Abstract Background This study clarified the synergistic relationship among annual changes to specify the changes in agro-meteorological factors, soil characteristics and peanut growth in saline-alkali land near the estuary of the Yellow River Delta. We aimed to find the key factors affecting peanut production to optimize and regulate peanut planting mode in saline alkali soil. Results The daily average temperature from early May to late September in Lijin and Kenli was above 24 °C, with 470–600 mm of precipitation. The sunshine duration was 7.9 h/day and 7.3 h/day and the accumulated temperature was 3742 °C and 3809 °C, in Lijin and Kenli, respectively. Agro-meteorological conditions were suitable for peanut growth and development with the consistent main developmental period in the two experiment regions. The best sowing period was when the soil temperature stabilized above 18 °C in early May, and the best harvest was in mid-September. The soil volumetric water content in Lijin concentrated among 25–40%. Salt was mainly distributed in the 40–60 cm soil layers, and increased rapidly to 2.5 g kg− 1 in 0–20 cm cultivation layer in mid-May due to lack of precipitation. In Kenli experiment region, the soil volumetric water content ranged from 10 to 35%. Soil salinity was mainly distributed in the 20 cm soil layer, and the changes in salinity was little affected by precipitation. From mid-July to mid-August, the effective accumulated temperature of 5 cm soil layer was above 520 °C in both regions, which could ensure the normal pod development. The slow dynamic growth of kernel, high unfilled pod rate (26.99%) and low shelling rate (66.0%) might be the main reasons for low peanut yield in Lijin. Conclusion Soil salinity was the main factor affecting pod development and yield. It was also a key point in optimizing the peanut planting mode in the saline alkali land of the Yellow River Delta.

Arbuscular mycorrhizae (AM) fungi form symbiotic associations with plant roots, providing nutritional benefits and promoting plant growth and defenses against various stresses. Metabolic changes in the roots during AM fungal colonization are key to understanding the development and maintenance of these symbioses. Here, we investigated metabolic changes in the roots of peanut (Arachis hypogaea L.) plants during the colonization and development of AM symbiosis, and compared them to uncolonized roots. The primary changes during the initial stage of AM colonization were in the contents and compositions of phenylpropanoid and flavonoid compounds. These compounds function in signaling pathways that regulate recognition, interactions, and pre-colonization between roots and AM fungi. Flavonoid compounds decreased by 25% when the symbiosis was fully established compared to the initial colonization stage. After AM symbiosis was established, general metabolism strongly shifted toward the formation of lipids, amino acids, carboxylic acids, and carbohydrates. Lipid compounds increased by 8.5% from the pre-symbiotic stage to well-established symbiosis. Lyso-phosphatidylcholines, which are signaling compounds, were only present in AM roots, and decreased in content after the symbiosis was established. In the initial stage of AM establishment, the content of salicylic acid increased two-fold, whereas jasmonic acid and abscisic acid decreased compared to uncolonized roots. The jasmonic acid content decreased in roots after the symbiosis was well established. AM symbiosis was associated with high levels of calcium, magnesium, and D-(+)-mannose, which stimulated seedling growth. Overall, specific metabolites that favor the establishment of AM symbiosis were common in the roots, primarily during early colonization, whereas general metabolism was strongly altered when AM symbiosis was well-established. In conclusion, specialized metabolites function as signaling compounds to establish AM symbiosis. These compounds are no longer produced after the symbiosis between the roots and AM becomes fully established.

Abstract Peanut (Arachis hypogaea) showcases geocarpic behavior, transitioning from aerial flowering to subterranean seed development. We recently obtained an atavistic variant of this species, capable of producing aerial and subterranean pods on a single plant. Notably, although these pod types share similar vigor levels, they exhibit distinct differences in their physical aspects, such as pod size, color, and shell thickness. We constructed 63 RNA-sequencing datasets, comprising three biological replicates for each of 21 distinct tissues spanning six developmental stages for both pod types, providing a rich tapestry of the pod development process. This comprehensive analysis yielded an impressive 409.36 Gb of clean bases, facilitating the detection of 42,401 expressed genes. By comparing the transcriptomic data of the aerial and subterranean pods, we identified many differentially expressed genes (DEGs), highlighting their distinct developmental pathways. By providing a detailed workflow from the initial sampling to the final DEGs, this study serves as an important resource, paving the way for future research into peanut pod development and aiding transcriptome-based expression profiling and candidate gene identification.

IntroductionPaclitaxel, a widely used chemotherapeutic agent for various cancers, induces peripheral neuropathy (PIPN) in approximately 80% of patients, severely affecting their quality of life. The role of vitamin D in pain perception has gained attention, but its correlation with PIPN remains unclear.MethodsThis study included 129 cancer patients who received adjuvant paclitaxel chemotherapy from January to June 2023. Neuropathic pain was assessed using the Douleur Neuropathique 4 Questions (DN4) questionnaire, and serum levels of vitamin D and glutathione (GSH) were measured to explore the correlation between vitamin D levels and neuropathic pain induced by paclitaxel chemotherapy.ResultsThe results showed a negative correlation between vitamin D deficiency and the occurrence of neuropathic pain (Spearman correlation coefficient of −0.324, P < 0.001). The receiver operating characteristic (ROC) curve analysis revealed that the area under the vitamin D curve for neuropathic pain was 0.681. Furthermore, after paclitaxel chemotherapy, there was a significant decrease in GSH levels in the serum of patients, with a more pronounced decline in the vitamin D-deficient group.DiscussionThe findings of this study indicate that higher levels of vitamin D are negatively associated with the occurrence of paclitaxel-induced neuropathic pain, suggesting that vitamin D might protect against oxidative stress. This discovery is significant for clinical treatment as it may help physicians better understand the mechanisms of pain during paclitaxel therapy and provide new strategies for the prevention and treatment of such pain. It also suggests that modulating vitamin D levels could reduce the neurotoxicity of paclitaxel, thereby improving patients’ quality of life and treatment compliance.

Peanuts grown in saline alkali soil are also subjected to drought stress caused by water scarcity. Therefore, we used HY25 (peanut variety) as an experimental material to investigate the effects of drought on the height of peanut main stems, length of the first lateral branch, leaf area per plant, SPAD value, net photosynthetic rate, and accumulation and distribution of photosynthetic products in saline alkali soil. The results showed that the combined stress of short-term drought and salt significantly reduced the main stem height, first lateral branch length, single plant leaf area, SPAD value, net photosynthetic rate (Pn), intercellular carbon dioxide concentration (Ci), and dry matter accumulation of peanuts, including a decrease in single plant pod yield, 100-pod weight, 100-kernel weight, and peanut yield. And the impact of drought stress on peanut yield varies at different growth stages. For example, under drought stress alone, the sensitive period is the 40th day after planting (40D) > 60th day after planting (60D) > 30th day after planting (30D). Short-term drought has the greatest impact on peanut yield at 40D, while in contrast, resuming watering after drought at 30D results in a slight but not significant increase in peanut yield in comparison with the control. Under the combined stress of drought and salt, the sensitive period of peanuts was 40D > 30D > 60D, and the single pod weight of peanuts was significantly reduced by 15.26% to 57.60% from the flowering stage to the pod stage under drought treatment compared to salt treatment, indicating a significant interaction between drought and salt stress, reducing the single leaf area and net photosynthetic rate of peanut leaves, ultimately leading to a decrease in peanut yield. Therefore, when planting peanuts in saline alkali soil, drought should be avoided, especially early drought, in order to prevent the combined effects of drought and salt stress from harming peanut yield.

Abstract Background Polyadenylation is a crucial process that terminates mRNA molecules at their 3′-ends. It has been observed that alternative polyadenylation (APA) can generate multiple transcripts from a single gene locus, each with different polyadenylation sites (PASs). This leads to the formation of several 3′ untranslated regions (UTRs) that vary in length and composition. APA has a significant impact on approximately 60–70% of eukaryotic genes and has far-reaching implications for cell proliferation, differentiation, and tumorigenesis. Results In this study, we conducted long-read, single-molecule sequencing of mRNA from peanut seeds. Our findings revealed that over half of all peanut genes possess over two PASs, with older developing seeds containing more PASs. This suggesting that the PAS exhibits high tissue specificity and plays a crucial role in peanut seed maturation. For the peanut acetyl-CoA carboxylase A1 (AhACCA1) gene, we discovered four 3′ UTRs referred to UTR1–4. RT-PCR analysis showed that UTR1-containing transcripts are predominantly expressed in roots, leaves, and early developing seeds. Transcripts containing UTR2/3 accumulated mainly in roots, flowers, and seeds, while those carrying UTR4 were constitutively expressed. In Nicotiana benthamiana leaves, we transiently expressed all four UTRs, revealing that each UTR impacted protein abundance but not subcellular location. For functional validation, we introduced each UTR into yeast cells and found UTR2 enhanced AhACCA1 expression compared to a yeast transcription terminator, whereas UTR3 did not. Furthermore, we determined ACC gene structures in seven plant species and identified 51 PASs for 15 ACC genes across four plant species, confirming that APA of the ACC gene family is universal phenomenon in plants. Conclusion Our data demonstrate that APA is widespread in peanut seeds and plays vital roles in peanut seed maturation. We have identified four 3′ UTRs for AhACCA1 gene, each showing distinct tissue-specific expression patterns. Through subcellular location experiment and yeast transformation test, we have determined that UTR2 has a stronger impact on gene expression regulation compared to the other three UTRs.

To explore the key factors and specific thresholds of water resources limiting economic development, and to provide technical support for water resources management in cities dominated by agriculture similar to Zhangjiakou. We used the Tapio elastic decoupling method to quantitatively evaluate the decoupling relationship between the water resources ecological footprint (WEF) and economic growth. Then the logarithmic mean Divisia index (LMDI) and mathematical statistics are used to identify the key factors and threshold effects. The results show a significant decreasing trend in the WEF and obvious spatial differences in Zhangjiakou between 2006 and 2015, with agricultural ecological footprint dominating all districts and counties (77.54 ± 14.35%). The changes in technological effect are a contributing factor to the decoupling between the WEF and the economy in Zhangjiakou, while the economic effect is the main restricting factor. In particular, there is a high correlation between the WEF and the number of water-saving irrigation machines and the total power of agricultural machinery. According to the findings, for water-scarce cities such as Zhangjiakou, where agriculture is the primary focus, it is suggested that increasing the number of agricultural machinery can effectively alleviate the problem of water scarcity constraining economic development. HIGHLIGHTS Combining the water resources ecological footprint theory with the Tapio decoupling model, this paper analyses the changes of water resources ecological footprint and decoupling state in different districts and counties of Zhangjiakou City in terms of time and space.; Combining LMDI modelling with mathematical and statistical methods to analyse the key factors affecting water resources and economic change in Zhangjiakou.;

The Ca2+/CaM signal transduction pathway helps plants adapt to environmental stress. However, our knowledge on the functional proteins of Ca2+/CaM pathway in peanut (Arachis hypogeae L.) remains limited. In the present study, a novel calmodulin 4 (CaM4)-binding protein S-adenosyl-methionine synthetase 1 (SAMS1) in peanut was identified using a yeast two-hybrid assay. Expression of AhSAMS1 was induced by Ca2+, ABA, and salt stress. To elucidate the function of AhSAMS1, physiological and phenotypic analyses were performed with wild-type and transgenic materials. Overexpression of AhSAMS1 increased spermidine and spermidine synthesis while decreased the contents of ethylene, thereby eliminating excessive reactive oxygen species (ROS) in transgenic lines under salt stress. AhSAMS1 reduced uptake of Na+ and leakage of K+ from mesophyll cells, and was less sensitive to salt stress during early seedling growth, in agreement with the induction of SOS and NHX genes Transcriptomics combined with epigenetic regulation uncovered relationships between differentially expressed genes and differentially methylated regions, which raised the salt tolerance and plants growth. Our findings support a model in which the role of AhSAMS1 in the ROS-dependent regulation of ion homeostasis was enhanced by Ca2+/CaM while AhSAMS1-induced methylation was regulated by CaM, thus providing a new strategy for increasing the tolerance of plants to salt stress.

The application of nitrogen fertilizer is crucial to the growth and biological nitrogen fixation of peanut, especially in the seedling stage where nodules have not yet formed. However, it is still uncertain how much initial nitrogen fertilizer should be applied to promote peanut root growth, nodule formation, and biological nitrogen fixation (BNF). There, a 2-year pot experiment was conducted using Huayu 22 (HY22, large-grain cultivar) and Huayu 39 (HY39, small-grain cultivar) as experimental materials to research the effects of different initial nitrogen fertilizer application rates on peanut root growth (root weight, root length, root mean diameter, root activity) and biological nitrogen fixation capacity (nodule number, nodule weight, biological nitrogen fixation, and nitrogen fixation potential per plant). N0, as control, four initial nitrogen fertilizer application rates were established: 15 kg·hm−2 (N15), 30 kg·hm−2 (N30), 45 kg·hm−2 (N45), and 60 kg·hm−2 (N60). The present results showed that the nodule number, nodule dry weight, nitrogenase activity, and biological nitrogen fixation of the HY22 cultivar under the N15 treatment were higher compared to those under other treatments over the two growing seasons. In addition, the cultivar of HY39 treated with the N15 treatment also increased the nitrogen fixation potential per plant and BNF relative to other treatments. Although the application of 60 kg·hm−2 nitrogen increased the root surface area and root volume, it decreased the nitrogenase activity, nodule dry weight, and nitrogen fixation potential per plant of HY22 and HY39 varieties in both growing seasons. Above all, an initial nitrogen application of 15 kg·hm−2 may be the optimal treatment for promoting peanut nodule formation and biological nitrogen fixation.

Short-term drought events occur more frequently and more intensively under global climate change. Biochar amendment has been documented to ameliorate the negative effects of water deficits on plant performance. Moreover, biochar can alter the soil microbial community, soil properties and soil metabolome, resulting in changes in soil functioning. We aim to reveal the extent of biochar addition on soil nutrients and the soil microbial community structure and how this improves the tolerance of legume crops (peanuts) to short-term extreme drought. We measured plant performances under different contents of biochar, set as a gradient of 2%, 3% and 4%, after an extreme experimental drought. In addition, we investigated how soil bacteria and fungi respond to biochar additions and how the soil metabolome changes in response to biochar amendments, with combined growth experiments, high-throughput sequencing and soil omics. The results indicated that biochar increased nitrites and available phosphorus. Biochar was found to influence the soil bacterial community structure more intensively than the soil fungal community. Additionally, the fungal community showed a higher randomness under biochar addition when experiencing short-term extreme drought compared to the bacterial community. Soil bacteria may be more strongly related to soil nutrient cycling in peanut agricultural systems. Although the soil metabolome has been documented to be influenced by biochar addition independent of soil moisture, we found more differential metabolites with a higher biochar content. We suggest that biochar enhances the resistance of plants and soil microbes to short-term extreme drought by indirectly modifying soil functioning probably due to direct changes in soil moisture and soil pH.

Water shortage and water pollution are two prominent issues in North China. Understanding hydrological cycle and water quality changes in response to pollution control measures is fundamental for a better water management there. Using coupled MIKE SHE/MIKE 11 modeling, various hydrological components in Yanghe Basin in a semiarid area of North China were quantified for three typical hydrological years and concentrations of COD and TP in a national monitoring section of Yanghe were evaluated with/without pollution control measures. The modeling results show that the underground water storage of Yanghe Basin becomes depleted due to evapotranspiration compensation and groundwater utilization regardless of hydrological condition, indicating an unsustainable in-situ water resource utilization. Water quality goals set for Yanghe (COD ≤ 20 mg/L and TP ≤ 0.2 mg/L) can hardly be achieved if pollution control measures are not taken, especially for a dry hydrological year. Depending on hydrological conditions, non-point source control technology-related projects in a 109-km2 village and a 7-km river-channel wetland in mainstream of Yanghe will have a positive effect or negligible effect on water quality improvement. To meet water quality goals, implementation of three wetlands is an effective and economic way. HIGHLIGHTS A coupled MIKE SHE/MIKE 11 model is constructed for Yanghe Basin in North China.; Yanghe Basin undergoes unsustainable water resource utilization, with evapotranspiration surpassing rainfall.; Demonstration water projects on a small scale have either a positive or negligible effect on water quality of Yanghe.; Implementation of three river-channel wetlands is an effective and economic way to achieve water quality goals.;

Triacylglycerols is the major storage lipid in most crop seeds. As the key enzyme catalyzing the final step of triacylglycerols biosynthesis, the activity of diacylglycerol acyltransferases directly related to oil content. It has been shown that certain amino acids are very important for enzyme activity, one amino acid variation will greatly change the enzyme activity. In this study, we identified three amino acid point mutations that affect the Arachis hypogaea diacylglycerol acyltransferase 2 enzyme activity, T107M, K251R and L316P. According to the three amino acid variations, three single-nucleotide-mutant sequences of Arachis hypogaea diacylglycerol acyltransferase 2a were constructed and transformed into yeast strain H1246 for function verification. Results showed that T107M and K251R could change the fatty acid content and composition of the transformed yeast strains, whereas L316P led to the loss of enzyme activity. By analyzing the 2D and 3D structures of the three variants, we found that the changes of spatial structure of T107M, K251R and L316P caused the changes of the enzyme activity. Our study could provide a theoretical basis for changing the enzyme activity of DGAT by genetic engineering, and provide a new idea for increasing the oil content of the crops.

Peanut (Arachis hypogaea L.) is an important oil and cash crop. An efficient utilization of saline–alkali soil resources, the development of peanut planting in saline–alkali soil, and obtaining high and stable yield have become urgent needs to ensure peanut production. Arbuscular mycorrhizal fungi (AMF) have been reported to develop the potential productivity of host plants and improve their salt resistance and tolerance. However, there is still limited research on promoting the growth and morphology of peanut roots. Therefore, in this study, seeds of salt-tolerant peanut variety “HY 25” were coated with commercial AMF inoculant before being planted in saline–alkali and normal soils to investigate the effects of AMF on peanut root growth and rhizosphere soil. The results showed that root morphological characteristics were significantly increased by the use of AMF at the podding stage in saline–alkali soil and from the flowering and pegging stage to the maturity stage in normal soil. Of note, the total root volume of peanut inoculated with AMF significantly increased by 31.57% during the podding stage in saline–alkali soil. Meanwhile, AMF significantly increased the phosphatase and invertase activities in the peanut rhizosphere of saline–alkali soil from the flowering stage to maturity stage and soil CAT activity at the maturity stage (41.16~48.82%). In normal soil, soil phosphatase and urease activities were enhanced by AMF at the flowering stage and the podding stage, respectively. AMF also increased the contents of soil organic matter, available phosphorus, and hydrolysable nitrogen, but decreased soil EC in saline–alkali soil. In addition to the significant increase in soil available phosphorus content, AMF had no significant effect on the physical and chemical properties of the soil and other soil nutrients in normal soil. AMF significantly increased pod biomass and yield in saline–alkali soil and normal soil, and improved their agronomic characteristics. In conclusion, peanut seeds coated with AMF improved the root morphological characteristics of peanuts and improved the physical and chemical properties in peanut rhizosphere, especially in saline–alkali soil. The process of rhizosphere soil nutrient transformation was also enhanced. Finally, AMF improved plant agronomic traits to increase the pod yield (16.5~21.9%). This study provides the theoretical basis and technical support for the application of AMF in peanut production in saline–alkali soil.

Metal fused-coating technology has the advantages of both low cost and high efficiency and is a new additive manufacturing technology in recent years. The previous studies were mainly aimed at the optimization of process parameters and the control of the surface quality of parts, while there were few theoretical analyses on the microstructure morphology after solidification. A three-dimensional transient numerical model was established to calculate temperature gradient and solidification rate, considering the changes in material physical properties with temperature during the calculation process. The temperature gradient on the substrate surface is jointly affected by the melt flowing out of the nozzle and the welding arc. It was found that the solidification front of the aluminum alloy was in an unstable state during the coating process. When the value of G/R decreases, the microstructure of the solidification interface gradually changes from columnar crystals to columnar dendrites and equiaxial crystals. The microstructure at the bottom of both the molten pool and coating layer is columnar crystal, while the microstructure at the upper part is equiaxed crystal.

Long-term excessive application of chemical fertilizers can cause many problems, such as soil degradation and environmental pollution. Therefore, we reduced conventional nitrogen fertilization and added organic fertilizers in some cases to investigate the response of photosynthetic characteristics, root nodules and yield on reduced nitrogen fertilization. Compared to conventional nitrogen fertilization, the 25% and 35% nitrogen reduction treatments reduced the leaf area index, net photosynthetic rate, 100-fruit weight, 100-kernel weight and the yield of peanut, but had no significant effect on the kernel rate. With constant N fertilizer, adding organic fertilization alone increased leaf area index, chlorophyll, net photosynthetic rate and yield of peanut. In compounded treatments of nitrogen and organic fertilizer, the highest yields were achieved in the 25% N reduction with the 3000 kg/hm−2 organic fertilizer treatment (T3) and the 4500 kg/hm−2 organic fertilizer treatment (T4); furthermore, the net photosynthetic rate, leaf area index, yield and fertilizer contribution were significantly higher in these two treatments than in the conventional fertilizer treatments. Nitrogen fertilizer had significant effects on the quantity and fresh weight of root nodules. Concretely, nitrogen reduction increased the quantity and fresh weight of root nodules of peanut in the early stage of fertility but decreased them in the harvest stage. Nitrogen reduction with an additional organic fertilizer in the late stage of fertility increased the quantity and fresh weight of root nodules of peanut. Considering the property of root nodules was significantly positively correlated with net photosynthetic rate and yield, the arguments above may be the mechanism of the highest yields found in T3 and T4. This work can provide empirical and instructional support for a balanced fertilization strategy in peanut agriculture and high-yielding and efficient cultivation of peanut.

Excessive nitrogen and insufficient calcium could significantly impact peanut yields. This study investigated the effects of nitrogen and calcium fertilizers on nutrient absorption, utilization, and yield; experiments were conducted using the peanut cultivar from Xianghua 2008 in a split-plot arrangement with two calcium fertilizer levels (Ca0: 0 and Ca1: 568 kg CaO ha−1) in the main plots and six nitrogen fertilizer gradients (N0: 0, N45.0: 45.0, N90.0: 90.0, N112.5: 112.5, N135.0: 135.0, and N157.5: 157.5 kg N ha−1) in subplots between 2015 and 2016 in Changsha, China. We examined the impact of different rates of calcium and nitrogen fertilizers on the net photosynthetic rate (Pn), agronomic traits, dry matter quality, yield and yield composition, nutrient accumulation, and distribution. The combined application of calcium and nitrogen fertilizers significantly affected the yield and yield components, Pn, main stem height, dry matter, and nutrient accumulation. Under the same calcium level, nitrogen application significantly increased the main stem height and Pn and promoted the accumulation of dry matter and nutrients in the plant, particularly in the kernel. Under the same nitrogen treatment, calcium significantly increased Pn and promoted the accumulation of dry matter, calcium, and magnesium. The pod yield increased gradually with an increasing nitrogen application rate (0–112.5 kg ha−1) and peaked at N112.5, increasing by 52.3–138.0% compared with N0. However, excessive nitrogen application (N > 112.5 kg ha−1) decreased the pod yield. Under different nitrogen fertilizer levels, calcium application increased pod yields by 11.5–29.6% by promoting Pn, nutrient uptake, accumulation in the individual plant, and nutrient accumulation in the kernel. Therefore, this study suggested that adjusting the calcium (568 kg ha−1) and nitrogen (112.5 kg ha−1) fertilizer rates significantly improved peanut growth and productivity by enhancing photosynthetic efficiency and nutrient accumulation in calcium-deficient acidic red soil.

Abstract Voltage sags and momentary interruptions of power are the most severe voltage disturbances. Here, a tansformerless active voltage quality regulator with the novel three‐leg circuit is proposed to mitigate long duration and deep voltage sags. The shunt rectifier and series inverter of this novel three‐leg circuit are all full‐bridge structures, so it has lower DC bus voltage and smaller filter parameters than the conventional half‐bridge topologies. The totem‐pole bridgeless boost circuit is formed among the three bridge arms and the DC bus voltage will be charged to exceed the peak value of the supply voltage. It can compensate for deep voltage sags (that may be more than 50% of the nominal voltage). Compared to the conventional transformerless series‐connected topology, the proposed topology has the merits of higher efficiency, smaller volume and lower cost. The validity and feasibility of the proposed topology are verified by theoretical analysis, simulation and experiment.

Quick differentiation of the circulating variants and the emerging recombinant variants of SARS-CoV-2 is essential to monitor their transmission. However, the widely used gene sequencing method is time-consuming and costly when facing the viral recombinant variants, because partial or whole genome sequencing is required. Allele-specific real time RT-PCR (qRT-PCR) represents a quick and cost-effective method in SNP genotyping and has been successfully applied for SARS-CoV-2 variant screening. In the present study, we developed a panel of 3 multiplex allele-specific qRT-PCR assays targeting 12 key differential mutations for quick differentiation of SARS-CoV-2 recombinant variants (XD and XE) and Omicron subvariants (BA.1 and BA.2). Two parallel multiplex qRT-PCR reactions were designed to separately target the protype allele and the mutated allele of the four mutations in each allele-specific qRT-PCR assay. The variation of Cp values (ΔCp) between the two multiplex qRT-PCR reactions was applied for mutation determination. The developed multiplex allele-specific qRT-PCR assays exhibited outstanding analytical sensitivities (with limits of detection [LoDs] of 2.97-27.43 copies per reaction), wide linear detection ranges (107-100 copies per reaction), good amplification efficiencies (82% to 95%), good reproducibility (Coefficient of Variations (CVs) < 5% in both intra-assay and inter-assay tests) and clinical performances (99.5%-100% consistency with Sanger sequencing). The developed multiplex allele-specific qRT-PCR assays in this study provide an alternative tool for quick differentiation of SARS-CoV-2 recombinant variants (XD and XE) and Omicron subvariants (BA.1 and BA.2).

The dual challenges of global climate change and reductions in the amount of arable land represent growing threats to the stability of global human populations. Efforts to further optimize cropping systems to maximize yields while minimizing greenhouse gas emissions in limited land areas have thus emerged as a focus in modern agriculture. Cereal-intercropping management strategies may represent a promising approach to simultaneously addressing both of these challenges in China. We aimed at comprehensively assessing changes in yield, carbon footprint, and net ecosystem economic benefit when transitioning from maize/peanut monoculture to intercropping in a field-scale study in an effort to aid in the development of low-carbon intercropping systems that do not have an adverse impact on Chinese grain yields. Beginning in June of 2018, a randomized complete block design with three treatments was used to initiate this study: (1) peanut monoculture (P), (2) maize monoculture (M), and (3) maize/peanut intercropping (MP). We compared yield, greenhouse gas emissions, carbon footprint and net ecosystem economic benefit. Results over four years showed that the land equivalent ratio associated with MP was greater than 1. All three of these cropping systems were net CO2 and N2O sources as well as net CH4 sinks, with MP generating significantly (p < 0.05) lower N2O and CO2 flux as well as smaller seasonal N2O and CO2 emissions relative to M. MP additionally reduced the carbon footprint associated with this cropping system by 11.11–31.65% and 30.37–43.62% relative to M and P, respectively. Consistently, MP treatment resulted in respective 70.69% and 26.25% net ecosystem economic benefit (NEEB) increases relative to the M and P conditions while simultaneously enhancing energy use efficiency. In summary, MP systems have potential economic benefit with lower environmental risk alternative to traditional peanut or maize monocropping systems. Converting from peanut or maize monocropping systems to MP systems practices contributed to improved farmland use efficiency, clean production and increased farmers’ income in an agricultural system.

Although the biological relationship between vitamin D (VD) deficiency and cognitive function has been recognized by many scholars, the theoretical mechanisms involved are still not well-understood. In this study, we demonstrated the role of VD in alleviating the cognitive dysfunction in aged mice caused by sevoflurane anesthesia. Forty female C57BL/6 mice aged 12 months were selected for the experiment. VD (-) and VD (+) mouse models and sevoflurane anesthesia models were established. Mice were randomly divided into normal elderly group (NC group), normal aged mice + sevoflurane anesthesia treatment group (NS group), aged VD (-) mice + sevoflurane anesthesia treatment group [VD (-) group], and aged VD (+) + sevoflurane anesthesia treatment group [VD (+) group]. To compare the emergence time after sevoflurane anesthesia in aged mice with different levels of VD and to test the cognitive function of four groups through the water maze. Inflammatory factor expression and cholinergic activity in hippocampus tissue of all mice were measured at the end of behavioral tests. These data show that, low levels of VD aggravated the delayed emergence and cognitive dysfunction in aged mice caused by sevoflurane anesthesia, while higher levels of VD mitigated this impairment by enhancing cholinergic activity and reducing inflammatory factor expression in the hippocampus.

PurposeVitamin D prevents hypocalcaemia, osteoporosis, and infections, among other problems, and is involved in the prevention and treatment of cardiovascular and neurological diseases. Recently, vitamin D was shown to improve cognitive dysfunction caused by Alzheimer's disease and vascular dementia. This study aims to explore the correlation between preoperative serum vitamin D and postoperative cognitive disorder (POCD) occurrence in elderly patients with gastrointestinal tumors to guide perioperative medication use and promote early patient recovery.MethodsThis study recruited 238 elderly patients (65 ≤ age ≤ 85) who underwent gastrointestinal tumor surgery; 117 cases were enrolled, and 55 controls of the same age and education level as the cases were included. Blood samples were taken preoperatively and at 7, 15, 30, and 90 days postoperatively, and plasma vitamin D (25OH-D3) and glutathione (GSH) was measured. Different from the previous diagnosis of POCD was obtained by telephone interview through Cognitive Status Modified Telephone Interview (TICS-m), mainly for memory impairment, a series of neuropsychological tests was used to evaluate cognitive function, Picture Recollect Test, Stroop Color-word Test, and Digit Symbol Substitution Test were used to comprehensively evaluate the three domains of cognitive function of patients, namely memory, attention and information processing ability. All neuropsychiatric assessments were performed at the bedside and completed face-to-face by the assessment staff and the patient.ResultsA total of 65.8% (77/117) of elderly patients undergoing gastrointestinal tumor surgery had preoperative vitamin D deficiency (serum 25OH-D concentration < 12 ng/ml), of whom 46.7% (36/77, 7 days after surgery), 31.2% (24/77, 15 days after surgery), 15.6% (12/77, 30 days after surgery), and 9% (7/77, 90 days after surgery) of patients developed POCD; 7.5% (3/40) of patients without vitamin D deficiency developed PNDs, which was detected only on the 7th day after surgery.ConclusionsVitamin D deficiency can increase neurocognitive disorder risk in elderly patients during the perioperative period, possibly because low vitamin D levels cannot effectively inhibit the postoperative oxidative stress increase.Trial RegistrationThis experiment was approved and registered by the China Clinical Trial Registration Center, registration number ChiCTR2100046900 (30/05/2021).

This paper focuses on how to make a charging guidance for urgent charging electric vehicles (EVs) to fast charging stations. To avoid charging overload in the spatial-temporal scale and excessive waiting time for EV users, a charging guidance strategy based on the virtual service range is proposed. Considering the mutual influence among EVs' charging selections, the proposed strategy can unify the temporal shift and spatial shift of the charging load into the same time scale to control rather than other works only do the temporal shift. Specifically, the proposed strategy has two stages: the charging arrangement and the spatial-temporal shift. The charging arrangement stage is to determine whether an EV can be allowed to charge at a certain time and a given charging station while the spatial-temporal shift stage is to regulate the charging time or charging location. Besides, a bucketsort-based algorithm is proposed to solve the formulated problem in the charging arrangement stage. Finally, three scenarios have been designed to demonstrate the feasibility of the proposed strategy and the performance of the presented algorithm is compared with the other decentralized algorithms.

With the rapid development of multispectral detection, infrared and visible compatible camouflage becomes necessary. Metafilms with dielectric/metal/dielectric (D/M/D) structures can be highly transparent in visible band (380 ∼ 780 nm) and highly reflective in infrared atmospheric windows (3 ∼ 5 μ m, 8 ∼ 14 μ m). The metafilm can be deposited on the equipment surface, and the high visible transmittance can make the original camouflage coating continue to achieve visible camouflage, while the low infrared emissivity can inhibit the infrared signal to achieve infrared camouflage. Compatible camouflage is urgently needed by high-temperature targets such as exhaust pipes and engine cabins. Therefore, the thermal stability of multilayer structure is very important. In this study, a D/M/D-structured metafilm with improved thermal performance is proposed. Al-doped zinc oxide (AZO) is selected as the material of the dielectric layers due to good thermal stability, and high visible transmittance is realized through the mechanism of admittance matching. Ag is selected as the material of the metal layer to increase infrared reflectance. The metafilm with the structure of AZO/Ag/AZO is rigorously designed and fabricated. The results from Fourier transform infrared spectrometer and spectrophotometer show that the integrated visible transmittance and infrared emissivity at room temperature is higher than 0.87 and lower than 0.05, respectively. The camouflage performance of the metafilm is demonstrated on a flexible polyethylene terephthalate (PET) substrate. The camouflage performance of metafilm samples at 20 ∼ 140 °C is tested on a model cabin. The metafilm does not affect the original camouflage coating, so it can achieve visible camouflage. The radiation temperature of the metafilm is approximately 80 °C lower than that of the control surface, and the infrared signature is significantly attenuated. In order to further investigate the thermal stability and thermal fatigue resistance of the metafilm, metafilm deposited on quartz substrate is continuously heated and periodically heated at different temperatures. It is found that the sample can withstand continuous heating at 450 °C for 4 h or repeated heating for 20 cycles. SEM (scanning electron microscope) and EDS (energy dispersive spectrometer) scanning shows that if heated at higher temperature or for more cycles, the AZO layer becomes blocky, and the proportion of Ag and O changes significantly. This leads to the decrease of visible transmittance and the increase of infrared emissivity of samples.

Waterlogging has negative effects on crop yield. Physiological and transcriptome data of two peanut cultivars [Zhongkaihua 1 (ZKH 1) and Huayu 39 (HY 39)] were studied under normal water supply and waterlogging stress for 5 or 10 days at the flowering stage. The results showed that the main stem height, the number of lateral branches, lateral branch length, and the stem diameter increased under waterlogging stress, followed by an increase in dry matter accumulation, which was correlated with the increase in the soil and plant analysis development (SPAD) and net photosynthetic rate (Pn) and the upregulation of genes related to porphyrin and chlorophyll metabolism and photosynthesis. However, the imbalance of the source–sink relationship under waterlogging was the main cause of yield loss, and waterlogging caused an increase in the sucrose and soluble sugar contents and a decrease in the starch content; it also decreased the activities of sucrose synthetase (SS) and sucrose phosphate synthetase (SPS), which may be due to the changes in the expression of genes related to starch and sucrose metabolism. However, the imbalance of the source–sink relationship led to the accumulation of photosynthate in the stems and leaves, which resulted in the decrease of the ratio of pod dry weight to total dry weight (PDW/TDW) and yield. Compared with ZKH 1, the PDW of HY 39 decreased more probably because more photosynthate accumulated in the stem and leaves of HY 39 and could not be effectively transported to the pod.

Background In China, double-seed (DS) sowing (i.e., sowing two seeds per hole) has been conventionally performed towards the erect-plant-type peanuts to increase the low germination rate due to poor seed preservation conditions. However, the corresponding within-hole plant competition usually limits the subsequent plant growth and the final yield. We developed a high-yield cultivation system of single-seed (SS) precision sowing to solve this paradox, saving 20% of seeds and increasing yields by more than 10% relative to the conventional DS sowing. Methods To explore the mechanisms of these two different cropping patterns in peanut yields, we conducted transcriptomic and physiological comparisons in the seeding plant leaf and root tissues between SS precision sowing and standard DS sowing treatments. Results After assembly, each library contained an average of 43 million reads and generated a total of 523,800, 338 clean reads. After GO and Kyoto Encyclopedia of Genes and Genomes pathway analysis, we found the key genes for biotic and abiotic stress showed higher expression in roots of plants grown under the SS precision sowing treatment, including genes encoding disease resistance, oxidation-reduction, hormone related, and stress response transcription factors and signaling regulation proteins. In particular, the resveratrol synthesis genes related to stress and disease resistance appeared induced in roots under the SS sowing treatment. Conclusion These data indicated that Aspergillus flavus resistance and stress tolerance in roots under SS precision sowing were enhanced compared with roots under the DS sowing treatment. This work benefits the development of underground pods and thus increasing peanut yields.

Abstract Background Balance performance is considered as an indicator of functional status in the elderly, a large scale population screening and evaluation in the community context followed by proper interventions would be of great significance at public health level. However, there has been no suitable balance testing scale available for large scale studies in the unique community context of urban China. Methods A balance scale named X16 balance testing scale was developed, which was composed of 3 domains and 16 items. A total of 1985 functionally independent and active community-dwelling elderly adults’ balance abilities were tested using the X16 scale. The internal consistency, split-half reliability, content validity, construct validity, discriminant validity of X16 balance testing scale were evaluated. Results Factor analysis was performed to identify alternative factor structure. The Eigenvalues of factors 1, 2, and 3 were 8.53, 1.79, and 1.21, respectively, and their cumulative contribution to the total variance reached 72.0%. These 3 factors mainly represented domains static balance, postural stability, and dynamic balance. The Cronbach alpha coefficient for the scale was 0.933. The Spearman correlation coefficients between items and its corresponding domains were ranged from 0.538 to 0.964. The correlation coefficients between each item and its corresponding domain were higher than the coefficients between this item and other domains. We aimed to elucidate the possible yield-increasing mechanisms through regulation of shade-avoidance responses at both physiological and molecular levels under monoseeding. Our results revealed that monoseeding decreased the main stem height but increased the main stem diameter and the number of branches and nodes compared to the traditional double- and triple-seeding patterns. The chlorophyll contents were higher under monoseeding than that under double- and triple-seeding. Further analysis showed that this, in turn, increased the net photosynthetic rate and reallocated higher levels of assimilates to organs. Monoseeding induced the expression patterns of Phytochrome B (Phy B) gene but decreased the expression levels of Phytochrome A (Phy A) gene. Furthermore, the bHLH transcription factors (PIF 1 and PIF 4) that interact with the phytochromes were also decreased under monoseeding. The changes in the expression levels of these genes may regulate the shade-avoidance responses under monoseeding. In addition, monoseeding increased pod yield at the same population density through increasing the number of pods per plant and 100-pod weight than double- and triple-seeding patterns. Thus, we inferred that monoseeding is involved in the regulation of shade-avoidance responsive genes and reallocating assimilates at the same population density, which in turn increased the pod yield.

Arch-cruciform DNA are self-assembled on AuNPs/VS2 scaffold as a highly sensitive and selective electrochemical biosensor for michigan cancer foundation-7 (MCF-7) breast cancer cells. In the construction, arch DNA is formed using two single-strand DNA sequences embedded with the aptamer for MCF-7 cells. In the absence of MCF-7 cells, a cruciform DNA labeled with three terminal biotin is bound to the top of arch DNA, which further combines with streptavidin-labeled horseradish peroxidase (HRP) to catalyze the hydroquinone-H2O2 reaction on the electrode surface. The presence of MCF-7 cells can release the cruciform DNA and reduce the amount of immobilized HRP, thus effectively inhibiting enzyme-mediated electrocatalysis. The electrochemical response of the sensor is negatively correlated with the concentration of MCF-7 cells, with a linear range of 10~1 × 105 cells/mL, and a limit of detection as low as 5 cells/mL (S/N = 3). Through two-dimensional materials and enzyme-based dual signal amplification, this biosensor may pave new ways for the highly sensitive detection of tumor cells in real samples.

In this work, a virtual submerged floating operational system (VSFOS) based on parallel and serial robotic platforms is proposed. The primary aim behind its development lies in carrying out simulated underwater manipulation experiments in an easier and safer way. This VSFOS is consisted of a six-degree-of-freedom (6-DOF) parallel platform, an ABB serial manipulator, an inertial sensor, and a real-time industrial computer. The 6-DOF platform is used to simulate the movement of an underwater vehicle, whose attitude is measured by the inertial sensor. The ABB manipulator, controlled by the real-time industrial computer, works as an operational tool to perform underwater manipulation tasks. In the control system architecture, software is developed to receive the data collected by the inertial sensor, to communicate and send instructions. Furthermore, the real-time status of the manipulator is displayed in this software. To validate the proposed system, two experiments have been conducted to test its performance. In the first experiment, the test is carried out to check the communication function of VSFOS, while in the second one, the manipulator is intended to follow the movement of the parallel platform and perform simulated operational task in the space. The obtained results from these two experiments show clearly the effectiveness and the performance of the proposed VSFOS.

Calcium not only serves as a necessary nutrient for plant growth but also acts as a ubiquitous central hub in a large number of signaling pathways. Free Ca2+ deficiency in the soil may cause early embryo abortion, which eventually led to abnormal development of peanut pod during the harvest season. To understand the mechanisms of Ca2+ regulation in pod development, transcriptome analysis of peanut gynophores and pods was performed by comparing the treatments between free Ca2+ sufficiency and free Ca2+ deficiency using Illumina HiSeq™ 2000. 9,903,082,800 nt bases are generated totally. After assembly, the average length of 102,819 unigenes is 999 nt, N50 is 1,782 nt. RNA-seq based gene expression profilings showed a large number of genes at the transcriptional level changed significantly between the aerial pegs and underground swelling pods under free Ca2+ sufficienct or deficiency treatments, respectively. Genes encoding key members of Ca2+ signaling transduction pathway, enzymes for hormone metabolism, cell division and growth, transcriptional factor as well as embryo development were highlighted. This information provides useful information for our further study. The results of digital gene expression (DGE) indicated that exogenous calcium might contribute to the development of peanut pod through its signal transduction pathway, meanwhile, promote the normal transition of the gynophores to the reproductive development.

Peanut is one of the calciphilous plants. Calcium serves as a ubiquitous central hub in a large number of signaling pathways. In the field, free calcium ion (Ca2+)-deficient soil can result in unfilled pods. Four pod stages were analyzed to determine the relationship between Ca2+ excretion and pod development. Peanut shells showed Ca2+ excretion at all four stages; however, both the embryo of Stage 4 (S4) and the red skin of Stage 3 (S3) showed Ca2+ absorbance. These results showed that embryo and red skin of peanut need Ca2+ during development. In order to survey the relationship among calcium, hormone and seed development from gene perspective, we further analyzed the seed transcriptome at Stage 2 (S2), S3, and S4. About 70 million high quality clean reads were generated, which were assembled into 58,147 unigenes. By comparing these three stages, total 4,457 differentially expressed genes were identified. In these genes, 53 Ca2+ related genes, 40 auxin related genes, 15 gibberellin genes, 20 ethylene related genes, 2 abscisic acid related genes, and 7 cytokinin related genes were identified. Additionally, a part of them were validated by qRT-PCR. Most of their expressions changed during the pod development. Since some reports showed that Ca2+ signal transduction pathway is involved in hormone regulation pathway, these results implied that peanut seed development might be regulated by the collaboration of Ca2+ signal transduction pathway and hormone regulation pathway.

The movement and deposition of particles that occur during their natural circulation in supercritical water exercise an important impact on the safe and stable operation of a supercritical water reactor (SCWR). When supercritical water flows in pipelines, a large number of corrosive particles may be generated due to pipeline corrosion or the purity of the fluid itself. The presence of particulate matter affects the heat transfer efficiency of the pipeline, increasing flow resistance and easily promoting heat transfer deterioration. ANSYS-CFX numerical analysis software was used to simulate the natural circulation loop of supercritical water, and micron particles were added in the initial flow field. The effects of heating power, particle concentration and particle diameter on particle deposition were obtained. Through this analysis, it can be concluded that the heating of the pipeline has a certain inhibitory effect on the deposition of particles. The rise in both initial particle concentration and particle diameter serve to reinforce the deposition of particles in the heating section. Depending on the degree of influence, the contributory parameters to particle deposition include particle diameter, particle concentration and heating power in turn.
