Abstract Demyelination occurs widely in the central nervous system (CNS) neurodegenerative diseases, especially the multiple sclerosis (MS), which with a complex and inflammatory lesion microenvironment inhibiting remyelination. Sirtuin6 (SIRT6), a histone/protein deacetylase is of interest for its promising effect in transcriptional regulation, cell cycling, inflammation, metabolism and longevity. Here we show that SIRT6 participates in the remyelination process in mice subjected to LPC-induced demyelination. Using pharmacological SIRT6 inhibitor or activator, we found that SIRT6 modulated LPC-induced damage in motor or cognitive function. Inhibition of SIRT6 impaired myelin regeneration, exacerbated neurological deficits, and decreased oligodendrocyte precursor cells (OPCs) proliferation and differentiation, whereas activation of SIRT6 reversed behavioral performance in mice, demonstrating a beneficial effect of SIRT6. Importantly, based on RNA sequencing analysis of the corpus callosum tissues, it was further revealed that SIRT6 took charge in regulation of glial activation during remyelination, and significant alterations in CHI3L1 were obtained, a glycoprotein specifically secreted by astrocytes. Impaired proliferation and differentiation of OPCs could be induced in vitro using supernatants from reactive astrocyte, especially when SIRT6 was inhibited. Mechanistically, SIRT6 regulates the secretion of CHI3L1 from reactive astrocytes by histone-H3-lysine-9 acetylation (H3K9Ac). Adeno-associated virus-overexpression of SIRT6 (AAV-SIRT6-OE) in astrocytes improved remyelination and functional recovery after LPC-induced demyelination, whereas together with AAV-CHI3L1-OE inhibits this therapeutic effect. Collectively, our data elucidate the role of SIRT6 in remyelination and further reveal astrocytic SIRT6/CHI3L1 as the key regulator for improving the remyelination environment, which may be a potential target for MS therapy.

Abstract Background Diurnal and nocturnal mammals have evolved distinct pathways to optimize survival for their chronotype-specific lifestyles. Conventional rodent models, being nocturnal, may not sufficiently recapitulate the biology of diurnal humans in health and disease. Although diurnal rodents are potentially advantageous for translational research, until recently, they have not been genetically tractable. The present study aims to address this major limitation by developing experimental procedures necessary for genome editing in a well-established diurnal rodent model, the Nile grass rat (Arvicanthis niloticus). Results A superovulation protocol was established, which yielded nearly 30 eggs per female grass rat. Fertilized eggs were cultured in a modified rat 1-cell embryo culture medium (mR1ECM), in which grass rat embryos developed from the 1-cell stage into blastocysts. A CRISPR-based approach was then used for gene editing in vivo and in vitro, targeting Retinoic acid-induced 1 (Rai1), the causal gene for Smith-Magenis Syndrome, a neurodevelopmental disorder. The CRISPR reagents were delivered in vivo by electroporation using an improved Genome-editing via Oviductal Nucleic Acids Delivery (i-GONAD) method. The in vivo approach produced several edited founder grass rats with Rai1 null mutations, which showed stable transmission of the targeted allele to the next generation. CRISPR reagents were also microinjected into 2-cell embryos in vitro. Large deletion of the Rai1 gene was confirmed in 70% of the embryos injected, demonstrating high-efficiency genome editing in vitro. Conclusion We have established a set of methods that enabled the first successful CRISPR-based genome editing in Nile grass rats. The methods developed will guide future genome editing of this and other diurnal rodent species, which will promote greater utility of these models in basic and translational research.

2-Ethylhexyl diphenyl phosphate (EHDPP) is a representative organophosphorus flame retardant (OPFR) that has garnered attention due to its widespread use and potential adverse effects. EHDPP exhibits cytotoxicity, genotoxicity, developmental toxicity, and endocrine disruption. However, the toxicity of EHDPP in mammalian oocytes and the underlying mechanisms remain poorly understood. Melatonin is a natural free radical scavenger that has demonstrated cytoprotective properties. In this study, we investigated the effect of EHDPP on mouse oocytes in vitro culture system and evaluated the rescue effect of melatonin on oocytes exposed to EHDPP. Our results indicated that EHDPP disrupted oocyte maturation, resulting in the majority of oocytes arrested at the metaphase I (MI) stage, accompanied by cytoskeletal damage and elevated levels of reactive oxygen species (ROS). Nevertheless, melatonin supplementation partially rescued EHDPP-induced mouse oocyte maturation impairment. Results of single-cell RNA sequencing (scRNA-seq) analysis elucidated potential mechanisms underlying these protective effects. According to the results of scRNA-seq, we conducted further tests and found that EHDPP primarily disrupts mitochondrial distribution and function, kinetochore-microtubule (K-MT) attachment, DNA damage, apoptosis, and histone modification, which were rescued upon the supplementation of melatonin. This study reveals the mechanisms of EHDPP on female reproduction and indicates the efficacy of melatonin as a therapeutic intervention for EHDPP-induced defects in mouse oocytes.

The high temperature superconductors enhanced magnetoplasmadynamic thruster is a promising device for various space mission scenarios. However, a critical issue hindering the enhancement of the thruster’s performance is the problem of anode power deposition. Previous studies have identified the anode fall voltage as a major contributor to this power deposition. This paper introduces an approximate analytical formula for calculating anode power deposition resulting from anode fall voltage. The paper further analyzes and calculates the variation of anode fall voltage with an applied magnetic field and discharge current. The findings reveal a direct correlation between applied magnetic field and both anode fall voltage and power, indicating an increase with higher magnetic field values. In contrast, while the anode fall voltage experiences a slight decline with increased discharge current, the anode power deposition remains elevated due to the transition from field ionization to thermal ionization. To gain a comprehensive understanding of anode power deposition, an experiment was conducted to obtain measurements under different operating conditions, involving varying levels of applied magnetic field and discharge current. The experimental results align with the theoretical predictions. These insights have the potential to serve as a valuable reference for enhancing the efficiency of magnetoplasmadynamic thrusters, bringing us closer to unlocking the full potential of these remarkable propulsion systems in the realm of space exploration.

Triocresyl phosphate (TOCP) was commonly used as flame retardant, plasticizer, lubricant, and jet fuel additive. Studies have shown adverse effects of TOCP on the reproductive system. However, the potential harm brought by TOCP, especially to mammalian female reproductive cells, remains a mystery. In this study, we employed an in vitro model for the first time to investigate the effects of TOCP on the maturation process of mouse oocytes. TOCP exposure hampered the meiotic division process, as evidenced by a reduction in the extrusion of the first polar body from oocytes. Subsequent research revealed the disruption of the oocyte cell cytoskeleton induced by TOCP, resulting in abnormalities in spindle organization, chromosome alignment, and actin filament distribution. This disturbance further extended to the rearrangement of organelles within oocytes, particularly affecting the mitochondria. Importantly, after TOCP treatment, mitochondrial function in oocytes was impaired, leading to oxidative stress, DNA damage, cell apoptosis, and subsequent changes of epigenetic modifications. Supplementation with nicotinamide mononucleotide (NMN) alleviated the harmful effects of TOCP. NMN exerted its mitigating effects through two fundamental mechanisms. On one hand, NMN conferred stability to the cell cytoskeleton, thereby supporting nuclear maturation. On the other hand, NMN enhanced mitochondrial function within oocytes, reducing the excess reactive oxygen species (ROS), restoring meiotic division abnormalities caused by TOCP, preventing oocyte DNA damage, and suppressing epigenetic changes. These findings not only enhance our understanding of the molecular basis of TOCP induced oocyte damage but also offer a promising avenue for the potential application of NMN in optimizing reproductive treatment strategies.

Abstract Hyperglycemia-induced aberrant glucose metabolism is a causative factor of neurodegeneration and cognitive impairment in diabetes mellitus (DM) patients. The pyruvate dehydrogenase kinase (PDK)–lactic acid axis is regarded as a critical link between metabolic reprogramming and the pathogenic process of neurological disorders. However, its role in diabetic neuropathy remains unclear. Here, we found that PDK1 and phosphorylation of pyruvate dehydrogenase (PDH) were obviously increased in high glucose (HG)-stimulated primary neurons and Neuro-2a cell line. Acetyl-coA, a central metabolic intermediate, might enhance PDK1 expression via histone H3K9 acetylation modification in HG condition. The epigenetic regulation of PDK1 expression provided an available negative feedback pattern in response to HG environment-triggered mitochondrial metabolic overload. However, neuronal PDK1 was decreased in the hippocampus of streptozotocin (STZ)-induced diabetic mice. Our data showed that the expression of PDK1 also depended on the hypoxia-inducible factor-1 (HIF-1) transcriptional activation under the HG condition. However, HIF-1 was significantly reduced in the hippocampus of diabetic mice, which might explain the opposite expression of PDK1 in vivo. Importantly, overexpression of PDK1 reduced HG-induced reactive oxygen species (ROS) generation and neuronal apoptosis. Enhancing PDK1 expression in the hippocampus ameliorated STZ-induced cognitive impairment and neuronal degeneration in mice. Together, our study demonstrated that both acetyl-coA-induced histone acetylation and HIF-1 are necessary to direct PDK1 expression, and enhancing PDK1 may have a protective effect on cognitive recovery in diabetic mice. Abstract Recently it has been shown that the cosmological dynamics of covariant f(Q) gravity depend on different affine connections. In this paper, two specific f(Q) models are investigated with SNe+CC+BAO+QSO observational data, and the spatial curvature of the universe is studied in covariant f(Q) gravity. It is found that the parameters $${\mathcal {X}}_0$$ X 0 and $${\mathcal {X}}'_0$$ X 0 ′ characterizing affine connections significantly affect the behavior of the effective equation of state $$w_Q$$ w Q and may drive it across the phantom divide line. These results imply some inertial effects of the universe change the cosmic dynamics. However based on the Bayesian evidence, the zero inertial effect is more favored in the flat universe. Moreover, a closed universe is favored not only in the $$\Lambda $$ Λ CDM model but also in covariant f(Q) gravity. The f(Q) models have less support evidence than the $$\Lambda $$ Λ CDM model in the non-flat universe.

Abstract Pancreatic cancer, as one of the neoplasms with the highest degree of malignancy, has become a main disease of concerns in recent years. BHLHE40, a critical transcription factor for remodeling of the tumor immune microenvironment, has been described to be substantially increased in a variety of tumor-associated immune cells. Nevertheless, the pro-cancer biological functions and underlying molecular mechanisms of BHLHE40 for pancreatic cancer and its unique microenvironment are unclear. Hereby, we investigated the pro-oncogenic role of BHLHE40 in the pancreatic cancer microenvironment by bioinformatics analysis and cell biology experiments and determined that the expression of BHLHE40 was obviously elevated in pancreatic cancer tissues than in adjacent normal tissues. In parallel, Kaplan–Meier survival analysis unveiled that lower expression of BHLHE40 was strongly associated with better prognosis of patients. Receiver operating characteristic (ROC) curve analysis confirmed the accuracy of the BHLHE40-related prediction model. Subsequent, spearman correlation analysis observed that higher expression of BHLHE40 might be involved in immunosuppression of pancreatic cancer. Silencing of BHLHE40 could inhibit proliferation, invasion, and apoptosis of pancreatic cancer in vitro and in vivo, implying that BHLHE40 is expected to be a potential therapeutic target for pancreatic cancer. In addition, we explored and validated the FGD5-AS1/miR-15a-5p axis as a potential upstream regulatory mode for high expression of BHLHE40 in pancreatic cancer. In summary, our data showed that ceRNA involved in the regulation of BHLHE40 contributes to the promotion of immunosuppressive response in pancreatic and is expected to be a diagnostic marker and potential immunotherapeutic target for pancreatic cancer.

Nanoarchitectonics has emerged as a post-nanotechnology concept. As one of the applications of nanoarchitectonics, this review paper discusses the control of stem cell fate and function as an important issue. For hybrid nanoarchitectonics involving living cells, it is crucial to understand how biomaterials and their nanoarchitected structures regulate behaviours and fates of stem cells. In this review, biomaterials for the regulation of stem cell fate are firstly discussed. Besides multipotent differentiation, immunomodulation is an important biological function of mesenchymal stem cells (MSCs). MSCs can modulate immune cells to treat multiple immune- and inflammation-mediated diseases. The following sections summarize the recent advances of the regulation of the immunomodulatory functions of MSCs by biophysical signals. In the third part, we discussed how biomaterials direct the self-organization of pluripotent stem cells for organoid. Bioactive materials are constructed which mimic the biophysical cues of in vivo microenvironment such as elasticity, viscoelasticity, biodegradation, fluidity, topography, cell geometry, and etc. Stem cells interpret these biophysical cues by different cytoskeletal forces. The different cytoskeletal forces lead to substantial transcription and protein expression, which affect stem cell fate and function. Regulations of stem cells could not be utilized only for tissue repair and regenerative medicine but also potentially for production of advanced materials systems. Materials nanoarchitectonics with integration of stem cells and related biological substances would have high impacts in science and technology of advanced materials.

Abstract Background Succinate‐CoA ligase/synthetase (SCS) deficiency is responsible for encephalomyopathy with mitochondrial DNA depletion and mild methylmalonic aciduria. Variants in SUCLG1, the nuclear gene encoding the alpha subunit of the SCS enzyme playing a pivotal role in maintaining mtDNA integrity and stability, are associated with mitochondrial DNA depletion syndrome 9 (MTDPS9). Methods In this study, we reported an infant with clinical features of MTDPS9 from China. Whole exome sequencing (WES) was used to identify the genetic cause. Bioinformatic analysis and mtDNA level detection were performed to assess pathogenicity. Results The proband manifested with hypotonia, lactic acidosis, mild methylmalonic aciduria, hearing loss and psychomotor retardation. WES identified new compound heterozygous SUCLG1 variants of c.601A>G (p.R201G) in exon 6 and c.871G>C (p.A291P) in exon 8. Computational analysis predicted that these missense variants might alter structure stability and mitochondrial translocation of SUCLG1. qRT‐PCR showed 68% depletion of mtDNA content in proband as compared to controls. Conclusion Novel compound heterozygous variants c.601A>G (p.R201G) and c.871G>C (p.A291P) in SUCLG1 may cause MTDPS9 in this family. Our finding should be helpful for molecular diagnosis, genetic counseling and clinical management of SCS deficiency disorders.

All-dielectric metasurfaces have been widely used in nanophotonics because of their properties, such as negligible Ohmic loss, stable properties, and flexible fabrication techniques. We propose an all-dielectric metasurface consisting of subwavelength particles, which supports a “trapped” mode after the introduction of adequate symmetry-breaking in the silicon meta-atoms. Coating the metasurface with monolayer graphene enables the adsorption of nucleic acids such as DNA and RNA. We compare the resonant states in the transmission spectrum and find that the proposed metasurface is ultra-sensitive to the surrounding environment and exhibits excellent biosensing performance. Our conclusions indicate that all-dielectric metasurfaces can be a promising platform to realize ultra-sensitive bio-sensors.

As one of the most popular social media platforms in China, Weibo has aggregated huge numbers of texts containing people’s thoughts, feelings, and experiences. Analyzing emotions expressed on Weibo has attracted a great deal of academic attention. Emotion lexicon is a vital foundation of sentiment analysis, but the existing lexicons still have defects such as a limited variety of emotions, poor cross-scenario adaptability, and confusing written and online expressions and words. By combining grounded theory and semi-automatic methods, we built a Weibo-based emotion lexicon for sentiment analysis. We first took a bottom-up approach to derive a theoretical model for emotions expressed on Weibo, and the substantive coding led to eight core emotion categories: joy, expectation, love, anger, anxiety, disgust, sadness, and surprise. Second, we built a new emotion lexicon containing 2,964 words by manually selecting seed words, constructing a word vector model to expand words, and making rules to filter words. Finally, we tested the effectiveness of our lexicon by using a lexicon-based approach to recognize the emotions expressed in Weibo text. The results showed that our lexicon performed better in Weibo emotion recognition than five other Chinese emotion lexicons. This study proposed a method to construct an emotion lexicon that considered both theory and application by combining qualitative research and artificial intelligence methods. Our work also provided a reference for future research in the field of social media sentiment analysis.

Cell metabolite detection is important for cell analysis. As a cellular metabolite, lactate and its detection play an important role in disease diagnosis, drug screening and clinical therapeutics. This paper reports a microfluidic chip integrated with a backflow prevention channel for cell culture and lactate detection. It can effectively realize the upstream and downstream separation of the culture chamber and the detection zone, and prevent the pollution of cells caused by the potential backflow of reagent and buffer solutions. Due to such a separation, it is possible to analyze the lactate concentration in the flow process without contamination of cells. With the information of residence time distribution of the microchannel networks and the detected time signal in the detection chamber, it is possible to calculate the lactate concentration as a function of time using the de-convolution method. We have further demonstrated the suitability of this detection method by measuring lactate production in human umbilical vein endothelial cells (HUVEC). The microfluidic chip presented here shows good stability in metabolite quick detection and can work continuously for more than a few days. It sheds new insights into pollution-free and high-sensitivity cell metabolism detection, showing broad application prospects in cell analysis, drug screening and disease diagnosis.

Highly active Ru nanoparticles (Ru NPs) supported on NiFe layered double hydroxide (Ru/NiFe-LDH) are prepared easily using ultrasound-assisted reduction method without chemical reductants and stabilizers. The plentiful hydroxyls on NiFe-LDH are excited into hydrogen radicals (·H) under the action of ultrasound for reducing Ru3+ to Ru0. Ru NPs with an average particle size of 1.26 nm highly disperse on the mesopore-like surface of NiFe-LDH, which improve the catalytic performance for N-ethylcarbazole (NEC) hydrogenation. The experimental results show that 5Ru/NiFe-LDH-300–60 exhibits the best catalytic performance with 100% conversion of NEC, 98.88% yield of dodecahydro-N-ethylcarbazole (12H-NEC) and 5.77 wt% mass hydrogen storage capacity under the reaction conditions of 110 ℃, 6 MPa and mRu:mNEC = 0.15 wt% for 80 min. The kinetics study shows that the apparent activation energy is only 25.15 kJ/mol, which is the lowest in the reported literatures. Ru complexes with O-contained groups on NiFe-LDH, improving the catalytic stability in NEC hydrogenation.

In this paper, we discuss about the possibility to enhance the tensor-to-scalar ratio r under the condition of Trans-Planckian censorship conjecture (TCC), thus r∼O(10−3) could be observable within the sensitivity of future experiments. We make use of the scalar-tensor theory where inflaton is nonminimally coupled to gravity. After demonstrating that the TCC condition could be modified in scalar-tensor theory, we show that due to the effects of modified gravity at the end of inflation, a large r∼O(10−3) could be allowed without violating the TCC. Moreover, the modification can give rise to a weak coupling of gravity to the inflation field. If such an effect has been present as early as inflation starts, it would imply that in our case, the Universe might have experienced an asymptotically safe period at its early time.

The propagation of electromagnetic (EM) wave along a plasma filament induced by the sufficiently intense femtosecond laser pulse is studied with the commercial simulation software XFDTD. The simulated results show that the EM wave can propagate along the plasma filament, with the electric field enhanced out of the waveguide and attenuated in the waveguide. Besides, we can conclude that the relative direction between the plasma filament and the electric field can influence the guiding performance of the plasma filament. Furthermore, a maximum EM wave intensity enhancement more than threefold the free space propagation is realized. The research results imply that the laser-induced plasma filament can provide efficient transmission of the EM wave energy. Keywords: EM wave, Femtosecond laser, Plasma filament, Propagation, XFDTD

Participation in adult education is a crucial component of the active aging framework. However, there have been limited studies exploring the association between participation in adult education and subjective wellbeing among older adults. Furthermore, the underlying mediating and moderating mechanisms in this association have not been extensively investigated. This study aims to explore the mediating roles of physical and digital isolation, as well as the moderating roles of educational level. A sample of 7,602 older adults (M = 70.97, SD = 7.13) was drawn from the 2018 wave of the China Longitudinal Aging Social Survey. Structural equation modeling was employed to test the mediating and moderating roles. The results revealed that participation in adult education is positively associated with higher levels of life satisfaction and lower levels of loneliness among older adults. Both physical isolation and digital isolation play mediating roles in these associations. Moreover, educational level moderates the association between participation in adult education and older adults' life satisfaction and loneliness. The linkages are significant for older adults with lower levels of education, but not significant for those with higher levels of education. These findings contribute to current knowledge and offer practical implications for improving older adults' subjective wellbeing through participation in adult education.
