The repair of organs and tissues has stepped into a prospective era of regenerative medicine. However, basic research and clinical practice in the lung regeneration remains crawling. Owing to the complicated three dimensional structures and above 40 types of pulmonary cells, the regeneration of lung tissues becomes a great challenge. Compelling evidence has showed that distinct populations of intrapulmonary and extrapulmonary stem/progenitor cells can regenerate epithelia as well as endothelia in various parts of the respiratory tract. Recently, the discovery of human lung stem cells and their relevant studies has opened the door of hope again, which might put us on the path to repair our injured body parts, lungs on demand. Herein, we emphasized the role of endogenous and exogenous stem/progenitor cells in lungs as well as artificial tissue repair for the injured lungs, which constitute a marvelous toolbox for the treatment of acute lung injury. Background Aberrant microRNA (miRNA) expression plays an essential role in the pathogenesis of Hepatocellular Carcinoma (HCC). However, specific involvement of miRNAs in HCC remains incompletely understood. The aim of this study was to explore the relevant microRNAs involved in the development of HCC. Methods MicroRNA microarray was used to screen for the differentially expressed miRNAs in cancerous tissue and adjacent non-cancerous control tissue from patients with HCC (n = 3).Quantitative PCR was subsequently used to verify the results of microarray. Based on the findings, we investigated the role of miR-492 in the pathogenesis of HCC in vitro and in vivo using three tumor cells lines. Furthermore, we analyzed the clinical correlation of miR-492 expression with patient survival (n = 28). Results We showed that microRNA-492 (miR-492) was elevated in HCC samples from patients with hepatic cancer. Knockdown of miR-492 attenuated the proliferation of cancer cell lines in vitro and inhibited primary tumor growth in vivo in SCID mice. We identified PTEN as a functional target for miR-492. Overexpression of miR-492 resulted in decreased PTEN expression and was associated with increased AKT activation in cancer cell lines. Moreover, miR-492-mediated increase of the proliferation of cancer cells was able to be suppressed by a PI3K inhibitor and an AKT inhibitor. The HCC patients with high miR-492/low PTEN had poorer survival. Conclusions miR-492 is implicated in the regulation of HCC progression through PTEN and AKT pathway. In this study, cellulose possessing β-cyclodextrin (β-CD) is employed as a host molecule, and cellulose possessing ferrocene (Fc) is used as a guest polymer. The properties due to the host-guest interactions are presented. The results show that β-CD-cellulose and Fc-cellulose can form inclusion complexes wherein hydrophobic interaction caused by β-CD-cellulose and Fc-cellulose significantly affect the performance of the cellulose gel. A physical gel based on cellulose that can autonomously heal between cut surfaces after 24 h was formed under mild conditions. Moreover, ferrocene redox status affects the hydrophobic interaction, such that the hydrophobic interaction can strengthen the gel strength and affect the self-healing property. Background Recently, interest in the utilization of corncob residue (CCR, with high lignin of 45.1%) as a feedstock for bioethanol has been growing. Surfactants have been one of the most popular additives intended to prevent the inhibitory effect of lignin on cellulolytic enzymes, thereby improving hydrolysis. In this study, the effects of biosurfactant tea saponin (TS) on the enzymatic hydrolysis of CCR and the bonding behavior of cellulolytic enzymes to the substrate were investigated. The surface tension in the supernatant was also detected to obtain-information about the characteristics and stability of TS. Results The glucose concentration was 17.15 mg/mL at 120 hours of hydrolysis with the low loading of cellulolytic enzymes (7.0 FPU/g cellulose and 10.5 BGU/g cellulose) and 5% CCR. The optimal dosage of TS was its critical micelle concentration (cmc, 1.80 mg/mL). The glucose yield was enhanced from 34.29 to 46.28 g/100 g dry matter by TS. The results indicate that TS can promote the adsorption of cellulolytic enzymes on the substrate and mediate the release of adsorbed enzymes. Meanwhile, TS improves the recovery of the cellulolytic enzymes after a hydrolysis cycle and prevents deactivation of the enzymes during the intense shaking process. The surface tension in supernatants of digested CCR with TS remained at 50.00 mN/m during the course of hydrolysis. It is interesting to note that biosurfactant TS can maintain the surface tension in supernatants, despite its digestibility by cellulolytic enzymes. Conclusions Serving as an accelerant of lignocellulose hydrolysis, TS can also be degraded by the cellulolytic enzymes and release glucose while retaining stability, which reduces the cost of both the cellulolytic enzymes and the additive. As the glucose from the TS could be utilized by yeast, further efforts will investigate the mechanism of function and the application of TS in the production of ethanol by simultaneous saccharification and fermentation (SSF). Major trauma is the leading cause of death in young adults. Despite advances in prehospital system and treatment in hospital, mortality rates have not improved significantly over the past decades. Victims of severe injuries who survive the initial hours have great risk for additional life-threatening complications, including uncontrollable infection (sepsis) and multiple organ dysfunction syndrome (MODS). Single nucleotide polymorphisms (SNPs) have been shown to affect susceptibility to the course of numerous diseases. Accumulating evidence suggests that genetic backgrounds also play important roles in posttraumatic complications. Genetic polymorphismsmay become powerful biomarkers for diagnosis and prognosis of trauma-induced complications. Recent advances in studies on associations between genetic polymorphisms and sepsis or MODS have led to better understanding of posttraumatic complications. Here we summarise recent findings on genetic variations in molecules of the innate immune system and other systems as well as their connection with susceptibility to posttraumatic complications. To study the long-term symptom burden among older COVID-19 survivors 2 years after hospital discharge and identify associated risk factors. The current cohort study included COVID-19 survivors aged 60 years and above, who were discharged between February 12 and April 10, 2020, from two designated hospitals in Wuhan, China. All patients were contacted via telephone and completed a standardized questionnaire assessing self-reported symptoms, the Checklist Individual Strength (CIS)-fatigue subscale, and two subscales of the Hospital Anxiety and Depression Scale (HADS). Of the 1,212 patients surveyed, the median (IQR) age was 68.0 (64.0-72.0), and 586 (48.3%) were male. At the two-year follow-up, 259 patients (21.4%) still reported at least one symptom. The most frequently self-reported symptoms were fatigue, anxiety, and dyspnea. Fatigue or myalgia, which was the most common symptom cluster (11.8%; 143/1212), often co-occurred with anxiety and chest symptoms. A total of 89 patients (7.7%) had CIS-fatigue scores = 27, with older age (odds ratio [OR], 1.08; 95% CI: 1.05-1.11, P < 0.001) and oxygen therapy (OR, 2.19; 95% CI: 1.06-4.50, P= 0.03) being risk factors. A total of 43 patients (3.8%) had HADS-Anxiety scores = 8, and 130 patients (11.5%) had HADS-Depression scores = 8. For the 59 patients (5.2%) who had HADS total scores = 16, older age, serious illness during hospitalization and coexisting cerebrovascular diseases were risk factors. Cooccurring fatigue, anxiety, and chest symptoms, as well as depression, were mainly responsible for long-term symptom burden among older COVID-19 survivors 2 years after discharge. PURPOSE: Hepatocellular carcinoma (HCC) is one of the most common solid tumors and ranks the third leading cause of cancer mortality worldwide. The purpose of this study was to examine the expression dynamics of GOLPH3 in HCC tissue samples, and explore the correlation between GOLPH3 expression and prognosis in patients with HCC. METHODS: The levels of Golgi phosphoprotein 3 (GOLPH3) mRNA and protein in liver cancer cell lines and fresh tissues were determined by qRT-PCR and Western blotting. Additionally, the protein expression of GOLPH3 was detected by immunohistochemistry. RESULTS: High GOLPH3 expression was positively correlated with serum AFP level (P=0.015) and tumor recurrence or metastasis (P=0.010). In addition, liver cancer patients with high GOLPH3 expression had significantly poorer overall survival (HR, 1.87; 95% CI, 1.19-2.94; P=0.006) and poorer disease-free survival (HR, 1.90; 95% CI, 1.21-2.98; P=0.005) than those with low GOLPH3 expression. The cumulative 5-year survival rate was only 35.19% (95% CI, 26.18%-44.20%) in the high GOLPH3 expression group, whereas it was 55.93% (95% CI, 43.26%-68.60%) in the low GOLPH3 expression group. Furthermore, multivariate Cox regression analysis demonstrated that the expression of GOLPH3, tumor size and tumor multiplicity were independent prognostic predictors for HCC patients. CONCLUSION: GOLPH3 was overexpressed in HCC at both the mRNA and protein levels, and the presence of high levels of expression of GOLPH3 could serve as a novel and potential prognostic biomarker for liver cancer patients. Cellulose possessing β-cyclodextrin (β-CD) was used as a host molecule and cellulose possessing ferrocene (Fc) as a guest polymer. Infrared spectra, differential scanning calorimetry (DSC), ultraviolet spectroscopy (UV), and contact angle analysis were used to characterise the material structure and the inclusion behaviour. The results showed that the β-CD-cellulose and the Fc-cellulose can form inclusion complexes. Moreover, ferrocene oxidation, and reduction of state can be adjusted by sodium hypochlorite (NaClO) as an oxidant and glutathione (GSH) as a reductant. In this study, a physical gel based on β-CD-cellulose/Fc-cellulose was formed under mild conditions in which autonomous healing between cut surfaces occurred after 24 hours. The physical gel can be controlled in the sol-gel transition. The compressive strength of the Fc-cellulose/β-CD-cellulose gel increased with increased cellulose concentration. The host-guest interaction between the side chains of cellulose could strengthen the gel. Phagocytic clearance of apoptotic cells by the macrophages (efferocytosis) is impaired in sepsis, but its mechanism is poorly understood. Extracellular cold-inducible RNA-binding protein (eCIRP) is a novel damage-associated molecular pattern that fuels inflammation. We identify that eCIRP-induced neutrophil extracellular traps (NETs) impair efferocytosis through a novel mechanism. Coculture of macrophages and apoptotic thymocytes in the presence of recombinant murine CIRP (rmCIRP)–induced NETs significantly inhibited efferocytosis. Efferocytosis was significantly inhibited in the presence of rmCIRP-treated wild-type (WT), but not PAD4-/- neutrophils. Efferocytosis in the peritoneal cavity of rmCIRP-injected PAD4-/-mice was higher than WT mice. Milk fat globule–EGF–factor VIII (MFG-E8), an opsonin, increased macrophage efferocytosis, whereas the inhibition of efferocytosis by NETs was not rescued upon addition of MFG-E8, indicating disruption of MFG-E8’s receptor(s) αvβ3 or αvβ5 integrin by the NETs. We identified neutrophil elastase in the NETs significantly inhibited efferocytosis by cleaving macrophage surface integrins αvβ3 and αvβ5. Using a preclinical model of sepsis, we found that CIRP-/- mice exhibited significantly increased rate of efferocytosis in the peritoneal cavity compared with WT mice. We discovered a novel role of eCIRP-induced NETs to inhibit efferocytosis by the neutrophil elastase–dependent decrease of αvβ3/αvβ5 integrins in macrophages. The DNA repair enzyme 8-oxoguanine DNA glycosylase 1 (OGG1), which excises 8-oxo-7,8-dihydroguanine lesions induced in DNA by reactive oxygen species, has been linked to the pathogenesis of lung diseases associated with bacterial infections. A recently developed small molecule, SU0268, has demonstrated selective inhibition of OGG1 activity; however, its role in attenuating inflammatory responses has not been tested. In this study, we report that SU0268 has a favorable effect on bacterial infection both in mouse alveolar macrophages (MH-S cells) and in C57BL/6 wild-type mice by suppressing inflammatory responses, particularly promoting type I IFN responses. SU0268 inhibited proinflammatory responses during Pseudomonas aeruginosa (PA14) infection, which is mediated by the KRAS-ERK1-NF-κB signaling pathway. Furthermore, SU0268 induces the release of type I IFN by the mitochondrial DNA-cGAS-STING-IRF3-IFN-β axis, which decreases bacterial loads and halts disease progression. Collectively, our results demonstrate that the small-molecule inhibitor of OGG1 (SU0268) can attenuate excessive inflammation and improve mouse survival rates during PA14 infection. The success of early Chinese blue-and-white porcelains relied heavily on imported cobalt pigment from the West. In contrast to art-historical concept, which contains both typological evidence and literature records, it is assumed that imported Sumali blue was completely superseded by domestic Chinese asbolane ore based on the analytical results of the Fe/Mn ratio in imperial productions from the Xuande reign (1426 to 1435 CE) onward. Using a focused ion beam transmission electron microscopy technique to reassess this hotly debated question, we have identified two classes of residual submicron pigment particles in the blue glaze with diagnostic differences in morphology, chemical composition, and distribution behavior. Compared with the microstructural features of the blue-and-white porcelains of the Yuan and Qing dynasties, we show that a mixture of imported and domestic cobalt pigments was used for aesthetic reasons, indicating that the overseas supply chain of imported pigment remained consistent and adequate even though the authorities had terminated official trade and tributary activities after the death of Admiral Zheng He. This discovery further suggests that the globalized trading network and cross-regional industrial chain had been extensively established in the 15th century. Moreover, we provide analytical evidence against the fundamental assumption of the current Fe/Mn provenancing criteria, implying that the failures of previous chemical analyses can be attributed to elemental differentiation between the silicate glaze and the arsenic pigment. We propose an innovative method for directly assessing original mineralogic information from submicron residual pigment particles that provides a more reliable way to trace cobalt circulation and holds great promise for provenance studies. Autonomous self-repairing properties can prolong the service life of materials. In this paper, galactomannan hydrogel with high mechanical strength was prepared by graft copolymerization of galactomannan with acrylamide and octadecyl methacrylate in aqueous solution. The microstructure, water absorption property, self-healing behavior, and mechanical properties of the hydrogels were investigated using Fourier transform infrared spectroscopy (FTIR) and scanning electron microscope (SEM), etc. The galactomannan hydrogel had the highest tensile strength of 49 KPa and strain of 3000%. The water absorption reached 2340%, and the removal rate of methylene blue was more than 80%. Galactomannan hydrogels demonstrated significant self-healing properties. The cut hydrogel was quite effective in self-repairing in a few minutes, and the self-repairing strength increased with increasing contact time of hydrogel cut surfaces. The healing efficiency of fracture strain could reach 92.7% of the original sample in 10 h. The maximum water absorption of hydrogel reached 2340%. The maximum removal rate of methylene blue by hydrogel reached 80.5%, and the maximum adsorption capacity was 19.3 mg/g. The novelty of the work lies in octadecyl methacrylate being used for galactomannan cross-linking with the ability to self-repair after fracture. Proper control of cell migration is critically important in many biologic processes, such as wound healing, immune surveillance, and development. Much progress has been made in the initiation of cell migration; however, little is known about termination and sometimes directional reversal. During active cell migration, as in wound healing, development, and immune surveillance, the integrin expression profile undergoes drastic changes. Here, we uncovered the extensive regulatory and even opposing roles of integrins in directional cell migration in electric fields (EFs), a potentially important endogenous guidance mechanism. We established cell lines that stably express specific integrins and determined their responses to applied EFs with a high throughput screen. Expression of specific integrins drove cells to migrate to the cathode or to the anode or to lose migration direction. Cells expressing αMβ2, β1, α2, αIIbβ3, and α5 migrated to the cathode, whereas cells expressing β3, α6, and α9 migrated to the anode. Cells expressing α4, αV, and α6β4 lost directional electrotaxis. Manipulation of α9 molecules, one of the molecular directional switches, suggested that the intracellular domain is critical for the directional reversal. These data revealed an unreported role for integrins in controlling stop, go, and reversal activity of directional migration of mammalian cells in EFs, which might ensure that cells reach their final destination with well-controlled speed and direction. Background: Efficient cofermentation of glucose and xylose is necessary for economically feasible bioethanol production from lignocellulosic biomass. Here, we demonstrate pretreatment of sugarcane bagasse (SCB) with green liquor (GL) combined with ethanol (GL-Ethanol) by adding different GL amounts. The common Saccharomyces cerevisiae (CSC) and thermophilic S. cerevisiae (TSC) strains were used and different yeast cell mass ratios (CSC to TSC) were compared. The simultaneous saccharification and cofermentation (SSF/SSCF) process was performed by 5-20% (w/v) dry substrate (DS) solid loadings to determine optimal conditions for the co-consumption of glucose and xylose. Results: Compared to previous studies that tested fermentation of glucose using only the CSC, we obtained higher ethanol yield and concentration (92.80% and 23.22 g/L) with 1.5 mL GL/g-DS GL-Ethanol-pretreated SCB at 5% (w/v) solid loading and a CSC-to-TSC yeast cell mass ratio of 1:2 (w/w). Using 10% (w/v) solid loading under the same conditions, the ethanol concentration increased to 42.53 g/L but the ethanol yield decreased to 84.99%. In addition, an increase in the solid loading up to a certain point led to an increase in the ethanol concentration from 1.5 mL GL/g- DS-pretreated SCB. The highest ethanol concentration (68.24 g/L) was obtained with 15% (w/v) solid loading, using a CSC-to-TSC yeast cell mass ratio of 1:3 (w/w). Conclusions: GL-Ethanol pretreatment is a promising pretreatment method for improving both glucan and xylan conversion efficiencies of SCB. There was a competitive relationship between the two yeast strains, and the glucose and xylose utilization ability of the TSC was better than that of the CSC. Ethanol concentration was obviously increased at high solid loading, but the yield decreased as a result of an increase in the viscosity and inhibitor levels in the fermentation system. Finally, the SSCF of GL-Ethanol-pretreated SCB with mixed S. cerevisiae strains increased ethanol concentration and was an effective conversion process for ethanol production at high solid loading. Oxygen is supplied as a supportive treatment for patients suffering from acute respiratory distress syndrome. Unfortunately, high oxygen concentration increases reactive oxygen species generation, which causes DNA damage and ultimately cell death in the lung. Although 8-oxoguanine-DNA glycosylase (OGG-1) is involved in repairing hyperoxia-mediated DNA damage, the underlying molecular mechanism remains elusive. In this study, we report that ogg-1-deficient mice exhibited a significant increase of proinflammatory cytokines (TNF-α, IL-6, and IFN-γ) in the lung after being exposed to 95% oxygen. In addition, we found that ogg-1 deficiency downregulated (macro)autophagy when exposed to hyperoxia both in vitro and in vivo, which was evident by decreased conversion of LC3-I to LC3-II, reduced LC3 punctate staining, and lower Atg7 expression compared with controls. Using a chromatin immunoprecipitation assay, we found that OGG-1 associated with the promoter of Atg7, suggesting a role for OGG1 in regulation of Atg7 activity. Knocking down OGG-1 decreased the luciferase reporter activity of Atg7. Further, inflammatory cytokine levels in murine lung epithelial cell line cells were downregulated following autophagy induction by starvation and rapamycin treatment, and upregulated when autophagy was blocked using 3-methyladenine and chloroquine. atg7 knockout mice and Atg7 small interfering RNA-treated cells exhibited elevated levels of phospho-NF-κB and intensified inflammatory cytokines, suggesting that Atg7 impacts inflammatory responses to hyperoxia. These findings demonstrate that OGG-1 negatively regulates inflammatory cytokine release by coordinating molecular interaction with the autophagic pathway in hyperoxia-induced lung injury. The seeds of Gleditsia sinensis and Gleditsia microphylla, widespread in China, are an important source of galactomannans. G. sinensis gum (GSG) and G. microphylla gum (GMG) were purified and precipitated using different concentrations of ethanol and isopropanol. The GSG and GMG, precipitated in different stages, presented different characteristics, including polymer recovery, mannose/galactose ratio, chemical composition, molecular weight, and morphological appearance. The galactomannan recovery of GSG and GMG in 33.3% ethanol was 81.7% and 82.5%, respectively, while that in 28.8% isopropanol was 81.3% and 82.9%, respectively. To achieve similar precipitation efficiency, the amount of isopropanol should be lower than that of ethanol because of the lower dielectric constant of isopropanol (20 vs. 25 for ethanol). The precipitation behavior of galactomannans in polar organic solvents was dependent on the molecular structures and properties of the solvent. Background: As the most abundant renewable resources, lignocellulosic materials are ideal candidates as alternative feedstock for bioethanol production. Cassava residues (CR) are byproducts of the cassava starch industry which can be mixed with lignocellulosic materials for ethanol production. The presence of lignin in lignocellulosic substrates can inhibit saccharification by reducing the cellulase activity. Simultaneous saccharification and fermentation (SSF) of furfural residues (FR) pretreated with green liquor and hydrogen peroxide (GL-H2O2) with CR saccharification liquid was investigated. The final ethanol concentration, yield, initial rate, number of live yeast cells, and the dead yeast ratio were compared to evaluate the effectiveness of combining delignificated lignocellulosic substrates and starchy substrates for ethanol production. Results: Our results indicate that 42.0 % of FR lignin removal was achieved on FR using of 0.06 g H2O2/g-substrate and 9 mL GL/g-substrate at 80 °C. The highest overall ethanol yield was 93.6 % of the theoretical. When the ratio of 0.06 g/g-H2O2-GL-pretreated FR to CR was 5:1, the ethanol concentration was the same with that ratio of untreated FR to CR of 1:1. Using 0.06 g/g-H2O2-GL-pretreated FR with CR at a ratio of 2:1 resulted in 51.9 g/L ethanol concentration. Moreover, FR pretreated with GL-H2O2 decreased the concentration of byproducts in SSF compared with that obtained in the previous study. Conclusions: The lignin in FR would inhibit enzyme activity and GL-H2O2 is an advantageous pretreatment method to treat FR and high intensity of FR pretreatment increased the final ethanol concentration. The efficiency of ethanol fermentation of was improved when delignification increased. GL-H2O2 is an advantageous pretreatment method to treat FR. As the pretreatment dosage of GL-H2O2 on FR increased, the proportion of lignocellulosic substrates was enhanced in the SSF of the substrate mixture of CR and FR as compared with untreated FR. The price of raw material, energy demand in the pretreatment step, and enzyme usage rate are the major cost factors in the process of converting biomass to bioethanol. Unwashed furfural residues (FRs) possess great potential for application in bioethanol production. Surfactant addition is an effective method to enhance the fermentation rate. In this study, unwashed FRs were used directly as raw materials to produce bioethanol. Tea-seed cake (TSC), tea seed residues that contained protein and saponin, was added in the simultaneous saccharification and fermentation (SSF) process. The effect of TSC dosage on SSF was compared. TSC was added at the dosage of 10 g/L, which resulted in a final ethanol yield of 87.2%. However, a high concentration of TSC could induce cytotoxicity in yeast. The surface tension (approximately 33.92 mM/m) at SSF using TSC-medium was much lower than that of other fermentation systems (about 64.67 mN/m). Further contact angle testing showed that TSC-medium (21.7°) had better wetting capacity than FRs (45.6°). This study provided a proposed process strategy that SSF with the addition of TSC could be a minimum consumption of chemicals and enzymes for future cellulosic ethanol production process. Sugarcane bagasse was pretreated by green liquor combined with ethanol (GL-Ethanol) and green liquor combined with H2O2 (GL-H2O2). After 72 h of enzymatic hydrolysis, the glucose yields of sugarcane bagasse pretreated with GL-Ethanol and GL-H2O2 were 97.7% and 41.7%, respectively. The reason that GL-Ethanol was more effective than GLH2O2 has not been elucidated clearly. In this study, the chemical composition of the sugarcane bagasse and chemical structure of the isolated lignins after these two pretreatment methods were characterized to investigate their correlation with the enzymatic hydrolysis of sugarcane bagasse. The removal of lignins with GL-Ethanol pretreatment was much higher than that of GL-H2O2. In addition, the decomposition of cellulose was lower in the case of GL-Ethanol than in that of GL-H2O2. According to Fourier transform infrared spectroscopy (FT-IR) and 1H-nuclear magnetic resonance (NMR) studies, the ester bonds (belonging to lignincarbohydrate complex) could be broken during GL-Ethanol treatment. Sugarcane bagasse (SCB) was modified by steam explosion pretreatment at 190℃ for 10 min and 210℃ for 5 min using green liquor (GL) combined with hydrogen peroxide (GL-H2O2) and ethanol (GL-Ethanol) for simultaneous saccharification and fermentation (SSF)- based ethanol production. The results showed that 85.02% and 100% of hemicelluloses were solubilized by steam explosion pretreatment at 190℃ for 10 min and 210℃ for 5 min, respectively. Moreover, 20.08% and 73.77% of the lignin was removed through GL-H2O2 and GL-Ethanol pretreatments, respectively. The steam explosion pretreatments greatly improved the specific surface area of the SCB and led to the highest ethanol yield of 92.20% at 190℃ for 10 min and 93.19% of 210℃ for 5 min, respectively. In addition, the ethanol yield reached 72.58% for the GL-Ethanol pretreatment, and about 70% of active lignin could be recovered from the pretreatment liquid. A combination of steam explosion (SE) and chemical pretreatments, such as acid sulfite (AS), alkali (AL), and alkaline sulfite (ALS), were evaluated using bamboo. Low pressure steam explosion at 1.25 MPa for 4 min was first applied to the bamboo. Then, the pretreated bamboo was delignified using three chemical pretreatments. Enzymatic hydrolysis was also compared among the pretreated bamboo samples. It was found that SE-ALS could be a potential method for bamboo pretreatment, which led to the reduction of lignin from 25.15% to 1.74% at 165 °C for 2 h in 5% (w/v) Na2SO3 and 0.7% (w/v) NaOH; however, little cellulose was solubilized during ALS pretreatment. A maximum glucose yield of 99.35% was achieved during the enzymatic hydrolysis process when combined with the SE-ALS pretreatment. The SE-ALS method resulted in a lower degree of lignin condensation and increased delignification compared to the SE-AS method. Background: The treatment of acute respiratory distress syndrome (ARDS), most commonly seen during the organ dysfunction remains unsatisfied. Presently, the stem/progenitor cell-based endogenous repair has been aroused attention enormously. This report investigated the effects of retinoic acid (RA) plus simvastatin (SS) with respect to dynamics of lung repair cells as well as to elucidate the underlying mechanism.Materials and Methods: The experimental Sprague-Dawley rats were divided randomly into normal control (control), sham operated (sham), ARDS, ARDS +  vehicle and ARDS + RA + SS groups. ARDS was reproduced through hemorrhagic shock/resuscitation (shock) and subsequent intratracheal LPS (4.5 mg/kg, Escherichia coli serotype O55: B5) injection. The rats were treated by intragastric administration of RA (2 mg/kg/day) and SS (2 mg/kg/day) for 5 days in the ARDS + RA + SS group. Seven days after the first RA-SS injection, a right lower lobe of lung was sampled for histological analysis concerning systemic uniform random sampling method. Immunohistochemistry of inflation-fixed lungs for alveolar type 1 (AT1), alveolar type 2 (AT2) and Clara cells was measured by AQP5, Pro-SPC and CCSP staining respectively. The alveolar cell proliferation and apoptosis were analyzed with Ki67 staining and terminal deoxylnucleotidyl transferase mediated-dUTP nick end labeling (TUNEL) method. Meanwhile, the alveolar cell numerical and surface density (alveolar cells, AT1, AT2, Clara, proliferating and apoptotic cells) were evaluated by stereology.Results: RA-SS compound exerted anti-inflammatory and pro-repairing effects on respiratory tracts in ARDS induced by hemorrhagic-endotoxin shock. The numerical density and surface density of alveolar cells, AT1 cell fraction, and numerical density of AT2 and Clara cells were significantly increased after treatment with RA-SS compound in ARDS. Concurrently, the Ki67+ alveolar cells were obviously increased while the TUNEL+ alveolar cells were reduced, which was correlated with the attenuation of inflammatory injury and functional repair in injured lung tissues.Conclusions: Our data convincingly indicated that the prophylactic and therapeutic treatment of RA plus SS had obvious beneficial effect on the remodeling/regeneration of injured pulmonary tissues, suggesting that the underlying mechanisms are related to the re-balance between regeneration and apoptosis in lung stem/progenitor cells. The present study investigated the role of microRNA (miR)-138-5p in regulating carcinoma migration and sensitivity to chemotherapy in pancreatic cancer. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) was used to assess the expression levels of miR-138-5p in pancreatic cancer cell lines and primary carcinoma tissues from human patients. A lentiviral vector, containing miR-138-5p mimics (lv-miR-138-m) or miR-138-5p inhibitor (lv-miR-138-i), was used to either upregulate or downregulate the expression levels of miR-138-5p in PANC-1 cells, respectively. The effects of miR-138-3p regulation on pancreatic cancer cell migration and sensitivity to chemotherapy were examined. The predicted targeting of miR-138-5p on vimentin (VIM) was assessed by western blotting in PANC-1 cells. VIM was subsequently downregulated using small interfering (si)RNA to determine its effect on miR-138-5p-modulated pancreatic cancer cell development. The expression levels of miR-138-5p were downregulated in pancreatic cancer cell lines and primary carcinoma tissues. In PANC-1 cells, lentivirus-mediated upregulation of miR-138-5p inhibited cancer cell migration and increased cell chemosensitivity to 5-fluorouracil (5-FU). By contrast, downregulation of miR-138-5p promoted cancer cell migration and decreased cell chemosensitivity to 5-FU. A luciferase assay revealed that VIM was a direct target of miR-138-5p. Western blotting demonstrated that VIM was downregulated upon the upregulation of miR-138-5p in PANC-1 cells. siRNA-mediated downregulation of VIM inhibited pancreatic cancer cell migration in the control and miR-138-5p downregulated PANC-1 cells. As a lignocellulosic material, wastepaper is a potential material for ethanol production. However, little research on the enzymatic hydrolysis of wastepaper pulp has been conducted. In this study, the enzymatic hydrolysis of different waste pulp fractions (R80 represents greater than 80-mesh wastepaper pulp, R80-180 represents the range of 80- to 180-mesh wastepaper pulp, and R180 represents smaller than 180-mesh waste paper pulp) were carried out at 50 °C, pH 4.8, for 96 h, with a substrate concentration of 5% (w/v) and cellulase loading of 18 FPU/g cellulose. In terms of the specific surface area, fiber structure, and surface morphology, R80-180 had the highest affinity to cellulase and therefore the highest glucose yield of 80.33%. R180 had the lowest glucose yield (55.36%) because of its high ash content (21.36%), which reduced the adsorption of cellulase to cellulose. The enzymatic hydrolysis of R80 mixed with R80 or R80-180 was also studied. Results indicated that adding R80-180 increased the glucose yield of R80. The highest glucose yield (82.57%) was obtained when 15% R80-180 was mixed with R80. However, the glucose content decreased when R180 was mixed with R80 because of its high ash content. Simultaneous saccharification and fermentation (SSF) is an attractive process configuration for bio-ethanol production. Further reductions in process cost of SSF are expected with the use of waste agricultural or industrial materials as feedstock. In the current study, two industrial lignocellulosic wastes, cassava residues (CR) and furfural residues (FR), were combined during SSF for ethanol production due to their value-added applications and positive environmental impacts. After CR were liquefied and saccharified, saccharification liquid was added to SSF of FR. The effect of substrate fractions was investigated in terms of ethanol yield, byproduct concentration and the number of yeast cells. Besides, a natural surfactant, Gleditsia saponin, was added to investigate the effect of FR lignin on SSF with 20% substrate concentration. The results showed that increasing the ratio of CR/FR improved the ethanol yield and that the ethanol yield was also increased gradually by increasing the substrate concentration from 6% to 12%. A high ethanol concentration of 36.0 g/L was obtained under the condition of CR:FR = 2:1 with 12% substrate concentration, reaching 71.1% of the theoretical yield. However, Gleditsia saponin did not affect the ethanol yield, indicating the insignificant effect of lignin in SSF with low lignin content in the reaction system. [ABSTRACT FROM AUTHOR]

Furfural residues (FRs) were pretreated with ethanol and a green liquor (GL) catalyst to produce fermentable sugar. Anthraquinone (AQ) was used as an auxiliary reagent to improve delignification and reduce cellulose decomposition. The results showed that 42.7% of lignin was removed and 96.5% of cellulose was recovered from substrates pretreated with 1.0 mL GL/g of dry substrate and 0.4% (w/w) AQ at 140 °C for 1 h. Compared with raw material, ethanol–GL pretreatment of FRs increased the glucose yield from 69.0% to 85.9% after 96 h hydrolysis with 18 FPU/g-cellulose for cellulase, 27 CBU/g-cellulose for β-glucosidase. The Brauner–Emmett–Teller surface area was reduced during pretreatment, which did not inhibit the enzymatic hydrolysis. Owing to the reduced surface area, the unproductive binding of cellulase to lignin was decreased, thus improving the enzymatic hydrolysis. The article presents a study which compared the bioethanol production from the furfural residue (FR) samples after delignification pretreatment. The relationship between hydrophilicity of the samples and bioethanol production was determined by measuring hydrophilic variation through conductometric titration. Results suggest that some new hydrophilic groups were formed during pretreated delignification which could benefit enzymatic hydrolysis.

Furfural residues (FRs) were pretreated with laccase or a laccasemediator (1-hydroxybenzotriazole, HBT) system to produce fermentable sugar for bioethanol production. Compared to laccase-only pretreatment, laccase-mediator pretreatment dissolved more lignin. Approximately 10.5% of the initially present lignin was removed when FRs were treated with a laccase loading of 100 U/g of dry substrate in 1% (w/w) HBT at 48 °C for 24 h in an acetate buffer (pH 4.8). The enzymatic saccharification process was carried out by a combined laccase or laccase-mediator pretreatment without washing of the treated solids. The results showed that active laccase had a negative effect on the rate and yield of enzymatic hydrolysis. Laccase-oxidized HBT seriously reduced glucose yield. However, non-oxidized HBT increased glucose yield when laccase was deactivated at 121 °C for 20 min prior to enzymatic hydrolysis. The highest glucose yield, 80.9%, was obtained from the substrate pretreated with 100 U/g of dry substrate laccase and 1% (w/w) HBT at 48 °C for 24 h in an acetate buffer (pH 4.8). Low-pressure steam explosion (LPSE) combined with alkaline peroxide (AP) pretreatment was first employed to separate hemicellulose from Lespedeza stalks. The monosaccharide composition and molecular weight distribution of the obtained hemicellulose fractions were characterized in this study. The results show that the hemicellulose extracted from Lespedeza stalks consisted of xylose, glucose, galactose, and mannose, which was a mixture of arabinoxylans and xyloglucans or β-glucans. The yield of hemicellulose fractions after AP pretreatment ranged from 11.2% (2.5% hydrogen peroxide (H2O2), w/v for 12 h) to 12.2% (3.3% H2O2, w/v for 72 h). The molecular weight of hemicellulose decreased from 2,458 g/mol to 1,984 g/mol after AP pretreatment, indicating its degradation reaction. The structure of hemicellulose was analyzed by Fourier transform infrared spectroscopy, proton nuclear magnetic resonance, and heteronuclear single quantum coherence. The AP pretreatment partially cleaved the backbone and the ether linkage between lignin and hemicelluloses. Also, branched-chain α-Darabinofuranosyl in which β-D-xylose substituted at the C-3 position (monosubstituted) was removed, illustrating a partial debranching reaction. Therefore, the combination of low-pressure steam explosion and alkaline peroxide pretreatment (LPSE-AP) is an effective pretreatment method to separate hemicellulose from Lespedeza stalk. Acorn starch was used for ethanol production by separate hydrolysis and fermentation (SHF) in this study. The influence of tannins on hydrolysis and fermentation was investigated using ultrasonic-assisted extraction (UAE) to decrease the amount of tannin before SHF. The tannin was shown to have a negative role in hydrolysis and fermentation, and UAE can improve the two processes. The tannin content of acorn starch decreased from 6.19% to 1.91% with the UAE pretreatment time of 200 min. When the pretreatment time was 120 min, the glucose concentration increased from 78.08 to 98.76 g/L after 24 h of hydrolysis. The highest ethanol concentration was 42.22 g/L, which was obtained from the same pretreated acorn flour fermented for 12 h. However, the maximum ethanol yield was 88.06% of the theoretical yield, while pretreatment time was 80 min. Scanning electron microscope images indicated that protein was separated from the starch granules by UAE, as well as by the molecular weight of starch which decreased significantly based on the results from gel permeation chromatography (GPC) analysis. Background: Enhancement of enzymatic digestibility by some supplementations could reduce enzyme loading and cost, which is still too high to realize economical production of lignocellulosic biofuels. A recent study indicates that yeast hydrolysates (YH) have improved the efficiency of cellulases on digestibility of furfural residues (FR). In the current work, the components of YH were separated by centrifugation and size exclusion chromatography and finally characterized in order to better understand this positive effect. Results: A 60.8% of nitrogen of yeast cells was remained in the slurry (YHS) after hydrothermal treatment. In the supernatant of YH (YHL), substances of high molecular weight were identified as proteins and other UV-absorbing compounds, which showed close molecular weight to components of cellulases. Those substances attributed to a synergetic positive effect on enzymatic hydrolysis of FR. The fraction of YHL ranged from 1.19 to 2.19 mL (elution volume) contained over 50% of proteins in YHL and had the best performance in stimulating the release of glucose. Experiment results proved the adsorption of proteins in YHL on lignin. Conclusions: Supplementation of cellulases with YH enhances enzymatic digestibility of FR mainly by a competitive adsorption of non-enzymatic substances on lignin. The molecular weight of these substances has a significant impact on their performance. Different strategies can be used for a good utilization of yeast cells in terms of biorefinery concept. A combination of steam explosion (SE) and alkaline peroxide (AP) used to pretreat bamboo was investigated. Steam explosion at 224 °C for 4 min was applied to bamboo, and the pretreated bamboo was delignified by alkaline peroxide. Enzymatic hydrolysis was compared in the pretreated samples. Steam pretreatment led to remarkable hemicellulose solubilization (63.2%). Lignin solubilization (93.1%) was achieved by alkaline peroxide treatment of steam-pretreated bamboo at 80 °C for 1 h in 0.88% (v/v) H2O2, whereas only 33.4% of lignin was solubilized when using raw bamboo. Pretreatment methods resulted in a low degree of polymerization and increased hydrolysis of cellulose. A maximum glucose yield of 90.5% was achieved with a combined steam explosion and alkaline peroxide pretreatment. The surface structure of treated bamboo and the adsorption of enzyme on the substrate were characterized by X-ray photoelectron spectroscopy. Delignification decreased enzyme adsorption and increased enzymatic conversion. Furfural residues produced from the furfural industry were investigated as a substrate for lactic acid production by simultaneous saccharification and fermentation (SSF). Alkaline peroxide was used for delignification of furfural residues to improve the final lactic acid concentration. The residue was treated with 1.3% to 1.7% hydrogen peroxide at 80 °C for 1 h with a substrate concentration of 3.33%. SSF of furfural residues with different delignification degrees were carried out to evaluate the effect of delignification degree on lactic acid production. Using corn hydrolysates/furfural residues as substrates, SSF with different media were carried out to investigate the effect of lignin on the interaction between enzymes and lactic acid bacteria. Lactic acid bacteria had a negative effect on cellulase, thus resulting in the reduction of enzyme activity. Lignin and nutrients slowed down the decreasing trend of enzyme activity. A higher delignification resulted in a slower fermentation rate and lower yield due to degradation products of lignin and the effect of lignin on the interaction between enzymes and lactic acid bacteria. Background: Corn cob residue (CCR) is a kind of waste lignocellulosic material with enormous potential for bioethanol production. The moderated sulphite processes were used to enhance the hydrophily of the material by sulfonation and hydrolysis. The composition, FT-IR spectra, and conductometric titrations of the pretreated materials were measured to characterize variations of the CCR in different sulfite pretreated environments. And the objective of this study is to compare the saccharification rate and yield of the samples caused by these variations. Results: It was found that the lignin in the CCR (43.2%) had reduced to 37.8%, 38.0%, 35.9%, and 35.5% after the sulfite pretreatment in neutral, acidic, alkaline, and ethanol environments, respectively. The sulfite pretreatments enhanced the glucose yield of the CCR. Moreover, the ethanol sulfite sample had the highest glucose yield (81.2%, based on the cellulose in the treated sample) among the saccharification samples, which was over 10% higher than that of the raw material (70.6%). More sulfonic groups and weak acid groups were produced during the sulfite pretreatments. Meanwhile, the ethanol sulfite treated sample had the highest sulfonic group (0.103 mmol/g) and weak acid groups (1.85 mmol/g) in all sulfite treated samples. In FT-IR spectra, the variation of bands at 1168 and 1190 cm-1 confirmed lignin sulfonation during sulfite pretreatment. The disappearance of the band at 1458 cm-1 implied the methoxyl on lignin had been removed during the sulfite pretreatments. Conclusions: It can be concluded that the lignin in the CCR can be degraded and sulfonated during the sulfite pretreatments. The pretreatments improve the hydrophility of the samples because of the increase in sulfonic group and weak acid groups, which enhances the glucose yield of the material.
