This study explores the response of flow around isolated cuboid buildings to variations in the incoming turbulence arising from changes in atmospheric boundary layer (ABL) stability using a building-resolving large-eddy simulation (LES) technique with explicit representation of building effects through an immersed body force method. An extensive suite of LES for a neutral ABL with different model resolution and advection scheme configurations reveals that at least 6, 12, and 24 grid points per building side are required in order to resolve building-induced vortex shedding, mean-flow features, and turbulence statistics, respectively, with an advection scheme of a minimum of third order. Using model resolutions that meet this requirement, 21 building-resolving simulations are performed under varying atmospheric stability conditions, from weakly stable to convective ABLs, and for different building sizes (H), resulting in LABL/H ≈ 0.1–10, where LABL is the integral length scale of the incoming ABL turbulence. The building-induced flow features observed in the canonical neutral ABL simulation, e.g., the upstream horseshoe vortex and the downstream arch vortex, gradually weaken with increasing surface-driven convective instability due to the enhancement of background turbulent mixing. As a result, two local turbulence kinetic energy peaks on the lateral side of the building in nonconvective cases are merged into a single peak in strong convective cases. By considering the ABL turbulence scale and building size altogether, it is shown that the building impact decreases with increasing LABL/H, as coherent turbulent structures in the ABL become more dominant over a building-induced flow response for LABL/H > 1.

Fugitive natural gas leaks from abnormal operations or failed containment at oil and gas production fields and gas supply chains are a significant source of atmospheric methane (CH4), a potent greenhouse gas. This realization of significant CH4 leaks has stimulated efforts at mitigation using cost effective detection methods. US DOE's ARPA-E MONITOR program has supported transformational research to locate and quantify fugitive methane leaks at natural gas facilities in order to achieve a 90% reduction in CH4 emissions. This paper describes the development, application, and evaluation of a MONITOR program sponsored source attribution technology solution, wherein an artificial neural network (ANN) infers as outputs, a leak location and release rate given inputs of measured sonic anemometer wind velocities and methane sensor time series. We discuss development of our ANN model, its training, computational implementation and use to design effective sampling strategies over a range of source and sensor locations, and meteorological conditions. We report on the deployment and performance of our ALFaLDS in blind tests at Colorado State University's METEC well pad facility in Ft. Collins. Test results demonstrate that our ALFaLDS locates the engineered methane leaks to better than 8% of the domain length scale and has the ability to quantify fluxes, more accurately at high flux rates, less so at low rates. Our ANN overestimates leak source rates by an average scale factor of about 1.83 for small pads and by an average of 1.77 for larger domains. The scale factor can be used operationally but needs more research to assess its generality. This capability of leak location with high skill, speed and accuracy at moderate cost promises new automatic affordable sampling of fugitive gas leaks at well pads and oil and gas fields.

Multiscale modeling of a diurnal cycle of real-world conditions is presented for the first time, validated using data from the CWEX-13 field experiment. Dynamical downscaling from synoptic-scale down to resolved three-dimensional eddies in the atmospheric boundary layer (ABL) was performed, spanning 4 orders of magnitude in horizontal grid resolution: from 111 km down to 8.2 m (30 m) in stable (convective) conditions. Computationally efficient mesoscale-to-microscale transition was made possible by the generalized cell perturbation method with time-varying parameters derived from mesoscale forcing conditions, which substantially reduced the fetch to achieve fully developed turbulence. In addition, careful design of the simulations was made to inhibit the presence of under-resolved convection at convection-resolving mesoscale resolution and to ensure proper turbulence representation in stably-stratified conditions. Comparison to in situ wind-profiling lidar and near-surface sonic anemometer measurements demonstrated the ability to reproduce the ABL structure throughout the entire diurnal cycle with a high degree of fidelity. The multiscale simulations exhibit realistic atmospheric features such as convective rolls and global intermittency. Also, the diurnal evolution of turbulence was accurately simulated, with probability density functions of resolved turbulent velocity fluctuations nearly identical to the lidar measurements. Explicit representation of turbulence in the stably-stratified ABL was found to provide the right balance with larger scales, resulting in the development of intra-hour variability as observed by the wind lidar; this variability was not captured by the mesoscale model. Moreover, multiscale simulations improved mean ABL characteristics such as horizontal velocity, vertical wind shear, and turbulence.

Mesoscale models are considered to be the state of the art in modeling mountain-wave flows. Herein, the authors investigate the role and accuracy of planetary boundary layer (PBL) parameterizations in handling the interaction between large-scale mountain waves and the atmospheric boundary layer. To that end, recent large-eddy simulation (LES) results of mountain waves over a symmetric two-dimensional bell-shaped hill are used and compared to four commonly used PBL schemes. It is found that one-dimensional PBL parameterizations produce reasonable agreement with the LES results in terms of vertical wavelength, amplitude of velocity, and turbulent kinetic energy distribution in the downhill shooting-flow region. However, the assumption of horizontal homogeneity in PBL parameterizations does not hold in the context of these complex flow configurations. This inappropriate modeling assumption results in a vertical wavelength shift, producing errors of approximately 10 m s−1 at downstream locations because of the presence of a coherent trapped lee wave that does not mix with the atmospheric boundary layer. In contrast, horizontally integrated momentum flux derived from these PBL schemes displays a realistic pattern. Therefore, results from mesoscale models using ensembles of one-dimensional PBL schemes can still potentially be used to parameterize drag effects in general circulation models. Nonetheless, three-dimensional PBL schemes must be developed in order for mesoscale models to accurately represent complex terrain and other types of flows where one-dimensional PBL assumptions are violated.

The impact of atmospheric boundary layer (ABL) interactions with large-scale stably stratified flow over an isolated, two-dimensional hill is investigated using turbulence-resolving large-eddy simulations. The onset of internal gravity wave breaking and leeside flow response regimes of trapped lee waves and nonlinear breakdown (or hydraulic-jump-like state) as they depend on the classical inverse Froude number, Fr−1 = Nh/Ug, is explored in detail. Here, N is the Brunt–Väisälä frequency, h is the hill height, and Ug is the geostrophic wind. The results here demonstrate that the presence of a turbulent ABL influences mountain wave (MW) development in critical aspects, such as dissipation of trapped lee waves and amplified stagnation zone turbulence through Kelvin–Helmholtz instability. It is shown that the nature of interactions between the large-scale flow and the ABL is better characterized by a proposed inverse compensated Froude number, = N(h − zi)/Ug, where zi is the ABL height. In addition, it is found that the onset of the nonlinear-breakdown regime, ≈ 1.0, is initiated when the vertical wavelength becomes comparable to the sufficiently energetic scales of turbulence in the stagnation zone and ABL, yielding an abrupt change in leeside flow response. Finally, energy spectra are presented in the context of MW flows, supporting the existence of a clear transition in leeside flow response, and illustrating two distinct energy distribution states for the trapped-lee-wave and the nonlinear-breakdown regimes.

Emissions estimates of anthropogenic methane (CH 4 ) sources are highly uncertain and many sources related to energy production are localized yet difficult to quantify. Airborne imaging spectrometers like the next generation Airborne Visible/Infrared Imaging Spectrometer (AVIRIS-NG) are well suited for locating CH 4 point sources due to their ability to map concentrations over large regions with the high spatial resolution necessary to resolve localized emissions. AVIRIS-NG was deployed during a field campaign to measure controlled CH 4 releases at the Rocky Mountain Oilfield Testing Center (RMOTC) in Wyoming, U.S. for multiple flux rates and flight altitudes. Two algorithms were applied to AVIRIS-NG scenes, a matched filter detection algorithm and a hybrid retrieval approach using the Iterative Maximum a Posteriori Differential Optical Absorption Spectroscopy (IMAP-DOAS) algorithm and Singular Value Decomposition. Plumes for releases as low as 14.16 m 3 /h (0.09 kt/year) were consistently observed by AVIRIS-NG at multiple flight altitudes and images of plumes were in agreement with wind directions measured at ground stations. In some cases plumes as low as 3.40 m 3 /h (0.02 kt/year) were detected, indicating that AVIRIS-NG has the capability of detecting a wide range of fugitive CH 4 source categories for natural gas fields. This controlled release experiment is the first of its kind using AVIRIS-NG and demonstrates the utility of imaging spectrometers for direct attribution of emissions to individual point source locations. This is particularly useful given the large uncertainties associated with anthropogenic CH 4 emissions, including those from industry, gas transmission lines, and the oil and gas sectors.

Landscape heterogeneity shapes species distributions, interactions, and fluctuations. Historically, in dry forest ecosystems, low canopy cover and heterogeneous fuel patterns often moderated disturbances like fire. Over the last century, however, increases in canopy cover and more homogeneous patterns have contributed to altered fire regimes with higher fire severity. Fire management strategies emphasize increasing within-stand heterogeneity with aggregated fuel patterns to alter potential fire behavior. Yet, little is known about how such patterns may affect fire behavior, or how sensitive fire behavior changes from fuel patterns are to winds and canopy cover. Here, we used a physics-based fire behavior model, FIRETEC, to explore the impacts of spatially aggregated fuel patterns on the mean and variability of stand-level fire behavior, and to test sensitivity of these effects to wind and canopy cover. Qualitative and quantitative approaches suggest that spatial fuel patterns can significantly affect fire behavior. Based on our results we propose three hypotheses: (1) aggregated spatial fuel patterns primarily affect fire behavior by increasing variability; (2) this variability should increase with spatial scale of aggregation; and (3) fire behavior sensitivity to spatial pattern effects should be more pronounced under moderate wind and fuel conditions.

The current study focuses on coupled dynamics and resultant geometry of fireline segments of various ignition lengths. As an example, for ignition lines of length scales typical for field experiments, fireline curvature is the result of a competition between the head fire and the flanks of the fire. A number of physical features (i.e. buoyancy and wind field divergence for example) arise in and around an incipient fire that defines the shape and spreading pattern of the flame zone. These features are explored using a numerical atmospheric dynamics model HIGRAD, and wildfire combustion physics model FIRETEC. HIGRAD/FIRETEC was designed to investigate wildfires and their interactions with the environment. In this study, the model was used to simulate grass fires that were initiated with a finite length, straight ignition line in homogeneous fuels. The dynamic evolutions of these firelines were analyzed to understand the individual events that evolve a wildfire. By understanding each individual process and how it interacts with other processes, information can be extracted to develop a theory about the mechanisms that combine to produce the observed wildfire behavior. In the current study, the flow field in the region of the simulated fires developed structures consistent with multiple buoyancy-induced vortex pairs. The series of stream-wise vortex pairs produce a regular alternating pattern of up-wash and down-wash zones, which allow air to penetrate the flame zone through troughs created in downwash regions. Consequently, this periodicity in the flow field within the fire resulted in a pattern of residual combustion where prolonged burning occurred in the up-wash zones separated by near-complete fuel depletion in the downwash zones. Some explanation is provided for why increased ignition line length leads to increased rate of spread (ROS) with some asymptotic limit.

Remote activation of specific cells of a heterogeneous population can provide a useful research tool for clinical and therapeutic applications. Here, we demonstrate that photostimulation of gold nanorods (AuNRs) using a tunable near-infrared (NIR) laser at specific longitudinal surface plasmon resonance wavelengths can induce the selective and temporal internalization of calcium in HEK 293T cells. Biotin-PEG-Au nanorods coated with streptavidin Alexa Fluor-633 and biotinylated anti-His antibodies were used to decorate cells genetically modified with His-tagged TRPV1 temperature-sensitive ion channel and AuNRs conjugated to biotinylated RGD peptide were used to decorate integrins in unmodified cells. Plasmonic activation can be stimulated at weak laser power (0.7-4.0 W/cm(2) ) without causing cell damage. Selective activation of TRPV1 channels could be controlled by laser power between 1.0 and 1.5 W/cm(2) . Integrin targeting robustly stimulated calcium signaling due to a dense cellular distribution of nanoparticles. Such an approach represents a functional tool for combinatorial activation of cell signaling in heterogeneous cell populations. Our results suggest that it is possible to induce cell activation via NIR-induced gold nanorod heating through the selective targeting of membrane proteins in unmodified cells to produce calcium signaling and downstream expression of specific genes with significant relevance for both in vitro and therapeutic applications. Leadership-scale scientific simulations running as tens of thousands of tightly-coupled MPI processes are vulnerable to interruption due to a single process or node failure. Due to the dependence of each state calculation on the successful completion of each of the prior state calculations, checkpoint-restart is the most widely-used technique to achieve fault tolerance. To write a consistent view of distributed state as a checkpoint, applications typically synchronize and pause while writing data to persistent media. In this paper we present a transactional protocol that enables asynchronous distributed creation of checkpoint data sets, and describe the conditions under which it is beneficial. With simulations, we demonstrate that scientific applications exhibiting computational variance without frequent synchronization can use our protocol to either reduce run time by up to 27% or reduce required storage system capability by up to 40%.

We have recently reported that activation of tumor-specific T cells by subcutaneous vaccination with irradiated T9 glioma cells of syngeneic rats with a pre-existing, intracranial (i.c.) T9 glioma (T9+vaccination) promotes the mobilization of myeloid suppressor cells (MSC) which inhibit T cell function resulting in unregulated tumor progression. The current study investigated if this immunological paradigm could be recapitulated in T cell deficient rats, in other rat glioma models or using a dendritic cell (DC) vaccine. When nude rats were used in the T9+vaccination model, the level of MSC tumor infiltration remained low in vaccinated and control groups and there was no significant difference in tumor size between the groups. Increased tumor infiltration by MSC after vaccination with respective irradiated tumor cells was observed in the 9L, F98 and D74 gliomas. RT-2 tumors were markedly infiltrated with MSC regardless of vaccination. Enhanced tumor progression in response to immunization and T cell activation was observed in rats bearing F98 and D74 gliomas, although less pronounced than in the T9 model, and there was a trend for increased tumor size in the 9L glioma model. Increased MSC infiltrate and augmented T9 glioma growth were observed when DC pulsed with T9 cell lysate was used as a vaccine. These results suggest that MSC infiltration and unregulated tumor growth in response to vaccination is T cell-dependent; is not unique to the T9 glioma; and can be recapitulated with an alternate immunization approach.

The purpose of this report is to outline code and solution verification activities applied to HIGRAD, a Computational Fluid Dynamics (CFD) solver of the compressible Navier-Stokes equations developed at the Los Alamos National Laboratory, and used to simulate various phenomena such as the propagation of wildfires and atmospheric hydrodynamics. Code verification efforts, as described in this report, are an important first step to establish the credibility of numerical simulations. They provide evidence that the mathematical formulation is properly implemented without significant mistakes that would adversely impact the application of interest. Highly accurate analytical solutions are derived for four code verification test problems that exercise different aspects of the code. These test problems are referred to as: (i) the quiet start, (ii) the passive advection, (iii) the passive diffusion, and (iv) the piston-like problem. These problems are simulated using HIGRAD with different levels of mesh discretization and the numerical solutions are compared to their analytical counterparts. In addition, the rates of convergence are estimated to verify the numerical performance of the solver. The first three test problems produce numerical approximations as expected. The fourth test problem (piston-like) indicates the extent to which the code is able to simulate a 'mild' discontinuity,more » which is a condition that would typically be better handled by a Lagrangian formulation. The current investigation concludes that the numerical implementation of the solver performs as expected. The quality of solutions is sufficient to provide credible simulations of fluid flows around wind turbines. The main caveat associated to these findings is the low coverage provided by these four problems, and somewhat limited verification activities. A more comprehensive evaluation of HIGRAD may be beneficial for future studies.« less
