2. Endurance exercise-induced histone methylation modification involved in skeletal muscle fiber type transition and mitochondrial biogenesis

Abstract Skeletal muscle is a highly heterogeneous tissue, and its contractile proteins are composed of different isoforms, forming various types of muscle fiber, each of which has its own metabolic characteristics. It has been demonstrated that endurance exercise induces the transition of muscle fibers from fast-twitch to slow-twitch muscle fiber type. Herein, we discover a novel epigenetic mechanism for muscle contractile property tightly coupled to its metabolic capacity during muscle fiber type transition with exercise training. Our results show that an 8-week endurance exercise induces histone methylation remodeling of PGC-1α and myosin heavy chain (MHC) isoforms in the rat gastrocnemius muscle, accompanied by increased mitochondrial biogenesis and an elevated ratio of slow-twitch to fast-twitch fibers. Furthermore, to verify the roles of reactive oxygen species (ROS) and AMPK in exercise-regulated epigenetic modifications and muscle fiber type transitions, mouse C2C12 myotubes were used. It was shown that rotenone activates ROS/AMPK pathway and histone methylation enzymes, which then promote mitochondrial biogenesis and MHC slow isoform expression. Mitoquinone (MitoQ) partially blocking rotenone-treated model confirms the role of ROS in coupling mitochondrial biogenesis with muscle fiber type. In conclusion, endurance exercise couples mitochondrial biogenesis with MHC slow isoform by remodeling histone methylation, which in turn promotes the transition of fast-twitch to slow-twitch muscle fibers. The ROS/AMPK pathway may be involved in the regulation of histone methylation enzymes by endurance exercise.

3. Phase 1b study of first-line fuzuloparib combined with modified FOLFIRINOX followed by fuzuloparib maintenance monotherapy in pancreatic adenocarcinoma

Abstract Background Chemotherapy remains the standard first-line treatment for pancreatic adenocarcinoma, but with limited efficacy. We aimed to explore the feasibility of adding the PARP inhibitor fuzuloparib to mFOLFIRINOX in the locally advanced/metastatic (LA/M) setting. Methods This was the dose-escalation and -expansion, phase 1b portion of a phase 1b/2 study. Patients were given oral fuzuloparib at escalating doses starting at 30 mg twice daily (BID) plus intravenous mFOLFIRINOX q2w for 8–12 cycles, followed by maintenance fuzuloparib at 150 mg BID. Cohorts at the maximal tolerated dose (MTD) and lower dose of fuzuloparib were expanded. Primary endpoints were dose-limiting toxicity (DLT), MTD, and recommended phase 2 dose (RP2D). Results As of data cutoff on Jan 15, 2023, 39 patients were recruited. 12 patients were enrolled during dose escalation (30 mg [n = 4]; 60 mg [n = 6]; 100 mg [n = 2]). DLT occurred in 1 patient in 60 mg cohort and 1 patient in 100 mg cohort. 60 mg BID was determined to be the MTD, and then 60 and 30 mg cohorts were expanded to 22 and 15 patients, respectively. The most common grade ≥ 3 treatment-related adverse events were hematologic toxicities. Efficacy in 60 mg cohort seemed to be most favorable, with an objective response rate of 50.0% (95% CI, 26.0–74.0) and disease control rate of 94.4% (95% CI, 72.7–99.9). Conclusions First-line fuzuloparib plus mFOLFIRINOX followed by maintenance fuzuloparib was generally safe and showed encouraging anti-tumor activity in patients with LA/M pancreatic adenocarcinoma. The RP2D of fuzuloparib combination was 60 mg BID. Trial registration ClinicalTrials.gov, NCT04228601.

4. Metabolome-driven microbiome assembly determining the health of ginger crop (Zingiber officinale L. Roscoe) against rhizome rot

Abstract Background Plant-associated microorganisms can be found in various plant niches and collectively comprise the plant microbiome. The plant microbiome assemblages have been extensively studied, primarily in model species. However, a deep understanding of the microbiome assembly associated with plant health is still needed. Ginger rhizome rot has been variously attributed to multiple individual causal agents. Due to its global relevance, we used ginger and rhizome rot as a model to elucidate the metabolome-driven microbiome assembly associated with plant health. Results Our study thoroughly examined the biodiversity of soilborne and endophytic microbiota in healthy and diseased ginger plants, highlighting the impact of bacterial and fungal microbes on plant health and the specific metabolites contributing to a healthy microbial community. Metabarcoding allowed for an in-depth analysis of the associated microbial community. Dominant genera represented each microbial taxon at the niche level. According to linear discriminant analysis effect size, bacterial species belonging to Sphingomonas, Quadrisphaera, Methylobacterium-Methylorubrum, Bacillus, as well as the fungal genera Pseudaleuria, Lophotrichus, Pseudogymnoascus, Gymnoascus, Mortierella, and Eleutherascus were associated with plant health. Bacterial dysbiosis related to rhizome rot was due to the relative enrichment of Pectobacterium, Alcaligenes, Klebsiella, and Enterobacter. Similarly, an imbalance in the fungal community was caused by the enrichment of Gibellulopsis, Pyxidiophorales, and Plectosphaerella. Untargeted metabolomics analysis revealed several metabolites that drive microbiome assembly closely related to plant health in diverse microbial niches. At the same time, 6-({[3,4-dihydroxy-4-(hydroxymethyl)oxolan-2-yl]oxy}methyl)oxane-2,3,4,5-tetrol was present at the level of the entire healthy ginger plant. Lipids and lipid-like molecules were the most significant proportion of highly abundant metabolites associated with ginger plant health versus rhizome rot disease. Conclusions Our research significantly improves our understanding of metabolome-driven microbiome structure to address crop protection impacts. The microbiome assembly rather than a particular microbe’s occurrence drove ginger plant health. Most microbial species and metabolites have yet to be previously identified in ginger plants. The indigenous microbial communities and metabolites described can support future strategies to induce plant disease resistance. They provide a foundation for further exploring pathogens, biocontrol agents, and plant growth promoters associated with economically important crops. Video Abstract

This paper reviews the combustion characteristics, explosion mechanisms, and explosion prevention experiments of hybrids. Current research results on explosion prevention technology are organized, and the current challenges in experimental studies of hybrids explosions are summarized, providing references for future experimental studies.

6. Mining key genes associated with phosphorus deficiency through genome-wide identification and characterization of cucumber SPX family genes

Abstract Background Proteins harboring the SPX domain are crucial for the regulation of phosphate (Pi) homeostasis in plants. This study aimed to identify and analyze the entire SPX gene family within the cucumber genome. Results The cucumber genome encompassed 16 SPX domain-containing genes, which were distributed across six chromosomes and categorized into four distinct subfamilies: SPX, SPX-MFS, SPX-EXS and SPX-RING, based on their structure characteristics. Additionally, gene duplications and synteny analysis were conducted for CsSPXs, revealing that their promoter regions were enriched with a variety of hormone-responsive, biotic/abiotic stress and typical P1BS-related elements. Tissue expression profiling of CsSPX genes revealed that certain members were specifically expressed in particular organs, suggesting essential roles in cucumber growth and development. Under low Pi stress, CsSPX1 and CsSPX2 exhibited a particularly strong response to Pi starvation. It was observed that the cucumber cultivar Xintaimici displayed greater tolerance to low Pi compared to black-spined cucumber under low Pi stress conditions. Protein interaction networks for the 16 CsSPX proteins were predicted, and yeast two-hybrid assay revealed that CsPHR1 interacted with CsSPX2, CsSPX3, CsSPX4 and CsSPX5, implying their involvement in the Pi signaling pathway in conjunction with CsPHR1. Conclusion This research lays the foundation for further exploration of the function of the CsSPX genes in response to low Pi stress and for elucidating the underlying mechanism.

7. Social network analysis of intergovernmental relations and policy tools in China’s coastal reclamation management

As an effective means of sea development and utilization, high-intensity coastal reclamation activities lead to increasingly prominent ecological problems. The accurate implementation of policies is paramount in managing and controlling coastal reclamation. We reviewed China’s coastal reclamation management and control policies from 1978 to 2022. Utilizing content analysis and social network analysis, we constructed a policy network to explore the evolution of intergovernmental relations and the habitual combination of policy tools in coastal reclamation management and control. The study shows that (1) The intergovernmental relations of reclamation management and control agencies have evolved from simple to complex, with key departments becoming increasingly prominent; (2) Environmental considerations form the mainstay of policy tools for reclamation control, highlighting an imbalance between supply-side and demand-side approaches; (3) Since 1999, the interactive network between intergovernmental relations and policy tools has begun to exhibit a distinct core-periphery structure, and the social circle has gradually expanded to form a social circle consisting of 23 administrative departments and most departmental policy tools, while there are also some independent working factions at different stages; (4) Policy sustainability is currently inadequate, and in the later stages of management and control, there is a noticeable conflict within the policy texts. The study shows that, China’s coastal reclamation management and controlstill faces challenges, It is necessary to continue to explore the policy visualization methods used in coastal reclamation to help balance policy tools and build efficient intergovernmental relations.

8. Lateral/caudal ganglionic eminence makes limited contribution to cortical oligodendrocytes

The emergence of myelinating oligodendrocytes represents a pivotal developmental milestone in vertebrates, given their capacity to ensheath axons and facilitate the swift conduction of action potentials. It is widely accepted that cortical oligodendrocyte progenitor cells (OPCs) arise from medial ganglionic eminence (MGE), lateral/caudal ganglionic eminence (LGE/CGE), and cortical radial glial cells (RGCs). Here, we used two different fate mapping strategies to challenge the established notion that the LGE generates cortical OPCs. Furthermore, we used a Cre/loxP-dependent exclusion strategy to reveal that the LGE/CGE does not give rise to cortical OPCs. Additionally, we showed that specifically eliminating MGE-derived OPCs leads to a significant reduction of cortical OPCs. Together, our findings indicate that the LGE does not generate cortical OPCs, contrary to previous beliefs. These findings provide a new view of the developmental origins of cortical OPCs and a valuable foundation for future research on both normal development and oligodendrocyte-related disease.

9. Evolution of the SPX gene family and its role in the response mechanism to low phosphorus stress in self-rooted apple stock

Abstract Background Phosphorus plays a key role in plant adaptation to adversity and plays a positive role in the yield and quality formation of apples. Genes of the SPX domain-containing family are widely involved in the regulation of phosphorus signalling networks. However, the mechanisms controlling phosphorus deficiency are not completely understood in self-rooted apple stock. Results In this study, 26 members of the apple SPX gene family were identified by genome-wide analysis, and further divided into four subfamilies (SPX, SPX-MFS, SPX-EXS, and SPX-RING) based on their structural features. The chromosome distribution and gene duplications of MdSPXs were also examined. The promoter regions of MdSPXs were enriched for multiple biotic/abiotic stresses, hormone responses and typical P1BS-related elements. Analysis of the expression levels of 26 MdSPXs showed that some members were remarkably induced when subjected to low phosphate (Pi) stress, and in particular MdSPX2, MdSPX3, and MdPHO1.5 exhibited an intense response to low Pi stress. MdSPX2 and MdSPX3 showed significantly divergent expression levels in low Pi sensitive and insensitive apple species. Protein interaction networks were predicted for 26 MdSPX proteins. The interaction of MdPHR1 with MdSPX2, MdSPX3, MdSPX4, and MdSPX6 was demonstrated by yeast two-hybrid assay, suggesting that these proteins might be involved in the Pi-signaling pathway by interacting with MdPHR1. Conclusion This research improved the understanding of the apple SPX gene family and contribute to future biological studies of MdSPX genes in self-rooted apple stock.

10. Professional values education for undergraduate nursing students: developing a framework based on the professional values growth theory

Background Education has been recognised as necessary in forming and internalising professional values. The system and instructors' content in existing educational institutions focus on developing students' knowledge, skills and practices. Still, the development of values has yet to achieve significant effects, leading to a crisis in students' professional identity. Aims To construct a professional values growth theory for undergraduate nursing students and develop a corresponding education framework. Methods Through the review, some databases(PubMed、CINAHL、Web of Science、Wiley and Google Scholars)were searched using a systematic search strategy to collect relevant literature on professional values education. Based on the nursing professional values growth theory (Li and Li, Nursing Ethics In press, 2022), a theory of professional values growth of nursing undergraduates was developed using the method of theory derivation. Two rounds of expert meetings were conducted to review and revise an education framework of professional values of nursing undergraduates derived from that theory. Findings A total of 10 studies were included. The contents of two themes were analysed: theories and models and the current status of the professional values development of nursing students. The resulting professional values growth theory for undergraduate nursing students consists of five parts: key aspects, decisive opportunities, drivers, embodiment (humanistic sentiments, moral emotions), and outcomes. A total of five experts in the relevant fields were invited to this study. After two rounds of expert meetings, an education framework for undergraduate nursing students was finally developed, which consists of four parts: education objectives, education process and content, environment and conditions, and evaluation. Conclusion The education framework developed in this study has practical implications for the development of professional values of undergraduate nursing students, providing educational strategies and methods for the growth and internalisation of professional values of undergraduate nursing students.

11. Large area single crystal gold of single nanometer thickness for nanophotonics

Abstract Two-dimensional single crystal metals, in which the behavior of highly confined optical modes is intertwined with quantum phenomena, are highly sought after for next-generation technologies. Here, we report large area (>104 μm2), single crystal two-dimensional gold flakes (2DGFs) with thicknesses down to a single nanometer level, employing an atomic-level precision chemical etching approach. The decrease of the thickness down to such scales leads to the quantization of the electronic states, endowing 2DGFs with quantum-confinement-augmented optical nonlinearity, particularly leading to more than two orders of magnitude enhancement in harmonic generation compared with their thick polycrystalline counterparts. The nanometer-scale thickness and single crystal quality makes 2DGFs a promising platform for realizing plasmonic nanostructures with nanoscale optical confinement. This is demonstrated by patterning 2DGFs into nanoribbon arrays, exhibiting strongly confined near infrared plasmonic resonances with high quality factors. The developed 2DGFs provide an emerging platform for nanophotonic research and open up opportunities for applications in ultrathin plasmonic, optoelectronic and quantum devices.

12. Patterns of neural activity in response to threatening faces are predictive of autistic traits: modulatory effects of oxytocin receptor genotype

Abstract Autistic individuals generally demonstrate impaired emotion recognition but it is unclear whether effects are emotion-specific or influenced by oxytocin receptor (OXTR) genotype. Here we implemented a dimensional approach using an implicit emotion recognition task together with functional MRI in a large cohort of neurotypical adult participants (N = 255, male = 131, aged 17–29 years) to establish associations between autistic traits and neural and behavioral responses to specific face emotions, together with modulatory effects of OXTR genotype. A searchlight-based multivariate pattern analysis (MVPA) revealed an extensive network of frontal, basal ganglia, cingulate and limbic regions exhibiting significant predictability for autistic traits from patterns of responses to angry relative to neutral expression faces. Functional connectivity analyses revealed a genotype interaction (OXTR SNPs rs2254298, rs2268491) for coupling between the orbitofrontal cortex and mid-cingulate during angry expression processing, with a negative association between coupling and autistic traits in the risk-allele group and a positive one in the non-risk allele group. Overall, results indicate extensive emotion-specific associations primarily between patterns of neural responses to angry faces and autistic traits in regions processing motivation, reward and salience but not in early visual processing. Functional connections between these identified regions were not only associated with autistic traits but also influenced by OXTR genotype. Thus, altered patterns of neural responses to threatening faces may be a potential biomarker for autistic symptoms although modulatory influences of OXTR genotype need to be taken into account.

13. Operative treatment of pulmonary primitive neuroectodermal tumor: a case report and literature review

Abstract Background Pulmonary primitive neuroectodermal tumor (PNET), a member of the Ewing sarcoma family of tumors, is a rare malignancy that is associated with a grim prognosis. To date, fewer than 30 cases of pulmonary PNET have been reported. In this case report, we present the clinical details of a 12-year-old girl with pulmonary PNET who underwent surgical treatment. We also conducted an analysis and summary of other relevant studies and the surgical outcomes. Case presentation In May 2018, a 12-year-old girl was admitted with symptoms of cough and blood-tinged phlegm. A computed tomography scan revealed a large mass, measuring 12.9 cm × 8.1 cm, in the right middle and lower lungs. A percutaneous lung biopsy confirmed poorly differentiated tumor cells with a nested growth pattern. Immunohistochemical staining demonstrated positive expression of CD99, CD56, Vimentin, and Synaptophysin. The patient was diagnosed with pulmonary PNET. Following three cycles of neoadjuvant chemotherapy, a substantial reduction in tumor volume was observed. Subsequently, the patient underwent a surgical procedure involving pneumonectomy and partial resection of the left atrium with the assistance of cardiopulmonary bypass. The patient was discharged 37 days after surgery. During a three-year follow-up period, she exhibited no signs of tumor recurrence and has successfully returned to school. Conclusions This case highlights the successful management of an advanced PNET with neoadjuvant chemotherapy, pneumonectomy, and partial resection of the left atrium employing cardiopulmonary bypass. The patient remained disease-free after three years. Our analysis of surgically treated cases indicates that neoadjuvant chemotherapy can contribute to improved prognoses for PNET patients. It is crucial to emphasize that complete surgical excision remains the cornerstone of treatment, underscoring the importance of surgeons considering radical surgical approaches whenever feasible for patients with pulmonary PNETs.

14. Synthesizing Landsat images using time series model-fitting methods for China’s coastal areas against sparse and irregular observations

Long historical records and free accessibility have made Landsat data valuable for time-series analysis. However, Landsat time-series analysis is restricted for coastal areas due to the lack of sufficient numbers of clear images. The generation of synthetic Landsat images using model-fitting methods is accepted as an effective means of overcoming this problem. But for coastal areas, available observations are typically sparse and irregular, leading to ill-fitted models that distort synthetic Landsat images. In this study, we propose a linear harmonic model with different orders and implement it on an annual basis to generate synthetic Landsat images based on the Google Earth Engine (GEE) platform. First, we incorporated the available observations from adjacent years and filtered them by season to address the problems of data sparsity and irregularity. Then, we combined the threshold segmentation and ridge regression approaches to address the fake cloud problem, which sometimes contaminates synthetic Landsat images in summer. All cloud-free real Landsat images from five typical sites along China’s coasts during 1986–2020 were used to evaluate the accuracy and robustness of the synthetic Landsat images generated using our proposed methods. The principal results are as follows: (1) the R2 value of the linear harmonic model averaged across different Landsat bands and land cover types was 0.640, and the model was especially successful in simulating the surface reflectance in near NIR bands in forest and grassland areas; (2) 81.1% of the synthetic Landsat images covered by fake clouds were effectively restored, and there was the particular need to remove fake clouds for the synthetic Landsat images at low latitudes; (3) The mean absolute error for our synthetic Landsat images was 0.015, with the rate of 0.124; this was achieved under the clear-sky probability of only 36% and the average number of annual observations below 9, indicating a good performance. Compared with the continuous change detection and classification (CCDC) and seasonal median composite (SMC) methods, our proposed method offers advantages in both the accuracy and integrity of synthetic Landsat images. Our proposed method for synthesizing images also has potential for application to global coastal areas and other satellite datasets.

15. Gut microbiota-derived trimethylamine N-Oxide: a novel target for the treatment of preeclampsia

ABSTRACTPre-eclampsia (PE) is the most common complication of pregnancy and seriously threatens the health and safety of the mother and child. Studies have shown that an imbalance in gut microbiota can affect the progression of PE. Trimethylamine N-oxide (TMAO) is an intestinal microbiota-derived metabolite that is thought to be involved in the occurrence of PE; however, its causal relationship and mechanism remain unclear. In this clinical cohort study, including 28 patients with eclampsia and 39 matched healthy controls, fecal samples were collected for 16S rRNA gene sequencing, and serum was collected for targeted metabolomics research. The results showed that the level of TMAO and the abundance of its source bacteria had significantly increased in patients with PE, and were positively correlated with the clinical progression of PE. Fecal microbiota transplantation (FMT) was applied to an antibiotic-depleted-treated mouse model and targeted inhibition of TMAO. The results of the FMT experiment revealed that mice that received fecal microbiota transplantation from patients with PE developed typical PE symptoms and increased oxidative stress and inflammatory damage, both of which were reversed by 3,3-Dimethyl-1-butanol (DMB), a TMAO inhibitor, which also improved pregnancy outcomes in the model mice. Similar results were obtained in the classical NG-Nitroarginine methyl ester (L-NAME) induced PE mouse model. Mechanistically, TMAO promotes the progression of PE by regulating inflammatory and oxidative stress-related signaling pathways, affecting the migration and angiogenesis of vascular endothelial cells, as well as the migration and invasion of trophoblast cells. Our results reveal the role and mechanism of gut microbiota and TMAO in the progression of PE, provides new ideas for exploring the pathogenesis and therapeutic targets of PE, and determines the potential application value of TMAO as a target for PE intervention.

IntroductionEnhancing road safety conditions alleviates socioeconomic hazards from traffic accidents and promotes public health. Monitoring progress and recalibrating measures are indispensable in this effort. A systematic and scientific decision-making model that can achieve defensible decision outputs with substantial reliability and stability is essential, particularly for road safety system analyses.MethodsWe developed a systematic methodology combining the entropy weight method (EWM), preference ranking organization method for enrichment evaluation (PROMETHEE), and density-based spatial clustering of applications with noise (DBSCAN)—referred to as EWM–PROMETHEE II–DBSCAN—to support road safety monitoring, recalibrating measures, and action planning. Notably, we enhanced DBSCAN with a machine learning algorithm (grid search) to determine the optimal parameters of neighborhood radius and minimum number of points, significantly impacting clustering quality.ResultsIn a real case study assessing road safety in Southeast Asia, the multi-level comparisons validate the robustness of the proposed model, demonstrating its effectiveness in road safety decision-making. The integration of a machine learning tool (grid search) with the traditional DBSCAN clustering technique forms a robust framework, improving data analysis in complex environments. This framework addresses DBSCAN’s limitations in nearest neighbor search and parameter selection, yielding more reliable decision outcomes, especially in small sample scenarios. The empirical results provide detailed insights into road safety performance and potential areas for improvement within Southeast Asia.ConclusionThe proposed methodology offers governmental officials and managers a credible tool for monitoring overall road safety conditions. Furthermore, it enables policymakers and legislators to identify strengths and drawbacks and formulate defensible policies and strategies to optimize regional road safety.

BackgroundChronic inflammation is implicated in the development of diabetic retinopathy (DR). The neutrophil-to-lymphocyte ratio (NLR) is a marker of systemic inflammation that has been linked to cardiovascular and diabetic kidney diseases. However, the link between NLR and DR remains unclear. As such, this study investigated the association between NLR and DR in Chinese patients.MethodA total of 857 adults diagnosed with type 2 diabetes mellitus (T2DM) without DR at baseline between 2018 and 2021, from a single center in Ningbo, China, were included. Baseline clinical data, including age, sex, T2DM duration, hypertension, smoking, drinking, glycated hemoglobin level, lipid profile, renal function, and NLR, were recorded and analyzed. Cox proportional hazard regression analysis was used to assess the association between NLR and the risk for incident DR.ResultsDuring a median follow-up of 3.0 years, 140 patients developed DR. The multivariable-adjusted hazard ratio (HR) for incident DR across ascending NLR quartiles (≤1.46 [reference], 1.47–1.90, 1.91–2.45 and > 2.45) were 1.000, 1.327 (95% confidence interval [CI] 0.754–2.334), 1.555 (95% CI 0.913–2.648) and 2.217 (95% CI 1.348–3.649), respectively. For each 1-standard deviation increase in NLR, the risk for DR increased by 29.2% (HR 1.292 [95% CI 1.112–1.501) after adjusting for confounding factors.ConclusionResults revealed that a higher NLR at baseline was associated with an increased risk for incident DR. NLR has the potential to be an inexpensive, reliable, and valuable clinical measure that merits further exploration in future studies.

As a high-quality fish with high protein and unsaturated fatty acids, Siniperca chuatsi is an important aquaculture species in Guangdong, with a stable aquaculture production that ranks first in the whole country, and the production in 2022 was 148 700 t, accounting for 37.05% of the total output of S. chuatsi in the country. With the increasing demand for high-quality aquatic products in domestic market, S. chuatsi is increasingly favored by domestic consumers. China's S. chuatsi production has grew steadily from 2018 to 2022, with Guangdong, Hubei and Anhui ranking the top three in terms of S. chuatsi production, and the degree of industry aggregation will further increase. With the overall production of S. chuatsi in Guangdong rising on a steady trend, the domestic market share of S. chuatsi has been gradually occupied by Guangdong. Compared with China's major freshwater aquaculture fish, the market wholesale price of S. chuatsi has a greater advantage. As a key breeding area of S. chuatsi industry in China, and the largest production area of S. chuatsi seedlings in the country, the major breeding species of S. chuatsi in Guangdong is S. chuatsi (Basilewsky), and the main production areas of the breeding are located in Foshan, Zhaoqing, Guangzhou, Qingyuan, and Jiangmen, etc. The major breeding modes are pond breeding, cage breeding, water tank breeding and factory feed breeding, with the mainly breeding way of feeding on live bait fish. There is gradual expansion of S. chuatsi aquaculture and market scale in Guangdong Province with future development trend of large-scale breeding. Taking the market price of S. chuatsi in 2022 as a reference, a cost-benefit analysis was conducted for the S. chuatsi cultured with live bait and feed.The results showed that the average total cost of feed S. chuatsi was much lower than that of live bait S. chuatsi, with a large profit margin. In regard to the deficiencies of seedling, breeding environment and feed-based mode of S. chuatsi breeding industry in Guangdong, countermeasures such as strengthening the S. chuatsi seed system, promoting the development of feed S. chuatsi stably, further building a quality and safety system for S. chuatsi, and actively pushing the transformation of S. chuatsi breeding mode were proposed to provide references for the development of S. chuatsi industry in Guangdong.

Sparse view computed tomography (SVCT) aims to reduce the number of X-ray projection views required for reconstructing the cross-sectional image of an object. While SVCT significantly reduces X-ray radiation dose and speeds up scanning, insufficient projection data give rise to issues such as severe streak artifacts and blurring in reconstructed images, thereby impacting the diagnostic accuracy of CT detection. To address this challenge, a dual-domain reconstruction network incorporating multi-level wavelet transform and recurrent convolution is proposed in this paper. The dual-domain network is composed of a sinogram domain network (SDN) and an image domain network (IDN). Multi-level wavelet transform is employed in both IDN and SDN to decompose sinograms and CT images into distinct frequency components, which are then processed through separate network branches to recover detailed information within their respective frequency bands. To capture global textures, artifacts, and shallow features in sinograms and CT images, a recurrent convolution unit (RCU) based on convolutional long and short-term memory (Conv-LSTM) is designed, which can model their long-range dependencies through recurrent calculation. Additionally, a self-attention-based multi-level frequency feature normalization fusion (MFNF) block is proposed to assist in recovering high-frequency components by aggregating low-frequency components. Finally, an edge loss function based on the Laplacian of Gaussian (LoG) is designed as the regularization term for enhancing the recovery of high-frequency edge structures. The experimental results demonstrate the effectiveness of our approach in reducing artifacts and enhancing the reconstruction of intricate structural details across various sparse views and noise levels. Our method excels in both performance and robustness, as evidenced by its superior outcomes in numerous qualitative and quantitative assessments, surpassing contemporary state-of-the-art CNNs or Transformer-based reconstruction methods.

Abstract Background Mycoplasma pneumoniae (MP) is one of the most common causes of community-acquired pneumonia in children. Most children have fever. In 2021, we found that the proportion of children without fever increased. The aim of this study is to summarize the differences in the clinical characteristics of children with MP pneumonia who are febrile or not, and to raise awareness of children who are not febrile. Method Demographic information of the children was collected on admission. Clinical manifestations during the course of the disease and the first laboratory, imaging, and pulmonary function tests before discharge were recorded and compared. Results From August to December, a total of 542 people were included in the study. We found that older children were more likely to have fever. Inflammatory indicators including procalcitonin (P = 0.030), C-reaction protein (P 0.05), especially the small airway function impairment, are no lower in afebrile children than in febrile children. Conclusion The fever rate is lower in younger children, but wheezing is more common. In afebrile children, the impairment of organ and lung function was no less than in febrile children. Therefore, attention should also be paid to children who are not febrile.

Abstract Acute kidney injury (AKI) constitutes a prevalent clinical syndrome characterized by elevated morbidity and mortality rates, emerging as a significant public health issue. This study investigates the interplay between endoplasmic reticulum (ER) stress, unfolded protein response (UPR), and ER-associated degradation (ER-phagy) in the pathogenesis of AKI. We employed four distinct murine models of AKI—induced by contrast media, ischemia–reperfusion injury, cisplatin, and folic acid—to elucidate the relationship between ER-phagy, ER stress, and apoptosis. Our findings reveal a marked decrease in ER-phagy coinciding with an accumulation of damaged ER, elevated ER stress, and increased apoptosis across all AKI models. Importantly, overexpression of DDRGK1 in HK-2 cells enhanced ER-phagy levels, ameliorating contrast-induced ER stress and apoptosis. These findings unveil a novel protective mechanism in AKI, wherein DDRGK1–UFL1-mediated ER-phagy mitigates ER stress and apoptosis in renal tubular epithelial cells. Our results thereby contribute to understanding the molecular underpinnings of AKI and offer potential therapeutic targets for its treatment.

AbstractContext The antidiabetic effects of flavonoids have been reported, but it is still unclear whether 5,7,3′,4′,5′-pentamethoxyflavone, isolated from Bauhinia championii Benth. (Fabaceae), also exhibits such properties.Objective To isolate 5,7,3′,4′,5′-pentamethoxyflavone from B. championii using high-speed countercurrent chromatography and examine its potential in treating diabetic nephropathy.Materials and methods The phytochemical constituents from the stems of B. championii were separated and purified with high-speed countercurrent chromatography; 5,7,3′,4′,5′-pentamethoxyflavone (PMF) was identified by mass spectrum, 1H-NMR, and 13C-NMR. After exposing mesangial cells to 30 mM glucose and either 5 μM or 10 μM PMF for 6 h, the levels of fibronectin (FN) and p-Smad2/3 were analyzed using Western blotting. Male Sprague–Dawley rats were injected intraperitoneally with 55 mg/kg streptozotocin to induce diabetes and then were randomized into three groups (n = 10): vehicle administration, low-dose (5 mg/kg) PMF, and high-dose (25 mg/kg) PMF by intragastric gavage for 3 months. A healthy group was included as the control.Results Compared to the diabetic group, low-dose and high-dose PMF treatment decreased the phosphorylation of Smad2/3 by 0.54- and 0.52-fold, and the accumulation of FN decreased by 0.82- and 0.77-fold in vitro; the phosphorylation of Smad2/3 was decreased by 0.39- and 0.37-fold, and the accumulation of FN decreased by 0.47- and 0.40-fold in vivo, respectively. Furthermore, PMF alleviated the glomerular basement membrane thickness and foot process fusion.Conclusion The findings suggest for the first time that PMF may be a promising treatment option for diabetic kidney fibrosis, which warrants additional clinical investigation.

23. c-di-GMP inhibits the DNA binding activity of H-NS in Salmonella

Abstract Cyclic di-GMP (c-di-GMP) is a second messenger that transduces extracellular stimuli into cellular responses and regulates various biological processes in bacteria. H-NS is a global regulatory protein that represses expression of many genes, but how H-NS activity is modulated by environmental signals remains largely unclear. Here, we show that high intracellular c-di-GMP levels, induced by environmental cues, relieve H-NS-mediated transcriptional silencing in Salmonella enterica serovar Typhimurium. We find that c-di-GMP binds to the H-NS protein to inhibit its binding to DNA, thus derepressing genes silenced by H-NS. However, c-di-GMP is unable to displace H-NS from DNA. In addition, a K107A mutation in H-NS abolishes response to c-di-GMP but leaves its DNA binding activity unaffected in vivo. Our results thus suggest a mechanism by which H-NS acts as an environment-sensing regulator in Gram-negative bacteria.

24. Mendelian randomization analyses explore the relationship between cathepsins and lung cancer

Abstract Lung cancer, a major contributor to cancer-related fatalities worldwide, involves a complex pathogenesis. Cathepsins, lysosomal cysteine proteases, play roles in various physiological and pathological processes, including tumorigenesis. Observational studies have suggested an association between cathepsins and lung cancer. However, the causal link between the cathepsin family and lung cancer remains undetermined. This study employed Mendelian randomization analyses to investigate this causal association. The univariable Mendelian randomization analysis results indicate that elevated cathepsin H levels increase the overall risk of lung cancer, adenocarcinoma, and lung cancer among smokers. Conversely, reverse Mendelian randomization analyses suggest that squamous carcinoma may lead to increased cathepsin B levels. A multivariable analysis using nine cathepsins as covariates reveals that elevated cathepsin H levels lead to an increased overall risk of lung cancer, adenocarcinoma, and lung cancer in smokers. In conclusion, cathepsin H may serve as a marker for lung cancer, potentially inspiring directions in lung cancer diagnosis and treatment.

Abstract Cholesterol is an essential structural component of membranes that contributes to membrane integrity and fluidity. Cholesterol homeostasis plays a critical role in the maintenance of cellular activities. Recently, increasing evidence has indicated that cholesterol is a major determinant by modulating cell signaling events governing the hallmarks of cancer. Numerous studies have shown the functional significance of cholesterol metabolism in tumorigenesis, cancer progression and metastasis through its regulatory effects on the immune response, ferroptosis, autophagy, cell stemness, and the DNA damage response. Here, we summarize recent literature describing cholesterol metabolism in cancer cells, including the cholesterol metabolism pathways and the mutual regulatory mechanisms involved in cancer progression and cholesterol metabolism. We also discuss various drugs targeting cholesterol metabolism to suggest new strategies for cancer treatment.

27. Characteristics of PM2.5 and CO2 Concentrations in Typical Functional Areas of a University Campus in Beijing Based on Low-Cost Sensor Monitoring

The air quality in educational campuses affects the health and work efficiency of teachers and students. Studies into this matter are of great significance for optimizing the management of campus living environments. Low-cost online sensors to monitor PM2.5 and CO2 levels were used in typical functional areas of a university campus in Beijing, China, including offices, dormitories, leisure spaces, canteens, and laboratories. By comparing the findings with data from nearby national monitoring stations, the seasonal and spatial variations in PM2.5 and CO2 concentrations were analyzed. Findings indicate PM2.5 levels within the campus were notably lower compared to the surrounding urban environment. There was variation in PM2.5 and CO2 concentrations across different functional areas. Typically, indoor PM2.5 levels were lower than outdoor ones, while CO2 concentrations in enclosed indoor spaces with human activities progressively escalated. The main internal emission sources affecting the PM2.5 level on campus included traffic emissions, dust generated by human activities, and emissions from catering. In contrast, in areas with better green coverage or where a lake system participates in the atmospheric circulation, the PM2.5 level and CO2/PM2.5 were lower. This indicates that the cleansing impact of plants and aquatic systems is instrumental in lowering PM2.5 concentrations, offering healthier leisure spaces. Seasonal variations also impact PM2.5 levels. During the non-heating period, less pollution source emissions led to decreased outdoor PM2.5 concentrations. The campus monitoring sites experienced an approximate 5 µg/m3 and 29 µg/m3 reduction in the average PM2.5 levels as compared to the PM2.5 of the surrounding urban environment, respectively, during the non-heating and heating period. During indoor activities or sleep, CO2 levels can build up to as high as 2303 ppm due to breathing. It is advisable to stay indoors on days when pollution levels are high, whereas on days with clean air, it is healthier to be outdoors or to air out indoor areas by opening windows. Our research provides clearer scientific evidence for incorporating behavioral strategies for improving air quality into both daily work and life. Moreover, the findings are quite meaningful for the widespread adoption of low-cost sensor monitoring in various environments, with applications beyond just the campus settings.

Selenium is a vital trace mineral that is crucial for maintaining regular biological processes in aquatic animals. In this study, a four-week dietary trial was carried out to assess the impact of bio-fermented selenium (Bio−Se) on the growth and immune response of Chinese mitten crabs, Eriocheir sinensis. The crabs were randomly allocated to five dietary treatment groups, each receiving a different dose of Bio−Se. The doses included 0, 0.3, 0.6, 1.5, and 3.0 mg/kg and were accurately measured in basal diet formulations. The results showed the weight gain rate (WGR), specific growth rate (SGR), and survival rate (SR) in the 1.5 mg/kg Bio−Se group were the highest, and 3.0 mg/kg of Bio−Se has an inhibitory effect on the WGR, SGR, and SR. The activities of the immune enzymes, including glutathione peroxidase (GPX), superoxide dismutase (SOD), and acid phosphatase (ACP), of the hepatopancreas were significantly (p < 0.05) increased in the 1.5 mg/kg Bio−Se group, while they decreased (p < 0.05) in the 3.0 mg/kg feeding group compared to the 0 mg/kg feeding group. The concentration of maleic dialdehyde (MDA) exhibited the opposite pattern. Similarly, the mRNA expression levels of antimicrobial peptides (ALF-1, Crus-1, and LYS), ERK, and Relish genes were also observed to be the highest in the 1.5 mg/kg Bio−Se group compared with the other groups. Furthermore, the administration of 1.5 mg/kg of Bio−Se resulted in an increase in the thickness of the intestinal plica and mucosal layer, as well as in alterations in the intestinal microbial profile and bacterial diversity compared to the dose of 0 mg/kg of Bio−Se. Notably, the population of the beneficial bacterial phylum Fusobacteria was increased after crabs were fed the 1.5 mg/kg Bio−Se diet. In conclusion, the oral administration of 1.5 mg/kg of Bio−Se improved the growth efficiency, antioxidant capabilities, immunity, and intestinal health of E. sinensis. Through a broken-line analysis of the WGR against dietary Bio−Se levels, optimal dietary Bio−Se levels were determined to be 1.1 mg/kg. These findings contribute valuable insights to the understanding of crab cultivation and nutrition.

Abstract Background Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy. Radical surgical resection offers the only potential cure. There is increasing agreement that radical antegrade modular pancreatosplenectomy (RAMPS) may benefit patients with tumors in the body and tail of the pancreas. To address this, the Chinese Study Group for Pancreatic Cancer (CSPAC)-3 trial is proposed to compare the effect of RAMPS and standard retrograde pancreatosplenectomy (SRPS) on patient survival and preoperative safety Methods The randomized controlled trial will be multicenter and two-armed with blinded outcomes and intention-to-treat analysis. Three hundred patients with resectable body and tail pancreatic adenocarcinoma will be enrolled and randomly assigned to RAMPS or SRPS. Adjuvant chemotherapy based on an initial regimen will be recommended 4–6 weeks after surgery if no serious complication occurs. The hypothesis that RAMPS improves survival outcomes compared with SRPS will be tested using a superiority trial. The primary outcome will be overall survival (OS). Secondary outcomes will include recurrence-free survival (RFS), R0 resection rate, the number of harvested lymph nodes, postoperative complications, and quality of life scores. Discussion The use of RAMPS has increased over the past decade. It is reported that RAMPS is superior to SRPS in improving both the rate of R0 resection and lymph node yield. Despite these advantages, however, there is little high-level documentation of the superiority of RAMPS in terms of survival and this needs to be investigated. To address this issue, CSPAC has instigated the first prospective, randomized phase III control trials, aiming to explore the optimal surgical strategy for improving the prognosis and OS of patients with left-sided pancreatic cancer Trial registration Chinese Clinical Trial Registry ChiCTR2100053844; pre-results. Objective: To explore the effect and mechanism of action of the Wenjing Zhitong recipe (WZR) in primary dysmenorrhea (PD) treatment. Methods: Uterine contractions were induced by estradiol benzoate and oxytocin in a PD model and WZR was administrated. The rate of change in uterine contractility and the writhing test were used to evaluate the effects of WZR. The serum levels of prostaglandin F2α (PGF2α) and prostaglandin E2 (PGE2), and the activity of cyclooxygenase-2 (COX2) were detected by enzyme-linked immunosorbent assay (ELISA). The changes in phosphor-phospholipase C (pPLC/PLC), phosphor-protein kinase C (pPKC/PKC), and connexin 43 (CX43) expression were detected using immunohistochemistry and western blot. Results: WZR significantly reduced the rate of change in uterine contractility and writhing times in the PD model. WZR treatment inhibited the enzymatic activity of COX2 and reduced the levels of PGF2α, PGF2α/PGE2 and COX2 in the PD model. WZR also significantly reduced the expression of pPLC/PLC, pPKC/PKC and CX43. Targeting the inhibition of COX2 activity, caffeic acid and 1-acetyl-β-carboline were validated as the active ingredients in WZR responsible for reducing uterine contractions. Conclusion: WZR attenuated PD by inhibiting COX2 activity, downregulating PGF2α/PGE2 expression, and inhibiting the PKC signaling pathway.

Abstract Narrow bandgap materials have garnered significant attention within the field of broadband photodetection. However, the performance is impeded by diminished absorption near the bandgap, resulting in a rapid decline in photoresponsivity within the mid‐wave infrared (MWIR) and long‐wave infrared (LWIR) regions. Furthermore, they mostly worked in cryogenic temperature. Here, without the assistance of any complex structure and special environment, it is realized high responsivity covering ultra‐broadband wavelength range (Ultraviolet (UV) to LWIR) in a single quasi‐1D pseudogap (PG) system (TaSe4)2I nanoribbon, especially high responsivity (From 23.9 to 8.31 A W−1) within MWIR and LWIR region at room temperature (RT). Through direct probing the carrier relaxation process with broadband time‐resolved transient absorption spectrum measurement, the underlying mechanism of majorly photoconductive effect is revealed, which causes an increased spectral weight extended to PG region. This work paves the way for realizing high‐performance uncooled MWIR and LWIR detection by using quasi‐1D PG materials.

The research on the mechanism and protective measures of MIC is becoming more and more important. In this paper, combined with the unique environment of the circulating water cooling system, the influence of the unique environmental factors of the system on MIC is analyzed. Several kinds of bacteria known to cause microbial corrosion are summarized, including sulfate-reducing bacteria, nitrate-reducing bacteria, iron oxidizing bacteria, etc. The current MIC mechanisms and related research progress are reviewed. Several common MIC monitoring methods are summarized. The commonly used protection methods and the ideas of MIC technologies in the recent stage are listed. Finally, the future research direction of the MIC is prospected.

Lung adenocarcinoma (LUAD) has a poor prognosis. Circadian genes such as TIMELESS have been associated with several pathologies, including cancer. The expression of TIMELESS and the relationship between TIMELESS, infiltration of tumors and prognosis in LUAD requires further investigation. In this study, we investigated the expression of TIMELESS and its association with survival across several types of human cancer using data from The Cancer Genome Atlas (TCGA) and the Genotype-Tissue Expression Program. Noncoding RNAs (ncRNAs) regulating overexpression of TIMELESS in lung adenocarcinoma (LUAD) were explored with expression, correlation, and survival analyses. Immune cell infiltration and biomarkers were analyzed between different TIMELESS expression levels. The relationship between TIMELESS expression and immunophenoscores, which were used to predict response to immunotherapy, was evaluated. TIMELESS was identified as a potential oncogene in LUAD. NcRNA analysis showed MIR4435-2HG/hsa-miR-1-3p may interact with TIMELESS in a competitive endogenous RNA network in LUAD tumor tissues. Most immune cells were significantly decreased in TCGA LUAD tumor tissues with high TIMELESS expression except for CD4+T cells and Th2 cells. TIMELESS expression in LUAD tumor tissues was significantly negatively correlated with neutrophil biomarkers, dendritic cell biomarkers (HLA-DPB1, HLA-DQB1, HLA-DRA, HLA-DPA1, CD1C) and an immunophenoscore that predicted outcomes associated with the use of immune checkpoint inhibitors. These findings imply that ncRNAs-mediated TIMELESS overexpression in LUAD tumor tissues correlated with poor prognosis, reduced immune cell infiltration in the tumor microenvironment, and poor response to immune checkpoint inhibitors.

36. Mariculture may intensify eutrophication but lower N/P ratios: a case study based on nutrients and dual nitrate isotope measurements in Sansha Bay, southeastern China

The mariculture industry has grown rapidly worldwide over the past few decades. The industry helps meet growing food demands and may provide an effective means of carbon sequestration; however, it may harm the marine ecological environment, and the extent of its impact depends on the type of mariculture. Here we focus on the impact of mariculture on the nutrient status and eutrophication in Sansha Bay, which is a typical aquaculture harbor in southeastern China that employs a combination of shellfish and seaweed farming. Nutrient concentrations and dual nitrate isotopes were measured in Sansha Bay during the winter of 2021. The average concentrations of nitrate and phosphate were 31.3 ± 10.5 and 2.26 ± 0.84 µM, respectively, indicating that the water was in a eutrophic state. However, the N/P ratios were relatively low (14.3 ± 2.2). Nitrate isotope measurements were 8.8‰–11.9‰ for δ15N-NO3− and 2.2‰–6.0‰ for δ18O-NO3−. Source analysis based on the nitrate isotope measurements indicates that nitrate in Sansha Bay is derived mainly from the excretion of organisms and sewage discharge from mariculture. The isotopic fractionation model of nitrate assimilation by organisms indicates that surface waters in Sansha Bay experience strong biological uptake of nitrate, which is likely related to seaweed farming in winter. The low N/P ratios may be attributed to excessive nitrogen uptake (relative to phosphorus) during shellfish and seaweed farming, as well as nitrogen removal through sediment denitrification, which is fueled by the sinking of particulate organic matter from mariculture. Overall, our study shows that mariculture activities dominated by shellfish and seaweed cultivation in Sansha Bay may exacerbate eutrophication but reduce N/P ratios in the water column in aquaculture areas.

As China's resource shortage and environmental pollution intensify, the demand for new energy and electric energy substitution is becoming higher and higher. Accurately predicting the investment scale of China's new energy projects is of great practical significance for improving the efficiency of resource allocation and economically meeting energy demand. This paper builds a scientific and precise investment model for new energy projects from both macro and micro perspectives. First, from a macro perspective, considering macro indicators such as the external environment and internal economy, an annual total investment forecast model based on multiple linear regression is constructed, in order to predict the annual total investment scale of new energy investment entities and achieve preliminary accurate investment; second, designed the evaluation index system of different project priorities from three perspectives of external environment, internal development of enterprises and social development, and constructed the comprehensive weight design method based on AN-EWM and the comprehensive evaluation method of TOPSIS, in order to realize the priority of differentiated projects. Sorting; finally, a new energy project located in a city in northern China is selected as the research subject, and a multi-scenario example analysis is carried out. The results show that the new energy project investment scale index system constructed in this paper can effectively evaluate the investment capacity of the main body of the new energy project, and can better predict the total investment of the new energy investment project, so that the deviation rate can be controlled within 5 %, and the priority evaluation model constructed in this paper can provide a complete calculation method and a reference method for the judgement of the investment priority, which can promote accurate investment.

38. Factors associated with nonattendance at annual diabetes check-up in Ningbo, China: a case-control study

BackgroundType 2 diabetes mellitus (T2DM) is a grave issue in China. The annual check-up is recommended in clinical guidelines on T2DM. It plays an important role in monitoring and managing the condition and detecting and managing any comorbidities and T2DM-related complications. However, people with T2DM may miss the annual check-up, and the benefits of this check-up are lost. Therefore, this study aimed to determine the factors associated with nonattendance at the annual T2DM check-up in Ningbo, China.MethodsA case-control study was conducted using the Ningbo National Metabolic Management Center dataset. Cases were people with T2DM who were alive but did not attend the first annual check-up, scheduled between 1 March 2019 and 28 February 2022 (n = 1,549). Controls were people with T2DM who were alive and attended the first annual check-up during the same period (n = 1,354). The characteristics of cases and controls were compared using logistic regressions.ResultsThe odds of being a female [odds ratio (OR) 1.26, 95% confidence interval (CI) 1.06–1.50], alcohol drinker (1.26, 1.06–1.49), and with glycated hemoglobin A1c (HbA1c) ≥7% (1.67, 1.42–1.97) were higher among case patients than controls. The odds of being a high school graduate (0.77, 0.66–0.89) and on standard treatments in addition to lifestyle modification (oral hypoglycemic drug 0.63, 0.42–0.96; oral hypoglycemic drug and injection therapy 0.48, 0.32–0.73) were lower among case patients than controls.ConclusionThe factors associated with nonattendance at the annual T2DM check-up in Ningbo, China were female sex, not a high school graduate, alcohol drinker, HbA1c ≥7%, and only on lifestyle modification. The study findings should be used for improving attendance at the annual check-up among people with T2DM in Ningbo.

Background Fusarium wilt, caused by Fusarium oxysporum f. sp. cubense race 4 (Foc4), is the most lethal disease of bananas in Asia. Methods To better understand the defense response of banana to Fusarium wilt, the transcriptome and metabolome profiles of the roots from resistant and susceptible bananas inoculated with Foc4 were compared. Results After Foc4 inoculation, there were 172 and 1,856 differentially expressed genes (DEGs) in the Foc4-susceptible variety (G1) and Foc4-resistant variety (G9), respectively. In addition, a total of 800 DEGs were identified between G1 and G9, which were mainly involved in the oxidation-reduction process, cell wall organization, phenylpropanoid biosynthesis, and lipid and nitrogen metabolism, especially the DEGs of Macma4_08_g22610, Macma4_11_g19760, and Macma4_03_g06480, encoding non-classical arabinogalactan protein; GDSL-like lipase; and peroxidase. In our study, G9 showed a stronger and earlier response to Foc4 than G1. As the results of metabolomics, lipids, phenylpropanoids and polyketides, organic acids, and derivatives played an important function in response to Fusarium wilt. More importantly, Macma4_11_g19760 might be one of the key genes that gave G9 more resistance to Foc4 by a lowered expression and negative regulation of lipid metabolism. This study illustrated the difference between the transcriptomic and metabolomic profiles of resistant and susceptible bananas. These results improved the current understanding of host-pathogen interactions and will contribute to the breeding of resistant banana plants.

Tungsten is one of the most promising candidate materials for plasma-facing materials (PFM) because of its high melting point, superior thermal conductivity, and low sputtering rate. However, its significant brittleness and high ductile-to-brittle transition temperature constrain its broader engineering applications. To address these limitations, we developed a novel tungsten fiber-reinforced tungsten (Wf//W) composite. This composite utilizes commercially available black tungsten wire mesh as the fiber reinforcement phase with a mass ratio of 1 %. The material was synthesized through a combined Powder Extrusion Printing and High-Pressure, High-Temperature sintering process, achieving a relative density exceeding 99 %. Additionally, the composite exhibits a maximum hardness of 589 ± 10 HV, attributable to the presence of high-density dislocations in the matrix. Its compressive strength reaches up to 1530 MPa, and the plastic deformation strain is as high as 15.8 %, both of which are outstanding compared to existing research. These findings offer an alternative route for producing high-density Wf//W materials, in addition to chemical vapor deposition method and powder metallurgy technique.

AbstractPrevious studies on carbon storage simulation had ignored the difference of carbon intensity among various vegetation types inner the same land use. In this paper, The PLUS model was used to predict the land use change under multi-scenarios from 2030 to 2060, and the vegetation type data were supplemented by CA model to obtain the land cover-vegetation datasets from 2030-2060. Combined with the carbon density table of vegetation type, the future land use carbon storage during 2030-2060 under multi-scenarios in Beijing-Tianjin-Hebei region were analyzed. The main conclusions were as follows: (1) The spatial distribution of carbon storage in Beijing-Tianjin-Hebei region showed a pattern of ‘high in northeast-southwest and low in southeast-northwest’; (2) The carbon storage in Beijing-Tianjin-Hebei region during 1990-2020 showed a decreasing trend; (3) During 2030-2060, the carbon storage in Beijing-Tianjin-Hebei region showed a continuous decreasing trend in the absence of policy intervention, while that under the ecological protection and farmland protection scenarios showed an increasing trend; (4) Under different development scenarios, there were obvious significances of carbon storage in spatial distribution.

The results show that MFDMA method can describe the multifractal behavior of sEMG signal, and the multifractal characteristics such as spectral width have significant differences between muscle fatigue and non-fatigue state. The proposed method can reliably characterize exercise-induced muscle fatigue, and provide some feature reference for muscle fatigue recognition model and rehabilitation medicine research.

Carrying out green energy transformation, implementing clean energy power replacement and supply, and developing a new power system are some primary driving forces needed to fulfill China’s carbon-peak and carbon-neutral strategic goals. The construction of new power systems in China’s provinces and cities is developing rapidly, and the lack of a typical model promotes the application. The new power system path design should be based on the actual development of the power grid in different regions, energy use characteristics, and other actual needs to carry out the differentiated path design. In this context, this study analyzes the characteristics of the new domestic power system based on the policy background of the new domestic power system, constructs a new model for power system development stage identification, and proposes the overall design of the new power system development path from the power supply, transmission and distribution, and load sides. It also uses the Hebei South Network as an example to explore the development stage of the Hebei South Grid based on actual development needs. Finally, this study designs a novel power system development path for the entire supply and demand chain for the Hebei South Grid to propose ideas for constructing a new power system in China and to help green energy transformation.

45. Quantum-coherence-free precision metrology by means of difference-signal amplification

Abstract The novel weak-value-amplification (WVA) scheme of precision metrology is deeply rooted in the quantum nature of destructive interference between the pre- and post-selection states. And, an alternative version, termed as joint WVA (JWVA), which employs the difference-signal from the post-selection accepted and rejected results, has been found possible to achieve even better sensitivity (two orders of magnitude higher) under some technical limitations (e.g. misalignment errors). In this work, after erasing the quantum coherence, we analyze the difference-signal amplification (DSA) technique, which serves as a classical counterpart of the JWVA, and show that similar amplification effect can be achieved. We obtain a simple expression for the amplified signal, carry out characterization of precision, and point out the optimal working regime. We also discuss how to implement the post-selection of a classical mixed state. The proposed classical DSA technique holds similar technical advantages of the JWVA and may find interesting applications in practice.

Abstract Background Uremic tumoral calcinosis (UTC) is a rare complication in hemodialysis patients, whose mechanism remains incompletely understood. We report two cases with UTC who experienced completely different patterns of regression following parathyroidectomy, although there were no significant differences in serum calcium levels, parathyroid hormone, or phosphorus production between the two patients. Case presentation Case 1 had a substantial improvement in soft tissue calcification. However, in Case 2, one calcified mass was partially absorbed, while the others were aggravated with severe microvascular calcification and subcutaneous extravascular calcification. Whole-exome sequencing data revealed five mutation sites associated with atherosclerosis. Conclusion The different outcomes in UTC patients after PTX are rare. Further studies are required to elucidate the mechanism of paradoxical changes occurring in patients with UTC after parathyroidectomy.

Abstract Chronic kidney disease affects approximately 14.3% of people worldwide. Tubulointerstitial fibrosis is the final stage of almost all progressive CKD. To date, the pathogenesis of renal fibrosis remains unclear, and there is a lack of effective treatments, leading to renal replacement therapy. Mitophagy is a type of selective autophagy that has been recognized as an important way to remove dysfunctional mitochondria and abrogate the excessive accumulation of mitochondrial-derived reactive oxygen species (ROS) to balance the function of cells. However, the role of mitophagy and its regulation in renal fibrosis need further examination. In this study, we showed that mitophagy was induced in renal tubular epithelial cells in renal fibrosis. After silencing BNIP3, mitophagy was abolished in vivo and in vitro, indicating the important effect of the BNIP3-dependent pathway on mitophagy. Furthermore, in unilateral ureteral obstruction (UUO) models and hypoxic conditions, the production of mitochondrial ROS, mitochondrial damage, activation of the NLRP3 inflammasome, and the levels of αSMA and TGFβ1 increased significantly following BNIP3 gene deletion or silencing. Following silencing BNIP3 and pretreatment with mitoTEMPO or MCC950, the protein levels of αSMA and TGFβ1 decreased significantly in HK-2 cells under hypoxic conditions. These findings demonstrated that HIF1α-BNIP3-mediated mitophagy played a protective role against hypoxia-induced renal epithelial cell injury and renal fibrosis by reducing mitochondrial ROS and inhibiting activation of the NLRP3 inflammasome.

Tool wear prediction is of great significance in industrial production. Current tool wear prediction methods mainly rely on the indirect estimation of machine learning, which focuses more on estimating the current tool wear state and lacks effective quantification of random uncertainty factors. To overcome these shortcomings, this paper proposes a novel method for predicting cutting tool wear. In the offline phase, the multiple degradation features were modeled using the Brownian motion stochastic process and a SVR model was trained for mapping the features and the tool wear values. In the online phase, the Bayesian inference was used to update the random parameters of the feature degradation model, and the future trend of the features was estimated using simulation samples. The estimation results were input into the SVR model to achieve in-advance prediction of the cutting tool wear in the form of distribution densities. An experimental tool wear dataset was used to verify the effectiveness of the proposed method. The results demonstrate that the method shows superiority in prediction accuracy and stability.

Dementia exists as a ‘progressive clinical syndrome of deteriorating mental function significant enough to interfere with activities of daily living’, with the most prevalent type of dementia being Alzheimer’s disease (AD), accounting for about 80% of diagnosed cases. AD is associated with an increased risk of comorbidity with other clinical conditions such as hypertension, diabetes, and neuropsychiatric symptoms (NPS) including, agitation, anxiety, and depression as well as increased mortality in late life. For example, up to 70% of patients diagnosed with AD are affected by anxiety. As aging is the major risk factor for AD, this represents a huge global burden in ageing populations. Over the last 10 years, significant efforts have been made to recognize the complexity of AD and understand the aetiology and pathophysiology of the disease as well as biomarkers for early detection. Yet, earlier treatment options, including acetylcholinesterase inhibitors and glutamate receptor regulators, have been limited as they work by targeting the symptoms, with only the more recent FDA-approved drugs being designed to target amyloid-β protein with the aim of slowing down the progression of the disease. However, these drugs may only help temporarily, cannot stop or reverse the disease, and do not act by reducing NPS associated with AD. The first-line treatment options for the management of NPS are selective serotonin reuptake inhibitors/selective noradrenaline reuptake inhibitors (SSRIs/SNRIs) targeting the monoaminergic system; however, they are not rational drug choices for the management of anxiety disorders since the GABAergic system has a prominent role in their development. Considering the overall treatment failures and side effects of currently available medication, there is an unmet clinical need for rationally designed therapies for anxiety disorders associated with AD. In this review, we summarize the current status of the therapy of AD and aim to highlight novel angles for future drug therapy in our ongoing efforts to alleviate the cognitive deficits and NPS associated with this devastating disease.

50. Penifuranone A: A Novel Alkaloid from the Mangrove Endophytic Fungus Penicillium crustosum SCNU-F0006

One previously undescribed alkaloid, named penifuranone A (1), and three known compounds (2–4) were isolated from the mangrove endophytic fungus Penicillium crustosum SCNU-F0006. The structure of the new alkaloid (1) was elucidated based on extensive spectroscopic data analysis and single-crystal X-ray diffraction analysis. Four natural isolates and one new synthetic derivative of penifuranone A, compound 1a, were screened for their antimicrobial, antioxidant, and anti-inflammatory activities. Bioassays revealed that penifuranone A (1) exhibited strong anti-inflammatory activity in vitro by inhibiting nitric oxide (NO) production in lipopolysaccharide-activated RAW264.7 cells with an IC50 value of 42.2 μM. The docking study revealed that compound 1 exhibited an ideal fit within the active site of the murine inducible nitric oxide synthase (iNOS), establishing characteristic hydrogen bonds.
