Abstract Research background and purpose Soil microorganisms that are closely related to plants are important factors affecting plant health. Therefore, elucidating the abundant and rare bacterial species in soil associated with plant diseases is crucial for understanding ecological processes, maintaining the stability of microecological environments, and formulating microbial strategies that are consistent with modern agricultural development. Results Tea leaf blight leads to an increase in bacterial diversity in the rhizosphere. Random processes dominate the assembly of abundant and rare taxa, while abundant taxa are also influenced by deterministic processes. In the co-occurrence network, the increase in bacterial community diversity mediated by tea cloud leaf blight enhances the stability of the network. Meanwhile, the proportion of positive correlation between rare taxa is relatively high, and the relationship between rare taxa and intermediate taxa is closer. This highly diverse bacteria community maintained the structure and stability of the community to a certain extent. Conclusion The rare taxa in the rhizosphere and the rhizosphere bacterial community mediated by tea leaf blight have high diversity, which is of great significance for maintaining the stability of the rhizosphere bacterial ecological network. In the future, we will further explore the dynamic changes and interaction patterns of the species in the rhizosphere soil affected by tea tree diseases, and their ecological functions and importance in areas of habitat fragmentation. Overall, there are many microbial resources in the rhizosphere microbiota under the influence of plant diseases that can be used for agricultural practice. The results of this study will enrich the insights into ecodynamics of bacteria in karst areas, especially in karst tea gardens.

Rice false smut, caused by Ustilaginoidea virens, is a devastating disease that greatly reduces rice yield and quality. However, controlling rice false smut disease is challenging due to the unique infection mode of U. virens. Therefore, there is a need for early diagnosis and monitoring techniques to prevent the spread of this disease. Lateral flow strip-based recombinase polymerase amplification (LF-RPA) overcomes the limitations of current U. virens detection technologies, which are time-consuming, require delicate equipment, and have a high false-positive rate. In this study, we used a comparative genomics approach to identify Uv_3611, a specific gene of U. virens, as the target for the LF-RPA assay. The designed primers and probe efffectively detected the genomic DNA (gDNA) of U. virens and demonstrated no cross-reactivity with related pathogens. Under optimal conditions, the LF-RPA assay demonstrated a sensitivity of 10 pg of U. virens gDNA. Additionally, by incorporating a simplified PEG-NaOH method for plant DNA extraction, the LF-RPA assay enabled the detection of U. virens in rice spikelets within 30 min, without the need for specialized equipment. Furthermore, the LF-RPA assay successfully detected U. virens in naturally infected rice and seed samples in the field. Therefore, the LF-RPA assay is sensitive, efficient, and convenient, and could be developed as a kit for monitoring rice false smut disease in the field.

Abstract Lanthanum (La(III)) is recognized for its ability to mitigate heavy metal stress in plants. However, the inorganic La(III) salts and lanthanum oxide nanoparticles (La2O3 NPs) extensively used in agriculture are prone to soil immobilization, thereby compromising their bioavailability and posing environmental risks. This study synthesized and characterized the lanthanum(III)-amino acid chelate (La(III)-AA) from soybean protein isolate (SPI) hydrolysates. Maximum chelating rate (94.95%) was achieved under the conditions of mole ratio 1:1.5, pH 8.0, 50 ℃ and 5 h. Glu, Asp and Pro represent the primary La(III)-binding ligands. UV-vis and FTIR demonstrated that amino nitrogen and carboxyl oxygen participate in metal-ligand recognition. Scanning and Transmission electron microscopy showed that La(III) chelates with amino acids in a core-shell structure of uniform size. Consequently, a proposed chemical structure for the La(III)-AA complex was presented. A concentration of 20 mg/L La(III)-AA outperforms inorganic La salts in growth promotion and Cu detoxification. La(III)-AA significantly reduces the content of Cu (II) in rice tissues and enhances seedling tolerance to Cu (II) stress. This study provides a novel La(III)-based candidate for crop protection and furthers our understanding of rare earth element-induced mitigation of heavy metal stress.

Abstract Pitaya (Hylocereus undulatus) is a significant cash crop in the karst region of Southwest China. Ecological stoichiometry is an essential method to research biogeochemical cycles and limiting elements. The purpose of this study was to explore the stoichiometric characteristics of C, N, and P in Karst pitaya orchards and fruit quality and to elucidate the mechanism and process of nutrient cycling. The results showed that: (1) Fruit quality was highest under the combination of chemical and organic fertilizers. Compared to the control, the contents of per-fruit weight, vitamin C, and soluble sugar increased significantly by 55.5%, 60.7%, and 23.0%, respectively, while the content of titratable acidity decreased significantly by 22.0%. (2) The content of soil nutrients under fertilization stress showed a downward trend in general, as did microbial biomass and extracellular enzyme activities. (3) Different fertilization treatments significantly affected the soil-microbial stoichiometry C:N ratio, C:P ratio, with research areas being significantly limited by C and P. (4) Spearman and PLS-SEM (partial least squares-structural equation model) analysis results showed that under the influence of fertilization, there was a significant positive effect between microorganisms and soil nutrients, but a significant negative effect between soil nutrients and quality. The results of this study offer an innovative perspective on pitaya quality research in Karst areas.

Abstract Determining the balance between DNA double strand break repair (DSBR) pathways is essential for understanding treatment response in cancer. We report a method for simultaneously measuring non-homologous end joining (NHEJ), homologous recombination (HR), and microhomology-mediated end joining (MMEJ). Using this method, we show that patient-derived glioblastoma (GBM) samples with acquired temozolomide (TMZ) resistance display elevated HR and MMEJ activity, suggesting that these pathways contribute to treatment resistance. We screen clinically relevant small molecules for DSBR inhibition with the aim of identifying improved GBM combination therapy regimens. We identify the ATM kinase inhibitor, AZD1390, as a potent dual HR/MMEJ inhibitor that suppresses radiation-induced phosphorylation of DSBR proteins, blocks DSB end resection, and enhances the cytotoxic effects of TMZ in treatment-naïve and treatment-resistant GBMs with TP53 mutation. We further show that a combination of G2/M checkpoint deficiency and reliance upon ATM-dependent DSBR renders TP53 mutant GBMs hypersensitive to TMZ/AZD1390 and radiation/AZD1390 combinations. This report identifies ATM-dependent HR and MMEJ as targetable resistance mechanisms in TP53-mutant GBM and establishes an approach for simultaneously measuring multiple DSBR pathways in treatment selection and oncology research.

Abstract Background Colorectal cancer (CRC) is a serious global health burden because of its high morbidity and mortality rates. Hypoxia and massive lactate production are hallmarks of the CRC microenvironment. However, the effects of hypoxia and lactate metabolism on CRC have not been fully elucidated. This study aimed to develop a novel molecular subtyping based on hypoxia-related genes (HRGs) and lactate metabolism-related genes (LMRGs) and construct a signature to predict the prognosis of patients with CRC and treatment efficacy. Methods Bulk and single-cell RNA-sequencing and clinical data of CRC were downloaded from the TCGA and GEO databases. HRGs and LMRGs were obtained from the Molecular Signatures Database. The R software package DESeq2 was used to perform differential expression analysis. Molecular subtyping was performed using unsupervised clustering. A predictive signature was developed using univariate Cox regression, random forest model, LASSO, and multivariate Cox regression analyses. Finally, the sensitivity of tumor cells to chemotherapeutic agents before and after hypoxia was verified using in vitro experiments. Results We classified 575 patients with CRC into three molecular subtypes and were able to distinguish their prognoses clearly. The C1 subtype, which exhibits high levels of hypoxia, has a low proportion of CD8 + T cells and a high proportion of macrophages. The expression of immune checkpoint genes is generally elevated in C1 patients with severe immune dysfunction. Subsequently, we constructed a predictive model, the HLM score, which effectively predicts the prognosis of patients with CRC and the efficacy of immunotherapy. The HLM score was validated in GSE39582, GSE106584, GSE17536, and IMvigor210 datasets. Patients with high HLM scores exhibit high infiltration of CD8 + exhausted T cells (Tex), especially terminal Tex, and oxidative phosphorylation (OXPHOS)−Tex in the immune microenvironment. Finally, in vitro experiments confirmed that CRC cell lines were less sensitive to 5-fluorouracil, oxaliplatin, and irinotecan under hypoxic conditions. Conclusion We constructed novel hypoxia- and lactate metabolism-related molecular subtypes and revealed their immunological and genetic characteristics. We also developed an HLM scoring system that could be used to predict the prognosis and efficacy of immunotherapy in patients with CRC.

The uncoordinated integration of numerous distributed resources poses significant challenges to the safe and stable operation of distribution networks. To address the uncertainties associated with the intermittent output of distributed power sources, we propose a multi-objective planning strategy for distribution networks based on distributionally robust model predictive control (MPC). Initially, an error fuzzy set is established on a Wasserstein sphere using historical data to enhance out-of-sample performance. Next, a multi-objective optimization framework is constructed, balancing returns and risks, and is subsequently converted into a single-objective solution using value-at-risk conditions. This is followed by the implementation of multi-step rolling optimization within the model predictive control framework. We have linearized the proposed model using the linearized power flow method and conducted a thorough validation on an enhanced IEEE 37-node test system. Distributionally robust optimization (DRO) has been shown to reduce costs by a significant 29.16% when compared to an RO method. Moreover, the energy storage capacity required has been notably reduced by 33.33% on the 29-node system and by 20% on the 35-node system. These quantified results not only demonstrate the substantial economic efficiency gains but also the enhanced robustness of our proposed planning under the uncertainties associated with renewable energy integration.

Abstract Nervous system diseases are among the most common diseases globally, posing a severe threat to patients' quality of life and placing a considerable burden on families and society. With improvements in miniaturization, intelligence, and the safety of biosensors, the combination of machinery and organisms is becoming increasingly common. In neuroscience research, biosensors of different macroscopic dimensions have been uniquely utilized to harness their relevant properties. One‐dimensional (1D) biosensors can achieve in situ real‐time monitoring of neural markers at the subcellular, single‐cell, ex vivo, and in vivo levels, with reduced impacts on organisms. Two‐dimensional (2D) biosensors can monitor the chemical behavior of cells and the neural activity of living animals. They are helpful for objectively identifying the characteristics of cells in response to external stimuli and studying the neural circuits of living animals. Three‐dimensional (3D) biosensors have shown unique advantages in point‐of‐care testing, liquid biopsy, drug screening, and mechanistic research. In clinical practice, brain‐computer interfaces (BCIs) and wearable devices have become important tools for monitoring and treatment. To date, there has been widespread adoption of BCIs in clinical practice. BCIs not only exhibit good efficacy in severe neurological and mental diseases but also provide a method for early diagnosis and treatment of these diseases. Wearable sensor devices can accurately assess the symptoms of movement disorders and play an active role in rehabilitation and treatment. In this review, we summarize the application of advanced biosensors in neuroscience research and clinical practice. The challenges and prospects of biosensors as applied to nervous system diseases under interdisciplinary promotion are also discussed in depth.

The fused in sarcoma (FUS) protein is a DNA/RNA binding protein from the ten-eleven translocation protein family that is associated with neurodegeneration, and it has been shown to promote cell proliferation through the growth hormone/insulin-like growth factor 1 (Gh/Igf-1) signaling pathway. The zig-zag eel (Mastacembelus armatus) is a newly discovered species exhibiting sexual dimorphism in growth, and the potential role of fus in the growth and development of this species remains largely unknown. Herein, we analyzed the homology, conserved domains, evolutionary characteristics, and conserved syntenies of fus in several teleost species. The expression of fus was predominant in the brain and exhibited sexual dimorphism in the brain, muscle, and liver of zig-zag eels. We found that microRNA (miR)-146-5p, miR-489-3p, and 24 other miRNAs were targeted to the fus 3′ untranslated region, which might affect muscle and bone development in adults. The igf1, insulin-like growth factor 1 receptor a (igf1ra), insulin-like growth factor 2 receptor (igf2r), growth hormone-releasing hormone-like receptor (ghrhrl), growth hormone secretagogue receptor type 1 (ghsr), and glucocorticoid receptor (gr) genes contained a higher abundance of GU-rich fus motifs compared to the other four genes analyzed in zig-zag eels. We also measured the expression of fus mRNA during fish culture at various stocking densities to further elucidate the relationship between fus expression and the Gh/Igf-1 axis. After 100 days of fish cultivation, the expression of fus and ghrhrl decreased and the expression of ghrh and gr increased as the culture density increased (p < 0.05). The expression of fus exhibited a remarkable positive correlation with a specific growth rate. These results indicate that fus mediates growth differences by regulating the expression of several growth-related genes including Gh/Igf-1 axis genes in zig-zag eels.

This study involves the fabrication and characterization of original, hydrophobic, superhydrophilic, superhydrophobic and hybrid biphilic pattern coupled with the flat and pillar-structured surfaces. The boiling performance of the seven wettability structured surfaces is experimentally and numerically investigated, aiming at analyzing the boiling mechanism of wetting pattern coupled with structures. The findings indicate that coupling superhydrophobicity pillar structure surface (SHO-PS) result in an excellent boiling performance, the superhydrophilicity have difficulty in improving the heat transfer coefficient and critical heat flux. Analysis of the bubble dynamics behaviors reveals a strong positive correlation between the bubble nucleation sites and bubble departure diameter, except for hydrophobic flat structured surface (HO-FS) and superhydrophobicity pillar structured surface (SHO-PS), which exhibit the negative correlation. The bubble departure frequency shows a linear correlation with the contact angle of all the modified surfaces. In the simulation, it is observed that the similar bubble dynamic behaviors in experiment.

Abstract Background Geographical factors affect the nutritional, therapeutic and commercial values of fruits. Dragon fruit (Hylocereus spp) is a popular fruit in Asia and a potential functional food with diverse pharmacological attributes. Although it is produced in various localities, the information related to the altitudinal variation of dragon fruit nutrients and active compounds is scarce. Hence, this study aimed to investigate the variations in metabolite profiles of H. polyrhizus (variety Jindu1) fruit pulps from three different altitudes of China, including Wangmo (WM, 650 m), Luodian (LD, 420 m), and Zhenning (ZN, 356 m). Jindu1 is the main cultivated pitaya variety in Guizhou province, China. Results The LC-MS (liquid chromatography-mass spectroscopy)-based widely targeted metabolic profiling identified 645 metabolites, of which flavonoids (22.64%), lipids (13.80%), phenolic acids (12.40%), amino acids and derivatives (10.39%), alkaloids (8.84%), and organic acids (8.37%) were dominant. Multivariate analyses unveiled that the metabolite profiles of the fruit differed regarding the altitude. Fruits from WM (highest altitude) were prime in quality, with higher levels of flavonoids, alkaloids, nucleotides and derivatives, amino acids and derivatives, and vitamins. Fruits from LD and ZN had the highest relative content of phenolic acids and terpenoids, respectively. We identified 69 significantly differentially accumulated metabolites across the pulps of the fruits from the three locations. KEGG analysis revealed that flavone and flavonol biosynthesis and isoflavonoid biosynthesis were the most differentially regulated. It was noteworthy that most active flavonoid compounds exhibited an increasing accumulation pattern along with the increase in altitude. Vitexin and isovitexin were the major differentially accumulated flavonoids. Furthermore, we identified two potential metabolic biomarkers (vitexin and kaempferol 3-O-[2-O-β-D-galactose-6-O-a-L-rhamnose]-β-D-glucoside) to discriminate between dragon fruits from different geographical origins. Conclusion Our findings provide insights into metabolic changes in dragon fruits grown at different altitudes. Furthermore, they show that growing pitaya at high altitudes can produce fruit with higher levels of bioactive compounds, particularly flavonoids.

Abstract Background This study reported height prediction and longitudinal growth changes in Chinese pediatric patients with acute myeloid leukemia (AML) during and after treatment and their associations with outcomes. Methods Changes in 88 children with AML in percentages according to the growth percentile curve for Chinese boys/girls aged 2–18/0–2 years for body mass index (BMI), height, and weight from the time of diagnosis to 2 years off therapy were evaluated. The outcomes of AML were compared among patients with different BMI levels. Results The proportion of underweight children (weight 75th percentile) after 2 years of drug withdrawal. Unhealthy BMI at the initial diagnosis and during intensive chemotherapy leads to poorer outcomes. For height, all patients were in the range of genetic height predicted based on their parents’ height at final follow-up. Conclusions Physicians should pay more attention to the changes in height and weight of children with AML at these crucial treatment stages and intervene in time.

Abstract Clinical ulcerative colitis (UC) is a heterogeneous condition. Moreover, medical interventions are nonspecific, and thus, treatment responses are inconsistent. The aim of this study was to explore the molecular subtypes and biological characteristics of UC based on ferroptosis and neutrophil gene sets. Multiple intestinal mucosa gene expression profiles of UC patients in the Gene Expression Omnibus (GEO) database were downloaded. Unsupervised clustering methods were used to identify potential molecular subtypes based on ferroptosis and neutrophil gene sets. Multiple immune infiltration algorithms were used to evaluate the biological characteristics of the molecular subtypes. Machine learning identifies hub genes for molecular subtypes and analyses their diagnostic efficacy for UC and predictive performance for drug therapy. The relevant conclusions were verified by clinical samples and animal experiments. Four molecular subtypes were identified according to the ferroptosis and neutrophil gene sets: neutrophil, ferroptosis, mixed and quiescent. The subtypes have different biological characteristics and immune infiltration levels. Multiple machine learning methods jointly identified four hub genes (FTH1, AQP9, STEAP3 and STEAP4). Receiver operating characteristic (ROC) curve analysis revealed that the four hub genes could be used as diagnostic markers for UC. The clinical response profile data of infliximab treatment patients showed that AQP9 and STEPA4 were reliable predictors of infliximab treatment response. In human samples the AQP9 and STEAP4 protein were shown to be increased in UC intestinal samples. In animal experiments, the ferroptosis and neutrophil phenotype were confirmed. Dual analysis of ferroptosis and neutrophil gene expression revealed four subgroups of UC patients. The molecular subtype-associated hub genes can be used as diagnostic markers for UC and predict infliximab treatment response.

Abstract Grinding, a critical precision machining process for difficult-to-cut alloys, has undergone continual technological advancements to improve machining efficiency. However, the sustainability of this process is gaining heightened attention due to significant challenges associated with the substantial specific grinding energy and the extensive heat generated when working with difficult-to-cut alloys, renowned for their exceptional physical and mechanical properties. In response to these challenges, the widespread application of massive coolant in manufacturing industries to dissipate grinding heat has led to complex post-cleaning and disposal processes. This, in turn, has resulted in issues such as large energy consumption, a considerable carbon footprint, and concerns related to worker health and safety, which have become the main factors that restrict the development of grinding technology. This paper provides a holistic review of sustainability in grinding difficult-to-cut alloys, encompassing current trends and future directions. The examination extends to developing grinding technologies explicitly tailored for these alloys, comprehensively evaluating their sustainability performance. Additionally, the exploration delves into innovative sustainable technologies, such as heat pipe/oscillating heat pipe grinding wheels, minimum quantity lubrication, cryogenic cooling, and others. These groundbreaking technologies aim to reduce dependence on hazardous coolants, minimizing energy and resource consumption and carbon emissions associated with coolant-related or subsequent disposal processes. The essence of these technologies lies in their potential to revolutionize traditional grinding practices, presenting environmentally friendly alternatives. Finally, future development trends and research directions are put forward to pursue the current limitation of sustainable grinding for difficult-to-cut alloys. This paper can guide future research and development efforts toward more environmentally friendly grinding operations by understanding the current state of sustainable grinding and identifying emerging trends.

Abstract Knowing the impacts of global climate change on the habitat suitability distribution of Limassolla leafhoppers contributes to understanding the feedback of organisms on climate change from a macroecological perspective, and provides important scientific basis for protecting the ecological environment and biodiversity. However, there is limited knowledge on this aspect. Thus, our study aimed to address this gap by analyzing Asian habitat suitability and centroid shifts of Limassolla based on 19 bioclimatic variables and occurrence records. Selecting five ecological niche models with the outstanding predictive performance (Maxlike, generalized linear model, generalized additive model, random forest, and maximum entropy) along with their ensemble model from 12 models, the current habitat suitability of Limassolla and its future habitat suitability under two Shared Socio‐economic Pathways (SSP1‐2.6 and SSP5‐8.5) in the 2050s and 2090s were predicted. The results showed that the prediction results of the five models are generally consistent. Based on ensemble model, 11 potential biodiversity hotspots with high suitability were identified. With climate change, the suitable range of Limassolla will experience both expansion and contraction. In SSP5‐8.52050s, the expansion area is 118.56 × 104 km2, while the contraction area is 25.40 × 104 km2; in SSP1‐2.62090s, the expansion area is 91.71 × 104 km2, and the contraction area is 26.54 × 104 km2. Furthermore, the distribution core of Limassolla will shift toward higher latitudes in the northeast direction, and the precipitation of warmest quarter was found to have the greatest impact on the distribution of Limassolla. Our research results supported our four hypotheses. Finally, this research suggests establishing ecological reserves in identified contraction to prevent habitat loss, enhancing the protection of biodiversity hotspots, and pursuing a sustainable development path with reduced emissions.

Background Hepatocellular carcinoma (HCC) is an aggressive malignancy with limited effective treatment options. Phenethyl isothiocyanate (PEITC) is a bioactive substance present primarily in the cruciferous vegetables. PEITC has exhibited anti-cancer properties in various cancers, including lung, bile duct, and prostate cancers. It has been demonstrated that PEITC can inhibit the proliferation, invasion, and metastasis of SK-Hep1 cells, while effectively inducing apoptosis and cell cycle arrest in HepG2 cells. However, knowledge of its anti-carcinogenic effects on Huh7.5.1 cells and its underlying mechanism remains elusive. In the present study, we aim to evaluate the anti-carcinogenic effects of PEITC on human HCC Huh7.5.1 cells. Methods MTT assay and colony formation assay was performed to investigate the anti-proliferative effects of PEITC against Huh7.5.1 cells. The pro-apoptosis effects of PEITC were determined by Annexin V-FITC/PI double staining assay by flow cytometry (FCM), mitochondrial transmembrane potential (MMP) measurement, and Caspase-3 activity detection. A DAPI staining and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL) assay was conducted to estimate the DNA damage in Huh7.5.1 cells induced by PEITC. Cell cycle progression was determined by FCM. Transwell invasion assay and wound healing migration assay were performed to investigate the impact of PEITC on the migration and invasion of Huh7.5.1 cells. In addition, transcriptome sequencing and gene set enrichment analysis (GSEA) were used to explore the potential molecular mechanisms of the inhibitory effects of PEITC on HCC. Quantitative real-time PCR (qRT-PCR) analysis was performed to verify the transcriptome data. Results MTT assay showed that treatment of Huh7.5.1 cells with PEITC resulted in a dose-dependent decrease in viability, and colony formation assay further confirmed its anti-proliferative effect. Furthermore, we found that PEITC could induce mitochondrial-related apoptotic responses, including a decrease of mitochondrial transmembrane potential, activation of Caspase-3 activity, and generation of intracellular reactive oxygen species. It was also observed that PEITC caused DNA damage and cell cycle arrest in the S-phase in Huh7.5.1 cells. In addition, the inhibitory effect of PEITC on the migration and invasion ability of Huh7.5.1 cells was assessed. Transcriptome sequencing analysis further suggested that PEITC could activate the typical MAPK, PI3K-Akt, and p53 signaling pathways, revealing the potential mechanism of PEITC in inhibiting the carcinogenic properties of Huh7.5.1 cells. Conclusion PEITC exhibits anti-carcinogenic activities against human HCC Huh7.5.1 cells by activating MAPK/PI3K-Akt/p53 signaling pathways. Our results suggest that PEITC may be useful for the anti-HCC treatment.

Li7La3Zr2O12 (LLZO) is considered as a promising solid-state electrolyte due to its high ionic conductivity, wide electrochemical window, and excellent electrochemical stability. However, its application in solid-state lithium metal batteries (SSLMBs) is impeded by the growth of lithium dendrites in LLZO due to some reasons such as its high electronic conductivity. In this study, lithium fluoride (LiF) was introduced into Ta-doped LLZO (LLZTO) to modify its grain boundaries to enhance the performance of SSLMBs. A nanoscale LiF layer was uniformly coated on the LLZTO grains, creating a three-dimensional continuous electron-blocking network at the grain boundaries. Benefiting from the electronic insulator LiF and the special structure of the modified LLZTO, the symmetric cells based on LLZO achieved a high critical current density (CCD) of 1.1 mA·cm−2 (in capacity-constant mode) and maintained stability over 2000 h at 0.3 mA·cm−2. Moreover, the full cells combined with a LiFePO4 (LFP) cathode, demonstrated excellent cycling performance, retaining 97.1% of capacity retention after 500 cycles at 0.5 C. Abstract Background Ovarian cancer is one of the most common gynecological malignancies due to the lack of early symptoms, early diagnosis and limited screening. Therefore, it is necessary to understand the molecular mechanism underlying the occurrence and progression of ovarian cancer and to identify a basic biomarker for the early diagnosis and clinical treatment of ovarian cancer. Methods The association between FBXO28 and ovarian cancer prognosis was analyzed using Kaplan‒Meier survival analysis. The difference in FBXO28 mRNA expression between normal ovarian tissues and ovarian tumor tissues was obtained from The Cancer Genome Atlas (TCGA), and Genotype-Tissue Expression (GTEx) cohorts. The expression levels of the FBXO28 protein in ovarian cancer tissues and normal ovarian tissues were measured via immunohistochemical staining. Western blotting was used to determine the level of FBXO28 expression in ovarian cancer cells. The CCK-8, the colony formation, Transwell migration and invasion assays were performed to evaluate cell proliferation and motility. Results We found that a higher expression level of FBXO28 was associated with poor prognosis in ovarian cancer patients. Analysis of the TCGA and GTEx cohorts showed that the FBXO28 mRNA level was lower in normal ovarian tissue samples than in ovarian cancer tissue samples. Compared with that in normal ovarian tissues or cell lines, the expression of FBXO28 was greater in ovarian tumor tissues or tumor cells. The upregulation of FBXO28 promoted the viability, proliferation, migration and invasion of ovarian cancer cells. Finally, we demonstrated that FBXO28 activated the TGF-beta1/Smad2/3 signaling pathway in ovarian cancer. Conclusions In conclusion, FBXO28 enhanced oncogenic function via upregulation of the TGF-beta1/Smad2/3 signaling pathway in ovarian cancer.

Abstract Metastasis is the leading cause of high mortality in colorectal cancer (CRC), which is not only driven by changes occurring within the tumor cells, but is also influenced by the dynamic interaction between cancer cells and components in the tumor microenvironment (TME). Currently, the exploration of TME remodeling and its impact on CRC metastasis has attracted increasing attention owing to its potential to uncover novel therapeutic avenues. Noteworthy, emerging studies suggested that tumor-associated macrophages (TAMs) within the TME played important roles in CRC metastasis by secreting a variety of cytokines, chemokines, growth factors and proteases. Moreover, TAMs are often associated with poor prognosis and drug resistance, making them promising targets for CRC therapy. Given the prognostic and clinical value of TAMs, this review provides an updated overview on the origin, polarization and function of TAMs, and discusses the mechanisms by which TAMs promote the metastatic cascade of CRC. Potential TAM-targeting techniques for personalized theranostics of metastatic CRC are emphasized. Finally, future perspectives and challenges for translational applications of TAMs in CRC development and metastasis are proposed to help develop novel TAM-based strategies for CRC precision medicine and holistic healthcare.

Objective To investigate the role of autophagy in contrast-induced acute kidney injury (CI-AKI) in rats and to explore its possible mechanism. Methods SD rats were randomly divided into control group, acute kidney injury model group (intravenous injection of contrast medium ioversol via tail vein; model), rapamycin(RAPA) group and hydroxychloroquine (HCQ)group. Blood urea nitrogen(BUN) and serum creatinine(Scr) contents were measured and the potential change foun in renal pathology was detected by HE staining and microscopy. Transmission electron microscopy was used to observe auto-phagy-related changes in ultrastructure.Western blot was used to observe the expression of microtubule-associated protein 1 light chain 3(LC3)Ⅱ/LC3Ⅰ,ubiquitin-binding protein p62 and Histone deacetylase 4(HDAC4). The expression of HDAC4 was also observed by RT-qPCR. Results Compared with control group, the level of BUN, Scr and HDAC4 expression in the model and HCQ group was increased (P＜0.01),the proximal tubules of the kidney were significantly damaged. In the model group, autophagososomes and autolysosomes increased, accompanied by an increase of LC3Ⅱ/LC3Ⅰ and a decrease in the p62 level (P＜0.05,P＜0.01); Compared with model group, there were more autophagosomes and autolysosomes were found in RAPA group(P＜0.01), accompanied by increased LC3Ⅱ/LC3Ⅰ ratio and decrease in the p62 and HDAC4(P＜0.05,P＜0.01). In contrast, the number of autophagy related structures decreased in HCQ group(P＜0.01), accompanied by the simultaneous increase of LC3Ⅱ/LC3Ⅰ, p62 and the increase of HDAC4(P＜0.01). Conclusions Ioversol may induce autophagy activation, while enhancing autophagy by RAPA alleviates CI-AKI induced renal dysfunction. The mechanism is potentially atributed to the regulation of HDAC4.

Abstract Under natural conditions, most Hibiscus syriacus L. individuals form very few mature seeds or the mature seeds that do form are of poor quality. As a result, seed yield is poor and seeds have low natural germinability. These phenomena strongly hinder utilization of the excellent germplasm resources of H. syriacus. The study has shown that pollen activity and stigma receptivity were high on the day of anthesis, and the pistils and stamens were fertile. Pollen release and stigma receptivity were synchronous. But in styles following self and cross-pollination, pollen tube abnormalities (distortion and twisting of the pollen tubes) and callose deposition were observed. Cross-pollinated pollen tubes elongated faster and fewer pollen tube abnormalities were observed compared with self-pollinated pollen tubes. And during embryo development, abnormalities during the heart-shaped embryo stage led to embryo abortion. Imbalance in antioxidant enzyme activities and low contents of auxin and cytokinin during early stages of embryo development may affect embryo development. Therefore, a low frequency of outcrossing and mid-development embryo abortion may be important developmental causes of H. syriacus seed abortion. Nutrient deficiencies, imbalance in antioxidant enzyme activities, and a high content of abscisic acid at advanced stages of seed development may be physiological causes of seed abortion.

Abstract. Background:. Type 2 diabetes mellitus (T2DM) is an independent risk factor for colorectal cancer (CRC), and the patients with CRC and T2DM have worse survival. The human gut microbiota (GM) is linked to the development of CRC and T2DM, respectively. However, the GM characteristics in patients with CRC and T2DM remain unclear. Methods:. We performed fecal metagenomic and targeted metabolomics studies on 36 samples from CRC patients with T2DM (DCRC group, n = 12), CRC patients without diabetes (CRC group, n = 12), and healthy controls (Health group, n = 12). We analyzed the fecal microbiomes, characterized the composition and function based on the metagenomics of DCRC patients, and detected the short-chain fatty acids (SCFAs) and bile acids (BAs) levels in all fecal samples. Finally, we performed a correlation analysis of the differential bacteria and metabolites between different groups. Results:. Compared with the CRC group, LefSe analysis showed that there is a specific GM community in DCRC group, including an increased abundance of Eggerthella, Hungatella, Peptostreptococcus, and Parvimonas, and decreased Butyricicoccus, Lactobacillus, and Paraprevotella. The metabolomics analysis results revealed that the butyric acid level was lower but the deoxycholic acid and 12-keto-lithocholic acid levels were higher in the DCRC group than other groups (P < 0.05). The correlation analysis showed that the dominant bacterial abundance in the DCRC group (Parvimonas, Desulfurispora, Sebaldella, and Veillonellales, among others) was negatively correlated with butyric acid, hyodeoxycholic acid, ursodeoxycholic acid, glycochenodeoxycholic acid, chenodeoxycholic acid, cholic acid and glycocholate. However, the abundance of mostly inferior bacteria was positively correlated with these metabolic acid levels, including Faecalibacterium, Thermococci, and Cellulophaga. Conclusions:. Unique fecal microbiome signatures exist in CRC patients with T2DM compared to those with non-diabetic CRC. Alterations in GM composition and SCFAs and secondary BAs levels may promote CRC development.

Shengnongjia Forestry District is the national natural reserve with abundant biological resources in China. Monitoring its variations of vegetation for the intimate connection with eco-environmental changes is of great significance. In this paper, the 16-day composite MODIS normalized difference vegetation index (NDVI) products (MOD13A1) with 500 m resolution from 2011 to 2020 were selected to investigate the vegetation dynamics in Shengnongjia Forestry District. To alleviate the cloud contamination in NDVI products, a spatio-temporal prefill method with harmonic analysis of time series (ST-HANTS) is proposed. ST-HANTS first prefills the raw NDVI time series using spatio-temporal information to reduce data gaps, which effectively improves the reconstruction performance of the subsequent HANTS algorithm. In the simulation experiments, ST-HANTS shows the highest average correlation coefficient and the lowest average root mean square error compared with other mainstream methods, including HANTS, Savitzky–Golay filter, wavelet transform, and data assimilation. The reconstruction curves are close to the upper envelope of the NDVI time series, which is more consistent with the vegetation phenology and can effectively capture the key points in the growth cycle. By analyzing the cloud-free NDVI time series reconstructed with ST-HANTS, results reveal the overall trend of Shennongjia vegetation coverage is high in the middle while low at the edge. Except for the population centers and Hongping Airport, the NDVI of most areas is above 0.7 and shows a remarkable increasing tendency. Moreover, the fluctuation degree of NDVI in the whole study area is low, indicating that the ecological environment of Shennongjia is relatively stable. The vegetation variations are influenced by land surface temperature and precipitation, and the vegetation growth response to precipitation exhibits an apparent hysteresis.

Abstract Temporal and spatial variations in the ocean surface mixed layer are important for the climate and ecological systems. During 1980–2019, the Southern Indian Ocean (SIO) mixed layer depth (MLD) displays a basin‐wide shoaling trend that is absent in the other basins within 40°S–40°N. The SIO MLD shoaling is mostly prominent in austral winter with deep climatology MLD, substantially weakening the MLD seasonality. Moreover, the SIO MLD changes are primarily caused by a southward shift of the subtropical anticyclonic winds and hence ocean gyre, associated with a strengthening of the Southern Annular Mode, in recent decades for both winter and summer. However, the poleward‐shifted subtropical ocean circulation preferentially shoals the SIO MLD in winter when the meridional MLD gradient is sharp but not in summer when the gradient is flat. This highlights the distinct subtropical MLD response to meridional mitigation in winds due to different background oceanic conditions across seasons.

Abstract This work aims to explore the long‐term prognosis of hepatitis B virus‐related acute‐on‐chronic liver failure (HBV‐ACLF). In this prospective study, eligible inpatients with HBV‐ACLF are enrolled and followed up from December 2012 to February 2023, for clinical events, laboratory tests at least every 6 months. Overall, the survival rates at 28 days, 90 days, 1 year, 5 years, and 8 years are 64.7%, 48.8%, 46.1%, 43.8%, and 42.2%, respectively. Among the 8‐year mortality and liver transplant cases, ACLF survivors (who survived over 90 days) accounted for 7.8% (9/115). Among 101 patients who survived for more than 90 days, 97.9% of patients achieve virologic response at 1 year. For HBeAg‐positive patients, the HBeAg seroconversion are 25.5%, 63.6%, and 76.9% at 1, 5, and 8 years, respectively. Alanine aminotransferase, aspartate aminotransferase, total bilirubin, INR, white blood cell count, and albumin levels gradually improve within the first year. Fibrosis biomarkers APRI, FIB‐4 and Chitinase‐3‐like protein 1 (CHI3L1) levels decreases within the first 5 years. The Cox proportional hazards regression reveal that high total bilirubin (HR = 1.008, p = 0.021) is the independent risk factor for 8‐year survival of ALCF survivors. The 90‐day period following of HBV‐ACLF represented a critical juncture for long‐term prognosis, revealing favorable outcomes beyond this timeframe.

Summary: Hexanucleotide repeat expansions in the C9orf72 gene are the most common cause of amyotrophic lateral sclerosis (ALS) and frontotemporal dementia. Due to the lack of trunk neuromuscular organoids (NMOs) from ALS patients’ induced pluripotent stem cells (iPSCs), an organoid system was missing to model the trunk spinal neuromuscular neurodegeneration. With the C9orf72 ALS patient-derived iPSCs and isogenic controls, we used an NMO system containing trunk spinal cord neural and peripheral muscular tissues to show that the ALS NMOs could model peripheral defects in ALS, including contraction weakness, neural denervation, and loss of Schwann cells. The neurons and astrocytes in ALS NMOs manifested the RNA foci and dipeptide repeat proteins. Acute treatment with the unfolded protein response inhibitor GSK2606414 increased the glutamatergic muscular contraction 2-fold and reduced the dipeptide repeat protein aggregation and autophagy. This study provides an organoid system for spinal neuromuscular pathologies in ALS and its application for drug testing.

Since 2011, the Gyrovirus galga 1 (GyVg1, previously recognized as avian gyrovirus 2) strain has extensively been detected worldwide. However, because there are no up-to-date reports of examining the distribution of GyVg1 in flocks in southern China, the epidemiology of this virus is unknown. To investigate the prevalence and genetic evolution of GyVg1, a total of 2,077 field samples collected from 113 chicken farms in 6 provinces in southern China during 2020 to 2022 were tested. Among them, 315 samples (315/2,077, 15.17%) were positive for GyVg1 by PCR. The positive rate of GyVg1 detection between different regions of southern China ranged from 11.69% (Guangdong) to 22.46% (Yunnan). The correlation between GyVg1 prevalence and sample source groups was analyzed, the results showing that the highest seroprevalence of GyVg1 was observed in visceral tissues (27.34%, 187/684), significantly higher (P < 0.05) than that of feather shafts (17.22%, 31/180), serums (8.85%, 78/881), and fecal (5.72%, 19/332). Additionally, the complete genomes of 10 GyVg1 strains were sequenced and analyzed, which showed nucleotide identities of 96.2 to 99.9%, 97.0 to 100.0%, 95.2 to 100.0%, and 95.7 to 99.8% in the complete genome, ORF1, ORF2, and ORF3, respectively, and 94.4 to 100.0%, 91.3 to 100.0%, and 98.7 to 100.0% amino acid similarity in the VP2, VP3, and VP1 proteins, respectively. Phylogenetic analysis of the whole genome showed that 10 GyVg1 strains belong to genotype I, and one strain belongs to genotype III. Sequence analysis showed several amino acid substitutions in both the VP1, VP2, and VP3 proteins. Our results enhance the understanding of the molecular characterization of GyVg1 infection in southern China. In conclusion, this study reveals the high prevalence and high genetic differentiation of GyVg1 in Chinese chickens and suggests that the potential impact of GyVg1 on the chicken industry may be of concern.

Background: Increasing evidence has shown a link between immune cells and Alzheimer's disease (AD). Comprehensive two-sample Mendelian randomization (MR) analysis was performed to determine the causal association between 731 immune cell signatures and AD in this study. Methods: We extracted genetic variants of 731 immune cell traits and AD from the publicly available GWAS dataset. The immune features included median fluorescence intensity (MFI), relative cellular (RC), absolute cellular (AC) and morphological parameters (MP). The inverse variance weighted (IVW) method was the main MR analysis method, and sensitivity analyses were used to validate the robustness, heterogeneity and horizontal pleiotropy of the results. Results: After FDR adjustment, seven immune phenotypes were found to be associated significantly with AD risk: HLA DR on CD33− HLA DR+ (OR = 0.938, PFDR = 0.001), Secreting Treg %CD4 (OR = 0.972, PFDR = 0.021), HLA DR+ T cell AC (OR = 0.928, PFDR = 0.041), Activated & resting Treg % CD4 Treg (OR = 1.031, PFDR = 0.002), CD33 on CD33dim HLA DR+ CD11b+ (OR = 1.025, PFDR = 0.025), CD33 on CD14+ monocyte (OR = 1.026, PFDR = 0.027) and CD33 on CD66b++ myeloid cell (OR = 1.027, PFDR = 0.036). Conclusions: These findings demonstrated seven immune phenotypes were significantly associated with AD risk. This may provide researchers with a new perspective in exploring the biological mechanisms of AD and may lead to the exploration of earlier treatment.

Abstract Background Osteosarcoma has the highest incidence among bone malignant tumors and mainly occurs in adolescents and the elderly, but the pathological mechanism is still unclear, which makes early diagnosis and treatment very difficult. Bone marrow mesenchymal stem cells (BMSCs) are considered to be one of the sources of osteosarcoma cells. Therefore, a full understanding of the gene expression differences between BMSCs and osteosarcoma cells is very important to explore the pathogenesis of osteosarcoma and facilitate the early diagnosis and treatment of osteosarcoma. Small noncoding RNAs (sncRNAs) are a class of RNAs that do not encode proteins but directly play biological functions at the RNA level. SncRNAs mainly include Piwi-interacting RNAs (piRNAs), small nucleolar RNAs (snoRNAs), small nuclear RNAs (snRNAs), repeat RNAs and microRNAs (miRNAs). Methods In this study, we compared the expression of sncRNAs in BMSCs and osteosarcoma cells by high-throughput sequencing and qPCR and looked for differentially expressed sncRNAs. CCK-8, clone formation and transwell assay were used to detect the effect of sncRNA in MG63 cells. Results We found that 66 piRNAs were significantly upregulated and 70 piRNAs were significantly downregulated in MG63 cells. As for snoRNAs, 71 snoRNAs were significantly upregulated and 117 snoRNAs were significantly downregulated in MG63 cells. As for snRNAs, 35 snRNAs were significantly upregulated and 17 snRNAs were significantly downregulated in MG63 cells. As for repeat RNAs, 6 repeat RNAs were significantly upregulated and 7 repeat RNAs were significantly downregulated in MG63 cells. As for miRNAs, 326 miRNAs were significantly upregulated and 281 miRNAs were significantly downregulated in MG63 cells. Overexpression of piRNA DQ596225, snoRNA ENST00000364830.2, snRNA ENST00000410533.1 and miRNA hsa-miR-369-5p inhibited the proliferation and migration of MG63 cells. Conclusions Our results provide a theoretical basis for the pathogenesis, early diagnosis and treatment of osteosarcoma.

Abstract Objective The aim of this study was to understand the prevalence and potential risk factors of Helicobacter pylori (H. pylori) infection in Hainan Province, China. Methods We conducted this study in 21 health service stations in 5 cities of Hainan Province from August 2022 to April 2023. We selected the various participants based on a stratified whole-group sampling method. The 14C-UBT was used to analyze H. pylori infection in 3632 participants. We also analyzed the possible relationship between variables and H. pylori infection based on chi-square test and multifactorial logistic regression. The model was evaluated by performing a Hosmer–Lemeshow goodness-of-fit test and plotting receiver operating characteristic(ROC) curves. Results In total, the results of 3632 eligible participants (age: 14 to 93 years) were included in the analysis. The total prevalence of H. pylori infection in Hainan Province was approximately 38.7%. The prevalence of H. pylori infection was found to increase with age, stabilized in the age group of 45 to 64 years, but peaked in the age group of 65 years and older. In multifactorial analysis, the prevalence of H. pylori infection was positively associated with middle-aged adults (45–64 years), older adults (≥ 65 years), drinking, farmers, natural labor, routinely share utensils, have habit of frequent betel nut consumption, upper gastrointestinal symptoms, and family history of gastric cancer. The factors negatively associated with prevalence included family size ≤ 3, washing hands often before meals, frequent exercise, regular meals, and frequent consumption of fruits and vegetables. In addition, the Hosmer–Lemeshow test showed a good fit (χ2 = 12.983, P = 0.112) and the area under ROC was 0.631 (95%CI: 0.613 ~ 0.649). Conclusion The prevalence of H. pylori infection in Hainan Province was observed to be moderate and closely related to age, local socioeconomic conditions, hygienic status and dietary habits.

The aim of this study is to assess the relationship of leptin (LEP) and adiponectin (ADPN) with other circulating fat markers, physical capacity, behaviors, and anthropometric indices in a population of overweight and obese Chinese university students. LEP and ADPN levels, as well as behavioral, anthropometric, biochemical, and performance characteristics, were measured. Method: A total of 17 anthropometric parameters, 8 questionnaires (investigating quality of life, sleep, eating, perceived functioning, stress, and depression), 9 biochemical parameters, and 12 functional parameters were investigated. Results: In contrast to previous studies, our work found an unusually strong relationship between LEP and ADPN (r = 0.961, p = 0.000) that can be related to ethnicity. We also found that LEP and ADPN were associated with stress and bodily pain. A total of 12 anthropometric measures were also associated with LEP/ADNP levels. Moreover, LEP and ADPN were found to be related to lower limb, hand, and abdominal strength; blood pressure; and basic metabolism. However, we did not find associations with sleep; eating habits; or cardiovascular fitness, which was measured in the form of resting heart rate and VO2max. Conclusion: This study reveals new relationships of LEP and ADPN with selected anthropometric and behavioral parameters in obese Chinese college students.

In the field of risk assessment, the traditional econometric models are generally used to assess credit risk. And with the introduction of the “dual-carbon” goals to promote the development of a low-carbon economy, the scale of green credit in China has rapidly expanded. But with the advent of the big data era, due to the poor interpretability of a traditional single machine learning model, it is difficult to capture nonlinear relationships, and there are shortcomings in prediction accuracy and robustness. This paper selects the adjusted ensemble learning model based on the homogeneous and heterogeneous factors for user default prediction, which can efficiently process large quantities of high-dimensional data. This article adjusts each model to adapt to the task and innovatively compares various models. In this paper, the missing value filling method, feature selection, and ensemble model are studied and discussed, and the optimal ensemble model is obtained. When comparing the predictions of single models and ensemble models, the accuracy, sensitivity, specificity, F1-Score, Kappa, and MCC of Categorical Features Gradient Boosting (CatBoost) and Random undersampling Boosting (RUSBoost) all reach 100%. The experimental results prove that the algorithm based on adjusted homogeneous and heterogeneous ensemble learning can predict the user default efficiently and accurately. This paper also provides some references for establishing a risk assessment index system.

BackgroundMucinous adenocarcinoma (MAC) is a unique subtype of colorectal cancer and its prognostic value remains controversial. This study aimed to compare the clinicopathological characteristics and prognostic differences between patients with MAC and non-mucinous adenocarcinoma (NMAC).Methods674 patients with NMAC, 110 patients with adenocarcinoma with mucinous component (ACWM) and 77 patients with MAC between 2016-2019 were enrolled in the study. Univariate and multivariate Cox regression were performed to analyze the factors associated with prognosis. Predictive nomograms of overall survival (OS) and cancer-specific survival (CSS) for patients with colorectal adenocarcinoma were constructed. Confounding factors were eliminated by propensity score matching (PSM).ResultsCompared with patients with NMAC, patients with MAC were more likely to have a tumor located at the proximal colon, present with a larger tumor diameter, more advanced T stage, higher frequency of metastasis, deficiency of mismatch repair, and elevated preoperative carcinoembryonic antigen. Background. Observational studies have suggested an association between inflammatory cytokines and Parkinson’s disease (PD). This Mendelian randomization (MR) was conducted to further assess the causal correlations between inflammatory cytokines and PD. Methods. Genetic instruments associated with inflammatory cytokines were extracted from a large summary genome-wide association studies (GWAS) involving 8,293 European participants. Summary-level statistics for PD were obtained from a large-sample GWAS containing 17 studies that involved European participants. Causalities of exposures and outcomes were explored mainly using inverse variance weighted (IVW) method. Results. The IVW method indicated that basic fibroblast growth factor (FGFBasic), interleukin-2 (IL-2), and macrophage migration inhibitory factor (MIF) may be suggestively associated with the risk of PD (OR: 0.71, 95%CI: 0.52–0.96, P = 0.027; OR: 1.18, 95%CI: 1.01–1.38, P = 0.041; and OR: 1.23, 95%CI: 1.04–1.46, P = 0.018). In the reverse direction, monokine induced by interferon gamma (MIG), beta nerve growth factor (bNGF), interleukin-17 (IL-17), and interferon gamma (IFNg) are suggested to be the consequences of PD. Conclusion. Our MR analysis indicated that suggestive associations between circulating levels of FGFBasic, IL-2, and MIF and PD risk. In addition, MIG, bNGF, IL-17, and IFNg are more likely to be involved in the development of downstream PD.

Highly productive and efficient biomass growth in bioreactors is an essential bioprocess outcome in many industrial applications. Large-scale biomass creation in the cultivated meat industry will be critical given the demand size in the conventional meat and seafood sectors. However, many challenges must be overcome before cultivated meat and seafood become commercially viable, including cost reductions of cell culture media, bioprocess design innovation and optimization, and scaling up in the longer term. Computational modeling and simulation can help to address many of these challenges and can be a far cheaper and faster alternative to performing physical experiments. Computer modeling can also help researchers pinpoint system interactions that matter and guide researchers to identify those parameters that should be changed in later designs for eventual optimization. This work developed a computational model that combines agent-based modeling and computational fluid dynamics to study biomass growth as a function of the operative conditions of stirred-tank bioreactors. The focus was to analyze how the mechanical stress induced by rotor speed can influence the growth of cells attached to spherical microcarriers. The computer simulation results reproduced observations from physical experiments that high rotor speeds reduce cell growth rates and induce cell death under the high mechanical stresses induced at these stir speeds. Moreover, the results suggest that modeling cell death and cell quiescence is required to recapitulate these observations from physical experiments. These simulation outcomes are the first step towards more comprehensive models that, combined with experimental observations, will improve our knowledge of biomass production in bioreactors for cultivated meat and other industries.

Abstract Malignant tumors pose a serious risk to human health. Ascorbic acid (AA) has potential for tumor therapy; however, the mechanism underlying the ability of AA to selectively kill tumor cells remains unclear. AA can cause redox disequilibrium in tumor cells, resulting in the release of abundant reactive oxygen species, represented by hydrogen peroxide (H2O2). Therefore, the detection of H2O2 changes can provide insight into the selective killing mechanism of AA against tumor cells. In this work, inspired by the ion‐exchange mechanism in coral formation, a flexible H2O2 sensor (PtNFs/CoPi@CC) is constructed to monitor the dynamics of H2O2 in the cell microenvironment, which exhibits excellent sensitivity and spatiotemporal resolution. Moreover, the findings suggest that dehydroascorbic acid (DHA), the oxidation product of AA, is highly possible the substance that actually acts on tumor cells in AA therapy. Additionally, the intracellular redox disequilibrium and H2O2 release caused by DHA are positively correlated with the abundance and activity of glucose transporter 1 (GLUT1). In conclusion, this work has revealed the potential mechanism underlying the ability of AA to selectively kill tumor cells through the construction and use of PtNFs/CoPi@CC. The findings provide new insights into the clinical application of AA.

The volume fraction and rafting degree of the γʹ-Ni3Al phase under stress and high temperature are the key characteristics of mechanical properties in Ni-based superalloys, the rafting and redissolution of γʹ phase caused by the creep at high temperature damage the morphology and properties of Ni-based superalloys. The phase-field simulation is performed to study the rafting accompany with the redissolution of γʹ phase under high temperature and loading stress in Ni–Al alloy, the driving force and kinetics evolution of the γʹ rafting were revealed. During the rafting under continuous heating, the elastic energy in the vertical γ channel is different to that of the horizontal γ channel, this difference in elastic energy drives the elements diffusion directionally to form the γʹ rafts morphology. With the increased tensile stress, the decrease of specific surface of the γʹ phase slows down the redissolution, a higher volume fraction is reserved for the rafted γʹ phase. With temperature increases, the interface of γ/γʹ phase becomes more diffusional and wider under stress. The results give an insight on the rafting mechanism of γʹ phase and the kinetics evolution in Ni-based superalloys under excess temperature.

The present study aimed to evaluate the perinatal outcomes and influencing factors in twin pregnancies undergoing emergency cervical cerclage. The present retrospective cohort study included clinical data that were recorded between January 2015 and December 2021 at The Second Affiliated Hospital and Yuying Children’s Hospital of Wenzhou Medical University (China). The study included data from 103 pregnancies (26 twin and 77 singleton pregnancies) that underwent emergency cerclage and 17 twin pregnancies that underwent expectant treatment. The median gestational age of twin emergency cerclage was significantly lower than that of singleton emergency cerclage, but higher than that of expectant treatment (28.5, 34.0 and 24.0 weeks, respectively). The median interval to delivery of twin emergency cerclage was significantly lower than that of singleton emergency cerclage, but significantly higher than that of expectantly treated twin pregnancies (37.0, 78.0 and 7.0 days, respectively).IMPACT STATEMENT What is already known on this subject? An important cause of premature birth is cervical insufficiency. Cervical cerclage extends the gestational period of women with cervical insufficiency. According to 2019 SOGC's No. 373-Cervical Insufficiency and Cervical Cerclage, both twin and single pregnancies benefit from emergency cerclage. However, there is minimal information about the pregnancy outcomes of emergency cerclage in twin pregnancies. What the results of this study add? This study shows that the outcomes of pregnancy in twin pregnancies undergoing emergency cerclage were better than that of expectant treatment but worse than that in singleton pregnancies undergoing emergency cerclage. What the implications are of these findings for clinical practice and/or further research? In this study, pregnant women with cervical insufficiency in twin pregnancies can benefit from emergency cerclage, we should treat those pregnant women as early as possible.

Abstract Aim The aim of this study was to explore the experience and perceptions of patients with Crohn’s disease in China. Methods Data mining was used to investigate posts in Crohn’s disease online medical communities. The data were collected through the crawler code, and latent Dirichlet allocation (LDA) and grounded theory were used to mine the theme features after data cleaning. Results In analyzing the topic characteristics of online posts, LDA divided 6757 posts into 15 topics on four aspects: seeking disease information, making decisions on medication use, psychological burden, and communicating about diet and nutrition. Conclusion Overall, social media is patient-centric and helps us better understand the experiences and perceptions of patients. This study can help medical staff predict the thoughts and concerns of Crohn's disease patients during the treatment process, facilitate doctor-patient communication, and assist in the formulation of medical policies.

Abstract Autism spectrum disorder (ASD) is a neurodevelopmental disorder that affects about 2% of children. Due to the shortage of clinicians, there is an urgent demand for a convenient and effective tool based on regular videos to assess the symptom. Computer-aided technologies have become widely used in clinical diagnosis, simplifying the diagnosis process while saving time and standardizing the procedure. In this study, we proposed a computer vision-based motion trajectory detection approach assisted with machine learning techniques, facilitating an objective and effective way to extract participants’ movement features (MFs) to identify and evaluate children’s activity levels that correspond to clinicians’ professional ratings. The designed technique includes two key parts: (1) Extracting MFs of participants’ different body key points in various activities segmented from autism diagnostic observation schedule (ADOS) videos, and (2) Identifying the most relevant MFs through established correlations with existing data sets of participants’ activity level scores evaluated by clinicians. The aim of this study was to find a purely natural and effective preservative that could be used in fresh-cut apples extracting proanthocyanidin (PC) components from black chokeberry (Aronia melanocarpa (Michx.) Elliott) using DESs (deep eutectic solvents). DM130 microporous resins were used in purification experiments to obtain proanthocyanidin purifiers with a higher content. To investigate the free radical scavenging ability (DPPH, ·OH, O2−, ABTS) of the resulting proanthocyanidin purifiers, experiments were conducted due to their potent antioxidant properties. The results show that the polyphenol extract from black chokeberry demonstrated an excellent antioxidant capacity, with more than 95% scavenging of DPPH, ABTS and superoxide anion at concentrations above 0.1 mg/mL. Furthermore, the antibacterial efficacy of the product was also evaluated. The results of this study on the inhibitory ability of bacteria demonstrated that PC exhibited superior inhibitory efficacy against E. coli and S. aureus, with inhibitory circles of 16 ± 0.11 mm and 18 ± 0.09 mm, respectively. Conversely, PC demonstrated no inhibitory efficacy against A. niger and Penicillium sp. Finally, the effect of PC in preserving fresh-cut apples was tested in terms of weight loss, hardness, appearance and total number of microorganisms. The results demonstrated a 26.44% reduction in weight loss and a 13.5% increase in hardness, as well as a 98% reduction in total bacterial counts in apple pieces treated with 5 mg/mL of PC in comparison to those left untreated.

Recently, non-orthogonal multiple access (NOMA)-based space–air–ground integrated networks (SAGINs) have gained increasing attention due to their robust communication, broader coverage, and resource-saving advantages. However, it is imperative to consider physical layer security as a crucial performance metric in NOMA-based SAGINs. This study addresses this concern by constructing a NOMA-based free space optical (FSO)/radio frequency (RF) dual-hop SAGIN system with eavesdroppers on both links. The two new fading channel models were proposed, considering the FSO link’s fog absorption and the RF link’s stochastic distribution based on Málaga and shadowed Rician distributions. The closed-form expressions for the secrecy outage probability are derived for the SAGIN system. Monte Carlo simulations were conducted to validate the theoretical findings. The results revealed the influence of fog absorption and the stochastic geometry distribution on the SAGIN system.

Traditional synthetic-aperture radar (SAR) imaging, while adept at capturing stationary scenes with clarity, often results in the blurring of moving targets due to Doppler spectrum disparities. To overcome this, this study introduces an innovative optical approach for imaging moving targets. By employing a spatial light modulator (SLM), the phase information of SAR data can be modulated onto a light beam. Then, the light propagation path is meticulously designed to take advantage of the free propagation of light to compensate for the phase differences in SAR data, thereby achieving focal clarity. Simulations and experimental validations have demonstrated that this method surpasses traditional digital imaging techniques in terms of focusing precision. Additionally, the processing delay is only as long as the time taken for light propagation. The optical path is simple, avoiding complex assembly and alignment. This novel approach opens up new possibilities for the SAR imaging of moving targets, offering potential applications in moving target extraction, separation, and velocity estimation.
