SERRATE (SE) is a core protein for microRNA (miRNA) biogenesis as well as for mRNA alternative splicing. Investigating the regulatory mechanism of SE expression is hence critical to understanding its detailed function in diverse biological processes. However, little about the control of SE expression has been clarified, especially through long noncoding RNA (lncRNA). Here, we identified an antisense intragenic lncRNA transcribed from the 3′ end of SE , named SEAIRa. SEAIRa repressed SE expression, which in turn led to serrated leaves. SEAIRa recruited plant U-box proteins PUB25/26 with unreported RNA binding ability and a ubiquitin-like protein related to ubiquitin 1 (RUB1) for H2A monoubiquitination (H2Aub) at exon 11 of SE . In addition, PUB25/26 helped cleave SEAIRa and release the 5′ domain fragment, which recruited the PRC2 complex for H3 lysine 27 trimethylation (H3K27me3) deposition at the first exon of SE . The distinct modifications of H2Aub and H3K27me3 at different sites of the SE locus cooperatively suppressed SE expression. Collectively, our results uncover an epigenetic mechanism mediated by the lncRNA SEAIRa that modulates SE expression, which is indispensable for plant growth and development.

The palm oil industry is gearing itself towards sustainability by implementing mandatory biogas capture for all palm oil mills in the effort to reduce greenhouse gases. This action also indicates that the anaerobic digestion of palm oil mill effluent (POME) will be one of the essential sources of biogas for energy generation. However, the availability of POME is limited during the low crop season due to the lower palm oil production. Concurrently, the solid wastes known as empty fruit bunches (EFB) are also produced from the oil mill. EFB has vast potential for power generation, though it is currently not being fully utilized. Therefore, this work aims to enhance the biogas production from anaerobic co-digestion (ACD) of EFB with POME by evaluating the effect of alkaline pretreatment of EFB, EFB-to-POME ratio, and temperature on the methane yield. The optimum EFB:POME ratio is 0.6:1 as the methane production of this ratio at both thermophilic and mesophilic conditions are relatively high (up to 74.02 ml CH4/g VS). Results show that ACD performs 2.36 times better than mono-digestion of POME at mesophilic conditions. The modified Gompertz model could predict the process behavior of the ACD of EFB with POME with high R2 (>0.95). Further investigation on other potential pretreatment methods and synergism analysis of co-digestion, which provides a balance between efficiency and economy could be carried out. The impact of this work will be towards a sustainable environment, which will significantly reduce the environmental impacts of palm oil production.

The rapid increase in demand for renewable energy has led to a need for more efficient and effective ways to produce biogas from palm oil mill effluent (POME), which is rich in biological and chemical oxygen demand (BOD and COD). Despite its potential as a source of biogas, POME is not always effectively utilized in biogas production due to a lack of optimization of the treatment process. This study aims to address this issue by identifying the critical parameters affecting biogas production from POME and optimizing the process for maximum biogas yield and COD removal. This study employed comparative analysis and response surface methodology to optimize the performance of palm oil mill effluent (POME)-based biogas plants in Malaysia. Historical data from three commercial POME-based biogas plants in Malaysia were analyzed to identify the most critical parameters for biogas yield and COD removal. Response surface methodology, using Box–Behnken design and Design-Expert software, was then used to optimize these parameters. Sensitivity analysis was performed to interpret the impact of parameters on biogas production, with Organic Loading Rate (OLR) found to be the most critical factor for methane yield. The results showed that the optimum conditions for maximum methane production were OLR of 1.23 kg/m3·day, inlet Total Solids (TS) of 46,370 mg/L, pH of 4.5, and temperature of 45.4 °C, resulting in a 39.6% increase in methane yield (0.335 m3 CH4/kgCODremoved) and a 1.1% increase in COD removal (93.4%).

The alarming rise of food waste all over the world due to population and economic growth must be tackled with better waste management and treatment methods. The current practice of landfilling has been scientifically proven to adversely impact environmental and societal health. A relatively new technology called hydrothermal carbonization (HTC) has the potential to solve this problem. It takes in high-moisture-content material, like food waste, and converts it into bio-coal with a heating value similar to normal coal. The present study explored the feasibility of HTC technology and bio-coal production in Malaysia. An in-depth study via desk research was conducted by implementing Porter’s five forces analysis to evaluate the feasibility of the bio-coal production project. A survey involving 215 respondents from different households that represent the average demography of Malaysia was also conducted to understand the behaviors and attitudes of different households towards food waste. The present study found that a typical Malaysian household disposes mostly of meal leftovers, with an average of 926 g of food waste per day. In addition, the 3 highest food categories that were disposed of were rice or noodles or pasta (13.0%), vegetables (12.2%) and curry and soup (10.1%). Meal leftovers such as curry and soup are high in moisture content, which is suitable for HTC. The survey on household waste provided adequate information to support the availability of a sufficient quantity of food waste in the country to sustain the raw material for the bio-coal project in Malaysia. Furthermore, a consumer survey involving seven industrial firms was conducted to determine the potential buyers of bio-coal. The responses from the industrial firms show that a bio-alternative for coal is important, and they are willing to transition to greener technologies. However, five out of seven firms stated that the main hurdle in adopting bio-coal is the high cost of production and incompatibility with existing industrial processes. Finally, interviews were conducted with key players in the industry to evaluate the adoptability of bio-coal into the wider market. The findings from the desk research and the primary research show that the outlook for bio-coal in the market is quite positive. In the long run, HTC is certainly profitable, but for immediate benefits, adequate government support and policy in favour of the use of HTC bio-coal in power plants are required.

The treatment of palm oil mill effluent (POME) has always been the topic of research in Malaysia due to the high concentration of total suspended solid (TSS) and oil and grease (O&G) ranges from 28 846 to 30 920 mg/L and 5614 to 8812 mg/L, respectively. Additionally, the release of methane gas during the treatment of POME in all of the palm oil mills has been the major contributor to greenhouse gas (GHG) effect. The main aim of this work is to evaluate the performance of the pilot-plant integrated anaerobic–aerobic bioreactor (IAAB) for POME treatment at two different phases: (a) startup and (b) long-term performance evaluation from an organic loading rate (OLR) of 10.5 to 32.5 gCOD/(L·day). Startup of the pilot-plant IAAB was successfully accomplished in 24 days with overall removal efficiencies in terms of biochemical oxygen demand (BOD), chemical oxygen demand (COD), and total suspended solid (TSS) of at least 90% at the OLR of 8.0 gCOD/(L·day). The pilot-plant IAAB achieved high BOD, COD, and TSS removal efficiencies of more than 99% for OLR up to 30.0 gCOD/(L·day) with methane yield ranges from 0.0406 LCH₄/gCODᵣₑₘₒᵥₑd to 0.1312 LCH₄/gCODᵣₑₘₒᵥₑd. The effluent quality remained stable with BOD of 16–107 mg/L with a high percent of compliance with the discharge limit of 100 mg/L. The coupling of the Grau second-order and Stover–Kincannon models (anaerobic system) with the Stover–Kincannon model (aerobic system) will completely define the pilot-plant IAAB system. This pilot-plant research will provide valuable data and experiences required to design and commission a full-scale IAAB.

Introduction: Flood syndrome is a rare and potentially fatal complication of liver cirrhosis with gross ascites. Case presentation: We present a case of Flood syndrome in a gentleman with alcoholic cirrhosis and ascites who had sudden spontaneous rupture of umbilical hernia, resulting in sudden gush of ascitic fluid from the hernia. The wound was cleaned and covered in sterile dressing and was admitted for further management. His umbilical hernia wound was closed at bedside by General Surgery team and he underwent ultrasound-guided ascitic drain insertion by Interventional Radiology. His stay was complicated by bacterial peritonitis which was treated with intravenous antibiotic. Patient eventually recovered and was discharged well. Discussion: Flood syndrome has high complication and mortality rate. Recent reported cases were reviewed, focusing on the causes of cirrhosis, complications of Flood syndrome, treatments provided and the outcomes. Conclusion: There is currently no standard guideline for the management of Flood syndrome which falls in the grey area between medical and surgical management. Proper medical management with early surgical consultation is important to reduce the morbidity and mortality for these patients.

INTRODUCTION It remains unclear which advanced airway device has better placement success and fewer adverse events in out-of-hospital cardiac arrests (OHCAs). This study aimed to evaluate the efficacy of the VBM laryngeal tube (LT) against the laryngeal mask airway (LMA) in OHCAs managed by emergency ambulances in Singapore. METHODS This was a real-world, prospective, cluster-randomised crossover study. All OHCA patients above 13 years of age who were suitable for resuscitation were randomised to receive either LT or LMA. The primary outcome was placement success. Per-protocol analysis was performed, and the association between outcomes and airway device group was compared using multivariate binomial logistic regression analysis. RESULTS Of 965 patients with OHCAs from March 2016 to January 2018, 905 met the inclusion criteria, of whom 502 (55.5%) were randomised to receive LT while 403 (44.5%) were randomised to receive LMA. Only 174 patients in the LT group actually received the device owing to noncompliance. Placement success rate for LT was lower than for LMA (adjusted odds ratio [OR] 0.52, 95% confidence interval [CI] 0.31–0.90). Complications were more likely when using LT (OR 2.82,0 95% CI 1.64–4.86). Adjusted OR for prehospital return of spontaneous circulation (ROSC) was similar in both groups. A modified intention-to-treat analysis showed similar outcomes to the per-protocol analysis between the groups. CONCLUSION LT was associated with poorer placement success and higher complication rates than LMA. The likelihood of prehospital ROSC was similar between the two groups. Familiarity bias and a low compliance rate to LT were the main limitations of this study.

Background and Purpose: Although VEGF 165 (vascular endothelial growth factor-165) is able to enhance both angiogenesis and neurogenesis, it also increases vascular permeability through the blood-brain barrier. Heparan sulfate (HS) sugars play important roles in regulating VEGF bioactivity in the pericellular compartment. Here we asked whether an affinity-purified VEGF 165 -binding HS (HS7) could augment endogenous VEGF activity during stroke recovery without affecting blood-brain barrier function. Methods: Both rat brain endothelial cell line 4 and primary rat neural progenitor cells were used to evaluate the potential angiogenic and neurogenic effects of HS7 in vitro. For in vivo experiments, male Sprague-Dawley rats were subjected to 100 minutes of transient focal cerebral ischemia, then treated after 4 days with either PBS or HS7. One week later, infarct volume, behavioral sequelae, immunohistochemical markers of angiogenesis and neural stem cell proliferation were assessed. Results: HS7 significantly enhanced VEGF 165 -mediated angiogenesis in rat brain endothelial cell line 4 brain endothelial cells, and increased the proliferation and differentiation of primary neural progenitor cells, both via the VEGFR2 (vascular endothelial growth factor receptor 2) pathway. Intracerebroventricular injection of HS7 improved neurological outcome in ischemic rats without changing infarct volumes. Immunostaining of the compromised cerebrum demonstrated increases in collagen IV/Ki67 and nestin/Ki67 after HS7 exposure, consistent with its ability to promote angiogenesis and neurogenesis, without compromising blood-brain barrier integrity. Conclusions: A VEGF-activating glycosaminoglycan sugar, by itself, is able to enhance endogenous VEGF 165 activity during the post-ischemic recovery phase of stroke.

Sediment is an endogenous pollution source, which often leads water systems to eutrophication due to the release of nutrients, especially phosphorus (P). Calcium nitrate (CN) was dosed to the water systems under different modes to control P release from the sediments in this study. A 63-day static laboratory test was conducted to explore the effects of intermittent dosing and one-time dosing modes of CN on P locking in the sediment and the concentrations of nitrogen (N) and P in waters. Results showed that 89% total phosphorus (TP) in the overlying water and 91% TP in the interstitial water of sediment were reduced in the intermittent dosing reactor, which were 4% and 13% higher than those in the one-time dosing reactor, respectively. Thus, the concentration of TP in the overlying water of the dosing reactors was both below 0.1 mg/L during the whole experiment. Meanwhile, the mean values of oxidation–reduction potential (ORP) in the sediment increased to − 110.7 ± 42.02 mV when CN was added intermittently, which were significantly higher than those of the one-time dosing reactor (− 158.3 ± 44.61 mV) and control reactor (− 320.7 ± 0.05 mV). Compared with one-time dosing mode, the intermittent dosing not only reduced the maximum concentrations of NO2−-N from 9.21 to 1.79 mg/L and NO3−-N from 92.42 to 27.58 mg/L but also shorten their retention time in the overlying water, which might depress the toxic threats to aquatic animals in water environments. Therefore, the intermittent dosing of CN could not only improve the P locking effect but also minimize the risks to aquatic animals in water environments under the premise of reasonable dosage selected. In a word, this research provided an effective operation mode for locking P with CN in the heavily polluted water bodies, which is also advantageous to avoid toxic threats to aquatic animals in water environment.

After stroke, peripheral immune cells are activated and these systemic responses may amplify brain damage, but how the injured brain sends out signals to trigger systemic inflammation remains unclear. Here we show that a brain-to-cervical lymph node (CLN) pathway is involved. In rats subjected to focal cerebral ischemia, lymphatic endothelial cells proliferate and macrophages are rapidly activated in CLNs within 24 h, in part via VEGF-C/VEGFR3 signalling. Microarray analyses of isolated lymphatic endothelium from CLNs of ischemic mice confirm the activation of transmembrane tyrosine kinase pathways. Blockade of VEGFR3 reduces lymphatic endothelial activation, decreases pro-inflammatory macrophages, and reduces brain infarction. In vitro, VEGF-C/VEGFR3 signalling in lymphatic endothelial cells enhances inflammatory responses in co-cultured macrophages. Lastly, surgical removal of CLNs in mice significantly reduces infarction after focal cerebral ischemia. These findings suggest that modulating the brain-to-CLN pathway may offer therapeutic opportunities to ameliorate systemic inflammation and brain injury after stroke., Brain damage induces systemic inflammation, but insights and implication of this induction is still unclear. Here the authors show, using rat and mouse focal cerebral ischemia models, that the damaged brain signals via the VEGF-C/VEFGR3 axis to activate inflammatory responses in the draining cervical lymph nodes to induce systemic inflammation.

This paper studies the effect of addition of Zn and ultrasound-assisted co-precipitation on Cu-Mn oxide catalysts for the oxidation of CO. Cu-Mn-Zn oxide catalysts were synthesised at different molar ratios and characterised and evaluated for CO oxidation performance. Scanning electron microscopy (SEM) imaging showed that the morphologies and textural properties of the catalysts were markedly affected by the different synthesis conditions, notably the formation of different geometries of various sizes depending on the Zn loading used. Characterisation results also revealed that both the addition of Zn and the use of ultrasound-assisted synthesis improved the dispersion of metal constituents in the mixed oxide phase, evident from the lower peak intensity and smaller crystallite sizes and more dispersed particles as seen in SEM imaging. Gas adsorption results revealed that Zn loading influences the porosity and adsorption capability of the catalyst. Zn-loaded samples exhibited a H3 hysteresis loop, which suggests the presence of slit-like pores. Temperature-programmed reduction results indicate that sonicated samples exhibit improved reduction capabilities which can increase overall catalytic activity. Results showed that Cu/Mn/Zn = 2:1:0.5 displayed the highest activity, showing 76.0% and 82.8% for CO and SO2 at 130 °C, respectively. This work has confirmed that the addition of 0.5 molar ratio of Zn and the presence of ultrasonic treatment during the catalyst synthesis have improved the overall catalytic performance, while further addition in Zn concentrations resulted in an adverse effect on the catalyst.

The purposes of this paper are to investigate Taiwanese undergraduate students’ responses to a selected fictional text, and to propose a critical intercultural pedagogical approach of reading global literature in the EFL educational context based on the insights from the research findings. The authors first critically analysed Xiaolu Guo’s third novel, A Concise Chinese-English Dictionary for Lovers, in terms of Occidentalism, in order to unpack the ideologies underlying her representation of the British and the Chinese. The analysis of the text was conducted through a qualitative approach of critical content analysis. After that, a semi-structured interview was adopted to scrutinise Taiwanese undergraduate students’ perceptions of the ideologies underlying the fiction. The analysis of the text identified three themes of British-Chinese binary opposition together with the juxtaposition of ethnocentric Occidentalism and reverse Occidentalism. The findings obtained from interviews indicated that two of the three dichotomies, i.e. individualism versus collectivism and cosmopolitan versus rooted, underlying the fictional text were commonly perceived by the interviewed undergraduates while they had diverse views on the second dichotomy, sexual freedom versus sexual reticence. Furthermore, some students’ responses to the fiction signified a subtle, implicit and delicate form of reverse Occidentalism. Based on the above research findings, the authors configured a critical intercultural pedagogy for raising students’ capability of decoding and deconstructing Occidentalist ideologies underlying global literature.

Endocrine disruptor compounds (EDCs) such as plasticisers, surfactants, pharmaceutical products, personal care products and pesticides are frequently released into the environmental waters. Therefore, a sensitive and environmentally friendly method is entailed to quantify these compounds at their trace level concentrations. This study encapsulated the β-cyclodextrin crosslinked with citric acid in a polypropylene membrane protected-μ-solid phase extraction (BCD-CA μ-SPE) device for preconcentrating the EDCs (triclosan, triclocarban, 2-phenylphenol, 4-tert-octylphenols and bisphenol A) in real water samples before the analysis by high-performance liquid chromatography. Pneumonia is one of the top ten causes of death. Early diagnosis of pneumonia is essential for subsequent effective treatment. Judging from the current development situation, many effective artificial intelligence (AI) algorithms are concentrated on diseases with a large amount of standardized data. After all, to achieve high-quality AI diagnosis, a large number of high-quality annotated images are required for pre-algorithm training.This research uses semi-supervised learning with Tensorflow and keras two frameworks to construct a disease location system. We use VGG16 and establish a deep convolutional generation confrontation network (DCGAN) [1], synthesize chest X-ray data, and amplify the data set. With an improved visualization effect, a heat map is used to display the lesions to assist clinical medical care personnel to fully grasp the patient's condition. We show examples of CAMs on the pneumonia detection task with no augmentation and DCGAN. It can be concluded that DCGAN performs the best results, the accuracy rate was increased by 11%.

The endogenous release of nutrients from sediments contributes to the eutrophication of landscape water to a certain degree, which depends on the characteristics of sediments. The study explored the characteristics of nitrogen (N) and phosphorus (P) released from two different sediments, which were deposited from reclaimed water (SRW) or surface water (SSW) respectively in Xi’an moat. This paper aimed to compare the effects of nutrient release from SRW and SSW on the water quality. Results showed that the maximum increase rates reached 1.21 mg TN/(L·day) and 0.11 mg TP/(L·day), respectively, in the overlying water of SRW, which were 1.6 and 2.8 times those of SSW. The released amounts of SRW were 0.192 mg TN/g and 0.038 mg TP/g, which were 4.1 and 12.7 times those of SSW. Meanwhile, the densities of benthic algae in SRW and SSW were 5.605 × 109 and 2.846 × 108 cells/L, respectively. Moreover, the species number and individual sizes of benthic algae in SRW were also larger than those in SSW, which played an important role in the nitrogen circulation. Unexpectedly, oxidation reduction potential (ORP) level of SRW was lower than that of SSW, although SRW has a higher dissolved oxygen level. Therefore, the N and P concentrations in the overlying water of SRW were considerably higher than those of SSW, which was mainly attributed to the higher nutrient contents and lower ORP in SRW.

As the population increases, energy demands continue to rise rapidly. In order to satisfy this increasing energy demand, biogas offers a potential alternative. Biogas is economically viable to be produced through anaerobic digestion (AD) from various biomass feedstocks that are readily available in Malaysia, such as food waste (FW), palm oil mill effluent (POME), garden waste (GW), landfill, sewage sludge (SS) and animal manure. This paper aims to determine the potential feedstocks for biogas production via AD based on their characteristics, methane yield, kinetic studies and economic analysis. POME and FW show the highest methane yield with biogas yields up to 0.50 L/g VS while the lowest is 0.12 L/g VS by landfill leachate. Kinetic study shows that modified Gompertz model fits most of the feedstock with R 2 up to 1 indicating that this model can be used for estimating treatment efficiencies of full-scale reactors and performing scale-up analysis. The economic analysis shows that POME has the shortest payback period (PBP), highest internal rate of return (IRR) and net present value (NPV). However, it has already been well explored, with 93% of biogas plants in Malaysia using POME as feedstock. The FW generation rate in Malaysia is approximately 15,000 tonnes per day, at the same time FW as the second place shows potential to have a PBP of 5.4 years and 13.3% IRR, which is close to the results achieved with POME. This makes FW suitable to be used as the feedstock for biogas production.
