Objective: To determine the role of RNA helicase DDX5 in sorafenib/multi-tyrosine kinase inhibitor (mTKI) response. Sorafenib and mTKIs downregulate DDX5 in vitro and preclinical hepatocellular carcinoma (HCC) models. In turn, sorafenib-mediated DDX5 downregulation activates Wnt/β-catenin and non-canonical NF-κB signaling, resulting in ferroptosis escape and mTKI resistance. Design and Results: Molecular, pharmacologic and bioinformatic approaches were employed in human HCC cell lines, preclinical HCC models, and HCCs from TCGA. Earlier studies linked sorafenib effectiveness to ferroptosis. Herein we demonstrate sorafenib/mTKIs downregulate DDX5 in vitro and in vivo. To understand the effect of DDX5 downregulation, we compared TCGA-derived HCCs expressing low vs. high DDX5 focusing on ferroptosis-related genes. Glutathione Peroxidase 4 (GPX4), a key ferroptosis regulator, was significantly overexpressed in DDX5LOW HCCs. Importantly, DDX5-knockdown (DDX5KD) HCC cell lines lacked lipid peroxidation by GPX4 inhibition, indicating DDX5 downregulation suppresses ferroptosis. RNAseq of wild type vs. DDX5KD cells untreated or treated with sorafenib, identified a unique set of genes repressed by DDX5 and upregulated by sorafenib. This set significantly overlaps genes from Wnt/β-catenin and non-canonical NF-κB pathways, including NF-κB inducing Kinase required for non-canonical NF-κB activation. Pharmacologic inhibition of these pathways in combination with sorafenib reduced DDX5KD cell viability. Mechanistically, sorafenib-mediated NIK expression induced NRF2 transcription, while DDX5KD extended NRF2 half/life by stabilizing p62/SQSTM1, enhancing GPX4 expression and ferroptosis escape. Conclusion: Sorafenib/mTKI-mediated DDX5 downregulation results in adaptive mTKI resistance by enhancing NRF2 expression, leading to ferroptosis escape. We propose inhibition of the pathways leading to NRF2 expression will enhance the therapeutic effectiveness of sorafenib/mTKIs.

Rationale: RNA helicase DDX5 is downregulated during hepatitis B virus (HBV) replication, and poor prognosis HBV-related hepatocellular carcinoma (HCC). The aim of this study is to determine the mechanism and significance of DDX5 downregulation for HBV-driven HCC, and identify biologics to prevent DDX5 downregulation. Methods: Molecular approaches including immunoblotting, qRT-PCR, luciferase transfections, hepatosphere assays, Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq), and RNA-seq were used with cellular models of HBV replication, HBV infection, and HBV-related liver tumors, as well as bioinformatic analyses of liver cancer cells from two independent cohorts. Results: We demonstrate that HBV infection induces expression of the proto-oncogenic miR17~92 and miR106b~25 clusters which target the downregulation of DDX5. Increased expression of these miRNAs is also detected in HBV-driven HCCs exhibiting reduced DDX5 mRNA. Stable DDX5 knockdown (DDX5KD) in HBV replicating hepatocytes increased viral replication, and resulted in hepatosphere formation, drug resistance, Wnt activation, and pluripotency gene expression. ATAC-seq of DDX5KD compared to DDX5 wild-type (WT) cells identified accessible chromatin regions enriched in regulation of Wnt signaling genes. RNA-seq analysis comparing WT versus DDX5KD cells identified enhanced expression of multiple genes involved in Wnt pathway. Additionally, expression of Disheveled, DVL1, a key regulator of Wnt pathway activation, was significantly higher in liver cancer cells with low DDX5 expression, from two independent cohorts. Importantly, inhibitors (antagomirs) to miR17~92 and miR106b~25 restored DDX5 levels, reduced DVL1 expression, and suppressed both Wnt activation and viral replication. Conclusion : DDX5 is a negative regulator of Wnt signaling and hepatocyte reprogramming in HCCs. Restoration of DDX5 levels by miR17~92 / miR106b~25 antagomirs in HBV-infected patients can be explored as both antitumor and antiviral strategy.

We demonstrate that DDX5 regulates STAT1 mRNA translation by resolving a G-quadruplex (rG4) RNA structure, proximal to the 5′ end of STAT1 5′UTR. We employed luciferase reporter assays comparing wild type (WT) versus mutant rG4 sequence, rG4-stabilising compounds, CRISPR/Cas9 editing of the STAT1-rG4 sequence and circular dichroism determination of the rG4 structure. STAT1-rG4 edited cell lines were resistant to the effect of rG4-stabilising compounds in response to IFN-α, while HCC cell lines expressing low DDX5 exhibited reduced IFN response. Ribonucleoprotein and electrophoretic mobility assays demonstrated direct and selective binding of RNA helicase-active DDX5 to the WT STAT1-rG4 sequence. Immunohistochemistry of normal liver and liver tumours demonstrated that absence of DDX5 corresponded to absence of STAT1. Significantly, knockdown of DDX5 in HBV infected HepaRG cells reduced the anti-viral effect of IFN-α.ConclusionRNA helicase DDX5 resolves a G-quadruplex structure in 5′UTR of STAT1 mRNA, enabling STAT1 translation. We propose that DDX5 is a key regulator of the dynamic range of IFN response during innate immunity and adjuvant IFN-α therapy.

TNF-related apoptosis-inducing ligand (TRAIL) possesses the capacity to induce apoptosis in a wide variety of tumor cells without affecting most normal cells. However, it has now emerged that many primary cancer cells are resistant to TRAIL monotherapy. Overcoming the intrinsic or acquired TRAIL resistance is desirable for TRAIL-mediated cancer therapy. In this study, we found that the miR-221/222 cluster was up-regulated in TRAIL-resistant liver cancer cells. Specific inhibitors of miR-221 and/or miR-222, called sponge, TuD and miR-Zip were constructed, and their ability to overcome TRAIL resistance was compared. Among them, AAV-mediated gene therapy using co-expression of TRAIL with miR-221-Zip showed the most synergistic activity in the induction of apoptosis in vitro. In vivo treatment of nude mice bearing human TRAIL-resistant liver cancer xenografts with AAV-TRAIL-miR-221-Zip also led to growth inhibition. This sensitizing effect of miR-221-Zip was associated with increased expression of PTEN, the miR-221 target, as well as with decreasing levels of Survivin. Moreover, miR-221 expression was concomitant with promotion of Survivin expression and suppression of PTEN expression. TRAIL sensitivity of cancer cells isolated from liver cancer tissues or from patients was significantly correlated with miR-221 expression. And miR-221 blood expression levels in liver cancer patients were correlated with TRAIL sensitivity, thus it had the potential to be a predictor of TRAIL sensitivity in liver cancer. These data suggested the potential of combining AAV-TRAIL with miR-221-Zip as a therapeutic intervention for liver cancer.

Chronic infections can lead to carcinogenesis through inflammation-related mechanisms. Chronic infection of the human gastric mucosa with Helicobacter pylori is a well-known risk factor for gastric cancer. However, the mechanisms underlying H. pylori-induced gastric carcinogenesis are incompletely defined. We aimed to screen and clarify the functions of long noncoding RNAs (lncRNAs) that are differentially expressed in H. pylori-related gastric cancer. We found that lncRNA SNHG17 was upregulated by H. pylori infection and markedly increased the levels of double-strand breaks (DSBs). SNHG17 overexpression correlated with poor overall survival in patients with gastric cancer. The recruitment of NONO by overabundant nuclear SNHG17, along with the role of cytoplasmic SNHG17 as a decoy for miR-3909, which regulates Rad51 expression, shifted the DSB repair balance from homologous recombination toward nonhomologous end joining. Notably, during chronic H. pylori infection, SNHG17 knockdown inhibited chromosomal aberrations. Our findings suggest that spatially independent deregulation of the SNHG17/NONO and SNHG17/miR-3909/RING1/Rad51 pathways upon H. pylori infection promotes tumorigenesis in gastric cancer by altering the DNA repair system, which is critical for the maintenance of genomic stability. Upregulation of SNHG17 by H. pylori infection might be an undefined link between cancer and inflammation.

Renal fibrosis represents a final common outcome of many renal diseases and has attracted a great deal of attention. To better understand whether lncRNAs could be a player in this process or be a biomarker for renal fibrosis diagnosis, we compared transcriptome sequencing data on renal tissues and urine respectively between UUO (unilateral ureteral obstruction) and shamed (Sham) rat model. Numerous genes including lncRNAs with significant changes in their expression were identified. 24 lncRNAs were up-regulated and 79 lncRNAs were down-regulated in the renal tissues of the UUO rats. 625 lncRNAs were up-regulated and 177 lncRNAs were down-regulated in urines of the UUO rats. Among the lncRNAs upregulated in renal tissue of UUO rats, 19 lncRNAs were predicted containing several conserved Smad3 binding motifs in the promoter. Among them, lncRNAs with putative promoter containing more than 4 conserved Smad3 binding motifs were demonstrated to be induced by TGF-β significantly in normal rat renal tubular epithelial NRK-52E cells. We further confirmed that lncRNA TCONS_00088786 and TCONS_01496394 were regulated by TGF-β stimulation and also can influence the expression of some fibrosis-related genes through a feedback loop. Based on transcriptome sequencing data, bioinformatics analysis and qRT-PCR detection, we also demonstrated lncRNA in urine are detectable and might be a novel biomarker of renal fibrosis. These results provide new information for the involvement of lncRNAs in renal fibrosis, indicating that they may serve as candidate biomarkers or therapeutic targets in the future.

Fatty acid binding protein 3 (H-FABP, FABP3) has been significantly associated with intramuscular fat (IMF) content in pigs, which is positively correlated with palatability of pork. However, its underlying function is not fully elucidated. We have investigated the effects of overexpression of the FABP3 gene on differentiation and adipogenesis of 3T3-L1 preadipocytes in the fat Banna mini-pig inbred line (fBMIL). Eukaryotic vectors that expressed the FABP3 protein were constructed, and stably established in the 3T3-L1 preadipocytes cell line. Cells were induced in a standard differentiation cocktail. Morphological changes and the degree of adipogenesis were measured by Oil Red O staining assay and triacylglycerol content measurement, respectively. mRNA expression levels of triacylglycerol metabolism-related genes were measured by qPCR. FABP3 significantly promoted differentiation of 3T3-L1 cells and enhanced triacylglycerol levels (P < 0.05). mRNA of the peroxisome proliferator-activated receptor γ (PPARγ), adipocyte fatty acid binding protein (422/aP2) and glycerol-3-phosphate dehydrogenase (GPDH) gene increased markedly (P < 0.05). In conclusion, expression of the FABP3 gene enhances adipogenesis in 3T3-L1 preadipocytes primarily by upregulating lipogenic PPARγ, 422/aP2 and GPDH genes.

Construction and characterization of a full-length infectious clone (pMEV) of mink enteritis virus are described. Feline kidney cells (F81) were transfected with pMEV containing an engineered BamHI site that served as a genetic marker. The rescued virus was indistinguishable from its parental virus. The availability of a MEV infectious clone will facilitate studies of viral replication and pathogenicity and will permit the elucidation of determinants of the host range of the parvovirus.

As the focus on developing new polymer electrolytes continues to intensify in the area of alternative energy conversion and storage devices, the rational design of polyelectrolytes with high single ion transport rates has emerged as a primary strategy for enhancing device performance. Previously, we reported a series of sulfonate based copolymer ionomers based on using mixed bulky quaternary ammonium cations and sodium cations as the ionomer counterions. This led to improvements in the ionic conductivity and an apparent decoupling from the Tg of the ionomer. In this article, we have prepared a new series of ionomers based on the homopolymer of poly(2-acrylamido-2-methyl-1-propane-sulfonic acid) using differing sizes of the ammonium counter-cations. We observe a decreasing Tg with increasing the bulkiness of the quaternary ammonium cation, and an increasing degree of decoupling from Tg within these systems. Somewhat surprisingly, phase separation is observed in this homopolymer system, as evidenced from multiple impedance arcs, Raman mapping and SEM. The thermal properties, morphology and the effect of plasticizer on the transport properties in these ionomers are also presented. The addition of 10 wt% plasticizer increased the ionic conductivity between two and three orders of magnitudes leading to materials that may have applications in sodium based devices.

In this study, LiNi0.5Mn1.5O4 (LNMO) nanoparticles were prepared as a 5 V cathode material via a rheological phase method and annealed at different temperatures: 680 °C, 750 °C, and 820 °C. The sample annealed at 750 °C shows the best performance. A room temperature ionic liquid (RTIL) containing 1 M lithium bis(trifluoromethanesulfonyl) imide (LiNTf2) in N-butyl-N-methyl-pyrrolidinium bis(trifluoromethanesulfonyl) imide (C4mpyrNTf2) was used as novel electrolyte in conjunction with the LNMO cathodes and their electrochemical properties have been investigated. The results show that the LNMO using RTIL as electrolyte has better coulombic efficiency and comparable discharge capacities to those of the cells assembled with standard liquid electrolyte (1 M LiPF6 in ethylene carbonate/diethyl carbonate). Electrochemical impedance spectroscopy shows that the RTIL is much more stable as the electrolyte for LiNi0.5Mn1.5O4 than the conventional electrolyte.

Long noncoding RNAs (lncRNAs) are emerging as important regulators in cellular processes, including the development, proliferation, and migration of cancer cells. We have demonstrated in a prior study that small nucleolar RNA host gene 5 (SNHG5) is dysregulated in gastric cancer (GC). To further explore the underlying mechanisms of SNGH5 function in the development of GC, in this study, we screened the microRNAs interacting with SNHG5 and elucidated their roles in GC. We showed that SNHG5 contains a putative miR-32-binding site and that deletion of this site abolishes the responsiveness to miR-32. Suppression of SNHG5 expression by miR-32 was found to be Argonaute (Ago)2-dependent. Immunoprecipitation showed that SNHG5 could be pulled down from the Ago-2 complex with miR-32. Furthermore, it was reported that Kruppel-like factor 4 (KLF4) is a target gene of miR-32. In agreement with SNHG5 being a decoy for miR-32, we showed that KLF4 suppression by miR-32 could be partially rescued by SNHG5 overexpression, whereas miR-32 mimic rescued SNHG5 overexpression-mediated suppression of GC cell migration. In addition, we identified a negative correlation between the expression of SNHG5 and miR-32 in GC tissues. Furthermore, KLF4 expression was significantly downregulated in GC specimens, and a negative correlation between miR-32 and KLF4 expression and a positive correlation between KLF4 and SNHG5 expression levels were detected. Overall, this study demonstrated, for the first time, that the SNHG5/miR-32/KLF4 axis functions as an important player in GC cell migration and potentially contributes to the improvement of GC diagnosis and therapy.-Zhao, L., Han, T., Li, Y., Sun, J., Zhang, S., Liu, Y., Shan, B., Zheng D., Shi, J. Addition of silica nanoparticles functionalised with lithium propane sulfonate to the organic ionic plastic crystal N-ethyl-N-methylpyrrolidinium bis(trifluoromethanesulfonyl)amide ([C2mpyr][NTf2]) results in a significant increase in ionic conductivity. Analysis of these nanocomposites by impedance spectroscopy, NMR, positron annihilation lifetime spectroscopy (PALS) and Raman spectroscopy suggests that this is the result of higher matrix mobility due to an increase in defect size and concentration. The effect of these functionalised nanoparticles is compared to that previously observed for unfunctionalised nanoparticles in the lithium-doped and pure plastic crystal.

Ionic liquid surface treatments are proposed as a method of controlling corrosion processes on magnesium alloys. An important magnesium alloy, ZE41 (nominally 4% Zn and 1% rare earth), was treated with the ionic liquid trihexyl(tetradecyl)phosphonium diphenylphosphate (P66614DPP). Impedance spectra were acquired at intervals during the treatment, indicating the development of a film and allowing a measure of the film formation process to be obtained over time. Mechanically polished and electro-polished surfaces were prepared; these surfaces, treated and untreated, were subsequently exposed to 0.1 M NaCl aqueous solutions. The corrosion behavior of the prepared surfaces were assessed using impedance spectroscopy and optical microscopy. The results indicated a significant role for the method of surface preparation used and, in both cases, the ionic liquid treatment produced a more corrosion-resistant surface.

V2O5 nanomaterials including nanoribbons, nanowires, and microflakes have been synthesized by an ultrasonic assisted hydrothermal method and combined with a post-annealing process. The as-annealed V2O5 nanomaterials are characterized by X-ray diffraction (XRD), Brunauer−Emmett−Teller (BET) N2 adsorption, scanning electron microscopy (SEM), transmission electron microscopy (TEM), and high resolution transmission electron microscopy (HRTEM). A room temperature ionic liquid (RTIL) has been used as an electrolyte ([C3mpyr][NTf2] containing 1 M LiNTf2) in rechargeable lithium metal batteries by combining V2O5 nanomaterials as cathode materials. The electrochemical tests including constant current charge−discharge, cyclic voltammetry (CV), and electrochemical impedance spectroscopy (EIS) show near theoretical specific capacity, improved cyclability, good high-rate capability, and enhanced kinetics. The NMR results indicate that SiO2 and Mg(OH)(2) interact strongly with the anion and cation of each IL through a mechanism of adsorption of the anion and subsequent close proximity of the cation in a surface double layer as observed through H-1-Si-29 cross polarization experiments). In contrast, Al2O3, MgO, ZnO, and ZrO2 appear less active, strongly suggesting the necessity of hydroxylated surface groups ill order to enhance the generation of these interfacial films. Using solid-state NMR to characterize Such interfaces not only has the potential to elucidate mechanisms of wear resistance and corrosion protection via iLs, but is also likely to allow their rapid screening for such durability applications.

In this work lithium modified silica (Li-SiO2) nano-particles were synthesized and used as a single ion lithium conductor source in gel electrolytes. It was found that Li-SiO2 exhibited good compatibility with DMSO, DMA/EC (a mixture of N,N-dimethyl acetamide and ethylene carbonate) and the ionic liquid, N-methyl-N-propyl pyrrolidinium bis(trifluoromethylsulfonyl) amide ([C3mpyr][NTf2]). Several gel electrolytes based on Li-SiO2 were obtained. These gel electrolytes were investigated by DSC, solid state NMR, conductivity measurements and cyclic voltammetry. Conductivities as high as 10−3 S/cm at room temperature were observed in these nano-particle gel electrolytes. The results of electrochemical tests showed that some of these materials were promising for using as lithium conductive electrolytes in electrochemical devices, with high lithium cycling efficiency evident.

Montmorillonites are composed of aluminosilicate layers stacked one above the other, and the layer thickness is approximately 1 nm. In this work lithium modified montmorillonite (Li-MMT) was prepared and used as a lithium macro-anion salt in gel electrolytes. It was found that Li-MMT exhibited good compatibility with poly(ethylene glycol), DMSO and the ionic liquid, 1-ethyl-3-methylimidazolium dicyanamide (EMIdca), and a few of novel gel electrolytes based on Li-MMT were obtained. These gel electrolytes were investigated by X-ray powder diffraction, solid state NMR, conductivity measurements and cyclic voltammetry. High conductivities up to 10− 4 to 10− 3 S/cm at room temperature were observed with these macro-anion gel electrolytes. These gel materials were promising to be used as lithium conductive electrolytes in electrochemical devices, such as lithium batteries.

Zwitterionic compounds such as those based on 1-butylimidazolium-3-(n-butanesulfonate) have previously been shown to have positive effects on the transport properties of polyelectrolytes. The addition of the zwitterion has been found to, in some cases, increase the dissociation of the lithium ion and enhance the conductivity by almost an order of magnitude. In this work, we report the effects of adding the above-mentioned zwitterion into the polyelectrolyte gel system poly(lithium methacrylate-co-N,N-dimethyl acrylamide); the anionic group being a stronger base leads to different behaviour for this copolymer compared to previous work. Polyelectrolyte gels based on dimethyl sulfoxide and polyether solvents were investigated to determine the breadth of applicability of the zwitterion in improving lithium ion transport. Impedance spectroscopy and pulse field gradient-NMR diffusion indicate an increase in the number of available charge carriers with zwitterion addition in some gel systems, however, the effect is not universal.

Novel polymer electrolyte materials based on a polyelectrolyte-in-ionic-liquid principle are described. A combination of a lithium 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPSLi) and N,N′-dimethylacrylamide (DMMA) are miscible with the ionic liquid, 1-ethyl-3-methylimidazolium dicyanamide (EMIDCA). EMIDCA has remarkably high conductivity (≥ 2 · 10−2 S · cm−1) at room temperature and acts as a good solvating medium for the polyelectrolyte. At compositions of AMPSLi less than or equal to 75 mol-% in the copolymer (P(AMPSLi-co-DMAA)), the polyelectrolytes in EMIDCA are homogeneous, flexible elastomeric gel materials at 10 − 15 wt.-% of total polyelectrolyte. Conductivities higher than 8 · 10−3 S · cm−1 at 30 °C have been achieved. The effects of the monomer composition, polyelectrolyte concentration, temperature and lithium concentration on the ionic conductivity have been studied using thermal and conductivity analysis, and pulsed field gradient nuclear magnetic resonance techniques.

Novel polymer-in-ionic liquid electrolytes (PILEs) have been developed for solid state electrochemical actuators based on polypyrrole. The active polymer electrodes are readily oxidized/reduced without degradation in the PILE. It was found that the actuator cycle life is significantly enhanced in the PILE as is the ‘shelf life’ of the device.

High conductivity in single ion conducting polymer electrolytes is still the ultimate aim for many electrochemical devices such as secondary lithium batteries. Achieving effective ion dissociation in these cases remains a challenge since the active ion tends to remain in close proximity to the backbone charge as a result of a low degree of ion dissociation. A unique aspect of this dissociation problem in polyelectrolytes is the repulsion between the backbone charges created by dissociation. It is found in this study that poly-[vinylpyrrolidone-co-(vinyl acetate)] (P(VP-c-VA) in 1-ethyk-3-methylimidazolium bis(trifluoromethyl sulfonyl) amide (EtMeIm + Tf2N - ) and poly(N,N-dimethyl acrylamide) (PDMAA) in trimethyl butyl ammonium bis(trifluoromethane sulfonyl) amide (N + 1114 Tf 2 N - ) produce ion-conducting liquids and gels. The P(VP-c-VA)/ EtMeIm + Tf 2 N - mixture has a conductivity around 10 -3 S . cm -1 at 22°C, for copolymer concentrations up to 30 wt.-%. Thermal analysis shows that the Tg of the PVP(VP-c-VA)/ EtMeIm + Tf 2 N - system is well described by the Fox equation as a function of polymer content. Poly(methyl methacrylate) (PMMA)/ EtMeIm + Tf 2 N - gel electrolytes were prepared by in-situ polymerisation of the monomer in the ionic liquid. In the presence of 0.5 - 2.0 wt.-% of a crosslinking agent, these PMMA-based electrolytes displayed elastromeric properties and high conductivity (ca. 10 -3 S. cm -1 ) at room temperature.

Characterization of a new family of salts, based on a number of 1-alkyl-2-methyl pyrrolinium cations and the bis(trifluoromethane sulfonyl) amide anion (TFSA), is presented. From the thermal analysis, conductivity and X-ray diffraction (XRD) measurements, at least one of the compounds of the family, 1-ethyl-2-methyl pyrrolinium TFSA, was found to exhibit plastic crystal phases before melting and to exhibit high conductivity in the solid state (1×10−4 S cm−1 at 25 °C). This plastic crystal behaviour is discussed in comparison with other members of this pyrrolinium salt family.

A range of low melting salts of the bis(trifluoromethanesulfonyl)amide (TFSA) anion with hindered organic cations (N-methyl-1-methylpyrrolinium, N,N-dimethylpyrrolidinium, N,N,N,-trimethylammonium, and N,N,N,N-tetrakis(n-propyl)ammonium) have been crystallized. Single-crystal X-ray diffraction data show these materials to consist of discrete ions with only weak C−H···O (or for Me3NH N−H···O) contacts between the constituent atoms of the cations and anions close to the limits of van der Waals separations. Consequently, the observed physical properties of these materials presumably result from the diffuse negative charge on the TFSA anion and inefficient packing of these large and irregularly shaped ions.

Novel lithium polyelectrolyte–ionic liquid systems, using poly(lithium 2-acrylamido-2-methyl propanesulfonate) and its copolymer with N-vinyl formamide, have been developed in this work. The ionic liquid used in this work is from a novel family of methanesulfonate ionic liquids, specifically N-hexyl-N,N,N-tributyl ammonium methanesulfonate, which is chosen because of the similarity with the anionic functionality of the polymer. The ionic liquid thereby acts as a good solvating medium for the polyelectrolyte. It was found that the copolymer-based polyelectrolyte–ionic liquid system exhibits two to three times higher conductivity than that of the homopolymer system. The results of solid-state 7Li-NMR have shown that lithium cations in the copolymer system are mobile whereas in the homopolymer, only a fraction appears to be mobile even at 80 °C. This supports the hypothesis that separation of the charged groups on the polymer backbone via the co-monomer encourages the dissociation of lithium cations from the sulfonate groups bonded to the polymer chains, and hence, results in an increase in conductivity of the polyelectrolyte material.

Mink enteritis virus (MEV) is one of the most important pathogens in the mink industry. Recent studies have shed light into the role of microRNAs (miRNAs), small noncoding RNAs of length ranging from 18–23 nucleotides (nt), as critical modulators in the host-pathogen interaction networks. We previously showed that miRNA miR-181b can inhibit MEV replication by repression of viral non-structural protein 1 expression. Here, we report that two other miRNAs (miR-320a and miR-140) inhibit MEV entry into feline kidney (F81) cells by downregulating its receptor, transferrin receptor (TfR), by targeting the 3′ untranslated region (UTR) of TfR mRNA, while being themselves upregulated.

Canine distemper virus (CDV) and rabies virus (RV) are two important pathogens of the dog. CDV, a member of the morbillivirus genus, has shown promise as an expression vector. The glycoprotein from RV is a main contributor to protective immunity and capable of eliciting the production of virus-neutralizing antibodies. In this study, we recovered an attenuated strain of canine distemper virus and constructed a recombinant virus, rCDV-RV-G, expressing a modified (R333Q) rabies virus glycoprotein (RV-G) of RV Flury strain LEP. RV-G expression by the recombinant viruses was confirmed. Furthermore, G was proved to be incorporated into the surface of CDV particles. While replication of the recombinant virus was slightly reduced compared with the parental CDV, it stably expressed the RV-G over ten serial passages. Inoculation of mice induced specific neutralizing antibodies against both RV-G and CDV. Therefore, the rCDV-RV-G has the potential as a vaccine that may be used to control rabies virus infection in dogs and other animals.

It was found in this work that silicotungstic acid hydrate could be mixed with phosphoric acid (H3PO4, 85%) to make a viscous paste material with high conductivity (10−2 S/cm at room temperature). The STA/H3PO4 paste samples were stable at 80°C in the atmosphere, and at 100°C under constant humidity over 10 days. The conductivity behavior of the paste samples has been investigated under various conditions, and it was found to be dependent on temperature, paste composition, and environment humidity.

In this work, two different polymer membrane systems based on Nafion and Teflon were investigated as proton conductors for polymer membrane fuel cells. Water-free Nafion117 membranes swollen with different non-aqueous solvents were prepared. The solvents included imidazole, imidazole–imidazolium salt solutions, room temperature molten salts and molten salt–acid solutions. Teflon films were treated with a surfactant, or a Nafion solution, to improve their surface properties, and were subsequently swollen with phosphoric acid. Conductivity measurements were carried out on both the Nafion and Teflon membranes. Conductivities in the range of 10−3 S cm−1 at around 100°C were obtained. This is still an order of magnitude lower than the corresponding water swollen Nafion at 80°C.

The free volume behaviour of the polyacrylonitrile/lithium triflate system is investigated over the composition range 0–75 wt % salt. The addition of salt, up to 45 wt %, to the PAN polymer substantially increases the free volume as measured by the orthopositronium pickoff lifetime, τ3. Beyond this salt concentration (i.e., 45–70 wt %) the free volume remains approximately constant. This constant free volume region corresponds to a region of high ionic conductivity in the glassy state, making these materials polymer-based fast ion conductors, that is, having a decoupling ratio Rτ ≫1. The high salt content in these fast ion conductors results in a high susceptibility to polar solvents such as water. For all compositions, water absorption results in a free volume increase attributed to plasticization; however, in the fast ion conducting region, a significantly larger free volume response due to plasticization is measured and may be connected to a percolation morphology in these samples. Salt addition is shown to lower the Tg, as measured by positron annihilation lifetime spectroscopy (PALS). Tg is 115°C for PAN and 85°C for 66 wt % lithium triflate. © 2000 John Wiley & Sons, Inc. Mink enteritis virus (MEV) is one of the most important viral pathogens in the mink industry. Recent studies have showed that microRNAs (miRNAs), small noncoding RNAs of length ranging from 18–23 nucleotides (nt) participate in host-pathogen interaction networks; however, whether or not miRNAs are involved in MEV infection has not been reported. Our study revealed that miRNA miR-181b inhibited replication of MEV in the feline kidney (F81) cell line by targeting the MEV non-structural protein 1 (NS1) messenger RNA (mRNA) coding region, resulting in NS1 translational repression, while MEV infection reduced miR-181b expression. This is the first description of cellular miRNAs modulating MEV infection in F81 cells, providing further insight into the mechanisms of viral infection, and may be useful in development of naturally-occurring miRNAs antiviral strategies.

Fatty acid binding protein 3 (H-FABP, FABP3) has been significantly associated with intramuscular fat (IMF) content in pigs, which is positively correlated with palatability of pork. However, its underlying function is not fully elucidated. We have investigated the effects of overexpression of the FABP3 gene on differentiation and adipogenesis of 3T3-L1 preadipocytes in the fat Banna mini-pig inbred line (fBMIL). Eukaryotic vectors that expressed the FABP3 protein were constructed, and stably established in the 3T3-L1 preadipocytes cell line. Cells were induced in a standard differentiation cocktail. Morphological changes and the degree of adipogenesis were measured by Oil Red O staining assay and triacylglycerol content measurement, respectively. mRNA expression levels of triacylglycerol metabolism-related genes were measured by qPCR. FABP3 significantly promoted differentiation of 3T3-L1 cells and enhanced triacylglycerol levels (P0.05). mRNA of the peroxisome proliferator-activated receptor γ (PPARγ), adipocyte fatty acid binding protein (422/aP2) and glycerol-3-phosphate dehydrogenase (GPDH) gene increased markedly (P0.05). In conclusion, expression of the FABP3 gene enhances adipogenesis in 3T3-L1 preadipocytes primarily by upregulating lipogenic PPARγ, 422/aP2 and GPDH genes.

The use of ionic liquids as additives to base oil for the lubrication of steel on aluminum was investigated. The miscibility and wear performance of various phosphonium, imidazolium, and pyrrolidinium ionic liquids in a range of polar and nonpolar base oils was determined. The structure and ion pairing of the ionic liquids was found to be important in determining their miscibility in the base oils. In wear tests, some of the miscible base oil/IL blends reduced the aluminum wear depth when compared to that found with the base oil alone. The nonpolar base oil/IL blends were able to withstand higher wear-test loads than the polar base oil/IL blends. At 10 N, as little as 0.01 mol/kg of IL, or 0.7-0.9 wt %, in the nonpolar base oils was enough to drastically reduce the wear depth on the aluminum. XPS analysis of the wear surfaces suggested that the adsorbing of the IL to the surface, where it can form low-shear layers and also react to form tribofilms, is important in reducing friction and wear. The largest reductions in wear at the highest load tested were found for a mineral oil/P6,6,6,14 (i)(C8)2PO2 blend.

MicroRNAs (miRNAs) are small regulatory RNAs that play a significant role in eukaryotes by targeting mRNAs for cleavage or translational repression. Recent studies have also shown them to be associated with cellular changes following viral infection. Mink enteritis virus (MEV) is one of the most important viral pathogens in the mink industry. To study the involvement of miRNAs in the MEV infection process, we used Illumina's ultrahigh throughput approach to sequencing miRNA libraries from the feline kidney (F81) cell line before and after infection with MEV. Using this bioinformatics approach we identified 196 known mammalian miRNA orthologs belonging to 152 miRNA families in F81 cells. Additionally, 97 miRNA*s of these miRNAs were detected. As well as known miRNAs, 384 and 398 novel miRNA precursor candidates were identified in uninfected and MEV-infected F81 cells respectively that have not been reported in other mammals. In MEV-infected cells 3 miRNAs were significantly down-regulated and 4 up-regulated including 3 significantly. The majority (12 of 16) of randomly selected miRNA expression profiles by qRT-PCR were consistent with those identified by deep sequencing. A total of 88 miRNAs were predicted to target interferon-associated genes; 6 appear to target the 3'UTR of MEV-specific receptor transferring receptor mRNAs; and 8 to target the MEV mRNA coding region. No miRNAs coded by MEV itself were detected.

The mechanical properties of urethane crosslinked poly(ethylene oxide-co-propylene oxide) glyceryl ether-plasticiser (tetraethylene glycol dimethyl ether, or methylformamide)-salt (LiClO4)-based polymer electrolytes have been studied. It was found that, with increasing concentration of salt, the elastic modulus and tensile strength of the materials unexpectedly decrease. This is interpreted in terms of a predominance of intramolecular coordination of the Li+ ions by the polymer.

Methoxy-ethylene glycol methacrylates, CH 2 CMeCOO(CH 2 CH 2 O) n Me ( n = 1, 2, 3), ethoxy-triethylene glycol methacrylate, CH 2 CMeCOO(CH 2 CH 2 O) 3 Et, and N,N-dimethylaminoethyl methacrylate, CH 2 CMeCOOCH 2 CH 2 NMe 2 , were used to synthesise the corresponding polymers. Conductivities of these polymers complexed with lithium perchlorate were investigated. Tetraethylene glycol dimethyl ether was used as plasticiser to increase the conductivity of the materials. A conductivity of 10 −5 S cm −1 was obtained at room temperature for the plasticised polymer samples. Effects of polymer structure, plasticiser, salt concentration and temperature on conductivity and glass transition temperature of the polymer electrolytes are discussed.

Conversion is near quantitative with activated magnesium as a coreductant and there is evidence for catalytic turnover in ytterbium on addition of a cyclopentadiene source. Reduction of 2,6-F 2 C 6 H 3 CO 2 H to o -FC 6 H 4 CO 2 H and of o -FC 6 H 4 CO 2 H to PhCO 2 H has also been achieved.

N,N-Dimethylpyrrolidinium hydroxide (P11OH·4H2O) was found to exhibit high ionic conductivity in the solid state (7 × 10−3 S cm−1 at 25 °C) and unusual thermal properties, and 2H solid state NMR measurements indicate liquid-like mobility of the deuterium species in the solid state of P11OD·4.5D2O.
