The Calocybe species possess notable economic and medicinal value, demonstrating substantial potential for resource utilization. The taxonomic studies of Calocybe are lacking in quality and depth. Based on the specimens collected from northeast China, this study provides a detailed description of two newly discovered species, namely Calocybe betulicola and Calocybe cystidiosa, as well as two commonly found species, Calocybe decolorata and Calocybe ionides. Additionally, a previously unrecorded species, C. decolorata, has recently been discovered in Jilin Province, China. The two newly discovered species can be accurately distinguished from other species within the genus Calocybe based on their distinct morphological characteristics. The primary distinguishing features of C. betulicola include its grayish-purple pileus, grayish-brown to dark purple stipe, smaller basidiomata, absence of cellular pileipellis, and its habitat on leaf litter within birch forests. Calocybe cystidiosa is distinguished by its growth on the leaf litter of coniferous forests, a flesh-pink pileus, a fibrous stipe with a white tomentose covering at the base, non-cellular pileipellis, larger basidiospores, and the presence of cheilocystidia. The reconstruction of phylogenetic trees using combined ITS, nLSU, and tef1-α sequences, employing maximum likelihood and Bayesian inference analyses, showed that C. betulicola formed a cluster with C. decurrens, while C. cystidiosa clustered with C. vinacea. However, these two clusters formed separate branches themselves, which also supported the results obtained from our morphological studies. A key to the Calocybe species reported from northeast China is provided to facilitate future studies of the genus.

The genus Gymnopus plays a significant role in ecological systems, with certain species holding potential as food or medicinal resources. However, the species diversity of Gymnopus in China remains unclear. In recent years, more than one thousand Gymnopus specimens have been collected across China. Thus, through the integration of ecological evidence, detailed morphological studies, and phylogenetic analysis using a multiloci dataset of ITS + nLSU + tef1-ɑ, four new species—Gymnopus longistipes, Gymnopus striatipileatus, Gymnopus viridiscus, and Gymnopus spadiceus—have been differentiated from known species. Gymnopus similis has been newly documented from Jiangxi Province, China. Detailed descriptions and vivid illustrations have been provided based on the newly collected specimens, along with comparisons to closely related species. Additionally, a key to the reported species of Gymnopus s.l. from East China has been included.

Neolentinus is a significant genus, belonging to Gloeophyllaceae, with important economic and ecological values, which are parasites on decaying wood of broad-leaf or coniferous trees, and will cause brown rot. However, the taxonomic study is lagging behind to other groups of macrofungi, especially in China. In view of this, we conducted morphological and molecular phylogenetic studies on this genus. We have discovered new types of cheilocystidia and with extremely long lamellae in Neolentinus, and, thus proposed it as a new species—Neolentinus longifolius. At the same time, we clarified the distribution of Neolentinus cyathiformis in China and provided a detailed description. Moreover, we also described two common species, viz. Neolentinus lepideus and Neolentinus adhaerens. All the species are described based on the Chinese collections. The key to the reported species of Neolentinus from China is provided. And the phylogeny of Neolentinus from China is reconstructed based on DNA sequences of multiple loci including the internal transcribed spacer (ITS) regions, the large subunit nuclear ribosomal RNA gene (nLSU), and the translation elongation factor 1-α gene (tef-1α). In addition, full morphological descriptions, illustrations, color photographs, taxonomic notes, and all the available sequences of Neolentinus species are provided.

In this contribution to the genus Craterellus in northern China, two new species are introduced: Craterellus connatus and C. striatus. These species and C. atrobrunneolus, initially described in south-western China, are highly similar and closely related. The species delimitation is molecularly supported by multigene phylogenetic analysis of the nr LSU and tef-1α region. Craterellus connatus is characterised by its medium-sized basidiomata, greyish-brown and smooth pileus with an off-white margin, the hymenophore with a strongly anastomosing vein, turning khaki upon drying, connate stipe, broad ellipsoid to ellipsoid basidiospores (6.1–7.8 × 4.8–5.9 μm), slender basidia with (2)4–6 sterigmata and the absence of clamp connection. Craterellus striatus is characterised by its small-sized basidiomata, fibrillose, greyish-brown to yellowish-brown, fully perforated pileus with a brown fringe, the hymenophore with a forking vein, the stipe inflated at the base, broad ellipsoid to ellipsoid basidiospores (6.8–8.0 × 5.1–6.0 μm), 2–6 spored basidia, encrusted hyphae and the absence of clamp connection. Detailed macroscopic and microscopic descriptions, accompanied by illustrations and a taxonomic discussion, are presented. A key to the Chinese Craterellus species is also provided.

Boletaceae, the largest family in Boletales, has been attracted by mycologists in the world due to its diverse morphology and complex history of evolution. Although considerable work has been done in the past decades, novel taxa are continually described. The current study aimed to introduce three new taxa and one new record of Boletaceae from China. The morphological descriptions, color photographs, phylogenetic trees to show the positions of the taxa, and comparisons with allied taxa are provided. The new genus Hemilanmaoa is unique in the Pulveroboletus group, and Hemilanmaoa retistipitatus was introduced as the type species. It can be distinguished by its bluing basidioma when injured, a decurrent hymenophore, a stipe covered with distinct reticulations, and a fertile stipitipellis. Porphyrellus pseudocyaneotinctus is characterized by its pileipellis consisting of broadly concatenated cells and thin-walled caulocystidia in Porphyrellus. In Phylloporus, Phylloporus biyangensis can be distinguished by its hymenophores that change to blue when injured and yellow basal mycelium. Lanmaoa angustispora, as a new record, is first reported in Northern China. Internal transcribed spacer (ITS), 28S rDNA (28S), translation elongation factor 1-alpha (tef1-α), RNA polymerase II subunit 1 (rpb1), and RNA polymerase II subunit 2 (rpb2) were employed to execute phylogenetic analyses.

Xinjiang Uyghur Autonomous Region in China embraces a unique geographical and ecological environment, and the macrofungi represent a rich resource. However, few studies on the genus Pluteus have been reported from Xinjiang. In 2021, the macrofungal resources in Xinjiang were surveyed, and 10 specimens belonging to the genus Pluteus were collected. Based on the morphological study and molecular analysis, three species were recognized, P. aletaiensis, P. brunneidiscus, and P. hongoi. Pluteus aletaiensis is proposed as a new species. It is characterized by its bright yellow lamellae and stipe, brittle texture, subfusiform to vesicular pleurocystidia, with short pedicels to broadly lageniform to obtuse at apices, a hymeniderm pileipellis, containing dark brown intracellular pigment, and it grows on the ground. Pluteus brunneidiscus, a new record to China, is characterized by uneven, smooth, grayish brown to brown pileus, with an entire margin, and pointed or flatter apices intermediate cystidia, without apical hooks. Pluteus hongoi, a new record to Xinjiang Uyghur Autonomous Region, China, is characterized by the apical hook’s structure (commonly bifid) of pleurocystidia. The nuclear internal transcribed spacer (nrITS) and translation elongation factor 1-alpha (TEF1-a) region were used for the molecular analysis. Phylogenetic trees were constructed using both the maximum likelihood analysis (ML) and Bayesian inference (BI). Detailed descriptions of the three species are presented herein. Finally, a key to the list of eight species of the genus Pluteus knew from Xinjiang is provided.

Gymnopus sect. Impudicae is a poorly studied group around the world. However, it is well known for its pungent smell—a total of five species from China belonging to sect. Impudicae were recorded, and included four species new to science—G. epiphyllus, G. cystidiosus, G. subdensilamellatus, and G. subpolyphyllus—which were delimited and proposed based on morphological and molecular evidences, and one new record from Henan, Jiangxi, and Gansu Province, China—G. densilamellatus. Detailed descriptions and illustrations were presented as well as comparisons to similar species. Phylogenetic analysis inferred from the ITS and nLSU dataset supported the Gymnopus as a monophyletic genus which was defined by Oliveira et al., and the novel species grouped as separate lineages within it. A Key to the reported species of Gymnopus sect. Impudicae is also provided.

Cordyceps species are notable medicinal fungi in China, which are pathogenic on insects and exhibit high biodiversity in tropical and subtropical regions. Recently, three new Cordyceps species, Cordyceps changchunensis and Cordyceps jingyuetanensis growing on pupae of Lepidoptera and Cordyceps changbaiensis growing on larvae of Lepidoptera, were found in Jilin Province, China and are described, based on morphological and ecological characteristics. These three new species are similar to the Cordyceps militaris group, but are distinctly distinguishable from the known species. Cordyceps changchunensis, characterised by its small and light yellow to orange stromata which is occasionally forked, covered with white mycelium at the base of stipe, globose to ovoid perithecia, is macroscopically similar to Cordyceps militaris. Cordyceps changbaiensis is clearly discriminated from other Cordyceps species by its white to orange and branched stromata, clavate to cylindrical fertile apical portion, immersed and globose to ovoid perithecia. Moreover, unbranched, clavate and orange to light red stromata, almond-shaped to ovoid and immersed perithecia separate Cordyceps jingyuetanensis from other Cordyceps species. nrITS, nrLSU and EF-1α sequences were undertaken and phylogenetic trees, based on Maximum Likelihood and Bayesian Inference analysis showed that the three new species clustered with Cordyceps militaris, but formed individual clades, as well as confirmed the results of our morphological study.

To explore the corrosion properties of magnesium alloys, the chemical behavior of a high strength Mg97Zn1Y2-1wt.%SiC alloy in different corrosion environments was studied. Three solutions of 0.2 mol·L-1 NaCl, Na2SO4 and NaNO3 were selected as corrosion solutions. The microstructures, corrosion rate, corrosion potential, and mechanism were investigated qualitatively and quantitatively by optical microscopy (OM), scanning electron microscopy (SEM), immersion testing experiment, and electrochemical test. Microstructure observation shows that the Mg97Zn1Y2-1wt.%SiC alloy is composed of α-Mg matrix, LPSO (Mg12ZnY) phase and SiC phase. The hydrogen evolution and electrochemical test results reflect that the Mg97Zn1Y2-1wt.%SiC in 0.2 mol·L-1 NaCl solution has the fastest corrosion rate, followed by Na2SO4 and NaNO3 solutions, and that the charge-transfer resistance presents the contrary trend and decreases in turn.

Macrofungi are essential in forest ecological functioning. Their distribution and diversity are primarily impacted by vegetation, topography, and environmental factors, such as precipitation and temperature. However, the composition and topographical changes of the macrofungi between the eastern mountainous area and central plains of Jilin Province are currently unknown. For this study, we selected six investigational sites representing three different topographical research sites in Jilin Province to assess macrofungal diversity, and applied a quadrat sampling method. Macro- and micro-morphological characteristics combined with the molecular method were used to identify the collected macrofungi. Meanwhile, selected meteorological data were obtained for statistical analysis. As a result, 691 species were identified, of which Agarics were the most common, accounting for 60.23%, while the Cantharelloid fungi were the least common (0.91%). Furthermore, most of the shared genera (species) were saprophytic. The α diversity showed that the species diversity and richness in Longwan National Forest Park (B2) were the highest at the genus level. The mycorrhizal macrofungi proportion revealed that Quanshuidong Forest Farm (A1) was the healthiest. Finally, species composition similarity decreased with the transition from mountainous to hilly plains. We concluded that the occurrence of macrofungi was most influenced by vegetation. The air humidity, precipitation, and wind velocity were also found to significantly impact the occurrence of macrofungi. Finally, the mycorrhizal:saprophytic ratios and species similarity decreased with the transition from the mountainous area to the plains. The results presented here help elucidate the macrofungi composition and their relationship with environmental factors and topography in Jilin Province, which is crucial for sustainable utilization and future conservation.

This paper reports a film-cooling effectiveness experiment on a turbine blade platform that combines two film hole shapes with three layouts. A linear cascade using the pressure-sensitive paint (PSP) technique was employed to measure the adiabatic film effectiveness and discharge coefficients. Three film hole layouts, including two double-row layouts and one dispersed layout, were designed based on the platform configurations. One double-row layout arranges both rows of holes on the pressure side. Another double-row layout arranges one row on the pressure side and one row on the suction side. The dispersed layout was designed with streamwise multirows using the same number of holes. A fan-shaped hole and a diffusion slot hole were tested and compared. The experiments were conducted at a mainstream Reynolds number of 7 x 105, a mainstream turbulence intensity of 3.6%, and a coolant-to-mainstream density ratio of 1.5. The blowing ratio ranged from 0.5 to 2.5. The results demonstrated that regardless of the hole shape, the dispersed layout performed better than the two double-row layouts. However, the effects of the layout on the film effectiveness and discharge coefficients are smaller for the diffusion slot hole. In the three layouts, the film effectiveness of the diffusion slot holes is remarkably greater than that of the fan-shaped holes, and the superiority increases as the blowing ratio increases. Alloys with long-period stacking ordered structures (LPSO) have good properties and are highly regarded. Mg-Zn-Y alloy containing LPSO phase was prepared by the traditional casting method, and the aging heat treatment was performed at different temperatures and times. The microstructure and phase constitutions of the alloy were observed by means of optical microscopy and scanning electron microscopy methods. Results show the microstructure of as-cast Mg95.5Zn1.5Y3 mainly consists of α-Mg, W phases and LPSO phases. During the aging treatment, fine lamellar-shaped 14H-LPSO phase is formed at the grain boundaries and precipitates from the supersaturated magnesium matrix, and the volume fraction increases as the aging time increases. By controlling the aging time, Mg-Zn-Y alloys with different volume fractions of 14H-LPSO phase were prepared. The aging kinetics equation of the 14H-LPSO phase is summarized, that is f = 1-exp(-0.2705 t0.6368). The phase transformation mechanism of 14H-LPSO in Mg95.5Zn1.5Y3 alloy can be described as the change of dislocation energy. [ABSTRACT FROM AUTHOR]

This study aimed to evaluate the antimicrobial activities and mechanism of sturgeon spermary protein extracts (SSPE) against Escherichia coli. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were determined. Cell structural change was analyzed using scanning electron microscopy-energy dispersive X-ray spectrometry and transmission electron microscope. Moreover, pH, zeta potential, membrane potential, intracellular ATP concentrations and the interaction of SSPE with genomic DNA were analyzed. Results showed that molecular weight of SSPE is 13.4 kDa, the content of basic amino acids is the highest, in which arginine accounts for 73.2%. The MIC and MBC of SSPE for E. coli were 0.05 and 5 mg/mL, respectively. After SSPE treatment, cell membrane permeability changes, zeta potential decrease and genomic DNA lysis occurred in E. coli, which indicated it exerted bacteriostatic effects either independently or simultaneously by destroying the cell membrane and genomic DNA. These findings indicated that SSPE has potential to be a natural antiseptic.

Icariside I, the glycosylation product of icaritin, is a novel effective anti-cancer agent with immunological anti-tumor activity. However, very limited natural icariside I content hinders its direct extraction from plants. Therefore, we employed a computer-aided protein design strategy to improve the catalytic efficiency and substrate specificity of the α-L-rhamnosidase from Thermotoga petrophila DSM 13995, to provide a highly-efficient preparation method. Several beneficial mutants were obtained by expanding the active cavity. The catalytic efficiencies of all mutants were improved 16–200-fold compared with the wild-type TpeRha. The double-point mutant DH was the best mutant and showed the highest catalytic efficiency (kcat/KM: 193.52 s−1 M−1) against icariin, which was a 209.76-fold increase compared with the wild-type TpeRha. Besides, the single-point mutant H570A showed higher substrate specificity than that of the wild-type TpeRha in hydrolysis of different substrates. This study provides enzyme design strategies and principles for the hydrolysis of rhamnosyl natural products.

Given the rapid development of genome mining in this decade, the substrate channel of paclitaxel might be identified in the near future. A robust microbial cell factory with gene dbat, encoding a key rate-limiting enzyme 10-deacetylbaccatin III-10-O-transferase (DBAT) in paclitaxel biosynthesis to synthesize the precursor baccatin III, will lay out a promising foundation for paclitaxel de novo synthesis. Here, we integrated gene dbat into the wild-type Escherichia coli BW25113 to construct strain BWD01. Yet, it was relatively unstable in baccatin III synthesis. Mutant gene dbatS189V with improved thermostability was screened out from a semi-rational mutation library of DBAT. When it was over-expressed in an engineered strain N05 with improved acetyl-CoA generation, combined with carbon source optimization of fermentation engineering, the production level of baccatin III was significantly increased. Using this combination, integrated strain N05S01 with mutant dbatS189V achieved a 10.50-fold increase in baccatin III production compared with original strain BWD01. Our findings suggest that the combination of protein engineering and metabolic engineering will become a promising strategy for paclitaxel production.

Electron-beam irradiation (EBI) is an efficient, safe, and nonthermal sterilization technique that is extensively used in food preservation research. Here we report the effects of different EBI doses (0, 4, 8 kGy) and preservation temperatures (room temperature [RT], 4 °C) on the muscle water distribution and muscle quality indices of silver carp chunks (SCCs). The highest entrapped water content was found in the 4-kGy-irradiated/4-°C-stored samples. The expressible moisture content (EMC) of the SCCs increased with increasing irradiation dose and was significantly lower in the RT group than in the 4 °C group. The irradiation dose and preservation temperature had no significant effect on the moisture content, whiteness value and protein content of SCCs (p > 0.05). When the irradiation dose reached 8 kGy, AV value, POV value and TVB value were significantly increased (p < 0.05). The myofibrillar protein content and actomyosin content of the SCCs in the 4 °C group was higher than that of the specimens in the RT group by 0.29–0.98 mg/mL (p < 0.05) and 36.21–296.58 μg/mL (p < 0.05), respectively. Overall, EBI treatment (4 kGy) and low-temperature preservation (4 °C) helped retain the muscle water content of the SCCs and preserve their quality, thereby endorsing the EBI treatment of silver carp products.

Bone remodeling is a process delicately balanced between osteoclastic bone resorption and osteoblastic bone formation. Osteoclasts (OCs) are multinucleated giant cells formed through the fusion of monocytic precursors of the hematopoietic stem cells lineage. OCs are the exclusive cells responsible for the resorption and degradation of the mineralized bone matrix. Pantoprazole (PPZ), a proton pump inhibitor (PPI), is commonly prescribed to reduce excess gastric acid production for conditions such as gastroesophageal reflux disease and peptic ulcer disease. Studies have found contradictory effects of PPI therapy on bone metabolism due to the lack of understanding of the exact underlying mechanism. In this study, we found that PPZ inhibits receptor activator of nuclear factor-κB (NF-κB) ligand- (RANKL-) induced osteoclastogenesis from bone marrow monocytic/macrophage (BMMs) precursors and the bone-resorbing activity of mature OCs. Correspondingly, the expression of OC marker genes was also attenuated. At the molecular level, PPZ treatment was associated with reduced activation of the ERK MAPK signaling pathways crucial to OC differentiation. Additionally, the in vivo administration of PPZ protected mice against lipopolysaccharide- (LPS-) induced inflammatory calvarial bone erosion, as a result of the reduced number and activity of OCs on the calvarial bone surface. Although PPI use is associated with increased risk of osteoporosis and bone fractures, our study provides evidence for the direct inhibitory effect of PPZ on OC formation and bone resorption in vitro and in vivo, suggesting a potential therapeutic use of PPZ in the treatment of osteolytic disease with localized bone destruction.
