6. Bone-Targeting HUVEC-Derived Exosomes Containing miR-503-5p for Osteoporosis Therapy.

Abstract

Osteoporosis (OP) is a systemic bone disease characterized by decreased bone mass, damaged bone microstructure, increased bone fragility, and increased risk of fractures. It is more common in postmenopausal women and elderly people. The development of drugs with a high bone-targeting ability is urgently needed. We prepared a bioactive nanoparticle that modified the exosomes derived from human umbilical vein endothelial cells (HUVEC-ExomiR‑503‑high) to contain a large amount of miR-503-5p, with a diameter range of 30–150 nm. The HUVEC-ExomiR‑503‑high that we constructed had the same characteristics as ordinary exosomes and could bind to osteoblasts/osteoclasts in vitro. Moreover, they exhibited better bone-targeting ability than exosomes derived from other sources of bone marrow stromal cells in vivo. Bioinformatics analysis showed that miR-503-5p is strongly associated with bone-related pathways. In vitro experiments showed that HUVEC-ExomiR‑503‑high effectively inhibited osteoclast differentiation and promoted osteogenic differentiation. In vivo experiments showed that injecting HUVEC-ExomiR‑503‑high into OP mice significantly increased bone density and prevented OP. HUVEC-ExomiR‑503‑high can be used for bone-targeted therapy of OP, providing an approach to the clinical prevention and treatment of OP. [ABSTRACT FROM AUTHOR]

15. Dynamics and biogeochemical significance of the physical environment in Lake Biwa

Abstract

We have examined two aspects of the dynamics and biogeochemical significance of the physical environment in Lake Biwa. One is the horizontal distribution of cyanobacteria as it relates to the ‘first gyre’ in the north basin of Lake Biwa. We could easily measure the first gyre using a vessel-mounted acoustic doppler currents profiler. We were able to quantify the dynamics of the horizontal and vertical structure of currents and water temperature of this gyre. The first gyre did not remain at a fixed position; it moved north and south according to the growth of the gyre. This may play a role in the redistribution of cyanobacteria from place to place in the north basin. The second important environmental dynamic we measured was oxygen consumption rates from 1994 to 2000. We found that the minimum oxygen concentration in the hypolimnion has a clear inverse relationship with the apparent oxygen consumption rates in the 80–85 m depth layer. As a reduction in oxygen concentration in the hypolimnion can have a serious impact on benthic organisms, we concluded that the Lake Biwa environment should be monitored carefully and systematically.

16. Solitary Internal Waves in Lake Biwa

Abstract

This paper describes the behavior of large-amplitude nonlinear internal waves observed in Lake Biwa. A thermistor chain was moored in November of 1992, when the water column in the lake was still well stratified. After a continuous strong north-west wind, a wave train with two distinctive parts was observed traveling from the west to the east. First, a sharp front which caused a temperature decrease of 3.5℃ within two minutes appeared. Next, 10 spatially coherent pulses and a lower-amplitude wave train that was less coherent then followed it. We interpret the initial pulses as solitons according to the nonlinear wave theory. The trailing waves can be well-explained by the infinitesimal wave theory. It can be shown by a two layer model (with a thick homogeneous epilimnion, a thin hypolimnion and the thermocline simplified to an interface) that the wave lengths of the solitons are about 51m to 63m and their speeds are estimated within a range of 18.92cm/s to 22.06cm/s. Thermocline elevations which are predicted by the two-layer model are consistent with the characteristics of the solitons. Subsequent analysis shows that the solitons with periods of 6 to 10 minutes were generated in the western coastal area approximately 5km away from the sampling station.

17. Chemical and thermal stratification in lakes

Abstract

An index that shows chemical stratification strength [IC-i; i = water quality item such as chlorophyll-a (Chl.a) and soluble phosphorus (SP)] was proposed and compared with one of thermal stratification strength indices, Schmidt’s stability index (SSI), in Shiozu Bay and Lake Biwa, Japan. The proposed indices of IC-i can be easily calculated with at least one set of each water quality data in both the epilimnion and the hypolimnion. The SSI was shown to be consistent with the traditional thermocline index of thermocline strength index (TSI), but SSI is used as the stability index of the whole lake, whereas TSI is used as the stability index near the thermocline. Analyses showed that chemical stratification strength is determined largely by thermal stratification strength. Totally different characteristics of IC-Chl.a and IC-phosphate (PO4) at high SSI in the main North Basin of Lake Biwa and in Shiozu Bay were possibly due to the difference in their volumes and hydrodynamic conditions. The proposed index and relationships are especially useful to roughly determine thermal and chemical stratification when only few water quality data are available.
