Abstract Background Gut bacteria are beneficial to the host, many of which must be passed on to host offspring. During metamorphosis, the midgut of holometabolous insects undergoes histolysis and remodeling, and thus risks losing gut bacteria. Strategies employed by holometabolous insects to minimize this risk are obscure. How gut bacteria affect host insects after entering the hemocoel and causing opportunistic infections remains largely elusive. Results We used holometabolous Helicoverpa armigera as a model and found low Lactobacillus load, high level of a C-type lectin (CTL) gene CD209 antigen-like protein 2 (CD209) and its downstream lysozyme 1 (Lys1) in the midgut of the wandering stage. CD209 or Lys1 depletion increased the load of midgut Lactobacillus, which further translocate to the hemocoel. In particular, CD209 or Lys1 depletion, injection of Lactobacillus plantarum, or translocation of midgut L. plantarum into the hemocoel suppressed 20-hydroxyecdysone (20E) signaling and delayed pupariation. Injection of L. plantarum decreased triacylglycerol and cholesterol storage, which may result in insufficient energy and 20E available for pupariation. Further, Lysine-type peptidoglycan, the major component of gram-positive bacterial cell wall, contributed to delayed pupariation and decreased levels of triacylglycerols, cholesterols, and 20E, in both H. armigera and Drosophila melanogaster. Conclusions A mechanism by which (Lactobacillus-induced) opportunistic infections delay insect metamorphosis was found, namely by disturbing the homeostasis of lipid metabolism and reducing 20E production. Moreover, the immune function of CTL − Lys was characterized for insect metamorphosis by maintaining gut homeostasis and limiting the opportunistic infections.

In this paper, with the method of epidemic dynamics, we assess the spread and prevalence of COVID-19 after the policy adjustment of prevention and control measure in December 2022 in Taiyuan City in China, and estimate the excess population deaths caused by COVID-19. Based on the transmission mechanism of COVID-19 among individuals, a dynamic model with heterogeneous contacts is established to describe the change of control measures and the population's social behavior in Taiyuan city. The model is verified and simulated by basing on reported case data from November 8th to December 5th, 2022 in Taiyuan city and the statistical data of the questionnaire survey from December 1st to 23rd, 2022 in Neijiang city. Combining with reported numbers of permanent residents and deaths from 2017 to 2021 in Taiyuan city, we apply the dynamic model to estimate theoretical population of 2022 under the assumption that there is no effect of COVID-19. In addition, we carry out sensitivity analysis to determine the propagation character of the Omicron strain and the effect of the control measures. As a result of the study, it is concluded that after adjusting the epidemic policy on December 6th, 2022, three peaks of infection in Taiyuan are estimated to be from December 22nd to 31st, 2022, from May 10th to June 1st, 2023, and from September 5th to October 13th, 2023, and the corresponding daily peaks of new cases can reach 400 000, 44 000 and 22 000, respectively. By the end of 2022, excess deaths can range from 887 to 4887, and excess mortality rate can range from 3.06% to 14.82%. The threshold of the infectivity of the COVID-19 variant is estimated 0.0353, that is if the strain infectivity is above it, the epidemic cannot be control with the previous normalization measures.

Abstract Background Studies on the choroid of myopic eyes with posterior staphyloma have shown that choroidal thickness decreased. This retrospective study further analysed the effects of posterior scleral staphyloma on choroidal blood vessels and matrix components compared to non-pathological myopia. Methods In this cross-sectional study, ninety-one eyes were divided into pathological (posterior staphyloma) and non-pathological myopia. The latter was further divided into three groups (Group 1: 26 mm ≤ axial length; Group 2: 24 mm ≤ axial length 26 mm.

PurposeWe aimed to explore the effects of percutaneous coronary intervention (PCI) on the ophthalmic artery (OA) hemodynamics in patients with acute coronary syndrome (ACS).MethodsA total of 73 participants (Group0: healthy controls, Group1: Patients with ACS underwent PCI < 3 months, Group2: Patients with ACS underwent PCI ≥ 3 months) were enrolled. Computed tomographic angiography images were used to construct three-dimensional models of participants' OAs. Numerical simulations based on computational fluid dynamics were used to acquire hemodynamic parameters.ResultsThe angle between the OA and internal carotid artery in Group2 was significantly larger compared with Group0 and Group1 (P = 0.003 and P = 0.044). Hemodynamic simulation showed a significantly slower OA blood velocity in Group1 than in the control (P < 0.001) and Group2 (P = 0.033). Lower wall shear stress was found in Group1 than that in control (P = 0.040). Patients after PCI had a higher wall pressure than healthy controls (P = 0.012 and P = 0.004). Mass flow ratios were decreased in Group1 and Group2 (P = 0.021 and P = 0.002). The hemodynamic parameters of OA were correlated with several clinical indicators.ConclusionsThe OA blood flow velocity of patients with ACS after PCI initially slowed down, which increased the risk of plaque formation, and then showed an increasing trend. There was a correlation between OA hemodynamic parameters and clinical indexes related to cardiac stress. Ischemia-reperfusion injury and changes in blood flow status after PCI may affect OA morphology and hemodynamics, leading to ocular lesions.Trial registrationChiCTR2100050428.

Abstract Background The insect hemolymph (blood-equivalent fluid), composed of a large number of hemocytes (blood cells) and a variety of soluble immune effectors, is hostile for pathogens including fungi. In order to survive in the insect hemocoel (body cavity), the entomopathogenic fungus (EPF) has evolved two classical coping strategies, namely evasion and suppression of the host immune reactions. However, it remains unclear whether EPF has other ways of coping with host immunity. Results In this study, we demonstrated that Metarhizium rileyi (an EPF) infection by injection of blastospores into the hemocoel enhanced the plasma antibacterial activity of cotton bollworm (Helicoverpa armigera), which was partially due to the enhanced expression of antimicrobial peptides (AMPs). The early stage of M. rileyi infection induced the translocation of gut bacteria into the hemocoel, where they were subsequently cleared due to the enhanced plasma antibacterial activity. Further, we showed that the enhanced plasma antibacterial activity and AMP expression were attributable to M. rileyi but not the invasive gut bacteria (opportunistic bacteria). Elevated ecdysone (major steroid hormone in insects) levels in the hemolymph at 48 h post-M. rileyi infection might contribute to the enhanced expression of AMPs. The fungus-elicited AMPs, such as cecropin 3 or lebocin, exhibited potent inhibitory activity against the opportunistic bacteria but not against hyphal bodies. In addition, the opportunistic bacteria competed with hyphal bodies for amino acid nutrients. Conclusions M. rileyi infection induced the translocation of gut bacteria, and then the fungi activated and exploited its host humoral antibacterial immunity to eliminate opportunistic bacteria, preventing them from competing for nutrients in the hemolymph. Unlike the classical strategies, EPF utilizes to evade or suppress host immunity, our findings reveal a novel strategy of interaction between EPF and host immunity. Video Abstract

Abstract Background Cerebral small vessel disease (CSVD) is a systemic disease, affecting not only the brain, but also eyes and other organs. The total CSVD score is a tool for comprehensive evaluation of brain lesions in patients with CSVD. The ophthalmic artery (OA) is a direct response to ocular blood flow. However, little is known about the correlation between CSVD and characteristics of OA. We investigated the OA morphologies and hemodynamics in patients with CSVD and the correlation between these changes and the total CSVD score. Methods This cross-sectional observational study included 34 eyes from 22 patients with CSVD and 10 eyes from 5 healthy controls. The total CSVD score was rated according to the CSVD signs on magnetic resonance imaging. OA morphological characteristics were measured on the basis of 3D OA model reconstruction. OA hemodynamic information was calculated using computational fluid dynamics simulations. Results The total CSVD score negatively correlated with the OA diameter, blood flow velocity, and mass flow ratio (all P 0.05). ObjectiveTo explore the prevalence and main influencing factors of fear of disease progression (FoP) in Chinese cancer survivors and to provide references for the prevention and control of mental health problems in cancer patients. MethodsWe searched Chinese National Knowledge Infrastructure (CNKI), China Biomedical Database (CBM), Wanfang Database, Weipu Chinese Science Database and Technology Periodical Database (VIP), PubMed, Cochrane Library Database, Web of Science Database, Embase Database, PsycINFO Database, and CINAHL Database to retrieve literatures on the prevalence and influencing factors of FoP in Chinese cancer survivors published up to August 31, 2021. Stata MP 16.0 software was used to perform a meta-analysis. ResultsFinally, 22 literatures (19 in Chinese and 3 in English) were included in the analysis, with a total of 6 461 participants. The meta-analysis results showed that the prevalence of FoP in Chinese cancer survivors was 51.0% (95% confidence interval [95% CI ]: 42.0% – 61.0%). The results of subgroup analysis revealed that the prevalence of FoP differed significantly among cancer survivors in different regions (χ2 = 843.3, P < 0.05 ), with the prevalence rates of 56.0%, 54.0% and 42.0% for cancer survivors in South, Central and East China; the prevalence also varied among the survivors suffering from cancer of various systems (χ2 = 843.3, P < 0.05 ) and the prevalence rate was 40.0%, 56.0%, 58.0% and 54.0% for the survivors with digestive tract, gynecological, hematological and respiratory malignancies, respectively. The meta-analysis revealed that age, average monthly income, clinical stage of the malignancy, educational level, marital status, employment, disease course, family history of cancer, and anxiety were influencing factors for FoP among the survivors. The results of meta-regression analysis demonstrated that the sources of heterogeneity of the included studies were associated with following characteristics of the survivors: aged < 40 years ( β = 4.29, P = 0.018 ), monthly income of 1 000 – 2 999 (RMB yuan, β = – 3.80, P = 0.032 ), monthly income of 3 000 – 4 999 ( β = – 4.56, P = 0.011 ), monthly income of ≥ 5 000 ( β = – 8.70, P = 0.000 ), and with the malignancy of clinical stage I ( β = – 5.55,P = 0.020 ). Sensitivity analysis indicated that the results of the meta-analysis were stable; the results of Egger′ test and funnel plot demonstrated that the publication bias was less likely to present for the studied indicators, except for family history. ConclusionThe prevalence of FoP is relatively high among Chinese cancer survivors and the prevalence varies among survivors living in different regions and with different clinical stage of the malignancy; the FoP prevalence is mainly influenced by survivors′ age, monthly income, clinical stage of the malignancy, education, marital status, employment, disease course, family history of malignancy, and anxiety symptom.

Abstract Background Ocular blood flow provides a new perspective for studying the effects of diabetes and ischemic heart disease on systemic blood flow, pathological mechanisms, and prognosis. Previous studies have analyzed the hemodynamic changes of the ophthalmic artery (OA) in patients with diabetes and ischemic heart disease, but the results remain controversial due to limited observation methods. We aimed to explore the morphological and hemodynamic features in the OA in patients with type 2 diabetes (T2D) with and without acute coronary syndrome (ACS). Methods In total, 134 participants, including 30 control participants, 34 with ACS only, 34 with T2D only, and 36 with both ACS and T2D, undergoing computed tomography angiography were enrolled. Three-dimensional OA models were reconstructed, and morphological parameters of the OA were measured. In addition, numerical simulations using computational fluid dynamics were used to acquire hemodynamic parameters of the OA. Results In this study, 134 OA models were reconstructed. Morphological measurements revealed a smaller initial OA diameter in the T2D group than in the other two ACS groups. Abstract Camrelizumab (SHR-1210) is a humanized IgG4 monoclonal anti-programmed cell death protein 1 (PD-1) antibody that has been shown to inhibit the binding of PD-1 to PD-L1, thereby blocking the immune escape of various types of cancer, including lung squamous cell carcinoma (LSCC). Reactive cutaneous capillary endothelial proliferation (RCCEP) is the most common adverse event in camrelizumab-treated patients. Here, we introduce a case of LSCC with recall RCCEP induced by stereotactic body radiation therapy (SBRT). A 76-year-old LSCC patient developed RCCEP when he received camrelizumab and chemotherapy. After discontinuing camrelizumab treatment, the RCCEP lesions spontaneously regressed and fell off. However, when the patient received subsequent SBRT, the RCCEP occurred again at the same sites. This case may provide clues for additional study of the immune reactivation effect of SBRT or the underlying mechanism of RCCEP.

ObjectiveTo examine changing trend in disease burden of lung cancer and related risk factors among residents in China from 1990 to 2019 for developing targeted strategies on lung cancer prevention and treatment. MethodsThe open access data relevant to disease burden of lung cancer and its associates among Chinese residents in 1990, 2005 and 2019 were extracted from the Global Burden of Disease (GBD) Study 2019. We used years lived with disability (YLDs), years of life lost (YLLs), and disability adjusted life years (DALYs) to describe changes of lung cancer burden and population-attributable faction (PAF) to analyze the disease burden attributable to risk factors and epidemiological transition among the residents of various age groups from 1990 to 2019. ResultsIn 2019 among Chinese male residents, the rates (1/100 000) of YLDs, YLLs, and DALYs of lung cancer were 19.10, 1 632.03, and 1 651.13 and all the rates were higher than those of 8.73, 731.14, and 739.87 among Chinese female residents; the three rates increased with age, with the highest rates of 76.17, 5 243.52, and 5 319.70 in the residents aged 70 years and over. Compared to those in 1990, the rates of YLDs, YLLs, and DALYs increased by 3.61%, 2.49%, and 2.50%, respectively in 2019. Among the 16 known risk factors of lung cancer, the top five with highest contributions to DALYs (million person years), DALYs rate (1/10 000) and PAF were smoking (10.75, 755.91 and 62.06%), environmental particulate pollution (3.86, 271.20 and 22.47%), second-hand smoke (1.35, 94.81 and 7.86%), high fasting plasma glucose (1.11, 77.87 and 6.47%) and home air pollution due to solid fuel use (0.85, 59.72 and 4.94%). Of the PAF for the 16 known risk factors during the period of 1990 – 2019, the PAFs of 8 factors increased, with the highest increase of 110.12% for environmental particulate pollution and other 8 factors′ PAF decreased, with the lowest decrease of 77.74% for home air pollution due to solid fuel use. ConclusionDuring 1990 – 2019 among residents in China, the disease burden of lung cancer was heavy and the YLDs, YLLs and DALYs rate of lung cancer increased; smoking is still a main risk factor for disease burden of lung cancer.

Abstract Background To investigate the structural and functional changes of the retina in patients with different degrees of internal carotid artery (ICA) stenosis. Methods This cross-sectional study included patients with varying degrees ICA stenosis. Clinical characteristics of 41 patients were collected after being divided into four groups according to the ICA stenosis indicated by computed tomographic angiography (Group 0: without ICA stenosis, Group 1: ipsilateral slight ICA stenosis, Group 2: ipsilateral moderate ICA stenosis, Group 3: ipsilateral severe ICA stenosis). Retinal vessel caliber (RVC) was measured quantitatively with the Integrative Vessel Analysis software. The retinal sensitivity was examined with the MP-3 microperimeter. The relationships among central retinal artery equivalent (CRAE), central retinal vein equivalent, arteriole to venule ratio (AVR), mean retinal sensitivity (MS) and ICA stenosis degree were analysed. Regional function identification and transformation mechanism exploration provide an entry point for a comprehensive understanding of the pattern and direction of rural development. This paper proposes a new analytical framework by establishing first, the rural production-living-ecological evaluation index; second, the optimal functional clustering of 186 counties in the Central Plains Urban Agglomeration area of China by combining the Self Organizing Feature Map network, Geodetector model, and Mann-Kendall test method; and third, the transformation mechanism by comparing the 2-phase zoning results with an interval of 10 years. An overall positive trend was found in the transformation of the rural function. The proportion of lagging development functional areas decreased by 20.5 %. The proportions of dominant function areas and composite coordination function areas increased by 5.4 % and 15.1 %, respectively. There was a synergistic relationship between production and living functions and an opposing relationship between ecological functions and the other two. The functional area with a synergistic relationship had the highest conversion rate, and most of the functional areas with trade-off relationships remained unchanged. The transfer-out rate of functional areas with synergistic and trade-off relations is negatively correlated with its lagging functional inferiority. The degree of difficulty in transitioning to functional areas with synergistic or trade-off relations is similar. The research reveals the mechanism and law of the evolution of rural regional functions and provides a methodology reference and policy planning basis for further research on rural areas.

The widespread use of graphene oxide-silver nanoparticle nanocomposites (GO-AgNPs) in biomedical sciences is increasing the chances of human and animal exposure to its chronic non-toxic doses. Exposure to AgNPs-related nanomaterials may result in the negative effect on the dam, fetus and offspring. However, there are only little available information for profound understanding of the epigenetic alteration in the cells and animals caused by low-dose chronic exposure of GO-AgNPs. The present study investigated the effect of 0.5 μg/mL GO-AgNPs for 10 weeks on the differential expression of circular RNAs (circRNAs) in caprine fetal fibroblast cells (CFFCs), and this dose of GO-AgNPs did not affect cell viability and ROS level. We predicted the functions of those differentially expressed (DE) circRNAs in CFFCs by bioinformatics analysis. Furthermore, we validated the expression of ten DE circRNAs using quantitative real-time reverse transcription-polymerase chain reaction (qRT-PCR) to ensure the reliability of the sequencing data. Our results showed that the DE circRNAs may potentially regulate the GO-AgNPs-inducing epigenetic toxicity through a regulatory network consisted of circRNAs, miRNAs and messenger RNAs (mRNAs). Therefore, the epigenetics toxicity is essential to assess the biosafety level of GO-AgNPs.

Background Trimethylamine-N-Oxide (TMAO) is a proatherogenic and prothrombotic metabolite. Our study examined the association of plasma TMAO level with cardiovascular and all-cause mortality in hemodialysis (HD) patients. Methods Patients who were at least 18 years-old and received HD for at least 6 months were enrolled within 6 months. Patients with coronary heart disease, congestive heart failure, arrhythmia, or stroke within 3 months before study onset were excluded. The primary endpoints were cardiovascular and all-cause death, and the secondary endpoint was cerebrovascular death. Results We recruited 252 patients and divided them into a high-TMAO group (>4.73 μg/mL) and a low-TMAO group (≤4.73 μg/mL). The median follow-up time was 73.4 months (interquartile range: 42.9, 108). A total of 123 patients died, 39 from cardiovascular disease, 19 from cerebrovascular disease, and 65 from other causes. Kaplan-Meier analysis indicated that the high-TMAO group had a greater incidence of cardiovascular death (Log-Rank: p = 0.006) and all-cause death (Log-Rank: p

Abstract Background Mermithid nematodes, such as Ovomermis sinensis, display a broad host range including some lepidopteran pests. Infective juveniles penetrate their host through the cuticle, complete their growth within the hemocoel and eventually kill the host upon their emergence. Hence, mermithid nematodes are considered potential biological control agents of insect pests. Our previous data indicate that the infection rate of O. sinensis on cotton bollworm (Helicoverpa armigera) is low, which may be largely due to the strong immune system of the host. However, current knowledge on the interactions of mermithid nematodes with their hosts and the mechanisms employed by hosts to defend themselves against mermithid nematodes is limited. Results Here, we investigated the response of H. armigera to O. sinensis infection. Parasitism by O. sinensis caused a sharp decline in the survival rate of H. armigera. The hemocytic phagocytosis ability, antibacterial activity, and phenoloxidase (PO) activity in plasma of H. armigera increased at 1 d post parasitism (dpp) but decreased at 3 dpp. Further, we investigated gene expression in the fat body of parasitized and non-parasitized H. armigera larvae at 1, 3, and 5 dpp using a digital gene expression system. In total, 41, 60 and 68 immune-related differentially expressed genes were identified at 1, 3, and 5 dpp, respectively. These genes encoded pattern recognition receptors (PRRs), antimicrobial peptides (AMPs), serine proteases (SPs), SP inhibitors, mucins and other immune-related proteins. The expression of most PRRs, AMPs, SPs, and mucins was upregulated in the fat body of larvae at 1 dpp, downregulated at 3 dpp, and then again upregulated at 5 dpp by O. sinensis. The increased expression of SP inhibitors may contribute to the inhibited PO activity at 5 dpp. Conclusions This study demonstrates that parasitism by O. sinensis modulates the immune reaction of the host H. armigera by altering the expression of immune-related genes. Our data provide a basis for future investigation of the molecular mechanisms employed by the mermithid nematode O. sinensis to modulate the immunity of the host H. armigera. These data will also likely facilitate the improvement of success in parasitism of H. armigera by O. sinensis.

Lymphopenia caused by disease or treatment is frequent in patients with cancer, which seriously affects the prognosis of these patients. Immune checkpoint inhibitors (ICIs) have garnered attention as one of the most promising strategies for the treatment of esophageal cancer (EC). The status of the immune system, such as, the lymphocyte count, is now considered to be an important biomarker for ICI treatments. Recognition of the significant impact of the lymphocyte count on the survival of patients with EC in the era of immunotherapy has revived interest in understanding the causes of lymphopenia and in developing strategies to predict, prevent and eliminate the adverse effect of lymphopenia. Here, we review what we have learned about lymphopenia in EC, including the prognostic and predictive value of lymphopenia in patients with EC, the predictors of lymphopenia, and the strategies to ameliorate the effect of lymphopenia in patients with EC.

Abstract The North Tianshan Orogenic Belt contains the youngest ophiolites in the Tianshan and provides some information on timing of the last closure of the Junggar-Balkhash Ocean. LA–ICP–MS zircon U–Pb dating was conducted to define the formation age of the Arbasay Formation in the Shichang Region of North Tianshan, which is exposed near the suture zone but its age remains debated. The Arbasay Formation is mainly composed of volcanic and volcaniclastic rocks with tuff interlayers. The zircons from the tuffs yield two age populations of 315 ± 3 Ma and 304 ± 2 Ma, constraining the commencement and demise timings of volcanism, respectively. Furthermore, zircon U–Pb age spectra of the tuffaceous sandstones display the youngest peak age at 308 Ma, indicating a

Abstract Background In China, stomach cancer is the third most common cancer and the third leading cause of cancer death. Few studies have examined Chinese stomach cancer patients’ medical expenses and their associated trends. The Cancer Screening Program in Urban China (CanSPUC) is a Major Public Health Project funded by the central government. Through this project, we have extracted patients’ medical expenses from hospital billing data to examine the costs of the first course treatments (which refers to 2 months before and 10 months after the date of cancer diagnosis) in Chinese patients with stomach cancer and the associated trends. Methods The expense data of 14,692 urban Chinese patients with stomach cancer were collected from 40 hospitals in 13 provinces. We estimated the inflation-adjusted medical expenses per patient during 2002–2011. We described the time trends of medical expenses at the country-level, and those trends by subgroup, and analyzed the compositions of medical expenses. We constructed the Generalized Linear Mixed (GLM) regression model with Poisson distribution to examine the factors that were associated with medical expenses per patient. Results The average medical expenses of the first course treatments were about 43,249 CNY (6851 USD) in 2011, more than twice of that in 2002. The expenses increased by an average annual rate of 7.4%. Longer stay during hospitalization and an increased number of episodes of care are the two main contributors to the expense increase. The upward trend of medical expenses was observed in almost all patient subgroups. Drug expenses accounted for over half of the medical expenses. Conclusions The average medical expenses of the first course (2 months before and 10 months after the date of cancer diagnosis) treatments per stomach cancer patient in urban China in 2011 were doubled during the previous 10 years, and about twice as high as the per capita disposable income of urban households in the same year. Such high expenses indicate that it makes economic sense to invest in cancer prevention and control in China.

Purpose. To investigate the outcome of pars plana vitrectomy (PPV) with suitable internal limiting membrane peeling (ILM) and air tamponade for patients with highly myopic foveoschisis-associated lamellar macular hole (MH). Methods. This retrospective interventional case series included 11 patients with highly myopic foveoschisis-associated lamellar MH who underwent PPV and indocyanine green-aided ILM peeling up to the temporal vascular arcades. Following air tamponade after surgery, all patients were instructed to maintain a face-down position. The patients were followed up for over 1 year and evaluated for MH closure and the best-corrected visual acuity before and after surgery. Results. The mean ± standard deviation values of patient age, axial length, and follow-up duration were 67.82 ± 6.54 years, 29.21 ± 1.95 mm, and 24.27 ± 8.11 months, respectively. After surgery, the lamellar MH closed in all eyes, and 10 eyes showed vision improvement at the 1-month, 3-month, and final follow-up evaluations. One patient showed decreased vision at 2 years after surgery, with patchy chorioretinal atrophy in the macular region. Myopic foveoschisis showed resolution in three eyes and alleviation in eight. Ten patients underwent cataract surgery during PPV. Conclusion. Extension of ILM peeling up to the temporal vascular arcades and air tamponade after PPV may improve the visual function and rate of MH closure for patients with highly myopic foveoschisis-associated lamellar MH.

Graphene oxide–silver nanoparticle (GO-AgNPs) nanocomposites have drawn much attention for their potential in biomedical uses. However, the potential toxicity of GO-AgNPs in animals and humans remains unknown, particularly in the developing fetus. Here, we reported the GO-AgNP-mediated cytotoxicity and epigenetic alteration status in caprine fetal fibroblast cells (CFFCs). In brief, the proliferation and apoptosis rate of GO-AgNP-treated CFFCs (4 and 8 µg/mL of GO-AgNPs) were measured using the cell-counting kit (CCK-8) assay and the annexin V/propidium iodide (PI) assay, respectively. In addition, the oxidative stress induced by GO-AgNPs and detailed mechanisms were studied by evaluating the generation of reactive oxygen species (ROS), superoxide dismutase (SOD), lactate dehydrogenase (LDH), malondialdehyde (MDA), and caspase-3 and abnormal methylation. The expression of pro- and anti-apoptotic genes and DNA methyltransferases was measured using reverse transcription followed by RT-qPCR. Our data indicated that GO-AgNPs cause cytotoxicity in a dose-dependent manner. GO-AgNPs induced significant cytotoxicity by the loss of cell viability, production of ROS, increasing leakage of LDH and level of MDA, increasing expression of pro-apoptotic genes, and decreasing expression of anti-apoptotic genes. GO-AgNPs incited DNA hypomethylation and the decreased expression of DNMT3A. Taken together, this study showed that GO-AgNPs increase the generation of ROS and cause apoptosis and DNA hypomethylation in CFFCs. Therefore, the potential applications of GO-AgNPs in biomedicine should be re-evaluated.

A large petrochemical complex, dubbed Petro–complex, situated in a rather rural region of Taiwan, was used as a test bed to detect emissions from the Petro–complex to its surroundings. Hourly observations of speciated non–methane hydrocarbons (NMHCs) by the photochemical assessment monitoring stations (PAMSs), as well as the total amounts of NMHCs, SO2, and NOx provided by the air quality stations (AQSs), were utilized to find useful petro–emission indication methods. The analytical aspect of NMHCs either as a speciated form or as total amounts was demonstrated through field comparison to illustrate data quality. Using ethyne to offset traffic influence, the ratios of ethene to ethyne (acetylene) (E/A) and propene to ethyne (P/A) were proven to be effective indicators of petro–emissions owing to pronounced emissions of ethene and propene, revealed as tall spikes in PAMS measurements. SO2 and NOx were also explored as petro–emission indicators mainly for stack sources. By coordinating with three–dimensional modeling, SO2 from petro–emissions could be distinguished from other prominent sources, such as coal–fired power plants. An attempt was also made to use SO2 and NOx as indicators of broader petro–emissions with stringent criteria to minimize traffic interference and increase specificity. Similar findings were observed with the three indicators, that is, volatile organic compounds (VOCs) ratios, SO2 and NOx, to identify the southwest area of the Petro–region as the most affected area, as represented by Taisi station (F2). The percent affected time of a year at F2 was 10%–14%, owing to the dominant wind field of northeast monsoonal (NEM) in the region, as compared with other sites in the east and north of 1–5%. Using VOC ratios as petro–emission indicators is more effective than using other gases, owing to the compositional advantage to minimize traffic interference.

Continuous measurements of ozone and its precursors were performed at sites in two Chinese megacities, i.e., an urban site in Beijing and a suburban site in the Pearl River Delta (PRD). At both sites, the total oxidants (O3 + NO2) varied with the ratio of ethylbenzene to m,p-xylenes, which serves as an indicator of photochemical aging. An observation-based method (OBM) was derived for calculating the photochemical consumption of individual non-methane hydrocarbons (NMHCs) based on the observed NMHC concentrations and the ratio of ethylbenzene to m,p-xylenes. The results show a strong correlation between the oxidant level and the derived consumption of precursors at the two sites (R2 = 0.81 for the PRD site and R2 = 0.83 for the Beijing site), demonstrating a strong cause–effect relationship. The relative “consumption efficiency” among NMHCs was calculated based on the integrated amount of hydroxyl radicals derived from the ratio of ethylbenzene to xylenes. Thus, the percent contributions to ozone formation from each individual NMHC can be calculated. This concept of consumption is purely observation-based and provides an easy way to bypass complicated modeling and the necessity of knowing instantaneous concentrations of hydroxyl radicals, which are highly illusive in nature.

The adaptability of soybean to be grown at a wide range of latitudes is attributed to natural variation in the major genes and quantitative trait loci (QTLs) that control flowering time and maturity. Thus, the identification of genes controlling flowering time and maturity and the understanding of their molecular basis are critical for improving soybean productivity. However, due to the great effect of the major maturity gene E1 on flowering time, it is difficult to detect other small-effect QTLs. In this study, aiming to reduce the effect of the QTL, associated with the E1 gene, on the detection of other QTLs, we divided a population of 96 recombinant inbred lines (RILs) into two sub-populations: one with the E1 allele and another with the e1nl allele. Compared with the results of using all 96 recombinant inbred lines, additional QTLs for flowering time were identified in the sub-populations, two (qFT-B1 and qFT-H) in RILs with the E1 allele and one (qFT-J-2) in the RILs with the e1nl allele, respectively. The three QTLs, qFT-B1, qFT-H and qFT-J-2 were true QTLs and played an important role in the regulation of growth period. Our data provides valuable information for the genetic mapping and gene cloning of traits controlling flowering time and maturity and will help a better understanding of the mechanism of photoperiod-regulated flowering and molecular breeding in soybean.

Purpose. To determine the effect of osthole on the retina in a chronic cerebral hypoperfusion (CCH) rat model and to investigate its therapeutic activity. Methods. Seventy-two rats were randomly allocated into 6 groups. CCH was induced by permanent bilateral common carotid artery occlusion (BCCAO) in five groups. Sham surgery was performed without occlusion of the artery in the sixth group (control group). Animals were administered with saline (model group), osthole (osthole-IG group), aspirin (aspirin group), or ginaton (ginaton group); the osthole-PI group was performed with peribulbar injection of osthole. Four rats in each group were sacrificed every 5 days after drug administration, and histopathology along with morphology of retina were observed. Fundus fluorescein angiography was performed before the animals were sacrificed at day 15. Retinal Akt, NF-κB, Bax, and Bcl-2 levels were assessed using immunohistochemistry, immunofluorescence, and reverse-transcription PCR; retinal injury was assessed using TUNEL in situ; retinal levels of superoxide dismutase (SOD) and malondialdehyde (MDA) were measured. Results. Fundus fluorescein angiography revealed the retinal vascular diameter in the osthole-IG group rats to be wider than that in the model, osthole-PI, aspirin, or ginaton group rats. Histological analysis of retinal tissue revealed an increase in retinal thickness in all treatment groups, and significant improvement was noticed in the osthole-IG group. TUNEL staining revealed fewer apoptotic cells in the osthole-IG and osthole-PI groups than in the other groups. For immunohistochemistry results, in the osthole-IG group, levels of NF-κB and Akt were lower than those in the other treated groups, while levels of the ratio Bcl-2/Bax were higher. Levels of MDA were lower and levels of SOD were higher in the osthole-IG group than in the other groups. Conclusions. Osthole protects the retina from ischemia injury secondary to CCH induced by BCCAO, mainly through anti-inflammatory, antioxidant, and antiapoptotic effects.

We measured levels of ambient volatile organic compounds (VOCs) at seven sites in the Pearl River Delta (PRD) region of China during the Air Quality Monitoring Campaign spanning 4 October to 3 November 2004. Two of the sites, Guangzhou (GZ) and Xinken (XK), were intensive sites at which we collected multiple daily canister samples. The observations reported here provide a look at the VOC distribution, speciation, and photochemical implications in the PRD region. Alkanes constituted the largest percentage (>40%) in mixing ratios of the quantified VOCs at six sites; the exception was one major industrial site that was dominated by aromatics (about 52%). Highly elevated VOC levels occurred at GZ during two pollution episodes; however, the chemical composition of VOCs did not exhibit noticeable changes during these episodes. We calculated the OH loss rate to estimate the chemical reactivity of all VOCs. Of the anthropogenic VOCs, alkenes played a predominant role in VOC reactivity at GZ, whereas the contributions of reactive aromatics were more important at XK. Our preliminary analysis of the VOC correlations suggests that the ambient VOCs at GZ came directly from local sources (i.e., automobiles); those at XK were influenced by both local emissions and transportation of air mass from upwind areas.

BACKGROUND: The heavy metal lead has been shown to be associated with a genotoxic risk. Drosophila melanogaster is a model organism commonly utilized in genetic toxicology testing. The endosymbionts--Wolbachia are now very common in both wild populations and laboratory stocks of Drosophila. Wolbachia may induce resistance to pathogenic viruses, filarial nematodes and Plasmodium in fruit fly and mosquito hosts. However the effect of Wolbachia infection on the resistance of their hosts to heavy metal is unknown. METHODOLOGY/PRINCIPAL FINDINGS: Manipulating the lead content in the diet of Drosophila melanogaster, we found that lead consumption had no different effects on developmental time between Wolbachia-infected (Dmel wMel) and -uninfected (Dmel T) flies. While in Pb-contaminated medium, significantly reduced amount of pupae and adults of Dmel wMel were emerged, and Dmel wMel adults had significantly shorter longevity than that of Dmel T flies. BACKGROUND: Wolbachia are obligate endosymbiotic bacteria that infect numerous species of arthropods and nematodes. Wolbachia can induce several reproductive phenotypes in their insect hosts including feminization, male-killing, parthenogenesis and cytoplasmic incompatibility (CI). CI is the most common phenotype and occurs when Wolbachia-infected males mate with uninfected females resulting in no or very low numbers of viable offspring. However, matings between males and females infected with the same strain of Wolbachia result in viable progeny. Despite substantial scientific effort, the molecular mechanisms underlying CI are currently unknown. METHODOLOGY/PRINCIPAL FINDINGS: Gene expression studies were undertaken in Drosophila melanogaster and D. simulans which display differential levels of CI using quantitative RT-PCR. We show that Hira expression is correlated with the induction of CI and occurs in a sex-specific manner. Hira expression is significantly lower in males which induce strong CI when compared to males inducing no CI or Wolbachia-uninfected males. A reduction in Hira expression is also observed in 1-day-old males that induce stronger CI compared to 5-day-old males that induce weak or no CI. In addition, Hira mutated D. melanogaster males mated to uninfected females result in significantly decreased hatch rates comparing with uninfected crosses. Interestingly, wMel-infected females may rescue the hatch rates. An obvious CI phenotype with chromatin bridges are observed in the early embryo resulting from Hira mutant fertilization, which strongly mimics the defects associated with CI. CONCLUSIONS/SIGNIFICANCE: Our results suggest Wolbachia-induced CI in Drosophila occurs due to a reduction in Hira expression in Wolbachia-infected males leading to detrimental effects on sperm fertility resulting in embryo lethality. These results may help determine the underlying mechanism of CI and provide further insight in to the important role Hira plays in the interaction of Wolbachia and its insect host.

Four-year ground-level measurements of the two primary greenhouse gases (carbon dioxide (CO _2 ) and methane (CH _4 )) were conducted at Dongsha Island (DSI), situated in the northern South China Sea (SCS), from March 2010 to February 2014. Their mean mixing ratios are calculated to be 396.3 ± 5.4 ppm and 1863.6 ± 50.5 ppb, with an annual growth rate of +2.19 ± 0.5 ppm yr ^–1 and +4.70 ± 4.4 ppb yr ^–1 for CO _2 and CH _4 , respectively, over the study period. Our results suggest that the Asian continental outflow driven by the winter northeast monsoon could have brought air pollutants into the northern SCS, as denoted by significantly elevated levels of 6.5 ppm for CO _2 and 59.6 ppb for CH _4 , which are greater than the marine boundary layer references at Cape Kumukahi (KUM) in the tropical northern Pacific in January. By contrast, the summertime CH _4 at DSI is shown to be lower than that at KUM by 19.7 ppb, whereas CO _2 is shown to have no differences (

Female mosquitoes require a vertebrate blood meal to activate reproduction, transmitting numerous devastating human diseases. Vitellogenesis is a central event of female reproduction that involves the massive production of vitellogenin (Vg) in the fat body and the maturation of ovaries. This process is controlled by the steroid hormone 20-hydroxyecdysone (20E); however, its molecular regulatory basis remains not completely understood. We found that the expression of Aedes aegypti muscle aponeurosis fibromatosis B (AaMafB), coding for a basic leucine zipper (bZIP) transcription factor, was significantly up-regulated after a blood meal. The 20E-bound ecdysone receptor-ultraspiracle heterodimer directly targeted the ecdysone response element in the promoter of AaMafB, activating its transcription. Coimmunoprecipitation assays illustrated the interaction between AaMafB and Cap "n" collar C (AaCncC), another bZIP transcription factor. RNA interference-mediated depletion of AaMafB or AaCncC led to impaired ovarian growth, decreased expression of AaVg and Halloween genes, and reduced 20E levels. The AaMafB-AaCncC heterodimer directly activated the transcription of AaVg and AaShade by targeting the antioxidant response element in their promoters. Together, our results indicate that AaMafB functions as an early 20E response gene, the product of which heterodimerizes with AaCncC to maintain high 20E levels and facilitates activation of AaVg in mosquitoes after a blood meal. Data-driven surrogate models that assist with efficient evolutionary algorithms to find the optimal development scheme have been widely used to solve reservoir production optimization problems. However, existing research suggests that the effectiveness of a surrogate model can vary depending on the complexity of the design problem. A surrogate model that has demonstrated success in one scenario may not perform as well in others. In the absence of prior knowledge, finding a promising surrogate model that performs well for an unknown reservoir is challenging. Moreover, the optimization process often relies on a single evolutionary algorithm, which can yield varying results across different cases. To address these limitations, this paper introduces a novel approach called the multi-surrogate framework with an adaptive selection mechanism (MSFASM) to tackle production optimization problems. MSFASM consists of two stages. In the first stage, a reduced-dimensional broad learning system (BLS) is used to adaptively select the evolutionary algorithm with the best performance during the current optimization period. In the second stage, the multi-objective algorithm, non-dominated sorting genetic algorithm II (NSGA-II), is used as an optimizer to find a set of Pareto solutions with good performance on multiple surrogate models. A novel optimal point criterion is utilized in this stage to select the Pareto solutions, thereby obtaining the desired development schemes without increasing the computational load of the numerical simulator. The two stages are combined using sequential transfer learning. From the two most important perspectives of an evolutionary algorithm and a surrogate model, the proposed method improves adaptability to optimization problems of various reservoir types. To verify the effectiveness of the proposed method, four 100-dimensional benchmark functions and two reservoir models are tested, and the results are compared with those obtained by six other surrogate-model-based methods.
