The Australian gummy shark Mustelus antarcticus is distributed across southern and eastern Australian waters and is the main target of a large shark fishery. Commercial harvest of the species is considered sustainable based on biomass estimates that show recovery from past overexploitation. However, the effective population size and genetic-based assessments of stock structure remain unresolved. We evaluated the genetic structure and effective population size (Ne) of the gummy shark using genome-wide single nucleotide polymorphisms. We found genetic divergence between individuals from the east coast and those along the south coast, resulting in 2 discrete populations with signatures of adaptive divergence. Spatial analyses revealed widespread gene flow within each of these populations, with some evidence for mild isolation-by-distance observed using individual-based tests. Demographic modelling of each population showed a comparatively rapid decline of Ne in the most recent past compared to more historical projections, with evidence that such declines have occurred to the point where genetic variation could be at risk. The identification of 2 divergent populations of gummy shark suggests that management should consider each individually to ensure the long-term sustainability of the species.

Members of the smoothhound shark genus Mustelus display a widespread distribution pattern across ocean basins with a high degree of sub-regional endemism. The patterns and processes that resulted in smoothhound biodiversity and present-day distribution remain largely unknown. We infer the phylogenetic relationships of the genus Mustelus, based on sequence data (3474 bp) from three mitochondrial genes (CR, NADH-2 and 12S-16SrRNA) and a nuclear gene (KBTBD2) from seven species of Mustelus distributed across the eastern Atlantic- and Indo-Pacific oceans. Using the CR and KBTBD2 dataset, we infer the phylogeographic placement of Old World Mustelus, with particular reference to species from southern Africa. Using a near-complete phylogeny of the genus including Old World and New World species of Mustelus and publicly available sequences of the NADH-2 gene, we found supporting evidence indicating a major cladogenic event separating placental and aplacental species. Biogeographical analyses further revealed that the radiation of Mustelus in the southern African region was driven primarily by long-distance dispersal during the upper Miocene to lower Pleistocene. The placement of the placental blackspotted smoothhound Mustelus punctulatus at the base of the placental non-spotted clade suggests the secondary loss of black spots in the genus, and this was also supported by the ancestral state reconstruction. The results furthermore suggest that the Southern Hemisphere species of the genus arose from multiple separate dispersal events from the Northern Hemisphere which is in line with the earliest record of Mustelus in the Northern Hemisphere.

The shark genus Mustelus is speciose, commercially important and systematically troublesome. We use a molecular approach combining inter and intra-specific data to investigate Mustelus species in the central Indo-Pacific and Australasia. Our analysis supports two Mustelus clades, one comprising species with no white spots and a placental reproductive mode and a second clade of white spotted, aplacental species. Levels of genetic divergence are low, especially among species in the white spotted, aplacental clade and this should be taken into account when employing molecular data to delineate species. Our data support the hypothesis of a radiation following dispersal from a northern hemisphere ancestor. Molecular dating suggests that localised speciation in Australasia may have occurred during the Pleistocene. We propose that some of the difficulties associated with Mustelus systematics relate to a recent radiation, particularly in the Australasian region.

Genetic stock structure information is needed to delineate management units and mon- itor trade in sharks, many of which are heavily exploited and declining. The dusky shark Carcha - rhinus obscurus is a large apex predator that is sought after for its fins and is considered highly sus- ceptible to overexploitation. The International Union for the Conservation of Nature (IUCN) classifies this species as 'Vulnerable' globally and 'Endangered' in the northwest Atlantic. We make the first assessment of global stock structure of C. obscurus by analyzing part of the mitochondrial control region (mtCR) in 255 individuals sampled from 8 geographically dispersed locations. We found 25 mtCR haplotypes and rejected a null hypothesis of panmixia (analysis of molecular variance, ΦST = 0.55, p < 0.000001), detecting significant differentiation between 3 management units: US Atlantic (USATL), South Africa (SAF), and Australia (AUS). We also found preliminary evidence of population structure between the USATL and southwest Atlantic (Brazil). There were no shared haplotypes between the western Atlantic and Indo-Pacific. These analyses suggest that replenishment of the col- lapsed USATL management unit via immigration of females from elsewhere is unlikely. Mixed stock analysis (MSA) simulations show that reconstruction of the relative contributions of USATL, SAF, and AUS management units to the Asian fin trade is possible using these mtCR sequences. We suggest avenues for obtaining samples to conduct MSA of the shark fin trade, which could enhance manage- ment of dusky sharks and other species that are exploited for their fins.

This study shows a range extension for the Australian blacktip shark Carcharhinus tilstoni, which was believed to be restricted to Australia's tropical waters, of >1000 km into temperate waters, revealing its vulnerability to a wider commercial fishery.

The development of genetic markers for studies of population structure and genetic diversity can be applied to improving management and monitoring of commercially fished species. Here we report on the use of high-throughput (454) sequencing to develop a panel of 12 novel polymorphic microsatellite markers for the commercially harvested gummy shark, Mustelus antarcticus. This constitutes the first suite of microsatellites developed for the genus Mustelus, a group which features in commercial fisheries around the world. We demonstrate the successful amplification of these markers in other species of Mustelus and test their utility in species from an additional nine genera of shark.

Elasmobranchs (sharks and rays) show an amazing diversity of reproductive modes and behaviors. Multiple paternity (MP) has been identified in all species where more than 1 litter has been investigated; yet neither direct nor indirect benefits from MP have been determined in elasmobranchs. This has led to the suggestion that MP in this group may simply be a product of convenience polyandry with variation in the frequency of MP driven by differences in mate encounter rates. Here, we use molecular markers to investigate polyandry and MP in 2 closely related and commercially important species of shark, Mustelus antarcticus and Mustelus lenticulatus. In total, 328 M. antarcticus embryos originating from 29 different mothers and 75 M. lenticulatus embryos originating from 19 different mothers were genotyped using 8 microsatellite loci. We find that MP occurs in both species. However, in both species, the majority of litters were sired by a single father. Our results do not support increased fecundity per se as a driver of MP. Further, our results do not suggest that high population densities with resulting high mate encounter rates generated by breeding aggregations necessarily lead to high frequencies of MP. Importantly, we note evidence of reproductive skew within polyandrous litters, which is a predicted outcome of postcopulatory mechanisms.

The copper or bronze whaler shark (Carcharhinus brachyurus) is a large, coastal top predator that is vulnerable to overexploitation. We test the null hypothesis that copper sharks are panmictic throughout the southern hemisphere. We analysed part of the mitochondrial control region (mtCR) in 120 individuals from eight sampling areas, defining 20 mtCR haplotypes (h = 0.76 ± 0.06, π = 0.016 ± 0.0007).
