Abstract Neovaginas are surgically constructed to correct uterovaginal agenesis in women with Mayer-Rokitansky-Küster-Hauser (MRKH) syndrome or as part of gender-affirming surgery for transfeminine individuals. Understanding the assembly of the neovaginal microbiota is crucial for guiding its management. To address this, we conducted a longitudinal study on MRKH patients following laparoscopic peritoneal vaginoplasty. Our findings reveal that the early microbial assemblage exhibited stochastic characteristics, accompanied with a notable bloom of Enterococcus faecalis and genital Mycoplasmas. While both the pre-surgery dimple microbiota and the fecal microbiota constituted the primary species pool, the neovaginal microbiota developed into a microbiota that resembled that of a normal vagina at 6–12 months post-surgery, albeit with a bacterial vaginosis (BV)-like structure. By 2–4 years post-surgery, the neovaginal microbiota had further evolved into a structure closely resembling with the homeostatic pre-surgery dimple microbiota. This concords with the development of the squamous epithelium in the neovagina and highlights the pivotal roles of progressive selective forces imposed by the evolving neovaginal environment and the colonization tropism of vaginal species. Notably, we observed that strains of Lactobacillus crispatus colonizing the neovagina primarily originated from the dimple. Since L. crispatus is generally associated with vaginal health, this finding suggests potential avenues for future research to promote its colonization.

Abstract Computer-aided implant surgery has undergone continuous development in recent years. In this study, active and passive systems of dynamic navigation were divided into active dynamic navigation system group and passive dynamic navigation system group (ADG and PDG), respectively. Active, passive and semi-active implant robots were divided into active robot group, passive robot group and semi-active robot group (ARG, PRG and SRG), respectively. Each group placed two implants (FDI tooth positions 31 and 36) in a model 12 times. The accuracy of 216 implants in 108 models were analysed. The coronal deviations of ADG, PDG, ARG, PRG and SRG were 0.85 ± 0.17 mm, 1.05 ± 0.42 mm, 0.29 ± 0.15 mm, 0.40 ± 0.16 mm and 0.33 ± 0.14 mm, respectively. The apical deviations of the five groups were 1.11 ± 0.23 mm, 1.07 ± 0.38 mm, 0.29 ± 0.15 mm, 0.50 ± 0.19 mm and 0.36 ± 0.16 mm, respectively. The axial deviations of the five groups were 1.78 ± 0.73°, 1.99 ± 1.20°, 0.61 ± 0.25°, 1.04 ± 0.37° and 0.42 ± 0.18°, respectively. Abstract Background Supracrestal gingival tissue dimensions (SGTDs) has been considered to be an essential element of periodontal phenotype (PP) components. This study aimed to explore the relationship between SGTDs and other PP components by digital superposition method that integrated cone beam computed tomography (CBCT) with intraoral scanning. Methods This cross-sectional study was conducted at the Stomatology Hospital of Fujian Medical University. Participants were recruited based on the inclusion and exclusion criteria. The data obtained from the digital scanner (TRIOS 3, 3Shape, Denmark) and CBCT images were imported into the TRIOS software (Implant Studio, 3Shape, Denmark) for computing relevant parameters. The significant level was set at 0.05. Results A total of 83 participants with 498 maxillary anterior teeth were finally included. The mean values of supracrestal gingival height (SGH) and the distance from the cementoenamel junction (CEJ) to the crest of the alveolar ridge (CEJ-ABC) on the buccal site were significantly higher than palatal SGH (SGH-p) and palatal CEJ-ABC (CEJ-ABC-p). Men exhibited taller CEJ-ABC and SGH-p than women. Additionally, tooth type was significantly associated with the SGH, SGH-p and CEJ-ABC-p. Taller SGH was associated with wider crown, smaller papilla height (PH), flatter gingival margin, thicker bone thickness (BT) and gingival thickness (GT) at CEJ, the alveolar bone crest (ABC), and 2 mm apical to the ABC. Smaller SGH-p displayed thicker BT and GT at CEJ, the ABC, and 2 and 4 mm apical to the ABC. Higher CEJ-ABC showed lower interproximal bone height, smaller PH, flatter gingival margin, thinner GT and BT at CEJ, and 2 mm apical to the ABC. Smaller CEJ-ABC-p displayed thicker BT at CEJ and 2 and 4 mm apical to the ABC. On the buccal, thicker GT was correlated with thicker BT at 2 and 4 mm below the ABC. Conclusion SGTDs exhibited a correlation with other PP components, especially crown shape, gingival margin and interdental PH. The relationship between SGTDs and gingival and bone phenotypes depended on the apico-coronal level evaluated. Abstract Microglia/macrophages are major contributors to neuroinflammation in the central nervous system (CNS) injury and exhibit either pro- or anti-inflammatory phenotypes in response to specific microenvironmental signals. Our latest in vivo and in vitro studies demonstrated that curcumin-treated olfactory ensheathing cells (aOECs) can effectively enhance neural survival and axonal outgrowth, and transplantation of aOECs improves the neurological outcome after spinal cord injury (SCI). The therapeutic effect is largely attributed to aOEC anti-inflammatory activity through the modulation of microglial polarization from the M1 to M2 phenotype. However, very little is known about what viable molecules from aOECs are actively responsible for the switch of M1 to M2 microglial phenotypes and the underlying mechanisms of microglial polarization. Herein, we show that Interleukin-4 (IL-4) plays a leading role in triggering the M1 to M2 microglial phenotype, appreciably decreasing the levels of M1 markers IL‑1β, IL‑6, tumour necrosis factor-alpha (TNF-α) and inducible nitric oxide synthase (iNOS) and elevating the levels of M2 markers Arg-1, TGF-β, IL-10, and CD206. Strikingly, blockade of IL-4 signaling by siRNA and a neutralizing antibody in aOEC medium reverses the transition of M1 to M2, and the activated microglia stimulated with the aOEC medium lacking IL-4 significantly decreases neuronal survival and neurite outgrowth. In addition, transplantation of aOECs improved the neurological function deficits after SCI in rats. More importantly, the crosstalk between JAK1/STAT1/3/6-targeted downstream signals and NF-κB/SOCS1/3 signaling predominantly orchestrates IL-4-modulated microglial polarization event. These results provide new insights into the molecular mechanisms of aOECs driving the M1-to-M2 shift of microglia and shed light on new therapies for SCI through the modulation of microglial polarization.

Abstract Currently, the classification of bone mineral density (BMD) in many research studies remains rather broad, often neglecting localized changes in BMD. This study aims to explore the correlation between peri-implant BMD and primary implant stability using a new artificial intelligence (AI)-based BMD grading system. 49 patients who received dental implant treatment at the Affiliated Hospital of Stomatology of Fujian Medical University were included. Recorded the implant stability quotient (ISQ) after implantation and the insertion torque value (ITV). A new AI-based BMD grading system was used to obtain the distribution of BMD in implant site, and the bone mineral density coefficients (BMDC) of the coronal, middle, apical, and total of the 1 mm site outside the implant were calculated by model overlap and image overlap technology. Our objective was to investigate the relationship between primary implant stability and BMDC values obtained from the new AI-based BMD grading system. There was a significant positive correlation between BMDC and ISQ value in the coronal, middle, and total of the implant (P 0.05). Background: Colpocleisis is one of traditional surgical procedures for elderly and frail women with advanced pelvic organ prolapse. The occurrence of de novo urinary incontinence following colpocleisis was considered to impair the postoperative quality of life. The incidence of de novo urinary incontinence after colpocleisis has been reported to be ranging from 6.6 % to 27 %. There was an absence of prospective large-sample study to investigate the accurate incidence of de novo urinary incontinence following colpocleisis and the impact on the quality of life till now. Purpose: s The primary objective was to report the incidence of de novo urinary incontinence after colpocleisis. The second objectives were to evaluate the long-term quality of life in patients with de novo urinary incontinence, and to conduct detailed pre- and post-operative evaluations of lower urinary tract symptoms. Methods: This prospective study included 253 patients with symptomatic pelvic organ prolapse who underwent colpocleisis between 2009 and 2021. De novo urinary incontinence was defined as the occurrence of urinary incontinence 3 months postoperatively. All patients were required to complete the Urinary Distress Inventory questionnaire and the Urinary Impact Questionnaire for the evaluation of patients' quality of life, and the Patient Global Impression of Improvement questionnaire for the evaluation of patients’ satisfaction. Results: 245 patients (245/253, 96·8 %) completed the 3-month follow-up, and were included in the final analysis. The incidence of de novo urinary incontinence was 5.4 % (10/185). There was no significant difference in the Urinary Distress Inventory −6 scores (22.50 vs. 10.30, P = 0.276) or the subjective satisfaction rate (100 % vs. 98.9 %, P = 0.250) between the patients with or without de novo urinary incontinence at the long-term follow-up. Abstract Background The outer membrane vesicles (OMVs) derived from Porphyromonas gingivalis (P. gingivalis) have long been acknowledged for their crucial role in the initiation of periodontitis. However, the implications of P. gingivalis OMVs in the context of cardiovascular disease (CVD) remain incompletely understood. This study aimed to clarify both the impact and the underlying mechanisms through which P. gingivalis OMVs contribute to the propagation of distal cardiovascular inflammation and trauma. Methods In this study, various concentrations (0, 1.25, 2.5, and 4.5 µg/µL) of P. gingivalis OMVs were microinjected into the common cardinal vein of zebrafish larvae at 48 h post-fertilization (hpf) to assess changes in cardiovascular injury and inflammatory response. Zebrafish larvae from both the PBS and the 2.5 µg/µL injection cohorts were harvested at 30 h post-injection (hpi) for transcriptional analysis. Real-time quantitative PCR (RT-qPCR) was employed to evaluate relative gene expression. Results These findings demonstrated that P. gingivalis OMVs induced pericardial enlargement in zebrafish larvae, caused vascular damage, increased neutrophil counts, and activated inflammatory pathways. Transcriptomic analysis further revealed the involvement of the immune response and the extracellular matrix (ECM)-receptor interaction signaling pathway in this process. Conclusion This study illuminated potential mechanisms through which P. gingivalis OMVs contribute to CVD. It accentuated their involvement in distal cardiovascular inflammation and emphasizes the need for further research to comprehensively grasp the connection between periodontitis and CVD.

Agroforestry systems can fully exploit the ecological benefits of an ecosystem’s component plants, and improve the ecological environment, soil quality, and land use efficiency, all of which have recently attracted the attention of many scholars. Southwest China’s karst graben basins have barren soil that needs immediate improvement. The karst graben basin in southwest China was used for this study to examine the impacts of several forest–grass composite systems of Bingtang orange–alfalfa–ryegrass on soil improvement, which is located in Jianshui County, Honghe Prefecture, Yunnan Province. The experiment had four treatments, Bingtang orange–alfalfa sowing (B2), Bingtang orange–ryegrass × alfalfa mixed sowing (A), Bingtang orange–ryegrass × alfalfa intercropping (R), and Bingtang orange monocropping (CK). The results showed that different forest–grass composite patterns had noticeable effects on improving the soil’s organic carbon (SOC), total nutrients, and available nutrients, especially in the rainy season when plants grew vigorously. Forage grass intercropping under forest exerted the best effect on soil improvement in the surface layer, and the effect decreased with the increase in soil depth. Alfalfa intercropping under Bingtang orange forest had the strongest effect on improving SOC, total N, ammonia nitrogen, and nitrate nitrogen, whose content increased by 30.7%, 27.3%, 35%, and 36.3%, respectively, in the dry season and 38%, 46.7%, 48.7%, and 55.3%, in the rainy season. However, the effect of alfalfa–ryegrass intercropping under the Bingtang orange forest on soil total P, total K, and available P was better than that of the Bingtang orange–alfalfa intercropping system. The C:N ratio is more suitable in the Bingtang orange–ryegrass-alfalfa composite system. Forage grass intercropping under the Bingtang orange forest effectively improved the soil NPK and C status, and the results of this study provided a basis for the selection of forest–grass composite patterns for soil improvement.

Background. Diabetic retinopathy (DR) is a diabetic microvascular complication and a leading cause of vision loss. However, there is a lack of effective strategies to reduce the risk of DR currently. The present study is aimed at assessing the causal effect of lipid-regulating targets on DR risk using a two-sample Mendelian randomization (MR) study. Method. Genetic variants within or near drug target genes, including eight lipid-regulating targets for LDL-C (HMGCR, PCSK9, and NPC1L1), HDL-C (CETP, SCARB1, and PPARG), and TG (PPARA and LPL), were selected as exposures. The exposure data were obtained from the IEU OpenGWAS project. The outcome dataset related to DR was obtained from the FinnGen research project. Inverse-variance-weighted MR (IVW-MR) was used to calculate the effect estimates by each target. Sensitivity analyses were performed to verify the robustness of the results. Results. There was suggestive evidence that PCSK9-mediated LDL-C levels were positively associated with DR, with OR (95% CI) of 1.34 (1.02-1.77). No significant association was found between the expression of HMGCR- and NPC1L1-mediated LDL-C levels; CETP-, SCARB1-, and PPARG-mediated HDL-C levels; PPARA- and LPL-mediated TG levels; and DR risk. Conclusions. This is the first study to reveal a genetically causal relationship between lipid-regulating drug targets and DR risk. PCSK9-mediated LDL-C levels maybe positively associated with DR risk at the genetic level. This study provides suggestive evidence that PCSK9 inhibition may reduce the risk of DR.

As bone and joint injuries from various causes become increasingly prominent, how to effectively reconstruct and repair bone defects presents a difficult problem for clinicians and researchers. In recent years, graphene and its derivatives have been the subject of growing body of research and have been found to promote the proliferation and osteogenic differentiation of stem cells. This provides a new idea for solving the clinical problem of bone defects. However, as as numerous articles address various aspects and have not been fully systematized, there is an urgent need to classify and summarize them. In this paper, for the first time, the effects of graphene and its derivatives on stem cells in solution, in 2D and 3D structures and in vivo and their possible mechanisms are reviewed, and the cytotoxic effects of graphene and its derivatives were summarized and analyzed. The toxicity of graphene and its derivatives is further reviewed. In addition, we suggest possible future development directions of graphene and its derivatives in bone tissue engineering applications to provide a reference for further clinical application.

ObjectiveThe immune response assumes a pivotal role in the underlying mechanisms of urticaria pathogenesis. The present study delves into an investigation of the genetic causal connections between urticaria and prevalent autoimmune afflictions, notably rheumatoid arthritis (RA), systemic lupus erythematosus (SLE), ulcerative colitis (UC), and Crohn’s disease (CD).MethodsA bidirectional two-sample Mendelian randomization (MR) analysis was conducted to investigate the causal relationships involving four autoimmune diseases and urticaria. The genome-wide association study (GWAS) summary data of four autoimmune disease were sourced from the IEU OpenGWAS database. The GWAS summary data for urticaria were derived from the Finnish consortium dataset. The principal analytical approach employed in this study was the random-effects inverse variance weighted (IVW) method. Subsequently, a series of sensitivity analyses were performed, encompassing assessments of heterogeneity, horizontal pleiotropy, outliers, “Leave-one-out” analyses, and tests for adherence to the assumption of normal distribution.ResultsThe random-effects IVW analysis indicate a positive genetic causal association between RA and urticaria (P < 0.001, OR 95% CI = 1.091 [1.051-1.133]). Conversely, SLE, UC, and CD do not exhibit a significant genetic causal relationship with urticaria. The reverse MR analysis reveals a positive genetic causal linkage between urticaria and SLE (P = 0.026, OR 95% CI = 1.289 [1.031-1.612]). However, the analysis demonstrates no substantial genetic causal relationship between urticaria and RA, UC, or CD. Importantly, the genetic causal assessment absence of heterogeneity, horizontal pleiotropy, and outliers. Furthermore, it remains unaffected by any individual single nucleotide polymorphism (SNP), demonstrating adherence to a normal distribution.ConclusionThis investigation establishing RA as a predisposing factor for urticaria. Moreover, urticaria as a plausible risk determinant for SLE. Heightened vigilance is recommended among RA patients to monitor the manifestation of urticaria within clinical settings. Similarly, individuals afflicted by urticaria should duly acknowledge the prospective susceptibility to SLE.

In the context of global warming, terrestrial ecosystems have undergone significant variations. China has implemented a variety of ecological engineering methods to enhance carbon stocks. However, understanding the spatial and temporal dynamics of carbon and water in drylands under climate change remains limited. Here, our research elucidates carbon and water dynamics in China’s drylands over the last two decades, with a focus on understanding spatial–temporal changes and the effects of ecological engineering on the carbon–water cycle. Furthermore, this study investigates the relationships among climate change, water use efficiency (WUE), and its components—Gross Primary Productivity (GPP) and Evapotranspiration (ET)—identifying key climatic drivers and assessing possible directions for enhancing WUE under changing climate conditions. Our research indicates that both GPP and ET have significantly increased over the past 20 years, with growth rates of 4.96 gC·m−2·yr−1 and 4.26 mm·yr−1, respectively. Meanwhile, WUE exhibited a slight declining trend, at a rate of −0.004 gC·mmH2O·yr−1. This confirms the positive impact of vegetation restoration efforts. We found that fluctuations in interannual WUE were influenced by human activities and climate change. Precipitation (Prec) was the key climatic factor driving the GPP increase. Both solar radiation (Solra) and Prec were crucial for the interannual variation of WUE. Interestingly, WUE was the main factor affecting GPP development. The decline in WUE in drylands is linked to interannual variability in WUE and increased Vapor Pressure Deficit (VPD) due to warming. Seasonal variations in how WUE responds to climatic factors were also observed. For instance, fall rainfall increased WUE, while spring rainfall decreased it. Fall WUE was highly sensitive to VPD. Spatially, we found higher WUE in China’s eastern and Xinjiang regions and lower in inland areas and the Tibetan Plateau. Geomorphologic factors and soil conditions were the main drivers of this spatial variability in WUE. Temperature (Tem), Solra, VPD, and relative humidity (Relah) also played significant roles. Our results show a generalized inverse persistence in WUE variability. This suggests a potential for increased WUE in the eastern regions and a risk of decreased WUE on the Tibetan Plateau. Addressing the threat of vegetation decline in arid regions, particularly within the Tibetan Plateau, is crucial. It is essential to adapt forestry practices to complement the carbon and water cycles in these landscapes.

ObjectiveThe purpose of this study was to prospectively evaluate the efficacy of a demineralized dentin matrix (DDM) in decreasing the initial inflammatory response of the gingiva and facilitating the repair and regeneration of soft tissue in alveolar ridge preservation.MethodsThis clinical study employed a split-mouth design. Fourteen patients with a total of forty-four sites underwent extraction and alveolar ridge preservation (ARP) procedures. A Bilaterally symmetrical extraction operation were conducted on the premolars of each patient. The experimental group received DDM as a graft material for ARP, while the control group underwent natural healing. Within the first month postoperatively, the pain condition, color, and swelling status of the extraction sites were initially assessed at different time points Subsequently, measurements were taken for buccal gingival margin height, buccal-lingual width, extraction socket contour, and the extraction socket area and healing rate were digitally measured. Additionally, Alcian Blue staining was used for histological evaluation of the content during alveolar socket healing.ResultsBoth groups experienced uneventful healing, with no adverse reactions observed at any of the extraction sites. The differences in VAS pain scores between the two groups postoperatively were not statistically significant. In the early stage of gingival tissue healing (3 days postoperatively), there were statistically significant differences in gingival condition and buccal gingival margin height between the two groups. In the later stage of gingival tissue healing (7, 14, and 30 days postoperatively), there were statistically significant differences in buccal-lingual width, extraction socket healing area, and healing rate between the two groups. Furthermore, the histological results from Alcian Blue staining suggested that the experimental group may play a significant role in promoting gingival tissue healing, possibly by regulating inflammatory responses when compared to the control group.ConclusionThe application of DDM in alveolar ridge preservation has been found to diminish initial gingival inflammation after tooth extraction. Additionally, it has shown the ability to accelerate early gingival soft tissue healing and preserve its anatomical contour.Clinical trial registrationchictr.org.cn, identifier ChiCTR2100050650.

After extensive ecological restoration in the karst region of Southwestern China, a potential zone for achieving “carbon neutrality” has emerged, despite facing water scarcity. We analyzed the dynamics of water use efficiency (WUE) and its correlations with soil moisture (SM) and leaf area index (LAI) from 2003 to 2017 using PML-V2 data and multiple datasets for SM and LAI. The effects of different enclosure years on soil physicochemical properties provide scientific basis for soil fertility evaluation and ecological restoration. However, the details on the effects of long-term fencing measures on soil physicochemical properties in alpine regions are still lacking. The soil physicochemical properties, soil nutrient content, and stoichiometric characteristics of desert grassland were examined in the desert steppe of northern Tibetan Plateau, where grazing exclusion using fences has been implemented. This study included conducting experiments on different soil layers during different duration (10, 11, and 12 years) of grazing exclusion using fences; natural plots with no grazing exclusion were used as a control variable. Results showed that several soil physicochemical indexes, such as soil organic matter, soil water content, soil capillary porosity, soil non-capillary porosity, and soil capillary water capacity, in plots under the fencing measures were higher than those in the natural plots. The soil bulk density of the 12- and 11-year grazing exclusion plots was lower than that of the natural plots. The clay content of the 10- and 12-year grazing exclusion plots was higher than that of the natural plots, and the sand content was lower than that of the natural plots. The soil nutrient content was especially low with less than the average level of soil nutrient content in China. The stoichiometric C/N, C/P, N/P, and C/K ratios were 10.28–16.39, 2.74–16.69, 0.24–1.53, and 0.27–0.82, respectively. This result indicates that the degree of organic carbon mineralization was relatively high, and there existed a certain N limitation in the study area. The implementation of enclosure is conducive to the accumulation of C and N. The results provide support for elucidating the physicochemical characteristics, soil nutrients, and nutrient cycling of desert steppe in the northern Tibetan Plateau and for the study of the allotted time of fencing project in the alpine region.

The assessment of mental overload is essential as mental overload has been proved to be a critical cause of many accidents. Mental overload is affected by various performance shaping factors (PSFs), and harsh PSFs will either increase the task demand or decrease the overload threshold, which is rarely considered in the current mental overload assessment method. This research proposes a VACP-based mental overload assessment model which considers the effects of PSFs. In the VACP model, mental overload can be identified when the sum of the task demand values of every unit task is greater than the given threshold. In human reliability analysis, PSFs are mainly used as weighting factors to modify basic human error probability. In our research, by virtue of the quantitative relationship between human error and mental workload, the weighting factors of PSFs are converted to modify the task demand values and threshold for VACP activities. Furthermore, Bayesian Network (BN) is used to model the influence of PSFs and to calculate the probability of mental overload. The proposed method is applied to an accident involving a helicopter crash that occurred in Maryland, and the results show that, in comparison with the VACP method, the proposed method can more effectively identify the state of mental overload and provide a more rational explanation of the process of the accident.

Abstract For complex dynamic interactive tasks (such as aviating), operators need to continuously extract information from areas of interest (AOIs) through eye movement to maintain high level of situation awareness (SA), as failures of SA may cause task performance degradation, even system accident. Most of the current eye movement models focus on either static tasks (such as image viewing) or simple dynamic tasks (such as video watching), without considering SA. In this study, an eye movement model with the goal of maximizing SA is proposed based on Markov decision process (MDP), which is designed to describe the dynamic eye movement of experienced operators in dynamic interactive tasks. Two top-down factors, expectancy and value, are introduced into this model to represent the update probability and the importance of information in AOIs, respectively. In particular, the model regards sequence of eye fixations to different AOIs as sequential decisions to maximize the SA-related reward (value) in the context of uncertain information update (expectancy). Further, this model was validated with a flight simulation experiment. Results show that the predicted probabilities of fixation on and shift between AOIs are highly correlated ( $$R = 0.928$$ R = 0.928 and $$R = 0.951$$ R = 0.951 , respectively) with those of the experiment data.

Cadmium (Cd) contamination in the soil potentially hampers microbial biomass and adversely affects their services such as decomposition and mineralization of organic matter. It can reduce nitrogen (N) metabolism and consequently affect plant growth and physiology. Further, Cd accumulation in plants can pose health risks through vegetable consumption. Here, we investigated consequences of Cd contamination on fertilizer value and associated health risks following the application of biogas residues (BGR) to various soil types. Our results indicate that the application of BGR to all soil types significantly increased dry matter (DM) yield and N uptake. However, the Cd contamination negatively affected DM yield and N recovery from BGR in a dose-dependent manner. Organic N mineralization from BGR also decreased in Cd-contaminated soils. The highest DM yield and N recovery were recorded in sandy soil, whereas the lowest values were observed in clay soil. Cadmium was accumulated in spinach, and health risk index (HRI) associated with its dietary intake revealed that consuming spinach grown in Cd-contaminated soil, with or without BGR, is unsafe. Among the soil types, values of daily intake of metals (DIM) and HRI were lowest in clay soil and highest in sandy soil. However, the application of BGR curtailed HRI across all soil types. Notably, the application of BGR alone resulted in HRI values < 1, which are under the safe limit. We conclude that soil contamination with Cd reduces fertilizer value and entails implications for human health. However, the application of BGR to the soil can decrease Cd effects.

Peanut is a major oilseed and food legume. Shelling percentage (SP), closely associated with seed yield, is a trait whose improvement is a major goal of peanut breeding. In this study, a mapping population (Xuhua 13 × Zhonghua 6) was used to map quantitative trait loci (QTL) controlling SP in four environments. Two stable major QTL for SP were mapped on both SSR- and SNP-based genetic maps. qSPA07.1 on chromosome A07 explained up to 31.7% of phenotypic variation, and qSPA08.2 on chromosome A08 explained up to 10.8%. Favorable alleles of qSPA07.1 and qSPA08.2 were derived from the female and male parents, respectively. Eight recombinant inbred lines (RILs) carrying both favorable alleles showed superiority in SP over the two parents in all environmental trials. A combination of the two favorable alleles using the linked markers was verified to increase SP by ∼5% in the RIL population and by ∼3% SP in diverse peanut cultivars. qSPA07.1 and qSPA08.2 were delimited to respectively a 0.73-Mb interval harboring 96 genes and a 3.93-Mb interval harboring 238 genes. Respectively five and eight genes with high expression in pods, including enzymes and transcription factors, were assigned as candidate genes for qSPA07.1 and qSPA08.2. These consistent major QTL provide an opportunity for fine mapping of genes controlling SP, and the linked markers may be useful for genetic improvement of SP in peanut.

Abstract Background Aflatoxin contamination caused by Aspergillus fungi has been a serious factor affecting food safety of peanut (Arachis hypogaea L.) because aflatoxins are highly harmful for human and animal health. As three mechanisms of resistance to aflatoxin in peanut including shell infection resistance, seed infection resistance and aflatoxin production resistance exist among naturally evolved germplasm stocks, it is highly crucial to pyramid these three resistances for promoting peanut industry development and protecting consumers’ health. However, less research effort has been made yet to investigate the differentiation and genetic relationship among the three resistances in diversified peanut germplasm collections. Results In this study, the Chinese peanut mini-mini core collection selected from a large basic collection was systematically evaluated for the three resistances against A. flavus for the first time. The research revealed a wide variation among the diversified peanut accessions for all the three resistances. Totally, 14 resistant accessions were identified, including three with shell infection resistance, seven with seed infection resistance and five with aflatoxin production resistance. A special accession, Zh.h1312, was identified with both seed infection and aflatoxin production resistance. Among the five botanic types of A. hypogaea, the var. vulgaris (Spanish type) belonging to subspecies fastigiata is the only one which possessed all the three resistances. There was no close correlation between shell infection resistance and other two resistances, while there was a significant positive correlation between seed infection and toxin production resistance. All the three resistances had a significant negative correlation with pod or seed size. A total of 16 SNPs/InDels associated with the three resistances were identified through genome-wide association study (GWAS). Through comparative analysis, Zh.h1312 with seed infection resistance and aflatoxin production resistance was also revealed to possess all the resistance alleles of associated loci for seed infection index and aflatoxin content. Conclusions This study provided the first comprehensive understanding of differentiation of aflatoxin resistance in diversified peanut germplasm collection, and would further contribute to the genetic enhancement for resistance to aflatoxin contamination.

IntroductionThe wild allotetraploid peanut Arachis monticola contains a higher oil content than the cultivated allotetraploid Arachis hypogaea. Besides the fact that increasing oil content is the most important peanut breeding objective, a proper understanding of its molecular mechanism controlling oil accumulation is still lacking.MethodsWe investigated this aspect by performing comparative transcriptomics from developing seeds between three wild and five cultivated peanut varieties.ResultsThe analyses not only showed species-specific grouping transcriptional profiles but also detected two gene clusters with divergent expression patterns between two species enriched in lipid metabolism. Further analysis revealed that expression alteration of lipid metabolic genes with co-expressed transcription factors in wild peanut led to enhanced activity of oil biogenesis and retarded the rate of lipid degradation. In addition, bisulfite sequencing was conducted to characterize the variation of DNA methylation between wild allotetraploid (245, WH 10025) and cultivated allotetraploid (Z16, Zhh 7720) genotypes. CG and CHG context methylation was found to antagonistically correlate with gene expression during seed development. Differentially methylated region analysis and transgenic assay further illustrated that variations of DNA methylation between wild and cultivated peanuts could affect the oil content via altering the expression of peroxisomal acyl transporter protein (Araip.H6S1B).DiscussionFrom the results, we deduced that DNA methylation may negatively regulate lipid metabolic genes and transcription factors to subtly affect oil accumulation divergence between wild and cultivated peanuts. Our work provided the first glimpse on the regulatory mechanism of gene expression altering for oil accumulation in wild peanut and gene resources for future breeding applications.

BackgroundRheumatoid arthritis (RA) is a chronic systemic autoimmune disease with symptoms characterized by typical circadian rhythmic changes. This study aimed to identify the hub circadian rhythm genes (CRGs) in RA and explore their association with immune cell infiltration and pathogenesis of RA.MethodsThe differentially expressed CRGs (DECRGs) between RA and normal control samples were screened from Datasets GSE12021 and GSE55235. Gene Ontology, Kyoto Encyclopedia of Genes and Genomes, and Gene Set Enrichment Analysis were used to explore the potential functional mechanisms of DECRGs in RA. Weighted Gene Co-expression Network Analysis and Least Absolute Shrinkage and Selection Operator regression analysis were performed to identify hub CRGs of RA. CIBERSORT was conducted to compare the infiltration level of immune cells in RA and control synovial tissue and their relationship with hub genes. In addition, the diagnostic value of hub biomarkers was evaluated by the area under the receiver operator characteristic curve. Further, a nomogram prediction model was constructed and its significance for clinical decision-making was evaluated.ResultsThe green module was identified as the hub module associated with RA. Four hub CRGs (EGR1, FOSL2, GADD45B, and NFIL3) were identified and showed that they had the highest specificity and sensitivity for RA diagnosis, respectively. The expression levels and diagnostic values of these genes were externally validated in the dataset GSE55457. A nomogram prediction model based on the four hub CRGs was constructed and proved to have a certain clinical decision value. Additionally, the correlation analysis of immune cells with hub genes showed that all hub genes were significantly positively correlated with activated mast cells, resting memory CD4+ T cells, and monocytes. Whereas, all hub genes were negatively correlated with plasma cells, CD8+ T cells, and activated memory CD4+ T cells. Meanwhile, FOSL2 and GADD45B were negatively correlated with Tfh cells.ConclusionFour hub CRGs were identified and showed excellent diagnostic value for RA. These genes may be involved in the pathological process of RA by disrupting the rhythmic oscillations of cytokines through immune-related pathways and could be considered molecular targets for future chronotherapy against RA.

Identifying the right accessories for installing the dental implant is a vital element that impacts the sustainability and the reliability of the dental prosthesis when the medical case of a patient is not comprehensive. Dentists need to identify the implant manufacturer from the x-ray image to determine further treatment procedures. Identifying the manufacturer is a high-pressure task under the scaling volume of patients pending in the queue for treatment. To reduce the burden on the doctors, a dental implant identification system is built based on a new proposed thinner VGG model with an on-demand client-server structure. We propose a thinner version of VGG16 called TVGG by reducing the number of neurons in the dense layers to improve the system’s performance and gain advantages from the limited texture and patterns in the dental radiography images. The outcome of the proposed system is compared with the original pre-trained VGG16 to verify the usability of the proposed system.

The annual phosphate (PO43−) utilization has increased, leading to a depletion of existing sources of phosphorus (P). To overcome this, digestate as a source to recover P is being investigated. Due to the abundance of nutrients, the digestate from an agricultural biogas plant is used as fertilizer for crops. The separation of solids and liquids from the digestate by a screw press is the simplest form of concentrating, therefore, recovering PO43−. This is the most commonly employed method in existing biogas plants. However, the separation is not very efficient as only 20–30% of P is recovered in the solid phase. The goal of this study is to increase the separation efficiency and recover more P into the solid phase, in order to improve the transportability. For this, separation trials at a laboratory scale were performed for five experimental groups, with biochar and straw flour as non-reactive additives and kieserite as a reactive additive. In addition, untreated digestate was studied as a control. The control and the treatment with biochar and straw flour were carried out at 25 °C, while the treatment with kieserite was performed at 25 °C and 50 °C. The separation trails were performed at treatment times of 0 h, 1 h, 2 h, 8 h, and 20 h. The results showed that the treatment with additives had a beneficial effect on the recovery of P. It was noted that kieserite treatment at 25 °C and 50 °C bound about 61% of the total P present in the digestate to the solid phase. A sequential extraction was performed to study the effect of additives on the recovery of different P species. The results concluded that, compared to biochar and straw flour, kieserite was efficient in recovering the non-labile fractions (NaOH-P and HCl-P) of P, which act as slow-release fertilizers. This study shows that the use of additives, especially kieserite, has a positive influence on recovering P from digestate, and further research to optimize the recovery process would be beneficial.

BackgroundThe morbidity and mortality of cardiovascular diseases (CVD) in patients with rheumatoid arthritis (RA) is significantly higher than those in the general population, leading to RA-related CVD has attracted broad attention and numerous articles have been published. However, no study has systematically examined this area from a scientometric perspective. This study aimed to visualize the knowledge structure and identify emerging research trends and potential hotspots in this field.Materials and methodsArticles and reviews on RA-CVD published from 2001 to 2021 were extracted from the Web of Science Core Collection database. CiteSpace and VOSviewer software were used to visualize the knowledge network of countries, institutions, authors, references and keywords in this field. SPSS and Microsoft Excel software were used for curve fitting and correlation analysis.ResultsA total of 2,618 articles and reviews were included. The number of publications about RA-related CVD significantly increased yearly. Publications were mainly concentrated in North America, Europe and East Asia. The United States contributed most with 699 publications, followed by the United Kingdom and Italy. Gross Domestic Product was an important factor affecting scientific output. University of Manchester and Professor Kitas George D. were the most prolific institutions and influential authors, respectively. Journal of Rheumatology was the most productive journal for RA-related CVD research. The research hotspots switched in the order of clinical features (cardiovascular events), mechanism exploration, anti-tumor necrosis factor therapy, risk factors, and antirheumatic drug safety, which can be observed from the keyword analysis and co-cited reference cluster analysis.ConclusionsThis study found that research on RA-related CVD is flourishing. The safety and cardiovascular pharmacological mechanisms of anti-rheumatoid drugs, especially targeted synthetic DMARDs, would be the focus of current research and developmental trends in future research.

Spinal cord injury (SCI) is a devastating type of neurological disorder of the central nervous system (CNS) with high mortality and disability. The pathological processes of SCI can usually be described as two stages, namely, primary and acute secondary injuries. Secondary injury produces more significant exacerbations of the initial injury. Among all the mechanisms of secondary damage, infection and inflammatory responses, as the principle culprits in initiating the second phase of SCI, can greatly contribute to the severity of SCI and numerous sequelae after SCI. Therefore, effectively antagonizing pro-inflammatory responses may be a promising treatment strategy to facilitate functional recovery after SCI. Olfactory ensheathing cells (OECs), a unique type of glial cells, have increasingly become potential candidates for cell-based therapy in the injured CNS. Strikingly, there is growing evidence that the mechanisms underlying the anti-inflammatory role of OECs are associated with the immune properties and secretory functions of these cells responsible for anti-neuroinflammation and immunoregulatory effects, leading to maintenance of the internal microenvironment. Accordingly, a more profound understanding of the mechanism of OEC immunological functions in the treatment of SCI would be beneficial to improve the therapeutic clinical applications of OECs for SCI. In this review, we mainly summarize recent research on the cellular and molecular immune attributes of OECs. The unique biological functions of these cells in promoting neural regeneration are discussed in relation of the development of novel therapies for CNS injury.

Abstract Owing to the limited range, battery electric vehicle drivers are likely to worry about being stranded in range‐critical situations, known as range anxiety. Research has revealed that the provision of accurate information through range‐related in‐vehicle information systems establishes a solution to overcome range anxiety. However, an important range‐related information i.e. the battery state of health, is seldom being considered in the research of range anxiety due to the difficulty in daily‐use acquisition. Therefore, this paper aims to explore the influence of state of health on the range anxiety of battery electric vehicle drivers by performing a driving experiment. Under the condition of state of health degradation, participants used simplified battery electric vehicle simulators equipped with different types of in‐vehicle information systems to finish a designated trip in a range‐critical situation. During and after the trip, they measured their experienced range anxiety level. Results show that participants perceive less range anxiety when the in‐vehicle information system provides the remaining range corrected by the state of health, and range anxiety can be further reduced when the extra correlated information (the state of health) is displayed on the in‐vehicle information systems. The findings provide useful inspirations that range anxiety can be reduced if the range‐related in‐vehicle information system is well designed in information content and presentation considering the state of health.

Abstract Background Peanut is one of the primary sources for vegetable oil worldwide, and enhancing oil content is the main objective in several peanut breeding programs of the world. Tightly linked markers are required for faster development of high oil content peanut varieties through genomics-assisted breeding (GAB), and association mapping is one of the promising approaches for discovery of such associated markers. Results An association mapping panel consisting of 292 peanut varieties extensively distributed in China was phenotyped for oil content and genotyped with 583 polymorphic SSR markers. These markers amplified 3663 alleles with an average of 6.28 alleles per locus. The structure, phylogenetic relationship, and principal component analysis (PCA) indicated two subgroups majorly differentiating based on geographic regions. Genome-wide association analysis identified 12 associated markers including one (AGGS1014_2) highly stable association controlling up to 9.94% phenotypic variance explained (PVE) across multiple environments. Interestingly, the frequency of the favorable alleles for 12 associated markers showed a geographic difference. Two associated markers (AGGS1014_2 and AHGS0798) with 6.90–9.94% PVE were verified to enhance oil content in an independent RIL population and also indicated selection during the breeding program. Conclusion This study provided insights into the genetic basis of oil content in peanut and verified highly associated two SSR markers to facilitate marker-assisted selection for developing high-oil content breeding peanut varieties.

The cultivated peanut (Arachis hypogaea L.), which is rich in edible oil and protein, is widely planted around the world as an oil and cash crop. However, aflatoxin contamination seriously affects the quality safety of peanuts, hindering the development of the peanut industry and threatening the health of consumers. Breeding peanut varieties with resistance to Aspergillus flavus infection is important for the control of aflatoxin contamination, and understanding the genetic basis of resistance is vital to its genetic enhancement. In this study, we reported the quantitative trait locus (QTL) mapping of resistance to A. flavus infection of a well-known resistant variety, J11. A mapping population consisting of 200 recombinant inbred lines (RILs) was constructed by crossing a susceptible variety, Zhonghua 16, with J11. Through whole-genome resequencing, a genetic linkage map was constructed with 2,802 recombination bins and an average inter-bin distance of 0.58 cM. Combined with phenotypic data of an infection index in 4 consecutive years, six novel resistant QTLs with 5.03–10.87% phenotypic variances explained (PVE) were identified on chromosomes A05, A08, B01, B03, and B10. The favorable alleles of five QTLs were from J11, while that of one QTL was from Zhonghua 16. The combination of these favorable alleles significantly improved resistance to A. flavus infection. These results could contribute greatly to the understanding of the genetic basis of A. flavus resistance and could be meaningful in the improvement of further resistance in peanuts.

Evapotranspiration (ET) is an essential component of the forest stand water budget, and the main factors controlling ET variations are meteorological conditions and soil moisture. A deep understanding of their impacts on ET is crucial for forest water management and sustainable development in changing environments. However, the independent effects of meteorological conditions and soil moisture on ET are not clear because their impacts occur concurrently under natural environmental conditions. In this study, a relatively simple but mechanism-based forest stand ET model that couples the effects of the reference ET (ETo) and relative extractable water (REW) in the 0–60 cm soil layer was developed to quantify the independent impacts of ETo and REW on ET. The model was established based on observed stand transpiration, forest floor ET, meteorological conditions, and soil water content obtained on non-rainfall days of the 2015, 2016, and 2019 growing seasons from a larch (Larix principis-rupprechtii) plantation in Northwest China. The following key results were obtained. (1) At this sub-humid site, the weather-dependent ET increased linearly with rising ETo; daily ET initially increased rapidly with rising REW, and then gradually reached a threshold; thereafter, it tended to be stable. The response could be described by an exponential saturation function. (2) The daily stand ET model that coupled these response functions to individual factors was determined as ET = 0. 785 × [ETo × (1 – EXP(–13.567REW))]. (3) The contributions of ETo and REW were further determined using the new ET model, and the results show that ETo was the main driving factor affecting the ET variations in three different hydrological years compared with REW and the interaction of ETo and REW. Compared with those in the normal year 2015, REW and ETo limited the ET variation in the dry year 2016 by 13.1% and 7.3%, respectively, and ETo limited the ET variation in the wet year 2019 by 8.8%. Our study provides a simple and feasible method for separating the meteorological conditions (i.e., ETo) and soil moisture controls on ET variations, and can improve the understanding of the impacts of varying environments on ET.

Peanut is an important legume crop worldwide. To uncover the genetic basis of yield features and assist breeding in the future, we conducted genome-wide association studies (GWAS) for six yield-related traits of the Chinese peanut mini-core collection. The seed (pod) size and weight of the population were investigated under four different environments, and these traits showed highly positive correlations in pairwise combinations. We sequenced the Chinese peanut mini-core collection using genotyping-by-sequencing approach and identified 105,814 high-quality single-nucleotide polymorphisms (SNPs). The population structure analysis showed essentially subspecies patterns in groups and obvious geographical distribution patterns in subgroups. A total of 79 significantly associated loci (P < 4.73 × 10–7) were detected for the six yield-related traits through GWAS. Of these, 31 associations were consistently detected in multiple environments, and 15 loci were commonly detected to be associated with multiple traits. Two major loci located on chromosomal pseudomolecules A06 and A02 showed pleiotropic effects on yield-related traits, explaining ∼20% phenotypic variations across environments. The two genomic regions were found 46 putative candidate genes based on gene annotation and expression profile. The diagnostic marker for the yield-related traits from non-synonymous SNP (Aradu-A06-107901527) was successfully validated, achieving a high correlation between nucleotide polymorphism and phenotypic variation. This study provided insights into the genetic basis of yield-related traits in peanut and verified one diagnostic marker to facilitate marker-assisted selection for developing high-yield peanut varieties.

The inheritance of pod- and seed-number traits (PSNT) in peanut (Arachis hypogaea L.) is poorly understood. In the present study, a recombinant inbred line (RIL) population of 188 lines was used to map quantitative trait loci (QTL) for number of seeds per pod (NSP), number of pods per plant (NPP), and numbers of one-, two-, and three-seeded pods per plant (N1PP, N2PP, and N3PP) in four environments. A total of 28 consensus QTL and 14 single QTL were identified, including 11 major and stable QTL. Four major and stable QTL including qN3PPA5.2, qN3PPA5.4, qN3PPA5.5, and qN3PPA5.7 each explained 12.3%–33.0% of phenotype variation. By use of another integrated linkage map for the A5 group (hereafter referred to as INT A5 group), QTL for PSNT were located in seven intervals of 0.73–9.68 Mb in length on chromosome A05, and candidate genes underlying N3PP were suggested. These findings shed light on the genetic basis of PSNT. Major QTL for N3PP could be used as candidates for further positional cloning. Keywords: Peanut, Number, Pod, Seed, QTL

Most Micro-Electro-Mechanical Systems (MEMS) experience natural degradation and random shocks simultaneously, and their failures are mainly the results of competing soft and hard failure processes. For some MEMS devices like micro-engines, considering that they have resistance against small shock loads, shocks can be categorized into three shock zones according to their magnitudes: safety zone, damage zone, and fatal zone. The fatal shocks can cause hard failure immediately, and the damage shocks can (i) increase the degradation level and (ii) reduce the hard failure threshold. In this paper, the effect (ii) is described by a dependence between the classifications of damage shocks and fatal shocks: after surviving a damage shock, the probability of a shock in fatal zone increases and in damage zone decreases. Due to the dependence, the Poisson process widely used in previous studies is unsuitable. A dependent zoned shock model is developed, where a Hindrance model, which is a special type of Markov point process, is first introduced to describe the damage shocks, and a Cox process is used to model the fatal shocks. Then, a general reliability model of micro-engines subject to degradation and dependent zoned shocks is developed. When the micro-engine degrades linearly, an analytical reliability model is derived. Finally, a numerical example is conducted to illustrate the effectiveness of the developed model.

Resveratrol (trans-3,4′,5-trihydroxystilbene) is a natural stilbene phytoalexin which is also found to be good for human health. Cultivated peanut (Arachis hypogaea L.), a worldwide important legume crop, is one of the few sources of human's dietary intake of resveratrol. Although the variations of resveratrol contents among peanut varieties were observed, the variations across environments and its underlying genetic basis were poorly investigated. In this study, the resveratrol content in seeds of a recombination inbred line (RIL) population (Zhonghua 6 × Xuhua 13, 186 progenies) were quantified by high performance liquid chromatography (HPLC) method across four environments. Genotypes, environments and genotype × environment interactions significantly influenced the resveratrol contents in the RIL population. A total of 8,114 high-quality single nucleotide polymorphisms (SNPs) were identified based on double-digest restriction-site-associated DNA sequencing (ddRADseq) reads. These SNPs were clustered into bins using a reference-based method, which facilitated the construction of high-density genetic map (2,183 loci with a total length of 2,063.55 cM) and the discovery of several chromosome translocations. Through composite interval mapping (CIM), nine additive quantitative trait loci (QTL) for resveratrol contents were identified on chromosomes A01, A07, A08, B04, B05, B06, B07, and B10 with 5.07–8.19% phenotypic variations explained (PVE). Putative genes within their confidential intervals might play roles in diverse primary and secondary metabolic processes. These results laid a foundation for the further genetic dissection of resveratrol content as well as the breeding and production of high-resveratrol peanuts.

ObjectivesCoronavirus disease 2019 (COVID-19) remains a global challenge. Corticosteroids constitute a group of anti-inflammatory and immunosuppressive drugs that are widely used in the treatment of COVID-19. Comprehensive reviews investigating the comparative proportion and efficacy of corticosteroid use are scarce. Therefore, we conducted a systematic review and meta-analysis of clinical trials to evaluate the proportion and efficacy of corticosteroid use for the treatment of COVID-19.MethodsWe conducted a comprehensive literature review and meta-analysis of research articles, including observational studies and clinical trials, by searching the PubMed, EMBASE, Cochrane Controlled Trials Registry, and China Academic Journal Network Publishing databases. Patients treated between December 1, 2019, and January 1, 2021, were included. The outcome measures were the proportion of patients treated with corticosteroids, viral clearance and mortality. The effect size with the associated 95% confidence interval is reported as the weighted mean difference for continuous outcomes and the odds ratio for dichotomous outcomes.ResultsFifty-two trials involving 15710 patients were included. The meta-analysis demonstrated that the proportion of COVID-19 patients who received corticosteroids was significantly lower than that of patients who did not receive corticosteroids (35.19% vs. 64.49%). In addition, our meta-analysis demonstrated no significant difference in the proportions of severe and nonsevere cases treated with corticosteroids (27.91% vs. 20.91%). We also performed subgroup analyses stratified by whether patients stayed in the intensive care unit (ICU) and found that the proportion of patients who received corticosteroids was significantly higher among those who stayed in the ICU than among those who did not. The results of our meta-analysis indicate that corticosteroid treatment significantly delayed the viral clearance time. Finally, our meta-analysis demonstrated no significant difference in the use of corticosteroids for COVID-19 between patients who died and those who survived. This result indicates that mortality is not correlated with corticosteroid therapy.ConclusionThe proportion of COVID-19 patients who received corticosteroids was significantly lower than that of patients who did not receive corticosteroids. Corticosteroid use in subjects with severe acute respiratory syndrome coronavirus 2 infections delayed viral clearance and did not convincingly improve survival; therefore, corticosteroids should be used with extreme caution in the treatment of COVID-19.

The aim of present study was to investigate the regulatory effects of an extract of the Grifola frondosa on glycolipid metabolism and the intestinal flora in high-fat diet fed rats. The ethyl acetate extract of G. frondosa (EAE), enriched with pseudobaptigenin and cyanidin 3-O-xylosylrutinoside, exerted a stronger inhibitory effect on lipase than the other polar fractions of G. frondosa in vitro. Supplementation with EAE (50 mg/kg) to rats for 8 weeks clearly ameliorated dyslipidaemia by suppressing the serum levels of glucose (Glu), total triglycerides (TG) and low-density lipoprotein cholesterol (LDL-C), and hepatic fat accumulation. Furthermore, EAE supplementation normalized the intestinal flora structure in high-fat diet fed rats. The relevance analysis suggested strong associations of Anaerotruncus, Barnesiella, Clostridiaceae, Erysipelotrichaceae, Paraprevotella, Clostridium_XVIII, Coprococcus and Turicibacter with the biochemical indexes. Simultaneously, the EAE treatment intervened the expression levels of the genes associated with glycolipid metabolism. Our results indicated that pseudobaptigenin and cyanidin-3-O-xylosylrutinoside can be used as pharmaceutical agents for the prevention of the disorder of glycolipid metabolism.

Objective: To describe the clinical and pathological features of endometrial carcinoma with extraperitoneal metastasis and examine whether surgery could improve the prognosis. Methods: The Surveillance, Epidemiology, and End Results database was used to analyze 730 patients who were diagnosed with extraperitoneal metastasis of endometrial cancer from 2010 to 2015, including metastasis to the lung, bone, or brain. Results: Of the 730 patients, 372 (50.96%) patients had single lung metastases, and 196(26.85%) patients had multiple organ metastases that included pulmonary invasion. Therefore, the lung was the most common target organ for extraperitoneal metastasis of endometrial cancer. In multivariate risk factor analysis, grade 3 tumor (odds ratio = 3.39, P < .001), positive peritoneal cytology (odds ratio = 2.02, P < .001), and cervical stromal invasion (odds ratio = 1.42, P = .030) were independent risk factors for extraperitoneal metastasis. Once metastasis occurred in the brain or multiple organs, the prognosis was often poor. Of the patients, 362 underwent surgery, and surgery was performed only for primary tumors of the reproductive organs in almost all patients (97.23%) with extraperitoneal metastasis. The median cancer-specific survival periods of patients with solitary pulmonary metastasis undergoing surgery and those without surgery were 23 (16.43-29.57) months and 9 (6.21-11.79) months, respectively ( P < .001), and survival superiority also existed in patients with bone metastasis (19 vs 8 months, P = .015) and multiple organs metastases (15 vs 4 months, P < .001). However, patients with brain metastasis had the same median survival period in the 2 groups (6 months, P = .146). Conclusions: The lung was the most common target organ for extraperitoneal metastasis in patients with endometrial cancer. Surgery was associated with improved survival in women with extraperitoneal metastasis, except for patients with brain metastasis.

Background: The aim of this study was to clarify the features and prognostic significance of isolated para-aortic lymphatic metastasis of endometrial cancer. Methods: A retrospective study of patients with stage IIIC endometrial cancer was performed based on the Surveillance, Epidemiology, and End Results (SEER) database. A total of 2767 patients were divided into three groups according to the lymphatic metastasis patterns: isolated pelvic lymphatic metastasis, isolated para-aortic lymphatic metastasis and dual lymphatic metastasis. The clinic-pathological characteristics and prognosis of patients were compared among the three groups. Result: The proportion of patients with isolated para-aortic lymphatic metastasis was 13.70%. Patients with isolated pelvic lymphatic metastasis or isolated para-aortic lymphatic metastasis shared similar histological characteristics, except that patients with isolated para-aortic lymphatic metastasis had a lower proportion of tumors over 5 cm in diameter than patients with isolated pelvic lymphatic metastasis (35.1% versus 45.7%, p = 0.001). Understanding the determinants of tree biomass allocation patterns among organs is crucial for both predicting the rate and potential of forest carbon sinks and guiding future multifunctional forest management. However, it is still not clear how the site conditions (e.g., elevation) and stand structure (e.g., tree dominance, stand density) affect the biomass allocation of single trees in forests. This study was implemented in the Liupan Mountains of the Loess Plateau of Northwest China by collecting the related information of biomass data of 110 sample trees with different dominance and influencing factors within 23 sample plots of larch plantations set up along the elevation gradient. Based on these data, the response tendency and functions of biomass allocation of single trees to individual influencing factors of site conditions and forest structure were analyzed. Moreover, the results illustrated that the ratio between root biomass and aboveground biomass decreased significantly with rising stand age and tree density, but increased significantly with rising elevation, and there was no significant relationship with the dominance of individual trees. The results of this study revealed the importance of considering the influencing factors of site conditions and stand structure when developing dynamic models of tree biomass allocation. The results and research methods used in this study provide useful tools for quantifying the biomass allocation and carbon storage partitioning in the study area and other similar regions.

Most previous reliability models of consecutive k-out-of-n system assume that the components are independent or their reliabilities are equal. The system reliability can be obtained according to the configuration of k consecutive failed components. However, for consecutive (2, k)-out-of-(2, n) systems with local load-sharing, workloads on failed components will be equally shared by both vertically and horizontally adjacent components. Consequently, the component failure probability density function (FPDF) may be unequal and dependent on the actual load (both original and sharing load), which is determined by the states of adjacent components. Furthermore, the component FPDF may be time-varying with the failure sequences of adjacent components. Therefore, reliability modeling must consider the component dynamic process determined by the failure sequences of adjacent components, instead of the static configuration determined by failure states of adjacent components in the traditional models. To solve the above issues, this paper develops a new reliability model for consecutive (2, k)-out-of-(2, n) systems with local load-sharing. The system and component dynamic processes are automatically searched and combined into a set of state transition paths. In each path, the component load-varying process can be confirmed, by considering the influence of adjacent failures on its shared load. Based on the state transition paths, the component and system reliability models are further derived, according to the confirmed load processes of each component. Finally, we present graphical illustrations of the reliability evaluation of cable-strut systems in long-span suspension bridges.

Carbon sequestration of plantations formed by three kinds of forestation (natural forest to plantation (NP), grassland to plantation (GP), and cropland to plantation (CP)) greatly depends on the change of soil organic carbon density (SOCD) compared with its initial SOCD before forestation. However, this dependence was rarely studied, especially in semi-humid/arid regions with strong site variation. This limits the precise assessment and management of SOCD. Therefore, the SOCD variations of 0–100 cm soil layers in these three kinds of plantations were studied in the semi-humid/arid Liupan Mountains in northwestern China. The NP with high initial SOCD showed firstly a decrease and then an increase of SOCD up to 293.2 t·ha−1 at 40 years. The CP and GP with low and relatively high initial SOCD showed negligible and slight SOCD decrease after forestation, but then an increase up to 154.5 and 266.5 t·ha−1 at 40 years. After detecting the main factors influencing SOCD for each forestation mode, statistic relationships were fitted for predicting SOCD variation. This study indicates that besides forest age and biomass growth, the effects of initial vegetation, site-dependent initial SOCD, and SOCD capacity, also precipitation and air temperature in some cases, should be considered for more precise assessment and management of SOCD of plantations.

The use of various veterinary antibiotics (VAs) in animal husbandry raises serious concerns about the development of antibiotic resistance. Antibiotics such as tetracycline, oxytetracycline, sulfadiazine, norfloxacin, and enrofloxacin are the most frequently used antimicrobial compounds in animal husbandry and generate large eco-toxicological effects; however, they are still difficult to determine in a complex matrix such as swine manure. This study has developed an effective method for detecting five VAs in swine manure using Ultra-High-Performance Liquid Chromatography–Diode Array Detector (UHPLC–DAD) coupled with on-line solid-phase extraction (SPE). The results show that the mobile phase of ACN/0.01 M oxalic acid was the optimum at pH 3.0. VAs in a swine manure matrix were extracted using solid extraction buffer solution (T3) with 97.36% recovery. Sensitivity, accuracy, and precision were also evaluated. The validity study showed good linearity (R2 > 0.99). Limit of detection (LOD) was found to be from 0.1 to 0.42 µg mL−1 in the liquid fraction and from 0.032 to 0.58 µg g−1 dw in the solid fraction. The corresponding values of the limit of quantification (LOQ) ranged from 0.32 to 1.27 µg mL−1 for the liquid fraction and from 0.096 to 1.77 µg g−1 dw for the solid fraction. Therefore, the proposed method showed the potential applicability for detecting different antibiotic compounds from swine manure samples.

In dryland regions, soil moisture is an important limiting factor for canopy transpiration (T). Thus, clarifying the impact of soil moisture on T is critical for comprehensive forest—water management and sustainable development. In this study, T, meteorological factors (reference evapotranspiration, ETref), soil moisture (relative soil water content, RSWC), and leaf area index (LAI) in a Larix principis-rupprechtii plantation of Liupan Mountains in the dryland region of Northwest China were simultaneously monitored during the growing seasons in 2017–2019. A modified Jarvis—Stewart model was established by introducing the impact of RSWC in different soil layers (0–20, 20–40, and 40–60 cm, respectively) to quantify the independent contribution of RSWC of different soil layers to T. Results showed that with rising ETref, T firstly increased and then decreased, and with rising RSWC and LAI, T firstly increased and then gradually stabilised, respectively. The modified Jarvis—Stewart model was able to give comparable estimates of T to those derived from sap flow measurements. The contribution of RSWC to T in different soil layers has obvious specificity, and the contribution rate of 20–40 cm (13.4%) and 0–20 cm soil layers (6.6%) where roots are mainly distributed is significantly higher than that of 40–60 cm soil layer (1.9%). As the soil moisture status changes from moist (RSWC0–60cm ≥ 0.4) to drought (RSWC0–60cm < 0.4), the role of the soil moisture in the 0–20 cm soil layer increased compared with other layers. The impacts of soil moisture that were coupled into the Jarvis—Stewart model can genuinely reflect the environmental influence and can be used to quantify the contributions of soil moisture to T. Thus, it has the potential to become a new tool to guide the protection and management of forest water resources.

Sucrose content is a crucial indicator of quality and flavor in peanut seed, and there is a lack of clarity on the molecular basis of sucrose metabolism in peanut seed. In this context, we performed a comprehensive comparative transcriptome study on the samples collected at seven seed development stages between a high-sucrose content variety (ICG 12625) and a low-sucrose content variety (Zhonghua 10). The transcriptome analysis identified a total of 8334 genes exhibiting significantly different abundances between the high- and low-sucrose varieties. We identified 28 differentially expressed genes (DEGs) involved in sucrose metabolism in peanut and 12 of these encoded sugars will eventually be exported transporters (SWEETs). The remaining 16 genes encoded enzymes, such as cell wall invertase (CWIN), vacuolar invertase (VIN), cytoplasmic invertase (CIN), cytosolic fructose-bisphosphate aldolase (FBA), cytosolic fructose-1,6-bisphosphate phosphatase (FBP), sucrose synthase (SUS), cytosolic phosphoglucose isomerase (PGI), hexokinase (HK), and sucrose-phosphate phosphatase (SPP). The weighted gene co-expression network analysis (WGCNA) identified seven genes encoding key enzymes (CIN, FBA, FBP, HK, and SPP), three SWEET genes, and 90 transcription factors (TFs) showing a high correlation with sucrose content. Furthermore, upon validation, six of these genes were successfully verified as exhibiting higher expression in high-sucrose recombinant inbred lines (RILs). Our study suggested the key roles of the high expression of SWEETs and enzymes in sucrose synthesis making the genotype ICG 12625 sucrose-rich. This study also provided insights into the molecular basis of sucrose metabolism during seed development and facilitated exploring key candidate genes and molecular breeding for sucrose content in peanuts.

Many systems often experience multiple failures resulting from simultaneous exposure to degradation processes and random shocks. For a load-sharing system, the dependencies among the degradation processes, random shocks and component failures potentially cause the system to fail more easily, which poses new challenging issues to evaluate the reliability. A novel reliability model for loadsharing systems subject to dependent degradation processes and random shocks is proposed. The new model extends previous models for simple parallel systems by considering the characteristics and specific dependencies of load-sharing systems. In a load-sharing system, the workload and shock load shared by each surviving component will increase after a component failure, leading to the higher degradation rate, the more serious sudden degradation damage caused by random shocks, and the greater probability of hard failure. In the model, the analytical expression is utilized to calculate the complex reliability. The complexity of this calculation is caused by the stochastic failure time of surviving components, the stochastic arriving time of shocks, and their interaction. A case of the load-sharing redundant micro-engines in micro-electromechanical system is presented to demonstrate the proposed model, and the result shows that the reliability of load-sharing system is lower than that of a simple parallel system.

Germany (DE) and China (CN) have different political approaches in supporting the biogas sector. Three German and three Chinese large-scale biogas plants (BGPs) were evaluated as part of a year-round monitoring program. Laboratory methods were utilized to analyze the chemical indicators. Results showed a stable anaerobic digestion process without system failures in all BGPs. The methane yield had a range of 0.23–0.35 m3CH4/kgODM for DE BGPs and 0.11–0.22 m3CH4/kgODM for CN BGPs, due to different substrates and working temperatures. Financial analyses indicated that DE BGPs are viable under their current feed-in tariffs contracts. Their financial internal rate of return (IRR) ranged between 8 and 22%. However, all CN BGPs had negative IRRs, indicating that they are financially unfeasible. Risk analyses illustrated that DE BGPs will face financial nonviability if benefits decrease by 9–33% or costs increase by 10–49%, or if a combined worse case (benefit decrease and cost increase) of 5–20% occurs. Incentives to BGP operations are particularly important in China, where the government should consider switching the construction-based subsidy to a performance-based subsidy system to motivate the operators. BGP monitoring is necessary to understand the performance, in addition to briefing policymakers in case a policy reform is needed.

Aflatoxin B1 (AFB1) and aflatoxin B2 (AFB2) are the most common aflatoxins produced by Aspergillus flavus in peanuts, with high carcinogenicity and teratogenicity. Identification of DNA markers associated with resistance to aflatoxin production is likely to offer breeders efficient tools to develop resistant cultivars through molecular breeding. In this study, seeds of 99 accessions of a Chinese peanut mini-mini core collection were investigated for their reaction to aflatoxin production by a laboratory kernel inoculation assay. Two resistant accessions (Zh.h0551 and Zh.h2150) were identified, with their aflatoxin content being 8.11%−18.90% of the susceptible control. The 99 peanut accessions were also genotyped by restriction site-associated DNA sequencing (RAD-Seq) for a genome-wide association study (GWAS). A total of 60 SNP (single nucleotide polymorphism) markers associated with aflatoxin production were detected, and they explained 16.87%−31.70% of phenotypic variation (PVE), with SNP02686 and SNP19994 possessing 31.70% and 28.91% PVE, respectively. Aflatoxin contents of accessions with “AG” (existed in Zh.h0551 and Zh.h2150) and “GG” genotypes of either SNP19994 or SNP02686 were significantly lower than that of “AA” genotypes in the mean value of a three-year assay. The resistant accessions and molecular markers identified in this study are likely to be helpful for deployment in aflatoxin resistance breeding in peanuts.
