Lanthanum lithium titanate is one of the promising electrolytes for solid-state lithium-ion batteries due to its high bulk ionic conductivity up to ∼10−3 S/cm. However, the practical application of this material has been bottlenecked by high grain boundary (GB) resistance, while the underlying mechanism is still under debate. Here we report a comprehensive study with direct evidence to reveal the origin of high GB resistance in La2/3–xLi3xTiO3 (LLTO). Atomic-scale observations via advanced scanning transmission electron microscopy show that the GBs are uniformly subject to subsurface segregation of La atoms to compensate for the excess surface charges. The La segregation leads to an abrupt decrease of charge carrier concentration neighboring GBs and hence is supposed to have deleterious effect on GB conductivity. The findings suggest a novel mechanism of space-charge-induced cation segregation, which shed lights on the intrinsic origin of low GB ionic conductivity in LLTO.

Abstract A rotor speed signature analysis‐ (RSSA‐)based inter‐turn short circuit (ITSC) fault diagnostic method is proposed, which is robust with regard to speed‐ and current‐loop bandwidths of permanent magnet synchronous machine (PMSM) drive systems. Conventional ITSC fault detection solutions that rely on current signals or extra devices. Thus, an initial step of investigation on the validity of RSSA is taken for residual insulation capacity monitoring of the ITSC fault at the incipient stage. The Vold–Kalman filtering order tracking method is employed for the real‐time extraction of fault features. Besides, the impact of speed‐ and current‐loop controller bandwidths on ITSC fault diagnosis is also analysed and an Adaline estimator is designed to decouple their impacts. Finally, the effectiveness of the proposed method is verified on a faulty PMSM, which exhibits satisfying results in tracking the degradation of residual insulation, even if the phase currents are significantly distorted due to low switching frequency of the inverter.

Abstract Background Inter-subspecific hybrid rice represents a significant breakthrough in agricultural genetics, offering higher yields and better resilience to various environmental stresses. While the utilization of these hybrids has shed light on the genetic processes underlying hybridization, understanding the molecular mechanisms driving heterosis remains a complex and ongoing challenge. Here, chromatin immunoprecipitation-sequencing (ChIP-seq) was used to analyze genome-wide profiles of H3K4me3 and H3K27me3 modifications in the inter-subspecific hybrid rice ZY19 and its parents, Z04A and ZHF1015, then combined them with the transcriptome and DNA methylation data to uncover the effects of histone modifications on gene expression and the contribution of epigenetic modifications to heterosis. Results In the hybrid, there were 8,126 and 1,610 different peaks for H3K4me3 and H3K27me3 modifications when compared to its parents, respectively, with the majority of them originating from the parental lines. The different modifications between the hybrid and its parents were more frequently observed as higher levels in the hybrid than in the parents. In ZY19, there were 476 and 84 allele-specific genes with H3K4me3 and H3K27me3 modifications identified, representing 7.9% and 12% of the total analyzed genes, respectively. Only a small portion of genes that showed differences in parental H3K4me3 and H3K27me3 modifications which demonstrated allele-specific histone modifications (ASHM) in the hybrid. The H3K4me3 modification level in the hybrid was significantly lower compared to the parents. In the hybrid, DNA methylation occurs more frequently among histone modification target genes. Additionally, over 62.58% of differentially expressed genes (DEGs) were affected by epigenetic variations. Notably, there was a strong correlation observed between variations in H3K4me3 modifications and gene expression levels in the hybrid and its parents. Conclusion These findings highlight the substantial impact of histone modifications and DNA methylation on gene expression during hybridization. Epigenetic variations play a crucial role in controlling the differential expression of genes, with potential implications for heterosis.

With Polygonatum cyrtonema tender buds as subjects, Polygonatum cyrtonema tender bud tea was prepared using the technology for processing flat green tea after fixation by microwave, pan frying and steaming, respectively. The effects of different fixation methods on the quality of Polygonatum cyrtonema tender bud tea was then explored through physicochemical composition, color and gloss of tea broth, volatile aroma component analysis and organoleptic evaluation. The results showed that the Polygonatum cyrtonema tender bud tea prepared by microwave fixation not only presented good organoleptic properties, but also had the highest contents of water extract, soluble sugar, tea polyphenols and total flavonoids, 48.04%, 14.49%, 1.32% and 1.66%, respectively. A total of 84 volatile olfactory compounds were detected in Polygonatum cyrtonema tender bud tea prepared by the three different fixation methods, mainly consisting of aldehydes (12 types), ketones (11 types), olefins (10 types), alcohols (8 types) and esters (8 types). There were 10, 12 and 7 key aroma components in Polygonatum cyrtonema tender bud tea with microwave, pan frying and steaming fixation, respectively. Among them, ethyl butyrate was a unique key aroma component to microwave fixation, while heptanal, pentanal, hexanal and 1-pentanol were the key aroma components unique to pan frying fixation. Overall, there are differences in the quality of Polygonatum cyrtonema tender bud tea prepared by different fixation methods, among which microwave fixation is more suitable for the preparation of Polygonatum cyrtonema tender bud tea.

Abstract Plant polyploidization increases the complexity of epigenomes and transcriptional regulation, resulting in genome evolution and enhanced adaptability. However, few studies have been conducted on the relationship between gene expression and epigenetic modification in different plant tissues after allopolyploidization. In this study, we studied gene expression and DNA methylation modification patterns in four tissues (stems, leaves, flowers and siliques) of Brassica napusand its diploid progenitors. On this basis, the alternative splicing patterns and cis-trans regulation patterns of four tissues in B. napus and its diploid progenitors were also analyzed. It can be seen that the number of alternative splicing occurs in the B. napus is higher than that in the diploid progenitors, and the IR type increases the most during allopolyploidy. In addition, we studied the fate changes of duplicated genes after allopolyploidization in B. napus. We found that the fate of most duplicated genes is conserved, but the number of neofunctionalization and specialization is also large. The genetic fate of B. napus was classified according to five replication types (WGD, PD, DSD, TD, TRD). This study also analyzed generational transmission analysis of expression and DNA methylation patterns. Our study provides a reference for the fate differentiation of duplicated genes during allopolyploidization.

BackgroundXiaojin Pill (XJP) is a traditional Chinese medicine prescribed for treating benign prostatic hyperplasia (BPH). It has been proven to have multiple effects, such as regulating sex hormone levels, exhibiting anti-tumor, anti-inflammatory, analgesic, and anti-platelet aggregation properties, and improving immunity. However, the material basis of XJP's therapeutic effect on BPH and its metabolic process in vivo remains to be clarified. At the same time, many microorganisms that exist in the urogenital tract, including those related to BPH, can also affect the health of the host.MethodsUsing ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), the chemical components of XJP were identified. A BPH model was created through bilateral testicular ablation and injections of testosterone propionate. A comprehensive evaluation of XJP efficacy was conducted using pathological ELISA, TUNEL, and immunohistochemical techniques. In addition, UPLC-MS metabolomics and 16S rRNA sequencing revealed the serum metabolic profile and intestinal microbiota composition. We performed a Spearman correlation coefficient analysis to highlight the interactions between “intestinal microbiota-serum factors” and “intestinal microbiota-metabolites.”ResultsXJP contains 91 compounds that alleviate pathologies of BPH in rats, decreasing prostate weight, index, and serum levels of Dihydrotestosterone (DHT), Prostate-Specific Antigen (PSA), epidermal growth factor (EGF), basic fibroblast growth factor (bFGF), and vascular endothelial growth factor (VEGF) levels. It inhibits prostatic epithelial cell apoptosis and downregulates Bax, TGF-β1, and IGF-1 proteins in the caspase-3 pathway. Metabolomics studies have revealed 10 upregulated and 10 downregulated metabolites in treated rats, with 5-methylcytosine, uracil, and cytosine enriched in pyrimidine metabolism. L-arginine plays a pivotal role in metabolic pathways encompassing pyrimidine metabolism, arginine biosynthesis, and the mammalian target of rapamycin (mTOR) signaling pathway. 16S rRNA sequencing revealed that XJP optimized the diversity and balance of intestinal flora in BPH rats by decreasing the Bacteroidetes/Firmicutes (B/F) ratio, enhancing the beneficial bacteria, such as Eggerthellaceae, Anaerovoracaceae, and Romboutsia, and suppressing the dysfunctional bacteria, such as Atopobiaceae, Prevotellaceae_NK3B31_group, Dorea, and Frisingicoccus. According to the Spearman correlation coefficient analysis, Lactobacillus was found to be most associated with serum factors, whereas Romboutsia showed the highest correlation with metabolites. This finding suggests that XJP modulates pyrimidine metabolism disorders in BPH rats, a regulation that aligns closely with Romboutsia, Prevotellaceae_NK3B31_group, Lactobacillus, Chujaibacter, and Enterorhabdus, thereby providing valuable biological insights.ConclusionIn summary, these findings indicate that XJP possesses a synergistic anti-BHP effect through its multi-component, multi-target, multi-gut microbiota, and multi-metabolic pathway properties. The effect involves the regulation of sex hormone levels, growth factors, and the anti-epithelial cell apoptosis process. The modulation of specific gut microbiota by the host and the involvement of multiple metabolic pathways are likely one of the significant mechanisms of XJP in treating BPH. Notably, pyrimidine metabolism and the intestinal microbial ecosystem are closely intertwined in this process.

As an energy-saving device for energy transduction, a steam ejector has been crucial in energy saving and emission reduction, energy consumption reduction, and energy reuse. An optimization method of full geometric parameters and a preferred method of working conditions were considered to rise the entrainment ratio of a steam ejector and improve its entrainment capacity. The related models of the ejector were established and numerically simulated. The influence of geometric parameters on the entrainment capacity of the ejector researched by adopting the control variable method and the 11-factor 5-level orthogonal test. The effect of working conditions on the entrainment ratio for the optimized ejector discussed, and the preferred method of working conditions of the ejector proposed. The results show that the entrainment ratio of the optimized ejector by the control variable method rises by 30.8 % to that of the original ejector, and the entrainment ratio of the new ejector by orthogonal test rises by 78 %. The constant cross-section diameter is a critical parameter affecting the entrainment capacity of the ejector. Contrasted with the original ejector, the maximum entrainment ratio under preferred working conditions rises by 242%–471 % via using the preferred method of working conditions. Considering the optimization of full geometric parameters and working conditions, the influence of the optimized process on the entrainment capacity of ejectors is evident, which provides an effective way and reference for the optimization research and development of other types of ejectors.

The gain-driven emission in gain–loss coupling systems attains significant interest. In this work, we study the monochromatic oscillation of a gain–loss coupling system through a circuit model, where the monochromacy is manifested as the gain-driven synchronization of two coupled oscillators. The synchronized oscillation in this model is theoretically studied and experimentally manifested. Benefiting from the conceptual simplicity of a circuit model, a pair of asymmetric long-living states is found among the unstable gain-driven monochromatic oscillations, corresponding to bound states in the continuum of coupling physics.

Along with the increasing low-carbon demand of the power system, the access of a high percentage of renewable energy resources to the distribution network has a large impact on the voltage fluctuation of the system and reduces the operational reliability. In this paper, we consider utilizing the reactive capacity of distributed resources to participate in system voltage regulation to reduce node loss of load probability (LOLP) caused by node overvoltage faults and propose an overvoltage risk regulation strategy for the interaction between distribution network operators (DSOs) and distributed users in the framework of the Stackelberg game. First, the nodes are clustered and analyzed based on the two-dimensional indexes of node voltage regulation ability, and different voltage regulation compensation tariffs are assigned. Second, the cost-benefit model of voltage regulation for the leader and follower sides and the node LOLP model are constructed to measure the reliability of the system. The Stackelberg game is used to co-optimize the two parties’ compensation tariffs and voltage regulation strategies. The optimal solution of voltage regulation under the equilibrium of the game is obtained by solving using the particle swarm optimization (PSO) algorithm. Based on the IEEE-33 node system, a case study is carried out to verify that the proposed overvoltage risk regulation strategy can maximize the benefits of the regulator participants while enhancing the operational reliability of the system.

Abstract Multi-dimensional heterojunction materials have attracted much attention due to their intriguing properties, such as high efficiency, wide band gap regulation, low dimensional limitation, versatility and scalability. To further improve the performance of materials, researchers have combined materials with various dimensions using a wide variety of techniques. However, research on growth mechanism of such composite materials is still lacking. In this paper, the growth mechanism of multi-dimensional heterojunction composite material is studied using quasi-two-dimensional (quasi-2D) antimonene and quasi-one-dimensional (quasi-1D) antimony sulfide as examples. These are synthesized by a simple thermal injection method. It is observed that the consequent nanorods are oriented along six-fold symmetric directions on the nanoplate, forming ordered quasi-1D/quasi-2D heterostructures. Comprehensive transmission electron microscopy (TEM) characterizations confirm the chemical information and reveal orientational relationship between Sb2S3 nanorods and the Sb nanoplate as substrate. Further density functional theory calculations indicate that interfacial binding energy is the primary deciding factor for the self-assembly of ordered structures. These details may fill the gaps in the research on multi-dimensional composite materials with ordered structures, and promote their future versatile applications. Graphical Abstract

Utilizing spatial diversity, the distributed multiple-input multiple-output (MIMO) radar has the potential advantage of improving system detection performance. In this paper, the incoherent detection performance of distributed multiple-input multiple-output (MIMO) radars is investigated for Rice fluctuating targets. To calculate the incoherent detection probability, the moment generating function (MGF) of the Rice variable is expanded as the infinite series form. By inverting the product of MGFs of multiple independent Rice variables, new closed-form expressions for the probability density function (PDF) of the sum of independent and weighted squares of Rice variables are proposed. The proposed PDF expression for the sum of independent, non-identically distributed (i.n.i.d.) Rice variables involves an infinite series in terms of the confluent Lauricella function. Specially, the PDF for the sum of independent identically distributed (i.i.d.) Rice is expressed as the confluent hypergeometric function-based infinite series. In addition, the uniform convergence of the proposed PDF expression is also validated. Using this proposed expression, the closed-form and approximate expressions of the incoherent detection probability of MIMO radar are derived, respectively. Numerically evaluated results are illustrated and compared with Monte Carlo (MC) simulations to validate the accuracy of the derivations.

Background In China, esophageal cancer is one of the most common cancers, and its incidence and mortality are more than twice the global average. However, most esophageal cancers in China are not detected until they are at moderate and advanced stages with poor prognosis, so early diagnosis and treatment are urgently needed. At present, the role of gut flora in the diagnosis and treatment of esophageal cancer has attracted much attention. Objective To explore the basic characteristics of gut flora in patients with esophageal squamous cell carcinoma. Methods From the Fourth Hospital of Hebei Medical University from April to August 2022, 35 patients with esophageal squamous cell carcinoma with no previous anti-cancer treatments were included as esophageal cancer group (EC group), and 35 healthy volunteers as control group. Stool samples of the two groups were collected to detect the gut flora using 16S rDNA sequence. Then an analysis was performed on the Alpha diversity, Beta diversity〔using the principal coordinate analysis (PCoA) 〕 and differential species of gut flora 〔using the linear discriminant analysis Effect Size (LEfSe) 〕 of two groups according to the species annotation of the test results. Results The Alpha diversity indices, such as Shannon index, Simpson index, Chao1 index, ACE index and Goods_coverage index demonstrated no significant differences between the two groups (P>0.05), while PCoA graph showed that the Beta diversity differed significantly between the two groups on the whole, and the community structures were different (t=10.837, P

Islands and the mainland are separated by seas, and the distances between them might be so long that the height on the mainland cannot be exactly translated to the islands, resulting in different height systems on the mainland and the islands. In this study, we used astrogeodetic deflections of the vertical and ellipsoidal heights of points on the mainland and island near their coastlines to implement height connection across sea areas. First, the modeled gravity and modeled astrogeodetic vertical deflections of segmentation points along connecting routes over the sea between the mainland and the island were determined by Earth Gravity Model (EGM), and the ellipsoidal heights of segmentation points were determined by the satellite altimetry data sets. Second, we used a linear interpolation model to increase the precision of the vertical deflections of segmentation points. Third, we computed the geopotential difference of points between the mainland and the island using a method derived from geopotential theory and the astronomical leveling principle. Finally, we estimated the normal height of the point on the island using the geopotential-difference iterative computation approach. Using observed data of normal heights, ellipsoidal heights, and astrogeodetic vertical deflections referring to height sites in Qingdao, Shandong Province, we conducted a numerical experiment involving the normal height connection across sea regions. We determined the data of the ellipsoidal heights and gravity of segmentation points along the connecting route across the water in the numerical experiment using DTU10. The distance of the height connection across the sea was approximately 10.5 km. According to China's official leveling specifications, the experimental results met the criterion of third-class leveling precision.

Abstract Tumor suppressor gene CHFR (The Checkpoint with Forkhead-associated and Ring finger domains) is a mitotic checkpoint and frequently hypermethylated in gastric cancer. Our previous study found CHFR played a certain extent pro-tumor function in gastric cancer. However, little is known about the underlying mechanism. In this study, we tried to further elucidate the role and mechanism for CHFR in gastric cancer (GC) by constructing CHFR stably expressed cell lines. As expected, the ectopic expression of CHFR slowed the cell proliferation in both two SGC-7901 and AGS cells, while significantly promoted the potential of cell migration and invasion. For the first time, our data indicated that stable expression of CHFR in SGC-7901 and AGS restrained cellular reactive oxygen species (ROS) generation and promoted the activation of AKT and ERK, two regulators of redox hemostasis. Furthermore, H2O2 treatment effectively elevated ROS level and reversed CHFR-induced cell invasion in stable SGC-7901 and AGS cells with the decreased phosphorylation of AKT and ERK. We also confirmed that CHFR exerted its function by promoting NRF2 expression. The most important is, the ectopic expression of CHFR significantly inhibited SGC-7901 cell-derived xenografts and obviously promoted lung metastasis of GC cell with NRF2, p-AKT and p-ERK increased. Taken together, our findings suggested that CHFR might take part in gastric cancer progression especially cancer metastasis by activating AKT and ERK via NRF2- ROS axis.

Early response assessment is critical for personalizing cancer therapy. Emerging therapeutic regimens with encouraging results in the wild-type (WT) KRAS colorectal cancer (CRC) setting include inhibitors of epidermal growth factor receptor (EGFR) and glutaminolysis. Towards predicting clinical outcome, this preclinical study evaluated non-invasive positron emission tomography (PET) with (4S)-4-(3-[18F]fluoropropyl)-L-glutamic acid ([18F]FSPG) in treatment-sensitive and treatment-resistant WT KRAS CRC patient-derived xenografts (PDXs). Tumor-bearing mice were imaged with [18F]FSPG PET before and one week following the initiation of treatment with either EGFR-targeted monoclonal antibody (mAb) therapy, glutaminase inhibitor therapy, or the combination. Imaging was correlated with tumor volume and histology. In PDX that responded to therapy, [18F]FSPG PET was significantly decreased from baseline at 1-week post-therapy, prior to changes in tumor volume. In contrast, [18F]FSPG PET was not decreased in non-responding PDX. These data suggest that [18F]FSPG PET may serve as an early metric of response to EGFR and glutaminase inhibition in the WT KRAS CRC setting.

Abstract Background Histologically, cytoplasmic deposits of lipids and glycogen are common in clear cell renal cell carcinoma (ccRCC). Owing to the significance of lipid deposition in ccRCC, numerous trials targeting lipid metabolism have shown certain therapeutic potential. The agonism of peroxisome proliferator-activated receptor-α (PPARα) via ligands, including WY-14,643, has been considered a promising intervention for cancers. Methods First, the effects of WY-14,643 on malignant behaviors were investigated in ccRCC in vitro. After RNA sequencing, the changes in lipid metabolism, especially neutral lipids and glycerol, were further evaluated. Finally, the underlying mechanisms were revealed. Results Phenotypically, the proliferation and migration of ccRCC cells treated with WY-14,643 were significantly inhibited in vitro. A theoretical functional mechanism was proposed in ccRCC: WY-14,643 mediates lipid consumption by recognizing carnitine palmitoyltransferase 1 A (CPT1A). Activation of PPARα using WY-14,643 reduces lipid deposition by increasing the CPT1A level, which also suppresses the NF-κB signaling pathway. Spatially, WY-14,643 binds and activates PPARα by targeting Gly335. Conclusion Overall, WY-14,643 suppresses the biological behaviors of ccRCC in terms of cell proliferation, migration, and cell cycle arrest. Furthermore, its anticancer properties are mediated by the inhibition of lipid accumulation, at least in part, through the PPARα/CPT1A axis by targeting Gly335, as part of the process, NF-κB signaling is also suppressed. Pharmacological activation of PPARα might offer a new treatment option for ccRCC.

Retroperitoneal tuberculosis is a rare disease that can mimic many conditions and lacks specific clinical manifestations, which makes it difficult to diagnose. As a consequence, it can be misdiagnosed as a malignant tumour. Endoscopic ultrasonography-guided fine-needle aspiration (EUS-FNA) can obtain specimens of the lesion site from areas that might be inaccessible using more traditional biopsy methods. A 60-year-old female patient was admitted with a 3-month history of intermittent upper abdominal pain accompanied by nausea. Imaging found pancreatic uncinate process and retroperitoneal lymph nodes at the horizontal part of the duodenum. EUS-FNA found necrotic matter, multinucleated giant cells and epithelioid cells consistent with the signs of tuberculosis bacilli infection, although typical noncaseous granuloma and Mycobacterium tuberculosis were not observed. Retroperitoneal tuberculosis was considered as the diagnosis. After anti-tubercular therapy, the signs and symptoms quickly improved and a repeat computed tomography scan found that the space-occupying lesion had reduced in size. By using EUS-FNA, the cytological and histopathological findings can be obtained in a timely manner to facilitate an earlier diagnosis and avoid unnecessary procedures such as laparotomy or surgery.

Background: Although immunotherapy greatly extends overall survival (OS) of patients with extensive-stage small cell lung cancer (ES-SCLC), a number of patients develop immunotherapy resistance (IR). Patterns of failure in ES-SCLC are not clarified. Our study aims to explore the clinical pattern of IR and prognostic factors for these patients. Methods: The study was conducted from 117 ES-SCLC patients with immunotherapy between 2018 and 2022. Chi-square tests and Fishers' exact tests was used to explore failure patterns in different populations. Survival analyses of different progression patterns and subsequent treatment regimens were conducted by Kaplan–Meier curves and log-rank test. Results: 86 (73.5%) patients experienced IR. The patients with smoking (never smoker vs. current or ex-smoker, 59.5 % vs. 81.3%, P = 0.010), liver metastasis (extrahepatic metastasis vs. intrahepatic metastasis, 73.6 % vs. 90.9%, P = 0.050), and distant metastasis status (no distant metastasis vs. distant metastasis, 39.1 % vs. 81.9%, P

Soil salinization negatively affects rice growth and yield; however, how different sludge sources regulate rice growth and yield under salt stress was rarely investigated. This study evaluated the performance of two salt-tolerant rice cultivars, Chaoyou 1000 and Longliangyou 506, grown in two sediment sources, pond sediment (PS) and river sludge (RS), under salt stress (56 ds m−1 brine irrigation) with conventional soil (CS) used as the control. The results showed that the rice yield under the PS and RS treatments was enhanced by 51.0% and 43.6% as compared with CS, respectively, owing to an improvement in spikelet per panicle, 1000-grain weight, dry matter accumulation, and the chlorophyll content in both rice cultivars. Compared with CS, the total nitrogen accumulation, nitrogen grain production efficiency, nitrogen harvest index, and nitrogen partial productivity under the PS and RS treatments were increased by 18.9–28.9%, 17.0–20.6%, 7.2–16.6%, and 43.8–50.9%, respectively. Moreover, rice grown in PS and RS showed higher activities of nitrogen metabolism-related enzymes (nitrate reductase, glutamine synthetase, and glutamate synthetase) at the heading stage and higher K+ and K+/Na+ contents in the leaves. Overall, a balanced utilization of sediment resources (especially pond sediment) can effectively alleviate salt stress and improve the yield and nitrogen use efficiency in rice.

The nanometric length traceability chain is the foundation of cutting-edge research at the nanoscale. The current traceability chain has limitations in its applications. However, the application performance of the nanometric traceability chain based on the ^7 S _3 - ^7 P _4 transition in chromium atoms has been fully validated. Addressing the issue that the ^7 S _3 - ^7 P _4 transition frequency at the traceability side of this chain has not been measured in a vacuum environment, this study used the fluorescence symmetry method under vacuum conditions to measure the ^7 S _3 - ^7 P _4 transition frequency, obtaining the corresponding vacuum wavelength of 425.5533474 nm and the vacuum wave number of 23498.81645 cm ^−1 .

Abstract Concerning the difficulty in determining the restart strategy for AC/DC interconnected systems, this paper proposes a method of calculating the optimal restart strategy for AC/DC interconnected systems based on dynamic energy. First, a dynamic energy function that characterises the stability of system is constructed. Then, the analytical expressions of the dynamic energy function are calculated separately for the deionisation stage, the restart stage, and the alpha retard stage of the entire restart process. On this basis, the variation rate of the system dynamic energy is defined as the energy accumulation rate, which quantifies the accumulation or consumption of dynamic energy during the restart process, and facilitates detailed analysis of the effects of the restart time and restart duration on the stability of system. Then, a method to calculate the optimal restart time is proposed with the goal of minimising the dynamic energy accumulated in the system. Finally, hardware‐in‐the‐loop tests are carried out on the RT‐LAB platform for simulation verification. The experimental results show that the dynamic energy function constructed in this paper can reflect the stability of system. Using the proposed optimal restart strategy can reduce the accumulation of dynamic energy and improve the stability of system.

Changes in chromatin accessibility occuring during the process of allopolyploidization of Brassica napus are analysed using ATAC and ChIPseq, with differences in asymmetric chromatin accessibility between subgenomes of B. napus investigated.

Objectives: Bone marrow-derived mesenchymal stem cells (BMSCs) are considered to have potential clinical application value in the treatment of acute lung injury (ALI). Myeloid-derived growth factor (MYDGF) can promote the proliferation of stem cell. We hypothesized that MYDGF may play a role in reducing lung injury in vitro and in vivo through bone marrow mesenchymal stem cells. Methods: An in vitro model of lipopolysaccharide (LPS)(MLE-12) was established, which was divided into five groups: A: MLE-12; B: MLE-12+LPS; C: MLE-12+LPS + BMSCs; D: MLE-12+LPS + MYDGF; and E: MLE-12+LPS + BMSCs + MYDGF. A Cell Counting Kit-8 was used to detect the OD value. And an ALI model was constructed by inducing mice with a lipopolysaccharide. Forty male Balb/c mice were randomly divided into five groups: A control group; B: model group; C: LPS + BMSCs; D: LPS + MYDGF; E: LPS +BMSCs +MYDGF. Specimens were collected after 24 h. Hematoxylin-eosin (HE)-staining was performed on the tissue sections. The protein concentration in the alveolar lavage fluid was measure by bicinchoninic acid (BCA). The NF-κB, p-Akt, Bax, and Bcl-2 protein expression was detected through Western blotting, and Enzyme linked immunosorbent assay (ELISA) was used to measure the expression of serum interleukin-6, interleukin-10, and TNF-α. Results: Compared with the model group, BMSCs and MYDGF can alleviate the ALI induced by lipopolysaccharide in vitro and vivo ( p < .05). Conclusion: We found that the combined treatment effect of MYDGF and BMSCs was better than using MYDGF or BMSCs alone. We speculate that a pretreatment with MYDGF after ALI in mice may improve the survival and growth of transplanted MSCs, thereby improving the curative effect of cell transplantation.

In this study, we not only optimized and improved the synthesis process of levobupivacaine hydrochloride (21) but also conducted a comprehensive exploration of critical industrial-scale production details, and a novel high-performance liquid chromatography (HPLC) analysis method was developed. Starting with the readily available and cost-effective (R,S)-N-(2,6-dimethylphenyl)piperidine-2-carboxamide (28) as the initial material and utilizing l-(–)-dibenzoyl tartaric acid (29) for chiral separation, and then through substitution and a salting reaction, levobupivacaine hydrochloride (21) was obtained with high purity (chemical purity of 99.90% and enantiomeric excess (ee) values of 99.30%). The total yield of the three steps was 45%. Structures of intermediates and the final product were confirmed using nuclear magnetic resonance (NMR) (1H NMR, 13C NMR), mass spectrometry (MS), and elemental analysis. The crystal structure of the final product was determined through differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and X-ray diffraction (XRD). Furthermore, we evaluated the risk of the substitution reaction using a reaction calorimeter and accelerating rate calorimetry (ARC). This process offers the advantages of simple operation, greenness, safety, controllable quality, and cost-effectiveness. It provides reliable technical support for the industrial-scale production of levobupivacaine hydrochloride (21), which is of significant importance in meeting clinical demands. Pilot-scale production has already been successfully completed by China National Medicines Guorui Pharmaceutical Co., Ltd., with a production scale of 20 kg.

Abstract Background Polyploidization promotes species formation and is widespread in angiosperms. Genome changes dramatically bring opportunities and challenges to plants after polyploidy. Methyl-CpG-Binding Domain (MBD) proteins can recognize and bind to methylation sites and they play an important role in the physiological process related to methylation in animals and plants. However, research on the influence of the allopolyploidization process on the MBD gene family is still lacking, so it is necessary to conduct a comprehensive analysis. Results In this study, twenty-two, ten and eleven MBD genes were identified in the genome of allotetraploid B. napus and its diploid ancestors, B. rapa and B. oleracea, respectively. Based on the clades of the MBD gene in Arabidopsis, rice and maize, we divided the new phylogenetic tree into 8 clades. Among them, the true MBD genes in Brassica existed in only 5 clades. Clade IV and Clade VI were unique in term of MBD genes in dicotyledons. Ka/Ks calculations showed that MBD genes underwent purifying selection in Brassica and may retain genes through sequence or functional differentiation early in evolution. In the process of allopolyploidization, the number of MBD gene introns increased, and the protein motifs changed. The MBD proteins had their own special motifs in each clade, and the MBD domains were only conserved in their clades. At the same time, the MBD genes were expressed in flower, leaf, silique, and stem tissues, and the expression levels of the different genes were significantly different, while the tissue specificity was not obvious. The allopolyploidization process may increase the number of cis-acting elements and activate the transposable elements. During allopolyploidization, the expression pattern of the MBD gene changes, which may be regulated by cis-acting elements and transposable elements. The number imbalance of cis-acting elements and transposable elements in An and Cn subgenomes may also lead to biased An subgenome expression of the MBD gene in B. napus. Conclusions In this study, by evaluating the number, structure, phylogeny and expression of the MBD gene in B. napus and its diploid ancestors, we increased the understanding of MBD genes in allopolyploids and provided a reference for future analysis of allopolyploidization.
