Abstract Energy system optimization models offer insights into energy and emissions futures through least-cost optimization. However, real-world energy systems often deviate from deterministic scenarios, necessitating rigorous uncertainty exploration in macro-energy system modeling. This study uses modeling techniques to generate diverse near cost-optimal net-zero CO2 pathways for the United States’ energy system. Our findings reveal consistent trends across these pathways, including rapid expansion of solar and wind power generation, substantial petroleum use reductions, near elimination of coal combustion, and increased end-use electrification. We also observe varying deployment levels for natural gas, hydrogen, direct air capture of CO2, and synthetic fuels. Notably, carbon-captured coal and synthetic fuels exhibit high adoption rates but only in select decarbonization pathways. By analyzing technology adoption correlations, we uncover interconnected technologies. These results demonstrate that diverse pathways for decarbonization exist at comparable system-level costs and provide insights into technology portfolios that enable near cost-optimal net-zero CO2 futures.

ABSTRACT Campylobacter infections are a leading cause of bacterial-derived gastroenteritis worldwide with particularly profound impacts on pediatric patients in low- and middle-income countries. It remains unclear how Campylobacter impacts these hosts, though it is becoming increasingly evident that it is a multifactorial process that depends on the host immune response, the gastrointestinal microbiota, various bacterial factors, and host nutritional status. Since these factors likely vary between adult and pediatric patients in different regions of the world, it is important that studies define these attributes in well-characterized clinical cohorts in diverse settings. In this study, we analyzed the fecal microbiota and the metabolomic and micronutrient profiles of asymptomatic and symptomatic pediatric patients in Colombia who were either infected or uninfected with Campylobacter during a case-controlled study on acute diarrheal disease. Here, we report that the microbiome of Campylobacter-infected children only changed in their abundance of Campylobacter spp. despite the inclusion of children with or without diarrhea. In addition to increased Campylobacter, computational models were used to identify fecal metabolites that were associated with Campylobacter infection and found that glucose-6-phosphate and homovanillic acid were the strongest predictors of infection in these pediatric patients, which suggests that colonocyte metabolism is impacted during infection. Despite changes to the fecal metabolome, the concentrations of intestinal minerals and trace elements were not significantly impacted by Campylobacter infection but were elevated in uninfected children with diarrhea.IMPORTANCEGastrointestinal infection with pathogenic Campylobacter species has long been recognized as a significant cause of human morbidity. Recently, it has been observed that pediatric populations in low- and middle-income countries are uniquely impacted by these organisms in that infected children can be persistently colonized, develop enteric dysfunction, and exhibit reduced development and growth. While the association of Campylobacter species with these long-term effects continues to emerge, the impact of infection on the gastrointestinal environment of these children remains uncharacterized. To address this knowledge gap, our group leveraged clinical samples collected during a previous study on gastrointestinal infections in pediatric patients to examine the fecal microbiota, metabolome, and micronutrient profiles of those infected with Campylobacter species and found that the metabolome was impacted in a way that suggests gastrointestinal cell metabolism is affected during infection, which is some of the first data indicating how gastrointestinal health in these patients may be affected.

Abstract Energy transition scenarios are characterized by increasing electrification and improving efficiency of energy end uses, rapid decarbonization of the electric power sector, and deployment of carbon dioxide removal (CDR) technologies to offset remaining emissions. Although hydrocarbon fuels typically decline in such scenarios, significant volumes remain in many scenarios even at the time of net-zero emissions. While scenarios rely on different approaches for decarbonizing remaining fuels, the underlying drivers for these differences are unclear. Here we develop several illustrative net-zero systems in a simple structural energy model and show that, for a given set of final energy demands, assumptions about the use of biomass and CO2 sequestration drive key differences in how emissions from remaining fuels are mitigated. Limiting one resource may increase reliance on another, implying that decisions about using or restricting resources in pursuit of net-zero objectives could have significant tradeoffs that will need to be evaluated and managed.

Abstract Supramolecular polymer networks contain non-covalent cross-links that enable access to broadly tunable mechanical properties and stimuli-responsive behaviors; the incorporation of multiple unique non-covalent cross-links within such materials further expands their mechanical responses and functionality. To date, however, the design of such materials has been accomplished through discrete combinations of distinct interaction types in series, limiting materials design logic. Here we introduce the concept of leveraging “nested” supramolecular crosslinks, wherein two distinct types of non-covalent interactions exist in parallel, to control bulk material functions. To demonstrate this concept, we use polymer-linked Pd2L4 metal–organic cage (polyMOC) gels that form hollow metal–organic cage junctions through metal–ligand coordination and can exhibit well-defined host-guest binding within their cavity. In these “nested” supramolecular network junctions, the thermodynamics of host-guest interactions within the junctions affect the metal–ligand interactions that form those junctions, ultimately translating to substantial guest-dependent changes in bulk material properties that could not be achieved in traditional supramolecular networks with multiple interactions in series.

Deep decarbonization requires fundamental changes in meeting energy service demands, with some efforts increasing overall costs. Examining abatement measures in isolation, however, fails to capture their interactive effects within the energy system. Here we show the abatement costs of decarbonization in the United States using an energy system optimization model to capture technological interactions, multi-decadal path dependence, and endogenous end-use technology selection. Energy-system-wide net-zero CO _2 -eq emissions are achieved in 2050 at a cost under $400 per tonne CO _2 -eq, led by emissions reductions in power generation, end-use electrification of ground transportation, space heating, and some industrial applications. Differences in mitigation costs and CO _2 geological storage potential lead to regional heterogeneities in mitigation rates and residual emissions. The marginal abatement cost curves show that additional decarbonization comes at higher incremental costs, this cost penalty decreases over time, and substantially greater abatement occurs in future time periods at the same abatement cost.

ABSTRACTCampylobacter jejuni colonizes the thick mucus layer lining the lower intestinal epithelium of hosts to promote commensalism in birds and animals or diarrheal disease in humans. C. jejuni displays higher swimming velocity as external viscosity increases, which likely benefits colonization and persistence in the intestinal mucus layer of hosts. C. jejuni produces one of the most structurally complex bacterial flagellar motors, which has been proposed to generate high torque for high swimming velocity. However, it is unknown how this flagellar motor alters output to impact swimming velocities in a viscosity-dependent manner. In this work, we identified viscosity-dependent determinant A (VidA) (Cjj81176_0996) and VidB (Cjj81176_1107) in modulating swimming velocity primarily in low-viscosity environments. C. jejuni ΔvidA cells were non-motile or swam slowly in low-viscosity media, but swam with high velocity similar to wild-type cells in both Newtonian and non-Newtonian media with high viscosity. These data indicate that VidA is required for swimming in low-viscosity environments. Suppressor mutants suggested that the lower swimming velocity of ΔvidA in low viscosity was due to unregulated activity of VidB, which we propose has a brake- or clutch-like activity to reduce swimming velocity in low viscosity. Unlike other bacterial flagellar brakes or clutches, we found no evidence for cyclic diguanylate monophosphate influencing VidB activity. Our analyses suggest that the mechanics of the C. jejuni flagellar motor has naturally evolved for high-velocity swimming in milieus with high viscosity and requires the unique combination of VidA and VidB to modulate its activity to slow swimming velocity in low-viscosity environments.IMPORTANCEBacteria can adapt flagellar motor output in response to the load that the extracellular milieu imparts on the flagellar filament to enable propulsion. In this work, we provide evidence that the mechanics of the C. jejuni flagellar motor has evolved to naturally promote high swimming velocity in high-viscosity environments. We found that C. jejuni produces VidA and VidB as auxiliary proteins to specifically affect flagellar motor activity in low viscosity to reduce swimming velocity. Our findings provide some of the first insights into different mechanisms that exist in bacteria to alter the mechanics of a flagellar motor, depending on the viscosity of extracellular environments.

Screening polymer electrolytes for batteries is extremely expensive due to the complex structures and slow dynamics. Here the authors develop a machine learning scheme to accelerate the screening and explore a space much larger than past studies.

Physical activity (PA) provides numerous health benefits for individuals with type 1 diabetes (T1D). However, the threat of exercise-induced hypoglycemia may impede the desire for regular PA. Therefore, we aimed to study the association between three common types of PA (walking, running, and cycling) and hypoglycemia risk in 50 individuals with T1D. Real-world data, including PA duration and intensity, continuous glucose monitor (CGM) values, and insulin doses, were available from the Tidepool Big Data Donation Project. Participants' mean (SD) age was 38.0 (13.1) years with a mean (SD) diabetes duration of 21.4 (12.9) years and an average of 26.2 weeks of CGM data available. We developed a linear regression model for each of the three PA types to predict the average glucose deviation from 70 mg/dl for the 2 h after the start of PA. This is essentially a measure of hypoglycemia risk, for which we used the following predictors: PA duration (mins) and intensity (calories burned), 2-hour pre-exercise area under the glucose curve (adjusted AUC), the glucose value at the beginning of PA, and total bolus insulin (units) within 2 h before PA. INTRODUCTION: We studied the relationship of acute ischemic stroke (AIS) large-vessel occlusion clot composition with vessel recanalization and preprocedure imaging. SUBJECTS AND METHODS: Individual clots from AIS patients who underwent mechanical thrombectomy (MT) between September 2016 and September 2018 were examined. Clot composition was analyzed histologically through a trichrome staining and image segmentation, and the area occupied by red blood cells (RBCs), fibrin, or mixed composition was quantified. RESULTS: Forty-three patients (65.4 ± 12.7 years, 39% of females) who underwent 92 retrieval passes (mean 2.14, range 1–6) were included in this study. Fibrin (44%) occupied the greatest area, followed by mixed composition (34%) and RBCs (22%). A stent retriever was deployed in 81% of cases, 20 patients (47%) achieved first-pass efficacy (FPE) (thrombolysis in cerebral infarction [TICI] 2b-3 after first pass), 41 (95%) achieved successful revascularization (TICI 2b-3), and 21 (49%) had good outcome (modified Rankin Scale [mRS] ≤2) at 90 days. Hyperdense artery sign (HAS) on initial computed tomography was correlated with mixed clot composition (P = 0.01) and lack of fibrin content (P = 0.03). In the univariate analysis, FPE was associated with RBC clot area, atrial fibrillation, and occlusion location but not with fibrin clot area, mixed clot area, stroke etiology, thrombectomy technique, distal emboli, or 90-day mRS. In the multivariate analysis, FPE was significantly correlated with low RBC clot area (odd ratio = 0.96, confidence interval [0.92.99], P = 0.034) but not with atrial fibrillation or location. CONCLUSION: Our results suggest that HAS is correlated with mixed clot composition and lower fibrin content and that lower RBC clot composition is associated with FPE in patients undergoing MT.

OBJECTIVE: Proper blood flow is essential for the maintenance of homeostasis for the human cerebrum. The dural venous sinuses comprise the dominant cerebral venous outflow path. Understanding the spatial configuration of the dural venous sinuses can provide valuable insight into several pathological conditions. Previously, only two-dimensional or cadaveric data have been used to understand cerebral outflow. For the first time, we applied three-dimensional rotational venography (3D-RV) to study and provide detailed quantitative morphological measurements of the terminal cerebral venous sinus system in several pathological states. SUBJECTS ANDMETHODS: Patients who underwent a 3D-RV procedure were identified by reviewing our local institution's endovascular database. Patients with high-quality angiographic images were selected. Eighteen patients were included (37.1 ± 3.8 years). Sinuses were divided into four segments, starting at the torcula and ending at the internal jugular vein. Segment length, 3D displacement, and cross-sectional area were measured. RESULTS: The transverse sinus (60.2 mm) was the longest segment, followed by the sigmoid sinus (55.1 mm). Cross-sectional areas were smallest at the middle of the transverse sinus (21.3 mm2) but increased at the sigmoid sinus (33.5 mm2) and at the jugular bulb (49.7 mm2). The only variation in displacements of venous flow was at the sigmoid-jugular junction, where 55% of cases had lateral displacements versus 45% medial, and 78% superior versus 22% inferior. CONCLUSIONS: We describe the terminal venous sinus system of patients with a variety of diagnoses, detailing segment length, cross-sectional area, and 3D path.

Summary: Achieving ambitious greenhouse gas mitigation targets will require technological advances and cost reductions in dispatchable carbon-free power generation sources that can provide load following flexibility to integrate high penetrations of variable wind and solar power. Several other sectors may be difficult to decarbonize and a net-zero or net-negative carbon economy may require the deployment of geologic carbon dioxide (CO2) storage. Utilizing CO2 as a working fluid for geothermal energy production and energy storage can achieve both goals: isolating CO2 from the atmosphere and providing valuable power system services to enable high penetrations of variable carbon-free electricity production. The use of CO2 as a working fluid facilitates access to low-grade heat in sedimentary basins, which are widely available and could allow for strategic citing near CO2 sources or where power system flexibility is needed. In this perspective piece, we summarize the state of knowledge for sedimentary basin CO2-geothermal, sometimes referred to as CO2 plume geothermal, and explore how it could support decarbonization of the energy sector. We also present the potential for using geologically stored CO2 for bulk energy storage which could provide valuable time-shifting and other services to the power grid. We explore the promise and challenges of these technologies, identify key research gaps, and offer a critical appraisal of the role that policy for a technology at the intersection of renewable energy, energy storage, and geologic CO2 storage may play in achieving broad deployment.

The wide scale deployment of variable renewable energy technologies (VREs) offers a pathway to decarbonize the electric grid. One challenge to reliably operating the grid is ensuring that sufficient generating capacity is available to meet demand at all hours. By determining an individual generator's contribution to resource adequacy based on its expected availability when power is needed, the capacity credit for these resources is estimated. The objective of this study is to quantify the contribution of VRE to resource adequacy as a function of VRE penetration, across several regions, technologies, and resources. A computational model was built using the effective load carrying capability (ELCC) method to calculate capacity credit values for regions spanning the contiguous United States. As the deployment of VRE increases, we show its marginal contribution to meeting peak load decreases, which in turn requires additional generating capacity to maintain reliability. In addition, a rapid approximation method is demonstrated to estimate solar and wind capacity credit, relying on the capacity factors during hours of peak net demand. We find that estimates with the lowest error relative to capacity credits calculated using the ELCC method occur using the average renewable resource capacity factors of the top net 10 demand hours, regardless of resource type. Using context-specific values for capacity credit can improve long-term decision making in generation capacity expansion, cultivating more economical long-term resource planning for deep decarbonization.

INTRODUCTION: Acute ischemic stroke (AIS) complicating cardiac interventions (CI) is well described. The use of mechanical thrombectomy (MT) for treatment of emergent large vessel occlusion (ELVO) in this setting, however, is not widely reported. METHODS: Cases of patients undergoing MT for AIS with ELVO at a single institution were reviewed. Cases preceded by recent CI were investigated retrospectively. Data was collected for patient demographics, type of cardiac intervention, stroke characteristics, neurovascular intervention, and patient outcomes. RESULTS: Between 2008 and 2017, registry analysis identified nine patients treated with MT for AIS complicating recent CI. Patients were more commonly male with a mean age of 67 years. A large majority had a known cardiac arrhythmia. Coronary artery bypass graft surgery (CABG) was the most identified CI, followed by valve repair, and cardiac ablations. Mean presenting NIHSS was 18. Most presented with hemiplegia. Seven cases were found to have MCA occlusions. Stent-retrievers were used in 6 cases with excellent recanalization in five MCA cases (TICI 2c or 3) and in two basilar cases. Despite immediate improvements in NIHSS scores in most cases, functional outcomes were poor in 7 cases (mRS of 4-6). Three cases were complicated by hemorrhage and three cases ended in mortality. CONCLUSION: AIS with ELVO following recent CI is associated with high rates of mortality and poor functional outcomes despite MT. Further work is needed to understand the key drivers to poor outcomes in this ELVO subgroup.

Forecasting ground magnetic field perturbations has been a long-standing goal of the space weather community. The availability of ground magnetic field data and its potential to be used in geomagnetically induced current studies, such as risk assessment, have resulted in several forecasting efforts over the past few decades. One particular community effort was the Geospace Environment Modeling (GEM) challenge of ground magnetic field perturbations that evaluated the predictive capacity of several empirical and first principles models at both mid- and high-latitudes in order to choose an operative model. In this work, we use three different deep learning models-a feed-forward neural network, a long short-term memory recurrent network and a convolutional neural network-to forecast the horizontal component of the ground magnetic field rate of change (dBH/dt) over 6 different ground magnetometer stations and to compare as directly as possible with the original GEM challenge. We find that, in general, the models are able to perform at similar levels to those obtained in the original challenge, although the performance depends heavily on the particular storm being evaluated. We then discuss the limitations of such a comparison on the basis that the original challenge was not designed with machine learning algorithms in mind.

China, the world’s largest greenhouse gas emitter in 2022, aims to achieve carbon neutrality by 2060. The power sector will play a major role in this decarbonization process due to its current reliance on coal. Prior studies have quantified air quality co-benefits from decarbonization or investigated pathways to eliminate greenhouse gas emissions from the power sector. However, few have jointly assessed the potential impacts of accelerating decarbonization on electric power systems and public health. Additionally, most analyses have treated air quality improvements as co-benefits of decarbonization, rather than a target during decarbonization. Here, we explore future energy technology pathways in China under accelerated decarbonization scenarios with a power system planning model that integrates carbon, pollutant, and health impacts. We integrate the health effects of power plant emissions into the power system decision-making process, quantifying the public health impacts of decarbonization under each scenario. We find that compared with a reference decarbonization pathway, a stricter cap (20% lower emissions than the reference pathway in each period) on carbon emissions would yield significant co-benefits to public health, leading to a 22% reduction in power sector health impacts. Although extra capital investment is required to achieve this low emission target, the value of climate and health benefits would exceed the additional costs, leading to $824 billion net benefits from 2021 to 2050. Another accelerated decarbonization pathway that achieves zero emissions five years earlier than the reference case would result in lower net benefits due to higher capital costs during earlier decarbonization periods. Treating air pollution impacts as a target in decarbonization can further mitigate both CO _2 emissions and negative health effects. Alternative low-cost solutions also show that small variations in system costs can result in significantly different future energy portfolios, suggesting that diverse decarbonization pathways are viable.

Existing studies on the economic feasibility of energy storage are system-specific without considering the decarbonisation of electricity production or impacts of GHG taxes. Here the authors applied an optimization model to investigate the economic viability of nice selected energy storage technologies in California and found that renewable curtailment and GHG reductions highly depend on capital costs of energy storage.

As a leading cause of bacterial-derived gastroenteritis worldwide, Campylobacter jejuni has a significant impact on human health in both the developed and developing worlds. Despite its prevalence as a human pathogen, the source of these infections remains poorly understood due to the mutation frequency of the organism and past limitations of whole genome analysis. Recent advances in both whole genome sequencing and computational methods have allowed for the high-resolution analysis of intraspecies diversity, leading multiple groups to postulate that these approaches may be used to identify the sources of Campylobacter jejuni infection. To address this hypothesis, our group conducted a regionally and temporally restricted sampling of agricultural and environmental Campylobacter sources and compared isolated C. jejuni genomes to those that caused human infections in the same region during the same time period. Through a network analysis comparing genomes from various sources, we found that human C. jejuni isolates clustered with those isolated from cattle and chickens, indicating these as potential sources of human infection in the region.

Calgranulin proteins are an important class of molecules involved in innate immunity. These members of the S100 class of the EF-hand family of calcium-binding proteins have numerous cellular and antimicrobial functions. One protein in particular, S100A12 (also called EN-RAGE or calgranulin C), is highly abundant in neutrophils during acute inflammation and has been implicated in immune regulation. Structure-function analyses reveal that S100A12 has the capacity to bind calcium, zinc, and copper, processes that contribute to nutritional immunity against invading microbial pathogens. S100A12 is a ligand for the receptor for advanced glycation end products (RAGE), toll-like receptor 4 (TLR4), and CD36, which promote cellular and immunological pathways to alter inflammation. We conducted a scoping review of the existing literature to define what is known about the association of S100A12 with digestive disease and health. Results suggest that S100A12 is implicated in gastroenteritis, necrotizing enterocolitis, gastritis, gastric cancer, Crohn’s disease, irritable bowel syndrome, inflammatory bowel disease, and digestive tract cancers. Together, these results reveal S100A12 is an important molecule broadly associated with the pathogenesis of digestive diseases.

Category: Arthroscopy Introduction/Purpose: While there has been an increasing emphasis on assessing resident competency, little has been published on how to best evaluate competency for ankle arthroscopy. The purpose of this study was to design and validate an objective model for assessing basic ankle arthroscopy knowledge and surgical skills on a cadaveric ankle. Methods: The Basic Ankle Arthroscopy Skills Scoring System was adapted from previously validated assessment tools for knee arthroscopy. The scoring system includes (1) an oral questionnaire (0-23 points), (2) a surgical task-specific checklist (0-19 points), and (3) a global surgical skills rating (12-60 points). Ten medical students and twenty-three residents participated. All participants performed a diagnostic ankle arthroscopy on a cadaveric ankle and were assessed by a single observer. Six participants were tested by two evaluators to determine inter-observer reliability. ABSTRACT The stator units of the flagellum supply power to the flagellar motor via ion transport across the cytoplasmic membrane and generate torque on the rotor for rotation. Flagellar motors across bacterial species have evolved adaptations that impact and enhance stator function to meet the demands of each species, including producing stator units using different fuel types or various stator units for different motility modalities. Campylobacter jejuni produces one of the most complex and powerful flagellar motors by positioning 17 stator units at a greater radial distance than in most other bacteria to increase power and torque for high velocity of motility. We report another evolutionary adaptation impacting flagellar stators by identifying FlgX as a chaperone for C. jejuni stator units to ensure sufficient power and torque for flagellar rotation and motility. We discovered that FlgX maintains MotA and MotB stator protein integrity likely through a direct interaction with MotA that prevents their degradation. Suppressor analysis suggested that the physiology of C. jejuni drives the requirement for FlgX to protect stator units from proteolysis by the FtsH protease complex. C. jejuni ΔflgX was strongly attenuated for colonization of the natural avian host, but colonization capacity was greatly restored by a single mutation in MotA. These findings suggest that the likely sole function of FlgX is to preserve stator unit integrity for the motility required for host interactions. Our findings demonstrate another evolved adaptation in motile bacteria to ensure the equipment of the flagellar motor with sufficient power to generate torque for motility. IMPORTANCE The bacterial flagellum is a reversible rotating motor powered by ion transport through stator units, which also exert torque on the rotor component to turn the flagellum for motility. Species-specific adaptations to flagellar motors impact stator function to meet the demands of each species to sufficiently power flagellar rotation. We identified another evolutionary adaptation by discovering that FlgX of Campylobacter jejuni preserves the integrity of stator units by functioning as a chaperone to protect stator proteins from degradation by the FtsH protease complex due to the physiology of the bacterium. FlgX is required to maintain a level of stator units sufficient to power the naturally high-torque flagellar motor of C. jejuni for motility in intestinal mucosal layers to colonize hosts. Our work continues to identify an increasing number of adaptations to flagellar motors across bacterial species that provide the mechanics necessary for producing an effective rotating nanomachine for motility.
