Abstract Background Metabolic dysfunction-associated steatotic liver disease (MASLD) patients are at an elevated risk of developing severe coronavirus disease 2019 (COVID-19). The objective of this study was to assess antibody responses and safety profiles six months after the third dose of the inactivated acute respiratory syndrome coronavirus 2 (SARS-CoV-2) vaccine in MASLD patients. Methods This study included MASLD patients and healthy volunteers without a history of SARS-CoV-2 infection. Blood samples were collected six months after receiving the third dose of the inactivated vaccine to measure the levels of neutralizing antibodies (NAbs) and anti-spike IgG antibodies against SARS-CoV-2. Results A total of 335 participants (214 MASLD patients and 121 healthy volunteers) were enrolled. The seroprevalence of NAb was 61.7% (132 of 214) in MASLD patients and 74.4% (90 of 121) in healthy volunteers, which was a significant difference (p = 0.018). Statistically significant differences in IgG seroprevalence were also observed between MASLD patients and healthy volunteers (p = 0.004). Multivariate analysis demonstrated that the severity of MASLD (OR, 2.97; 95% CI, 1.32–6.68; p = 0.009) and age (OR, 1.03; 95% CI, 1.01–1.06; p = 0.004) were independent risk factors for NAb negativity in MASLD patients. Moderate/severe MASLD patients had a lower NAb seroprevalence than mild MASLD patients (45.0% vs. 65.5%, p = 0.016). Conclusion Lower antibody responses were observed in MASLD patients six months after their third dose of the inactivated vaccine than in healthy volunteers, providing further assistance in monitoring patients who are more vulnerable to hypo-responsiveness to SARS-CoV-2 vaccines.

The healing of large skin defects remains a significant challenge in clinical settings. The lack of epidermal sources, such as autologous skin grafting, limits full-thickness skin defect repair and leads to excessive scar formation. Skin organoids have the potential to generate a complete skin layer, supporting in-situ skin regeneration in the defect area. In this study, skin organoid spheres, created with human keratinocytes, fibroblasts, and endothelial cells, showed a specific structure with a stromal core surrounded by surface keratinocytes. We selected an appropriate bioink and innovatively combined an extrusion-based bioprinting technique with dual-photo source cross-linking technology to ensure the overall mechanical properties of the 3D bioprinted skin organoid. Moreover, the 3D bioprinted skin organoid was customized to match the size and shape of the wound site, facilitating convenient implantation. When applied to full-thickness skin defects in immunodeficient mice, the 3D bioprinted human-derived skin organoid significantly accelerated wound healing through in-situ regeneration, epithelialization, vascularization, and inhibition of excessive inflammation. The combination of skin organoid and 3D bioprinting technology can overcome the limitations of current skin substitutes, offering a novel treatment strategy to address large-area skin defects.

To enhance flow stability and reduce hydrodynamic noise caused by fluctuating pressure, a quasi-periodic elastic support skin composed of flexible walls and elastic support elements is proposed for fluid noise reduction. The arrangement of the elastic support element is determined by the equivalent periodic distance and quasi-periodic coefficient. In this paper, a dynamic model of skin in a fluid environment is established. The influence of equivalent periodic distance and quasi-periodic coefficient on flow stability is investigated. The results suggest that arranging the elastic support elements in accordance with the quasi-periodic law can effectively enhance flow stability. Meanwhile, the hydrodynamic noise calculation results demonstrate that the skin exhibits excellent noise reduction performance, with reductions of 10 dB in the streamwise direction, 11 dB in the spanwise direction, and 10 dB in the normal direction. The results also demonstrate that the stability analysis method can serve as a diagnostic tool for flow fields and guide the design of noise reduction structures.

Senecaviurs A (SVA) infection, an emerging infectious disease in pig populations, is characterized by vesicular lesions predominantly affecting the mouth, snout, and hooves of infected pigs, similar to the symptoms of Foot and Mouth Disease Virus (FMDV). This disease first spread into China in 2015, causing great panic in the pig breeding industry. To determine the prevalence of SVA in pig herds in China from 2018 to 2021, a total of 4,901 pig tissue samples were collected from 18 provinces, autonomous regions and municipalities (P.A.M.s) for epidemiological investigation, virus isolation and genetic analysis. In 2021, the individual positive rates (IPRs) from the perspective of spatial distribution in East China, South China, Central China, North China, Southwest China, Northwest China, and Northeast China were 0, 0, 1.69, 0.94, 11.70, 3.31 and 2.21%, respectively. The herd positive rates (HPRs) were 0, 0, 9.52, 9.09, 50.00, 7.69 and 23.08%. From the perspective of temporal distribution, the IPR showed an overall downwards trend from 2018 to 2021, with only a slight increase in 2020. Moreover, the HPR decreased from 36.63 to 10.07%. From the perspective of population distribution in 2021, the IPR (2.62%) and HPR (12.00%) in apparently healthy pig herds (slaughterhouses) were greater than those in non-healthy pig herds (2.10 and 5.13%, respectively), consistent with the results in 2019. To characterize the prevalent strains, 10 SVA strains isolated from positive samples in 2019 were clustered in Clades I and VII; SVA-FJ039-2019, SVA-HuN032-2019, SVA-GX011-2019, SVA-FJ036-2019, SVA-GXF011-2019 and SVA-GXF053-2019 were clustered in Clade I; and SVA-FJ018-2019, SVA-SD069-2019, SVA-SD072-2019, and SVA-SD074-2019 were clustered in Clade VII. In conclusion, until 2021, the prevalence of SVA in pig herds in China was still relatively high, the contaminated area was still large, and there were a number of hidden infections. In the future, the epidemic status of SVA in pig herds in China must be closely monitored and the prevention and control measures must be adjusted in a timely manner.

Abstract To investigate the effect of “Small Private Online Course” (SPOC) based on flipped classroom teaching model on the students in the course of fundamental operations in surgery. A prospective study. 8-year program students (juniors) majored in clinical medicine in Navy medical university. The mastery of theoretical knowledge and operational skill of the students, the comparison of final test examination score between traditional teaching method and “SPOC + flipped classroom” model and the feedback completed by students. Our study found that SPOC + flipped classroom could significantly increase the efficacy of the class and enhance the ability of the students compared with the traditional method. The new teaching model could have a positive influence for medical students on their basic knowledge and operational skill.

Thermochromic textiles possess the capability to indicate ambient temperature through color changes, enabling real-time temperature monitoring and providing temperature warnings for body heat management. In this study, three thermochromic dyes—blue, red, and yellow—were synthesized using crystalline violet lactone (CVL), 6′-(diethylamino)-1′,3′-dimethyl-fluoran (DDF), and 3′,6′-dimethoxyfluoran (DOF) as leuco dyes, respectively, with biomass tea polyphenol serving as the color developer and tetradecanol as the phase change material. The chemical structures of these dyes were characterized using UV spectroscopy, infrared spectroscopy, Raman spectroscopy and 1H NMR. The thermochromic mechanisms were investigated, revealing that the binding bonds between the leuco dyes and the color developer broke and reorganized with temperature changes, imparting reversible thermochromic property. Polyester fabrics were dyed using an impregnation method to produce three reversible thermochromic fabrics in blue, red, and yellow. The structure and properties of these fabrics were analyzed, showing a significant increase in the UPF value from 26.3 to approximately 100, indicating enhanced UV resistance. Water contact angle measurements revealed that the contact angle of undyed polyester fabrics was 139°, while that of dyed polyester fabrics decreased by about 40°, indicating improved hydrophilicity. Additionally, the fabric inductive static tester showed that the static voltage half-life of dyed polyester fabric was less than 1 s, demonstrating a significant antistatic effect. Infrared thermal imaging results indicated that during the warming and cooling process, the thermochromic polyester fabric exhibited specific energy storage and insulation effects at 38 °C, close to the human body temperature. This study presented a novel approach to developing smart color-changing textiles using biomass-derived thermochromic dyes, offering diverse materials for personal thermal management, and intelligent insulation applications.

BackgroundIt remains unclear whether BPIV3 infection leads to stress granules formation and whether G3BP1 plays a role in this process and in viral replication. This study aims to clarify the association between BPIV3 and stress granules, explore the effect of G3BP1 on BPIV3 replication, and provide significant insights into the mechanisms by which BPIV3 evades the host’s antiviral immunity to support its own survival.MethodsHere, we use Immunofluorescence staining to observe the effect of BPIV3 infection on the assembly of stress granules. Meanwhile, the expression changes of eIF2α and G3BP1 were determined. Overexpression or siRNA silencing of intracellular G3BP1 levels was examined for its regulatory control of BPIV3 replication.ResultsWe identify that the BPIV3 infection elicited phosphorylation of the eIF2α protein. However, it did not induce the assembly of stress granules; rather, it inhibited the formation of stress granules and downregulated the expression of G3BP1. G3BP1 overexpression facilitated the formation of stress granules within cells and hindered viral replication, while G3BP1 knockdown enhanced BPIV3 expression.ConclusionThis study suggest that G3BP1 plays a crucial role in BPIV3 suppressing stress granule formation and viral replication.

Diabetes mellitus is a chronically inflamed disease that predisposes to delayed fracture healing. Macrophages play a key role in the process of fracture healing by undergoing polarization into either M1 or M2 subtypes, which respectively exhibit pro-inflammatory or anti-inflammatory functions. Therefore, modulation of macrophage polarization to the M2 subtype is beneficial for fracture healing. Exosomes perform an important role in improving the osteoimmune microenvironment due to their extremely low immunogenicity and high bioactivity. In this study, we extracted the M2-exosomes and used them to intervene the bone repair in diabetic fractures. The results showed that M2-exosomes significantly modulate the osteoimmune microenvironment by decreasing the proportion of M1 macrophages, thereby accelerating diabetic fracture healing. We further confirmed that M2-exosomes induced the conversion of M1 macrophages into M2 macrophages by stimulating the PI3K/AKT pathway. Our study offers a fresh perspective and a potential therapeutic approach for M2-exosomes to improve diabetic fracture healing.

A decoupled model predictive control (MPC) power balancing strategy for a coupled inductor-based flying capacitor DC–DC converter (FCDC) is a proposed to solve the power imbalance caused by the parameter differences in the coupled inductor. The decoupled mathematical model of coupled inductor FCDC is firstly derived by analyzing the converter operation state under various modes. On this basis, the control relationship between inductor current and flying capacitor (FC) voltage is redefined and an MPC power balance strategy based on the inductor current with single-step optimization is proposed. The proposed MPC strategy not only achieves decoupled power balancing control but also solves multi-objective dynamic optimization control of the inductor current and FC voltage, greatly reducing the computation load. A detailed theoretical analysis of the proposed strategy is presented and the balancing performance is effectively verified through the experiments.

Background: Differentiating adrenal adenomas from metastases poses a significant challenge, particularly in patients with a history of extra-adrenal malignancy. This study investigates the performance of three-phase computed tomography (CT) based robust federal learning algorithm and traditional deep learning for distinguishing metastases from benign adrenal lesions. Material and methods: This retrospective analysis includes 1187 instances who underwent three-phase CT scans between January 2008 and March 2021, comprising 720 benign lesions and 467 metastases. Utilizing the three-phase CT images, both a Robust Federal Learning Signature (RFLS) and a traditional Deep Learning Signature (DLS) were constructed using the Least Absolute Shrinkage and Selection Operator (LASSO) logistic regression. Their diagnostic capabilities were subsequently validated and compared using metrics such as the Areas Under the Receiver Operating Curve (AUCs), Net Reclassification Improvement (NRI), and Decision Curve Analysis (DCA). ObjectivesIn patients with hepatocellular carcinoma (HCC), accurately predicting the preoperative microvascular invasion (MVI) status is crucial for improving survival rates. This study proposes a multi-modal domain-adaptive fusion model based on deep learning methods to predict the preoperative MVI status in HCC.Materials and methodsFrom January 2008 to May 2022, we collected 163 cases of HCC from our institution and 42 cases from another medical facility, with each case including Computed Tomography (CT) images from the pre-contrast phase (PCP), arterial phase (AP), and portal venous phase (PVP). We divided our institution’s dataset (n=163) into training (n=119) and test sets (n=44) in an approximate 7:3 ratio. Additionally, we included cases from another institution (n=42) as an external validation set (test1 set). We constructed three single-modality models, a simple concatenated multi-modal model, two current state-of-the-art image fusion model and a multi-modal domain-adaptive fusion model (M-DAFM) based on deep learning methods. We evaluated and analyzed the performance of these constructed models in predicting preoperative MVI using the area under the receiver operating characteristic curve (AUC), decision curve analysis (DCA), and net reclassification improvement (NRI) methods.ResultsIn comparison with all models, M-DAFM achieved the highest AUC values across the three datasets (0.8013 for the training set, 0.7839 for the test set, and 0.7454 for the test1 set). Notably, in the test set, M-DAFM’s Decision Curve Analysis (DCA) curves consistently demonstrated favorable or optimal net benefits within the 0-0.65 threshold probability range. Additionally, the Net Reclassification Improvement (NRI) values between M-DAFM and the three single-modal models, as well as the simple concatenation model, were all greater than 0 (all p < 0.05). Similarly, the NRI values between M-DAFM and the two current state-of-the-art image fusion models were also greater than 0. These findings collectively indicate that M-DAFM effectively integrates valuable information from multi-phase CT images, thereby enhancing the model’s preoperative predictive performance for MVI.ConclusionThe M-DAFM proposed in this study presents an innovative approach to improve the preoperative predictive performance of MVI.

Our study aims to investigate the water quality and distribution patterns of phytoplankton communities in the Jinjiang River Basin in Quanzhou, as well as their relationship with environmental factors. We integrated data from the national water quality databases of the two main tributaries of the West and East Jinjiang Rivers between 2020 and 2023, supplemented by field surveys. Redundancy analysis was used to explore the effect of environmental factors on phytoplankton communities. Our findings revealed that the West Jinjiang River experienced a significant influence from excessive fertilizer use in tea cultivation, leading to an increase in TN concentrations compared to the East Jinjiang River. The abundance of phytoplankton in the Jinjiang River Basin was 105 cells·L−1, with phytoplankton being dominated by Chlorophyta, Cyanphyta, and diatoms, accounting for an average of 50%, 20%, and 19% of the total phytoplankton abundance, respectively. Redundancy analysis indicated that temperature, pH, and nutrient concentrations were important factors influencing the phytoplankton communities. With increasing temperature and nutrients concentrations, the abundance of Chlorophyta and Dinophyta significantly increased. This study provides a solid foundation for the regular “health diagnosis” of crucial rivers and lakes in Quanzhou and supports the establishment of a health guarantee system for rivers and lakes.

In the design of flapping-wing micro-size air vehicles capable of hovering, wings serve as the primary source of hovering power, making the analysis of aerodynamics and aerodynamic efficiency crucial. Traditional quasi-steady models treat the wings as single rigid plane, neglecting the deformable characteristics of flexible wings. This paper proposes a multi-plane method that, in conjunction with various design parameters of flexible wings in a two-dimensional plane, analyzes their deformation characteristics under the assumption of multiple planes in three-dimensional space, and describes the deformation of wings during flapping. By combining the quasi-steady aerodynamic model, aerodynamic analysis of the deformed wings can be conducted. The relationship between the slack angle, wing flapping position, and wing deformation are analyzed, along with their effects on aerodynamics and aerodynamic efficiency. Experiments validate the deformation patterns of wings during flapping and compare the simulated aerodynamic forces with measured ones. The results indicate that wing deformation can be accurately described by adjusting the parameters in the multi-plane method and that the aerodynamic analysis using this method closely approximates the average lift results. Additionally, the multi-plane method establishes a connection between wing morphology and aerodynamic forces and efficiency, providing valuable insights for aerodynamic analysis.

Skeletal muscle disorders have posed great threats to health. Selective delivery of drugs and oligonucleotides to skeletal muscle is challenging. Aptamers can improve targeting efficacy. In this study, for the first time, the human skeletal muscle-specific ssDNA aptamers (HSM01, etc.) were selected and identified with Systematic Evolution of Ligands by Exponential Enrichment (SELEX). The HSM01 ssDNA aptamer preferentially interacted with human skeletal muscle cells in vitro. The in vivo study using tree shrews showed that the HSM01 ssDNA aptamer specifically targeted human skeletal muscle cells. Furthermore, the ability of HSM01 ssDNA aptamer to target skeletal muscle cells was not affected by the formation of a disulfide bond with nanoliposomes in vitro or in vivo, suggesting a potential new approach for targeted drug delivery to skeletal muscles via liposomes. Therefore, this newly identified ssDNA aptamer and nanoliposome modification could be used for the treatment of human skeletal muscle diseases.

Abstract Background Cardiac-resident mesenchymal stem cells (cMSCs) can exhibit fibrotic, proinflammatory, and proangiogenic phenotype in response to myocardial ischemia (Isch). How their phenotypic fate decisions are determined remains poorly understood. Here, we demonstrate that the cooperation of Oct4 and c-Myc in cMSCs creates a preferable mesenchymal-to-endothelial transition (MEndoT) to promote angiogenesis and consequent myocardial repair. Methods We collected MSCs from cardiac and peripheral blood of rat with left ventricular Isch (LV Isch) 30 days after myocardial infarction (MI) or sham operation. After a comparison of characterization between cMSCs and peripheral blood MSCs (pbMSCs), we conducted transcriptome analysis and RNA sequencing of cMSCs. Using loss/gain-of-function approaches to understand the cooperation of c-Myc and Oct4 on MEndoT of cMSCs under hypoxic condition, we explored the mechanisms through transcriptome and functional experiment, and chromatin immunoprecipitation. Next, we transplanted male cMSCs with overexpression or inhibition of c-Myc/Oct4 into the infarcted myocardium of female rats and evaluated infarct size, cell retention, inflammation, remodeling, and function after 30 days. Results LV Isch switched cMSCs toward both inflammatory and proangiogenic phenotypes, with increased secretion of inflammatory cytokines as well as decreased expression of proangiogenic factors. The effect of LV Isch on pbMSCs was less remarkable. Gene expression heatmap showed imbalance in expression of Oct4 and c-Myc regulating genes associated with remodeling of cMSCs. We provided evidence that cMSCs-specific c-Myc- versus Oct4-overexpression showed divergent genomic signatures, and their corresponding target genes play an important role in regulating cMSCs phenotypic changes. In particular, Oct4 accelerated angiogenesis induced by c-Myc overexpression in cMSCs and inhibited their phenotypic transition into inflammatory cells and fibroblast. Mechanistically, exogenous Oct4 caused c-Myc to translocate from the nucleus to the cytoplasm and activated some of its target signalings including VEGF signaling. Although transplantation of cMSCs alone did not improve LV remodeling and function, cMSCs co-transfected with c-Myc and Oct4 promoted a more positive effect in their survival and reparative properties, increased animal survival, reduced infarct size, decreased scar thickness, inhibited LV remodeling, and improved heart function 30 days after MI. Significantly, Oct4 promoted MEndoT (“Rescue me” signal) of cMSCs after both c-Myc stimulation in vitro and transplantation into the infarcted heart. Conclusions Myocardial Isch drives resident cMSCs toward multiple phenotypes. Oct4 interacts with c-Myc to promote MEndoT capacity of cMSCs and improve their survival and reparative effects through upregulation of angiogenesis-related signaling pathways. These findings may identify novel targets for stem cell therapy.

Most of the driving mechanisms used in micro flapping wing vehicles are gear and linkage transmission mechanisms, which convert the unidirectional rotation of the motor into the reciprocating flapping of the driving mechanism. However, gear and linkage transmission mechanisms occupy a larger space and weight with certain energy losses. This article introduces a drive mechanism that is different from gear and linkage mechanisms, namely the string-based mechanism. In this study, the working principle and various parameters in string-based mechanisms are analyzed, and the flapping trajectory and amplitude of the mechanism is simulated. Following that, this article proposes an improved method of the cam mechanism, designs the trajectory of a cam mechanism, and a physical design is carried out with a driving mechanism width of 22 mm. Subsequently, the experiments are conducted to compare the flapping trajectory of the actual mechanism with the simulation trajectory, thereby verifying the simulation accuracy. The results prove that, in a string-based mechanism, the ratio of the radius of the pulley to the installation radius of the double-layer bearings has a direct impact on the flapping amplitude, and optimizing the design of cam mechanisms can effectively reduce the tensile and relaxation phenomena in string-based mechanisms. The above conclusion was verified in physical experiments, where the modified cam mechanism effectively reduced the stuck phenomenon in the string-based mechanism.

Abstract Background Lumbar disc herniation (LDH) is a common and frequently occurring disease in clinics. Low back pain and sciatica are the presenting symptoms of LDH. To some extent, it can be considered that measures with the capability to improve low back pain or sciatica have the potential to treat LDH. Ma’s bamboo-based medicinal moxibustion therapy can effectively reduce the degree of low back pain and has been widely used. Studies of small sample size have seen significant improvement on pain relief. The aim of this trial is to evaluate the clinical efficacy and safety of Ma’s bamboo-based medicinal moxibustion therapy in the treatment of LDH low back pain. Methods/design The trial is a multicenter, randomized, parallel-group, non-inferiority study. Three hundred and twelve patients will be randomly assigned to a Ma’s bamboo-based medicinal moxibustion group (n=156) and an acupuncture group (n=156). Patients in each group will receive treatment every day, 6 times a week, 12 times in total. Follow-up will be conducted 14 days after treatment. The primary outcome will be the visual analog scale(VAS) at baseline, after 6 times of treatment, end of treatment, and follow-up. The secondary outcomes will include Oswestry disability indexes (ODI), modified Japanese Orthopaedic Association low back pain (M-JOA) score, serum β-endorphin (β-EP), and serum substance P (SP). β-EP and SP, as well as safety evaluation indexes (routine blood, liver, and kidney function and electrocardiogram), will be measure at baseline and after the end of treatment. The number, nature, and severity of adverse events will be recorded. Discussion The results of the trial will compare the efficacy of low back pain in LDH between Ma’s bamboo-based medicinal moxibustion group and the acupuncture group and will be expected to make a systematic and objective evaluation of the clinical efficacy and safety of Ma’s bamboo-based medicinal moxibustion therapy. Abstract Background To evaluate the specificity of the expression patterns of microRNAs (miRNAs) in circulating CD4+ T cells in aged patients with atherosclerosis obliterans (ASO). Methods A comprehensive miRNA expression study was conducted using a miRNA microarray of CD4+ T cells isolated from peripheral blood mononuclear cells (PBMCs) of 33 patients with ASO and 24 healthy donors. A t test was used for statistical analysis, and the average linkage method was used for hierarchical clustering. The results were validated by qRT–PCR. Putative targeted pathways associated with validated miRNAs were predicted with the online software DIANA miRPath. Results We identified 44 miRNAs based on a cutoff value of a 1.3-fold change in expression between the two groups, with 18 miRNAs showing a false discovery rate (FDR) p value

Abstract Background The health benefits of anthocyanins impel researchers and food producers to explorer new methods to increase anthocyanin contents in plant foods. Our previous studies revealed a positive role of nitric oxide (NO) in anthocyanin accumulation in radish (Raphanus sativus L.) sprouts. The application of hemin, an inducer of heme oxygenase-1 (HO-1), can effectively elevate NO production in vivo. Hemin treatment also improves plant growth and stress tolerance. This study is aimed to assess the effects of hemin treatment on anthocyanin production in radish sprouts, and to investigate whether NO signalling is involved in this process. Results The application of hemin significantly up regulated the expressions of many anthocyanins biosynthesis related structure and regulatory genes, leading to increased anthocyanins accumulation in radish hypocotyls. Hemin treatment also raised NO contents in radish sprouts, probably through enhancing nitrate reductase (NR) activity and Nitric Oxide-Associated 1 (NOA1) expression. Comparing the effects of Zinc Protoporphyrin (ZnPP, HO-1 activity inhibitor), Sodium Nitroprusside (SNP, NO donor) and carboxy-PTIO (cPTIO, NO-scavenger) on anthocyanin and NO production, a positive role of NO signalling has been revealed in hemin-derived anthocyanin accumulation. A positive feedback loop between HO-1 and NO may be involved in regulating this process. Conclusions Hemin induced anthocyanin accumulation in radish sprouts through HO-1 and NO signalling network.

Porcine reproductive and respiratory syndrome virus (PRRSV) is globally prevalent and seriously harms the economic efficiency of pig farming. Because of its immunosuppression and high incidence of mutant recombination, PRRSV poses a great challenge for disease prevention and control. Nonstructural protein 2 (NSP2) is the most variable functional protein in the PRRSV genome and can generate NSP2N and NSP2TF variants due to programmed ribosomal frameshifts. These variants are broad and complex in function and play key roles in numerous aspects of viral protein maturation, viral particle assembly, regulation of immunity, autophagy, apoptosis, cell cycle and cell morphology. In this paper, we review the structural composition, programmed ribosomal frameshift and biological properties of NSP2 to facilitate basic research on PRRSV and to provide theoretical support for disease prevention and control and therapeutic drug development.

The diffusion growth of intermetallic compounds in Al-Er alloys are closely related to the properties of the alloys. The current work aims at explaining the dominance of Al3Er in the Al-Er alloys precipitation phases and the interface thin layer phenomenon by diffusion couple technique, estimating the parabolic growth constant and diffusion activation energy of intermetallic compound in Al-Er diffusion couples to provide theoretical guidance for the design of new Al-Er alloys. In this work, Al-Er diffusion couples were successfully prepared by casting-cladding method in the atmosphere. The growth of Al-Er intermetallic compounds at diffusion couple interface during annealing were observed and recorded by High-Temperature Laser-Scanning Confocal Microscopy at 673, 698, 723 and 748 K respectively. The results show that the growth characteristics of Al-Er intermetallic compounds were accord with layer-terraced growth during annealing. The thickness of intermetallic compound was linear with the square root of time at experimental temperature. The intermetallic compound layer was composed of Al3Er and a very thin AlEr phase. The parabolic growth constants of Al3Er phase at 673, 698, 723 and 748 K were 1.017 × 10−14, 1.609 × 10−14, 3.111 × 10−14 and 4.76 × 10−14 respectively. The activation energy of Al3Er phase was (88.4 ± 5.3) kJ/mol and the pre-exponential factor was 7.126 × 10−8 m2/s.

Abstract Background Inflammation markers have an important effect on tumor proliferation, invasion, and metastasis. Oligometastatic disease (OMD) is an intermediate state between widespread metastases and locally confined disease, where curative strategies may be effective for some patients. We aimed to explore the predictive value of inflammatory markers in patients with oligometastatic colorectal cancer (OMCC) and build a nomogram to predict the prognosis of these patients. Methods Two hundred nine patients with OMCC were retrospectively collected in this study. The Kaplan-Meier survival curves and Cox regression analysis were used to estimate overall survival (OS) and progression-free survival (PFS). A multivariate Cox analysis model was utilized to establish the nomogram. The concordance index (C-index), calibration curve, and receiver operating characteristics (ROC) were established to verify the validity and accuracy of the prediction model. Results According to the multivariate analysis, decreased platelet-to-lymphocyte ratio (PLR) might independently improve OS in patients with OMCC (HR = 2.396, 95% CI 1.391–4.126, P = 0.002). Metastases of extra-regional lymph nodes indicated poor OS (HR = 2.472, 95% CI 1.247–4.903, P = 0.010). While the patients with early N stage had better OS (HR = 4.602, 95% CI 2.055–10.305, P = 0.001) and PFS (HR = 2.100, 95% CI 1.364–3.231, P = 0.007). Primary tumor resection (HR = 0.367, 95% CI 0.148–0.908, P = 0.030) and lower fibrinogen (HR = 2.254, 95% CI 1.246–4.078, P = 0.007) could significantly prolong the OS in patients with OMCC. PLR, metastases of extra-regional lymph nodes, N stage, primary tumor resection, and fibrinogen were used to make up the nomogram. The C-index and area under the curve (AUC) of the ROC in nomogram were 0.721 and 0.772 respectively for OS, showed good consistency between predictive probability of OS and actual survival. Conclusions Decreased PLR could predict a good prognosis in patients with OMCC. The nomogram including inflammatory factors and clinicopathological markers was credible and accurate to predict survivals in patients with OMCC.

The cellular composition and underlying spatiotemporal transformation processes of subchondral bone in osteoarthritis (OA) remain unknown. Herein, various cell subsets from tibial plateau of patients with OA are identified, and the mechanism of subchondral microstructure alteration is elaborated using single-cell RNA sequencing technique. We identified two novel endothelial cell (EC) populations characterised by either exosome synthesis and inflammation response or vascular function and angiogenesis. Three osteoblast (OB) subtypes are introduced, separately related to vascularisation, matrix manufacturing and matrix mineralisation. The distinct roles and functions of these novel phenotypes in OA development are further discussed as well as interaction network between these subpopulations. The variation tendency of each population is testified in a destabilisation of the medial meniscus mouse model. The identification of cell types demonstrates a novel taxonomy and mechanism for ECs and OBs inside subchondral bone area provides new insights into the physiological and pathological behaviours of subchondral bone in OA pathogenesis.

PurposeWe used bibliometric methods to assess the global scientific output on the IRAEs for colorectal cancer and to explore the current status and trends in the field over the last three decades.MethodsStudies on immune-related adverse events for colorectal cancer published from 1996 to 2022 were retrieved from the Web of Science. For quantitative and qualitative assessments of publication outputs and author contributions, the R bibliometrix package was used. VOSviewer was used to construct networks based on the co-authorship of countries/institutions/authors, co-citation analysis of journals/references, citation analysis of documents, and co-occurrence of keywords.ResultsA total of 237 relevant articles were included in the final analysis. The number of publications has increased significantly over time. The countries and institutions that contributed most to the field were the USA and the University of Texas MD Anderson Cancer Center. Jefferey Schlom was the most productive author, ranking first in cited authors. The most cited document was Topalian et al. in The New England Journal of Medicine (2012). The journals with the highest number of selected articles and citations were The New England Journal of Medicine and the Journal of Clinical Oncology, respectively. Co-occurrence analysis showed that IRAEs for colorectal cancer were associated with immunotherapy, open-label, chemotherapy, nivolumab, and PD-1. Trend analysis showed that immune checkpoint inhibitors, gut-microbiota, inflammatory-bowel disease, and PD-1has been on the rise in recent years to IRAEs for colorectal cancer.ConclusionOur bibliometric analysis showed that studying IRAEs for colorectal cancer is increasingly a hot topic. The focus of the research had evolved from traditional treatment modalities such as targeted therapy to gut microbiota. Inflammatory bowel disease may be a future research hotspot of IRAEs for colorectal cancer.

The power conversion of salinity gradient energy (SGE) between concentrated brine from seawater desalination and seawater by reverse electrodialysis (RED) benefits energy conservation and also dilutes the discharge concentration to relieve the damage to coastal ecosystems. However, two key performance indexes of the maximum net power density and energy conversion efficiency for a RED stack harvesting the energy usually cannot reach the optimal simultaneously. Here, an optimization study on the two indexes was implemented to improve the performance of RED in harvesting the energy. A RED model for capturing the SGE between concentrated brine and seawater was constructed, and the correlation coefficients in the model were experimentally determined. Based on the model, the effects of a single variable (concentration, flow rate, temperature, thickness of the compartment, length of the electrode) on the performance of a RED stack are analyzed. The multi-objective optimization method based on the genetic algorithm was further introduced to obtain the optimal solution set, which could achieve the larger net power density and energy conversion efficiency with coordination. The ranges of optimal feed parameters and stack size were also obtained. The optimal flow velocity of the dilute solution and the concentration of the dilute solution are approximately 7.3 mm/s and 0.4 mol/kg, respectively.

Abstract Background The administration of mesenchymal stem cells (MSCs) remains the most promising approach for cardiac repair after myocardial infarct (MI). However, their poor survival and potential in the ischemic environment limit their therapeutic efficacy for heart repair after MI. The purpose of this study was to investigate the influence of FoxC1-induced vascular niche on the activation of octamer-binding protein 4 (Oct4) and the fate of MSCs under hypoxic/ischemic conditions. Methods Vascular microenvironment/niche was induced by efficient delivery of FoxC1 transfection into hypoxic endothelial cells (ECs) or infarcted hearts. MSCs were cultured or injected into this niche by utilizing an in vitro coculture model and a rat MI model. Survival and neovascularization of MSCs regulated by Oct4 were explored using gene transfer and functional studies. Results Here, using gene expression heatmap, we demonstrated that cardiac ECs rapidly upregulated FoxC1 after acute ischemic cardiac injury, contributing to an intrinsic angiogenesis. In vitro, FoxC1 accelerated tube-like structure formation and increased survival of ECs, resulting in inducing a vascular microenvironment. Overexpression of FoxC1 in ECs promoted survival and neovascularization of MSCs under hypoxic coculture. Overexpression of Oct4, a FoxC1 target gene, in MSCs enhanced their mesenchymal-to-endothelial transition (MEndoT) while knockdown of Oct4 by siRNA altering vascularization. In a rat MI model, overexpression of FoxC1 in ischemic hearts increased post-infarct vascular density and improved cardiac function. The transplantation of adOct4-pretreated MSCs into these ischemic niches augments MEndoT, enhanced vascularity, and further improved cardiac function. Consistently, these cardioprotective effects of FoxC1 was abrogated when Oct4 was depleted in the MSCs and was mimicked by overexpression of Oct4. Conclusions Together, these studies demonstrate that the FoxC1/Oct4 axis is an essential aspect for survival and neovascularization of MSCs in the ischemic conditions and represents a potential therapeutic target for enhancing cardiac repair.

Abstract We evaluate the topographic distribution of diffuse midline gliomas and hemispheric high-grade gliomas in children with respect to their normal gene expression patterns and pathologic driver mutation patterns. We identified 19 pediatric patients with diffuse midline or high-grade glioma with preoperative MRI from tumor board review. 7 of these had 500 gene panel mutation testing, 11 patients had 50 gene panel mutation testing and one 343 gene panel testing from a separate institution were included as validation set. Tumor imaging features and gene expression patterns were analyzed using Allen Brain Atlas. Twelve patients had diffuse midline gliomas and seven had hemispheric high-grade gliomas. Three diffuse midline gliomas had the K27M mutation in the tail of histone H3 protein. All patients undergoing 500 gene panel testing had additional mutations, the most common being in ACVR1, PPM1D, and p53. Hemispheric high-grade gliomas had either TP53 or IDH1 mutation and diffuse midline gliomas had H3 K27M-mutation. Gene expression analysis in normal brains demonstrated that genes mutated in diffuse midline gliomas had higher expression along midline structures as compared to the cerebral hemispheres. Our study suggests that topographic location of pediatric diffuse midline gliomas and hemispheric high-grade gliomas correlates with driver mutations of tumor to the endogenous gene expression in that location. This correlation suggests that cellular state that is required for increased gene expression predisposes that location to mutations and defines the driver mutations within tumors that arise from that region.

On June 9，2020，an earthquake measuring ML3.4 occurred in Shapotou District，Zhongwei City，Ningxia. The earthquake occurred in the meizoseismal area of the 1709 M7½ earthquake of South Zhongwei，and its epicenter is located in an area where there were relatively few weak earthquakes in the past. In this paper，based on the waveform records of the Ningxia regional seismic network，the focal mechanism solution and focal centroid depth of the Zhongwei ML3.4 earthquake on June 9，2020 are inversed using the gCAP method. The focal mechanism solution is also calculated by the Hash method，and the central focal mechanism of these two methods is obtained. The result shows that the focal mechanism solution of gCAP method is strike 255°，dip 79°，rake −20° for nodal plane I，and strike 348°，dip 70°，rake −160° for nodal plane II，and the centroid depth is 12 km，and that the focal mechanism solution of Hash method is strike 344°，dip 89°，rake 176° for nodal plane I，and strike 74°，dip 86°，rake 1° for nodal plane II. The central focal mechanism solution of the two methods is strike 255°，dip 87°，rake −11° for nodal plane I，and strike 346°，dip 80°，rake −176° for nodal plane II，and the principal stress axis is mainly in NE direction. The minimum 3-D rotation angle between the gCAP method result and the central focal mechanism is relative small，which is 12.09°. Combing the past geologic structure data，we deduces that the main dislocation of Zhongwei ML3.4 earthquake on June 9，2020 is the left-lateral slip，and that the fault is more likely to belong to the nodal plane in NEE direction.

Blizzard is a severe weather-related disaster with significant environmental, economic, and social impacts. What is worse, the blizzard is increasingly frequent along with climate change. To enhance resilience, it is important to accurately estimate the blizzards. While some studies have reported the blizzard formation and characteristics, limited studies have not well presented the backflow blizzard associated with the cold vortex in Northeast China. In order to overcome this research gap, this study aims to analyze the characteristics of a backflow blizzard in Liaoning, China, and reveal the reasons behind the spatial heterogeneity of snowstorm intensity and duration. With observation data and mesoscale numerical simulation, this study discovered that the northeast cold vortex was the combined results of airflows from the Sea of Japan transported by the easterly airflow, the East China Sea, and the Yellow Sea transported by the southeast airflow, and the low-level southwest airflow, and the vortex was an important driver to the blizzard. Results further indicate that the interactions of airflow movement, water vapor variation, and frontogenesis occurrence at different layers caused the differences in snowfall intensity, duration, and volume at Zhangwu and Huanren stations. In particular, at Zhangwu station, there was an interaction of warm-wet and cold-dry airflows, but low-layer water vapor content was small, under which background only the cold air pushed the warm air as the southwest wind did not increase significantly, leading warm-humid air to be forced to climb and resulting in frontogenesis at 700–925 hPa. Therefore, the situation of no frontogenesis and poor humidity conditions in the low-level cold air contributed to the weak and a short-duration of snowfall at Zhangwu Station. The water vapor content of warm-wet and cold-wet airflows over Huanren station was better than that at Zhangwu so that during cold and warm air interaction, there was frontogenesis at 500–925 hPa. Near the top of the inverted trough in the ground layer, there was a convergence of the southeasterly wind and the northeasterly winds, resulting in the occurrence of horizontal frontogenesis throughout the layer of 500 hPa and forming an upward movement column. Moreover, a deep near-saturated water vapor layer formed below 600 hPa due to the upward movement column. Overall, the abundant water vapor combined with the deep dynamic uplift led to the heavy and long duration of snowfall at the Huanren station. Overall, this study is an important reference for understanding the backflow blizzard and its mechanism. Moreover, it is conducive to the accurate estimation of backflow blizzards in Northeast China and eastern China, the eastern coast of the Korean peninsula, and other similar areas where on the east part of the continent is the ocean.

Senecavirus A (SVA) is an emerging picornavirus. Its genome is one positive-sense, single-stranded RNA. The viral protein (VPg) is covalently linked to the extreme 5′ end of the SVA genome. A complex hairpin-pseudoknot-hairpin (HPH) RNA structure was computationally predicted to form at the 5′ end of the SVA genome. A total of three extra “U” residues (UUU) served as a linker between the HPH structure and the VPg, causing putative UUU–HPH formation at the extreme 5′ end of the SVA genome. It is unclear how the UUU–HPH structure functions. One SVA cDNA clone (N0) was constructed previously in our laboratory. Here, the N0 was genetically tailored for reconstructing a set of 36 modified cDNA clones (N1 to N36) in an attempt to rescue replication-competent SVAs using reverse genetics. The results showed that a total of nine viruses were successfully recovered. Out of them, five were independently rescued from the N1 to N5, reconstructed by deleting the first five nucleotides (TTTGA) one by one from the extreme 5′ end of N0. Interestingly, these five viral progenies reverted to the wild-type or/and wild-type-like genotype, suggesting that SVA with an ability to repair nucleotide defects in its extreme 5′ end. The other four were independently rescued from the N26 to N29, containing different loop-modifying motifs in the first hairpin of the HPH structure. These four loop-modifying motifs were genetically stable after serial passages, implying the wild-type loop motif was not a high-fidelity element in the first hairpin during SVA replication. The other genetically modified sequences were demonstrated to be lethal elements in the HPH structure for SVA recovery, suggesting that the putative HPH formation was a crucial cis-acting replication element for SVA propagation.

Anthocyanins are water-soluble plant pigments mainly stored in the plant vacuoles. Glutathione S-transferases (GSTs) are a multifunctional enzyme family, which can regulate substance metabolism and biological and abiotic stresses in plants. However, few reports were focused on the involvement of GSTs in anthocyanin sequestration in red skin radish. Here, we identified a glutathione S-transferase gene RsGSTF12 that played roles in anthocyanin sequestration in radish. The bioinformatics analysis revealed that RsGSTF12 belonged to the phi (F) class of glutathione S-transferases and showed a high homology with AtGSTF12, followed by AtGSTF11. The subcellular localization assay showed that RsGSTF12 was located in the endoplasmic reticulum and tonoplast. Temporal and spatial gene expression-specific analyses uncovered a strong correlation of RsGSTF12 with anthocyanin accumulation in radish sprouts. The anthocyanin solubility assay found RsGSTF12 was capable of improving cyanidin water solubility in vitro. Transiently expressing RsGSTF12 in radish cotyledons was able to increase their anthocyanin sequestrations. Furthermore, the functional complementation and overexpression of the Arabidopsis thaliana tt19 mutant and wild type demonstrated that RsGSTF12 might play an indispensable role in anthocyanin accumulation in radish. Taken together, we provide compelling evidence that RsGSTF12 functions critically in how anthocyanins are sequestrated in radish, which may enrich our understanding of the mechanism of anthocyanin sequestration.

Strain observation is the most intuitive observation equipment to monitor stress change in the crust. Strain equipment near the epicenter area of the Jiuzhaigou earthquake (JZGEQ) provides a lot of reliable pre-seismic and post-seismic data for the study of stress change. In this study, the load/unload response ratio (LURR) method is used to study the stress state of rocks by calculating the ratio of strain observation during the loading phase and unloading phase. Results show that the LURR method based on strain observation is an effective method to describe the dynamic change of the constitutive relationship of the rocks in the crust. Different from the strain observation, the LURR anomaly evolution process is more continuous regardless of the time series curve or spatial and temporal distribution characteristics. The LURR curves of different stations begin to increase above 1.0 gradually from 6 months to 1 year prior to the occurrence of the JZGEQ, reaching the maximum value in 1–3 months before the JZGEQ, and subsequently return to a low level. The maximum value of the LURR anomalies decreases with the distance from the epicenters. At the same time, the spatial and temporal evolution characteristics of the LURR anomalies show us the process of “extension—enhance—weaken” in the epicenter of JZGEQ and its peripheral area prior to the earthquake. The concentration areas of the aforementioned LURR anomalies are all distributed in the pre-seismic normal stress-loading zone, which indicates that the faults are in the process of decoupling, and microfracture may exist in the stage of rock dilatancy.

Abstract Background Elongated rostra play an important role in the egg-laying of weevils, and its emergence plays a key role in the adaptive radiation of weevils. Eucryptorrhynchus scrobiculatus Motschulsky and E. brandti Harold co-occur on the same only host Ailanthus altissima, while their oviposition sites are different. In order to understand the adaptation between the rostra of the two weevils and their oviposition sites, the structural differentiation of the rostra in E. scrobiculatus and E. brandti was compared. Results The present study reveals that: (1) The rostra length of E. scrobiculatus and E. brandti was found to be correlated with body size, larger weevils have a correspondingly longer rostrum. The increase of rostra length may be a byproduct of larger weevils. (2) There were significant differences in the external shape of the two rostra, especially the shape of the mandibles of the mouthparts at the apex of the rostra used to excavate an oviposition cavity. (3) There was no difference in the size of the abductor muscles that control the extension of the mandibles, but there were significant differences in the size of the adductor muscles that control the contraction of the mandibles. Conclusions These structural differences reflect the functional potential ovipositional tactics of rostra, which is considered to be a response to the ecological demands of egg deposition, and also provide new insights into the coexistence of two weevil species in the same host A. altissima.

Highlights An artificial solid–electrolyte interface composed of a perovskite type material, BaTiO3, is introduced to Zn anode surface in aqueous zinc ion batteries. The BaTiO3 layer endowing inherent character of the switched polarization can regulate the interfacial electric field at anode/electrolyte interface. Zn dendrite can be restrained, and Zn metal batteries based on BaTiO3 layer show stable cycling. Abstract Aqueous zinc ion batteries show prospects for next-generation renewable energy storage devices. However, the practical applications have been limited by the issues derived from Zn anode. As one of serious problems, Zn dendrite growth caused from the uncontrollable Zn deposition is unfavorable. Herein, with the aim to regulate Zn deposition, an artificial solid–electrolyte interface is subtly engineered with a perovskite type material, BaTiO3, which can be polarized, and its polarization could be switched under the external electric field. Resulting from the aligned dipole in BaTiO3 layer, zinc ions could move in order during cycling process. Regulated Zn migration at the anode/electrolyte interface contributes to the even Zn stripping/plating and confined Zn dendrite growth. As a result, the reversible Zn plating/stripping processes for over 2000 h have been achieved at 1 mA cm−2 with capacity of 1 mAh cm−2. Furthermore, this anode endowing the electric dipoles shows enhanced cycling stability for aqueous Zn-MnO2 batteries. The battery can deliver nearly 100% Coulombic efficiency at 2 A g−1 after 300 cycles.

Abnormal CSF accumulation in the brain can lead to hydrocephalus. The mechanisms regulating CSF clearance during early development are unclear. Here, the authors show that NKCC1 regulates the clearance of both CSF K+ and fluid volume through the choroid plexus during postnatal development in mice.

In 2020, an unexpectedly large outbreak of the coronavirus disease 2019 (COVID-19) epidemic was reported in mainland China. As we known, the epidemic was caused by imported cases in other provinces of China except for Hubei in 2020. In this paper, we developed a differential equation model with tracing isolation strategy with close contacts of newly confirmed cases and discrete time imported cases, to perform assessment and risk analysis for COVID-19 outbreaks in Tianjin and Chongqing city. Firstly, the model behavior without imported cases was given. Then, the real-time regeneration number in Tianjin and Chongqing city revealed a trend of rapidly rising, and then falling fast. Finally, sensitivity analysis demonstrates that the earlier with Wuhan lock-down, the fewer cases in these two cities. One can obtain that the tracing isolation of close contacts of newly confirmed cases could effectively control the spread of the disease. But it is not sensitive for the more contact tracing isolation days on confirmed cases, the fewer cases. Our investigation model could be potentially helpful to provide model building technology for the transmission of COVID-19.

Senecavirus A (SVA) is an emerging virus that belongs to the genus Senecavirus in the family Picornaviridae. Its genome is a positive-sense and single-stranded RNA, containing two untranslated regions (UTRs). The 68-nt-long 3′ UTR is computationally predicted to possess two higher-order RNA structures: a kissing-loop interaction and an H-type-like pseudoknot, both of which, however, cannot coexist in the 3′ UTR. In this study, we constructed 17 full-length SVA cDNA clones (cD-1 to -17): the cD-1 to -7 contained different point mutations in a kissing-loop-forming motif (KLFM); the cD-8 to -17 harbored one single or multiple point mutations in a pseudoknot-forming motif (PFM). These 17 mutated cDNA clones were independently transfected into BSR-T7/5 cells for rescuing recombinant SVAs (rSVAs), named rSVA-1 to −17, corresponding to cD-1 to −17. The results showed that the rSVA-1, -2, -3, -4, -5, -6, -7, -9, -13, and -15 were successfully rescued from their individual cDNA clones. Moreover, all mutated motifs were genetically stable during 10 viral passages in vitro. This study unveiled viral abilities of tolerating mutations in the computationally predicted KLFM or PFMs. It can be concluded that the putative kissing-loop structure, even if present in the 3′ UTR, is unnecessary for SVA replication. Alternatively, if the pseudoknot formation potentially occurs in the 3′ UTR, its deformation would have a lethal effect on SVA propagation.

ObjectivesTo systematically review, assess the reporting quality of, and discuss improvement opportunities for studies describing machine learning (ML) models for glioma grade prediction.MethodsThis study followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses of Diagnostic Test Accuracy (PRISMA-DTA) statement. A systematic search was performed in September 2020, and repeated in January 2021, on four databases: Embase, Medline, CENTRAL, and Web of Science Core Collection. Publications were screened in Covidence, and reporting quality was measured against the Transparent Reporting of a multivariable prediction model for Individual Prognosis Or Diagnosis (TRIPOD) Statement. Descriptive statistics were calculated using GraphPad Prism 9.ResultsThe search identified 11,727 candidate articles with 1,135 articles undergoing full text review and 85 included in analysis. 67 (79%) articles were published between 2018-2021. The mean prediction accuracy of the best performing model in each study was 0.89 ± 0.09. The most common algorithm for conventional machine learning studies was Support Vector Machine (mean accuracy: 0.90 ± 0.07) and for deep learning studies was Convolutional Neural Network (mean accuracy: 0.91 ± 0.10). Only one study used both a large training dataset (n>200) and external validation (accuracy: 0.72) for their model. The mean adherence rate to TRIPOD was 44.5% ± 11.1%, with poor reporting adherence for model performance (0%), abstracts (0%), and titles (0%).ConclusionsThe application of ML to glioma grade prediction has grown substantially, with ML model studies reporting high predictive accuracies but lacking essential metrics and characteristics for assessing model performance. Indole-3-carbinol (I3C), an important secondary metabolite with strong anti-cancer ability, is widely found in cruciferous plants. Light and phytohormones are one of the most important external and internal signals, respectively, that control the growth, development, and secondary metabolism of the plant life cycle. However, there are few studies about the influence of the blue light and salicylic acid (SA) on the regulation of I3C accumulation. In this study, a negative correlation was found between the content of I3C and SA in different species. Among this, broccoli and Arabidopsis thaliana were chosen for further studies. We observed that blue light treatment increased the accumulation of I3C, and exogenous SA treatment significantly inhibited the accumulation of I3C in broccoli sprouts. Based on the RNA sequence, the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis indicated that blue light promoted the enrichment of differentially expressed genes (DEGs) in plant hormone signal transduction pathways. More specifically, downregulated expression of genes related to SA biosynthesis and upregulated expression of I3C genes related to metabolic pathway were observed under blue light. Taken together, these results suggested that SA negatively regulates blue light-induced I3C accumulation in broccoli sprouts.

Biotic stresses, including fungal infections, result in increased production of flavonoid compounds, including 3-deoxyanthocyanidins (3-DAs), in the leaf tissues of Sorghum bicolor. Our objectives were to determine whether sorghum genotypic variation influenced root flavonoid and 3-DA concentrations and rhizosphere microbial communities and to identify how these relationships were affected by abiotic stress. We evaluated root chemicals and rhizosphere microbiomes of five near-isogenic lines of sorghum before and after a late-season frost. Roots were analyzed for total flavonoids, total phenolics, 3-DA concentrations, and antioxidant activity. Amplicon sequencing of 16S ribosomal RNA genes and internal transcribed spacer regions was performed on rhizosphere soils. Concentrations of luteolinidin (a 3-DA) and total flavonoids differed between several lines before frost; however, these relationships changed after frost. Luteolinidin increased in three lines after frost, whereas total flavonoids decreased in all the lines after frost. Lines that differed in luteolinidin and total flavonoid concentrations before frost were different from those after frost. Rhizosphere community compositions also differed before and after frost but only fungal community compositions differed among sorghum lines. Bacterial community compositions were highly correlated with total flavonoid and luteolinidin concentrations. Furthermore, a greater number of bacterial taxa were correlated with total flavonoids and luteolinidin compared with fungal taxa. Collectively, this study provides evidence that plant genotypic variation influences root flavonoids and rhizosphere community composition and that these relationships are affected by frost. Plant–microbe interactions and secondary metabolite production may be important components to include for selective breeding of sorghum for frost stress tolerance.

Abstract Background The original version of Victorian Institute of Sport Assessment-Patella Questionnaire (VISA-P) is developed in English, and aimed to assess the severity of patellar tendinopathy symptoms. Before used in China, it should be translated to Chinese version. Objectives Our aim is to make a translation/cross-culturally adaption for the VISA-P into simplified Chinese version (VISA-PC). And primarily validate the VISA-PC in Chinese speaking population. Methods The translation process of VISA-P questionnaire into simplified Chinese version (VISP-PC) followed the International recognized guideline. Cross-cultural adaptation was carried out with a clinical measurement study. A total of 128 projects which consisted 33 healthy students, 39 patients with patellar tendinopathy and 56 military students (receive military training as at-risk population) were included into this study. Internal consistency was evaluated with Cronbach’s alpha, and test-retest reliability was assessed with intraclass correlation coefficients (ICCs). Construct validity and floor and ceiling effects were also tested. Results The scores were 95.84 ± 5.97 of healthy group, 91.87 ± 9.03 of at-risk group, 62.49 ± 11.39 of pathological group. There is no ceiling and floor effect of VISA-PC. The Cronbach’s alpha (0.895) and ICC (0.986) values showed good internal consistency and reliability. There were high correlations between VISA-PC and Kujala patellofemoral score (r = 0.721). VISA-PC score also had good correlation with the relevant SF-36 items. Conclusion The VISA-PC was well translated into simplified Chinese version (VISA-PC), which is reliable and valid for Chinese-speaking patients with patellar tendinopathy. Level of evidence II.

Abstract Background Knee osteoarthritis (KOA) seriously reduces quality of life and is a major threat to the health of the middle-aged and elderly. This study aimed to assess the efficacy of Miao crossbow needle therapy vs. acupuncture for KOA therapy. Methods This multicenter, randomized controlled trial was performed at three hospitals between April 2016 and December 2016. The patients were randomized to receive crossbow needle (CN) or acupuncture (AT). All treatments were completed within 46 days. Evaluation of treatment was conducted on the 46th, 62nd, and 77th days. The primary endpoint was change of Western Ontario and McMaster Osteoarthritis Index (WOMAC) score on the 46th day. The secondary endpoints included WOMAC score, the Lysholm knee score, the Japanese Orthopedic Association (JOA) knee score, visual analog scale (VAS), and the MOS 36-item short-form health survey (SF-36), on the 46th, 62nd, and 77th day. Results Finally, data of 301 participants were analyzed for the efficacy of treatment. Abstract Background Rice black-streaked dwarf virus (RBSDV) causes one of the most important rice virus diseases of plants in East Asia. However, molecular mechanism(s)controlling rice resistance to infection is largely unknown. Results In this paper, we showed that RBSDV infection in rice significantly induced nitric oxide (NO) production. This finding was further validated through a genetic approach using a RBSDV susceptible (Nipponbare) and a RBSDV resistant (15HPO187) cultivar. The production of endogenous NO was muchhigher in the 15HPO187 plants, leading to a much lower RBSDV disease incidence. Pharmacological studies showed that the applications of NO-releasingcompounds (i.e., sodium nitroprusside [SNP] and nitrosoglutathione [GSNO]) to rice plants reduced RBSDV disease incidence. After RBSDV infection, the levels of OsICS1, OsPR1b and OsWRKY 45 transcripts were significantly up-regulated by NO in Nipponbare. The increased salicylic acid contents were also observed. After the SNP treatment, protein S-nitrosylation in rice plants was also increased, suggesting that the NO-triggered resistance to RBSDV infection was partially mediated at the post-translational level. Although Osnia2 mutant rice produced less endogenous NO after RBSDV inoculation and showed a higher RBSDV disease incidence, its RBSDV susceptibility could be reduced by SNP treatment. Conclusions Collectively, our genetic and molecular evidence revealed that endogenous NO was a vital signal responsible for rice resistance to RBSDV infection.

To compare the clinical efficacy of gastric bypass in obese patients with T2DM with different BMI. Serum leptin, adiponectin, triglyceride (TG), cholesterol (CHOL) were measured as the indicators to show clinical efficacy after laparoscopic gastric bypass surgery (LRYGB). For patients with high BMI and patients with low BMI, the therapeutic effect of LRYGB surgical diabetes is more significant. The postoperative remission rate of diabetes in the high BMI group was not correlated with the preoperative lipid metabolism index but was positively correlated with the postoperative lipid metabolism index CHOL, TG, leptin, adiponectin. The postoperative remission rate of diabetes in the low BMI group was positively correlated with the preoperative abnormal lipid metabolism of the patients, and positively correlated with the postoperative remission of leptin and adiponectin, but was not correlated with the postoperative remission of total CHOL and TG. The increase of serum adiponectin level and the decrease of leptin resistance after LRYGB surgery restored the metabolic balance of leptin and adiponectin, improved insulin resistance (IR), and thus improved blood glucose level. Therefore, LRYGB has a definite therapeutic effect on obese patients with T2DM, and elevated adiponectin and improved leptin resistance are some of the mechanisms of surgical treatment of diabetes.

Cadmium (Cd) is one of the toxic heavy metals in soil, which not only suppresses crop production but also threatens human health. In this study, we aim to clarify the biological function of Cd-related gene BcHIPP16, so as to provide potential genetic solutions to decrease the Cd levels of pak choi. Tissue expression analysis showed that BcHIPP16 expressed in almost all the plant bodies. The transcriptional level of BcHIPP16 in roots was higher than that in shoots, which was significantly induced by copper (Cu) deficiency and Cd exposure conditions. Subcellular localization revealed that BcHIPP16 localized in plasma membrane. Expressing BcHIPP16 in yeast cells improved the sensitivity to Cu and Cd and improved their accumulation in yeast. Furthermore, the Cu and Cd content of Arabidopsis seedlings were increased and complemented, respectively when expressing BcHIPP16 in wild type (WT) and hip16 mutants. Non-invasive Micro-test Technology (NMT) was used to measure the real-time Cd2+ influx from the root surface of BcHIPP16 transgenic Arabidopsis lines, and the result demonstrated that BcHIPP16 promoted Cd2+ influx into Arabidopsis root cells. Taken together, our study showed that BcHIPP16 contributed to absorbing nutrient metal Cu and heavy metal Cd in planta.

Hollow nanocone arrays are fabricated by a low-cost and efficient colloidal lithography (CL) technique. The hollow nanocone arrays are then reversed to make only the tips contact the substrate. The optical properties of the obverse and inverse hollow nanocone arrays are determined by the surrounding environment, showing different reflection spectra and structure dependence. The inverse hollow nanocone arrays show a relative index sensitivity of 70% per RIU with strict linearity. The fluorescence of fluorophore or staining cells can be facilely enhanced by placing them on the tips of the hollow nanocone arrays, while having no quenching effect. The study of the obverse and inverse hollow nanocone arrays can benefit the understanding of the effect of the environment on the plasmonic resonances. The hollow nanocone arrays are promising to serve as high-performance plasmonic sensors and versatile substrates for surface-enhanced fluorescence imaging.

The beet webworm (Loxostege sticticalis L.) is an important agricultural pest and can tolerate harsh environmental conditions by entering diapause. The diapause mechanism of beet webworm is unknown. Therefore, we conducted a transcriptomic study of the process from diapause induction to diapause release in beet webworms. The results revealed 393 gene modules closely related to the diapause of beet webworm. The hub gene of the red module was the HDACI gene, which acts through histone deacetylase (HDAC) enzymes. HDAC enzyme activity was regulated by the light duration and influenced the JH content under induced beet webworm diapause conditions (12 h light:12 h dark). In addition, transcriptomic data suggested that circadian genes may not be the key genes responsible for beet webworm diapause. However, we showed that the photoperiod affects HDAC enzyme activity, and HDAC can regulate the involvement of JH in beet webworm diapause. This study provided a new module for studying insect diapause and links histone acetylation and diapause at the transcriptome level.

Abstract Rice stripe virus (RSV) causes one of the most important rice virus diseases of plants in East Asia. However, the molecular mechanisms controlling rice resistance to RSV infection are largely unknown. Recently, several studies presented a novel model that melatonin (MT) and nitric oxide (NO) participate in the plant-pathogen interaction in a synergetic manner. In this study, there was a difference in MT content between two rice varieties that correlated with one being susceptible and one being resistant to RSV, which suggested that MT is related to RSV resistance. In addition, a test with two NO biosynthesis inhibitors revealed that NO inhibitor were able to increase the disease incidence of RSV. A pharmacological experiment with exogenous MT and NO showed that increased MT and NO in the MT-pretreated plants led to lower disease incidences; however, only NO increased in a NO-releasing reagent [sodium nitroprusside (SNP)] pretreated plants. The expressions level of OsPR1b and OsWRKY 45 were significantly induced by MT and NO. These results suggest that rice resistance to RSV can be improved by increased MT through a NO-dependent pathway.

Human error has been statistically proven to be the primary cause of road accidents. This undoubtedly is a contributory cause of the rising popularity of autonomous vehicles as they are presumably able to maneuver appropriately/optimally on the roads while diminishing the likelihood of human error and its repercussion. However, autonomous vehicles are not ready for widespread adoption because their safety and security issues are yet to be thoroughly investigated/addressed. Little literature could be found on collaborative analysis of safety and security of autonomous vehicles. This paper proposes a framework for analyzing both safety and security issues, which includes an integrated safety and security method (S&S) with international vehicle safety and security standards ISO 26262 and SAE J3061. The applicability of the proposed framework is demonstrated using an example of typical autonomous vehicle model. Using this framework, one can clearly understand the vehicle functions, structure, the associated failures and attacks, and also see the vulnerabilities that are not yet addressed by countermeasures, which helps to improve the in-vehicle safety and security from researching and engineering perspectives.

The negative thermal expansion (NTE) or nearly zero thermal expansion (NZTE) behaviors have been found in some magnetic metallic compounds with great application prospects. However, it is a big challenge for applications that the responding temperature range is not broad enough or far away from room temperature. Herein, a new method is put forward to design the wide temperature range of NTE and NZTE Behavior by NTE/NZTE multi-phase composite. In this method, the optimized volume fraction of the components achieves a concordant collaboration of multi-phase in the counteraction of NTE and positive thermal expansion (PTE) at several adjacent temperature ranges and thus the temperature range is broadened as expected. By this method, MnCuGeN-based NTE composite (290 K ≤ T ≤ 385 K, ΔT = 95 K) and MnCuZnSiGeN-based NZTE composite (200 K ≤ T ≤ 400 K, ΔT = 200 K) with excellent stability have been successfully designed and prepared. This idea will open an avenue to the multi-phase composite with designable NTE/NZTE properties, which would have important applications in aerospace and other modern industries.
