This paper deeply investigated the gate leakage and dielectric breakdown mechanisms of the Normally-OFF GaN MOSFET with LPCVD-SiNx/PEALD-AlN dual Gate Dielectric and in-situ H2(15%)/N2(85%) plasma pretreatment. The in-situ plasma pretreatment was performed in a PEALD system prior to the PEALD-AlN deposition. Experimental studies showed that the in-situ H2(15%)/N2(85%) plasma pretreatment is effective in improving the quality of the AlN/GaN interface, the LPCVD-SiNx/PEALD-AlN dual Gate Dielectric exhibits not only high breakdown electric field but low leakage current. Experiments have proved dominant mechanism of the leakage current through LPCVD-SiNx/PEALD-AlN Gate Dielectric is identified to be Poole-Frenkel emission at low fields and Fowler-Nordheim tunneling at high fields. Further, gate dielectric time-dependent dielectric breakdown of electric-field-accelerated and temperature-accelerated was investigated.

The effects of plant volatiles on parasitoids are important with regards to the tri-trophic interactions among host plants, insect herbivores, and their natural enemies. However, the effects of plant volatiles on the important parasitoid (Microplitis pallidipes Szepligeti; Hymenoptera: Braconidae) of noctuids have not been determined at the molecular, laboratory, and outdoor scales with complementary approaches. The odorant binding protein 8 of M. pallidipes (MpOBP8) with an open reading frame of 459 bp was cloned and successfully expressed in Escherichia coli (Eubacteriales: Enterobacteriaceae). The purified protein MpOBP8 had a relatively high binding capacity to the plant volatile β-ionone (Ki value was 28.86 μM) in the fluorescence competitive binding assay. Meanwhile, Y-tube olfactometer experiments indicated that β-ionone selectively attracts the parasitic wasp as the ratio of M. pallidipes selecting β-ionone vs. control was 2:1. Likewise, field screenhouse experiments verified the above results as the ratio of M. pallidipes selecting β-ionone vs. control was close to 2:1. This study suggested that β-ionone is a plant volatile that attracts the parasitoid M. pallidipes, and MpOBP8 may play a key role in the discrimination of M. pallidipes in searching for hosts via β-ionone. In general our results suggest that β-ionone has the potential to enhance the biocontrol effect of parasitoids on insect herbivores.

Increasing studies have suggested that some cardiac glycosides, such as conventional digoxin (DIG) and digitoxin, can induce immunogenic cell death (ICD) in various tumors. We previously found that 3'-epi-12β-hydroxyfroside (HyFS), a novel cardenolide compound isolated by our group, could induce cytoprotective autophagy through inactivation of the Akt/mTOR pathway. However, whether HyFS can induce ICD remains unknown. In this study, we extend our work to further investigate whether HyFS could induce both autophagy and ICD, and we investigated the relationship between autophagy and ICD in three TNBC cell lines. Unexpectedly, compared to DIG, we found that HyFS could induce complete autophagy flux but not ICD in three human triple-negative breast cancer (TNBC) cell lines and one murine TNBC model. Inhibition of HyFS-induced autophagy resulted in the production of ICD in TNBC MDA-MB-231, MDA-MB-436, and HCC38 cells. A further mechanism study showed that formation of RIPK1/RIPK3 necrosomes was necessary for ICD induction in DIG-treated TNBC cells, while HyFS treatment led to receptor-interacting serine-threonine kinase (RIPK)1/3 necrosome degradation via an autophagy process. Additionally, inhibition of HyFS-induced autophagy by the autophagy inhibitor chloroquine resulted in the reoccurrence of ICD and reversion of the tumor microenvironment, leading to more significant antitumor effects in immunocompetent mice than in immunodeficient mice. These findings indicate that HyFS-mediated autophagic degradation of RIPK1/RIPK3 necrosomes leads to inactivation of ICD in TNBC cells. Moreover, combined treatment with HyFS and an autophagy inhibitor may enhance the antitumor activities, suggesting an alternative therapeutic for TNBC treatment.

Previous meditation-related studies included considerably more women than men, potentially concealing gender differences in attention training effects. Here, we present a study exploring potential gender differences in short-term meditation training (MT) effects on performance in an attentional blink (AB) task with a balanced sampling of men and women. A total of 165 college students were randomized into three groups: MT, video game training (VGT), and control (CON). Participants completed a series of pre-intervention assessments, including the Five-Facet Mindfulness Questionnaire (FFMQ), the State-Trait Anxiety Inventory (S-TAI), and self-reported mood, arousal level, and daily time spent playing video games. Participants then completed MT, completed VGT, or had no intervention (CON group). The MT and VGT sessions consisted of 4 days of 20 min of training per day. After training, participants re-rated their arousal state and emotional state, and then performed the AB task. Relative to the CON group, the MT or VGT groups showed a significantly reduced AB. Interestingly, regarding effects on AB performance, MT was more effective in women, whereas VGT was more effective in men. MT, but not VGT, decreased anxiety scores from before to after the intervention. The current results indicate that gender plays an important role in the benefits of attention training, and thus that clinicians should take gender into consideration when implementing meditation-based therapies.

Practitioners with years of mindfulness training (MT) have a reduced attentional blink (AB), relative to novices and non-practitioners. There is still a lack of evidence regarding the impact of relatively short-term intensive traditional MT on AB. Moreover, it is unknown which component(s) of mindfulness underlie mindfulness-associated reductions in AB. Eighty-nine Chinese subjects were randomized into a MT group (N = 44) and a control (CON) group (N = 45). The participants in the CON group were waitlisted for the opportunity to participate in the training program during the 8 weeks that the MT group underwent training. Before (pretest) and after (posttest) the MT program, the subjects completed the Five-Facet Mindfulness Questionnaire (FFMQ), Peace-of-mind questionnaire (POM), and an AB task. Ultimately, 73 participants (MT group/CON group: 35/38) completed the study. From the pretest to the posttest, the MT group exhibited increases in FFMQ scores (total scores, observing subscores, and non-reactivity subscores) and POM scores, as well as improved accuracy in the AB task and a reduction in the lag-1 sparing effect. Increases in T2 accuracy in the AB correlated with improved non-reactivity. These findings broaden our understanding of the association between mindfulness and AB, and support the supposition that the skill of mindful non-reactivity plays an important role in reducing AB.

N6-methyladenosine (m6A) demethylase fat mass and obesity-associated gene(FTO), previously recognized to be related with obesity and diabetes, was gradually discovered to be dysregulated in multiple cancers and plays an oncogenic or tumor-suppressive role. However, the specific expression and pro- or anti-cancer role of FTO in various cancers remained controversial. In this review, through summarizing the available literature, we found that FTO single nucleotide polymorphisms (SNPs) were closely related with cancer risk. Additionally, the dysregulation of FTO was implicated in multiple biological processes, such as cancer cell apoptosis, proliferation, migration, invasion, metastasis, cell-cycle, differentiation, stem cell self-renewal and so on. These modulations mostly relied on the communications between FTO and specific signaling pathways, including PI3K/AKT, MAPK and mTOR signaling pathways. Furthermore, FTO had great potential for clinical application by serving as a prognostic biomarker.

Excessive theta (θ) frequency oscillation and synchronization in the basal ganglia (BG) has been reported in elderly parkinsonian patients and animal models of levodopa (L-dopa)-induced dyskinesia (LID), particularly the θ oscillation recorded during periods when L-dopa is withdrawn (the off L-dopa state). To gain insight into processes underlying this activity, we explored the relationship between primary motor cortex (M1) oscillatory activity and BG output in LID. We recorded local field potentials in the substantia nigra pars reticulata (SNr) and M1 of awake, inattentive resting rats before and after L-dopa priming in Sham control, Parkinson disease model, and LID model groups. We found that chronic L-dopa increased θ synchronization and information flow between the SNr and M1 in off L-dopa state LID rats, with a SNr-to-M1 flow directionality. Compared with the on state, θ oscillational activity (θ synchronization and information flow) during the off state were more closely associated with abnormal involuntary movements. Our findings indicate that θ oscillation in M1 may be consequent to abnormal synchronous discharges in the BG and support the notion that M1 θ oscillation may participate in the induction of dyskinesia.

There has been a recent surge of interest in the development of the Judgement Bias Test (JBT) as a translational assay and objective measure of depressive interpretation bias. Judgement bias, namely the tendency of favoring an approach/avoidance response towards ambiguous cues, is thought to reflect a biased interpretation or anticipation of the cues. However, no study has examined the relationship between judgement bias and depressive interpretation bias. We validated the JBT against established measurements of interpretation bias in a college sample (Study 1) and an adolescent sample (Study 2). In both samples, a negative judgement bias was significantly associated with a depressive interpretation bias. However, the strength of the associations was only moderate and failed to meet the minimum qualification of convergent validity. Our results suggest that judgement bias and interpretation bias might be related but distinct. The JBT should not be used as an indirect assessment of interpretation bias in its current form, and it would be premature to claim that animal studies using the JBT could inform our understanding of cognitive biases in mood disorders. Nevertheless, the JBT might be useful for studying specific mechanisms, such as reward processing under ambiguity.

Purpose The goal of this study was to review relevant randomized controlled trials or case-control studies to determine radical resection compared with simple cholecystectomy for gallbladder carcinoma. Methods Using appropriate keywords, we identified relevant studies using PubMed, the Cochrane Library, and Embase. Key pertinent sources in the literature were also reviewed, and all articles published through September 2019 were considered for inclusion. For each study, we assessed odds ratios, mean difference, and 95% confidence interval (CI) to assess and synthesize outcomes. Results We included 19 studies with a total of 1791 patients in the radical resection group and 3014 in the simple cholecystectomy group. Compared with simple cholecystectomy, radical resection significantly improved the 5-year disease-free survival rate [relative risk (RR): 1.36, 95% CI: 1.02-1.81], the 1-year overall survival (OS) rate (RR: 1.27, 95% CI: 1.04-1.54), and the 3-year OS rate (RR: 1.71, 95% CI: 1.02-2.85). However, there was no significant difference in the recurrence rate (RR: 1.04, 95% CI: 0.87-1.23), and in the 5-year OS rate (RR: 1.05, 95% CI: 0.92-1.19) between the 2 groups. Conclusion Compared with simple cholecystectomy, radical resection has advantages in improving the 5-year disease-free survival rate, and the 1- and 3-year OS rate of gallbladder carcinoma patients.

Host plants have an effect on the susceptibility of lepidopteran insects to nucleopolyhedrovirus (NPV), but the role of the protein in peritrophic membrane (PM) of larvae has not been investigated. We determined a key PM gene (SeIIM8 gene) in Spodoptera exigua (Hübner), which plays a role in protecting against NPV infection. Then we quantified the activity of chitin synthase (SeCHS), the content of insect intestinal mucin (SeIIM) and the expression of their genes in S. exigua fed one of four foods (artificial diet, Brassica oleracea (L.), Glycine max (L.), or Ipomoea aquatica (Forsskal)) with or without inoculation of Spodoptera exigua multiple nucleopolyhedrovirus (SeMNPV). Our results showed that the larval mortality was highest for NPV-infected with siRNA-injected larvae, intermediate for NPV-infected larvae, and lowest for uninfected larvae. The SeCHS activity, SeIIM content, and their relative gene expressions of S. exigua fed G. max or artificial diet were lower compared to caterpillars fed other diets. The relative SeCHS and SeIIM8 gene expression levels of larvae depended on the interaction of NPV treatment, diet, and time. This study concluded that plants affected the susceptibility of insects to NPV by altering the enzyme activity and gene expression of insects.

Summary Urbanization is a pressing challenge for earth’s humans because it is changing not only natural environments but also agricultural lands. Yet, the consequences of cropland loss on pest insect populations that largely depend on these habitats remain largely unclear. We used a 17-year data set to investigate the dynamics of three moth pest species (i.e., striped stem borer, yellow stem borer, and pink stem borer) and their driving forces across the largest mega-urban region of China. Total abundance of three pest species is declined by about 80%, which was strongly associated with cropland loss during rapid urbanization. Our findings indicate that not only the increasing conversion of natural areas to human-dominated landscapes but also that of agricultural lands to urban landscapes can be critical to insect populations. Time perception is an important ability that is related closely to humans’ and animals’ daily activities. It can be distorted by various emotional states. In human studies, experimental pain has been shown to prolong the perception of time. However, related animal studies are lacking. In this study, we used a temporal bisection task to investigate how acute inflammatory pain (induced by hind-paw formalin injection) and chronic neuropathic pain [induced by spinal nerve ligation (SNL)] affected time perception in rats. Rats were trained to recognize “short” (1200-ms) and “long” (2400-ms) anchor-duration pure tones and were rewarded for corresponding lever presses. During testing, rats perceived a series of intermediate-duration and anchor-duration pure tones, and selected levers corresponding to the “short” and “long” tones. After formalin injection, rats gave more “long” lever-press responses than after saline injection. The point of subjective equality after formalin injection also increased, suggesting that formalin-induced acute pain extended time perception. In contrast, rats that had undergone SNL gave fewer “long” lever-press responses compared with the sham surgery group. This animal study suggests that formalin-induced pain and neuropathic pain may have different effects on time perception.

Plant‐insect herbivore‐entomopathogen interactions are one of the hot topics in biological control and humoral immunity, and biochemical metabolism are important responses of herbivores to pathogen infection. Entomopathogens are key biocontrol agents of caterpillars, but how plants affect the responses of caterpillars to these organisms is not well understood. We studied hormonal immunity (lysozyme and phenoloxidase activities) and biochemical metabolism (total protein and lipid contents) of Beauveria bassiana‐infected beet armyworm (Spodoptera exigua) larvae that feed on five different host plants (soya bean, Chinese cabbage, edible amaranth, water convolvulus and pepper). Results indicated that plant species differentially affected lysozyme and phenoloxidase activity and lipid content, but had no effect on protein content of pathogen‐infected caterpillars. Both lysozyme and phenoloxidase activities were generally higher in entomopathogen‐infected larvae that feed on edible amaranth or water convolvulus compared with the other three plants from days 1 to 5 after treatment. Plant species did not affect in regular changes during the 5 days in the lipid content of infected or non‐infected caterpillars. Our study reveals that plants fail to affect the biochemical metabolism but plants can mediate the humoral immunity of caterpillars to defend against pathogens. This study provides insight into plant‐mediated effects on the response of herbivores to pathogens.

The therapeutic goals of patients with chronic pain are not only to relieve pain but also to improve the quality of life. Chronic pain negatively affects various aspects of daily life, such as by decreasing the motivation to work and reward sensitivity, which may lead to difficulties in daily life or even unemployment. Human and animal studies have shown that chronic pain damages reward processing; the exploration of associated internal mechanisms may aid the development of treatments to repair this damage. Incentive salience theory, used widely to describe reward processing, divides this processing into “liking” (reward-induced hedonic sensory impact) and “wanting” (reward-induced motivation) components. It has been employed to explain pathological changes in reward processing induced by psychiatric disorders. In this review, we summarize the findings of studies of reward processing under chronic pain and examine the effects of chronic pain on “liking” and “wanting.” Evidence indicates that chronic pain compromises the “wanting” component of reward processing; we also discuss the neural mechanisms that may mediate this effect. We hope that this review aids the development of therapies to improve the quality of life of patients with chronic pain.

Oscillatory activities within basal ganglia (BG) circuitry in L-DOPA induced dyskinesia (LID), a condition that occurs in patients with Parkinson disease (PD), are not well understood. The aims of this study were firstly to investigate oscillations in main BG input and output structures—the dorsolateral striatum (dStr) and substantia nigra pars reticulata (SNr), respectively— including the direction of oscillation information flow, and secondly to investigate the effects of 5-HT1A/B receptor agonism with eltoprazine on oscillatory activities and abnormal involuntary movements (AIMs) characteristic. To this end, we conducted local field potential (LFP) electrophysiology in the dStr and SNr of LID rats simultaneous with AIM scoring. The LFP data were submitted to power spectral density, coherence, and partial Granger causality analyses. AIM data were analyzed relative to simultaneous oscillatory activities, with and without eltoprazine. We obtained four major findings. 1) Theta band (5–8 Hz) oscillations were enhanced in the dStr and SNr of LID rats. 2) Theta power correlated with AIM scores in the 180-min period after the last LID-inducing L-DOPA injection, but not with daily summed AIM scores during LID development. 3) Oscillatory information flowed from the dStr to the SNr. 4) Chronic eltoprazine reduced BG theta activity in LID rats and normalized information flow directionality, relative to that in LID rats not given eltoprazine. These results indicate that dStr activity plays a determinative role in the causal interactions of theta oscillations and that serotonergic inhibition may suppress dyskinesia by reducing dStr-SNr theta activity and restoring theta network information flow.

Based on the self-terminating thermal oxidation-assisted wet etching technique, two kinds of enhancement mode Al2O3/GaN MOSFETs (metal–oxide–semiconductor field-effect transistors) separately with sapphire substrate and Si substrate are prepared. It is found that the performance of sapphire substrate device is better than that of silicon substrate. Comparing these two devices, the maximum drain current of sapphire substrate device (401 mA/mm) is 1.76 times that of silicon substrate device (228 mA/mm), and the field-effect mobility (μ FEmax) of sapphire substrate device (176 cm2/V⋅s) is 1.83 times that of silicon substrate device (96 cm2/V⋅s). The conductive resistance of silicon substrate device is 21.2Ω ⋅mm, while that of sapphire substrate device is only 15.2Ω ⋅mm, which is 61% that of silicon substrate device. The significant difference in performance between sapphire substrate and Si substrate is related to the differences in interface and border trap near Al2O3/GaN interface. Experimental studies show that (i) interface/border trap density in the sapphire substrate device is one order of magnitude lower than in the Si substrate device, (ii) Both the border traps in Al2O3 dielectric near Al2O3/GaN and the interface traps in Al2O3/GaN interface have a significantly effect on device channel mobility, and (iii) the properties of gallium nitride materials on different substrates are different due to wet etching. The research results in this work provide a reference for further optimizing the performances of silicon substrate devices.

Coupling reagents play crucial roles in the iterative construction of amide bonds for the synthesis of peptides and peptide-based derivatives. The novel DIC/Oxyma condensation system featured with the low risk of explosion displayed remarkable abilities to inhibit racemization, along with efficient coupling efficiency in both manual and automated syntheses. Nevertheless, an ideal reaction molar ratio in DIC/Oxyma condensation system and the moderate reaction temperature by manual synthesis remain to be further investigated. Herein, the synthetic efficiencies of different reaction ratios between DIC and Oxyma under moderate reaction temperature were systematically evaluated. The robustness and efficiency of DIC/Oxyma condensation system are validated by the rapid synthesis of linear centipede toxin RhTx. Different folding strategies were applied for the construction of disulfide bridges in RhTx, which was further confirmed in assays of circular dichroism and patch-clamp electrophysiology evaluation. This work establishes the DIC/Oxyma-based accelerated synthesis of peptides under moderate condensation conditions, which is especially useful for the manual synthesis of peptides. Besides, the strategy presented here provides robust technical supports for the large-scale synthesis and oxidative folding of RhTx.

Due to its size, shape, and inherent expression of pathogen-associated molecular patterns and invasion-assistant adhesion proteins, Burkholderia pseudomallei can easily attach to, and then be internalized by, dendritic cells (DCs), leading to more efficient antigen cross-presentation if modified as carrier. Herein, we engineered Burkholderia pseudomallei as a porous/hollow carrier (SB) for loading tumor lysates (L) and CpG (C) to be used as a tumor vaccine (SB-LC). We found that the adhesion proteins of Burkholderia pseudomallei promote internalization of the SB-LC vaccine by DCs, and result in enhanced DC maturation and antigen cross-presentation. SB-LC induces robust cellular and humoral antitumor responses that synergistically inhibit tumor growth with minimal adverse side effects in several tumor models. Moreover, SB-LC vaccination reverses the immunosuppressive tumor microenvironment, apparently as a result of CD8+-induced tumor ferroptosis. Thus, SB-LC is a potential model tumor vaccine for translating into a clinically viable treatment option.

Neuropathic pain is difficult to cure and is often accompanied by emotional and psychological changes. Exploring the mechanisms underlying neuropathic pain will help to identify a better treatment for this condition. The insular cortex is an important information integration center. Numerous imaging studies have documented increased activity of the insular cortex in the presence of neuropathic pain; however, the specific role of this region remains controversial. Early studies suggested that the insular lobe is mainly involved in the processing of the emotional motivation dimension of pain. However, increasing evidence suggests that the role of the insular cortex is more complex and may even be related to the neural plasticity, cognitive evaluation, and psychosocial aspects of neuropathic pain. These effects contribute not only to the development of neuropathic pain, but also to its comorbidity with neuropsychiatric diseases. In this review, we summarize the changes that occur in the insular cortex in the presence of neuropathic pain and analgesia, as well as the molecular mechanisms that may underlie these conditions. We also discuss potential sex-based differences in these processes. Further exploration of the involvement of the insular lobe will contribute to the development of new pharmacotherapy and psychotherapy treatments for neuropathic pain.

The use of ground cover vegetation is becoming a prominent intervention for promoting biodiversity and associated ecosystem services in Chinese orchards. Despite the large number of studies that have examined the effects of ground cover vegetation on promoting natural enemy populations and related natural pest control, it is less understood whether enhanced natural pest control translates to increase yield and reduced pesticide use. We conducted a 2-year experiment comparing three cover vegetation (ryegrass, clover, and hairy vetch) practices versus a bare ground control in commercial pear orchards in the Yangtze River Delta of East China (YRDEC), China. Natural enemy density (predator and parasitoid abundance), invertebrate herbivore performance (piercing-sucking herbivore abundance and branch-boring and fruit-boring percentage), pesticide input and pear fruit yield were recorded. The results indicated that cover vegetation decreased herbivore abundance and boring percentage by 49.95 and 63.6% respectively, and thus decreased pesticide use by 26.10%. Meanwhile, we found that cover vegetation increased the abundance of natural enemies by 620.75%, and increased pear fruit yield by 6.82%. Piecewise structural equation modelling indicated that increased natural enemy densities, decreased herbivore performance and pesticide use, while increasing fruit yield. Our results confirm that the use of ground cover vegetations (especially with clover and hairy vetch) can promote ecological intensification and biological pest control in pear orchards.

MicroRNAs (miRNAs) are small non-coding RNAs (18~25 nt in length) that act as master regulators of eukaryotic gene expression. They might play an oncogenic or tumor-suppressive role in multiple cancers. In recent decades, several studies have focused on the functions and mechanisms of miR-335 in cancer. The expression level of miR-335 in tissues and cells varies with cancer types, and miR-335 has been proposed as a potential biomarker for the prognosis of cancer. Besides, miR-335 may serve as an oncogene or tumor suppressor via regulating different targets or pathways in tumor initiation, development, and metastasis. Furthermore, miR-335 also influences tumor microenvironment and drug sensitivity. MiR-335 is regulated by various factors such as lncRNAs and microRNAs. In this review, we reveal the functions and targets of miR-335 in various cancers and its potential application as a possible biomarker in prognostic judgment and treatment of malignant tumors.

Background Tri-trophic interactions among plants, insect herbivores and entomopathogens are one of the hot topics in ecology. Although plants have been shown to impact the interactions between herbivores and entomopathogens, it is still unclear how plants affect the cellular immunity of herbivores to entomopathogens. Results The number of hemocytes and the proportion of two main cell types (granular hemocytes and plasmatocytes), plasmatocyte-spreading rate, apoptosis rate, two Spodoptera exigua caspase (SeCasp1, 5) activities and gene expressions were all higher and the activities and gene expression of S. exigua inhibitor of apoptosis protein (SeIAP) were lower in nucleopolyhedrovirus (NPV)-infected caterpillars fed I. aquatica than those fed other plants or artificial diet. Scanning electron microscopy images were consistent with molecular patterns of immune responses. Conclusion This study suggests that host plants affect the immune responses of herbivores to entomopathogens by manipulating the composition, morphology and apoptosis of herbivore hemocytes, which sheds light on the mechanisms that allow host plants to influence multi-trophic interactions. Urbanization is a pressing challenge for earth's human because it is transforming and fragmenting not only natural environments, but also rural lands. Yet, the consequences of rural land-use change on insect populations that depend on these habitats remain largely unclear. We used a 17-year data set to investigate the dynamics of three dominant native farmland moths and its driving forces across the largest megaurban region of China. Total moth abundance declined by about 80%, which was strongly associated with loss and fragmentation of croplands during rapid urbanization. Our findings indicate that urbanization, through loss of traditional agricultural lands, can be a further threat to insect populations. It is therefore really critical to monitor and understand the agrobiodiversity dynamics in rapidly urbanizing regions, which are currently found in many developing countries worldwide.

BackgroundThe oncolytic Newcastle disease virus (NDV) is inherently able to trigger the lysis of tumor cells and induce the immunogenic cell death (ICD) of tumor cells and is also an excellent gene-engineering vector. The macrophage inflammatory protein-3α (MIP-3α) is a specific chemokine for dendritic cells (DCs). Thus, we constructed a recombinant NDV expressing MIP-3α (NDV-MIP3α) as an in vivo DC vaccine for amplifying antitumor immunities.MethodsThe recombinant NDV-MIP3α was constructed by the insertion of MIP-3α cDNA between the P and M genes. Western blotting assay and ELISA were used to detect MIP-3α, HMGB1, IgG, and ATP in the supernatant and sera. The chemotaxis of DCs was examined by Transwell chambers. The phenotypes of the immune cells (eg, DCs) were analyzed by flow cytometry. The antitumor efficiency of NDV-MIP3α was observed in B16 and CT26 tumor-bearing mice. Immunofluorescence and immunohistochemistry were applied to observe the ecto-calreticulin (CRT) and intratumoral attraction of DCs. Adoptive transfer of splenocytes and antibodies and depletion of T-cell subsets were used to evaluate the relationship between antitumor immunities and the role of the T-cell subtype.ResultsThe findings show that NDV-MIP3α has almost the same capabilities of tumor lysis and induction of ICD as the wild-type NDV (NDV-WT). MIP-3α secreted by NDV-MIP3α could successfully attract DCs in vitro and in vivo. Both B16 and CT26 cells infected with NDV-MIP3α could strongly promote DC maturation and activation. Compared with NDV-WT, intratumoral injection of NDV-MIP3α and the adoptive transfer of T lymphocytes from mice injected with NDV-MIP3α resulted in a significant suppression of B16 and CT26 tumor growth. The NDV-MIP3α-induced production of tumor-specific cellular and humoral immune responses was dependent on CD8+ T cells and partially on CD4+ T cells. A significant reversion of tumor microenvironments was found in the mice injected with NDV-MIP3α.ConclusionsCompared with NDV-WT, the recombinant NDV-MIP3α as an in vivo DC vaccine demonstrates enhanced antitumor activities through the induction of stronger system immunities and modulation of the tumor microenvironment. This strategy may be a potential approach for the generation of an in vivo DC vaccine.

Nucleopolyhedrovirus (NPV) and parasitic wasps are two important biological control agents of lepidoptera caterpillars, and the compatibility of parasitism and NPV infection remains a complicated process. In this study, we examined the effects of different learning experiences by the endoparasitoid Microplitis pallidipes on their ability to discriminate NPV-infected from healthy caterpillars (larvae of beet armyworm, Spodoptera exigua). Compared to the parasitoids with no prior host contact (control), parasitoids with oviposition experience with healthy caterpillars had shorter searching time, shorter attacking time, a greater number of attacks, higher parasitism rate and greater comprehensive discrimination ability to both non-infected and NPV-infected caterpillars. They also had a higher percentage of first attacks on non-infected caterpillars than NPV-infected caterpillars, and these differences were more obvious with increasing time post-virus-inoculation. However, the values of these six host-discrimination indicators on NPV-infected caterpillars were not significant between the control parasitoids and parasitoids with oviposition experience on NPV-infected caterpillars from hours 24 to 96 after virus inoculation. Compared to parasitoids with oviposition experience on healthy caterpillars, parasitoids with oviposition experience on NPV-infected caterpillars generally had longer searching time, longer attacking time, fewer attacks, lower parasitism rate, and lower comprehensive discrimination ability to both non-infected and NPV-infected caterpillars, and a lower percentage of first attacks on non-infected vs. NPV-infected caterpillars. These results suggest that experience with healthy hosts helps parasitoids distinguish NPV-infected caterpillars from healthy caterpillars, but that experience with NPV-infected caterpillars did not convey the same discrimination ability.
