Bacterial wound infections are prevalent in daily life. However, conventional tissue adhesives lack antimicrobial properties. In this study, a redox method was employed to prepare a nano-silver solution with tannic acid as a dispersant. Subsequently, the nano-silver solution was combined with the precursor solution of the hydroxyethyl methacrylate/vinylpyrrolidone (HEMA/NVP) hydrogel. Finally, it was put under ultraviolet light to produce the hydrogel. The hydrogel exhibits remarkable extendibility (1223 %), an elastic modulus compatible with human skin tissue (3.7 ± 0.5 kPa), the strong adhesion to porcine skin tissue (24.67 ± 1.15 kPa) markedly exceeds that achieved by clinically utilized fibrin glue, low swelling ratio (75 ± 1.55 %), and demonstrates good in vitro antimicrobial properties against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). Furthermore, it displays excellent biocompatibility with fibroblast cells (NIH/3T3) with cell viability above 80 %, favorable blood compatibility with goat blood, and moderate coagulation ability. It provides more possibilities for clinical wound repair.

The accurate assessment of node influence is of vital significance for enhancing system stability. Given the structural redundancy problem triggered by the network topology deviation when an empirical network is copied, as well as the dynamic characteristics of the empirical network itself, it is difficult for traditional static assessment methods to effectively capture the dynamic evolution of node influence. Therefore, we propose a heuristic-based spatiotemporal feature node influence assessment model (HEIST). First, the zero-model method is applied to optimize the network-copying process and reduce the noise interference caused by network structure redundancy. Second, the copied network is divided into subnets, and feature modeling is performed to enhance the node influence differentiation. Third, node influence is quantified based on the spatiotemporal depth-perception module, which has a built-in local and global two-layer structure. At the local level, a graph convolutional neural network (GCN) is used to improve the spatial perception of node influence; it fuses the feature changes of the nodes in the subnetwork variation, combining this method with a long- and short-term memory network (LSTM) to enhance its ability to capture the depth evolution of node influence and improve the robustness of the assessment. Finally, a heuristic assessment algorithm is used to jointly optimize the influence strength of the nodes at different stages and quantify the node influence via a nonlinear optimization function. The experiments show that the Kendall coefficients exceed 90% in multiple datasets, proving that the model has good generalization performance in empirical networks.

The wild strawberry (Fragaria vesca L.; F. vesca) represents a resilient and extensively studied model organism. While the AP2/ERF gene family plays a pivotal role in plant development, its exploration within F. vesca remains limited. In this study, we characterized the AP2/ERF gene family in wild strawberries using the recently released genomic data (F. vesca V6.0). We conducted an analysis of the gene family expansion pattern, we examined gene expression in stem segments and leaves under cold conditions, and we explored its functional attributes. Our investigation revealed that the FvAP2/ERF family comprises 86 genes distributed among four subfamilies: AP2 (17), RAV (6), ERF (62), and Soloist (1). Tandem and segmental duplications significantly contributed to the growth of this gene family. Furthermore, predictive analysis identified several cis-acting elements in the promoter region associated with meristematic tissue expression, hormone regulation, and resistance modulation. Transcriptomic analysis under cold stress unveiled diverse responses among multiple FvAP2/ERFs in stem segments and leaves. Real-time fluorescence quantitative reverse transcription PCR (RT-qPCR) results confirmed elevated expression levels of select genes following the cold treatment. Additionally, overexpression of FvERF23 in Arabidopsis enhanced cold tolerance, resulting in significantly increased fresh weight and root length compared to the wild-type control. These findings lay the foundation for further exploration into the functional roles of FvAP2/ERF genes.

Clusters and their progenitors (protoclusters) at z ∼ 2 − 4, the peak epoch of star formation, are ideal laboratories to study the formation process of both the clusters themselves and their member galaxies. However, a complete census of their member galaxies has been challenging due to observational difficulties. Here we present new JWST/NIRCam observations targeting the distant cluster CLJ1001 at z = 2.51 from the COSMOS-Web program, which, in combination with previous narrowband imaging targeting H α emitters and deep millimeter surveys of CO emitters, provide a complete view of massive galaxy assembly in CLJ1001. In particular, JWST reveals a population of massive, extremely red cluster members in the long-wavelength bands that were invisible in previous Hubble Space Telescope (HST)/F160W imaging (HST-dark members). Based on this highly complete spectroscopic sample of member galaxies, we show that the spatial distribution of galaxies in CLJ1001 exhibits a strong central concentration, with the central galaxy density already resembling that of low- z clusters. Moreover, we reveal a “top-heavy” stellar mass function for the star-forming galaxies (SFGs), with an overabundance of massive SFGs piled up in the cluster core. These features strongly suggest that CLJ1001 is caught in a rapid transition, with many of its massive SFGs likely soon becoming quiescent. In the context of cluster formation, these findings suggest that the earliest clusters form from the inside out and top to bottom, with the massive galaxies in the core assembling first, followed by the less massive ones in the outskirts.

ObjectiveThe purpose of this study was to investigate whether the combination of abnormal systemic immune-inflammation index (SII) levels and hyperglycemia increased the risk of cognitive function decline and reduced survival rate in the United States.MethodsThis cross-sectional study used data from the National Health and Nutrition Examination Survey (NHANES) database from 2011-2014 and enrolled 1,447 participants aged 60 years or older. Restricted cubic splines (RCS), linear regression and kaplan-meier(KM) curve were employed to explore the combined effects of abnormal SII and hyperglycemia on cognitive function and survival rate, and subgroup analysis was also conducted.ResultsThe RCS analysis revealed an inverted U-shaped relationship between lgSII levels and cognitive function. Linear regression analysis indicated that neither abnormal SII nor diabetes alone significantly contributed to the decline in cognitive function compared to participants with normal SII levels and blood glucose. However, when abnormal SII coexisted with diabetes (but not prediabetes), it resulted to a significant decline in cognitive function. After adjusting for various confounding factors, these results remained significant in Delayed Word Recall (β:-0.76, PConclusionThe findings suggest that the impact of diabetes on cognitive function/survival rate is correlated with SII levels, indicating that their combination enhances predictive power.

Abstract Background Pneumocephalus is rare in vaginal deliveries. Pneumocephalus may be asymptomatic or present with signs of increased intracranial pressure. However, parturients who received epidural anesthesia with air in their brains may experience low intracranial pressure headaches after giving birth, causing the diagnosis of pneumocephalus to be delayed. We report a case of a parturient who developed post-dural puncture headache combined with pneumocephalus secondary to vaginal delivery following epidural anesthesia. Case presentation A 24-year-old G1P0 Chinese woman at 38 weeks gestation was in labor and received epidural anesthesia using the loss of resistance to air technique and had a negative prior medical history. She presented with postural headache, neck stiffness and auditory changes 2 h after vaginal delivery. The head non-contrast computed tomography revealed distributed gas density shadows in the brain, indicating pneumocephalus. Her headache was relieved by bed rest, rehydration, analgesia, and oxygen therapy and completely disappeared after 2 weeks of postpartum bed rest. Conclusions This is the first report that positional headaches after epidural anesthesia may not indicate low intracranial pressure alone; it may combine with pneumocephalus, particularly when using the loss of resistance to air technique. At this moment, head computed tomography is essential to discover other conditions like pneumocephalus.

The NAC (NAM, ATAF1/2, CUC2) family of transcription factors (TFs) is a vital transcription factor family of plants. It controls multiple parts of plant development, tissue formation, and abiotic stress response. We cloned the FvNAC29 gene from Fragaria vesca (a diploid strawberry) for this research. There is a conserved NAM structural domain in the FvNAC29 protein. The highest homology between FvNAC29 and PaNAC1 was found by phylogenetic tree analysis. Subcellular localization revealed that FvNAC29 is localized onto the nucleus. Compared to other tissues, the expression level of FvNAC29 was higher in young leaves and roots. In addition, Arabidopsis plants overexpressing FvNAC29 had higher cold and high-salinity tolerance than the wild type (WT) and unloaded line with empty vector (UL). The proline and chlorophyll contents of transgenic Arabidopsis plants, along with the activities of the antioxidant enzymes like catalase (CAT), peroxidase (POD), and superoxide dismutase (SOD) under 200 mM NaCl treatment or −8 °C treatment, were higher than those activities of the control. Meanwhile, malondialdehyde (MDA) and the reactive oxygen species (ROS) content were higher in the WT and UL lines. FvNAC29 improves transgenic plant resistance to cold and salt stress by regulating the expression levels of AtRD29a, AtCCA1, AtP5CS1, and AtSnRK2.4. It also improves the potential to tolerate cold stress by positively regulating the expression levels of AtCBF1, AtCBF4, AtCOR15a, and AtCOR47. These findings suggest that FvNAC29 may be related to the processes and the molecular mechanisms of F. vesca response to high-salinity stress and LT stress, providing a comprehensive understanding of the NAC TFs.

Ginseng peptide (GP) (100, 200, 400 mg/kg bw) lowered blood glucose levels, explored the relationship between GP and gut microbiota-based hypoglycemic effects. GP significantly reduced fasting blood glucose levels (25.38–13.37 mM), improved insulin resistance (IR) (9.78–5.02), considerably increased liver glycogen content (64.71–97.95 mg/g), glycolytic and gluconeogenesis enzyme activities in T2DM mice, significantly reduced free fatty acids (1.65–0.93 mM), triglycerides (1.55–0.66 mM), total cholesterol (14.75–5.43 mM), and low-density lipoprotein (4.12–2.36 mM) in the serum, significantly increased high-density lipoprotein levels (6.20–17.44 mM) to improve lipid metabolism disorders. According to colonic 16S rDNA sequencing results, GP-H regulated gut microbiota disorders and short chain fatty acid metabolism. Western blot analysis showed that GP could regulate glycogen synthesis, gluconeogenesis, and glucose transport by activating IRS-1/PI3K/Akt and AMPK signaling pathways and improve IR by inhibiting the mitochondrial apoptosis signaling pathway.

The Malus baccata (L.) Borkh is a species of apple in the Rosaceae family, which grows in northern China and has excellent drought and cold resistance. It is an excellent apple rootstock for obtaining well-resistant apple varieties and has certain economic value. WRKY-TFs act as regulators in response to abiotic stress, so it is necessary to research the function of WRKY gene in Malus baccata in order to study whether the resistance of Malus baccata is related to WRKY TFs. The MbWRKY40 was isolated for the first time. Through qRT-PCR analysis, it was found that there were differences of MbWRKY40 in all organs of plants. At the same time, its expression increased rapidly under multiple stresses, but was higher under cold and drought stresses. The response mechanism of MbWRKY40 under cold and drought stresses was analyzed through genetic transformation and qRT-PCR research methods, and the relevant physiological indicators of transgenic Arabidopsis were measured. Using GraphPad Prism software to analyze the measured data, it was found that transgenic plants with heterologous expression of MbWRKY40 showed improved tolerance to stress. Transgenic plants had higher chlorophyll and proline contents, but lower malondialdehyde (MDA) content and electrolyte leakage rate (EL). Overexpression of MbWRKY40 can also effectively scavenge reactive oxygen species by positively regulating anti-oxidant enzymes activities under stress treatments. Meanwhile, the high expression of MbWRKY40 promoted the accumulation of AtKIN1, AtDREB2A, AtRD29A, AtERD10, AtCOR47A, and AtRD29B genes in Arabidopsis. Based on these data, it can be inferred that this gene may up-regulate downstream stress-related genes through ABA-induced pathways, thereby improving the survival ability of transgenic plants. The results not only enrich the progress of WRKY TFs in Malus baccata in abiotic stress, but also provide a new possibility for molecular breeding of apple resistance.

In this investigation, we leverage the combination of the Dark Energy Spectroscopic Instrument (DESI) Legacy Imaging Surveys Data Release 9, Survey Validation 3, and Year 1 data sets to estimate the conditional luminosity functions and conditional stellar mass functions (CLFs and CSMFs) of galaxies across various halo mass bins and redshift ranges. To support our analysis, we utilize a realistic DESI mock galaxy redshift survey (MGRS) generated from a high-resolution Jiutian simulation. An extended halo-based group finder is applied to both MGRS catalogs and DESI observation. By comparing the r - and z -band luminosity functions (LFs) and stellar mass functions (SMFs) derived using both photometric and spectroscopic data, we quantified the impact of photometric redshift (photo- z ) errors on the galaxy LFs and SMFs, especially in the low-redshift bin at the low-luminosity/mass end. By conducting prior evaluations of the group finder using MGRS, we successfully obtain a set of CLF and CSMF measurements from observational data. We find that at low redshift, the faint-end slopes of CLFs and CSMFs below ∼10 ^9 h ^−2 L _⊙ (or h ^−2 M _⊙ ) evince a compelling concordance with the subhalo mass functions. After correcting the cosmic variance effect of our local Universe following Chen et al., the faint-end slopes of the LFs/SMFs turn out to also be in good agreement with the slope of the halo mass function.

The roles that mass and environment play in galaxy quenching are still under debate. Leveraging the Photometric objects Around Cosmic webs method, we analyze the excess surface distribution ${\bar{n}}_{2}{w}_{{\rm{p}}}({r}_{{\rm{p}}})$ of photometric galaxies in different colors (rest-frame u − r ) within the stellar mass range of 10 ^9.0 ∼ 10 ^11.0 M _⊙ around spectroscopic massive central galaxies (10 ^10.9 ∼ 10 ^11.7 M _⊙ ) at the redshift interval of 0 < z _s < 0.7, utilizing data from the Hyper Suprime-Cam Subaru Strategic Program and the spectroscopic samples of Slogan Digital Sky Survey (i.e., Main, LOWZ, and CMASS samples). We find that both mass and environmental quenching contribute to the evolution of companion galaxies. To isolate the environmental effect, we quantify the quenched fraction excess (QFE) of companion galaxies encircling massive central galaxies within 0.01 h ^−1 Mpc < r _p < 20 h ^−1 Mpc, representing the surplus quenched fraction relative to the average. We find that the high-density halo environment affects star formation quenching up to about three times the virial radius, and this effect becomes stronger at lower redshift. We also find that even after being scaled by the virial radius, the environmental quenching efficiency is higher for more massive halos or for companion galaxies of higher stellar mass, though the trends are quite weak. We present a fitting formula that comprehensively captures the QFE across central and companion stellar mass bins, halo-centric distance bins, and redshift bins, offering a valuable tool for constraining galaxy formation models. Furthermore, we have made a quantitative comparison with IllustrisTNG that underscores some important differences, particularly in the excessive quenching of low-mass companion galaxies (

‘Beta’ is a hybrid of Vitis riparia L. and V. labrusca and has a strong ability to adapt to adverse growth environments and is mainly cultivated and used as a resistant rootstock. At present, the most extensively studied MYB TFs are R2R3-type, which have been found to be involved in plant growth, development, and stress response processes. In the present research, VhMYB15, a key transcription factor for abiotic stress tolerance, was screened by bioinformatics in ‘Beta’ rootstock, and its function under salinity and drought stresses was investigated. VhMYB15 was highly expressed in roots and mature leave under salinity and drought stresses. Observing the phenotype and calculating the survival rate of plants, it was found that VhMYB15-overexpressing plants exhibited relatively less yellowing and wilting of leaves and a higher survival rate under salinity and drought stresses. Consistent with the above results, through the determination of stress-related physiological indicators and the expression analysis of stress-related genes (AtSOS2, AtSOS3, AtSOS1, AtNHX1, AtSnRK2.6, AtNCED3, AtP5CS1, and AtCAT1), it was found that transgenic Arabidopsis showed better stress tolerance and stronger adaptability under salinity and drought stresses. Based on the above data, it was preliminarily indicated that VhMYB15 may be a key factor in salinity and drought regulation networks, enhancing the adaptability of ‘Beta’ to adverse environments.

The two-point correlation function (2PCF) is a powerful statistical tool to measure galaxy clustering. Although 2PCF has also been used to study the clustering of stars on subparsec to kiloparsec scales, its physical implication is not clear. In this study, we use the Illustris-TNG50 simulation to study the connection between the 2PCF of accreted halo stars and the assembly histories of Milky Way-mass galaxies. We find, in general, that the 2PCF signal increases with the increase in galactocentric radii, r , and with the decrease in the pair separations. Galaxies that assemble late on average have stronger 2PCF signals. With z _1/4 , z _1/2 , and z _3/4 defined as the redshifts when galaxies accreted one-fourth, half, and three-fourths of their ex situ stellar mass today, we find they all show the strongest correlations with the 2PCF signals at r < ∼ 0.2 R _200 . z _3/4 shows the strongest correlations than those of z _1/4 or z _1/2 . However, the correlations have large scatters. The 2PCFs in the velocity space show weaker correlations with the galaxy formation times within ∼0.35 R _200 than real-space 2PCFs, and the scatter is considerably large. Both the real- and velocity-space 2PCFs correlate with the assembly histories of the host dark matter halos as well. Within 0.3 R _200 , the real-space 2PCF shows stronger correlations with the galaxy formation histories than with the halo formation histories. We conclude that it is difficult to use 2PCF alone to precisely predict the formation times or assembly histories of galaxies.

Abstract Background Growing evidence shows that C-Type Lectin Domain Containing 7A (Clec7a) may be involved into neuroinflammatory injury of various neurological diseases. However, its roles in neuropathic pain remain unclear. Methods A chronic constriction injury (CCI) rat model was constructed, and gene expression profilings in spinal cord tissues of CCI-insulted rats were detected by both microarray and RNA-seq studies. A series of bioinformatics analyses identified C/EBPβ-Clec7a to be a candidate axis involved into neuropathic pain. Then, its roles in mechanical allodynia, and pathological and molecular changes during CCI progression were determined by various gain-of-function and loss-of-function experiments in vivo and in vitro. Results Significant upregulation of Clec7a at both mRNA and protein levels were verified in spinal cord tissues of CCI-insulted rats. Clec7a knockdown markedly attenuated CCI-induced mechanical allodynia, obstructed Syk, ERK and JNK phosphorylation, inhibited NLRP3 inflammasome and caspase-1 activation, GSDMD cleavage, and consequently reduced the release of pro-inflammatory cytokines (all P

As an excellent grafting material, Malus baccata (L.) Borkh is native to Liaoning, Jilin, Heilongjiang and other regions in China, with a strong adverse environmental adaptability. As a typical transcription factor, the NAC gene acts as a regulator in many molecular pathways responding to abiotic stress. However, research of NAC in the Malus baccata has just begun. In the present research, a new NAC transcription factor, MbNAC22, was obtained from the seedlings of Malus baccata, and its function in drought and salt treatments was studied by heterologous expression. The open reading frame of the MbNAC22 gene is 768, encoding 255 amino acids (aa). Through confocal microscopy, MbNAC22 was found to be located in the nucleus. The heterologous expression of MbNAC22 in Arabidopsis showed that it enhanced the viability of Arabidopsis under drought and salt treatments. Under stresses, the chlorophyll content of the plants decreased, but the decline of the overexpressed-MbNAC22 Arabidopsis was relatively low. Through phenotypic observation and determination of stress-related physiological indicators, it was found that compared with WT Arabidopsis, overexpressed-MbNAC22 Arabidopsis had a higher tolerance to stresses. Under stresses, the overexpression of MbNAC22 positively regulated ion-transport-related genes (AtNHX1 and AtSOS1), the key genes of the ABA pathway (AtNCED3 and AtDREB2A), the proline synthesis gene (AtP5CS2) and the drought-induced gene (AtERD11), while the expression of the leaf senescence-associated gene (AtSAG21) and programmed cell death related gene (AtAEP1) was inhibited. Therefore, we speculate that MbNAC22 responds positively to drought and salt stresses by regulating the expression of stress-related genes.

We investigate the impact of binary orbital motions on the dynamical modeling of dwarf galaxies with intrinsic line-of-sight velocity dispersions ( ${\sigma }_{{v}_{r}}$ ) of 1–9 km s ^−1 . Using dwarf galaxies from the auriga level-2 and level-3 simulations, we apply the Jeans Anisotropic Multi-Gaussian Expansion modeling to tracer stars before and after including binaries to recover the dynamical masses. The recovered total masses within the half-mass radius of tracers, M (< r _half ), are always inflated due to binary motions, with greater inflations occurring for smaller ${\sigma }_{{v}_{r}}$ . However, many dwarf galaxies experience central density deflated due to binary motions, with little dependence on ${\sigma }_{{v}_{r}}$ . This is due to the negative radial gradients in the velocity dispersion profiles, with the fractional inflation in ${\sigma }_{{v}_{r}}$ due to binaries more significant in outskirts. An extreme binary fraction of 70% can lead to central density deflation of up to 10%–20% at 3 km s ^−1 < ${\sigma }_{{v}_{r}}$ < 8 km s ^−1 , with M ( < r _half ) inflated by 4% at 9 km s ^−1 and up to 15% at 3 km s ^−1 . A lower binary fraction of 36% leads to similar deflations, with the inflations decreasing to approximately 10% at 3 km s ^−1 and becoming statistically insignificant. The choice of binary orbit distribution models does not result in significant differences, and observational errors tend to slightly weaken the deflations in the recovered central density. Two observations separated by 1 yr to exclude binaries lead to almost zero inflations/deflations for a binary fraction of 36% over 3 km s ^−1 < ${\sigma }_{{v}_{r}}\lt 9$ km s ^−1 . For ${\sigma }_{{v}_{r}}\sim 1$ km s ^−1 to 3 km s ^−1 , a binary fraction of 70% (36%) still results in 60% (30%) to 10% (1%) of inflations in M ( < r _half ), even with two-epoch observation.

We investigate the buildup of the halo profile out to large scale in a cosmological simulation, focusing on the roles played by the recently proposed depletion radii. We explicitly show that halo growth is accompanied by the depletion of the environment, with the inner depletion radius demarcating the two. This evolution process is also observed via the formation of a trough in the bias profile, with the two depletion radii identifying key scales in the evolution. The ratio between the inner depletion radius and the virial radius is approximately a constant factor of 2 across redshifts and halo masses. The ratio between their enclosed densities is also close to a constant of 0.18. These simple scaling relations reflect the largely universal scaled mass profile on these scales, which only evolves weakly with redshift. The overall picture of the boundary evolution can be broadly divided into three stages according to the maturity of the depletion process, with cluster halos lagging behind low-mass ones in the evolution. We also show that the traditional slow and fast accretion dichotomy of halo growth can be identified as accelerated and decelerated depletion phases, respectively.

Malus baccata (L.) Borkh is an apple rootstock with good drought and cold resistance. The ICE gene is a key factor in the molecular mechanisms of plant drought and cold resistance. In the present research, the function of drought- and cold-induced MbICE1 of Malus baccata was investigated in Arabidopsis. According to GFP fluorescence images, MbICE1 was determined to be a nuclear protein. The MbICE1 was transferred to Arabidopsis, showing enhanced tolerance to drought and cold stresses. Under drought and cold treatments, the transgenic Arabidopsis had higher chlorophyll content and free proline content than WT plants, but the Malondialdehyde (MDA) content and electrolyte leakage (EL) were lower than those of WT plants. In addition, drought and cold led to a large accumulation of ROS (H2O2 and O2−) content in Arabidopsis, while overexpression of MbICE1 enhanced the antioxidant enzyme activity in Arabidopsis and improved the plant’s resistance to stresses. Moreover, the accumulation of MbICE1 promoted the expression of AtCBF1, AtCBF2, AtCBF3, AtCOR15a, AtCOR47 and AtKIN1 genes in Arabidopsis. These data indicate that MbICE1 is a key regulator of drought and cold and can be used as a backup gene for breeding Malus rootstocks.

In the pathogenesis of major depressive disorder, chronic stress-related neuroinflammation hinders favorable prognosis and antidepressant response. Mitochondrial DNA may be an inflammatory trigger, after its release from stress-induced dysfunctional central nervous system mitochondria into peripheral circulation. This evidence supports the potential use of peripheral mitochondrial DNA as a neuroinflammatory biomarker for the diagnosis and treatment of major depressive disorder. Herein, we critically review the neuroinflammation theory in major depressive disorder, providing compelling evidence that mitochondrial DNA release acts as a critical biological substrate, and that it constitutes the neuroinflammatory disease pathway. After its release, mitochondrial DNA can be carried in the exosomes and transported to extracellular spaces in the central nervous system and peripheral circulation. Detectable exosomes render encaged mitochondrial DNA relatively stable. This mitochondrial DNA in peripheral circulation can thus be directly detected in clinical practice. These characteristics illustrate the potential for mitochondrial DNA to serve as an innovative clinical biomarker and molecular treatment target for major depressive disorder. This review also highlights the future potential value of clinical applications combining mitochondrial DNA with a panel of other biomarkers, to improve diagnostic precision in major depressive disorder.

Apple, as one of the most important economic forest tree species, is widely grown in the world. Abiotic stress, such as low temperature and high salt, affect apple growth and development. Ethylene response factors (ERFs) are widely involved in the responses of plants to biotic and abiotic stresses. In this study, a new ethylene response factor gene was isolated from Malus baccata (L.) Borkh and designated as MbERF11. The MbERF11 gene encoded a protein of 160 amino acid residues with a theoretical isoelectric point of 9.27 and a predicated molecular mass of 17.97 kDa. Subcellular localization showed that MbERF11 was localized to the nucleus. The expression of MbERF11 was enriched in root and stem, and was highly affected by cold, salt, and ethylene treatments in M. baccata seedlings. When MbERF11 was introduced into Arabidopsis thaliana, it greatly increased the cold and salt tolerance in transgenic plant. Increased expression of MbERF11 in transgenic A. thaliana also resulted in higher activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), higher contents of proline and chlorophyll, while malondialdehyde (MDA) content was lower, especially in response to cold and salt stress. Therefore, these results suggest that MbERF11 probably plays an important role in the response to cold and salt stress in Arabidopsis by enhancing the scavenging capability for reactive oxygen species (ROS).

NAC (no apical meristem (NAM), Arabidopsis thaliana transcription activation factor (ATAF1/2) and cup shaped cotyledon (CUC2)) transcription factors play crucial roles in plant development and stress responses. Nevertheless, to date, only a few reports regarding stress-related NAC genes are available in Malus baccata (L.) Borkh. In this study, the transcription factor MbNAC25 in M. baccata was isolated as a member of the plant-specific NAC family that regulates stress responses. Expression of MbNAC25 was induced by abiotic stresses such as drought, cold, high salinity and heat. The ORF of MbNAC25 is 1122 bp, encodes 373 amino acids and subcellular localization showed that MbNAC25 protein was localized in the nucleus. In addition, MbNAC25 was highly expressed in new leaves and stems using real-time PCR. To analyze the function of MbNAC25 in plants, we generated transgenic Arabidopsis plants that overexpressed MbNAC25. Under low-temperature stress (4 °C) and high-salt stress (200 mM NaCl), plants overexpressing MbNAC25 enhanced tolerance against cold and drought salinity conferring a higher survival rate than that of wild-type (WT). Correspondingly, the chlorophyll content, proline content, the activities of antioxidant enzymes superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) were significantly increased, while malondialdehyde (MDA) content was lower. These results indicated that the overexpression of MbNAC25 in Arabidopsis plants improved the tolerance to cold and salinity stress via enhanced scavenging capability of reactive oxygen species (ROS).

Objective: To investigate the impact of metabolic dysfunction-associated steatotic liver disease (MASLD) on the efficacy of immune checkpoint inhibitor (ICI)-based therapy in patients with chronic hepatitis B (CHB)-related hepatocellular carcinoma (HCC). Methods: A total of 155 patients with CHB-related HCC who received ICI–based therapy (in the Department of Hepatology, Tianjin Second People’s Hospital and Department of Hepatobiliary Oncology, Tianjin Medical University Cancer Institute & Hospital) between April 2021 and December 2023 were evaluated. Patients were divided into two groups: MASLD concurrent with CHB [MASLD-CHB] (n = 38), and CHB (n = 117). Results: The median progression-free survival (PFS, 6.9 months vs. 9.3 months; P = 0.001), progressive disease (57.89% vs. 37.61%; P = 0.028), and disease control rate (42.11% vs. 62.39%; P = 0. 028) in the MASLD-CHB group were significantly worse than the CHB group. The median overall survival was not attained. The percentage of CD4+PD1+ (17. 56% vs. 8.89%; P < 0.001) and CD8+PD1+ T cells (10.50% vs. 7.42%; P = 0.005) in patient samples from the MASLD-CHB group were significantly higher than the CHB group. Concurrent MASLD [hazard ratio (HR) = 1.921; 95% CI, 1.138–3.245; P = 0.015] and alpha-fetoprotein levels after 3 months of treatment (HR = 2.412; 95% CI, 1.360–4.279; P = 0.003) were independent risk factors for PFS in all patients. Conclusions: ICI-based therapy in patients with CHB-related HCC and concurrent MASLD resulted in poorer efficacy and shorter PFS compared to patients with CHB-related HCC alone. High-harmonic generation from a gas target exhibits sharp spectral features and rapid phase variation near the Cooper minimum. By applying spectral filtering, shaped isolated attosecond pulses can be generated where the pulse is split into two in the time domain. Using such shaped extreme-ultraviolet (XUV) pulses, we theoretically study attosecond transient absorption (ATA) spectra of helium 2s2p autoionizing state which is resonantly coupled to the 2s² dark state by a time-delayed infrared laser. Our simulations show that the asymmetric 2s2p Fano line shape can be readily tuned into symmetric Lorentzian within the time delay of a few tens of attoseconds. Such efficient control is due to the destructive interference in the generation of the 2s2p state when it is excited by a strongly shaped XUV pulse. This is to be compared to prior experiments where tuning the line shape of a Fano resonance would take tens of femtoseconds. We also show that the predicted ATA spectral line shape can be observed experimentally after propagation in a gas medium. Our results suggest that strongly shaped attosecond XUV pulses offer the opportunity for controlling and probing fine features of narrow resonances on the few-ten attoseconds timescale.
