Testicular tumors represent a common form of solid tumor in young men, with choriocarcinoma of the testis being a rare, non-granulomatous germ cell tumor. It accounts for less than 0.3% of all testicular germ cell tumors. Pelvic and pulmonary metastases originating from testicular choriocarcinoma are exceptionally uncommon in men. This study describes a case of a 27-year-old male diagnosed with testicular choriocarcinoma, presenting initially with nausea, vomiting, and abdominal pain. Furthermore, this review encompasses cases of testiclar choriocarcinoma in individuals aged 30 years and below, both in China and internationally, over the past 20 years.

Therapeutic strategies medicated by M2 phenotypic macrophages or M2 macrophage inducer have been widely used to promote tissue remodeling. In this study, ultrasound-responsive bioactive microdroplets (MDs) encapsulated with bioactive molecule interleukin-4 (IL4, hereafter designated MDs-IL4) were fabricated to regulate macrophage polarization and potentiate the osteogenic differentiation of human mesenchymal stem cells (hBMSCs).Materials and Methods: The MTT assay, live and dead staining, and phalloidin/DAPI dual staining were used to evaluate biocompatibility in vitro. H&E staining was used to evaluate biocompatibility in vivo. Inflammatory macrophages were further induced via lipopolysaccharide (LPS) stimulation to mimic the pro-inflammatory condition. The immunoregulatory role of the MDs-IL4 was tested via macrophage phenotypic marker gene expression, pro-inflammatory cytokine level, cell morphological analysis, and immunofluorescence staining, etc. The immune-osteogenic response of hBMSCs via macrophages and hBMSCs interactions was further investigated in vitro.Results: The bioactive MDs-IL4 scaffold showed good cytocompatibility in RAW 264.7 macrophages and hBMSCs. The results confirmed that the bioactive MDs-IL4 scaffold could reduce inflammatory phenotypic macrophages, as evidenced by changing in morphological features, reduction in pro-inflammatory marker gene expression, increase of M2 phenotypic marker genes, and inhibition of pro-inflammatory cytokine secretion. Abstract Aiming at meetiing the need to filtering flight trajectory data for aircraft testing, a novel adaptive cubature Kalman filter (CKF) is proposed based on the maximum correntropy and Gaussian‐sum in this paper. Firstly, based on the traditional CKF algorithm, we introduced a Gaussian‐sum method to approximate non‐Gaussian noise to get more accurate filtering results in view of the problem of reduced filtering accuracy caused by the inherent non‐Gaussian nature of the noise and the system non‐linearity. Secondly, the maximum correntropy criterion is introduced to solve further the problem of improving the filtering accuracy of the system in the case of non‐linearity. Simulation results and actual data verification showed that the Square‐root cubature Kalman filter algorithm based on the maximum correntropy and Gaussian‐sum has higher accuracy than traditional filtering algorithms, which verified the algorithm's effectiveness in the application.

Before the implementation of conversion therapy, all patients underwent CT scan to assess the presence of hepatic steatosis and divided into the HS group (n = 124) and the non-HS group (n = 282). After using propensity score matching (PSM) to eliminate the potential confounding bias of the two groups, the conversion hepatectomy rate and long-term oncological survival in two groups were compared.Results: After 1:1 PSM, no significant difference was observed at baseline between patients in the HS group (n = 119) and the non-HS group (n = 119). Patients in the HS group had higher conversion hepatectomy rate from MDT evaluation (31.1% vs 18.5%, P = 0.029) and actual hepatectomy rate (30.2% vs 18.5%, P = 0.030), when compared with patients in the non-HS group, respectively. In addition, the HS group achieved better progression-free survival (PFS, P = 0.047) and overall survival (OS, P = 0.035) than that of the non-HS group. Multivariate logistic analysis confirmed that pretreatment HS was an independent predictor for conversion hepatectomy rate (OR, 2.393; 95% CI, 1.463–4.315, P = 0.001), and multivariate Cox analysis revealed that HS was an independent prognostic factor for PFS (HR, 0.493, 95% CI 0.281–0.866, P = 0.014) and OS (HR, 0.559, 95% CI 0.398–0.785, P = 0.001).Conclusion: For CLLM patients who underwent conversion therapy, hepatic steatosis could be an effective predictor for conversion hepatectomy rate and an independent prognostic factor for PFS and OS.Keywords: colorectal cancer, liver metastases, conversion therapy, hepatic steatosis, objective response rate, survival

Abstract Background T helper 17 (Th17) cells have proven to be crucial in the pathogenesis of neutrophils-dominant asthma. Hypoxia inducible factor-1α (HIF-1α) is involved in allergic responses in asthma. Our previous studies indicated that Methtyl-CpG binding domain protein 2 (MBD2) expression was increased in asthma patients. The aim of the present study is to understand how MBD2 interacts with HIF-1α to regulate Th17 cell differentiation and IL-17 expression in neutrophils-dominant asthma. Methods A neutrophils-dominant asthma mouse model was established using female C57BL/6 mice to investigate Th17 cell differentiation and MBD2 and HIF-1α expression regulation using flow cytometry, western blot or qRT-PCR. MBD2 and HIF-1α genes were silenced or overexpressed through lentiviral transduction to explore the roles of MBD2 in Th17 cell differentiation and IL-17 release in neutrophils-dominant asthma. Results A neutrophilic inflammatory asthma phenotype model was established successfully. This was characterized by airway hyperresponsiveness (AHR), increased BALF neutrophil granulocytes, activated Th17 cell differentiation, and high IL-17 levels. MBD2 and HIF-1α expression were significantly increased in the lung and spleen cells of mice with neutrophils-dominant asthma. Through overexpression or silencing of MBD2 and HIF-1α genes, we have concluded that MBD2 and HIF-1α regulate Th17 cell differentiation and IL-17 secretion. Moreover, MBD2 was also found to regulate HIF-1α expression. Conclusions Our findings have uncovered new roles for MBD2 and HIF-1α, and provide novel insights into the epigenetic regulation of neutrophils-dominant asthma.

Patients and methods: From June 2008 to December 2014, patients with RAS wild-type CLMs treated at Zhongshan Hospital, Fudan University were included. DNA extracted from formalin-fixed paraffin-embedded tissue of primary tumors was sequenced with next-generation sequencing for single-nucleotide polymorphism of 96 genes according to custom panel. Mutations were compared between synchronous and metachronous liver metastases and correlated with clinical characteristics. Results: A total of 161 patients were included: 93 patients with synchronous CLM and 68 patients with metachronous CLM. Patients with metachronous CLM were obviously elder. For pathology of primary tumors, synchronous CLMs were larger in size, poorly differentiated, and more frequently local advanced and lymph node positive. For evaluation of liver metastases, synchronous CLM had more and larger metastatic lesions. A rumen simulation technique apparatus with eight 800 mL fermentation vessels was used to investigate the effects of rumen degradable protein (RDP) level and non-fibre carbohydrate (NFC) type on ruminal fermentation, microbial growth, and populations of ruminal cellulolytic bacteria. Treatments consisted of two NFC types (starch and inulin) supplemented with 0 g/d (low RDP) or 1.56 g/d (high RDP) sodium caseinate. No significant differences existed among dietary treatments in the apparent disappearance of dietary nutrients except for dietary N, which increased with increased dietary RDP (PRuminococcus flavefaciens (P≤0.010). Non-fibre carbohydrate source did not affect daily microbial N flow regardless of dietary RDP, but ammonia N production was lower for inulin than for starch treatments under high RDP conditions (PFibrobacter succinogenes in solid fraction (P=0.023) and R. flavefaciens in liquid (P=0.017) and solid fractions (P=0.007), but it increased the carboxymethylcellulase activity in solid fraction (P=0.045). Current results suggest that starch and inulin differ in ruminal volatile fatty acid fermentation but have similar effects on ruminal digestion and microbial synthesis in vitro, although inulin suppressed the growth of partial ruminal cellulolytic bacteria.

BackgroundExposure of the developing brain to propofol results in cognitive deficits. Recent data suggest that inhibition of neuronal apoptosis does not prevent cognitive defects, suggesting mechanisms other than neuronal apoptosis play a role in anaesthetic neurotoxicity. Proper neuronal growth during development is dependent upon growth cone morphology and axonal transport. Propofol modulates actin dynamics in developing neurones, causes RhoA-dependent depolymerisation of actin, and reduces dendritic spines and synapses. We hypothesised that RhoA inhibition prevents synaptic loss and subsequent cognitive deficits. The present study tested whether RhoA inhibition with the botulinum toxin C3 (TAT-C3) prevents propofol-induced synapse and neurite loss, and preserves cognitive function.MethodsRhoA activation, growth cone morphology, and axonal transport were measured in neonatal rat neurones (5-7 days in vitro) exposed to propofol. Synapse counts (electron microscopy), dendritic arborisation (Golgi-Cox), and network connectivity were measured in mice (age 28 days) previously exposed to propofol at postnatal day 5-7. Memory was assessed in adult mice (age 3 months) previously exposed to propofol at postnatal day 5-7.ResultsPropofol increased RhoA activation, collapsed growth cones, and impaired retrograde axonal transport of quantum dot-labelled brain-derived neurotrophic factor, all of which were prevented with TAT-C3. Adult mice previously treated with propofol had decreased numbers of total hippocampal synapses and presynaptic vesicles, reduced hippocampal dendritic arborisation, and infrapyramidal mossy fibres. These mice also exhibited decreased hippocampal-dependent contextual fear memory recall. All anatomical and behavioural changes were prevented with TAT-C3 pre-treatment.ConclusionInhibition of RhoA prevents propofol-mediated hippocampal neurotoxicity and associated cognitive deficits.

In Chinese culture, certain digits are lucky and others unlucky.We test howsuch numerological superstition affects financial decision in the China initial public offering (IPO) market. We find that the frequency of lucky numerical stock listing codes exceeds what would be expected by chance. Also consistent with superstition effects, newly listed firms with lucky listing codes experience inferior post-IPO abnormal returns. Further tests suggest that our conclusions are not driven by endogeneity.

Newly-developed spectrographs with increased resolving powers, particularly those covering the near-IR range, allow the characterization of more and more absorption lines in stellar spectra. This includes the identification and confirmation of absorption lines and the calibration of oscillator strengths. In this study, we provide empirical values of |${{\rm log} gf}$| based on the abundances of classical Cepheids obtained with optical spectra to establish the consistency between optical and infrared abundance results. Using time series spectra of classical Cepheids obtained with WINERED spectrograph (0.97–1.35  |$\mu$| m, R |$\sim$| 28000), we demonstrate that we can determine the stellar parameters of the observed Cepheids, including effective temperature (⁠|${T_\mathrm{eff}}$|⁠), surface gravity (⁠|${{\rm log} g}$|⁠), microturbulence (⁠|$\xi$|⁠), and metallicity (⁠|${\rm {[{\mathrm{ M}}/\mathrm{ H}]}}$|⁠). With the newly calibrated relations of line-depth ratios, we can achieve accuracy and precision comparable to optical studies, with uncertainties of |$\sim$| 90 K and 0.108 dex for |${T_\mathrm{eff}}$|⁠ , and |${{\rm log} g}$|⁠ , respectively. Finally, we created a new atlas of absorption lines, featuring precise abundance measurements of various elements found in the atmosphere of Cepheids, including neutron-capture elements whose |${{\rm log} gf}$| values have been astrophysically calibrated. Purpose: To present updated information on odontogenic keratocyst (OKC) classification, etiology, genetic and molecular alterations, epidemiology, clinical presentation, radiographic characteristics, histological and immune histochemical features, differential diagnosis, treatment, and controversies, as well as a literature review of case frequencies in different countries. Materials and Methods: Studies were selected using the key words 'odontogenic keratocyst,' 'odontogenic cysts,' 'odontogenic keratocyst and clinical study'. Full-text papers were reviewed on the basis of the inclusion and exclusion criteria. The literature search aimed to find articles that would show the frequency of OKC, dentigerous cyst, radicular cyst, and other cysts. Results: OKC presents local aggression and high recurrence; therefore, a better understanding of its clinical characteristics and the genetic and molecular factors involved in this peculiar and controversial lesion is required. It is always essential to discuss treatment alternatives. Although OKC is an entity with a high recurrence, aggressive treatment is not advisable in all cases because factors such as commitment to anatomical structures and possible complications should be considered. However, periodic radiographic controls are advised. Conclusion: To reduce the high number of present cases worldwide, it is important to improve knowledge on this pathology so that accurate diagnoses can be achieved and appropriate treatment can be provided. Abstract Prehistoric settlement patterns on the Tibetan Plateau, particularly in the Qaidam Basin, have drawn significant scholarly interest. Yet, limited data have hindered a comprehensive understanding the Bronze Age chronology and landscape exploitation in this region. This paper presents 24 newly obtained radiocarbon dates from eight archaeological sites within the Qaidam Basin on the Tibetan Plateau. Integrated with existing 14 C data and spatial analysis, these findings reveal an overall continuous occupation of the basin from ca. 3600–2500 calBP, with a gradual shift from expansive lowland settlements to smaller, ephemeral high-altitude structures, identified around 3350 calBP. This change is consistent with demonstrated periods of climate degradation which likely drove strategic adaptations in landscape management and subsistence methods. Overall, this research not only establishes a more robust chronology for the Qaidam Basin’s Bronze Age, but also advances the discussion on human-environment interactions in high-altitude areas during late prehistory.

Abstract The formation of human collagen requires the presence of Prolyl 3-hydroxylase 1 (P3H1), but the regulatory mechanism of P3H1 remained insufficiently understood. Our study aimed to identify the role of P3H1 in clear cell renal cell carcinoma (ccRCC). P3H1 expression in ccRCC was validated using multiple databases and in vitro experiments. We performed a correlation analysis of P3H1 with drug sensitivity, immune checkpoints, and immune cell infiltration using transcriptome and single-cell sequencing. Drawing upon the Encyclopedia of RNA Interactomes database, we selected P3H1 as the focal point of our investigation, meticulously uncovering the intricate network of microRNAs and lncRNAs that potentially orchestrate ceRNA mechanisms. This study employs a multidimensional approach integrating vitro assays and multi-omics bioinformatics analyses to investigate P3H1’s impact on ccRCC prognosis, immune modulation, immune checkpoints, ceRNA regulatory network, drug sensitivity, and therapeutic responses, aiming to uncover new insights into its therapeutic potential and inform future clinical strategies.

The effects of bran types and solid-state fermentation conditions on the enzyme activity of bran-koji inoculated with Penicillium oxalicum M1816 were investigated, and the bran-koji with the highest enzyme activity was applied to Huangjiu production to evaluate its effects on the phenolic content, physicochemical indexes, and sense of Huangjiu. The results showed that when the diameter of bran was 20-mesh, the enzyme activity of bran-koji was corn bran>wheat bran>oat bran>rye bran. The highest enzyme activity in corn bran-koji was achieved under the following conditions: Glacial acetic acid was used to adjust the pH of the solid medium to 6.5, the initial moisture content of the bran-koji was 85% (V/W), the incubation temperature was 30 ℃, and the incubation time was 120 h. Compared with factory Huangjiu, the total amount of phenolic acids in the Huangjiu produced by adding corn bran-koji was 116.48 mg/L, which increased to 1.1-folds in comparison with that of the Huangjiu produced by adding no-corn bran-koji (55.53 mg/L). Abstract To improve ore recovery ratio and the quality of medium-length hole blasting in non-pillar sublevel caving method, this study first investigated the effects of brow line failure on rock movement and ore recovery indexes based on rock blasting fragmentation mechanisms, the granular dynamics theory and ore drawing theory. Subsequently, control strategies to enhance blasting quality and reduce brow line failure were explored through energy impact crushing experiments, experiments on the radial rock-breaking effects of blast holes with different detonation velocities, and underground physical simulation experiments using full blast hole length detonating cord blasting. The experimental results indicate that as the degree of brow line failure increases, the ore recovery ratio gradually decreases, while the waste ratio gradually increases. Bottom initiation and the use of the full blast hole length detonating cord were identified as the optimal initiation method and charging structure for medium-length hole blasting in non-pillar sublevel caving method, effectively improving blasting quality and reducing damage to the brow line.

Abstract Background Obesity in humans can lead to chronic diseases such as diabetes and cardiovascular disease. Similarly, subcutaneous fat (SCF) in pigs affects feed utilization, and excessive SCF can reduce the feed efficiency of pigs. Therefore, identifying factors that suppress fat deposition is particularly important. Numerous studies have implicated the gut microbiome in pigs’ fat deposition, but research into its suppression remains scarce. The Lulai black pig (LL) is a hybrid breed derived from the Laiwu pig (LW) and the Yorkshire pig, with lower levels of SCF compared to the LW. In this study, we focused on these breeds to identify microbiota that regulate fat deposition. The key questions were: Which microbial populations reduce fat in LL pigs compared to LW pigs, and what is the underlying regulatory mechanism? Results In this study, we identified four different microbial strains, Eubacterium siraeum, Treponema bryantii, Clostridium sp. CAG:413, and Jeotgalibaca dankookensis, prevalent in both LW and LL pigs. Blood metabolome analysis revealed 49 differential metabolites, including tanshinone IIA and royal jelly acid, known for their anti-adipogenic properties. E. siraeum was strongly correlated with these metabolites, and its genes and metabolites were enriched in pathways linked to fatty acid degradation, glycerophospholipid, and glycerolipid metabolism. In vivo mouse experiments confirmed that E. siraeum metabolites curb weight gain, reduce SCF adipocyte size, increase the number of brown adipocytes, and regulate leptin, IL-6, and insulin secretion. Finally, we found that one important pathway through which E. siraeum inhibits fat deposition is by suppressing the phosphorylation of key proteins in the PI3K/AKT signaling pathway through the reduction of tyrosine. Conclusions We compared LW and LL pigs using fecal metagenomics, metabolomics, and blood metabolomics, identifying E. siraeum as a strain linked to fat deposition. Oral administration experiments in mice demonstrated that E. siraeum effectively inhibits fat accumulation, primarily through the suppression of the PI3K/AKT signaling pathway, a critical regulator of lipid metabolism. These findings provide a valuable theoretical basis for improving pork quality and offer insights relevant to the study of human obesity and related chronic metabolic diseases. Video Abstract

Abstract Background Transfusion-dependent β-thalassemia (TDT) is a severe inherited disorder. Without regular treatment, patients with TDT experience complications that can significantly shorten life expectancy and severely impact both their quality of life and that of their families. The condition has attracted significant attention in global health discussions. Due to the challenges of blood supply shortages, the high costs of iron chelation therapy, and hematopoietic stem cell transplantation (HSCT), TDT presents a serious health risk to patients and imposes a substantial burden on families and society. However, research on the health-related quality of life (HRQoL) of thalassemia patients in China remains limited. This study evaluated the factors affecting the HRQoL of these patients, with the goal of developing strategies to improve their quality of life. Methods In this cross-sectional study, children with TDT were recruited from five treatment centers in Guangxi, a province with a high prevalence of thalassemia in China. Structured questionnaires were employed to gather relevant data on sociodemographic variables, disease characteristics, treatments, and associated costs. The HRQoL was assessed using the Transfusion-Dependent Quality of Life (TranQoL) questionnaire, with a proxy version for patients aged 0–11 years and a child version for those aged 12–18 years. Abstract Explicit identification of hydrochemical processes and their controlling factors within groundwater systems is critical for the sustainable utilization of water resources in coastal urban areas. This study was undertaken in the North Coastal Region of Jiaozhou Bay (NCRJB), located in the eastern part of Shandong Province, China, an area grappling with significant issues of groundwater quality degradation and water scarcity. A total of 105 groundwater samples and 34 surface water samples, collected from 2020 to 2024, were analyzed and studied using various hydrogeological tools, multivariate statistical analyses, and water quality assessment methods. These include the Piper diagram, hydrochemical facies evolution diagram (HFE-D), Principal Components Analysis (PCA), correlation analysis, stable isotope analysis, Water Quality Index (WQI), and USSL diagrams. The results indicated that all surface water and pore groundwater samples were categorized as Na-Cl type, exhibiting high Total Dissolved Solids (TDS) and Electrical Conductivity (EC) values, characteristics that render them poor to unsuitable for drinking and irrigation purposes. The fracture groundwater is predominantly of the Ca-Na-Cl mixed type, with average suitability for irrigation and a limited proportion (22.5%) deemed suitable for drinking. Seawater intrusion, primarily through the surface water system, and the impact of human activities were identified as the predominant controlling factors con-tributing to the degradation of the local groundwater environment. Field empirical investigations further validated the results derived from hydrogeological assessments, multivariate statistical analyses, and isotopic approaches. The long-term shifts in hydrochemical properties, along with the latent threat of seawater intrusion, exhibit an upward trend during the dry season and show a certain degree of mitigation during the wet season. This study highlights that field investigations, in conjunction with hydrochemical tools, multivariate statistical analyses, and stable isotope analysis, can successfully furnish reliable insights into the predominant mechanisms governing regional groundwater evolution within the context of long-term and intricate envi-ronmental settings.

Plants play an important role in maintaining the ecological balance of the biosphere, but often suffer from pathogenic fungi during growth. During our continuing mycological surveys of plant pathogens from terrestrial plants in Jiangxi and Yunnan provinces, China, 24 strains of Pestalotiopsis isolated from diseased and healthy tissues of plant leaves represented eight new species, viz. P. alpinicola, P. camelliicola, P. cyclosora, P. eriobotryae, P. gardeniae, P. hederae, P. machiliana and P. mangifericola. Multi-locus (ITS, tef1-α and tub2) phylogenetic analyses were performed using maximum-likelihood and Bayesian inference to reveal their taxonomic placement within Pestalotiopsis. Both molecular phylogenetic analyses and morphological comparisons supported them as eight independent taxa within Pestalotiopsis. Illustrations and descriptions of these eight taxa were provided, in conjunction with comparisons with closely related taxa in the genus. This work highlights the large potential for new fungal species associated with diseased plant leaves.

The traditional net-laying operation is highly dependent on manual labor, which not only has high labor intensity and high safety risks, but also has low efficiency, which has become an important factor restricting the balance of mining and excavation. Therefore, the core objective of this study is to design and implement a standardized mesh-laying strategy for underground coal mine robots, which improves the efficiency of mesh-laying operation, reduces the labor load of workers, enhances the safety of operation through the introduction of automation and intelligent technology, and then promotes the efficient, green and sustainable development of coal mine production. In order to realize the above research objectives, this study adopts a combination of simulation and experimental methods, based on the special environment and operating characteristics of underground, and designs a remote-controlled manipulator structure that assists the net-laying operation of digging-anchor integrated machine. The structure adopts a seven-axis articulated robotic arm with a top gripper in order to realize the multi-angle and high-precision laying of anchor nets. In addition, advanced computer simulation technology is used to analyze the dynamic performance, motion trajectory planning and force characteristics of the robotic arm, and the structural design and control algorithm of the robotic arm are optimized iteratively to ensure its stability and reliability in the complex underground environment. At the same time, a set of adaptive adjustment strategy is developed for the motion control of the robotic arm, so that the robotic arm can automatically adjust the motion parameters according to the actual working conditions (tunnel shape, mesh material), and realize accurate and efficient net-laying operation. The results show that the automated net-laying equipment significantly improves the efficiency of net-laying operation, compared with the traditional manual net-laying method, the efficiency is increased by more than 20%. Through automated operation, it realizes the goal of reducing the number of workers by more than 33%, which effectively alleviates the problem of tense human resources in underground coal mines. The labor intensity of workers has been greatly reduced, reducing the high degree of climbing work, reducing the risk of work-related injuries, and the labor intensity has been reduced by more than 80%. At the same time, the automated net-laying process reduces the possibility of human error and significantly improves operational safety. In the future, with the continuous iteration and improvement of the technology, this technology is expected to play a more important role in the intelligent construction of coal mines, and promote the development of coal mine production in the direction of safer, greener and more efficient.

Abstract Defect tolerance is a critical enabling factor for efficient lead-halide perovskite materials, but the current understanding is primarily on band-edge (cold) carriers, with significant debate over whether hot carriers can also exhibit defect tolerance. Here, this important gap in the field is addressed by investigating how intentionally-introduced traps affect hot carrier relaxation in CsPbX3 nanocrystals (X = Br, I, or mixture). Using femtosecond interband and intraband spectroscopy, along with energy-dependent photoluminescence measurements and kinetic modelling, it is found that hot carriers are not universally defect tolerant in CsPbX3, but are strongly correlated to the defect tolerance of cold carriers, requiring shallow traps to be present (as in CsPbI3). It is found that hot carriers are directly captured by traps, instead of going through an intermediate cold carrier, and deeper traps cause faster hot carrier cooling, reducing the effects of the hot phonon bottleneck and Auger reheating. This work provides important insights into how defects influence hot carriers, which will be important for designing materials for hot carrier solar cells, multiexciton generation, and optical gain media.

Abstract Genome differential positioning within interphase nuclei remains poorly explored. We extended and validated Tyramide Signal Amplification (TSA)-seq to map genomic regions near nucleoli and pericentric heterochromatin in four human cell lines. Our study confirmed that smaller chromosomes localize closer to nucleoli but further deconvolved this by revealing a preference for chromosome arms below 36-46 Mbp in length. We identified two lamina associated domain subsets through their differential nuclear lamina versus nucleolar positioning in different cell lines which showed distinctive patterns of DNA replication timing and gene expression across all cell lines. Unexpectedly, active, nuclear speckle-associated genomic regions were found near typically repressive nuclear compartments, which is attributable to the close proximity of nuclear speckles and nucleoli in some cell types, and association of centromeres with nuclear speckles in human embryonic stem cells (hESCs). Our study points to a more complex and variable nuclear genome organization than suggested by current models, as revealed by our TSA-seq methodology.

Abstract Despite the pivotal role of stannum doping in achieving ultrahigh piezoelectric performance in barium titanate-based ceramics, the fundamental mechanisms underlying this enhancement remain elusive. Here, we introduce a single variable nonstoichiometric stannum strategy in lead-free barium titanate-based ceramics with giant piezoelectricity, revealing that stannum doping contributes intrinsically and extrinsically to enhance piezoelectricity. Density functional theory calculations elucidate the intrinsic enhancement of polarization arising from lattice distortion and increased space for titanium-oxygen bonds induced by optimal stannum doping, which is corroborated by Rayleigh analysis. A phase transition from ferroelectric multiphase coexistence to paraelectric phase is observed, alongside a rapid miniaturized and eventually disappeared domains with increasing stannum doping. This evolution in phase structure and domain configuration induces a nearly vanishing polarization anisotropy and low domain wall energy, facilitating easy polarization rotation and domain wall motion, thereby significantly contributing to the extrinsic piezoelectric response. Consequently, the origins of ultrahigh performance can be attributed to the synergistic effect of stannum-induced intrinsic and extrinsic contributions in barium titanate-based ceramics. This study provides fundamental insights into the role of doping elements and offers guidance for the design of high-performance piezoelectrics.

Objective The stray current harm in urban rail transit lines is increasingly non-ignorable. Stray current is not easy to be detected, and the amount of the rail-ground transition resistance is a crucial factor affecting stray current. Therefore, measuring the amount of the rail-ground transition resistance is an effective stray current control method to judge the influence degree of stray current. Method Based on EMTP (electromagnetic transient program) software, a simulation model of the track circuit distributed parameters is built. The respective influence patterns of the distributed capacitance, distributed inductance, longitudinal resistance of the steel rail, and the rail-ground transition resistance on the time-varying characteristic curve of impulse impedance are obtained by using the single control variable method. The method of using impulse impedance time-varying ratio curve to judge whether the rail-ground transition resistance meets the requirement is proposed, and further verified by actual measurement of an urban rail transit line in Chengdu. Result & Conclusion Within 7~20 μs, only the rail-ground transition resistance parameter may affect the time-varying characteristics of the impulse impedance. Therefore, it can be used to reflect the condition of the rail-ground transition resistance and make valid judgement on whether the rail-ground transition resistance meets the requirement in the specification or not. Test results show that the measuring method is feasible and practical.

Nanobody (Nb) is derived from the variable domain of heavy-chain antibody (HCAb), naturally displaying notable properties like nano-scale size, exceptional stability, high specificity, low immunogenicity, and cryptic epitope accessibility. These features contribute to its great therapeutic potential as a valuable research tool across diverse diseases, especially autoimmune diseases (AIDs). Caplacizumab (Cablivi®) is the first nanobody drug approved for treating acquired thrombotic thrombocytopenic purpura (aTTP). This review summarizes the biomolecular structure, usage of Nb as a foundation of recombinant constructs, and biochemical properties of nanobodies. As attractive therapeutic candidates, many clinical trials of Nbs have been conducted, elucidating potential therapeutic strategies for AIDs. Therefore, the preclinical development and application of Nbs in AIDs are emphasized throughout this review.

Abstract In acid fracturing, acid is injected into artificial fracture to dissolve the rock, and the rock surface is shaped into the non-uniform morphology. This non-uniform morphology is the main cause of fracture conductivity after acid etching. The effects of acid type, flow rate, contact time and rock type on the non-uniform morphology of acid-etched rock surface have been studied. However, when the direction of acid flow is determined, the difference of dissolution characteristics in different directions of rock surface is not investigated. In this paper, acid rock reaction experiment and analysis of rock surface morphology after acid etching are carried out by using rock samples with flat surface. The difference of morphological characteristic parameters of rock surface in different directions under the condition of acid flow is discussed. It was found that the mean drop height, drop height range and roughness coefficient increase with the increase of acid rock reaction rate on flat rock surface. The morphological characteristic parameters of acid-etched rock face of limestone are greater than those of dolomite, which is conducive to forming acid-etched channels. According to the statistics of all experimental data, the larger average drop height of rock surface is distributed in the direction of parallel acid flow, the larger drop height range is distributed in the direction of vertical acid flow and other directions, and the larger roughness coefficient is distributed in other directions.

Lipopolysaccharide (LPS) plays an important role in Raw264.7 macrophage ferroptosis and inflammation. Purine-rich element-binding protein B (Purb) influences cellular processes by regulating gene expression as a transcription factor. However, the effect and molecular mechanism of Purb in regulating Raw264.7 macrophage ferroptosis is still unknown. In this study, we used malondialdehyde, glutathione (GSH) assays, Fe2+ fluorescence, reactive oxygen species staining, and western blotting to determine the effect of Purb on LPS-induced Raw264.7 macrophage ferroptosis. Pharmacological inhibitor of ferroptosis was utilized to explore its potential effects for inflammation by reverse transcription-quantitative polymerase chain reaction analysis. Meanwhile, chromatin immunoprecipitation was performed to verify the binding of Purb and the GSH-dependent peroxidase 4 (Gpx4). The results showed that LPS-induced inflammation in Raw264.7 macrophages was inhibited by ferroptosis inhibitor Fer-1 treatment. LPS inhibited the expression of Purb in Raw264.7 macrophages. In addition, Purb overexpression relieved the ferroptosis, and inflammatory response of Raw264.7 macrophages induced by LPS. Mechanistically, the binding of Purb to the Gpx4 promoter was decreased after LPS stimulation. Therefore, we concluded for the first time that Purb played a critical role in LPS-induced ferroptosis and inflammatory response by regulating the transcription of Gpx4. These results provide a theoretical basis for further research on the macrophage ferroptosis and inflammation.

ObjectivesThis study seeks to explore the influence of water depth on the drag reduction effect and optimal drag reduction height of an interceptor. Based on the numerical solution approach of the RANS equations, the flow field around a high-speed monohull ship with various height interceptors is numerically simulated. MethodsFirst, the validity of the numerical method is verified by a DTMB 5415 high-speed ship. Next, the flow field around a high-speed monohull ship is numerically simulated under different interceptor heights and water depths. Based on the numerical results, the resistance, navigation state and flow field characteristics under different working conditions are analyzed. ResultsThe results show that the drag reduction effect of the interceptor has an obvious shallow water effect. The optimal drag reduction height and reduction of trim angle of the interceptor is unchanged with the change in water depth. The interceptor has a certain effect on reducing wave-making behind the ship, but the effect decreases with the decrease in water depth. ConclusionsThis study has certain practical reference value for the application of interceptors in shallow water.

Abstract Chloroplasts are key players in photosynthesis and immunity against microbial pathogens. However, the precise and timely regulatory mechanisms governing the control of photosynthesis-associated nuclear genes (PhANGs) expression in plant immunity remain largely unknown. Here we report that TaPIR1, a Pst-induced RING-finger E3 ubiquitin ligase, negatively regulates Pst resistance by specifically interacting with TaHRP1, an atypical transcription factor histidine-rich protein. TaPIR1 ubiquitinates the lysine residues K131 and K136 in TaHRP1 to regulate its stability. TaHRP1 directly binds to the TaHRP1-binding site elements within the PhANGs promoter to activate their transcription via the histidine-rich domain of TaHRP1. PhANGs expression induces the production of chloroplast-derived ROS. Although knocking out TaHRP1 reduces Pst resistance, TaHRP1 overexpression contributes to photosynthesis, and chloroplast-derived ROS production, and improves disease resistance. TaPIR1 expression inhibits the downstream activation of TaHRP1 and TaHRP1-induced ROS accumulation in chloroplasts. Overall, we show that the TaPIR1-mediated ubiquitination and degradation of TaHRP1 alters PhANGs expression to disrupt chloroplast function, thereby increasing plant susceptibility to Pst.
