1. Comprehensive analysis of H3K27me3 LOCKs under different DNA methylation contexts reveal epigenetic redistribution in tumorigenesis.

Abstract

Background: Histone modification H3K27me3 plays a critical role in normal development and is associated with various diseases, including cancer. This modification forms large chromatin domains, known as Large Organized Chromatin Lysine Domains (LOCKs), which span several hundred kilobases. Result: In this study, we identify and categorize H3K27me3 LOCKs in 109 normal human samples, distinguishing between long and short LOCKs. Our findings reveal that long LOCKs are predominantly associated with developmental processes, while short LOCKs are enriched in poised promoters and are most associated with low gene expression. Further analysis of LOCKs in different DNA methylation contexts shows that long LOCKs are primarily located in partially methylated domains (PMDs), particularly in short-PMDs, where they are most likely responsible for the low expressions of oncogenes. We observe that in cancer cell lines, including those from esophageal and breast cancer, long LOCKs shift from short-PMDs to intermediate-PMDs and long-PMDs. Notably, a significant subset of tumor-associated long LOCKs in intermediate- and long-PMDs exhibit reduced H3K9me3 levels, suggesting that H3K27me3 compensates for the loss of H3K9me3 in tumors. Additionally, we find that genes upregulated in tumors following the loss of short LOCKs are typically poised promoter genes in normal cells, and their transcription is regulated by the ETS1 transcription factor. Abstract

Night cultural and tourism consumption areas (NCTCAs) have become an important carrier to light up the city's night-time economic (NTE). Improving the consumption experience of NCTCAs is an effective way to promote the high-quality development of NCTCAs and release their economic potential. Based on the review data of NCTCAs and Ctrip travel platform above the provincial level, this paper analyzes the spatial distribution characteristics of NCTCAs and their Internet word-of-mouth (IWOM) by combining nearest neighbor index and kernel density estimation methods, and further discusses the influencing factors of IWOM of NCTCAs based on Geodetector. It is found as follows. ➀ The NCTCAs in China are mainly distributed in the southeast of Hu Line, and the different types of NCTCAs have spatial differences. ➁ The IWOM of the NCTCAs is generally good and presents a spatial pattern of two poles and multiple cores. The high-value areas of IWOM are basically distributed in the diamond arrangement area in east of Hu Line, while the core areas and sub-core areas are mostly located in the T-shaped development axis zone in China. ➂ Socio-economic factors such as urban consumption power, tertiary industry and population have a strong influence on IWOM of NCTCAs. Among them, total retail sales of consumer goods are the dominant factor, which together with the Baidu search index constitute the leading interactive factors of IWOM for various NCTCAs. Therefore, in the process of construction and development of NCTCAs, we should reasonably cultivate and lay out the economic growth poles at night, improve consumer satisfaction and promote the formation of high-quality NCTCAs. 12. Virtual data generation method for simulation scenes based on scene flow prediction

Abstract

A virtual data generation method based on scene flow prediction is proposed to address the problem of high false positives and inability to ensure feature consistency after virtual data encoding in simulation scenarios. Firstly, a variational auto-encoder is introduced to determine the relationship between virtual data in different dimensions through back-propagation performance; then, with the help of deep semantic information, automatic annotation of simulation scene data is completed; finally, different displacement changes in the three-dimensional space of the data object are determined to obtain dense scene streams. Based on this, the scene stream prediction method is used to encode the entire virtual data and fuse it. Combined with the matching of encoded features, the consistency of the encoded virtual data features is ensured. Finally, the objective function is introduced, and a data generator is set up to achieve virtual data generation in simulation scenarios. The results show that the proposed method reduces the false positive rate of virtual data generation in simulation scenarios and has good performance.

27. Optimization of Injection Molding Process Parameters for Automobile Gearlever Based on Orthogonal Experiment.

Abstract

Aiming at various quality defects in the injection molding process of automobile gear lever, the warpage was selected as an optimization objective. The relationship between the experimental factors of mold temperature, melt temperature, injection molding time, packing pressure and holding time and the warpage deformation as experimental index were studied by means of the Taguchi experiment design method and Moldflow software. The order of influence degree for each factor on the warpage deformation of gear lever as well as the optimum combination of factors and levels were found. Then numerical simulation and injection molding were carried out to verify the validity of these results. As a result, the goal for improving the injection quality and shortening the production cycle of gear lever products was achieved. [ABSTRACT FROM AUTHOR]

Overall, little is known about the toxicological effects of SiNPs on the cardiovascular system. In this study, SiNPs with a 58 nm diameter were used to study their interaction with human umbilical vein endothelial cells (HUVECs). Dose- and time-dependent decrease in cell viability and damage on cell plasma-membrane integrity showed the cytotoxic potential of the SiNPs. SiNPs were found to induce oxidative stress, as evidenced by the significant elevation of reactive oxygen species generation and malondialdehyde production and downregulated activity in glutathione peroxidase. SiNPs also stimulated release of cytoprotective nitric oxide (NO) and upregulated inducible nitric oxide synthase (NOS) messenger ribonucleic acid, while downregulating endothelial NOS and ET-1 messenger ribonucleic acid, suggesting that SiNPs disturbed the NO/NOS system. SiNP-induced oxidative stress and NO/NOS imbalance resulted in endothelial dysfunction. SiNPs induced inflammation characterized by the upregulation of key inflammatory mediators, including IL-1β, IL-6, IL-8, TNFα, ICAM-1, VCAM-1, and MCP-1. In addition, SiNPs triggered the activation of the Nrf2-mediated antioxidant system, as evidenced by the induction of nuclear factor-κB and MAPK pathway activation. This study is aimed to clarify the biological behavior and influence of silica nanoparticles (Nano-SiO2) on endothelial cell function. The results showed that the Nano-SiO2 were internalized into endothelial cells in a dose-dependent manner. Monodansylcadaverine staining, autophagic ultrastructural observation, and LC3-I/LC3-II conversion were employed to verify autophagy activation induced by Nano-SiO2, and the whole autophagic process was also observed in endothelial cells. In addition, the level of nitric oxide (NO), the activities of NO synthase (NOS) and endothelial (e)NOS were significantly decreased in a dose-dependent way, while the activity of inducible (i)NOS was markedly increased. The expression of C-reactive protein, as well as the production of proinflammatory cytokines (tumor necrosis factor α, interleukin [IL]-1β, and IL-6) were significantly elevated. Moreover, Nano-SiO2 had an inhibitory effect on the phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt)/mammalian target of rapamycin (mTOR) signaling pathway. Our findings demonstrated that Nano-SiO2 could disturb the NO/NOS system, induce inflammatory response, activate autophagy, and eventually lead to endothelial dysfunction via the PI3K/Akt/mTOR pathway.
