26. Nucleus accumbens D1/D2 circuits control opioid withdrawal symptoms in mice

Abstract

The nucleus accumbens (NAc) is the most promising target for drug use disorder treatment. Deep brain stimulation (DBS) of NAc is effective for drug use disorder treatment. However, the mechanisms by which DBS produces its therapeutic effects remain enigmatic. Here, we define a behavioral cutoff criterion to distinguish depressive-like behaviors and nondepressivelike behaviors in mice after morphine withdrawal. We identified a basolateral amygdala (BLA) to NAc D1 medium spiny neuron (MSN) pathway that controls depressive-like behaviors after morphine withdrawal. Furthermore, the paraventricular nucleus of thalamus (PVT) to NAc D2 MSN pathway controls naloxone-induced acute withdrawal symptoms. Optogenetically induced long-term potentiation with K-opioid receptor (KOR) antagonism enhanced BLA to NAc D1 MSN signaling and also altered the excitation/inhibition balance of NAc D2 MSN signaling. We also verified that a new 50 Hz DBS protocol reversed morphine withdrawal-evoked abnormal plasticity in NAc. Importantly, this refined DBS treatment effectively alleviated naloxone-induced withdrawal symptoms and depressive-like behaviors and prevented stress-induced reinstatement. Abstract

Background: Although current studies have identified sleep disorders as an independent risk factor for suicide, the relationship between sleep disorders and suicide risk has not been well established. This study explored whether anxiety and depressive symptoms are used as mediators to participate in the impact of sleep quality on suicide risk. Methods: This is a cross-sectional study. We administered a psychological questionnaire to the participants, using a combination of self-assessment and psychiatrist assessment.Sleep quality, suicide risk, level of anxiety and depressive symptoms were assessed by PSQI, NGASR, SAS and SDS.The study subjects were 391 hospitalized COVID-19 patients from Wuhan hospitals. We used model 6 in the PROCESS (version 3.5) plug-in of SPSS software to conduct mediation test with sleep quality as the independent variable, suicide risk as the dependent variable, level of anxiety and depressive symptoms as intermediate variables. Results: The severity of anxiety and depressive symptoms and the risk of suicide in the sleep disorder group (63.15 ± 13.71, 59.85 ± 13.38, 6.52 ± 3.67) were higher than those in the non-sleep disorder group (49.83 ± 13.14, 44.87 ± 10.19, 2.87 ± 3.26) (P

Abstract

Nitrated aromatic compounds (NACs) and oxygenated derivatives of polycyclic aromatic hydrocarbons (OPAHs) play vital roles within brown carbon (BrC), influencing both climate dynamics and human health to a certain degree. The concentrations of these drug classes were analyzed in PM 2.5 from an urban area in Beijing during the autumn and winter of 2017 and 2018. There were four heavy haze episodes during the campaign, two of which happened prior to heating and the other two during heating. During the entire course of sampling, the mean total concentrations of the nine NACs and the eight OPAHs were 1.2–263 and 2.1–234 ng m -3 , respectively. The concentrations of both NACs and OPAHs were approximately 2–3 times higher in the heating period than before heating. For NACs, the relative molecular composition did not change significantly before and during heating, with 4-nitrocatechol and 4-nitrophenol demonstrating the highest abundance. For OPAHs, 1-naphthaldehyde was the most abundant species before and during heating, while the relative proportion of anthraquinone increased by more than twice, from 13 % before heating to 31 % during the heating. In Beijing's urban area during autumn and winter, significant sources of NACs and OPAHs have been traced back to automobile emissions and biomass burning activities. Interestingly, it was observed that the contribution from coal combustion increased notably during heating. It is worth noting that the secondary generation of BrC was important throughout the whole sampling period, which was manifested by the photochemical reaction before heating and the aqueous reaction during heating. It was further found that the haze in autumn and winter was nitrate-driven before heating and secondary organic carbon (SOC)-driven during heating, and the secondary formation of BrC increased significantly in pollution events, particularly during heating.
