Potato tuber is easy to be damaged and caused browning, which will affect its nutrition, sense quality and safety. Different potato varieties have different resistances to browning. In this study, 27 different potato germplasm resources,were used to measure four indexes including polyphenol oxidase (PPO), browning degree (BD), browning index and after cooking darkening (ACD), for indentifying and selecting anti-browning potato varieties (lines). The 27 materials could be divided into four groups. High anti-browning, group including 09306-82 and 13041-52 had high resistance to browning. Anti-browning group, included Yanshu 4, Q 8, 12-1, Chunshu 3, 19-1, 14018-142, Atlantic, Longshu 12, Weiyu 5, Lishu 6, Chunshu 5, and Zaodabai, had some obvious deficiencies in the related anti-browning indicators. Medium anti-browning group, including 09001-136, Zhongshu 5, Longshu 4, Dongnong 310, Qingshu 9, 15-1, Yunshu 505, BF006, Minshu 1, and Qianyu 8, had poor resistance to browning. Easy browning group, containing Chunshu 6 and Favorita, had very weak resistance to browning. The selected varieties (lines) with very high or high resistance to browning can be used in potato breeding for browning resistance , providing variety support for potato processing industry, and the easy browning materials can be used to study the browning mechanism of potato.

A series of rhodanine derivatives RB1–RB23 were synthesized through a two-round screening. Their Mycobacterial tuberculosis (Mtb) InhA inhibitory activity and Mtb growth blocking capability were evaluated. The most potent hit compound RB23 indicated comparable InhA inhibiton (IC50 = 2.55 μM) with the positive control Triclosan (IC50 = 6.14 μM) and Isoniazid (IC50 = 8.29 μM). Its improved growth-blocking effect on Mtb and low toxicity were attractive for further development. The docking simulation revealed the possible binding pattern of this series and picked the key interacted residues as Ser20, Phe149, Lys165 and Thr196. The 3D-QSAR model visualized the SAR discussion and hinted new information. Modifying the surroundings near rhodanine moiety might be promising attempts in later investigations.

Human serum albumin (HSA) is a biological macromolecule with important physiological functions; abnormal HSA levels are associated with coronary heart disease, multiple myeloma, diabetes, nephropathy, neurometabolic disorders, liver cirrhosis and other diseases. Therefore, accurate and quantitative detection of HAS have extremely important research and application value in biological science, molecular biology, clinical medicine and other fields. As for the detection method of HSA, dye-binding method and immune method are the first to be used, and have been applied in clinical detection. In recent years, many new detection technologies have emerged, such as fluorescent probe detection method, nano-materials for HSA detection, biosensor and so on. Although there are many methods developed recently to detect HSA, comprehensive reviews for HSA detection methods are still rare. Thus, writing this review to fill in the blank is in need. In order to highlight the recent progress in the field of HSA detection, in this review, the methods used to detect HSA are summarized and sorted, the advantages and disadvantages of these detection methods are also listed, then the research progress of small molecular fluorescence probe method is emphatically introduced in this paper. Then, we briefly discussed the challenges and future development directions in this field.

A novel fluorescent sensor CysW-1 was introduced for cysteine detection via the cleavage reaction of two fluorophores. It exhibited advantages including high sensitivity, high selectivity and low toxicity. By optimizing the conditions, a relatively steady and practical white light emitting system was successfully generated. The system was applicable in a wide pH range (7.0–12.0) and a long period (over 12 h). The most attractive point of CysW-1 was its biocompatibility, which ensured the application of the white light emitting system in living cell imaging and further pre-clinical potential.

The antibacterial agents and therapies today are facing serious problems such as drug resistance. Introducing dual inhibiting effect is a valid approach to solve this trouble and bring advantages including wide adaptability, favorable safety and superiority of combination. We started from potential DNA Gyrase inhibitory backbone isatin to develop oxoindolin derivatives as atypical dual Gyrase (major) and FabH (assistant) inhibitors via a two-round screening. Aiming at blocking both duplication (Gyrase) and survival (FabH), most of synthesized compounds indicated potency against Gyrase and some of them inferred favorable inhibitory effect on FabH. The top hit I18 suggested comparable Gyrase inhibitory activity (IC50 = 0.025 μM) and antibacterial effect with the positive control Novobiocin (IC50 = 0.040 μM). FabH inhibitory activity (IC50 = 5.20 μM) was also successfully introduced. Docking simulation hinted possible important interacted residues and binding patterns for both target proteins. Adequate Structure-Activity Relation discussions provide the future orientations of modification. With high potency, low initial toxicity and dual inhibiting strategy, advanced compounds with therapeutic methods will be developed for clinical application.

To develop a high-efficiency desulfurizer for the production of pipeline steel with the LD-RH-CC process, the desulfurization efficiency and mechanism of CaO-saturated slag were studied using a vacuum-induction furnace and Si-Mo electric resistance furnace. The results show that the desulfurization ability for slag with a small amount of solid CaO was highly enhanced under conditions of satisfied kinetics. The desulfurization reaction of CaO-saturated slag depended on the liquid slag rather than the solid CaO, as sulfur was not detected inside the solid CaO. The desulfurization efficiency of CaO-saturated slag was also influenced by the amount of residual liquid slag and its sulfur absorption ability. The sulfur absorption ability of CaO-CaF2 slag was analyzed to be much higher than that of CaO-Al2O3-CaF2-(SiCO2) slag. Moreover, the effect of solid CaO on the desulfurization kinetics was evaluated and the application conditions of CaO-saturated slag were discussed.

Organelles like lysosomes usually exhibit dynamic changes of locations, morphologies, and components, which could be reflected by the local viscosity. For monitoring lysosomal viscosity, a novel rotor P7-B was introduced with an improved auxochromic dynamic motor of azepanyl. All through the evaluating processes including optical tests, wave-band optimization and intracellular applications, P7-B exhibited comparable targeting capability and better auxochroming potential than morpholine-containing rotors. More than offering practical lysosomal viscosity monitoring tools or referential auxochromic dynamic motors, this work raised significant information for transcending the structural limits in future investigations.

Broadened antibacterial activity was introduced to rhodanine derivatives targeting Mycobacterial tuberculosis enoyl-acyl carrier protein reductase (Mtb InhA) by recruiting feature of xacins to bring DNA Gyrase B inhibitory capability. This is significant for preventing further bacterial injections in the tuberculosis treatment. The most potent compound Cy14 suggested comparable bioactivity (IC50 = 3.18 µM for Mtb InhA; IC50 = 10 nM for DNA Gyrase B) with positive controls. Structure-activity relationship discussion and molecular docking model revealed the significance of rhodanine moiety and derived methoxyl on meta-position, pointing out orientations for future modification.

The establishment of independent control area for JiBei grid will improve the flexibility of regulation for local units, and absorb more wind power and other clean energy. However, due to the high proportion of wind power capacity and the fluctuation and intermittent of wind power, it presents a challenge to the power supply safety. In this paper, based on the prediction of regulation demand and regulation speed in different time scales, dynamic hierarchical control method is proposed. Several strategies of hierarchical control under the emergency of steady climbing, reverse and fluctuations are established. Hence, it improves the capacity of power supply safety and the proportion of wind absorption. Finally, based on the measurement of characteristics of wind fluctuation, the value of reserve capacity in different control levels is calculated.

Potato late blight, caused by the oomycete pathogen Phytophthora infestans , is the most serious disease of potato worldwide. The adoption of varieties with resistance genes, especially broad-spectrum resistance genes, is the most efficient approach to control late blight. Solanum demissum is a well-known wild potato species from which 11 race-specific resistance genes have been identified, however, no broad-spectrum resistance genes like RB have been reported in this species. Here, we report a novel reisistance locus from S. demissum that potentially confer broad-spectrum resistance to late blight. A small segregating population of S. demissum were assessed for resistance to aggressive P. infestans isolates (race 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, and 11). This coupled with nucleotide binding site (NBS) profiling analyses, led to the identification of three fragments that linked to the potential candidate resistance gene(s). Cloning and sequence analysis of these fragments suggested that the identified resistance gene locus is located in the region containing R2 resistance gene at chromosome 4. Based on the sequences of the cloned fragments, a co-segregating sequence characterized amplified region (SCAR) marker, RDSP, was developed. The newly identified marker RDSP will be useful for marker assisted breeding and further cloning of this potential resistance gene locus.

In china, increasing large-scale wind power integration has brought much more peak load regulation demand, which is the bottleneck of the integration of wind power to systems. This paper analyzed the influence mechanism of large-scale wind power on peak load regulation demand. And critical indicators are constructed based on the daily peak-valley difference to evaluate peak load regulation demand caused by wind power integration. Statistical analyses based on large number of actual dates reflect the influence of large-scale wind power integration on peak load regulation demand, which can help provide more decision-making information for planning and operation.

Based on the phase distribution which is defined in this paper, a new classification algorithm of M-ary phase shift keying (PSK) signals is proposed. To classify the modulation type of the M-ary PSK signal, the phase distribution of the unclassified signal is calculated firstly, and then the characters of the signal modulation are extracted by computing the FFT of the phase distribution. Moreover, the method is improved in this paper that it is extended to MPSK baseband signals with frequency offset, and the calculation complexity is reduced. Simulation result shows that the accuracy rate of the classification of BPSK, QPSK and 8PSK signals can reach 98.5% when the symbol length is 500, SNR is 3dB, and 16PSK signals can also be well classified when the SNR improves to 9dB.

Generation scheduling is one of the major contents of power dispatch and has an important impact on the safe and efficient operation of power grid. As a newly-established provincial power grid, Jibei power grid bears features like multiple regional wind power installations and high rate of external power receiving, etc., and it urgently needs to develop a secured and economic power scheduling system with comprehensive consideration of various constraints. Aiming at features of Jibei power grid, this paper gives an overall design scheme of the day-ahead power scheduling system. The application indicates that this system can efficiently optimize generation schedule and power flow distribution, intensify the decision ability and intelligence level of power dispatch.

Based on security constraint economic dispatch module, and with wind power prior utilization as target, the day-ahead optimum dispatching model of wind-thermal power is constructed to improve the acceptance ability of intermittent new energy generation. The practical application in Jibei power grid indicates that this model can meet the compiling requirements of day-ahead generation schedule, realizes the prior acceptance of wind power and contributes to the energy saving and emissions reduction in power field under the power grid security constraint.

In high speed continuous casting of peritectic steel slabs, mold fluxes with high basicity are required for less surface defect product. However, the basicity of remaining liquid slag film tends to decrease in casting process because of the crystallization of 3CaO • 2SiO2 • CaF2. Thus, a way is put forward to improve mold fluxes properties by raising the original basicity. In order to confirm the possibility of this method, the effect of rising original basicity on the properties of mold fluxes is discussed. Properties of high fluorine based mold fluxes with different basicities and contents of CaF2, Na2O, and MgO were measured, respectively. Then, properties of higher basicity mold fluxes were discussed and compared with traditional ones. The results show that increasing the basicity index can improve the melting and flow property of mold fluxes. With the increasing basicity, crystallization rate of mold fluxes increases obviously and crystallization temperature tends to decrease when the basicity exceeds 1. 35. The method presented before is proved as a potential way to resolve the contradiction between horizontal heat transfer controlling and solidified shell lubricating for peritectic steel slab casting. But further study on improving the flow property of liquid slag is needed. This work can be used to guide mold fluxes design for high speed continuous casting of peritectic steel slabs.

Stearoyl-acyl carrier protein desaturase (SAD), locating in the plastid stroma, is an important fatty acid biosynthetic enzyme in higher plants. SAD catalyzes desaturation of stearoyl-ACP to oleyl-ACP and plays a key role in determining the homeostasis between saturated fatty acids and unsaturated fatty acids, which is an important player in cold acclimation in plants. Here, four new full-length cDNA of SADs (ScoSAD, SaSAD, ScaSAD and StSAD) were cloned from four Solanum species, Solanum commersonii, S. acaule, S. cardiophyllum and S. tuberosum, respectively. The ORF of the four SADs were 1182 bp in length, encoding 393 amino acids. A sequence alignment indicated 13 amino acids varied among the SADs of three wild species. Further analysis showed that the freezing tolerance and cold acclimation capacity of S. commersonii are similar to S. acaule and their SAD amino acid sequences were identical but differed from that of S. cardiophyllum, which is sensitive to freezing. Furthermore, the sequence alignments between StSAD and ScoSAD indicated that only 7 different amino acids at residues were found in SAD of S. tuberosum (Zhongshu8) against the protein sequence of ScoSAD. A phylogenetic analysis showed the three wild potato species had the closest genetic relationship with the SAD of S. lycopersicum and Nicotiana tomentosiformis but not S. tuberosum. The SAD gene from S. commersonii (ScoSAD) was cloned into multiple sites of the pBI121 plant binary vector and transformed into the cultivated potato variety Zhongshu 8. A freeze tolerance analysis showed overexpression of the ScoSAD gene in transgenic plants significantly enhanced freeze tolerance in cv. Zhongshu 8 and increased their linoleic acid content, suggesting that linoleic acid likely plays a key role in improving freeze tolerance in potato plants. This study provided some new insights into how SAD regulates in the freezing tolerance and cold acclimation in potato.

To employ (18)F-fluoro-2-deoxyglucose ((18)F-FDG) and (3-N-[(11)C] methylspiperone)(11)C-NMSP microPET to assess the changes of regional cerebral glucose metabolism and the expression of dopamine receptor type 2 (DRD(2)) in a rat model of Parkinson's disease (PD).A hemiparkinsonian model was established in rats by unilateral pretreatment with 6-hydroxydopamine (6-OHDA). At 2 weeks after 6-OHDA insult, (18)F-FDG and (11)C-NMSP microPET scan were performed to compare the differences of regional cerebral glucose metabolism and the expression of DRD(2) between the PD and control groups respectively. The immunohistochemical staining was used to detect the expression of tyrosine hydroxylase in two groups.In the PD model, the glucose metabolism rates were 88.2% ± 2.2%, 94.5% ± 4.5% and 96.2% ± 5.8% respectively, in right striatum, hippocampus and sensorimotor cortex. And they were significantly lower than those in the control group [92.7% ± 2.8% (P0.01), 98.9% ± 3.9% (P0.01)102.8% ± 2.1% (P0.01)]. The expression of DRD(2) in right striatum was significantly higher in the PD group than that in the control group (112.9% ± 9.0% vs 102.3% ± 1.4%, P0.01).In the PD rats, glucose metabolism decreases in injured side striatum, hippocampus and sensorimotor cortex while and the expression of DRD(2) increases in injured side striatum.(18)F-FDG and (11)C-NMSP microPET can effectively assess the regional cerebral glucose metabolism and the expression of DRD(2) in PD. They may serve as effective molecular imaging tools for an early diagnosis of PD.

Symbol rate is an important parameter in digital communication. In the area of non-cooperative communication, the precision of symbol rate estimation is essential to the success of subsequent signal processes such as demodulation. This paper proposes an approach for estimation of symbol rate for M-ary FSK signals. In this approach, an M-ary FSK signal is decomposed into M single tones, and the symbol rate is estimated by extracting the spectral line corresponding to the symbol rate in the spectrum of the summation of each single tone envelope signal. The implementation of the methodology does not require any apriori information. Computer simulations and analysis show that the approach provides high estimation accuracy and still keeps a good performance even at low input SNR.
