1. Translation velocity determines the efficacy of engineered suppressor tRNAs on pathogenic nonsense mutations

Abstract

Abstract Nonsense mutations – the underlying cause of approximately 11% of all genetic diseases – prematurely terminate protein synthesis by mutating a sense codon to a premature stop or termination codon (PTC). An emerging therapeutic strategy to suppress nonsense defects is to engineer sense-codon decoding tRNAs to readthrough and restore translation at PTCs. However, the readthrough efficiency of the engineered suppressor tRNAs (sup-tRNAs) largely varies in a tissue- and sequence context-dependent manner and has not yet yielded optimal clinical efficacy for many nonsense mutations. Here, we systematically analyze the suppression efficacy at various pathogenic nonsense mutations. We discover that the translation velocity of the sequence upstream of PTCs modulates the sup-tRNA readthrough efficacy. The PTCs most refractory to suppression are embedded in a sequence context translated with an abrupt reversal of the translation speed leading to ribosomal collisions. Moreover, modeling translation velocity using Ribo-seq data can accurately predict the suppression efficacy at PTCs. These results reveal previously unknown molecular signatures contributing to genotype-phenotype relationships and treatment-response heterogeneity, and provide the framework for the development of personalized tRNA-based gene therapies.

Abstract

Understanding the mechanisms that underlie de novo mutations (DNMs) can be essential for interpreting human evolution, including aspects such as rapidly diverging genes, conservation of non-coding regulatory elements, and somatic DNA adaptation, among others. DNM accumulation in Homo sapiens is often limited to evaluation of human trios or quads across a single generation. Moreover, human SNPs in exons, pseudogenes, or other non-coding elements can be ancient and difficult to date, including polymorphisms attributable to founder effects and identity by descent. In this report, we describe multigenerational evolution of a human coding locus devoid of natural selection, and delineate patterns and principles by which DNMs have accumulated over the past few thousand years. We apply a data set comprising cystic fibrosis transmembrane conductance regulator (CFTR) alleles from 2,393 individuals homozygous for the F508del defect. Additional polymorphism on the F508del background diversified subsequent to a single mutational event during recent human history. Because F508del CFTR is without function, SNPs observed on this haplotype are effectively attributable to factors that govern accumulating de novo mutations. We show profound enhancement of transition, synonymous, and positionally repetitive polymorphisms, indicating appearance of DNMs in a manner evolutionarily designed to protect protein coding DNA against mutational attrition while promoting diversity.

Abstract

Abstract Background Purine nucleoside phosphorylase (PNP) gene transfer represents a promising approach to treatment of head and neck malignancies. We tested recombinant adenovirus already in phase I/II clinical testing and leading‐edge patient‐derived xenografts (PDX) as a means to optimize this therapeutic strategy. Methods Our experiments investigated purine base cytotoxicity, PNP enzyme activity following treatment of malignant tissue, tumor mass regression, viral receptor studies, and transduction by tropism‐modified adenovirus. Results Replication deficient vector efficiently transduced PDX cells and mediated significant anticancer effect following treatment with fludarabine phosphate in vivo. Either 6‐methylpurine or 2‐fluoroadenine (toxic molecules generated by the PNP approach) ablated head and neck cancer cell proliferation. High levels of adenovirus‐3 specific receptors were detected in human tumor models, and vector was evaluated that utilizes this pathway. Conclusions Our studies provide the scientific foundation necessary to improve PNP prodrug cleavage and advance a new treatment for head and neck cancer.

Abstract

We report an estimation of the injected mass composition of ultra-high energy cosmic rays (UHECRs) at energies higher than 10 EeV. The composition is inferred from an energy-dependent sky distribution of UHECR events observed by the Telescope Array surface detector by comparing it to the Large Scale Structure of the local Universe. In the case of negligible extra-galactic magnetic fields the results are consistent with a relatively heavy injected composition at E ~ 10 EeV that becomes lighter up to E ~ 100 EeV, while the composition at E > 100 EeV is very heavy. The latter is true even in the presence of highest experimentally allowed extra-galactic magnetic fields, while the composition at lower energies can be light if a strong EGMF is present. The effect of the uncertainty in the galactic magnetic field on these results is subdominant., Comment: 8 pages, 3 figures, accepted for publication in PRL

Abstract

We use a new method to estimate the injected mass composition of ultrahigh cosmic rays (UHECRs) at energies higher than 10 EeV. The method is based on comparison of the energy-dependent distribution of cosmic ray arrival directions as measured by the Telescope Array experiment (TA) with that calculated in a given putative model of UHECR under the assumption that sources trace the large-scale structure (LSS) of the Universe. As we report in the companion letter, the TA data show large deflections with respect to the LSS which can be explained, assuming small extra-galactic magnetic fields (EGMF), by an intermediate composition changing to a heavy one (iron) in the highest energy bin. Here we show that these results are robust to uncertainties in UHECR injection spectra, the energy scale of the experiment and galactic magnetic fields (GMF). The assumption of weak EGMF, however, strongly affects this interpretation at all but the highest energies E > 100 EeV, where the remarkable isotropy of the data implies a heavy injected composition even in the case of strong EGMF. This result also holds if UHECR sources are as rare as $2 \times 10^{-5}$ Mpc$^{-3}$, that is the conservative lower limit for the source number density., Comment: 18 pages, 11 figures, accepted for publication in PRD

Abstract

We report on an observation of the difference between northern and southern skies of the ultrahigh energy cosmic ray energy spectrum with a significance of ${\sim}8\sigma$. We use measurements from the two largest experiments$\unicode{x2014}$the Telescope Array observing the northern hemisphere and the Pierre Auger Observatory viewing the southern hemisphere. Since the comparison of two measurements from different observatories introduces the issue of possible systematic differences between detectors and analyses, we validate the methodology of the comparison by examining the region of the sky where the apertures of the two observatories overlap. Although the spectra differ in this region, we find that there is only a $1.8\sigma$ difference between the spectrum measurements when anisotropic regions are removed and a fiducial cut in the aperture is applied., Comment: 8 pages, 6 figures

Thus, we aimed to identify the potential risk factors of non-adherence and evaluate whether utilizing a usual source of care was associated with medication adherence in immigrants.Methods: We utilized the Korea National Health Insurance Claims Database between 2012 and 2015. Cases were immigrants who had antihypertensive prescriptions at the time of hypertension diagnosis in 2012. Controls were native-born Koreans with hypertension who were 1:1 matched to immigrants by age, sex, and Charlson comorbidity index. We used the medication possession ratio for three years to assess the adherence to antihypertensive drugs. The likelihood of non-adherence was evaluated between cases and controls by multivariate linear regression models stratified by age, sex, and number of clinic visits. We assessed the potential risk factors of non-adherence in immigrants by multivariate linear regression and logistic regression models, respectively.Results: In total, 4114 immigrants and 4114 matched native-born Koreans with hypertension were included. The mean MPR was significantly lower in immigrants (56% vs 70%, p< 0.0001). Immigrants showed almost two times the level of non-adherence as native-born Koreans (odds ratio [OR], 2.01; 95% confidence interval [CI], 1.83– 2.21). Stratified analyses on non-adherence presented the highest non-adherence (2.28 times) in immigrants in the younger group (30– 49 years old) and the lowest non-adherence in immigrants in 65 and old group where the risk was 1.69 times higher than native Korean with the same age. The absence of a usual source of care significantly increased medication non-adherence by 1.31 to 1.58 times among immigrants.Conclusion: When the number of visited clinics increased, the degree of non-adherence increased consistently. Therefore, the systematization of registering with primary care (a usual source of care) might be a modifiable health care strategy to improve health care outcomes in immigrants.Keywords: usual source of care, hypertension, immigrants, health care strategy

8. A colon-specific prodrug of metoclopramide ameliorates colitis in an experimental rat model

Methods: MCP azo-linked to 5-ASA (5-[4-chloro-2-{2-(diethylamino)ethylcarbamoyl}–1-methoxyphenyl]azosalicylic acid, MCP-azo-ASA) was synthesized, where 5-ASA was used as a colon-targeting carrier and an anti-colitic agent, and the ability of MCP-azo-ASA to target the colon in vitro and in vivo was evaluated. Results: Our results indicate that MCP-azo-ASA was cleaved to MCP and 5-ASA in the cecal contents, but not in the contents of the small intestine. Oral gavage with equimolar concentrations of MCP-azo-ASA and sulfasalazine (SSZ, a colon-specific prodrug of 5-ASA widely used clinically) demonstrated that the two prodrugs delivered comparable amounts of 5-ASA to the cecum. MCP was barely detected in the blood after oral gavage with MCP-azo-ASA. In a rat model of 2,4-dinitrobenzene sulfonic acid hydrate (DNBS)-induced colitis, MCP-azo-ASA alleviated colonic damage in a dose-dependent manner. Moreover, MCP-azo-ASA reduced the concentrations of inflammatory mediators in the inflamed colon. At low equimolar doses, MCP-azo-ASA, but not SSZ, resulted in significant anti-colitic effects, which indicates that MCP has anti-colitic activity. MCP-azo-ASA had anti-colitic effects equal to those of SSZ at high equimolar doses. Conclusion: Thus, our results indicate that MCP-azo-ASA is a colon-specific prodrug of MCP. Targeted delivery of MCP to the colon ameliorated DNBS-induced colitis in rats, and we did not observe any synergistic effects of MCP after co-delivery with 5-ASA. However, studies using small molecule NO donors (N-diazeniumdiolate, S-nitrosothiol, etc) have yet to meet clinical requirements due to the short half-life and initial burst-release profile of NO donors. In this study, we report the feasibility of methoxy poly(ethylene glycol)-b-poly(lactic-co-glycolic acid) (mPEG-PLGA) nanoparticles (NPs) as NO-releasing polymers (NO-NPs) for inducing angiogenesis.Materials and methods: The mPEG–PLGA copolymers were synthesized by typical ring-opening polymerization of lactide, glycolide and mPEG as macroinitiators. Double emulsion methods were used to prepare mPEG–PLGA NPs incorporating hydrophilic NONOate (diethylenetriamine NONOate).Results: This liposomal NP encapsulates hydrophilic diethylenetriamine NONOate (70%±4%) more effectively than other previously reported materials. The application of NO-NPs at different ratios resulted in varying NO-release profiles with no significant cytotoxicity in various cell types: normal cells (fibroblasts, human umbilical vein endothelial cells and epithelial cells) and cancer cells (C6, A549 and MCF-7). The angiogenic potential of NO-NPs was confirmed in vitro by tube formation and ex vivo through an aorta ring assay. Tubular formation increased 189.8% in NO-NP–treated groups compared with that in the control group. Rat aorta exhibited robust sprouting angiogenesis in response to NO-NPs, indicating that NO was produced by polymeric NPs in a sustained manner.Conclusion: These findings provide initial results for an angiogenesis-related drug development platform by a straightforward method with biocompatible polymers. The dramatic increase in the prevalence of diabetes is largely due to the incidence of type 2 diabetes in low- and middle-income countries (LMIc) besides high-income countries (HIc). We aimed to evaluate whether pharmacist care (PC) service model in LMIc and HIc could improve clinical outcomes in diabetic patients by performing a meta-analysis. Methods: PubMed, Embase, and ProQuest Dissertations Unlimited Published Literature database were searched to find publications pertaining to pharmacist-led intervention in patients with diabetes. The inclusion criteria were as follows: 1) randomized controlled trials, 2) confirmed diabetic patients (type 1 or type 2), 3) pharmaceutical care intervention by clinical pharmacist or/and multidisciplinary team, and 4) reporting HbA1c at baseline and end of study or the mean change in these values. Results: A total of 37 articles were included in the meta-analysis. Abstract

The Extreme Universe Space Observatory on a Super Pressure Balloon 1 (EUSO-SPB1) was launched in 2017 April from Wanaka, New Zealand. The plan of this mission of opportunity on a NASA super pressure balloon test flight was to circle the southern hemisphere. The primary scientific goal was to make the first observations of ultra-high-energy cosmic-ray extensive air showers (EASs) by looking down on the atmosphere with an ultraviolet (UV) fluorescence telescope from suborbital altitude (33~km). After 12~days and 4~hours aloft, the flight was terminated prematurely in the Pacific Ocean. Before the flight, the instrument was tested extensively in the West Desert of Utah, USA, with UV point sources and lasers. The test results indicated that the instrument had sensitivity to EASs of approximately 3 EeV. Simulations of the telescope system, telescope on time, and realized flight trajectory predicted an observation of about 1 event assuming clear sky conditions. The effects of high clouds were estimated to reduce this value by approximately a factor of 2. A manual search and a machine-learning-based search did not find any EAS signals in these data. Autophagy is an intracellular degradation system that has been associated with the progression of neurodegenerative disorders. Although induction of autophagic flux by GO has been reported, the underlying signaling pathway in neurodegenerative disorders and how this is involved in neuroprotection remain obscure. We show that GO itself activates autophagic flux in neuronal cells and confers a neuroprotective effect against prion protein (PrP) (106–126)-mediated neurotoxicity. GO can be detected in SK-N-SH neuronal cells, where it triggers autophagic flux signaling. GO-induced autophagic flux prevented PrP (106–126)-induced neurotoxicity in SK-N-SH cells. Moreover, inactivation of autophagic flux blocked GO-induced neuroprotection against prion-mediated mitochondrial neurotoxicity. This is the first study to demonstrate that GO regulates autophagic flux in neuronal cells, and that activation of autophagic flux signals, induced by GO, plays a neuroprotective role against prion-mediated mitochondrial neurotoxicity. These results suggest that the nanomaterial GO may be used to activate autophagic flux and could be used in neuroprotective strategies for treatment of neurodegenerative disorders, including prion diseases. Thus, in an attempt to control the colonic distribution of the drug, a polymeric colon-specific prodrug, MTDZ conjugated to dextran via a succinate linker (Dex-SA-MTDZ), was designed. Upon treatment with dextranase for 8 h, the degree of Dex-SA-MTDZ depolymerization (%) with a degree of substitution (mg of MTDZ bound in 100 mg of Dex-SA-MTDZ) of 7, 17, and 30 was 72, 38, and 8, respectively, while that of dextran was 85. Depolymerization of Dex-SA-MTDZ was found to be necessary for the release of MTDZ, because dextranase pretreatment ensures that de-esterification occurs between MTDZ and the dextran backbone. In parallel, Dex-SA-MTDZ with a degree of substitution of 17 was found not to release MTDZ upon incubation with the contents of the small intestine and stomach of rats, but it released MTDZ when incubated with rat cecal contents (including microbial dextranases). Moreover, Dex-SA-MTDZ exhibited prolonged release of MTDZ, which contrasts with drug release by small molecular colon-specific prodrugs, MTDZ sulfate and N-nicotinoyl-2-{2-(2-methyl-5-nitroimidazol-1-yl)ethyloxy}-d,l-glycine. These prodrugs were eliminated very rapidly, and no MTDZ was detected in the cecal contents. Consistent with these in vitro results, we found that oral gavage of Dex-SA-MTDZ delivered MTDZ (as MTDZ conjugated to [depolymerized] dextran) to the distal colon. However, upon oral gavage of the small molecular prodrugs, no prodrugs were detected in the distal colon. Collectively, these data suggest that dextran conjugation is a potential pharmaceutical strategy to control the colonic distribution of drugs susceptible to colonic microbial metabolism. Abstract

Dark GRBs constitute a significant fraction of the GRB population. In this paper, we present the multiwavelength analysis of an intense two-episodic GRB 150309A observed early on to ~114 days post-burst. Despite the strong gamma-ray emission, no optical afterglow was detected for this burst. However, we discovered near-infrared afterglow ($K_{\rm S}$-band), ~5.2 hours post burst, with the CIRCE instrument mounted at the 10.4m GTC. We used Fermi observations of GRB 150309A to understand the prompt emission mechanisms and jet composition. We performed the early optical observations using the BOOTES robotic telescope and late-time afterglow observations using the GTC. A potential faint host galaxy is also detected at optical wavelength using the GTC. We modelled the potential host galaxy of GRB 150309A in order to explore the environment of the burst. The time-resolved spectral analysis of Fermi data indicates a hybrid jet composition consisting of a matter-dominated fireball and magnetic-dominated Poynting flux. GTC observations of the afterglow revealed that the counterpart of GRB 150309A was very red, with H-$K_{\rm S}$ > 2.1 mag (95 $\%$ confidence). The red counterpart was not discovered in any bluer filters of Swift UVOT, indicative of high redshift origin. This possibility was discarded based on multiple arguments, such as spectral analysis of X-ray afterglow constrain z < 4.15 and a moderate redshift value obtained using spectral energy distribution modelling of the potential galaxy. The broadband afterglow SED implies a very dusty host galaxy with deeply embedded GRB (suggesting $A_{\rm V}$ $\gtrsim$ 35 mag). The environment of GRB 150309A demands a high extinction towards the line of sight, demanding dust obscuration is the most probable origin of optical darkness and the very red afterglow of GRB 150309A. Cinacalcet effectively controls SHPT without causing hypercalcemia and hyperphosphatemia. However, there is significant inter-individual response variance to cinacalcet treatment. Therefore, we aimed to evaluate the genetic effects related with parathyroid hormone regulation as factors for cinacalcet response variance. Methods: Patients with a diagnosis of SHPT based on intact parathyroid hormone (iPTH) >300 pg/mL on dialysis were included in this study. They were over 18 years and have been treated by cinacalcet for more than 3 months. Responders and nonresponders were grouped by the serum iPTH changes. Twenty-four single nucleotide polymorphisms of CASR, VDR, FGFR1, KL, ALPL, RGS14, NR4A2, and PTHLH genes were selected for the pharmacogenetic analysis. Results: After adjusting for age, sex, and calcium level, CASR rs1042636 (odds ratio [OR]: 0.066, P=0.027) and rs1802757 (OR: 10.532, P=0.042) were associated with cinacalcet response. The association of haplotypes of CASR rs1042636, rs10190, and rs1802757; GCC (OR: 0.355, P=0.015); and ATT (OR: 2.769, P=0.014) with cinacalcet response was also significant. Conclusion: We obtained supporting information of the associations between cinacalcet response and CASR polymorphisms. CASR single nucleotide polymorphisms (SNPs) rs1802757, rs1042636, and haplotypes of rs1042636, rs10190, and rs1802757 were significantly associated with cinacalcet response variance. Abstract

We consider a reconfigurable intelligent surface (RIS) assisted multiple-input multiple-output (MIMO) system in the presence of scattering objects. The MIMO transmitter and receiver, the RIS, and the scattering objects are modeled as mutually coupled thin wires connected to load impedances. We introduce a novel numerical algorithm for optimizing the tunable loads connected to the RIS, which does not utilize the Neumann series approximation. The algorithm is provably convergent, has polynomial complexity with the number of RIS elements, and outperforms the most relevant benchmark algorithms while requiring fewer iterations and converging in a shorter time.

We investigated whether colon-targeted delivery of PCT could enhance anti-colitic effects and how therapeutic enhancement occurred at the molecular level. Molecular effects of PCT were examined in human colon carcinoma cells and inflamed colons. The anti-colitic effects of PCT in a colon-targeted capsule (colon-targeted PCT) were compared with PCT in a gelatin capsule (conventional PCT) in a trinitrobenzene sulfonic acid-induced rat colitis model. Colon-targeted PCT elicited greatly enhanced recovery of the colonic inflammation. In HCT116 cells, PCT inhibited nuclear factor kappaB while activating anti-colitic transcription factors, nuclear factor-erythroid 2 (NF-E2) p45-related factor 2, and hypoxia-inducible factor-1. Colon-targeted PCT, but not conventional PCT, modulated production of the target gene products of the transcription factors in the inflamed colonic tissues. Rectal administration of PCT, which simulates the therapeutic action of colon-targeted PCT, also ameliorated rat colitis and reproduced the molecular effects in the inflamed colonic tissues. Colon-targeted delivery increased therapeutic efficacy of PCT against colitis, likely resulting from multitargeted effects exerted by colon-targeted PCT. The drug delivery technique may be useful for therapeutic optimization of anti-colitic lead compounds including natural products. Glycylcelecoxib (GC), N-glycylaspart-1-ylcelecoxib (N-GA1C), and C-glycylaspart-1-ylcelecoxib (C-GA1C) were synthesized and their structures identified using infrared and proton nuclear magnetic resonance spectrometer. The celecoxib derivatives were chemically stable in pH 6.8 and 1.2 buffers. GC and C-GA1C were resistant to degradation in the small intestinal contents, while N-GA1C was substantially cleaved to release celecoxib. In contrast, all the celecoxib derivatives were degraded to liberate celecoxib in the cecal content. These results suggest that GC and C-GA1C could be delivered to and liberate celecoxib and glycine in the large intestine. In human colon carcinoma HCT116 and murine macrophage RAW264.7 cells, combined celecoxib–glycine chloramine treatment additively suppressed the production of proinflammatory NFκB target gene products. However, the cardiovascular toxicity of celecoxib limits its routine use in the clinic. Generally, colon-specific delivery of a drug both increases the therapeutic availability in the large intestine and decreases the systemic absorption of the drug, most likely resulting in enhanced therapeutic effects against colonic diseases such as colitis and reduced systemic side effects. To develop a colon-specific prodrug of celecoxib that could reduce its cardiovascular toxicity and improve its therapeutic activity, dextran–glutamic acid–celecoxib conjugate (glutam-1-yl celecoxib-dextran ester [G1CD]) was prepared and evaluated. While stable in pH 1.2 and 6.8 buffer solutions and small-intestinal contents, G1CD efficiently released celecoxib in cecal contents. Oral administration of G1CD to rats delivered a larger amount of celecoxib to the large intestine than free celecoxib. G1CD prevented the systemic absorption of celecoxib and did not decrease the serum level of 6-ketoprostaglandin F1α, an inverse indicator of cardiovascular toxicity of celecoxib. Collectively, G1CD may be a polymeric colon-specific celecoxib prodrug with therapeutic and toxicological advantages. Abstract

Small molecules that correct the folding defects and enhance surface localization of the F508del mutation in the Cystic Fibrosis Transmembrane conductance Regulator (CFTR) comprise an important therapeutic strategy for cystic fibrosis lung disease. However, compounds that rescue the F508del mutant protein to wild type (WT) levels have not been identified. In this report, we consider obstacles to obtaining robust and therapeutically relevant levels of F508del CFTR. For example, markedly diminished steady state amounts of F508del CFTR compared to WT CFTR are present in recombinant bronchial epithelial cell lines, even when much higher levels of mutant transcript are present. In human primary airway cells, the paucity of Band B F508del is even more pronounced, although F508del and WT mRNA concentrations are comparable. Therefore, to augment levels of "repairable" F508del CFTR and identify small molecules that then correct this pool, we developed compound library screening protocols based on automated protein detection. First, cell-based imaging measurements were used to semi-quantitatively estimate distribution of F508del CFTR by high content analysis of two-dimensional images. We evaluated ~2,000 known bioactive compounds from the NIH Roadmap Molecular Libraries Small Molecule Repository in a pilot screen and identified agents that increase the F508del protein pool. Second, we analyzed ~10,000 compounds representing diverse chemical scaffolds for effects on total CFTR expression using a multi-plate fluorescence protocol and describe compounds that promote F508del maturation. Together, our findings demonstrate proof of principle that agents identified in this fashion can augment the level of endoplasmic reticulum (ER) resident "Band B" F508del CFTR suitable for pharmacologic correction. As further evidence in support of this strategy, PYR-41-a compound that inhibits the E1 ubiquitin activating enzyme-was shown to synergistically enhance F508del rescue by C18, a small molecule corrector. Our combined results indicate that increasing the levels of ER-localized CFTR available for repair provides a novel route to correct F508del CFTR.

24. Robust Stimulation of W1282X-CFTR Channel Activity by a Combination of Allosteric Modulators.

Abstract

W1282X is a common nonsense mutation among cystic fibrosis patients that results in the production of a truncated Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) channel. Here we show that the channel activity of the W1282X-CFTR polypeptide is exceptionally low in excised membrane patches at normally saturating doses of ATP and PKA (single channel open probability (PO) < 0.01). However, W1282X-CFTR channels were stimulated by two CFTR modulators, the FDA-approved VX-770 and the dietary compound curcumin. Each of these compounds is an allosteric modulator of CFTR gating that promotes channel activity in the absence of the native ligand, ATP. Although W1282X-CFTR channels were stimulated by VX-770 in the absence of ATP their activities remained dependent on PKA phosphorylation. Thus, activated W1282X-CFTR channels should remain under physiologic control by cyclic nucleotide signaling pathways in vivo. VX-770 and curcumin exerted additive effects on W1282X-CFTR channel gating (opening/closing) in excised patches such that the Po of the truncated channel approached unity (> 0.9) when treated with both modulators. VX-770 and curcumin also additively stimulated W1282X-CFTR mediated currents in polarized FRT epithelial monolayers. In this setting, however, the stimulated W1282X-CFTR currents were smaller than those mediated by wild type CFTR (3-5%) due presumably to lower expression levels or cell surface targeting of the truncated protein. Combining allosteric modulators of different mechanistic classes is worth considering as a treatment option for W1282X CF patients perhaps when coupled with maneuvers to increase expression of the truncated protein.

Abstract

In this paper, we present the first high-speed video observation of a cloud-to-ground lightning flash and its associated downward-directed Terrestrial Gamma-ray Flash (TGF). The optical emission of the event was observed by a high-speed video camera running at 40,000 frames per second in conjunction with the Telescope Array Surface Detector, Lightning Mapping Array, interferometer, electric-field fast antenna, and the National Lightning Detection Network. The cloud-to-ground flash associated with the observed TGF was formed by a fast downward leader followed by a very intense return stroke peak current of -154 kA. The TGF occurred while the downward leader was below cloud base, and even when it was halfway in its propagation to ground. The suite of gamma-ray and lightning instruments, timing resolution, and source proximity offer us detailed information and therefore a unique look at the TGF phenomena.

Abstract

For several decades, the origin of ultra-high-energy cosmic rays (UHECRs) has been an unsolved question of high-energy astrophysics. One approach for solving this puzzle is to correlate UHECRs with high-energy neutrinos, since neutrinos are a direct probe of hadronic interactions of cosmic rays and are not deflected by magnetic fields. In this paper, we present three different approaches for correlating the arrival directions of neutrinos with the arrival directions of UHECRs. The neutrino data is provided by the IceCube Neutrino Observatory and ANTARES, while the UHECR data with energies above $\sim$50 EeV is provided by the Pierre Auger Observatory and the Telescope Array. All experiments provide increased statistics and improved reconstructions with respect to our previous results reported in 2015. The first analysis uses a high-statistics neutrino sample optimized for point-source searches to search for excesses of neutrinos clustering in the vicinity of UHECR directions. The second analysis searches for an excess of UHECRs in the direction of the highest-energy neutrinos. The third analysis searches for an excess of pairs of UHECRs and highest-energy neutrinos on different angular scales. None of the analyses has found a significant excess, and previously reported over-fluctuations are reduced in significance. Abstract

Within the frame of the commissioning of a new experimental apparatus EXPADES we undertook the measurement of the elastic scattering angular distribution for the system 17O+208Pb at energy around the Coulomb barrier. The reaction dynamics induced by loosely bound Radioactive Ion Beams is currently being extensively studied [4]. In particular the study of the elastic scattering process allows to obtain direct information on the total reaction cross section of the exotic nuclei. In order to understand the effect of the low binding energy on the reaction mechanism it is important to compare radioactive weakly bound nuclei with stable strongly-bound nuclei. In this framework the study of the 17O+208Pb elastic scattering can be considered to be complementary to a previous measurement of the total reaction cross section for the system 17F+208Pb at energies of 86, 90.4 MeV [5, 6]. The data will be compared with those obtained for the neighboring systems 16,18O+208Pb and others available in literature.

Abstract

There is a paucity of data regarding molecular subtypes of pure ductal carcinoma in situ (pDCIS). We evaluated the expression of ER, PR, HER2, Ki67, and p53 and DNA ploidy in 118 pDCIS and 100 invasive breast carcinomas (IBCAs) by routine IHC and classified them according to molecular subtypes. Quantification of biomarkers and DNA ploidy was performed by image analysis. Expression of ER, PR, and high ki67 was more frequent in pDCIS compared to IBCA. High-grade tumors had lower ER and PR expression, high Ki67, overexpression of HER2 and p53, and DNA aneuploidy. Luminal A and HER2 subtypes were more common in pDCIS, and triple negative was more prevalent in IBCA. In both groups, HER2 and triple negative subtypes were characterized by high ki67, overexpression of p53, and DNA aneuploidy compared to luminal subtypes. Molecular subtypes of IBCA are distinct from those of pDCIS. Invasion is characterized by change in phenotype in some tumors.

Abstract

We present the first observation by the Telescope Array Surface Detector (TASD) of the effect of thunderstorms on the development of cosmic ray single count rate intensity over a 700 km$^{2}$ area. Observations of variations in the secondary low-energy cosmic ray counting rate, using the TASD, allow us to study the electric field inside thunderstorms, on a large scale, as it progresses on top of the 700 km$^{2}$ detector, without dealing with the limitation of narrow exposure in time and space using balloons and aircraft detectors. In this work, variations in the cosmic ray intensity (single count rate) using the TASD, were studied and found to be on average at the $\sim(0.5-1)\%$ and up to 2\% level. These observations were found to be both in excess and in deficit. They were also found to be correlated with lightning in addition to thunderstorms. These variations lasted for tens of minutes; their footprint on the ground ranged from 6 to 24 km in diameter and moved in the same direction as the thunderstorm. With the use of simple electric field models inside the cloud and between cloud to ground, the observed variations in the cosmic ray single count rate were recreated using CORSIKA simulations. Depending on the electric field model used and the direction of the electric field in that model, the electric field magnitude that reproduces the observed low-energy cosmic ray single count rate variations was found to be approximately between 0.2-0.4 GV. This in turn allows us to get a reasonable insight on the electric field and its effect on cosmic ray air showers inside thunderstorms.

Abstract

The Telescope Array Collaboration has observed an excess of events with $E \ge 10^{19.4} ~{\rm eV}$ in the data which is centered at (RA, dec) = ($19^\circ$, $35^\circ$). This is near the center of the Perseus-Pisces supercluster (PPSC). The PPSC is about $70 ~{\rm Mpc}$ distant and is the closest supercluster in the Northern Hemisphere (other than the Virgo supercluster of which we are a part). A Li-Ma oversampling analysis with $20^\circ$-radius circles indicates an excess in the arrival direction of events with a local significance of about 4 standard deviations. The probability of having such excess close to the PPSC by chance is estimated to be 3.5 standard deviations. This result indicates that a cosmic ray source likely exists in that supercluster., Comment: 8 pages, 4 figures, 1 table

Abstract

We present and perform a detailed analysis of multi-wavelength observations of \thisgrb, an optical bright GRB with an observed reverse shock (RS) signature. Observations of this GRB were acquired with the BOOTES-4 robotic telescope, the \fermi, and the \swift missions. Time-resolved spectroscopy of the prompt emission shows that changes to the peak energy (\Ep) tracks intensity and the low-energy spectral index seems to follow the intensity for the first episode, whereas this tracking behavior is less clear during the second episode. The fit to the afterglow light curves shows that the early optical afterglow can be described with RS emission and is consistent with the thin shell scenario of the constant ambient medium. The late time afterglow decay is also consistent with the prediction of the external forward shock (FS) model. We determine the properties of the shocks, Lorentz factor, magnetization parameters, and ambient density of \thisgrb, and compare these parameters with another 12 GRBs, consistent with having RS produced by thin shells in an ISM-like medium. The value of the magnetization parameter ($R_{\rm B} \approx 18$) indicates a moderately magnetized baryonic dominant jet composition for \thisgrb. We also report the host galaxy photometric observations of \thisgrb obtained with 10.4m GTC, 3.5m CAHA, and 3.6m DOT telescopes and find the host (photo $z$ = $2.8^{+0.7}_{-0.9}$) to be a high mass, star-forming galaxy with a star formation rate of $20 \pm 10 \msun$ $\rm yr^{-1}$., Comment: 27 pages, 16 figures, 12 tables, accepted for publication in MNRAS

Abstract

Telescope Array (TA) is the largest ultrahigh energy cosmic-ray (UHECR) observatory in the Northern Hemisphere. It explores the origin of UHECRs by measuring their energy spectrum, arrival-direction distribution, and mass composition using a surface detector (SD) array covering approximately 700 km$^2$ and fluorescence detector (FD) stations. TA has found evidence for a cluster of cosmic rays with energies greater than 57 EeV. In order to confirm this evidence with more data, it is necessary to increase the data collection rate.We have begun building an expansion of TA that we call TAx4. In this paper, we explain the motivation, design, technical features, and expected performance of the TAx4 SD. Abstract

The feasibility of the multi-nucleon transfer (MNT) reaction in the 136Xe +198Pt system was investigated for synthesizing neutron-rich heavy nuclei around mass number A∼200 of astrophysical interest. Identification of the projectile-like fragments (PLF’s) was successfully performed by using VAMOS++ spectrometer. The isotopic distributions of the PLF’s produced by two-proton stripping and pickup reactions were compared with each other, demonstrating the properties of the MNT reactions observed in the present system. The two-proton pickup channels strongly populated than the twoproton stripping channels were observed. The isotopic distributions for different total kinetic energy loss (TKEL) ranges are also discussed. The overall enhancement of the strength of the two-proton pickup compared with the two-proton stripping appears to be related to the MNT reactions with relatively moderate energy dissipation.

Abstract

Elastic scattering has been studied for the collisions induced by 17O on 58Ni target at energies around and above the Coulomb barrier. The elastic scattering angular distributions were measured for several energies and were analyzed within the framework of the optical model to obtain total reaction cross sections. The reaction cross-sections of the tightly bound 17O were compared with those of weakly bound 17F on the same targets in order to investigate the effects of the low binding energy in the reaction dynamics.

Abstract

We describe the evolution of of Swift J1644+57, whose unique X-ray properties have led several groups to interpret its behavior as corresponding to an extraordinary event of tidal disruption of a star by a supermassive black hole in the nucleus of a (z = 0.3545) galaxy, as derived by GTC. Multiwavelength observations (X-rays, optical, millimetre, centimitre) are proving to be essential to reveal the long term nature of the emission in this source. In particular, we identify for the first time the properties of a forming relativistic jet. In our interpretation of the phenomenon, we leave the still open possibility that it may correspond to the onset of a dormant AGN, but this may only be tested with longer term X-ray, millimetre and centimetre monitoring.

Abstract

The BL Lacertae object OJ 287 (z = 0.306) has unique double-peaked optical outbursts every ~12 years, and it presents one of the best cases for a small-separation binary supermassive black hole (SMBH) system, with an extremely massive primary log (M_BH/M_Sun) ~ 10.3. However, the host galaxy is unresolved or only marginally detected in all optical studies so far, indicating a large deviation from the bulge mass - SMBH mass relation. We have obtained deep, high spatial resolution i-band and K-band images of OJ~287 when the target was in a low state, which enable us to detect the host galaxy. We find the broad-band photometry of the host to be consistent with an early type galaxy with M_R = -22.5 and M_K = -25.2, placing it in the middle of the host galaxy luminosity distribution of BL Lacertae objects. The central supermassive black hole is clearly overmassive for a host galaxy of that luminosity, but not unprecedented, given some recent findings of other ``overmassive'' black holes in nearby galaxies., Comment: Accepted for publication in Astrophysical Journal

Abstract

In this paper we report the first close, high-resolution observations of downward-directed terrestrial gamma-ray flashes (TGFs) detected by the large-area Telescope Array cosmic ray observatory, obtained in conjunction with broadband VHF interferometer and fast electric field change measurements of the parent discharge. The results show that the TGFs occur during strong initial breakdown pulses (IBPs) in the first few milliseconds of negative cloud-to-ground and low-altitude intracloud flashes, and that the IBPs are produced by a newly-identified streamer-based discharge process called fast negative breakdown. The observations indicate the relativistic runaway electron avalanches (RREAs) responsible for producing the TGFs are initiated by embedded spark-like transient conducting events (TCEs) within the fast streamer system, and potentially also by individual fast streamers themselves. The TCEs are inferred to be the cause of impulsive sub-pulses that are characteristic features of classic IBP sferics. Additional development of the avalanches would be facilitated by the enhanced electric field ahead of the advancing front of the fast negative breakdown. In addition to showing the nature of IBPs and their enigmatic sub-pulses, the observations also provide a possible explanation for the unsolved question of how the streamer to leader transition occurs during the initial negative breakdown, namely as a result of strong currents flowing in the final stage of successive IBPs, extending backward through both the IBP itself and the negative streamer breakdown preceding the IBP., Comment: Typo fixed and reference added. Abstract

Motivated by the detection of a significant dipole structure in the arrival directions of ultrahigh-energy cosmic rays above 8 EeV reported by the Pierre Auger Observatory (Auger), we search for a large-scale anisotropy using data collected with the surface detector array of the Telescope Array Experiment (TA). With 11 years of TA data, a dipole structure in a projection of the right ascension is fitted with an amplitude of 3.3+- 1.9% and a phase of 131 +- 33 degrees. The corresponding 99% confidence-level upper limit on the amplitude is 7.3%. At the current level of statistics, the fitted result is compatible with both an isotropic distribution and the dipole structure reported by Auger., Comment: 6 pages, 3 figures, 1 table, Proofed title. Abstract

Ultra high energy cosmic rays provide the highest known energy source in the universe to measure proton cross sections. Though conditions for collecting such data are less controlled than an accelerator environment, current generation cosmic ray observatories have large enough exposures to collect significant statistics for a reliable measurement for energies above what can be attained in the lab. Cosmic ray measurements of cross section use atmospheric calorimetry to measure depth of air shower maximum ($X_{\mathrm{max}}$), which is related to the primary particle's energy and mass. The tail of the $X_{\mathrm{max}}$ distribution is assumed to be dominated by showers generated by protons, allowing measurement of the inelastic proton-air cross section. In this work the proton-air inelastic cross section measurement, $\sigma^{\mathrm{inel}}_{\mathrm{p-air}}$, using data observed by Telescope Array's Black Rock Mesa and Long Ridge fluorescence detectors and surface detector array in hybrid mode is presented. $\sigma^{\mathrm{inel}}_{\mathrm{p-air}}$ is observed to be $520.1 \pm 35.8$[Stat.] $^{+25.0}_{-40}$[Sys.]~mb at $\sqrt{s} = 73$ TeV. The total proton-proton cross section is subsequently inferred from Glauber formalism and is found to be $\sigma^{\mathrm{tot}}_{\mathrm{pp}} = 139.4 ^{+23.4}_{-21.3}$ [Stat.]$ ^{+15.0}_{-24.0}$[Sys.]~mb.

Abstract

Evidence for a large-scale supergalactic cosmic ray multiplet (arrival directions correlated with energy) structure is reported for ultra-high energy cosmic ray (UHECR) energies above 10$^{19}$ eV using seven years of data from the Telescope Array (TA) surface detector and updated to 10 years. Previous energy-position correlation studies have made assumptions regarding magnetic field shapes and strength, and UHECR composition. Here the assumption tested is that, since the supergalactic plane is a fit to the average matter density of the local Large Scale Structure (LSS), UHECR sources and intervening extragalactic magnetic fields are correlated with this plane. This supergalactic deflection hypothesis is tested by the entire field-of-view (FOV) behavior of the strength of intermediate-scale energy-angle correlations. These multiplets are measured in spherical cap section bins (wedges) of the FOV to account for coherent and random magnetic fields. The structure found is consistent with supergalactic deflection, the previously published energy spectrum anisotropy results of TA (the hotspot and coldspot), and toy-model simulations of a supergalactic magnetic sheet. The seven year data post-trial significance of this supergalactic structure of multiplets appearing by chance, on an isotropic sky, is found by Monte Carlo simulation to be 4.2$\sigma$. The ten years of data post-trial significance is 4.1$\sigma$. Furthermore, the starburst galaxy M82 is shown to be a possible source of the TA Hotspot, and an estimate of the supergalactic magnetic field using UHECR measurements is presented.

48. Magnetic skyrmion diode: Unidirectional skyrmion motion via symmetry breaking of potential energy barriers

Abstract

We realize a magnetic skyrmion diode operated by a unidirectional skyrmion transport that flows in only one direction, which is highly significant for information processing in spintronic and nanoelectronic devices. We easily control the skyrmion transport by engineering asymmetric shapes of geometric structures. Here, we present a simple method to describe the underlying mechanism behind the unidirectional skyrmion transport by characterizing the topography of potential energy surfaces from a purely geometric perspective. Our approach enables a deeper physical insight into skyrmion transport manipulation and efficient design of skyrmion-based devices in geometric structures.

49. The hidden value of large-rotor, tall-tower wind turbines in the United States

Abstract

The significant upscaling of wind turbine size (nameplate capacity, rotor diameter, and tower height) has, to date, been driven primarily by a goal of minimizing the levelized cost of energy. But with wind’s levelized cost of energy now comparable with that of other generating resources, other design considerations besides cost-minimization have grown in importance—particularly as wind’s increasing market penetration begins to impose challenges on the electric grid. We find that taller towers and larger rotors (relative to nameplate capacity) can enhance the value of wind energy to the electricity system and provide other “hidden” benefits. Specifically, in regions where wind penetration has reached around 20%, we find a boost in wholesale market value of US$2–US$3/MWh. This is augmented by transmission, balancing, and financing benefits that sum to roughly US$2/MWh. The aggregate potential value enhancement of US$4–US$5/MWh is comparable with a 10%–15% reduction in levelized costs.

50. Solar and wind grid system value in the United States: The effect of transmission congestion, generation profiles, and curtailment

Abstract

The value of electricity generated from wind and solar sources declines as supply increases. This decline in value has varied over time and across regions, indicating that strategies to mitigate value decline will need to be carefully targeted. To help guide development of these strategies, we empirically determine wind and solar value at ∼2,100 plants within United States wholesale markets by using local prices and plant-specific generation profiles. We determine how each plant loses (or gains) value because of its output profile, transmission congestion, and curtailment. In regions where wind or solar account for roughly 20% of electricity generation, its value is 30% to 40% below the regional average value of a flat output profile at all plants. Solar value reductions are most sensitive to output profile and wind value reductions are sensitive to both profile and congestion, region dependent. Curtailment was not a major source of value reduction.
