrated the poisonousness of TiO2 in vivo and in vitro, the underlying molecular mechanisms have not been fully revealed. Methods: Characterization of TiO2 by FTIR, XRD, TEM and DLS. The NCM460 cell line, representing normal colon epithelial cells, was utilized as a model to assess the impact of TiO2 nanoparticles (TiO2-NPs) on cell proliferation and apoptosis. The potential molecular mechanisms underlying its toxic effects were investigated through transcriptome analysis, RT-qPCR, and western blot experiments. Results: The particle size of the TiO2-NPs used is about 25 nm, which has typical characteristics of anatase. TiO2-NPs at a concentration of 30–60 μg/mL will cause changes in colon cell morphology, decreased proliferation ability, and increased number of apoptotic cells. TiO2-NPs at a concentration of 6 μg/mL did not significantly modify the transcriptome expression profile of colon cells; while 30 μg/mL had a significant effect, leading to up-regulation of gene expression. The differentially expressed genes predominantly modulate the MAPK signaling pathway, TNF signaling pathway, cytokine-cytokine receptor interaction, and other related pathways. Further, western blot analysis revealed that higher concentrations of TiO2-NPs (30–60 μg/mL) could up-regulate the expression of P53, P21 and Bax, while down-regulating the expression of Bcl2 by regulating the MAPK (ERK, P38) signaling pathway. Simultaneously, it also promoted the decreased in Fos protein expression and inhibited the phosphorylation of Jun and Fos. Conclusion: This study demonstrates that TiO2-NPs may exert potential toxic effects on colon cells, and therefore the intake of TiO2-NPs should be strictly regulated in practical applications.

In order to investigate the differences of volatile compounds in sprouted garlic, an electronic nose and trap headspace-gas chromatography-mass spectrometry (HS-Trap-GC-MS) combined with orthogonal partial least squares discriminant analysis (OPLS-DA), aroma activity value, differential heat map, and correlation analysis were employed to analyze the differences in volatile compounds of garlic sprouts at 0, 24, 48, 72 and 96 hours. A predictive model with a prediction accuracy of 96.00% was established by combining the electronic nose with OPLS-DA. GC-MS analysis revealed that sulfur compounds were the major common volatile compounds in garlic sprouts at different sprouting stages. The relative content of sulfur compounds showed a decreasing trend with the prolongation of sprouting time, while the variety exhibited an increasing trend. Allyl disulfide was the compound that experienced the most significant decrease in content during the sprouting process. Allyl tetrasulfide and allyl mercaptan were identified as common key compounds in the samples. Differential heat map analysis demonstrated that in addition to differences in the content of common substances, the absence of propyl sulfide, hexanal, di-tert-butyl dicarbonate, propenol, 6-methyl-2-heptyne, 5-methylthiadiazole, 2-ethy-1,3-dithiane, 2-prop-2-ynylsulfonylpropane, 2,5-dimethylthiophene, 2,5-dimethylfuran, 1-hexen-3-ol, and 1,3-dithiane further amplified the differences in odor between non-sprouted and sprouted garlic. The types of main common volatile compounds in sprouted garlic showed an increasing trend with the extension of sprouting time. There was a significant correlation between the major volatile compounds of garlic and most sensors of the electronic nose. The odor intensity of garlic gradually decreased with the prolongation of sprouting time.

Abstract Background Bamboo charcoal has garnered wide recognition as an adsorbent owing to its stability and availability. Phosphoric acid modification contributes to forming specific functional groups on the surface of bamboo charcoal, culminating in the augmentation of its adsorption capabilities. Results The modification process in this study involved impregnating the bamboo charcoal in a 45% mass fraction of phosphoric acid solution with a 1:10 ratio (bamboo charcoal powder to phosphoric acid), followed by activation at a temperature of 500 °C for a duration of 1.5 h. The modification resulted in the formation of hydroxyl, carboxyl, and phosphate groups on the surface of the bamboo charcoal. Furthermore, the specific surface area of the modified bamboo charcoal surged from 4.91 m2/g to 115.18 m2/g, the average aperture decreased from 7.15 nm to 1.99 nm, and the pore volume escalated by 6.5 times. On this basis, a comparison in the adsorption capability for chlorogenic acid, a natural small-molecule active ingredient, was drawn before and after the modification of bamboo charcoal. The results indicate that the modified bamboo charcoal showcases an impressive 46.8-fold increase in its adsorption capacity when juxtaposed with the unmodified bamboo charcoal. Through the calculations based on density functional theory (DFT), the addition of functional groups on the surface of the modified bamboo charcoal module allows for the formation of hydrogen bonds with the chlorogenic acid module, with the highest binding energy reaching − 57.59 kJ/mol. Conclusion As evidenced by the adsorption kinetic studies, the curve of modified bamboo charcoal for chlorogenic acid abides by a first-order kinetic equation. Additionally, adsorption thermodynamic studies reveal that the curve of modified bamboo charcoal for chlorogenic acid conforms to the Boltzmann equation. These results shed light on the mechanism of selectively adsorbing specific constituents using phosphoric acid-modified bamboo charcoal, thereby providing a solid theoretical foundation for the application of bamboo charcoal. BackgroundThe coordinative relationship between medical treatment and the economy is an important part of promoting the sustainable and healthy development of a social economy, and it is conducive to promoting the coordinated development of the two systems.MethodsBased on constructing a comprehensive evaluation index system for the relationship between traditional Chinese medicine medical services and economic development, the entropy method, coupling coordination model, and Tobit model are used to calculate the comprehensive development level, coupling coordination degree, and influencing factors of the two systems in 31 provinces of China from 2015 to 2021. Suggestions are proposed for the coordinated development of traditional Chinese medicine medical services, and economy.ResultsThe results showed that the development level of traditional Chinese medicine was higher than that of social and economic development, the relative development gap between the two systems was gradually narrowed, and the coupling coordination degree increased steadily; however, the regional differences were large. The relative development level and coupling coordination degree of each region show a development pattern of eastern > central > western > northeast; the coupling coordination degree of the two systems in the spatial region was positively correlated and was enhanced year by year. From the perspective of influencing factors, the level of economic development is the decisive factor in the coupling and coordination between traditional Chinese medicine medical treatment and the social economy. Factors such as human capital and health level in traditional Chinese medicine have an inhibitory effect on the coordinated development of the two systems.ConclusionSo, China should focus on optimizing the regional allocation of traditional Chinese medicine medical personnel, improving the quality of medical services, and enhancing the health level of medical personnel.

Liver cancer is a heterogeneous disease characterized by poor responses to standard therapies and therefore unfavourable clinical outcomes. Understanding the characteristics of liver cancer and developing novel therapeutic strategies are imperative. Ferroptosis, a type of programmed cell death induced by lipid peroxidation, has emerged as a potential target for treatment. Naringenin, a natural compound that modulates lipid metabolism by targeting AMPK, shows promise in enhancing the efficacy of ferroptosis inducers. In this study, we utilized liver cancer cell lines and xenograft mice to explore the synergistic effects of naringenin in combination with ferroptosis inducers, examining both phenotypic outcomes and molecular mechanisms. Our study results indicate that the use of naringenin at non-toxic doses to hepatocytes can significantly enhance the anticancer effects of ferroptosis inducers (erastin, RSL3, and sorafenib). The combination index method confirmed a synergistic effect between naringenin and ferroptosis inducers. In comparison to naringenin or ferroptosis inducers alone, the combined therapy caused more robust lipid peroxidation and hence more severe ferroptotic damage to cancer cells. The inhibition of aerobic glycolysis mediated by the AMPK-PGC1α signalling axis is the key to naringenin's effect on reducing ferroptosis resistance in liver cancer, and the synergistic cytotoxic effect of naringenin and ferroptosis inducers on cancer cells was reversed after pretreatment with an AMPK inhibitor or a PGC1α inhibitor. Taken together, these findings suggest that naringenin could boost cancer cell sensitivity to ferroptosis inducers, which has potential clinical translational value.

BackgroundThe coordination of health service supply and regional economy is an integral path to promote China’s prosperity.MethodsBased on the coupling mechanism of health service supply and regional economy, we sampled the data from 30 provinces in China from 2009 to 2021 in this study and constructed the evaluation index system. Additionally, we calculated the coupling coordination degree (HED) of the two through the coupling coordination degree model. We further used the kernel density estimation, Moran’s I index, and spatial β convergence model to assess the dynamic evolution trends, spatial aggregation effect, and spatial convergence characteristics of coupling coordination.Conclusion(1) HED in China showed a rising trend during the study period but with large regional differences, forming a gradient distribution pattern of “high in the east and low in the west.” (2) The results of Kernel density estimation show that HED has formed a gradient differentiation phenomenon within each region in China. (3) HED has modeled spatial clustering characteristics during the study period, with high-value clusters mainly appearing in the eastern region and low-value clusters appearing in the northwestern region. (4) There are absolute β-convergence and conditional β-convergence trends in HED in China and the three major regions during the study period, but there is an obvious regional heterogeneity in the control factors. The research provides a reference for accurately implementing policies according to different levels of health service supply and economic development, in addition to narrowing the regional differences of the coupling coordination between the regional economy and health service supply.

To mitigate the continued impact of SARS-CoV-2, influenza A, and influenza B viruses on human health, a smartphone-based point-of-care testing (POCT) system was designed to detect respiratory pathogens through a nucleic acid test. This compact, light-weight, highly automated, and universal system enables the differential diagnosis of SARS-CoV-2, influenza A, and influenza B in approximately 30 min in a single-tube reaction. Numerous hospitals and disease control and prevention center assessed the triple POCT system’s detection threshold, sensitivity, specificity, and stability, and have concluded that all the assessments were comparable to those of fluorescent quantitative polymerase chain reaction (PCR)-based testing. The triple POCT system is suitable as an onsite rapid-diagnosis device, as well as for pathogen screening at airports and customs.

Objective As an integral part of emergency medical rescue teams during public health events, understanding the core competencies that nursing personnel should possess—including theoretical knowledge, practical skills, comprehensive abilities and personal traits—can provide a practical basis for better preparation and targeted training for future emergency rescue works. Thus, this study aims to provide a scientific and applicable reference for perfecting the routine training strategy of nursing personnel assembled by emergency medical rescue teams and improving the overall guarantee ability level of this group.Design This is a qualitative study conducted using individual semi-structured interviews. All interviews were recorded and transcribed verbatim for the purpose of thematic analysis and extraction.Setting Participants were recruited from February to March 2023, from four comprehensive hospitals in Chongqing China with the highest number of emergency relief works.Participants A sample of experts (N=15) with extensive experience in emergency relief works was recruited in Chongqing, China.Results 60% of the experts held master’s degrees or higher, 73.3% held senior or higher titles, 36.7% had participated in work execution more than five times and 73.3% held leadership positions in their current units and in the execution of emergency relief works. Four main themes and 22 corresponding subthemes were derived for the core competencies required for nursing personnel selected for emergency medical rescue teams in public health events, including theoretical knowledge, practical skills, comprehensive abilities and personal traits.Conclusions Our study revealed that through interviews with 15 experts with extensive experience in the public health event, the essential elements of core competencies for nursing personnel assigned to emergency medical rescue teams during the public health event were identified. These can serve as a reference standard for the selection of nursing personnel in public health events, and provide a basis for the cultivation and evaluation of competency for nursing personnel assigned to emergency medical rescue teams in the public health event in China and globally.

Plant glycoside hydrolase family 9 genes (GH9s) are widely distributed in plants and involved in a variety of cellular and physiological processes. In the current study, nine GH9 genes were identified in the mulberry and were divided into two subfamilies based on the phylogenetic analysis. Conserved motifs and gene structure analysis suggested that the evolution of the two subfamilies is relatively conserved and the glycoside hydrolase domain almost occupy the entire coding region of the GH9s gene. Only segmental duplication has played a role in the expansion of gene family. Collinearity analysis showed that mulberry GH9s had the closest relationship with poplar GH9s. MaGH9B1, MaGH9B6, MaGH9B5, and MaGH9B3 were detected to have transcript accumulation in the stalk of easy-to drop mature fruit drop, suggesting that these could play a role in mulberry fruit drop. Multiple cis-acting elements related to plant hormones and abiotic stress responses were found in the mulberry GH9 promoter regions and showed different activities under exogenous abscisic acid (ABA) and 2,4- dichlorophenoxyacetic acid (2,4-D) stresses. We found that the lignin content in the fruit stalk decreased with the formation of the abscission zone (AZ), which could indirectly reflect the formation process of the AZ. These results provide a theoretical basis for further research on the role of GH9s in mulberry abscission.

AbstractBackground Phosphatidylinositol 3-kinase (PI3K) and protein kinase B (AKT) are involved in the clearance of Mycobacterium tuberculosis (MTB) by macrophages.Aim This study aimed to investigate the effects of polymorphisms in the PI3K/AKT genes and the gene-smoking interaction on susceptibility to TB.Methods This case-control study used stratified sampling to randomly select 503 TB patients and 494 control subjects. Logistic regression analysis was used to determine the association between the polymorphisms and TB. Simultaneously, the marginal structure linear dominance model was used to estimate the gene-smoking interaction.Results Genotypes GA (OR 1.562), AA (OR 2.282), and GA + AA (OR 1.650) at rs3730089 of the PI3KR1 gene were significantly associated with the risk to develop TB. Genotypes AG (OR 1.460), GG (OR 2.785), and AG + GG (OR 1.622) at rs1130233 of the AKT1 gene were significantly associated with the risk to develop TB. BackgroundThe association of caesarean section (CS) for postpartum depression (PPD) remains controversial. This study aims to explore the relationship between CS and the risk of PPD, in order to provide a basis for preventing PPD.Material and methodsWe searched PubMed, Web of Science, Cochrane Library, and EMBASE databases for literature about the correlation between CS and PPD published as of February 2024. The combined odds ratios (ORs) and 95% confidence intervals (Cls) were obtained by flexible use of fixed-effects models or random-effects models.ResultsA total of 18 publications were ultimately included in the analysis. Among these, 14 were cohort studies and 4 were case-control reports, encompassing 844,328 total cases. All of the included studies were deemed to be of moderate or higher quality. The meta-analysis indicated that the prevalence of PPD among parturients undergoing CS was 13.4% (95% CI: 6.5%-25.5%).The adjusted odds ratio (OR) for the association between CS and PPD was 1.12 (95% CI: 1.04-1.20) compared to the natural vaginal delivery (NVD) group. Specifically, the adjusted OR for the association between CS and PPD was 1.29 (95% CI: 1.18-1.40) during the first 1-6 months postpartum, and 1.23 (95% CI: 1.14-1.33) after 6 months postpartum. Furthermore, in comparison to the NVD group, the adjusted OR for elective caesarean section (ELCS) and emergency caesarean section (EMCS) were 0.96 (0.83, 1.10) and 1.20 (1.08, 1.34), respectively.ConclusionOur findings suggest that PPD risk may rise in the presence of CS. In particular, the risk was increased by 20% in the EMCS group, and the risk of PPD within one to six months postpartum after CS increases by 6% compared to that at six months postpartum. For China's urban industrial layout and green innovation, determining which types of producer service agglomeration externalities (PSAE) are most conducive to the rise of green innovation efficiency (GIE) has theoretical and practical relevance. Based on the panel data of 274 prefecture-level cities in China from 2006 to 2019, this paper adopted the super-SBM model with unexpected output to evaluate the urban GIE. It used the Moran I index to judge the spatial auto-correlation of GIE, followed by the spatial Durbin model (SDM) to examine the spatial spillover effect of PSAE on the urban GIE. Then we used a comprehensive threshold effects model to investigate the non-linear relationship. The results demonstrate that Marshall-Arrow-Romer externality (MARE) and Jacobs externality (JACE) are curbing the urban GIE. Porter externality (PORTERE) inhibits the region and promotes the surrounding area. Furthermore, these effects also depend on the stage of economic development.

Today, population aging is the main trend of population development. Home-based care is mainly adopted in Chinese society, and scholars have paid ample attention to the effect of intergenerational support on the mental health of older people. However, research conclusions differ. This study uses data from the 2018 China Health and Pension Tracking Survey (CHARLS), which we analyzed with STATA software to construct least squares regression and two-stage least squares regression models. The regression model included 6,647 respondents to investigate the mental health status of older people based on depression status. Intergenerational support was defined as economic support, emotional support, and daily care provided by the children of older people. We studied the impact of three aspects of intergenerational support on the mental health of the elderly. We performed a robustness test using the variable replacement and propensity score matching methods, and analyzed age, gender, and urban-rural heterogeneity. The results showed that economic support had no significant impact on the mental health of older people, while emotional support and daily care had a positive effect. The heterogeneity results indicated that the relationship between intergenerational support and mental health of older people differed significantly based on age, gender, and urban and rural areas. Therefore, children should raise their awareness of supporting their parents, pay attention to their parents' mental health, and provide emotional support and daily care. Furthermore, community work improves family relations, creates a good social environment, and encourages young people to respect and be filial to older people. The government should improve the medical security system and old-age service system, and provide policy support to help the mental health of older people.

Background. Antihypertensive therapy is crucial for preventing stroke in hypertensive patients. However, the efficacy of antihypertensive therapy varies across individuals, partially due to therapy-related genetic variations among individuals. We investigated the association of antihypertensive drug-related gene polymorphism with stroke in patients with hypertension. Methods. Demographic information, medication, and outcome data were obtained from a hypertensive patient management system, and a PCR fluorescence probe technique was used to detect 7 gene polymorphic loci (CYP2D6∗10, ADRB1, CYP2C9∗3, AGTR1, ACE, CYP3A5∗3, and NPPA), and these loci were compared between patients with and without stroke. Logistic regression was performed to analyze the association of these genetic variations with stroke risk in hypertensive patients while controlling for potential factors. Results. The prevalence of stroke in the hypertensive population in Changsha County of Hunan Province was 2.75%. The mutation frequencies of ADRB1 (1165G > C), CYP2D6∗10, CYP2C9∗3, AGTR1 (1166A > C), ACE (I/D), NPPA (2238T > C), and CYP3A5∗3 were 74.43%, 57.23%, 4.26%, 5.71%, 31.62%, 1.17%, and 69.58%, respectively. Univariate analysis revealed that ADRB1 polymorphism was associated with stroke (χ2 = 8.659, P C), ACE (I/D), CYP3A5∗3, and NPPA (2238T > C) polymorphisms and stroke. Conclusions. ADRB1 (1165G > C) gene polymorphism is associated with the risk of stroke in Chinese hypertensive patients. The CC genotype is correlated with a higher risk of stroke than the GC + GG genotype.

This study focuses on exploring the potential of using Long Short-Term Memory networks (LSTMs) for low-flow forecasting for the Rhine River at Lobith on a daily scale with lead times up to 46 days ahead. A novel LSTM-based model architecture is designed to leverage both historical observation and forecasted meteorological data to carry out multi-step discharge time series forecasting. The feature and target selection for this deep learning (DL) model involves evaluating the use of different spatial resolutions for meteorological forcing (basin-averaged or subbasin-averaged), the impact of incorporating past discharge observations, and the use of different target variables (discharge Q or time-differenced discharge dQ). Then, the model is trained using the ERA5 dataset as meteorological forcing, and employed for operational forecast with ECMWF seasonal forecast (SEAS5) data. The forecast results are compared to a benchmark process-based model, wflow_sbm. This study also explores the flexibility of the DL model by fine-tuning the pretrained model with limited SEAS5 dataset. Key findings from feature and target selection include: (1) opting for subbasin-averaged meteorological variables significantly improves model performance compared to a basin-averaged approach. (2) Utilizing dQ as the target variable greatly boosts short-term forecast accuracy compared to using Q, with a mean absolute error (MAE) of 25 m3 s−1 and mean absolute percentage error (MAPE) of 0.02 for the first lead time, ensuring reliability and accuracy at the onset of the forecast horizon. (3) While incorporating historical discharge improves the forecasting of Q, its impact on predicting dQ is less pronounced for short lead times. In the operational forecast with SEAS5, compared to the wflow_sbm model, the DL model exhibits skill in forecasting low flows as evidenced by Continuous Ranked Probability Skill Score (CRPSS) median values of all lead times above zero, and better accuracy in forecasting drought events within short lead times. The wflow_sbm model shows higher accuracy for longer lead times. In the exploration of fine-tuning approach, the fine-tuned model generates marginal short-term enhancements in forecasting low-flow events over a non-fine-tuned model. Overall, this study contributes to advancing the field of low-flow forecasting using deep learning approach.

Objective To establish an enzyme-linked immunosorbent assay (ELISA) method for the determination of residual human coagulation factor Ⅺ in human prothrombin complex and validate the method. Methods Human factor Ⅺ was reacted with the capture antibody coated on the microtiter plate. After appropriate washing steps, biotinylated primary antibody was bound to the captured protein. Excess primary antibody was washed away and bound antibody was reacted with horseradish peroxidase conjugated streptavidin. TMB substrate was used for color development at 450 nm. The dilution reliability, accuracy, specificity, repeatability, intermediate precision, linearity, range and durability were verified. Results The verification results showed that the accuracy and specificity of this method met the experimental requirements, with an average recovery rate of 109.2% and RSD of 6.93%. The repeatability RSD was 6.78%, and the intermediate precision RSD was 6.75%, indicating good precision. The linear regression correlation coefficient of standard curve was 0.999 9, showing good accuracy and precision within the linear range. The durability was verified by the incubation time and the validity period of reagent kit opening. The results showed that the RSD of the incubation time change was 6.62%, indicating that the incubation time of this detection method was controlled between 28 to 32 minutes, and there was no significant impact on the results. The RSD of the detection results before and after the reagent kit was opened and stored under conditions for 7 days was 3.84%, indicating that the preservation of the reagent kit according to the conditions for 7 days after opening has no effect on the FⅪ detection results. Both indicated that the method had good durability. The dilution reliability results showed that there was a "hook" effect in the detection of FⅪ residue in human prothrombin complex, which could be solved by diluting 100 to 200 times. Conclusion This method can be used for the determination of FⅪ residues of human prothrombin complex in laboratory.

Abstract Objective It is critical to early monitor and manage small-for-gestational age (SGA) infants with truly adverse outcomes not detected by conventional methods. We aimed to explore the value of diffusion-weighted imaging (DWI)-based virtual magnetic resonance elastography (vMRE) and intravoxel incoherent motion (IVIM)-based biexponential and stretched exponential parameters in predicting adverse outcomes of SGA infants. Methods Twenty SGA infants with adverse outcomes and forty without adverse outcomes were included in this prospective study. One DWI-based vMRE parameter [the stiffness value (μ diff)], five IVIM–based parameters [true diffusion coefficient (D), pseudo-diffusion coefficient (D * ), perfusion fraction (f), diffusion distribution coefficient (DDC), and diffusion heterogeneity index (Alpha)] and apparent diffusion coefficient (ADC) were calculated and compared between groups. The predictive efficiency was compared by the logistic regression analysis and receiver operating characteristic curve analysis. The relationship between the μ diff value with gestational age was also evaluated. Results The placental μ diff value was remarkably higher, and the f, DDC, and ADC values were considerably lower in the SGA infants with adverse outcomes compared with those without adverse outcomes. The μ diff and f value were predictive risk factors for SGA infants with adverse outcomes. A combined predictive model (μ diff and f) improved the predictive efficacy. Moreover, there was no statistically significant correlation between the placental stiffness value and gestational age. Conclusions Functional MRI parameters to quantify placenta elastography and microcirculation in SGA patients. This might be a useful tool to assess placental function and a vital non-invasive supplement for predicting adverse outcomes of SGA infants. Critical relevance statement This prospective study shows DWI-based virtual magnetic resonance elastography and intravoxel incoherent motion-based functional parameters to quantify placenta elastography and microcirculation in small-for-gestational-age patients, which could complement existing non-invasive methods for monitoring and predicting neonatal perinatal adverse outcome. The volatile substances among Malus toringoides (Rehd.) Hughes black tea，green tea and Kang brick tea were detected by electronic nose (E-nose), trap head space-gas chromatography-mass spectrometry (HS Trap GC-MS) and the differences of the volatile substances were analyzed by orthogonal partial least squares discriminant analysis (OPLS-DA), relative odor activity value (ROAV) and differential thermography in order to explore the odor differences of Malus toringoides (Rehd.) Hughes teas with three different processes. Result showed that E-nose could effectively distinguish three kinds of Malus toringoides (Rehd.) Hughes teas, and the prediction ability of the model established by E-nose combined with OPLS-DA was 95.70%. GC-MS analysis showed that aldehydes, esters and hydrocarbons were the main volatile substances, while alcohols were the main common volatile substances of black tea and green tea. The differences in the contents of aldehydes, esters, hydrocarbons and alcohols account for the variations in the aroma of Malus toringoides (Rehd.) Hughes teas produced by different processes. ROAV value combined with aroma type analysis showed that the volatile components of fruit aroma had a relatively large contribution to the formation of the odor of Malus toringoides (Rehd.). Isovaleraldehyde might be the key aroma substance to Malus toringoides (Rehd.) Hughes teas. Differential thermogram analysis showed that trimethoxyester, isovaleraldehyde and 2-ethylfuran were the main common volatile substances in Malus toringoides (Rehd.) Hughes teas.

Abstract Background Health literacy (HL), self-efficacy (SE), social support (SS) and fear of disease progression (FOP) are all important factors affecting health-related quality of life (HRQoL) in cancer patients. However, their synergistic effects and underlying mechanisms on HRQoL in cancer patients remain unclear. Therefore, the purpose of this study was to construct a structural equation model (SEM) to explore the underlying mechanism of factors affecting HRQoL. It is hoped that this study will provide a theoretical basis for future interventions. Methods A cross-sectional design and convenience sampling method were used to investigate cancer inpatients in two general hospitals in Chongqing and Chengdu. Data were collected using structured scales, including HL, SE, SS, FOP and HRQoL. Finally, the SEM was constructed, and P ≤ 0.05 was considered significant. Active power-assist lower limb exoskeleton robots aim to enhance wearer assistance while ensuring wearer comfort and simplifying the exoskeleton’s design and control. This study proposes an active assistance method known as Equivalent Force on Connection (EFOC). The EFOC method effectively addresses the limitations encountered in conventional Joint Torque Proportional Compensation (JTPC) approaches. These limitations include the necessity for exoskeleton robot configurations to align with human limb structures for parallel assistance at each lower limb joint, as well as the exoskeleton’s inability to contribute a greater proportion of assistance due to the excessive load on specific skeletal and muscular structures, resulting in wearer discomfort. Furthermore, the effectiveness of the EFOC method is evaluated and validated for assistance during both the stance and swing phases of single-leg movements. Finally, the proposed EFOC method is implemented on a hydraulic-driven lower limb exoskeleton robot to assist wearers in squatting, stepping, and jumping locomotion. The experimental results demonstrate that the proposed EFOC method can effectively achieve the desired assistance effect.

Objective To search compatible and suitable platelets for platelet transfusion refractoriness (PTR) patient caused by compound antibodies against HLA and CD36. Methods ELISA method was used to detect the antibody against platelet antigens and the specificity of HLA-I antibody in PTR patients. The specificity of HLA-I antibody and corresponding epitopes of patients were analyzed using MATCH IT! and HLA Matchmaker software. The HLA genotype of both donor and patient was obtained by HLA-SSO method. Compatible or suitable donor platelets for PTR patients were searched through cross-reactive group (CREG) of HLA-I and HLA epitope-matched approach (Eplet). The matching degree was identified using monoclonal antibody-specific immobilization of platelet antigens (MAIPA) and the platelet suspension immunofluores-cence test (PIFT). Finally, the transfusion effect was evaluated according to the corrected count increment (CCI). Results Compound antibodies against both CD36 and HLA-I antigens were detected in two PTR patients, and their phenotype of CD36 was conformed to be type I deficient. Through LSA testing, high-frequency of HLA -I antibodies was found in these two patients, and the panel reactive antibody in patients 1 and 2 was 56% (54/96) and 53% (51/96), respectively. According to HLA-CREG and Eplet matching strategies, one donor of grade C-matching with patient 1 and one donor of grade D-matching with patient 2 were screened from the CD36 deficiency donor bank, respectively. And the selected donors avoided the antigen of HLA-I antibody epitope. These results of MAIPA and PIFT also confirmed that no immune response was detected between the patient and the donor. And a CCI of >4.5 within 24 hour of transfusion of compatible platelets was obtained in patient 2. Conclusion For PTR patients caused by HLA and CD36 compound antibodies, a combination strategy including serological cross-matching, HLA-CREG and Eplet approach should be used to select the CD36 deficient donor platelets which evaded the antigen corresponding to HLA-I antibodies and had the compatible HLA epitopes.

In order to optimize the cooking technology of flowering broad beans, four processing parameters including soaking temperature, soaking time, frying temperature and frying time were selected for single factor and orthogonal optimization experiments, and the indexes of color difference, texture, energy and nutrition, and volatile substances were detected. The results showed that the optimal process for producing flowering broad beans were: Soaking temperature 25 ℃, soaking time 15 h, frying temperature 180 ℃ and frying time 6 min. Color difference characteristics: L* 71.05±1.22, a* 10.59±0.67, b* 29.31±1.46, texture characteristics: Crispness 20.74±1.46 N, hardness 26.67±1.33 N, nutritional label: Each 100 g of flowering broad beans contained 1869.33±0.58 kJ of energy, 18.83±0.40 g of protein, 26.93±0.21 g of fat, 33.40±0.10 g of carbohydrate. The GC-MS test results showed that there were 75 volatile substances in the two groups of samples, mainly benzene and its derivatives, alcohols, olefins, etc, including 37 in the blank group, 40 in the optimal group, and 2 kinds of volatile substances in the two groups. The characteristic substance of the blank group was p-isopropyltoluene, and the characteristic substance of the optimal group was 1-isopropenyl-3-methylbenzene. The GC-IMS test results showed that there were 57 volatile substances in the two groups of samples, mainly alcohols, esters and heterocyclic, etc, including 44 in the blank group, 42 in the optimal group, and 29 kinds of volatile substances in the two groups. After processing, the types and contents of alcohols, aldehydes and ketones decreased in varying degrees, while esters and ethers increased in varying degrees. The results of GC-IMS and GC-MS were consistent. The flowering broad beans under the optimized technology are golden in color, crisp and delicious, and rich in flavor substances.

Objective: This study was aimed at analyzing the efficacy and safety of an injectable form of chlorogenic acid (CGA) in patients with recurrent high-grade glioma after standard of care treatments, through a first-in-human, open-label, dose-escalation phase I trial. Methods: A total of 26 eligible patients were enrolled, received intramuscular CGA injections at 5 dose levels, and were followed up for 5 years. CGA was well tolerated, and the maximum tolerated dose was 5.5 mg/kg. Results: The most common treatment-related adverse events occurred at the sites of injection. No grade 3 or 4 adverse events (e.g., drug allergy) were reported for these patients except for induration at the injection sites. A clinical pharmacokinetic study showed that CGA was rapidly eliminated from the plasma, with a t1/2 of 0.95–1.27 h on day 1 and 1.19–1.39 h on day 30, and no detectable CGA was observed on days 9, 11, 13, 23, 25, 27, and 29 before CGA administration. After the first treatment cycle, 52.2% of patients (12 of 23) achieved stable disease. Long-term follow-up indicated an estimated median overall survival of 11.3 months for all 23 evaluable patients. Of the 18 patients with grade 3 glioma, the median overall survival was 9.5 months. Two patients remained alive at the cutoff day. Conclusions: This phase I study demonstrated that CGA has a favorable safety profile (with no severe toxicity), and provides preliminary clinical benefits for patients with high grade glioma relapsing after prior standard therapies, thus shedding light on the potential clinical application of CGA for recurrent grade 4 glioma.

White blood cells (WBCs) play essential roles against inflammatory disorders, bacterial infections, and cancers. Inspired by nature, WBC membrane-camouflaged nanocarriers (WBC-NCs) have been developed to mimic the “dynamic” functions of WBCs, such as transendothelial migration, adhesion to injured blood vessels, etc, which make them promising for diverse medical applications. WBC-NCs inherit the cell membrane antigens of WBCs, while still exhibiting the robust inflammation-related therapeutic potential of synthetic nanocarriers with excellent (bio)physicochemical performance. This review summarizes the proposed concept of cell membrane engineering, which utilizes physical engineering, chemical modification, and biological functionalization technologies to endow the natural cell membrane with abundant functionalities. In addition, it highlights the recent progress and applications of WBC-NCs for inflammation targeting, biological neutralization, and immune modulation. Finally, the challenges and opportunities in realizing the full potential of WBC-NCs for the manipulation of inflammation-related therapeutics are discussed.

The prevalence rate of allergic rhinitis (AR) is high worldwide. The inhalation of allergens induces AR, which is an immunoglobulin E-mediated and type 2 inflammation-driven disease. Recently, the role of neuroimmune communication in AR pathogenesis has piqued the interest of the scientific community. Various neuropeptides, such as substance P (SP), vasoactive intestinal peptide (VIP), calcitonin gene-related peptide (CGRP), nerve growth factor (NGF), and neuromedin U (NMU), released via “axon reflexes” or “central sensitization” exert regulatory effects on immune cells to elicit “neurogenic inflammation,” which contributes to nasal hyperresponsiveness (NHR) in AR. Additionally, neuropeptides can be produced in immune cells. The frequent colocalization of immune and neuronal cells at certain anatomical regions promotes the establishment of neuroimmune cell units, such as nerve-mast cells, nerve-type 2 innate lymphoid cells (ILC2s), nerve-eosinophils and nerve-basophils units. Receptors expressed both on immune cells and neurons, such as TRPV1, TRPA1, and Mas-related G protein-coupled receptor X2 (MRGPRX2) mediate AR pathogenesis. This review focused on elucidating the mechanisms underlying neuroimmune communication in AR.

BackgroundThe impact of weight loss and/or improved metabolic status on the risk of non-alcoholic fatty liver disease (NAFLD) has yet to be determined.MethodsA total of 35,322 participants without NAFLD were followed. NAFLD risk was compared between consistently metabolically healthy non-obese (MHNO) and non-MHNO who lost weight to become non-obese and/or improved their metabolic health, using Cox proportional hazards and logistic regression models.ResultsFollowing 148,186 person-years, 8,409 participants had onset NAFLD, with an incidence rate of 56.75 (95% CI: 55.57, 57.94) per 1,000 person-years. Metabolically healthy obese (MHO), metabolically unhealthy obese (MUO), and metabolically unhealthy non-obese (MUNO) at baseline were associated with increased NAFLD risk, with hazard ratios of 4.48 (95%CI:4.24, 4.73), 8.85 (95%CI:7.95, 9.84), and 10.70 (95%CI:9.73, 11.78). Weight loss and/or metabolic status improvements could significantly reduce NAFLD risk by 79.46 to 41.46%. Specifically, after weight loss from MHO to MHNO, the reduction in NAFLD risk [OR decreased from 12.01 (95%CI:9.40, 15.35) to 4.14 (95%CI:3.08, 5.57)] was greater than that of the MUNO subgroup whose metabolic status improved to MHNO [OR decreased from 5.53 (95%CI:5.15, 5.94) to 2.71 (95%CI:2.50, 3.93)]. This paper reports on a research project developed in partnership with Fáilte Isteach, an initiative which welcomes migrants and promotes integration through volunteer-led English conversation classes in local communities across Ireland. The research considers the impact of the COVID-19 pandemic on this community-based language support for adult migrants by investigating the experience of online learning in Fáilte Isteach classes. It also explores the potential of technology-enhanced approaches to learning in these informal language classes in the post-pandemic environment. This mixed-methods study involved an initial survey of Fáilte Isteach learners and tutors to understand their perspectives on online learning during the pandemic, followed by a series of training workshops for Fáilte Isteach tutors, in which they could engage with digital tools and create technology-enhanced language learning resources for use with their own classes. Findings from the analysis of the qualitative and quantitative data collected through the survey and the workshop series revealed challenges associated with online learning during the pandemic, particularly regarding access to technology, digital skills, and the lack of social interaction in the online environment. Nevertheless, both learners and tutors identified positive aspects of their online experience and expressed interest in blended approaches as a complement to face-to-face learning. The findings of this research demonstrate the value of training in relation to digital engagement, as evident in the development of technology-enhanced resources through the workshop series. They further indicate how the use of accessible digital tools may benefit adult migrant learners and volunteer tutors in community-based language classes in the post-pandemic context. They also highlight the increasing need for English language support for adult migrants in Ireland and the valuable role of informal volunteer-led classes in local integration.

Objective: The aim of this study was to investigate the feasibility and safety of no placement of urinary catheter after single-port laparoscopic surgery in patients with benign ovarian tumor. Materials and Methods: Patients with benign ovarian tumor who received ovarian cystectomy or oophorectomy via single-port laparoscopic surgery in our department were screened between July 2019 and March 2021. Patients were divided into placement of urinary catheter group or no-placement of urinary catheter group according to whether an indwelling catheter was used after single-port laparoscopic surgery, and length of hospital stay, occurrence of postoperative urinary retention, incidence of urinary tract infection and re-insertion rate of urinary catheters were compared. Results: There was no significant difference in the rate of urinary catheter re-insertion between the two groups (P = 0.431), but a higher incidence of urinary catheter re-insertion was found in the group of dwelling urinary catheter placement. Simultaneously, there were no significant differences in the rates of urinary tract infection and postoperative urinary retention (1.6% vs 0.6%; P = 0.391 and 4.3% vs 6.9%; P = 0.295, respectively) between the two groups, whereas a significant shorter length of hospital stay was observed in the non-urinary catheter group when compared to the urinary catheter group (4.61 ± 1.40 vs 5.23 ± 1.72; p

This study aimed to investigate the volatile flavor compounds and tastes of six kinds of sauced pork from the southwest and eastern coastal areas of China using gas chromatography–ion mobility spectroscopy (GC-IMS) combined with an electronic nose (E-nose) and electronic tongue (E-tongue). The results showed that the combined use of the E-nose and E-tongue could effectively identify different kinds of sauced pork. A total of 52 volatile flavor compounds were identified, with aldehydes being the main flavor compounds in sauced pork. The relative odor activity value (ROAV) showed that seven key volatile compounds, including 2-methylbutanal, 2-ethyl-3, 5-dimethylpyrazine, 3-octanone, ethyl 3-methylbutanoate, dimethyl disulfide, 2,3-butanedione, and heptane, contributed the most to the flavor of sauced pork (ROAV ≥1). Multivariate data analysis showed that 13 volatile compounds with the variable importance in projection (VIP) values > 1 could be used as flavor markers to distinguish six kinds of sauced pork. Pearson correlation analysis revealed a significant link between the E-nose sensor and alcohols, aldehydes, terpenes, esters, and hetero-cycle compounds. The results of the current study provide insights into the volatile flavor compounds and tastes of sauced pork. Additionally, intelligent sensory technologies can be a promising tool for discriminating different types of sauced pork.

Salt is a key ingredient that can both enhance the taste and extend the shelf life of fermented vegetables. However, it is important to note that excessive salt levels can have adverse effects on consumer health. This study aimed to investigate the impact of various salt additions (2%, 4%, 6%, 8%, and 10% wt/wt) on the flavor profile of fermented ciba pepper, a traditional Chinese fermented chili sauce, using gas chromatography–ion mobility spectrometry (GC-IMS) in combination with an electronic nose (E-nose). Fermented ciba pepper samples were prepared with different salt additions: 2% (LJA), 4% (LJB), 6% (LJC), 8% (LJD), and 10% (LJE) (wt/wt). The physicochemical and sensory properties of the fermented ciba pepper samples were evaluated. Sensory evaluation indicated that LJC and LJD received higher scores compared to the other groups. The total acid and amino acid nitrogen contents displayed contrasting trends with the salt additions (p < 0.05). The E-nose analysis successfully differentiated the flavor profiles of the ciba pepper samples fermented with varying salt additions. Additionally, the GC-IMS analysis identified a total of 72 volatile compounds, including 14 alcohols, 21 esters, nine aldehydes, four acids, eight ketones, three terpenes, and eight other substances. Notably, the ciba pepper samples with lower salt additions exhibited higher levels of alcohols, aldehydes, and esters. In conclusion, the addition of salt during the fermentation process significantly influenced the formation of flavor compounds in ciba pepper. This study provides valuable insights into ciba pepper fermentation with different salt additions and offers prospects for the development of low-salt fermented ciba pepper products.

Face gear transmission is the transmission mechanism composed of cylindrical gear and face gear. At present, the tooth making method of face gear is still tooth cutting, with low material utilization and low production efficiency. The metal streamlines are cut off, resulting in low fatigue strength of gear teeth. In this paper, the forming of hot rolling was numerically simulated, with which the characteristics of equivalent stress field, equivalent strain field, rolling force variation, and metal flow in the process of rolling were analyzed. Finally, a hot rolling experimental device was set up, by which the face gear test piece was rolled. Then its tooth surface precision was investigated. The research reveals that hot rolling spur face gears is technological feasible and has promising prospects in industrial applications.

Mulberry holds significant economic value. However, during the ripening stage of its fruit, the phenomenon of abscission, resulting in heavy fruit drop, can severely impact the yield. The formation of off-zone structures is a critical factor in the fruit abscission process, and this process is regulated by multiple transcription factors. One such key gene that plays a significant role in the development of the off-zone in the model plant tomato is JOINTLESS, which promotes the expression of abscission-related genes and regulates the differentiation of abscission zone tissue cells. However, there is a lack of information about fruit abscission mechanism in mulberry. Here, we analyzed the MaJOINTLESS promoter and identified the upstream regulators MaABF1 and MaABI5. These two regulators showed binding with MaJOINTLESS promoter MaABF1 (the ABA Binding Factor/ABA-Responsive Element Binding Proteins) activated the expression of MaJOINTLESS, while MaABI5 (ABSCISIC ACID-INSENSITIVE 5) inhibited the expression of MaJOINTLESS. Finally, the differentially expressed genes (DEGs) were analyzed by transcriptome sequencing to investigate the expression and synergistic relationship of endogenous genes in mulberry during abscission. GO classification and KEGG pathway enrichment analysis showed that most of the DEGs were concentrated in MAPK signaling pathway, flavonoid biosynthesis, citric acid cycle, phytohormone signaling, amino acid biosynthesis, and glycolysis. These results provide a theoretical basis for subsequent in-depth study of physiological fruit abscission in mulberry.

BackgroundThe morbidity of thyroid cancer has been increasing in the last decades all over the world. In addition to the more sensitive thyroid nodule screening technology, several social and environmental factors might represent credible candidates for this increase. They include psychological stress, lifestyle-associated risk factors, nutritional deficiencies, and environmental pollutants. Foremost, psychological stress had gained high interest as a possible promoter and a modifiable risk factor for thyroid nodules in recent years. The present study was to investigate the clinical characteristics and psychological status of the population during the peak of coronavirus disease 2019 (COVID-19) and assessed the association of psychosocial determinants and the ultrasonic characteristics of thyroid nodules.MethodsIn this cross-sectional study, 490 adult subjects who had received at least two doses of COVID-19 vaccine and were not infected with COVID-19, and did not know whether they had thyroid nodules, received thyroid color ultrasound examination and psychological questionnaire survey. Depression, anxiety, and stress were assessed using Depression Anxiety Stress Scales-21 (DASS-21). Sleep quality was rated using the Pittsburgh sleep quality index (PQSI). The characteristics of 243 subjects with thyroid nodules were described and recorded in detail by thyroid color ultrasound, and the correlations between anxiety, depression, sleep quality, clinical indicators, and thyroid nodule ultrasound characteristics were analyzed. Associations between psychological status (mutually adjusted predictors) and ultrasonic characteristics of thyroid nodules (outcome) were modeled using binary logistic regression controlling for sex, age, BMI, TSH, FT3, and FT4.ResultsDepression was positively correlated with thyroid hypoechoic nodule (OR = 3.720, 95%CI 1.615–8.570), microcalcification of thyroid nodule (OR = 3.638, 95%CI 1.476–8.966), the aspect ratio of thyroid nodule>1 (OR = 3.860, 95%CI 1.052–14.161), the unclear boundary of thyroid nodule (OR = 4.254, 95%CI 1.359–13.312), and the irregular edge of thyroid nodule (OR = 4.134, 95%CI 1.810–9.439). Anxiety was positively correlated with microcalcification of thyroid nodules (OR = 4.319, 95%CI 1.487–11.409). Stress was positively correlated with thyroid hypoechoic nodules (OR = 4.319, 95%CI 1.487–11.409), microcalcification of thyroid nodules (OR = 2.724, 95%CI 1.038–7.151), and the irregular edge of thyroid nodules (OR = 2.478, 95%CI 1.077–5.705).ConclusionThis study demonstrates that depression, anxiety, and stress were associated with the morbidity of thyroid nodules and thyroid ultrasound characteristics. During COVID-19, people’s negative emotions increased significantly compared to before. Negative emotions might be harmful to thyroid health. Therefore, during periods of high stress, strategies to prevent psychological problems should be implemented to improve thyroid health.

Abstract Introduction Sleep abnormalities are highly correlated with neurodevelopmental disorders, such as intellectual disability, attention deficit hyperactivity disorder, and autism spectrum disorders (ASD). The severity of behavioral abnormalities is correlated with the presence of sleep abnormalities. Based on previous research, we investigated that Ctnnd2 gene deletion in mice lead to ASD‐like behaviors and cognitive defects. Given the importance of sleep in individuals with ASD, this study aimed to determine the effects of chronic sleep restriction (SR) on wild‐type (WT) mice and on Ctnnd2 deletion‐induced, neurologically related phenotypes in mice. Method WT and Ctnnd2 knockout (KO) mice were both subjected to manual SR (5 h per day) for 21 consecutively days separately, then we compared neurologically related phenotypes of WT mice, WT mice subjected to SR, KO mice, and KO mice subjected to SR using a three‐chamber assay, direct social interaction test, open‐field test, Morris water maze, Golgi staining, and Western blotting. Results The effects of SR on WT and KO mice were different. After SR, social ability and cognition were impaired in both WT and KO mice. Repetitive behaviors were increased, and exploration abilities were decreased in KO mice but not in WT mice. Moreover, SR reduced the density and area of mushroom‐type dendritic spines in WT rather than KO mice. Finally, the PI3K/Akt‐mTOR pathway was found to be involved in the effects induced by SR‐impaired phenotypes in WT and KO mice. Conclusion Overall, results of the present study may have implications for the role of disrupted sleep in patients with CTNND2 gene‐related autism and the evolution of neurodevelopmental disorders.

Background At present, the avoidance of drug shortages mainly relies on expert experience. This study aimed to establish an evaluation index system for the risk of drug shortages in medical institutions in China and to apply the system to guide the graded management of drugs in short supply.Methods A two-round Delphi process was conducted to determine the indicators in the index system. The weight value of each indicator was calculated using analytic hierarchy process (AHP) methods. The data of drugs in short supply from January 1 to December 31, 2020 in Hunan province were collected and evaluated using this index system. The evaluation scores, which ranged from 0 to 100, were calculated.Results A three-level index system with four first-level indicators, 11 second-level indicators, and 36 third-level indicators was constructed by the two rounds of the Delphi process. The expert authority coefficient (Cr) of the first and second rounds of consultation were 0.88 and 0.90, respectively. The Kendall’s coefficients of concordance (Kendall’s W) for the two rounds of consultation were 0.44 and 0.50, respectively (P < .05). For the first-level indicators ‘supply stability,’ ‘causes of shortage,’ ‘medicine availability in medical institution’ and ‘pharmaceutical properties,’ the weight values were 0.3253, 0.2489, 0.2398, and 0.1860, respectively. Based on the risk evaluation score, drugs (dosage strength) at high risk of shortage included sodium thiosulfate (0.64 g), posterior pituitary lobe hormones (1 mL:6 IU), protamine sulfate (5 mL:50 mg), thrombin (500 U), urokinase (10 WU), and rotundine sulfate (2 mL:60 mg).Conclusion An indexed system for the risk assessment of drug shortages in China was established to guide the graded response to drug shortages in medical institutions and the implementation of differential management strategies to address these shortages.
